Full textWe want to analyze the responses of biological systems to the introduction of perturbations and spatio-temporal modulations. More specifically, in order to develop innovative strategies for the study of biological systems, we propose to use microfluidic tools. We design adapted microsystems that can locally influence biological behaviors, so that a macroscopic experimenter can control the external environment of biological objects whose scale is microscopic. This engineering strategy is generic and multidisciplinary. In this thesis, it has been implemented in two collaborative projects, on one hand, on the scale of the E.coli bacterium and on the other hand on that of the embryo of mouse at an early stage post-implantation. The selected study objects are characteristic in many respects of the biological fields concerned: size, representativeness, complexity.We extended our expertise in fluidic device design and manufacturing to the service of the DISCO beamline of the synchrotron SOLEIL and the IRIBHM mouse embryology team. The key point of my work has been to design and manufacture reusable microfluidic tools for generic research, which allow biologists to dispense with the use of a clean room.More precisely, the project of microbiology at SOLEIL had for object the study of the influx and the efflux of antibiotic molecules in bacteria. To do this, we have developed a reusable device for immobilizing microorganisms and changing their chemical environment during UV imaging on epifluorescence microscopy. This study is carried out using two typical partners: the Escherichia coli bacterium and a drug from the fluoroquinolone family. The embryology project relied on the localized electroporation of nucleic acids within mouse embryos and the monitoring of cellular migrations.In this thesis, we have developed not only reusable micro-devices but also experimental protocols adapted to the use of these miniaturized instruments.More precisely, the microbiology project at SOLEIL focused on the influx and the efflux of antibiWe have developed a reusable device for immobilizing those microorganisms and changing their chemical environmentduring UV imaging on an epifluorescence microscopy. Full textLe gène Evi1 code pour un facteur de transcription à doigts à zinc notamment exprimé dans le métanéphros des vertébrés supérieurs et jouant un rôle majeur mais mal compris dans le développement. Chez l’embryon de xénope, le gène Evi1 est exprimé dès la fin de la neurulation dans la région ventro-postérieure de l’ébauche pronéphrique à l’origine du tubule distal et du canal. Son expression est inhibée par le facteur de transcription xWT1 et l’activation de la voie de signalisation Notch. Via des expériences de surproduction de la protéine Evi1 sauvage ou d’une fusion Evi1-VP16, fonctionnant de manière antagoniste à la protéine sauvage, nous avons montré que la protéine Evi1 joue un rôle important dans la néphrogenèse précoce en inhibant la formation du glomus et des tubules proximaux dans les cellules à l’origine du segment distal du néphron.
Afin de déterminer l’importance du facteur de transcription xWT1 dans la régulation de l’expression du gène Evi1 ainsi que dans la formation du glomus, des expériences de sous-expression ont été réalisées. Ces expériences montrent que la sous-expression de xWT1 inhibe la formation du glomus mais n’induit cependant pas d’expansion de l’expression du gène Evi1 ni celle d’autres marqueurs du pronéphros, suggérant l’existence d’au moins un autre facteur répresseur exprimé au niveau de la couche médiane du mésoderme pronéphrique.
Ensuite, nous avons comparé les profils de gènes codant notamment pour des protéines spécialisées dans le transport de solutés dans les reins embryonnaires de xénope et de poisson zèbre. Nos résultats montrent que le pronéphros du poisson zèbre, bien que présentant une structure uniforme, peut être subdivisé en quatre différents segments. Ces observations suggèrent que les reins embryonnaires du xénope et du poisson zèbre présentent des organisations similaires.
Dans la dernière partie de ce travail, nous avons entamé l’étude du promoteur du gène CLC-K, codant pour un canal chlore exprimé dans du segment distal du néphron, par la réalisation d’embryons transgéniques de xénope. Les résultats préliminaires obtenus indiquent qu’un fragment de 11kb du promoteur du gène CLC-KB humain est suffisant pour diriger une expression correcte du transgène chez le xénope.

