Vaccination is one of the most effective strategies for preventing infectious diseases. However, the efficacy of most vaccines is variable, due to genetic variations in the host and probably also to variations in the gut microbiota. Studying the combined effects of these two factors on vaccine response is a novel and integrated strategy. This project was based on the study of 4 lines of laying hens, including both experimental and commercial lines. We studied the impact of a strong disturbance of the intestinal microbiota (by a model of treatment with antibiotics) and of a change in the rearing environment (with access to an outdoor range) on the vaccine response and, more generally, on immunocompetence. To assess animal welfare, we also conducted behavioral studies. The aims of this thesis were: (1) to assess the impact of genetics on gut microbiota, vaccine response, immunocompetence and behavior; (2) to study the impact of microbiota disturbance on these parameters; (3) to assess the impact of outdoor rearing on these parameters. Finally, this work has contributed to the identification of the best levers (genetics, microbiota, rearing environment) that can be combined to raise more robust animals, with better health and optimized welfare

Este projeto curricular consiste num estudo acerca do efeito da geometria da aresta de corte na superfície maquinada e do seu respetivo desgaste, através da realização de ensaios de fresagem a alta velocidade numa operação de acabamento a seco no aço AISI D2, em dois estados endurecidos, 55 e 61 HRc. Foram utilizadas três ferramentas de corte de metal duro revestidas por PVD, duas delas por AlTiN e outra por TiAlN + TiN. A diferença entre as ferramentas de corte estava na sua geometria. Raio de ponta, zona da aresta de corte a 90 (face alisadora) e preparação da aresta negativa (chanfro) foram as características a serem estudadas. Foram realizados ensaios de corte longos e curtos. Os primeiros visaram avaliar o desgaste da ferramenta (VB) de modo a obter a curva de desgaste, numa operação onde as ferramentas maquinam em superfícies planas com diferentes durezas. Os ensaios curtos permitiram avaliar a rugosidade da superf ície maquinada onde a ferramenta maquinou em dois tipos de superfície, nomeadamente plana vertical e 3D. Efetuou-se um estudo e levantamento geométrico da ferramenta de corte já existente, que por sua vez, foi modelada com recurso ao software CAD, So- lidWorks. Posteriormente, no âmbito do projeto curricular, desenvolveu-se uma nova ferramenta de corte que pudesse conjugar todas as características das ferramentas de corte utilizadas, nomeadamente o raio de ponta, a face alisadora e chanfro. Concluiu-se que, para maquinagens em superfície plana vertical, a ferramenta com a geometria com menor raio de ponta e face alisadora apresentou maior tempo de maquinagem e menor valor de rugosidade, mesmo com o aumento da dureza do material. No entanto, para maquinagens em superfícies 3D, a referida ferramenta não é aconselhável. Ferramentas de corte com raios de ponta elevados, com faces alisadoras e chanfros representaram ferramentas capazes de maquinar superfícies 3D e, por sua vez, obter valores de rugosidade média aritmética abaixo do valor proposto. Contudo, para maquinagens em superfícies planas, estas apresentaram os menores tempos de maquinagem até a obtenção do V Bmax e maiores valores de rugosidade na superfície maquinada.
This curricular project consists on a study about the effect of the cutting edge's geometry in the machined surface and its respective wear, through the completion of milling tests with a high speed machine in a dry nishing operation on AISI D2 steel, in two hardened states, 55 and 61 HRc. Three hard metal cutting tools, coated with PVD, two of them with AlTiN and another with TiAlN +TiN. The difference between the cutting tools were in their geometry. Radius edge, cutting egde zone at 90 (face straightener) and the negative preparation edge (chamfer) were the characteristics to be studied. Long and short cut tests were conducted. The first aimed to evaluate the tool wear (VB) in order to obtain the wear curve, in an operation where the tools are used on at surfaces with different hardness values. The short test allowed to evaluate the roughness of the machined surface on two types of surface, vertical at and 3D. It was performed a geometric survey of a cutting tool available in the market which, in turn, was modeled using a CAD software,SolidWorks. Later, in the context of the curricular project, a new cutting tool was developed, in a way that could bring together all the characteristics of the cutting tools used, mainly the radius, the face straightener and chamfer. It was concluded that, while machining vertical at surfaces, a tool geometry with smaller radius of edge and face straightener, there was an increased machining time and the lowest value of surface roughness, even with the increase of the material hardness. This tool allowed, for the same conditions, to obtain a better surface finish with good dimensional accuracy. However, for 3D machining, this tool is not advisable. Tools with high edge radius, face straightener and chamfer are tools capable of 3D surface machining and get values of arithmetic surface roughness below the expected. However, format surface machining, this tool obtained smaller machining time until the achievement of the V Bmax and higher values of surface roughness in the machined surface.

