Abstract:
A number of different types of glycoconjugate are found associated with joint tissue and fluids, comprising glycoproteins, glycolipids and glycosaminoglycans. Oligosaccharide chains of glycoconjugates are degraded by exoglycosidases, and the dominant exoglycosidase found in human blood, synovial fluid, the synovial membrane and chondrocytes of articular cartilage is HEX (N-acetyl-β-hexosaminidase). HEX is localized mostly intracellularly in synovial cells. Serum activity of HEX may be used to monitor the course and efficiency of treatment of Lyme arthritis, and activity of HEX, above 10 μkat/kg of protein in the synovial fluid, suggests rheumatoid disease. There is a shortage of HEX inhibitors able to penetrate synoviocytes, so the development of drugs which inhibit synthesis and/or the activity of HEX will be a promising field for future investigations.

The article presents the issues of arthroplasty of several large joints in rheumatoid arthritis (RA), including the difficulties of perioperative management of young patients. The effectiveness of arthroplasty of several (from 2 to 4) large joints is shown in the management of patients with RA at the orthopedist and rheumatologist. Тhe absence of serious postoperative complications was demonstrated. The article presents а 20-year clinical case of a young patient with active RA and arthroplasty of 4 large joints. This example demonstrates the success and safety of multiple arthroplasty with a complete return of joint function and a decrease in RA activity during long-term dynamic follow-up at the orthopedist and rheumatologist.

Abstract:
By manipulating trunk angle in ergometer cycling, we studied the effect of body configuration on muscle recruitment and joint kinematics. Changing trunk angle affects the length of muscles that span the hip joint. It is hypothesized that this affects the recruitment of the muscles directly involved, and as a consequence of affected joint torque distributions, also influences the recruitment of more distal muscles and the kinematics of distal joints. It was found that changing the trunk from an upright position to approximately 20 deg forward or backward affected muscle activation patterns and kinematics in the entire lower limb. The knee joint was the only joint not affected by manipulation of the lengths of hip joint muscles. Changes in trunk angle affected ankle and hip joint kinematics and the orientation of the thigh. A similar pattern has been demonstrated for muscle activity: Both the muscles that span the hip joint and those acting on the ankle joint were affected with respect to timing and amplitude of EMG. Moreover, it was found that the association between muscle activity and muscle length was adapted to manipulation of trunk angle. In all three conditions, most of the muscles that were considered displayed some eccentric activity. The ratio of eccentric to concentric activity changed with trunk angle. The present study showed that trunk angle influences muscle recruitment and (inter)muscular dynamics in the entire limb. As this will have consequences for the efficiency of cycling, body configuration should be a factor in bicycle design.

Abstract:
Effectiveness of a joint-stock company is realized through the skills and ability to take into account the wishes and needs of stakeholders and the organization in full. This work is based on the alignment of performance criteria with the organization’s strategy and capabilities. Management plays a key role in this process. It is they who must understand what the shareholders, customers, employees, lenders, and regulators want from the company, and that each category requires organizations to meet their own needs. At the same time, the performance measurement and management system should be flexible enough to be transparent and able to keep track of processes occurring within the organization and stakeholder engagement. The activity of a joint-stock company includes general elements and special elements that arise from the specifics of the company’s operation. The peculiarities of the activity of the company are due also to industry trends, which are partially derived from specific elements of management. In this regard, when identifying problems of functioning of joint-stock companies of the dairy industry, it is necessary to take into account the specific nature of their activity.

Abstract:
The joint reconstruction of nonsparse multi-sensors data with high quality is a challenging issue in human activity telemonitoring. In this study, we proposed a novel joint reconstruction algorithm combining distributed compressed sensing with multiple block sparse Bayesian learning. Its basic idea is that based on the joint sparsity model, the distributed compressed sensing technique is first applied to simultaneously compress the multi-sensors data for gaining the high-correlation information regarding activity as well as the energy efficiency of sensors, and then, the multiple block sparse Bayesian learning technique is employed to jointly recover nonsparse multi-sensors data with high fidelity by exploiting the joint block sparsity. The multi-sensors acceleration data from an open wearable action recognition database are selected to assess the practicality of our proposed technique. The sparse representation classification model is used to classify activity patterns using the jointly reconstructed data in order to further examine the effectiveness of our proposed method. The results showed that when compression rates are selected properly, our proposed technique can gain the best joint reconstruction performance as well as energy efficiency of sensors, which greatly contributes to the best sparse representation classification–based activity classification performance. This has a great potential for energy-efficient telemonitoring of human activity.

Abstract:
This study measures the efficiency of higher education institutions with respect to teaching activity, research activity, and overall activities and also provides a direction for low-performing institutions to improve their performance. In many instances, the efficiency of the institution is a sum up of the efficiency of its activities. However, when the activities consume some resources in common, it requires the allocation of shared resources among the activities. Since sums up the efficiency of each activity does not give the institution's overall efficiency, we use a joint data envelopment analysis that takes into account the institution's internal operations to measure the teaching efficiency, research efficiency, and overall efficiency of the institutions. The empirical results indicate that the institutions that execute both activities simultaneously become more productive rather than adopt a single activity. Hence, this study suggests a useful and measurable action to improve the institution’s performance.