Full textNumerous studies in the mouse embryo have characterised morphological events during preimplantation development. However, molecular mechanisms driving cell fate ad differentiation are not yet well understood. The objective of our group is to analyse the mechanisms of cell differentiation by focusing on primitive endoderm (PrE) formation, the second cell type to differentiate in the embryo. PrE is an extraembryonnic tisssue that appears before implantation. Although PrE does not contribute to the embryo proper, it is necessary for both survival and correct development of the embryo. In order to identify new genes implicated in PrE formation, I analysed microarrays performed on early embryonic stages. This resulted in the identification of new early genes that are potentially specific to PrE. Using RT-PCR and in situ localisation, I analysed two of the selected genes : Lrp2 and Dkkl. A detailed study of LRP2 expression between E2. 5 and E4. 5 gave rise to a model of epithelial differentiation. In parallel with this work, I developed a new method for in vivo functional analysis. It combines electroporation, RNA interference and embryo culture and open perspectives new for studing early embryogenesis

Full textThe European sea bass Dicentrarchus labrax is a euryhaline species which usually spawns in seawater. Due to its life cycle that includes migrations to lagoon and estuaries, young stages can be exposed early to salinity variations. Osmoregulatory patterns are well known in prelarvae, larvae and adults D. labrax in seawater (SW, 38) and in dilute seawater (DSW, 5), but the possibility and mechanisms of embryonic osmoregulation are still unknown. The goal of this study was to investigate the occurence of the omoregulatory function in the embryos of D. Labrax.First, the embryonic development of the different osmoregulatory sites was described, focusing on the digestive system including the pharynx and the first gill slits. The formation of these structures is initialized at stage 12 somites (S) and was described throughout the opening of the mouth five days after hatching.Secondarily, the time and the location of the occurrence of the first osmoregulatory cells, the ionocytes were followed. These cells were identified at stage 12S on the yolk sac membrane and at stage 14S in the first gill slits and in the posterior primitive gut. The functionality of these cells was studied, using immunostaining of the main ionic transporters involved in osmoregulation [the Na+/K+ ATPase (NKA), the Na+/K+/2Cl- cotransporter (NKCC) and the chloride channel (CFTR)], and through ultrastructural investigations. Potentially functional ionocytes are present from stage 25S in the yolk sac membrane and in the gut, but gill slits ionocytes are not fully functional at hatching. Passive drinking is suspected to regulate water balance in D. labrax.Finally, the embryonic osmoregulation in SW and DSW was investigated. Nanoosmometry measurements of the embryonic fluids demonstrated some capabilities of hyper- and hypo-osmoregulation. However, in DSW, qRT-PCR and imunostaining of NKA, NKCC and CFTR, reveal that hyper-osmoregulatory mechanisms can only limit ion loss but are not efficient enough to allow a full acclimation at this early life stage

Full textEarly during development, the digestive tract is a simple, uniform, tubular structure, and the region-specific differentiation along its antero-posterior axis is driven by a reciprocal signaling between the mesoderm and the endoderm. In order to identify the molecular mechanisms that control the differentiation of the visceral mesenchyme in the vertebrate stomach, we developed microarrays to analyze the gene expression profiles of undifferentiated and differentiated avian stomachs. We identify Scleraxis, a basic-helix-loop-helix transcription factor, as a new marker of stomach mesenchyme and find that expression of Scleraxis defines the presence of two tendons closely associated to the two visceral smooth muscles. Using targeted gene misexpression, we show that FGF signaling is sufficient to induce Scleraxis expression and to establish two tendon domains adjacent to the smooth muscle strucutres. We also demonstrate that tendon organization is pertubed by altering Scleraxis expression or function. Moreover, using cells culture experiments, we find taht undifferentiated stomach mesenchyme can give rise to both SMCs and tendon cells. These data show that upon FGF activation, selected stomach mesenchymal cells are primed to express Scleraxis and to diffrentiate into tendon cells. Our discovery of the role of Scx in tendon differentiation during stomach organogenesis is unexpected and novel, and led to the publication of e research article in development on which I am first author. Recently we highlighted the expression of Fz8, a receptor of the Wnt pathway, in these tendinous structures, and are now characterizing how this pathway contributes to their correct development

