Abstract:
In this work, the characterization of endoprotease (EP) isoenzymes in peroxisomes is reported for the first time in cell organelles purified from pea leaves (Pisum sativum L.). A comparative analysis of the endo-proteolytic activity in peroxisomes purified from young (15-day-old) and senescent (50-day-old) leaves was carried out. Peroxisomes purified from senescent leaves showed a much higher endo-proteolytic activity than organelles from young plants. A 16 h incubation with exogenous substrates was the threshold time for the detection of a linear increase in the endo-proteolytic activity of peroxisomes from senescent leaves. Three EP isoenzymes (EP2, EP4 and EP5), having molecular masses of 88, 64 and 50 kDa respectively, were found in young plants by using SDS/polyacrylamide-gradient gels co-polymerized with gelatin. However, four additional isoenzymes (EP1, EP3, EP6 and EP7), with molecular masses of 220, 76, 46 and 34 kDa respectively, were detected in senescent plants. All the isoenzymes detected in peroxisomes from both young and senescent leaves were neutral proteases. By using different class-specific inhibitors, the electrophoretically separated EP isoenzymes were characterized as three serine-proteinases (EP1, EP3 and EP4), two cysteine-proteinases (EP2 and EP6) and a metallo-proteinase (EP7), and EP5 might be a metal-dependent serine-proteinase. Moreover, a peroxisomal polypeptide of 64 kDa was recognized by an antibody against a thiol-protease. The serine-proteinase isoenzymes (EP1, EP3 and EP4), which represent approx. 70% of the total EP activity of peroxisomes, showed a notable thermal stability, not being inhibited by incubation at 50 °C for 1 h.

Abstract:
The fact that the specifier of T0 is subject both to the Extended Projection Principle (EPP) and to the Empty Category Principle (ECP) has remained an unexplained accident within Government-Binding Theory. I propose a principled account of this correlation. The EPP is a selectional requirement of functional heads (e.g., T, Top, C) that applies at PF—an instance of p-selection for an overt element. Like all selectional requirements, it applies to the head of the selected phrase, explaining why null heads cannot appear in EPP positions (thus deriving certain representational ECP effects). A wide range of empirical results follow, all unified by the exclusion of null-headed phrases from EPP positions: subject-object asymmetries in the distribution of bare nouns in Romance and sentential complements; failure of certain adjuncts to occur in clause-initial position; resistance of indirect objects to Ā-movement; and phonological doubling of heads of fronted categories. I argue against the agreement/checking view of the EPP and show that only the selectional construal allows a natural explanation of its puzzling properties.

Abstract:
PGE2 is a potent stimulator of renin release. So far, the contribution of each of the four PGE2 receptor subtypes (EP1–EP4) in the regulation of renin release has not been characterized. Therefore, we investigated the effects PGE2 on renin secretion rates (RSR) from isolated, perfused kidneys of EP1−/−, EP2−/−, EP3−/−, EP4−/−, and wild-type mice. PGE2 concentration dependently stimulated RSR from kidneys of all four knockout strains with a threshold concentration of 1 nM in EP1−/−, EP2−/−, EP3−/−, and wild-type mice, whereas the threshold concentration was shifted to 10 nM in EP4−/− mice. Moreover, the maximum stimulation of RSR by PGE2 at 1 μM was significantly reduced in EP4−/− (12.8-fold of control) and EP2−/− (15.9-fold) compared with wild-type (20.7-fold), EP1−/− (23.8-fold), and EP3−/− (20.1-fold). In contrast, stimulation of RSR by either the loop diuretic bumetanide or the β-adrenoceptor agonist isoproterenol was similar in all strains. PGE2 exerted a dual effect on renal vascular tone, inducing vasodilatation at low concentrations (1 nmol/) and vasoconstriction at higher concentrations (100 nmol/) in kidneys of wild-type mice. In kidneys of EP2−/− as well as EP4−/− mice, vasodilatation at low PGE2 concentrations was prevented, whereas vasoconstriction at higher concentrations was augmented. In contrast, the vasodilatatory component was pronounced in kidneys of EP1 and EP3 knockout mice, whereas in both genotypes the vasoconstriction at higher PGE2 concentrations was markedly blunted. Our data provide evidence that PGE2 stimulates renin release via activation of EP2 and EP4 receptors, whereas EP1 and EP3 receptors appear to be without functional relevance in juxtaglomerular cells. In contrast, all four receptor subtypes are involved in the control of renal vascular tone, EP1 and EP3 receptors increasing, and EP2 as well as EP4 receptors, decreasing it.