Abstract:
Substrates for studying V(D)J recombination in human cells and two human pre-B-cell lines that have active V(D)J recombination activity are described. Using these substrates, we have been able to analyze the relative efficiency of signal joint and coding joint formation. Coding joint formation was five- to sixfold less efficient than signal joint formation in both cell lines. This imbalance between the two halves of the reaction was demonstrated on deletional substrates, where each joint is assayed individually. In both cell lines, the inversional reaction (which requires formation of both a signal and a coding joint) was more than 20-fold less efficient than signal joint formation alone. The signal and coding sequences are identical in all of these substrates. Hence, the basis for these differential reaction ratios appears to be that coding joint and signal joint formation are both inefficient and their combined effects are such that inversions (two-joint reactions) reflect the product of these inefficiencies. Physiologically, these results have two implications. First, they show how signal and coding joint formation efficiencies can affect the ratio of deletional to inversional products at endogenous loci. Second, the fact that not all signal and coding joints go to completion implies that the recombinase is generating numerous broken ends. Such unresolved ends may participate in pathologic chromosomal rearrangements even when the other half of the same reaction may have proceeded to resolution.

Abstract:
Introduction. In joint-stock companies there is a problem of shortage of money not only for carrying out of investment activity, but also for maintenance of sufficient level of the operational activity. One of the main instruments of a market economy that contributes to the formation of a cash flow system is the stock market. The purpose ofthe article is to study some aspects ofcash flow management in the system of financial management, to characterize cash flows of joint stock companies, which determine the potential of forming their internal source of financing for their development and provide the formation of additional investment resources for the implementation of financial investments in the securities market. Results. One of the important tasks of the Ukrainian economy development is the development of mechanisms for the formation of investment-attractive and innovative-oriented joint-stock companies. The development of market relations requires an increase in the effectiveness of their activities. The assessment of the effective activity of the entity was determined by analyzing the profit of the enterprise, and subsequently, economists supplemented its coefficients of liquidity, solvency, financial stability. The analysis and identification ofthe reserves for increasing profits is also carried out by analysts of the stock market in orderto furtherstimulate the investor. Conclusion. We believe that the analysis of cash flows by their dynamics and structure allows us to identify the negative factors of the organization of financial and economic activity of the entity, immediately reflects the size, quality and direction of change in the financial state of the joint-stock company, and also contributes to the development of concrete measures for its improvement. Since inbound and outbound cash flows of investments are the main factors in creating the value ofenterprises, which ultimately reflects the management of the result of investment activity in terms of Modern Value Approach, therefore, in our view, the process of reproduction of capital and the formation of investment flows can be expressed through the movement of value in the field Investment activity. In the process of studying investment flows of enterprises, it was found that the level of investment attractiveness of enterprises depends on the efficiency of the movement oftheirinvestment cash flows.

Abstract:
Objective.Patient self-report outcomes and physician-performed joint counts are important measures of disease activity and treatment response. This metaanalysis examines the degree of concordance in joint counts between trained assessors and patients with rheumatoid arthritis (RA).Methods.Studies eligible for inclusion met the following criteria: English language; compared patient with trained assessor joint counts; peer-reviewed; and RA diagnosis determined by board-certified or board-eligible specialist or met 1987 American College of Rheumatology criteria. We searched PubMed and Embase to identify articles between 1966 and January 1, 2008. We compared measures of correlation between patients and assessors for either tender/painful or swollen joint counts. We used metaanalysis methods to calculate summary correlation estimates.Results.We retrieved 462 articles and 18 were included. Self-report joint counts were obtained by a text and/or mannequin (picture) format. The summary estimates for the Pearson correlation coefficients for tender joint counts were 0.61 (0.47 lower, 0.75 upper) and for swollen joint counts 0.44 (0.15, 0.73). Summary results for the Spearman correlation coefficients were 0.60 (0.30, 0.90) for tender joint counts and 0.54 (0.35, 0.73) for swollen joint counts.Conclusion.A self-report tender joint count has moderate to marked correlation with those performed by a trained assessor. In contrast, swollen joint counts demonstrate lower levels of correlation. Future research should explore whether integrating self-report tender joint counts into routine care can improve efficiency and quality of care, while directly involving patients in assessment of RA disease activity.

Abstract:
Aim. The article deals with developing and assessing the efficiency of a complex nutritional support program for the joint-ligament system in athletes and physically active people. Materials and methods. The clinical evidence of biologically active substance (BAS) efficiency and functional orientation was obtained from the representative group of people with knee-joint osteoarthrosis. The main group of participants received a BAS complex with synergistic properties in terms of metabolism correction during osteoarthrosis: BAS 1 – 2 pills, BAS 2 – 1 capsule (2 times per day, 12 weeks), and BAS 3 – 1 capsule (2 times per day, 8 weeks). Nonsteroidal anti-inflammatory drugs were also prescribed to participants from the main and control groups. We used the general and special methods for assessing the quality and functional properties of specialized products. Knee joints were studied by using the Lequesne index (scores) characterizing pain syndrome, maximal distance, and daily movement activity. The intensity of pain syndrome was assessed with the visual analog scale (VAS, in mm). Results. We provided a scientific justification for the qualitative and quantitative content of BAS recipes for complex nutritional support of the joint-ligament system: BAS 1 – chondroprotective orientation; BAS 2 – an optimal source of minerals and vitamins; BAS 3 – polyunsaturated fatty acids. The results obtained revealed that specialized products improve the movement activity of participants with osteoarthrosis and significantly decrease pain syndrome (36 %). Apart from a chondroprotective effect such nutrition provides an anti-inflammatory effect and allows decreasing the intake of Nonsteroidal anti-inflammatory drugs in the absence of side effects. We revealed an insignificant number of disease recurrence – 6.7% (in the control group – 36.5 %) and established the mechanisms of such an influence. Conclusion. The application of biologically active complexes could serve as a reliable means of preventing and treating the diseases of the joint-ligament system, providing physical performance, and preserving health in athletes and physically active people.