High nitrogen austenitic stainless steels containing 0.9 wt-% N and 20 wt- % N were tested in a ultrasonically induced vibratory cavitation testing device. Incubation times for damage initiation and mass losses were periodically measured during the cavitation-erosion tests. Scanning Electron Microscopy observation of the damaged surfaces allowed identifying the wear mechanisms operating during each step of the cavitation-erosion test. 0.9 wt-% N specimens were obtained through High Temperature Gas Nitriding UNS S31803 duplex stainless steel, at temperatures between 1050 and 1200 oC. Three groups of specimens were obtained: solubilized with all nitrogen in solid solution, solubilized and work hardened specimens and nitride containing specimens. The 20 wt- % N specimens were obtained through Low Temperature Plasma Nitriding the already High Temperature Gas Nitrided specimens and getting an expanded austenite layer at the surface. The specimens were firstly characterized by Electron Backscattered Diffraction - EBSD techniques and then submitted to the cavitation-erosion tests in distilled water. When compared to conventional UNS S30403 lean nitrogen solubilized austenitic stainless steel specimens, greater incubation times and smaller maximum wear rates were observed. The maximum wear rates (compared to those of the solubilized UNS S30403 steel) were: for the nitride containing specimen 6.9 times smaller; for textured and non-textured all nitrogen in solid solution specimens 26.8 and 25 times smaller, respectively; for the solubilized and work hardened specimen 145 times smaller; for the expanded austenite layer, with circa 20 wt- % N, specimens 300 times smaller. The beneficial effect of nitrogen on the cavitation-erosion resistance of the studied specimens was attributed to: (i) an increase in resistance to plastic deformation; (ii) a more homogeneous distribution of the plastic deformation; and (iii) an increase of the relative participation of energy consuming mass loss mechanisms. Plastic deformation accompanied by formation of micro relief at the surface and slip bands protrusions were clearly identified, during the first stages of cavitation erosion. The first evidences of mass loss (detected by SEM observations) were seen as particles detaching from micro cracks and micro pits formed at the grain surface. Nucleation and growth of cavitation damage was heterogeneously distributed at the grain scale. Slip bands protrusions, grain boundary protrusions and nitride matrix interfaces sites were more prone to nucleating the damage. Increasing nitrogen contents in solid solution increased the relative contribution of grain boundary nucleated damage, compared to the total amount of nucleation sites. Grains with 100 and 111 crystallographic planes approximately parallel to the surface were more prone to nucleation and growth of cavitation damage. Grains with 101 planes // surface were much more resistant to cavitation-erosion damage. These results are discussed considering differences of critical resolved shear stresses for grains with different orientations. Cavitation erosion damage occurs preferentially at grain boundaries across which steep stress gradients arise. Particularly, CSL -3 twin boundaries are much more susceptible to cavitation erosion damage incubation than other types of CSL boundaries and non CSL boundaries.

Muitos estudos têm mostrado que os ácidos graxos poliinsaturados, especialmente ômega-3 ou n-3, são importantes para a boa saúde e desenvolvimento normal. Tais compostos e seus metabólitos ativos são importantes para a manutenção estrutural e fisiológica do Sistema Nervoso Central durante todo o desenvolvimento, principalmente durante as fases iniciais da vida. Sendo assim, avaliamos os efeitos da suplementação com n-3 em mães e ninhadas submetidas ao procedimento de separação materna longa (SML) do dia pós-natal (dpn) 2 ao 14, por um período de 360 min/dia, que constitui um estresse para ambas as partes. Ratas Wistar prenhes com 10 semanas de idade foram distribuídas em 6 grupos: 1) Controle (CTL) + dieta regular; 2) CTL + óleo de soja 3) CTL + óleo de peixe (n-3); 4) Separação materna (SM) + dieta regular; 5) SM + óleo de soja; 6) SM + n-3. A suplementação foi realizada do nascimento até o desmame dos filhotes, que aconteceu no dpn 21. Três dias após o desmame, as mães foram submetidos ao teste de labirinto em cruz elevado (LCE), assim como os filhotes no dpn 31; e ainda três dias após o LCE estas foram avaliadas no teste do nado forçado (TNF). A partir dpn 83, os filhotes, machos e fêmeas, foram desafiados no LCE e TNF. As conseqüências comportamentais da separação foram expressivas em mães, porém com pouca influência na emocionalidade da prole tanto juvenil quanto e adulta. Surpreendentemente o n-3 produziu efeito ansiogênico nas mães, mas não produziu qualquer efeito sobre o comportamento dos filhotes, exceto por uma redução nas concentrações basais de corticosterona da prole feminina adulta e um aumento de reatividade ao estresse em machos e fêmeas adolescentes.
