Full textRetinal ciliopathies are a group of rare diseases caused by mutations of ciliary genes. Defects in ciliary genes can cause defects in proteins traffics and induces apoptosis of photoreceptor cells caused by stress of the endoplasmic reticulum (ER) .We studied retinal ciliopathies by mice models, Leber congenital amaurosis, Xlinked retinitis pigmentosa, Bardet-Biedl syndrome and Alström Syndrome. The Bbs1-/-, Bbs10-/- and CEP290-/- mice exhibited a decrease in retinal function caused by ER stress. Rd9/y and Alms1foz/foz mice showed a late onset and a low rate of retinal degeneration and they could be caused by other mechanisms. The GV-Ret treatment based on ER stress could save both the function and morphology of the retina in BBS mice

Full textThe present Thesis intends to give a new approach to the reading of texts from the period known as Baroque and, for that, it was necessary to create new terms that corresponded to this expectation: the iconophotology and photographic poems. In order to enable a reader to read and understand texts of that period, he would have to have access to a signical key to which only readers of that moment had access: the iconologies. However, such reference has been lost; therefore, we have to replace it by another one, from the imagetic-photographic collection we have, which is called iconophotological. With this premise, it will be possible to read, under the contemporary point of view (not under the baroque point of view), the so- called photographic poems.

Full textThe present dissertation aims to study the term "Emblematic subjects" as a theoric contribution to the Theory of the Identity in construction, sustained in the syntagma identity-metamorphosis-emancipation proposed by Ciampa (1987). According to a survey carried out during the process of this work, the term "Emblematic subjects" have been used and built since 1996 in discussions at NEPIM (Center of Study and Research of Identity-Metamorphosis) of Pontifical Catholic University of São Paulo (PUC-SP). To carry out this study, we began, so, with academic productions of NEPIM that have had completed the term "Emblematic subjects" and, as a methodology, we used the history of life contained in the eight academic works that served as a base for short account that are presented here as contextualization and illustration of what is discussed now. This work designed in the syntagma identity-metamorphosis-emancipation proposed by Ciampa (1987) and the arising from productions of NEPIM, and has as a theoric reference Jürgen Habermas and Ciampa, especially the post-conventional subject of Habermas (1983) and the "come to be" and "be to itself" of Ciampa (1987). As we treat it like a theoric work, this dissertation ends with the final considerations broaching synthetically, what has been discussed throughout the work such as exposing inherent gaps to the academic production as possible spaces of appropriation of the Academy
A presente dissertação visa estudar o termo sujeitos emblemáticos como contribuição teórica à Teoria de Identidade em construção sustentada no sintagma identidade-metamorfose-emancipação proposto por Ciampa (1987). Segundo pesquisa realizada durante a construção desse trabalho, o termo sujeitos emblemáticos tem sido usado e construído desde 1996 em discussões no NEPIM (Núcleo de Estudos e Pesquisa em Identidade-Metamorfose) da Pontifícia Universidade Católica de São Paulo (PUC-SP). Para efetivar esse estudo, parte-se então das produções acadêmicas do NEPIM que tenham contemplado o termo sujeitos emblemáticos e, como metodologia, utilizou-se as histórias de vida contidas nos oito trabalhos acadêmicos que serviram de base para breves relatos que são aqui apresentados como contextualização e ilustração do que ora é discutido. Esse trabalho pauta-se no sintagma identidade-metamorfose-emancipação proposto por Ciampa (1987) e nas produções oriundas do NEPIM, tendo como referenciais teóricos Jürgen Habermas e Ciampa, especialmente o sujeito pós-convencional de Habermas (1983) e o vir-a-ser e o ser-para-si de Ciampa (1987). Por tratar-se de um trabalho teórico, essa dissertação se encerra com as considerações finais abordando sinteticamente o que fora discutido ao longo do trabalho, bem como expondo lacunas inerentes à produção acadêmica como espaços possíveis de apropriação da Academia