Abstract:
The article clarifies that results of collaborative efforts in construction, which are received without establishing a legal entity, are presented by constructed buildings that belong to owners as a jointly-owned property. It is revealed that a completed real estate object is put into operation by the operator of collaborative activities. It is pointed out that results of collaborative efforts are distributed according to percentage rates of co-owners and in accordance with the purpose of the joint project. The procedure of completing documents on jointly constructed building which is based on the aims of the collaborative activities is analyzed. It is determined that the aim of joint construction is to obtain a part of ownership in a constructed building by every co-owner (the aim is achieved at the time of signing the act of distributing housing space) or to get profits from jointly constructed buildings. The goal of collaborative activities includes constructing a building, its sale and distributing profits from collaborative activities (the aim is achieved at the time of distributing profits among participants of collaborative activities). It is found that a collaborative activity is discontinued upon achieving the aim. It is proposed to establish a standard act of distributing housing space of a jointly constructed building in accordance with an agreement on collaborative activities without setting up a legal entity.

Traumatic injuries of the knee joint of middle and elderly age account for the majority of injuries of the lower limb. There is a large number of developments in the rehabilitation of the knee joint after injuries, but unified approaches to the rehabilitation algorithm have not been formed. The purpose of this work was to evaluate the effectiveness of a set of physical exercises for the treatment and prevention of traumatic injuries of the knee joint in middle-aged and elderly patients (ICD-10: S80-S89). The claimed complex has been tested in 148 patients in whom the duration of remission was 12-36 months. The early use of physical exercises for injuries of the knee joint in middle-aged and elderly people is associated, first of all, with the anticipation of probable complications, stimulation of compensatory-adaptive reactions of the activity of organs and systems of the body.

Abstract:
Abstract Background People with cerebral palsy (CP) experience increased muscle stiffness, muscle weakness, and reduced joint range of motion. This can lead to an abnormal pattern of gait, which can increase the energy cost of walking and contribute to reduced participation in physical activity. Objective The aim of the study was to examine associations between lower-body joint angles, moments, power, and walking efficiency in adolescents with CP. Design This was a cross-sectional study. Methods Sixty-four adolescents aged 10 to 19 years with CP were recruited. Walking efficiency was measured as the net nondimensional oxygen cost (NNcost) during 6 minutes of overground walking at self-selected speed. Lower-body kinematics and kinetics during walking were collected with 3-dimensional motion analysis, synchronized with a treadmill with integrated force plates. The associations between the kinematics, kinetics, and NNcost were examined with multivariable linear regression. Results After adjusting for age, sex, and Gross Motor Function Classification System level, maximum knee extension angle (β = −0.006), hip angle at midstance (β = −0.007), and maximum hip extension (β = −0.008) were associated with NNcost. Age was a significant modifier of the association between the NNcost and a number of kinematic variables. Limitations This study examined kinetic and kinematic variables in the sagittal plane only. A high interindividual variation in gait pattern could have influenced the results. Conclusions Reduced knee and hip joint extension are associated with gait inefficiency in adolescents with CP. Age is a significant factor influencing associations between ankle, knee, and hip joint kinematics and gait efficiency. Therapeutic interventions should investigate ways to increase knee and hip joint extension in adolescents with CP.

However, many physiological mechanisms of the restorative effect of traditional systems still need to be clarified. We recruited 38 girls aged 10–16 years (mean age 12.5 years, interquartile range [10.8; 13.2] years) involved in different types of physical activity (rhythmic gymnastics and choreography) needing to recover their knee joint function after overuse injury. The subjects were divided into two groups: the main group (physical rehabilitation using a specially developed set of tai chi movements) and the control group (standard course of physical rehabilitation taking place in a physical therapy room). After 2 weeks of rehabilitation, the participants underwent isokinetic testing of the knee joint using the Biodex System 4 Pro (USA). The test results obtained by cluster analysis were typified into three characteristic patterns (χ2 test, p = 0.004): 1) insufficiency of strength parameters (peak torque (PT), peak torque to body mass (PT/BM)) at high speed and insufficiency of functional parameters (PT angle, deceleration time) at high speed; 2) insufficiency of functional parameters (time to PT, deceleration time); 3) insufficiency of strength parameters (PT, PT/BM). It was shown that while the classical means of physical rehabilitation are good for restoring the dynamic muscle parameters (PT), tai chi exercises optimally restore the functional parameters of muscle activation (TP angle and deceleration time). Abstract:
V(D)J recombination in lymphoid cells is a site-specific process in which the activity of the recombinase enzyme is targeted to signal sequences flanking the coding elements of antigen receptor genes. The order of the steps in this reaction and their mechanistic interdependence are important to the understanding of how the reaction fails and thereby contributes to genomic instability in lymphoid cells. The products of the normal reaction are recombinant joints linking the coding sequences of the receptor genes and, reciprocally, the signal ends. Extrachromosomal substrate molecules were modified to inhibit the physical synapsis of the recombination signals. In this way, it has been possible to assess how inhibiting the formation of one joint affects the resolution efficiency of the other. Our results indicate that signal joint and coding joint formation are resolved independently in that they can be uncoupled from each other. We also find that signal synapsis is critical for the generation of recombinant products, which greatly restricts the degree of potential single-site cutting that might otherwise occur in the genome. Finally, inversion substrates manifest synaptic inhibition at much longer distances than do deletion substrates, suggesting that a parallel rather than an antiparallel alignment of the signals is required during synapsis. These observations are important for understanding the interaction of V(D)J signals with the recombinase. Moreover, the role of signal synapsis in regulating recombinase activity has significant implications for genome stability regarding the frequency of recombinase-mediated chromosomal translocations.