Objective.Psoriatic arthritis (PsA) has been recognized as a severe erosive disease. However, some patients do not develop erosions. We aimed to determine the prevalence, characteristics, and predictors of erosion-free patients (EFP) as compared with erosion-present patients (EPP) among patients with PsA followed prospectively.Methods.This is a retrospective analysis conducted on patients from the Toronto PsA cohort. Patients with at least 10 years of followup and radiographs were analyzed. Radiographs were scored with the modified Steinbrocker method. Baseline (first visit to clinic) characteristics were used to predict the development of erosions with logistic regression models. To examine the effect of time-varying covariates, Cox regression models were fit for the time to development of erosions from baseline.Results.Among 290 patients, 12.4% were EFP and 87.6% were EPP over the study period. The mean time to development of erosion in the EPP over the course of followup was 6.8 ± 6.1 years. EFP were diagnosed with psoriasis at a younger age compared with EPP. In both models, actively inflamed joints and clinically damaged joints were predictive of the development of erosion, whereas a longer duration of psoriasis at baseline decreased the odds of developing erosion. EPP had a higher percentage of unemployment as compared with EFP at baseline and followup visits.Conclusion.Among patients with PsA followed for at least 10 years, 12.4% never develop erosions. The clinical and radiographic findings can ultimately assist in the stratification of a patient’s prognosis regarding the development of erosions.

Prostaglandin E2 is a major renal cyclooxygenase metabolite of arachidonate and interacts with four G protein-coupled E-prostanoid receptors designated EP1, EP2, EP3, and EP4. Through these receptors, PGE2modulates renal hemodynamics and salt and water excretion. The intrarenal distribution and function of EP receptors have been partially characterized, and each receptor has a distinct role. EP1 expression predominates in the collecting duct where it inhibits Na+ absorption, contributing to natriuresis. The EP2 receptor regulates vascular reactivity, and EP2 receptor-knockout mice have salt-sensitive hypertension. The EP3 receptor is also expressed in vessels as well as in the thick ascending limb and collecting duct, where it antagonizes vasopressin-stimulated salt and water transport. EP4 mRNA is expressed in the glomerulus and collecting duct and may regulate glomerular tone and renal renin release. The capacity of PGE2 to bidirectionally modulate vascular tone and epithelial transport via constrictor EP1 and EP3 receptors vs. dilator EP2 and EP4 receptors allows PGE2 to serve as a buffer, preventing excessive responses to physiological perturbations.

Of various PGs, PGE1 and PGE2 are shown to be the most potent stimulators of osteoclastogenesis in vitro. PGE receptors have been classified into four subtypes, EP1-EP4. Little is known about PGE receptors functioning in bone cells. In this study, using mouse marrow culture, we investigated which PGE receptors are important in osteoclast-like cell (OCL) formation induced by PGE. 11-deoxy-PGE1 (EP2, EP3 and EP4 agonist) stimulated OCL formation potently. Butaprost (EP2 agonist) stimulated it slightly, while sulprostone (EP1 and EP3 agonist) and ONO-AP-324-01 (EP3 agonist) did not. AH23848B (EP4 antagonist) inhibited PGE2-induced OCL formation in a dose-dependent manner. The expression of EP4 mRNA in mouse bone marrow was confirmed by RT-PCR. The results indicate an important role of EP4 in PGE2-induced OCL formation in marrow cultures and suggest therapeutic potential of EP4 antagonists in some clinical conditions with accelerated bone resorption.