Several studies have shown that polyunsaturated fatty acids, specially omega-3, or n-3, are essential for good health and for normal development. Such compounds, along with their active metabolites, are also important to the structural and physiological maintenance of the central nervous system throughout development, mainly during the initial phases of life. Therefore, we studied the effects of n-3 supplementation on dams and pups submitted the long maternal separation (LMS, 360 min long), from 2 to 14 post natal day (pnd). This is a stressfull stimulus to both parts. Pregnant female Wistar rats, 10-weeks old, were separated into 6 groups: 1) Control (CTL) + regular diet; 2) CTL + soy oil; 3) CTL + fish oil (n-3); 4) LMS + regular diet; 5) LMS + soy oil; 6) LMS + n-3. The supplementation was kept from birthday to weaning, which occurred on pnd 21. Three days after weaning, dams were exposed to the Elevated Plus Maze (EPM), as did the pups, on pnd 31, and three days after the Elevated Plus Maze, dams were evaluated on the Forced Swimming Test (FTS). From PND 83, male and female pups were challenged on EPM and FST. The behavioral consequences of the separation were expressive on dams, but had little influence on the emotionality of young males and females. Surprisingly, n-3 caused an ansiogenic effect on mothers, and failed to cause any effect on pups’ behavior. However, there was a reduction of the basal concentration of corticosterone in young females and an increased reactivity to stress in both male and female adolescents.

Introdu??o: A falha na fun??o da bomba muscular da panturrilha ? considerada a principal causa da insufici?ncia venosa cr?nica (IVC) e a hipertens?o venosa prolongada, leva ao enfraquecimento da musculatura da panturrilha. A avalia??o espec?fica dessa musculatura pode ser feita de diferentes formas, entre elas a utiliza??o do teste de eleva??o do calcanhar (TEC). Atualmente, v?rios protocolos foram desenvolvidos para a realiza??o do TEC em diferentes popula??es, sem uma padroniza??o bem detalhada. Objetivos: Primariamente avaliar a confiabilidade e reprodutibilidade tanto intra quanto inter-avaliadores do TEC em adultos saud?veis, de dois diferentes protocolos com diferentes est?mulos: auto-cadenciado (TECAC) e externamente cadenciado (TECEC); Secundariamente, avaliar o desempenho e a atividade el?trica dos m?sculos dos membros inferiores durante a realiza??o do TECEC em sujeitos com IVC comparando com indiv?duos saud?veis pareados. M?todos: Dois estudos foram realizados e ambos foram do tipo observacional, de car?ter transversal. Para o Estudo 1, dois protocolos do TECAC e TECEC foram realizados em sujeitos jovens e saud?veis. Para o Estudo 2, foram recrutados sujeitos com idade entre 35 e 65 anos, com diagn?stico cl?nico de IVC para realiza??o do TECEC associado ? Eletromiografia de Superf?cie (EMGs). Resultados: No estudo 1, 33 indiv?duos saud?veis foram recrutados (16H), com idade de 23,03 anos (?2,71). Observamos que o TECAC obteve um melhor resultado tanto no que diz respeito ao desempenho (53,01 eleva??es) quanto ? reprodutibilidade relativa inter-avaliador (CCI=0,77) e, o TECEC obteve melhor reprodutibilidade intra-avaliador (CCI=0,69). Foi observado que ao final da realiza??o de ambos os testes houve aumento dos sintomas de fadiga (p<0,01); entretanto, com manuten??o do desempenho de eleva??es de calcanhar nos tr?s TECAC (p=0,76) e diminui??o do desempenho no TECEC quando comparados T1, T2 e T3 (p<0,01). No Estudo 2, foram recrutados 44 sujeitos com IVC, dos quais 17 atingiram todos os crit?rios de inclus?o e compuseram a amostra do Grupo IVC, versus 17 sujeitos saud?veis pareados por idade, sexo e IMC que compuseram o Grupo Controle. Houve diferen?a significante na compara??o intergrupo do desempenho do TECEC (p<0,01) e tamb?m na compara??o intergrupo do tempo gasto para realiza??o do TECEC (p<0,05). A plantiflex?o no membro inferior direito foi significativamente menor (p<0,05) no Grupo IVC. Para avalia??o da atividade el?trica, o TECEC foi dividido em quatro momentos (25%, 50%, 75% e 100%). Na avalia??o da atividade el?trica muscular durante o TECEC, foram encontradas diferen?as significantes nos momentos 50% e 75% (p<0,05) do m?sculo tibial anterior direito e nos momentos 25% e 50% (p<0,05) do m?sculo tibial anterior esquerdo. Conclus?o: Nossos resultados do Estudo 1 demonstram que ambos os TEC (auto-cadenciado e externamente cadenciado) podem ser utilizados para fornecer resultados de desempenho muscular. O TECEC potencialmente determina uma maior e mais intensa atividade muscular e o TECAC demonstrou melhor reprodutibilidade e maior concord?ncia, quando avaliado o desempenho nos tr?s momentos. Em rela??o ao Estudo 2, ? poss?vel observar que os indiv?duos com IVC apresentam um desempenho inferior no TECEC comparados a sujeitos saud?veis, com menor atividade el?trica muscular em tibial anterior bilateralmente j? em est?gios iniciais da doen?a.