Abstract:
Intra-articular administration of drugs to the joint in the treatment of joint disease has the potential to minimize the systemic bioavailability and the usual side-effects associated with oral drug administration. In this work, a drug delivery system is proposed to achieve an anti-inflammatory local effect using resveratrol (RSV). This study aims to develop microcapsules made of poly-(ε-caprolactone) (PCL) by ultrasonic atomization to preserve the antioxidant activity of RSV, to prevent its degradation and to suppress the inflammatory response in activated RAW 264.7 macrophages. An experimental design was performed to build a mathematical model that could estimate the effect of nozzle power and polymer concentration on particle size and encapsulation efficiency. RSV-loaded microcapsules showed adequate morphology, particle size, and loading efficiency properties. RSV formulations exhibited negligible cytotoxicity and an efficient amelioration of inflammatory responses, in terms of Nitric Oxide (NO), ROS (Reactive Oxygen Species), and lipid peroxidation in macrophages. Thus, RSV-loaded microcapsules merit consideration as a drug delivery system suitable for intra-articular administration in inflammatory disorders affecting the joint.

Abstract:
Background. Osteoarthrosis is characterized by wide prevalence, especially among patients of older age groups, a high risk of restrictions on the musculoskeletal system function, which results in the patients impaired ability to work and reduced quality of life. Aim: investigate the possibilities of thermographic assessment of efficiency of magnetotherapy of the articular syndrome in osteoarthrosis of the knee joints. Methods. The study included 57 patients (46 women and 11 men) with osteoarthrosis of the knee joints (mean age 61.27.4 years). The patients were divided into two groups. In the group 1 (n=29), magnetotherapy with the ALMAG-01 apparatus was used, while in the group 2 (n=28), the placebo apparatus was applied. Results. The inclusion of magnetotherapy in the complex therapy of osteoarthrosis of the knee joints, according to thermographic data, increases significantly the efficiency of treatment and differs significantly from the placebo magnetotherapy procedures. In patients with osteoarthrosis of the knee joints of the X-ray stages I and II, a combination of course treatment using a traveling pulsed magnetic field and standard drug therapy revealed a positive dynamics in normalization of the temperature balance of the joints. In patients with osteoarthrosis of the X-ray stage I, the minimum efficiency of magnetotherapy was revealed according to thermography, which indicates low severity of the inflammatory joint syndrome. Conclusion. The method of infrared thermography can be used as a diagnostic technique, as it reflects the degree of activity of the inflammatory process in the joints with osteoarthrosis.

This study examines whether sell-side analysts fully incorporate into their earnings forecasts the joint effects between accounting conservatism and changes in real investment on the quality of current earnings. Our results indicate that sell-side analysts do not fully incorporate such effects when they forecast future earnings so that they overestimate (underestimate) future earnings when current earnings are inflated (depressed) by those effects. Thus, we conclude that sell-side analysts do not recognize fully the joint effects between accounting conservatism and real activity on the earnings quality and that they need to mitigate their bias to enhance market efficiency by providing investors with a good benchmark for their earnings expectation.

Abstract:
Human Pose Estimation (HPE) is defined as the problem of human joints’ localization (also known as keypoints: elbows, wrists, etc.) in images or videos. It is also defined as the search for a specific pose in space of all articulated joints. HPE has recently received significant attention from the scientific community. The main reason behind this trend is that pose estimation is considered as a key step for many computer vision tasks. Although many approaches have reported promising results, this domain remains largely unsolved due to several challenges such as occlusions, small and barely visible joints, and variations in clothing and lighting. In the last few years, the power of deep neural networks has been demonstrated in a wide variety of computer vision problems and especially the HPE task. In this context, we present in this paper a Deep Full-Body-HPE (DFB-HPE) approach from RGB images only. Based on ConvNets, fifteen human joint positions are predicted and can be further exploited for a large range of applications such as gesture recognition, sports performance analysis, or human-robot interaction. To evaluate the proposed deep pose estimation model, we apply it to recognize the daily activities of a person in an unconstrained environment. Therefore, the extracted features, represented by deep estimated poses, are fed to an SVM classifier. To validate the proposed architecture, our approach is tested on two publicly available benchmarks for pose estimation and activity recognition, namely the J-HMDBand CAD-60datasets. The obtained results demonstrate the efficiency of the proposed method based on ConvNets and SVM and prove how deep pose estimation can improve the recognition accuracy. By means of comparison with state-of-the-art methods, we achieve the best HPE performance, as well as the best activity recognition precision on the CAD-60 dataset.