Abstract:
Prostaglandin (PG)E derivatives are widely used for treating gastric mucosal injury. PGE receptors are classified into four subtypes, EP1, EP2, EP3, and EP4. We have tested which EP receptor subtypes participate in gastric mucosal protection against ethanol-induced gastric mucosal injury and clarified the mechanisms of such protection. The gastric mucosa of anesthetized rats was perfused at 2 ml/min with physiological saline, agonists for EP1, EP2, EP3, and EP4, or 50% ethanol, using a constant-rate pump connected to a cannula placed in the esophagus. The gastric microcirculation of the mucosal base of anesthetized rats was observed by transillumination through a window made by removal of the adventitia and muscularis externa. PGE2 and subtype-specific EP agonists were applied to the muscularis mucosae at the window. Application of 50% ethanol dilated the mucosal arterioles and constricted the collecting venules. Collecting venule constriction by ethanol was completely inhibited by PGE2 and by EP2 and EP4 agonists (100 nM) but not by an EP1 or an EP3 agonist. Ethanol-induced mucosal injury was also inhibited by EP2 and EP4 agonists. When leukotriene (LT)C4 levels in the perfusate of the gastric mucosa were determined by ELISA, intragastric ethanol administration elevated the LTC4 levels sixfold from the basal levels. These elevated levels were significantly (60%) reduced by both EP2 and EP4 agonists but not by other EP agonists. Since LTC4 application at the window constricted collecting venules strongly, and an LTC antagonist reduced ethanol-induced mucosal injury, reductions in LTC4 generation in response to EP2 and EP4 receptor signaling may be relevant to the protective action of PGE2. The present results indicate that EP2 and EP4 receptor signaling inhibits ethanol-induced gastric mucosal injury through cancellation of collecting venule constriction by reducing LTC4 production.

Federal mandates (e.g., Every Student Succeeds Act [ESSA], 2015) require special educators to use evidence-based practices (EBP) when working with K-12 students. However, for this expectation to become a reality, teacher educators must make changes in educator preparation program (EPP) curriculum, policy, coursework, and clinical experiences (Kolb et al., 2018). The need for changesin EPP clinical experiences has been underscored by the Council for Exceptional Children’s (CEC’s) shift from knowledge to practice-based standards for special educators (CEC, 2020). Real-time performance feedback (PF) delivered via online bug-in-ear (BIE) technology is an EBP (Sinclair, 2020) for coaching and supervising during early, mid, and late clinical experiences. In this article,we offer a rationale for making widespread, digital-age changes to coaching and supervising, through online BIE; provide an overview of relevant research; and offer guidance and recommendations for successful online BIE integration during EPP clinical experiences.

16,16-Dimethyl-PGE2 (PGE2) may interact with one of four prostaglandin type E (EP) receptors, which signal via cAMP (via EP2 or EP4 receptors) or intracellular Ca2+ (via EP1 receptors). Furthermore, EP3 receptors have several splice variants, which may signal via cAMP or intracellular Ca2+. We sought to determine the PGE2 receptor interactions that mediate recovery of transmucosal resistance ( R) in ischemia-injured porcine ileum. Porcine ileum was subjected to 45 min of ischemia, after which the mucosa was mounted in Ussing chambers. Tissues were pretreated with indomethacin (5 μM). Treatment with the EP1, EP2, EP3, and EP4 agonist PGE2 (1 μM) elevated R twofold and significantly increased tissue cAMP content, whereas the EP2 and EP4 agonist deoxy-PGE1 (1 μM) or the EP1 and EP3 agonist sulprostone (1 μM) had no effect. However, a combination of deoxy-PGE1 and sulprostone stimulated synergistic elevations in R and tissue cAMP content. Furthermore, treatment of tissues with deoxy-PGE1 and the Ca2+ ionophore A-23187 stimulated synergistic increases in R and cAMP, indicating that PGE2 triggers recovery of R via EP receptor cross talk mechanisms involving cAMP and intracellular Ca2+.