Background: Failure of calf muscle pump function is considered a major cause of venous insufficiency (CVI) and prolonged venous hypertension leads to weakening of the calf muscles. The evaluation can be done from different forms, from a heel lift test (TEC). Currently, several protocols have been developed for an ECT test in different populations, without a very detailed standardization. Objectives: Firstly to evaluate the reliability and reproducibility of intra-r inter-raters in healthy adults, from two different protocols with different stimuli: self-paced (TECAC) and externally cadenced (TECEC); secondarily, to evaluate the performance and electrical activity of lower limb muscles during an TECEC in subjects with CVI compared with healthy paired subjects. Methods: Two studies were performed and both were of the observational type, with a transversal character. For Study 1, two TECAC and TECEC protocols were performed on healthy young subjects. For Study 2, subjects with ages between 35 and 65 years old were recruited, with a clinical diagnosis of CVI to perform TECEC associated with Surface Electromyography (EMGs). Results: In study 1, 33 healthy subjects were recruited (16H), aged 23.03 years (? 2.71). We observed that the TECEC obtained a better result both in terms of performance (53.01 elevations) and relative inter-rater reproducibility (ICC = 0.77) and the TECEC obtained better intra-rater reproducibility (ICC = 0, 69). It was observed that at the end of both tests there was an increase in fatigue symptoms (p <0.01); (P = 0.76) and decreased performance in the TECEC when compared to T1, T2 and T3 (p <0.01). In Study 2, 44 subjects with CVI were recruited, of whom 17 met all inclusion criteria and composed the sample of Group IVC versus 17 healthy subjects matched for age, sex and BMI who composed the Control Group. There was a significant difference in the intergroup comparison of the TECEC performance (p <0.01) and also in the intergroup comparison of the time spent to perform the TECEC (p <0.05). The flexure in the right lower limb was significantly lower (p <0.05) in Group IVC. For evaluation of electrical activity, the TECEC was divided into four moments (25%, 50%, 75% and 100%). In the evaluation of the muscular electrical activity during the TECEC, significant differences were found in the moments 50% and 75% (p <0.05) of the right anterior tibial muscle and moments 25% and 50% (p <0.05) of the muscle Anterior tibialis. Conclusion: Our results from Study 1 demonstrate that both (self-paced and externally paced) ECT can be used to provide muscle performance results. The TECEC potentially determines a greater and more intense muscular activity and the TECAC demonstrated better reproducibility and greater agreement, when evaluated the performance in the three moments. Regarding Study 2, it is possible to observe that individuals with CVI present inferior performance in the TECEC compared to healthy subjects, with lower muscular electrical activity in the anterior tibial bilaterally already in the initial stages of the disease.