Abstract:
The operation of the bank is an important economic activity, which has contributed significantly to the development of the national economy over the past years. Therefore, the efficiency in the business operations of joint stock commercial banks is a topic that needs to be addressed, as it directly affects the efficiency in providing capital to businesses, as well as the stability and development of the financial market. Determining the factors affecting the profitability of joint-stock commercial banks will help managers to operate the bank's operations more efficiently. The purpose of this paper is to examine whether the profitability of a joint stock commercial bank in Vietnam is affected by its size and operating costs. All parameter estimates of the regression model are based on regression analysis of the random effects table of REM. Data of the study included 28 joint stock commercial banks in Vietnam for the period of 2012-2019. Empirical research results show that the bank's profit is affected by financial leverage and more particularly, there exists a positive correlation of the bank size and the operating cost on the bank's profit. The fact that larger banks are more competitive has the ability to diversify their asset portfolios, focus on higher profit margins, and spend money to investment, upgrading of service quality and facilities lead to increased operating costs, thereby reducing risks and increasing profits.

Abstract:
Context: Roller massagers are used as a recovery and rehabilitative tool to initiate muscle relaxation and improve range of motion (ROM) and muscular performance. However, research demonstrating such effects is lacking. Objective: To determine the effects of applying a roller massager for 20 and 60 seconds on knee-joint ROM and dynamic muscular performance. Design: Randomized controlled clinical trial. Setting: University laboratory. Patients or Other Participants: Ten recreationally active men (age = 26.6 ± 5.2 years, height = 175.3 ± 4.3 cm, mass = 84.4 ± 8.8 kg). Intervention(s): Participants performed 3 randomized experimental conditions separated by 24 to 48 hours. In condition 1 (5 repetitions of 20 seconds) and condition 2 (5 repetitions of 60 seconds), they applied a roller massager to the quadriceps muscles. Condition 3 served as a control condition in which participants sat quietly. Main Outcome Measure(s): Visual analog pain scale, electromyography (EMG) of the vastus lateralis (VL) and biceps femoris during roller massage and lunge, and knee-joint ROM. Results: We found no differences in pain between the 20-second and 60-second roller-massager conditions. During 60 seconds of roller massage, pain was 13.5% (5.7 ± 0.70) and 20.6% (6.2 ± 0.70) greater at 40 seconds and 60 seconds, respectively, than at 20 seconds (P < .05). During roller massage, VL and biceps femoris root mean square (RMS) EMG was 8% and 7%, respectively, of RMS EMG recorded during maximal voluntary isometric contraction. Knee-joint ROM was 10% and 16% greater in the 20-second and 60-second roller-massager conditions, respectively, than the control condition (P < .05). Finally, average lunge VL RMS EMG decreased as roller-massage time increased (P < .05). Conclusions: Roller massage was painful and induced muscle activity, but it increased knee-joint ROM and neuromuscular efficiency during a lunge.

The paper analyzes the effectiveness of magnetic resonance imaging with cartilage diagram in diagnosing signs of professional deforming arthrosis of knee joints in miners working in conditions of significant physical loading. Aim of the research – to determine of diagnostic efficiency of indicators of magnetic resonance imaging of the knee joint and cartilage diagram in miners of the main occupations suffering from deforming arthrosis. Methods. The research is conducted in 30 miners of basic occupations: 20 mining workers of breakage face (MWBF) and 10 machinists of shearer mining machines (МSMM) have been treated in the inpatient department of occupational pathology of the Lviv Regional Clinical Hospital in 2015-2017 due to deforming arthrosis. Damages of the main anatomical elements of the knee joint with arthrosis were analyzed, visualized initially with the help of MRI, and then - cartilage diagram. Results. According to the MRI data, in miners of the main occupations with arthrosis of the knee joint the posterior cross-shaped ligament are most commonly affected (in 75.0±9.7 % MWBF and 70.0±14.5 % МSMM), damage to the medial collateral ligament are diagnosed less frequently (in 5.0±4.9 % in the MWBF and in 10.0±9.5 % in the МSMM). On average 3.8±0.4 modified elements of the knee joint are visualized in patients, whereas 4.8±0.1 affected areas are visualized on the cartilage diagram (р<0.05). In 86.7±6.2 % patients, in the analysis of cartilage diagram, changes in all five analyzed areas are diagnosed, indicating a higher efficiency of the diagnosis of changes in the structures of the joint with DA of the professional etiology of the method of cartilage diagram compared with MRI. According to the cartilage diagram the most significant changes are noted in the hypertrophy of the femur: among all miners 62.5±0.3 ms (medial) and 62.6±0.4 ms (lateral), in the MWBF group the average time of Т2-delay is the largest in the area of the medial hypertrophy of the femur is 60.9±2.3 ms, in the МSMM group – in the area of the lateral hypertrophy of the femur: 66.7±3.3 ms, which can be linked to the peculiarities of the forced working position of miners of these professions and the kinetics of joint structures. These results can be used to diagnose the initial lesions of joint structures with DA of professional genesis, as well as the creation of prognostic models for determining the the degree of risk of development of knee joint damage, which will allow to improve the system of personified approach to diagnostic and preventive measures in working persons in conditions of considerable physical activity and forced working position.