Abstract BACKGROUND: Cell derived MPs are small membrane vesicles released during cell activation or apoptosis. They express an inside-out membrane, which exposes the negatively charged phospholipid layer outside, allowing clotting factors to be anchored and generate thrombin. Among various species of MPs, RMP (red cell MPs), PMP (platelet MPs), LMP (leukocytes), EMP (endothelial cells) are of special interest. They play an important role in hemostasis, thromboses, and inflammation. MPs mirror early injury of parent cells and are sensitive early biomarkers of underlying disorders. TTP is a microangiopathy mediated by antibody-induced depletion of ADAMTS13, a von Willebrand factor-cleaving protease. Endothelial injury promotes platelet clumping and formation of platelet rich microthrombi in the microcirculation. Subsequent platelet sequestration leads to impaired microcirculation, thrombocytopenia and red cell fragmentation with a microangiopathic hemolytic anemia. Exchange plasmapheresis (EPP) is the standard therapy. It removes antibodies to ADAMTS13, replacing it with ADAMTS13 rich plasma. It is possible that EPP removes thrombogenic MPs to improve the clinical course of TTP. In this study, we investigated MP profiles in active and remission phase of TTP and the effect of EPP on MP profiles. We also aimed to determine if MP profiles may be a useful measure for monitoring clinical course and tracking progress of therapy. METHODS: A retrospective study was conducted evaluating MP assays in patients with TTP. MP profiles were reviewed in acute and remission phases of TTP. Acute phase was defined as thrombocytopenia, clinical evidence of microangiopathy and hemolytic anemia and low ADAMTS13 activity. Remission was defined as sustained normalization of laboratory parameters and no further microangiopathy for at least one month. Patients were studied longitudinally, with MP assays before and after EPP. EMP were measured by CD31+/CD42b− (EMP31), CD62E+ (EMP62); PMP by CD31+/CD42b+ (PMP42) and CD41+ (PMP41). All were measured in platelet-poor plasma by flow cytometry. All MP data are presented in units of x105/µL. The differences in MP patterns among TTP patients in active and remission phases of disease, as well as the effect of EPP on MP profiles were assessed. RESULTS: Among 20 patients with TTP, 8 (40%) were in acute phase and 12 (60%) in remission. An average of 10.7 EPP were performed. The average platelet count prior to EPP was 50.6x103/µL, which increased to 248 x103/µL following the last EPP. ADAMTS13 activity was generally <10% at the onset. For patients in the acute phase, PMPs were low: PMP41 0.32 (±0.11), PMP42 3.11 (±2.56), but consistently increased following EPP to the point of statistical significance at the last treatment: PMP41 1.26 (±0.71), p=0.005 and PMP42 7.65 (±5.03), p=0.039. Moreover, levels were higher in the remission phase for both PMPs, but only statistically significant for PMP41: 1.39, p=0.034. Conversely, EMP62E was initially elevated on presentation, but declined with successive EPP: 6.07 (±3.02) prior to initiation of EPP to 4.5 (±3.1) upon the last day of EPP. Furthermore, this pattern continued into remission, with EMP62E of 2.72 (±2.11), p=0.009. Similar to the trend in EMP62E, RMP was elevated in the acute phase (25.1 [±18.5]) before steadily declining with EPP (19.65 [±12.53]). This progressive drop in RMP persisted for those in remission phase of TTP 10 (±5.42), p=0.015. DISCUSSION/CONCLUSION: Taken collectively, cell derived MP's were found to reflect disease activity and response to therapy. There was a linear correlation between PMP levels and platelet count. Low PMPs in the acute phase reflects the initial thrombocytopenia characteristic of TTP. When EPP was initiated, the disease improved and there was a rise in PMP which mirrored the rise in platelets. This rise was sustained in the remission phase. Conversely, EMP62E and RMP are increased in acute phase TTP due to endothelial activation, microthrombi deposition and red cell fragmentation. After EPP the disease is quiescent, hence the progressive decline of EMP62E and RMP. These results show promising utility of cell derived MP profiles as a clinical tool to surveil chronic TTP patients and to predict disease relapse at an early stage. After EPP is initiated, change in MP Profiles may be used to monitor response to therapy and determine the appropriate time to wean EPP. Disclosures No relevant conflicts of interest to declare.

En el contexto de la administración de nombres de dominio, existe preocupación por NIC Chile respecto de la robustez y del rendimiento de la plataforma de software actual de su Registro. Se predice un aumento de sus canales de inscripción de nombres de dominio en el corto plazo, por lo que mejorar estos atributos es una prioridad para la organización. Este trabajo fue enfocado en el análisis, diseño e implementación de una nueva plataforma de software para NIC Chile, siguiendo un modelo de arquitectura distinto, que separa las responsabilidades de las distintas piezas de software. Junto a la implementación de esta nueva plataforma de software, se crearon herramientas de medición que permitieron realizar una comparativa entre el software de Registro que NIC Chile venía utilizando en producción desde el año 2013 y la nueva plataforma de software producto del trabajo de esta tesis. La evaluación del producto final demostró que la nueva arquitectura permite un incremento en la robustez y rendimiento, además de agregar una mayor flexibilidad para diseñar procedimientos de escalamiento frente a situaciones de alta demanda. Este conjunto de mejoras permitirán que NIC Chile pueda generar las estrategias y procedimientos que conserven el servicio y prestaciones de primer nivel que caracteriza a la organización.