Full textThe purpose of this dissertation is the analysis of some aspects presented in Conrad Gesner´s Historia animalium (1551-1558), such as some information included in animal´s descriptions, its sources and images, as a vehicle of knowledge transmission. We intend to demonstrate that the content in Gesner´s work, as well as in his contemporary fellows, showed particular features which were consisted not only of anatomical and physiological animal´s descriptions, but they were especially comprised of information which today would be considered additional, as for example, what the ancients would say about each animal, the legends, the fables, its use in medicine, in culinary, in art, etc. All this correlated information was called, by some scholars of this period, the emblematic world view and they determined the scope of animal History in the sixteenth century. In order to demonstrate the interrelated information in Gesner´s writings, we selected not only his writings but also some works of other writers. One of them is Andrea Alciati´s Emblematum Libellus (1531), to which is attributed the dissemination of the emblems - a group of text and image designed to be deciphered, bringing intricate topics such as religion, love, betrayal, politics, moral, among others, which were widely covered in Gesner´s work. We also selected some examples of medieval bestiary to demonstrate how animals were described and illustrated in this kind of work, as well as Ovid´s (43 a.C 17 d.C) Ars Amatoria and the Methamorphoses, aiming at investigating this type of literature and, in later editions, the kind of images involving animals in these poems, in order to accomplish the case study about the mythical beast known as the Minotaur
O objetivo dessa Dissertação é analisar alguns aspectos na obra História animalium (1551-1558) de Conrad Gesner (1516-1565), entre eles, o tipo de informação contida na descrição dos animais, algumas de suas fontes e as imagens como via de transmissão de conhecimentos. Nossa intenção é demonstrar que o conteúdo da obra elaborada por Gesner, assim como por alguns de seus contemporâneos, apresentava características particulares e era composta não apenas por descrições acerca da anatomia e fisiologia das bestas, mas eram especialmente carregadas de informações adicionais, como por exemplo, o que os antigos diziam sobre cada animal, lendas, fábulas, uso na medicina, na culinária, na arte, etc. Todas essas informações interligadas faziam parte de uma rede de conhecimentos chamada por alguns estudiosos desse período de visão emblemática de mundo e de natureza, que caracterizou a História dos animais no século XVI. Para demonstrar algumas informações inter-relacionadas na obra de Gesner, selecionamos, além de seus escritos, trabalhos de outros autores. Um deles foi o Emblematum Libellus (1531) de Andrea Alciati (1492-1550), ao qual é atribuída a difusão dos emblemas - um conjunto de texto e imagem criados para serem decifrados, que traziam intrinsicamente mensagens de diversos temas como religião, amor, traição, política, moral entre outros, e que foram amplamente difundidos nas descrições de Gesner. Selecionamos também alguns exemplos de bestiários medievais para demonstrar como os animais eram descritos e representados nesse tipo de trabalho, bem como A arte de amar e as Metamorfoses de Públio Ovídio (43 a.C 17d.C) para averiguar o tipo de literatura, e em edições posteriores, de imagens que envolviam animais contidos nesses poemas, peculiarmente para realizar o estudo de caso sobre a besta mítica conhecida como minotauro. Contudo, foi possível expor ao longo do nosso trabalho o tipo de informação trazida nos textos e nas imagens das diferentes obras selecionadas

Full textLas tramas de Billy Budd y La hoguera de las vanidades están organizadas íntegramente alrededor de un juicio. En Los papeles póstumos del Club Pickwick, el proceso es una parte importante de la obra, pero también existen aventuras relacionadas en las que participan los diversos personajes. En los tres juicios se juzga a un inocente. En Los papeles póstumos del Club Pickwick, el autor busca presentar el funcionamiento real del sistema legal, en el cual el modus operandi de abogados inescrupulosos, quienes emplean únicamente métodos tramposos y fraudulentos, determina el origen y el resultado del proceso. En Billy Budd, un inocente es condenado a muerte para preservar un supuesto interés mayor: el bien común. En La hoguera de las vanidades, factores políticos, intereses personales, resentimientos y otros elementos de carácter mundano determinan el resultado del proceso. En los tres casos, el mecanismo de relojería que parece ser el proceso es totalmente sobrepasado por factores externos al mismo.

Full textOften forgotten comparing with the splendours of the court by which the dukes of Burgundy of the House of Valois exalted their power and wealth, war was nonetheless an instrument of political and symbolic communication just as important to these princes: a tool of conquest, essential in the development of the State, war was also a showcase of power, which allowed them to build and enforce a fundamental aspect of it, their status as war chiefs. This manipulation of war took two major forms, the image, that is how the dukes of Burgundy appeared on the battlefield, and the representations created after the event. As a result, it became clear that an imaginary prince existed alongside the real prince. In the present work, we will study these two aspects of the prince as war chief and think about how they coexisted. Firstly, this reflection will be based on written sources, starting with financial records, to identify the real prince, that is how the dukes appeared and acted on the battlefield. Iconographic sources will allow us to characterize the imaginary prince: it will be an opportunity for us to update our knowledge about the representations of the dukes of Burgundy by building a corpus which attempts to be as complete and diversified as possible. Through this work focusing on image and representations, we will be able to study the dukes’ relationship with war, their view on the role of war chief, but also the use of war in their propaganda: to this end, we will look at their involvement in the process of creation and dissemination of the various representations. Eventually, this work will enable us to develop a better understanding of what a war chief was by the end of the Middle Ages, through the example of the dukes of Burgundy