Abstract:
Aims Unicompartmental knee arthroplasty (UKA) and bicompartmental knee arthroplasty (BCA) have been associated with improved functional outcomes compared to total knee arthroplasty (TKA) in suitable patients, although the reason is poorly understood. The aim of this study was to measure how the different arthroplasties affect knee extensor function. Methods Extensor function was measured for 16 cadaveric knees and then retested following the different arthroplasties. Eight knees underwent medial UKA then BCA, then posterior-cruciate retaining TKA, and eight underwent the lateral equivalents then TKA. Extensor efficiency was calculated for ranges of knee flexion associated with common activities of daily living. Data were analyzed with repeated measures analysis of variance (α = 0.05). Results Compared to native, there were no reductions in either extension moment or efficiency following UKA. Conversion to BCA resulted in a small decrease in extension moment between 70° and 90° flexion (p < 0.05), but when examined in the context of daily activity ranges of flexion, extensor efficiency was largely unaffected. Following TKA, large decreases in extension moment were measured at low knee flexion angles (p < 0.05), resulting in 12% to 43% reductions in extensor efficiency for the daily activity ranges. Conclusion This cadaveric study found that TKA resulted in inferior extensor function compared to UKA and BCA. This may, in part, help explain the reported differences in function and satisfaction differences between partial and total knee arthroplasty. Cite this article: Bone Joint Res 2021;10(1):1–9.

Transfection efficiencies varied among the cell types tested (bCH 28.4% ± 22.87, rTDSPC 18.13% ± 12.07, hBMSC 18.23% ± 14.80, hSDSC 26.63% ± 8.81) and while an increase of NPs with a constant amount of mRNA generally improved the transfection efficiency, an increase of the mRNA loading ratio (2:50, 4:50, or 6:50 w/w mRNA:NPs) had no impact. However, metabolic activity of bCHs and rTDSPCs was significantly reduced when using higher amounts of NPs, indicating a dose-dependent cytotoxic response. Finally, we demonstrate the feasibility of transfecting extracellular matrix-rich 3D cell culture constructs using the nanoparticle system, making it a promising transfection strategy for musculoskeletal tissues that exhibit a complex, dense extracellular matrix.

Abstract:
Objective: to search for an association between basal expression of the AMP-activated protein kinase (adenosine monophosphate-activated protein kinase, AMPK) gene in the blood of patients with rheumatoid arthritis (RA) and disease activity, as well as joint destruction before and after 24 months of methotrexate (MT) therapy.Patients and methods. The study included 40 patients with RA who met the 1987 ACR classification criteria, with a disease duration of not more than 2 years, with a mean age of 47.5±15.5 years. All patients received 15 mg/week of MT. The number of swollen joints (SJC) out of 44, the number of tender joints (TSC) out of 53, the duration of morning stiffness (in minutes) and RA activity were assessed using the DAS28 index. The concentration of CRP and IgM rheumatoid factor (RF), antibodies to cyclic citrullinated peptide (ACCP) were assessed. To evaluate the radiological progression of RA, the Sharp method modified by van der Heijde was used. Gene expression in peripheral blood cells was determined by reverse transcriptase reaction and real-time polymerase chain reaction. Results and discussion. During MT therapy, there was a decrease in disease activity by DAS28 index, CRP level, duration of morning stiffness, SJC and TJC. After 2 years, there were no statistically significant changes in the number of erosions, while the number of joints with narrowing of the joint space increased by the end of the study (p=0.004). Treatment with MT resulted in a decrease in AMPK expression. At the same time, patients who achieved remission had the highest level of AMPK before therapy, as did seronegative patients compared to seropositive ones. Based on the ROC analysis, the threshold value of AMPK gene expression was determined, which makes it possible to predict a high probability of a positive effect of MT therapy.Conclusion. AMPK gene expression is associated with disease activity. Its high initial blood level in RA patients is associated with greater efficiency of MT and, possibly, plays a protective role. With an AMPK gene expression index greater than 3.83, the probability of a positive response to MT is high.

Abstract:
Abstract. The prompt detection of explosive volcanic activity is crucial since this kind of activity can release copious amounts of volcanic ash and gases into the atmosphere, causing severe dangers to aviation. In this work, we show how the joint analysis of seismic and infrasonic data by wavelet transform coherence (WTC) can be useful to detect explosive activity, significantly enhancing its recognition that is normally done by video cameras and thermal sensors. Indeed, the efficiency of these sensors can be reduced (or inhibited) in the case of poor visibility due to clouds or gas plumes. In particular, we calculated the root mean square (RMS) of seismic and infrasonic signals recorded at Mt. Etna during 2011. This interval was characterised by several episodes of lava fountains, accompanied by lava effusion, and minor strombolian activities. WTC analysis showed significantly high values of coherence between seismic and infrasonic RMS during explosive activity, with infrasonic and seismic series in phase with each other, hence proving to be sensitive to both weak and strong explosive activity. The WTC capability of automatically detecting explosive activity was compared with the potential of detection methods based on fixed thresholds of seismic and infrasonic RMS. Finally, we also calculated the cross correlation function between seismic and infrasonic signals, which showed that the wave types causing such seismo-acoustic relationship are mainly incident seismic and infrasonic waves, likely with a common source.