Esta tese discute o sujeito nulo no português do Brasil (PB), em contextos oracionais finitos e infinitivos, com foco no fenômeno denominado controle. O controle é um fenômeno de correferência (obrigatória) entre um sujeito (nulo) de uma oração (subordinada) e um argumento da oração matriz. Esse fenômeno é frequentemente observado, nas línguas em geral, em contextos não finitos (entre os quais, incluem-se as orações infinitivas). Diversos estudos, entretanto, mostram que no PB esse fenômeno é observado, também, em contextos oracionais finitos, principalmente em orações encaixadas indicativas, quando o sujeito nulo é de 3ª pessoa. Essa propriedade do PB destaca-se no âmbito das línguas românicas (de sujeito nulo), já que nessas línguas o sujeito nulo (de qualquer pessoa) recebe uma interpretação definida/específica. O objetivo desta tese é prover análise que explique a ocorrência do controle nesses contextos. Para tanto, adota-se o quadro teórico da gramática gerativa, mais especificamente aquele desenvolvido por Manzini & Savoia (2005, 2007), o qual unifica os níveis de análise morfológico e sintático. A proposta desenvolvida neste trabalho consiste em considerar que o afixo de pessoa realiza o sujeito no PB e que a interpretação de controle encontrada em orações finitas decorre das especificações lexicais do morfema de concordância de 3ª pessoa, o que leva a crer que há, nessa língua, uma cisão de pessoa. Essa cisão, que alinha, de um lado, 1ª e 2ª pessoas, opondo-as, por outro lado, à 3ª pessoa, abrange outros contextos linguísticos, especialmente aqueles que envolvem a realização de funções gramaticais da oração por pronomes. A análise de que o sujeito é realizado pelo afixo de concordância número-pessoal é estendida ao infinitivo flexionado. Em relação às orações infinitivas, propõe-se, ainda, que o morfema -r é especificado para modo irrealis. Nesse caso, se o infinitivo é impessoal, considera-se que o argumento EPP desse predicado não é realizado sintaticamente, o que significa que esse argumento é uma variável a ser valorada (na interface interpretativa). A valoração dessa variável pode se dar por meio da ligação com um DP na oração matriz ou da ligação por um operador responsável pela interpretação genérica. A interpretação ligada é obrigatória em um subgrupo de orações infinitivas. Isso se deve, por hipótese, à aplicação do fenômeno da reestruturação, observado em contextos de dependência entre oração encaixada (sem tempo) e oração matriz. O controle, então, é tratado como decorrente do fato de o argumento EPP de um predicado ser uma variável. _________________________________________________________________________________ ABSTRACT
This thesis addresses the syntax of null subjects in BP in finite and non-finite contexts, focusing on the phenomenon of control. Control consists of an obligatory coreferential relation between the null subject of an embedded clause and an argument in the matrix clause. This phenomenon is pervasively found crosslinguistically in non-finite contexts, among which the infinitive clauses are found. Nevertheless, various studies show that in BP this phenomenon is also found in finite clauses, mainly in indicative ones, whenever the subject is a null 3rd person one. This property of BP is quite isolated among (null subject) Romance languages, given that in the latter the null subject always receives a definite/specific interpretation, regardless of the person specification. The aim of this thesis is to provide an analysis for the occurrence of control in these contexts in BP, as opposed to the other contexts involving control, which are also found in the other Romance languages. In the analysis, the framework developed in Manzini & Savoia (2005, 2007) is adopted, which is based on the assumption that the morphological and syntactic levels are unified. The analysis presently proposed is that the person affix realizes the subject in BP, the control interpretation found in finite clauses being determined by the lexical specifications of the 3rd person agreement morpheme, which implies that a person split is found in this language. This split, which groups 1st and 2nd person, on the one hand, and 3rd person, on the other hand, is found in other syntactic contexts, particularly those involving the realization of grammatical functions by pronouns. The approach according to which the subject is realized by the person-number affix is extended to the contexts involving the inflected infinitive. Regarding infinitive clauses, it is further proposed that the morpheme –r is specified for irrealis mood. Accordingly, if the infinitive is non-inflected, it is assumed that the EPP argument of this predicate is not syntactically projected, which implies that this argument is interpreted as a variable, which will be assigned an interpretation at the appropriate interface. The variable will receive a value through binding either by an argument in the matrix clause or by a generic operator. The bound interpretation is obligatory in a subset of infinitive clauses. By hypothesis, this is so because of the application of restructuring, found in contexts of tense dependency between the embedded clause (which is not marked for tense) and the matrix clause. Control is thus analysed as arising in contexts in which the EPP argument of a predicate is a variable.