Full textI analyzed the function of two genes, Angiomotin like 2 (Amotl2) and FrzA (SFRP1, Secreted Frizzled Related Protein 1) and demonstrated that they play an essential role in controlling the cell fates and cell movements during the embryogenesis of the zebrafish (Danio rerio). Amotl2 is involved in the control of embryonic cell movements. I demontrated that Amotl2 interacts preferentially with and facilitates outward translocation of the phosphorylated c-Src, which may in turn regulates the membrane architecture. Amotl2 is under the control of the FGF signaling pathway and this gene is also required for the patterning of the embryo. Preliminary data suggest that Amotl2 is required for the secretion of Wnt8. FrzA is also required for proper cell movements and embryo patterning. Analysis of FrzA function indicates that this gene negatively regulates maternal β-catenin signaling. FrzA appeard to act in a paracrine manner and acts at short range. I established that FrzA acitivity is different from those of FrzB and DKK1. Furthermore, I showed that the inhibition of the maternal β-catenin signaling by FrzA is mediated by the Frizzled 2 receptor

Full textLa première étape de cette étude a consisté en des analyses morphologiques et morphométriques afin de vérifier les perturbations morphologiques communes entre les rats Dumbo et les syndromes malformatifs humains :la brièveté des os zygomatique, maxillaire, mandibulaire et la position basse des oreilles. Ces analyses ont été réalisées sur les squelettes embryonnaires âgés de 16 jours à 21 jours de rats Dumbo et Wistar à l’aide d’une coloration in toto au Bleu Alcian – Alizarine. La deuxième étape de cette étude consistait en une analyse cytogénétique. Pour ce faire, nous avons établi le caryotype du rat Dumbo et nous l’avons comparé avec le caryotype du rat Wistar. L’étape suivante fut de procéder à l’analyse histologique des malformations crânio-faciales chez le rat Dumbo en observant la chondrogenèse pendant la morphogenèse crânio-faciale. Enfin, l’examen de l’expression des gènes Msx1 sens (S) ,Msx1 antisens (AS) et Dlx1 dans l’extrémité céphalique des rats Dumbo a été réalisé par les techniques de RT–PCR (Reverse Transcription Polymerase Chain Reaction method). Des estimations semi-quantitatives ont été validées en utilisant des dilutions ADNc du rat Wistar. Des densitométries de la densité d’amplicons fluorescence ont été réalisées à l’aide du logiciel VilberLourmat Bio1D software.
Les résultats obtenus ont permis de caractériser de manière précise les malformations crânio-faciales chez le rat Dumbo.

Full textThis study combines experimental, analytical and numerical approaches on the role of mechanics in lung development. We show that an increase of the lung internal pressure, by clamping embryonic lung trachea in culture, induces twice as many branching events and a three fold increase in the branches growth speed as compared to non clamped lungs. This raise of the lung internal pressure affects the cellular expression of key lung morphogenesis proteins. From these results, a model, linking genetics and mechanical factors, is presented. Numerical simulations, using the finite elements method, reproduce qualitatively the three steps (elongation, stop and branching) of branches growth and the localisation of development proteins with a good agreement to the experiments. By taking into account mechanical aspects, we establish a mecanogenetic model of lung morphogenesis

Full textA cDNA-AFLP approach was used to identify Linum usitatissimum (flax) genes that are specifically expressed during the seed maturation process in this specie. Flax was chosen because its extended embryo maturation phase when compared to the Arabidopsis model. The cDNA-AFLP reactions were performed on flax seed harvested at different developmental stages, from post-morphogenesis embryo growth phase to mature seed, using a vegetative control. This screen allows us to isolated 487 cDNA-AFLP tags that, when compared to the publicly available databases, revealed for the half of them a clear similarity with known plant sequences. The results obtained confirmed the accuracy of the approach as, for example, a ABI3 flax ortholog was identified. Our interest focused mainly on sequences presenting, when translated, a strong similarity with factors involved in the control of expression, either at the transcriptional or even post-transcriptional and translational levels. On the basis of their strong similarity with their Arabidopsis putative counterparts, 13 of these flax sequences were selected. Using a real-time RT-PCR approach, their embryo specific expression and kinetics were confirmed. Among these EST, two flax paralogs of the non-LEC1 type AHAP3 were shown to be expressed in the embryo procambium as well as in its meristems using an in situ hybridization approach. Moreover, we show that their Arabidopsis orthologs are also specifically expressed in the embryo at the maturation stage. In the same way, more investigations were done on other genes of interest similar to a MYB-factor, an amino acid permease and a prolyl endopeptidase