Abstract:
The objectives of each economic organization can only be achieved as a result of the joint work of all its staff. They are most effectively achieved, provided they are in tune with the external environment and the factors in it. Also the motivation of staff plays critical role in this, and not the least, the selection of the staff and its objectvely examined competencies. The purpose of recruiting staff into the organization is to create a job reserve, taking into account the following circumstances: future change in organizational and staff relations, dismissals, relocations, retirement, premature termination of employment contracts, modification of the nature of the production program. Before making a decision to recruit new emploees, it is appropriate to identify other alternative options that can be used in the company. These include: extra work and increased work intensity; structural reorganization and use of new production schemes; reengineering; attracting specialized companies to carry out certain types of activities. Recruitment is a responsible task and the optimal number of staff needs to be taken into account when deciding how to do so. An insufficient number of workers, can have consequences, some of which may be a failure of the production program, occurrence of accidents at work, conflict situations in the collective. Labor surpluses lead to an increase in financial costs related to wages, a reduction in the interest in qualitative and highly skilled labor, the leave of skilled workers. The principal and main resource of each organization is the people who work in it. Therefore, the greatest importance should be given to the study of issues relating to the management of their behavior at work, the motivation of their activities. It is workers who create the production of the organization, and although machines and appliances have become "masters" of many technological processes, the role of man in the organization not only does not diminish but is constantly increasing. In today's dynamic economic development, people working in companies are too important, but also costly. That is why the people working in the organization are the main subject of management. The management of the company creates the personnel, establishes the system of relations between the people in the organization, allows to create the necessary conditions for the fullest manifestation of the employees' productive capacities and their intensive development, contributes to their training and growth. Management should create conditions in which workers feel satisfied with their work in the organization.

The virtual environment allows solving new educational problems, but also generates new educational, psychological and pedagogical effects, new experience. E-learning, like traditional learning, is realised with the help of three most common technologies: self-learning technology; one-to-one learning technology; one-to-many learning technology. At the same time, e-learning interaction involves self-organisation of the individual and the organisation of joint distributed activities of students. Self-organisation of the individual is understood as the ability to structure personal time, tactically plan own activities, purposefully act in solving problems. The effectiveness of educational activities of students in the conditions of e-learning depends on how students are guided by their own internal principles, the ability to create conditions for the optimization of learning tasks and actions. The article presents the results of theoretical and empirical research of self-organisation of the individual in the context of effective distribution of time resources, describes the psychological strategies of network activity of students, highlights the features of educational styles, levels of psychological efficiency of interaction with information technology.

Abstract:
Abstract A consistent theme in swine production is to increase efficiency and reduce input costs. In this study, sow activity traits and lesions were analyzed to identify associations with lameness and gilt retention. Gilts (n = 73) were culled based on structural unsoundness as determined by an experienced herdsman. Females (n = 132) that had been retained for breeding, but either did not show estrus at an appropriate age or were excessed due to limited farrowing space were used as controls. Gilts were recorded with the NUtrack System for 1 week prior to selection. The NUtrack System records distance travelled (m), time standing (s), eating (s), and laying (s), angle rotated (degrees) and average speed (m/s). Animals were humanely harvested in a USDA inspected abattoir. Both ends of the humerus and the femur head were evaluated for osteochondrosis or osteoarthritis lesions. Joint lesions were categorized as Major or Minor lesions. Major lesions were severe osteochondrosis lesions where cartilage was severely fractured or cartilage exhibited a region of necrosis larger than 2 millimeters in diameter. Minor lesions were defined creases or indentations in the cartilage or where abnormal blood flow existed in bone tissue underneath cartilage. Ninety-eight animals were identified with minor or major lesions (28 cull and 70 control). Unexpectedly, chi-square analysis did identify control animals as having higher incidences of minor or major lesions than cull animals (p < 0.05). Data were analyzed using logistic regression (RStudio V1.2.5033) with farrowing group included in the model. No NUtrack trait was predictive of joint lesions (P > 0.1). However, time standing (P < 0.001) and average speed (P < 0.05) were significantly associated with being retained for breeding. These data suggest that animal behavior and movement, as analyzed by NUtrack, can enhance herdsman efforts in making culling decisions of breeding animals.