I denna kvalitativa studie ombads 5 psykodynamiskt inriktade terapeuter att i skrift beskriva sin upplevelse av journalskrivning. Materialet analyserades med hjälp av EPP-(Empirical Phenomenological Psychological) metoden. Resultatet lyfter fram generella kännetecken och särskiljande drag inom upplevelsen av journalskrivning. Gemensamt för terapeuternas upplevelse är att journalskrivandet har olika funktioner. Det är ett arbetsverktyg för att kommunicera med andra vårdgivare, det omfattas av vissa styrfaktorer och en upplevd osäkerhet om journalskrivandets regelverk. Det egna journalskrivandet uppvisar en hög värdering av den personliga upplevelsen och känslan av att utlämna patienten i journalen. Däremot skiljer sig terapeuterna åt i sättet att hantera känslor av konflikt inom journalskrivandet vilket påverkar den egna upplevelsen av journalskrivning. Två upplevda alternativ att hantera konflikt presenteras. I den ena gruppen försöker terapeuterna att omforma journalen så att den bättre passar den egna terapeutiska identiteten. När det inte längre är möjligt uttrycks en känsla av att något som inte går att ersätta försvinner. I den andra gruppen ökar terapeuterna sin reflektion och bearbetning av de delar inom journalskrivningen som terapeuten upplever orsakar konflikt, vilket bidrar till en känsla av att något positivt tillförts den terapeutiska identiteten.
In this qualitative study 5 psychotherapists with psychodynamic specialization were asked to reflect on their experiences concerning the writing of patient records. This material was analyzed using the so-called EPP-method (empirical phenomenological psychological method). The results show both general and distinctive features in how different therapists feel about their patient records routines. All agree that the process of writing patient records contains a number of different functions. It is a tool to communicate with other medical staff and facilities; it is influenced by certain control factors and also by experienced uncertainty concerning the formal rules related to the routines of patient records. Record routines show a high degree of evaluation of personal emotions and experiences together with a fear of revealing too much information about a patient. On the other hand, there is a substantial distinction in how different therapists handle their feelings of conflict related to medical records. The study presents two different possible ways of handling such conflicts. One group of therapists try to reshape the medical journal to make it better adjusted to their personal therapeutic identity. When that becomes impossible they express a feeling of something being lost. In the second group the therapists work harder with those parts in patient record routines that are causing them problems. This results in a feeling that something valuable is being added to their therapeutic identity.

Abstract:
In this paper, a cathodic electrolytic plasma process (EPP) is introduced as a new corrosion prevention method. It is a green and cost-effective method of metal surface treatment. The EPP procedure and the EPP-treated 1018 steel are described in detail. On EPP treated surfaces, a spontaneous pattern due to localized melting and quenching could be obtained. Scanning electron microscopy and energy dispersive X-ray spectroscopy were used to characterize the special surface morphology and chemical patterns on EPP resultant features. Standard corrosion tests were conducted in EPP-treated samples. The corrosion analysis indicates that the EPP treatment could effectively improve the uniform corrosion of 1018 steel in 3.5% NaCl solution. With longer time of EPP treatment (15 minutes), the passivation was significantly extended and hence the localized corrosion resistance is improved. Also, the higher corrosion resistance was found to be sustainable (in 48 hours corrosion test, NaCl solution) on all the EPP treated samples.

Abstract This paper discusses the development of design charts for creep-fatigue evaluation using an integrated elastic-perfectly plastic analysis (EPP) and Simplified Model Test (SMT) approach. The creep-fatigue damage evaluation approach in current ASME Section III, Division 5 rules requires separate evaluation of creep and fatigue damages and then considers the effect of creep-fatigue interaction with a creep-fatigue interaction diagram, also known as D-diagram. The EPP+SMT approach avoids the use of D-diagram by directly comparing the strain range determined from an EPP analysis with design charts, conceptually developed directly from creep-fatigue and SMT tests, to determine the creep-fatigue damage. The EPP+SMT approach provides much simpler creep-fatigue design evaluation process compared to current methods and is also expected to reduce the over-conservatism in current methods. Generating the EPP+SMT design charts requires extrapolating high strain range, short hold time test data to low strain range and long hold times, typically experienced by structural components in high temperature nuclear service. This work investigates several extrapolation approaches in the development of methodologies for constructing EPP+SMT design charts for Alloy 617 at temperatures between 800°C and 950°C. The design charts are validated using failure data from pressurized SMT tests and by comparing the EPP+SMT method with current design methods for representative high temperature nuclear reactor components.