El presente trabajo se centrará en la comparación de la representación alegórica de la relación entre el amor y la muerte en cuatro emblemas del pintor, dibujante y humanista, Otto van Veen, también conocido como Otto Vaenius. Específicamente nos centraremos en dos de sus tres libros de emblemas Amorum Emblemata (1608) y Amoris Divini Emblemata (1615). El género emblemático, inaugurado por el Emblematum Liber (1531) de Alciato, es poco conocido hoy en día, pero tuvo una gran importancia en los siglos XVI y XVII, pues respondía a las inquietudes pasatistas de la época, siguiendo, por una parte, la influencia de los epigramas latinos y por otra, la interpretación que tuvieron en este tiempo los jeroglíficos egipcios.

Full textThis Phd thesis is about horticulture, relations to plants, human relations around plants, and conceptions of life and living things in Japan. It presents an ethnographic fieldwork conducted among two groups of cultivators in the Ōsaka metropolitan area (2013-2017): volunteers cultivating a wisteria (Wisteria floribunda) in the Fukushima district of Ōsaka, and professional producers growing plum, peach and cherry in the Higashino neighborhood in Itami. In order to monitor these emblematic plants to “bloom” (sakaseru), the cultivators practice traditional horticultural characterized by a fastidious shaping of each plant, and they skillfully display the flowers. However, upsetting events such as lack of bloom or the outbreak of a plant disease (specifically a plum virus) put into question a technical control of plants that was taken for granted. Cultivators are led to modify their practices, and also the way they of appreciate the plants. These actions and assessments lead to inferences, allowing to question the status of plants, and how they work. There are two main purposes in this research. On the one hand, an ethnography of Japanese horticulture, focusing on a description of local social life and the technical actions observed during fieldwork. On the other hand, drawing on an "anthropology of life" engaged with the concrete manipulations of living beings that are both organisms and artefacts, the aim is to grasp the conceptions of life and living things, may that be a flower festival or part of a phytosanitary plan

Education and communication are the two key elements on which is grafted the discourse on environmental and agrofood sustainability that is developed in this research. In relation to sustainable and responsible lifestyles, the communication and its messages can have strong influence on personal behavior but there is a need to provide authentic information, relevant and scientifically founded. “A sustainability communication, pedagogically oriented, what educational contribution can offer to people and communities for the adoption of responsible lifestyles according to integral ecology?” The reference to pedagogy of educational communication and emblematic cases under study highlight the need to build alliances oriented to the creation of an educational and generative agreement between research, institutions, businesses and consumers. Lifelong learning is the inescapable interpretive key that the combination of themes in this research suggests. The notion of integral human formation and the perspectives of the Social Doctrine of the Church are taken as an itinerary to explore the possible heuristic connections between pedagogy and civil economy, with reference to the educability of the individual and human ecology. The orientation towards integral ecology urges pedagogical reflection, in dialogue with other sciences, to generate unprecedented civil planning and new educational options to achieve authentic fair and solidarity-based development. In the context of the business pedagogy, organisational models such as social enterprises, community cooperatives and Benefit Societies are considered as protagonists in the responsible commitment to building the community, enhancing the flourishing of the human person and the care of creation. Human education and integral ecology become an alliance capable of developing emblematic green capabilities for building an inclusive and vital future, in a perspective of common good.