Full textNodal signalling plays a crucial role during germ layer formation in the vertebrate embryo. To identify target genes of the Nodal pathway in the zebrafish, I have established a subtractive cloning approach. Quantitative analysis of the subtracted probe pools using macroarrays that contain a normalised cDNA library led to the identification of 132 genes enriched by activation of the Nodal pathway. Among these genes are proteins involved in cellular metabolism and the cytoskeleton, which suggests an important role for this signalling pathway in cell physiology during gastrulation. Further genes identified include molecules implicated in different signalling pathways as well as transcription factors. I studied more in detail one of these factors, a novel sox gene family member, and demonstrated that it is disrupted in the casanova mutant which is blocked in a crucial step of endoderm development. Other genes cloned are the anti-dorsalizing morphogenetic protein, a BMP family member implicated in organizer formation, and the helix-loop-helix gene id3. A similar approach was applied to identify genes specifically expressed in the notochord and the floor plate of the neural tube, the formation of which depends on Nodal signalling in the zebrafish. Cells were isolated from embryos expressing GFP under the control of tissue specific promoters and a pool of clones enriched in notochord and floor plate specific genes was obtained. More than 650 clones have been tested for expression using a robot-aided in situ approach, allowing the identification of coexpression groups, including a notochord/floor plate/hypochord group comprising enzymes involved in the collagen synthesis and maturation pathway

Full textJaw morphogenesis is a complex process involving the migration and differentiation of cephalic neural crest cells (CNCCs) and mesodermal cells to the first pharyngeal arch (PA1). Before reaching their destination CCNCs are not yet engaged in specific morphogenetic pathways. Molecular signalling and cellular interactions occurring during PA1 colonization determine the genesis of facial structures. Endothelin-1 (Edn1) signaling is necessary and sufficient to specify “lower jaw” identity; the mechanism through which this specification takes place is not yet completely elucidated. Intake of retinoic acid (RA) or of its precursor, vitamin A, during early pregnancy is associated with increased incidence of craniofacial lesions. Before my thesis, the origin of these teratogenic effects was enigmatic as in cranial neural crest cells (CNCCs), the RA-transduction pathway is not active. In the first two articles we show that, in mouse and Xenopus laevis, RA acts on the signalling epithelium of PA1 reducing the expression of Edn1 and Fgf8. Depending on the time of administration and on the dose and length of treatment, RA reduces progressively the expression of these two molecular signals. Edn1 and Fgf8 are instrumental in activating Dlx genes in incoming CNCCs, thereby triggering the morphogenetic programs, which specify different jaw elements. We report that, RA treatments provokes dramatically different craniofacial malformations when administered at slightly different developmental times within a narrow temporal interval corresponding to the colonization of the 1st PA by CNCCs. The third article provides new insight into the spatio-temporal and gene-dosage dynamics of the jaw morphogenetic process proposing a link between the alteration of this process and the origin of human malformation collectively known as "First Arch Syndromes". This thesis contributed also to five other studies on the role and regulation of Dlx5 and Dlx6 during osteogenesis, limb development, genital tubercle morphogenesis and Leydig cells differentiation

Full textThe study of early patterning mechanisms in a chondrichtyan species, the catscharkScyliorhinus canicula, has to goal to gain insight about their evolution in gnathostome. The results show a biphasic endoderm specification, leading to extra‐embryonic and embryonic component respectively, as in amniote. The early step of neuroectoderm differentiation exhibits a pre‐neural state, with a molecular identity close to amniote. These works highlight also that early catshark embryo is a suitable model for quantification of signaling pathway activities. All these data suggest a deep unity of early development mechanisms in gnathostomes and shed in light the interest of catshark model to study their mode of evolution