An ASME Section III Division 5 code case, N-861, for the evaluation of strain limits based on the elastic-perfectly plastic (EPP) methodology has recently been published. A key feature of the EPP methodology is the application of the EPP finite element analysis method with a pseudo yield stress to bound component response under elevated temperature cyclic service. The simplified inelastic approach in Division 5, Appendix HBB-T that is based on the elastic analysis results is not applicable at the elevated temperature range where creep and plasticity cannot be distinguished and unified viscoplastic model is required to describe the deformation behavior. The EPP strain limits code case overcomes such limitations. It also has the distinct advantage that stress classification, which is required by the simplified inelastic approach, is not needed. Thus, it is ideally suited for modern-day finite element technology. The conservatism of the EPP strain limits code case was verified for some simple geometries. In this paper, a viscoplastic constitutive model calibrated to the experimental data for 316H stainless steel is used to conduct a full inelastic analysis. The calculated strain accumulation is compared with that obtained from using the EPP code case approach.

Abstract:
Limit load solutions obtained by elastic-perfectly plastic finite element analysis (EPP-FEA) are compared to results of tests of low-alloy steel perforated plate geometries loaded to full plastic collapse. Results are given for two plastic-collapse tests of flat circular disks with circular penetrations arranged in a triangular pattern and drilled normal to the surface of the plate. The ligament efficiency (minimum distance between holes divided by the distance between the centers of the holes) of the pattern is 0.32 and the plate thickness is 2.39 inches (60.7 mm). The tests were designed so that a transverse load generated plastic collapse in the outer row of penetrations due to a combination of transverse shear and in-plane bending. Limit-load solutions were obtained using EPP-FEA with small-strain, small-deflection linear geometry assumptions. Two FEA models are used: one where the perforated region is modeled using an equivalent solid plate (EQS) representation and another where each hole is explicitly modeled by FEA. The results presented in this paper demonstrate that the deformation patterns produced by the EPP-FEA solutions match exactly with the deformation patterns produced by the test. The EQS-EPP FEA solution is about 15% lower than the explicit-hole EPP-FEA solution. Using one-third the actual ultimate strength of the material as the strength parameter in the limit load calculation produces a calculated limit load that is greater than a factor of three less than the mean measured plastic-collapse load obtained in the tests. This paper adds to the qualification of the use of limit-load solutions obtained using small-strain, small deflection EPP-FEA programs for the calculation of the limit load for perforated plates.

We carry out punching experiments on low-density elastic polyether polyurethane (EPP) foams. The punch is rigid and wedge-shaped, and the specimens are either cubic or brick-shaped. The experimental results display some striking features, most notably a mechanical response that remains linear up to a penetration of the punch of about 40% of the height of the specimen. At higher penetrations, the mechanical response turns abruptly nonlinear: for the cubic specimens, there is a momentary loss of stiffness; for the brick-shaped specimens, there is a sizable increase in stiffness. We formulate a simple theoretical model of the punching of low-density EPP foams. In this model, a cell of the microstruture of an EPP foam behaves as a bistable elastic structure under compression, so that there are two preferred values of strain corresponding to two configurational phases of the foam. We use the model to explain the most salient experimental observations. We also formulate a micromechanical, mean-field model of EPP foams, implement the model in a general-purpose finite element code, and carry out computational simulations of the punching experiments. The computational results are in good accord with the experimental results, and they allow us to verify some of the assumptions on which the theoretical model is predicated.

Abstract:
Design approaches using elastic perfectly-plastic (EPP) analysis have recently been approved as Code Cases for the Section III, Division 5 design of high-temperature nuclear reactor components made from austenitic stainless steel. These methods bound the ratcheting strain and creep-fatigue damage accumulated over the life of a component with a simplified, elastic-perfectly plastic analysis using a special pseudo-yield stress — often not equal to the true material yield stress. The austenitic materials specified in the existing Code cases are cyclic-hardening for all allowable operating temperatures. However, other Section III, Division 5 materials, such as Grade 91 steel, are cyclic softening at expected advanced reactor operating temperatures. This work describes the extension of EPP methods to cyclic softening materials through the use of a postulated saturated material state and softening factors to be applied to the pseudo yield stress. We demonstrate the conservatism of the modified EPP method against a series of inelastic simulations of two bar tests, using a constitutive model that captures work and cyclic softening.