Full textThe development of high-throughput DNA sequencing has greatly facilitated the screening of genetic variations within patient genome. Henceforth, one of the main challenges in medical genetics is no longer the detection of variations, but their functional and clinical interpretation. Recently, we showed by using splicing reporter minigene assays, that although splicing mutations, and in particular those affecting its regulation, are more prevalent than initially estimated, they could now be predicted by using dedicated bioinformatics tools. We thus extended the evaluation of the predictive power of these four newly developed computational approaches by a comparative study of the scores obtained by these approaches with experimental data for a total of about 1200 exonic variations. Our findings have demonstrated the reliability of these approaches, used alone or in combination, and allow to offer recommendations for their use as a filtration tool to prioritize the variations to be analysed as a priority in splicing-dedicated functional assays. Nevertheless, an exhaustive mutational analysis targeting MLH1 exon 7, has highlighted the apparent failure of these approaches, yet validated by studies focused on BRCA2 exon 7, MAPT exon 10 and MSH2 exon 5, suggesting that these methods might not be equivalently applicable to all exons and/or genes. Indeed, we have shown that this exon has particular characteristics, i.e. remarkably strong splice sites, conferring it a total resistance to splicing regulation mutations and defeating prediction tools. These findings help to better determine the limitations of these bioinformatics tools while contributing to their improvement. In spite of these advances, the pathogenicity assessment of splicing mutations remains complicated, especially of those leading to in-frame and/or partial splicing anomalies. By using variant-induced partial BRCA2 exon 3 skipping as a model system, we showed that BRCA2 tumor suppressor function tolerates a substantial reduction in expression level, as BRCA2 allele producing as much as 70% of transcript encoding deficient protein may not necessarily confer high-risk of developing cancer. Altogether, these data have important implications in the molecular diagnosis and clinical management of patients and their relatives, with a direct benefit for hereditary cancer-suspected families and should contribute to the interpretation of VSI identified by high throughput sequencing in any other genetic disease

Full textThis dissertation is about the emblematical expression in the work of Jean Baudoin. At first Marguerite de Valois’s lecturer, he was sent to England by Marie de Médicis in order to translate L’Arcadie de la Comtesse de Pembroke, in 1624. Baudoin belonged to the House of the Cardinal de Richelieu and was one of chancelier Séguier’s protégés. As soon as 1634, hejoined the first members of the Académie française and took part in the writing of its statutes. Eighty six pieces of work are attributed to this prolific author and translator, most of which are translations significant of the orientation he gave to his work: the “Discours politiques” on Tacite’s work by Scipio Ammirato (1618), La Sagesse Mystérieuse des Anciens by Francis Bacon (1619), Mythologie by Natale Conti (1627) or Monita et Exempla politica by Justus Lipsus (1606 & 1650). The iconographical tradition is paramount in Jean Baudoin’s work, as well as the emblematical expression. The Recueil d’Emblemes divers, first published in Paris in 1638-1639 is the focus of the present dissertation. This collection of emblems was designed by Jean Baudoin, but he was largely influenced in its writing by previous translations. Two other pieces of work translated by Baudoin are essential to the thorough study of the Recueil d’Emblemes divers: the adaptation of Cesare Ripa’s Iconologia (First part 1636-1637, First and second part 1644) and the translation of Esope Phrygien’s Fables in 1631. The present work will first focus on how Baudoin constantly put himself and his work at the service of the reinforcement and affirmation of the French monarchy and of the moral edification of the reader. Then, we will study the connections between these three major works which enable to understand the importance of emblematical expression in Jean Baudoin. To do so, three main aspects will be studied: the renewal of the emblematical form/ genre, the creation of an allegorical vocabulary whose aim was the codification of the representation of power and monarchy, and a study of engraving, of it functioning in the text and of its production processes

Full textThe practice of Community Cultural Development (CCD) is shaped by ideals, ethical standards and socially democratic values. Principles of doing good, altruism, social change, participation, collaboration, even ‘community’ itself are embedded as the dominant operating paradigms. CCD practice has thus attracted a number of assumptions and operational touchstones in its brief history. Drawing on stories from 40 years of CCD work, I question how well have I lived up to these standards in my own practice. These stories focus on several specific projects including: The Ribbons of Steel project marking the closure of BHP Steelmaking in Newcastle in the Australian state of New South Wales (NSW), my writer-in-residency at Windale (NSW), my time with Pipi Storm Childrens’ Circus, and my work with Australian Aboriginal communities. In undertaking this journey the essential questions I address are: How can stories reveal a deeper understanding of the principles of our work? What can be understood from these stories of our relationship to the communities in which we work? Such questions require a deep reflection upon and analysis of my own career. Identifying and investigating the underlying philosophical principles, interrogating how well or not I have applied them, and what lessons I have learned in the struggle to cleave to a principled approach lies at the core of this thesis. While I approach this examination from a particularly individual perspective, I situate my work in the broader practice of CCD to demonstrate how the stories may be emblematic – seams that we can mine for knowledge. In so doing I arrive at a deeper understanding and clarity around what are the essential and foundation impulses and principles of CCD work.