Introduction: Previous studies have found that cerebral cooling is effective to decrease the elevation in core temperature during exercises performed in hot environment. Furthermore, other have shown that the rate of increase in core temperature may vary according to the exercise mode under consideration; in contrast to self-paced exercise, controled-pace exercise requires greater muscle recruitment, thereby inducing higher increase in core temperature. The aim of this study was to examine the acute effects of cranial-cervical cooling in the rate of increase of core temperature, and physiological and psychological responses during controlled-pace and self-paced exercises performed in hot environment. Methods: Ten cyclists took part in the study, they underwent 3 preliminary sessions under ~21 ??C and 4 experimental sessions in hyperthermia (~33 ??C), that is: self-paced exercise with or without central cooling (ESP33 e ESP33C); and controlled-pace exercise with or without central cooling (ECP33 e ECP33C). In both the exercise modes they completed 20km cycling trial either in a self-paced manner anchored in ratings of perceived exertion (RPE = 16), or in a controlled-pace manner with intensity set at the mean power output recorded during preliminary self-paced exercise session under ~21 ??C. Core and peripheral temperature, cardiopulmonary and perceptive measures were obtained during exercises, while maximal voluntary muscle torque, body mass and urine density were obtained prior and post exercises. Mean responses, as well as the rate of increase in core temperature and RPE along the exercises with or without cooling were compared. Results: The control trials showed that ESP e ECP were submaximal, and affected by the elevation in environment temperature (21 ??C vs 33??C). Regarding the responses to the cranial-cervical cooling, there were cooling main effects due to reduction in exercise time, increases in power output, reduction in mean core temperature and its rate of increase. Additionally, there was lower RPE and thermal sensation. Only the oxygen uptake showed exercise mode main effects, with greater values in ECP. No exercise mode by cooling interaction effects was observed. Conclusion: The central cooling was effective to increase the motor performance, likely due to the attenuated increase in core temperature and RPE. However, the exercise mode did not determine to these responses.
Introdu????o: Estudos anteriores observaram que o resfriamento central ?? efetivo para atenuar a taxa de eleva????o da temperatura central ao longo do exerc??cio realizado em hipertermia. Demonstram tamb??m, que a taxa de eleva????o da temperatura central pode depender do modelo de exerc??cio praticado; exerc??cios com ritmo controlado, ao contr??rio de exerc??cios com ritmo auto-ajustado, requerem maior recrutamento muscular e, consequentemente, apresentam maior eleva????o da temperatura central. O objetivo deste estudo foi examinar os efeitos agudos do resfriamento cr??nio-cervical sobre a taxa de eleva????o da temperatura central, respostas fisiol??gicas e perceptivas associadas, durante exerc??cios de ritmo auto-ajustado e ritmo controlado, realizados em ambiente quente. M??todos: Participaram do estudo dez ciclistas experientes e aclimatados ao calor, os quais foram submetidos a 4 sess??es preliminares em 21 ??C e 4 sess??es experimentais em hipertermia (33 ??C), a saber: exerc??cio com ritmo auto-ajustado sem e com resfriamento cr??nio-cervical (EAA33 e EAA33R); e exerc??cio com ritmo controlado sem e com resfriamento cr??nio-cervical (ECON33 e ECON33R). Em ambos os modelos, os participantes completaram 20 km; ou com ritmo auto-ajustado, controlado pela percep????o subjetiva de esfor??o (PSE = 16), ou ritmo controlado, em intensidade fixa, correspondente ?? intensidade m??dia gerada na sess??o preliminar de exerc??cio com ritmo auto-ajustado em 21 ??C. Medidas de temperatura central e perif??rica, cardiopulmonares e psicol??gicas foram obtidas durante a realiza????o dos exerc??cios, enquanto medidas do torque muscular volunt??rio m??ximo, massa corp??rea e densidade da urina, foram realizadas antes e ap??s os exerc??cios. Respostas m??dias e a taxa de modifica????o da temperatura retal (TR) e percep????o subjetiva de esfor??o (PSE) ao longo dos diferentes modelos de exerc??cio, sem e com resfriamento, foram comparadas. Resultados: Os testes controle evidenciaram que o EAA e ECON foram de intensidade subm??xima, e foram influenciados pelo aumento da temperatura ambiente (21 ??C vs 33??C). Quanto as respostas frente ao resfriamento cr??nio-cervical, foram verificados efeitos principais, em fun????o da redu????o do tempo total, eleva????o da pot??ncia mec??nica produzida, atenua????o da TR e de sua taxa de eleva????o; al??m da menor PSE (geral e local) e sensa????o t??rmica. Com rela????o aos efeitos principais do modelo de exerc??cio, somente o consumo de oxig??ncio foi modificado com maiores valores em ECON. Efeitos do resfriamento foram detectados sobre a TR e sua taxa de eleva????o, pois foram reduzidas em ambos os modelos de exerc??cio. Nenhum efeito de intera????o foi observado nas vari??veis medidas. Conclus??o: O resfriamento central foi efetivo na eleva????o do desempenho motor, possivelmente mediado por uma atenua????o da eleva????o da temperatura central e altera????o na PSE. Entretanto, o modelo de exerc??cio n??o foi determinante para a observa????o destas respostas.