Abstract:
Objective: to assess the long-term safety and efficiency of tocilizumab (TCZ) therapy in patients with early rheumatoid arthritis (RA) of moderate and high activity, who have completed the basic WA19926 study, as well as the rate of sustained drug-free remission.Subjects and methods. A long-term open-label multicenter Phase III extension study (ML28124) was conducted using a group of 49 patients (36 (73.5%) women and 13 (26.5%) men; mean age 53.3±10.8 years) with early RA of moderate and high activity. All the patients received an intravenous infusion of TCZ 8 mg/kg every 4 weeks for 104 weeks (a total of 27 infusions). The safety assessment criteria were the rate and severity of all adverse events (AE), serious AE (SAE), and AE of particular interest; the rate of AE causing drug dosage changes or withdrawal from the study; the frequency of clinically significant laboratory abnormalities. The analysis of efficiency (secondary end points) included changes of DAS8, which was calculated using erythrocyte sedimentation rate (ESR) (DAS28-ESR) and SDAI, the of tender joint count (TJC) and swollen joint count (SJC); the number of patients who had achieved drug-free remission; and the time to a RA exacerbation in patients who had achieved non-drug remission. Results and discussion. The total rate of AE was found to be 69.4%; that of SAE was 10.2%; SAE were 6.9% of the the total number of AE; AE of particular interest were 17.2% of the total number of AE. More than one-third (35.6%) of the AE caused drug dose changes or therapy discontinuation or complete cessation. The laboratory clinically significant abnormalities included those in complete blood cell counts (blood alanine aminotransferase, aspartate aminotransferase, total and direct bilirubin, creatinine, and glucose). The evaluation efficiency analysis showed that the main disease activity measures (DAS28-ESR, SDAI, SJC, and TJC) decreased at 104-week follow-up versus at baseline. The rate of drug-free remission was 71.4%; that of recurrence was 40.0% (the median time to recurrence was 23 weeks). The findings suggest that the safety profile of TCZ is acceptable in the long-term treatment of early RA and correspond to earlier results.

Abstract:
Abstract. Ice crystal formation in the mixed-phase region of deep convective clouds can affect the properties of climatically important convectively generated anvil clouds. Small ice crystals in the mixed-phase cloud region can be formed by heterogeneous ice nucleation by ice-nucleating particles (INPs) and secondary ice production (SIP) by, for example, the Hallett–Mossop process. We quantify the effects of INP number concentration, the temperature dependence of the INP number concentration at mixed-phase temperatures, and the Hallett–Mossop splinter production efficiency on the anvil of an idealised deep convective cloud using a Latin hypercube sampling method, which allows optimal coverage of a multidimensional parameter space, and statistical emulation, which allows us to identify interdependencies between the three uncertain inputs. Our results show that anvil ice crystal number concentration (ICNC) is determined predominately by INP number concentration, with the temperature dependence of ice-nucleating aerosol activity having a secondary role. Conversely, anvil ice crystal size is determined predominately by the temperature dependence of ice-nucleating aerosol activity, with INP number concentration having a secondary role. This is because in our simulations ICNC is predominately controlled by the number concentration of cloud droplets reaching the homogeneous freezing level which is in turn determined by INP number concentrations at low temperatures. Ice crystal size, however, is more strongly affected by the amount of liquid available for riming and the time available for deposition growth which is determined by INP number concentrations at higher temperatures. This work indicates that the amount of ice particle production by the Hallett–Mossop process is determined jointly by the prescribed Hallett–Mossop splinter production efficiency and the temperature dependence of ice-nucleating aerosol activity. In particular, our sampling of the joint parameter space shows that high rates of SIP do not occur unless the INP parameterisation slope (the temperature dependence of the number concentration of particles which nucleate ice) is shallow, regardless of the prescribed Hallett–Mossop splinter production efficiency. A shallow INP parameterisation slope and consequently high ice particle production by the Hallett–Mossop process in our simulations leads to a sharp transition to a cloud with extensive glaciation at warm temperatures, higher cloud updraughts, enhanced vertical mass flux, and condensate divergence at the outflow level, all of which leads to a larger convectively generated anvil comprised of larger ice crystals. This work highlights the importance of quantifying the full spectrum of INP number concentrations across all mixed-phase altitudes and the ways in which INP and SIP interact to control anvil properties.

Abstract:
The article clarifies the essence and significance of the concept of «community policing» in the activities of the National Police of Ukraine. It is established that the specified socially oriented concept provides endowment of territorial communities with the administrative levers which allow to influence quality of rendering of police law enforcement services. It was stated that the implementation of the concept of «community policing» involves the development of strategic directions to combat criminal and administrative offenses by joint efforts of the police and the community, increasing the number of patrols, involving representatives of local communities, creating legal conditions for public control and police accountability. at the local level. Forms of interaction between territorial communities and the police are classified depending on: the implementation of the tasks facing the participants of the partnership; meaningful content of tasks; spheres of activity; the nature of the interaction; duration of interaction; initiator of interaction. It is noted that a characteristic feature of the directions of administrative activity of the police is their versatility, which indicates that the police equally interacts both with state authorities and public entities. Therefore, in the context of our study, the external organizational direction of the police administrative activity, within which the police interacts with territorial communities. The legislative level of the police and territorial communities are carried out in the form of concluding joint projects between the specified institutions, the procedure for the implementation of which is established in Article 89 of the Law of Ukraine «On National Police». In particular, part 1 of this article provides for the preparation and implementation of joint projects between local communities and the police in the process of their interaction to meet the needs of the population and improve the efficiency of the police tasks assigned to it. Abstract:
Aims The purpose of this exploratory study was to investigate if the 24-hour activity profile (i.e. waking activities and sleep) objectively measured using wrist-worn accelerometry of patients scheduled for total hip arthroplasty (THA) improves postoperatively. Patients and Methods A total of 51 THA patients with a mean age of 64 years (24 to 87) were recruited from a single public hospital. All patients underwent THA using the same surgical approach with the same prosthesis type. The 24-hour activity profiles were captured using wrist-worn accelerometers preoperatively and at 2, 6, 12, and 26 weeks postoperatively. Patient-reported outcomes (Hip Disability and Osteoarthritis Outcome Score (HOOS)) were collected at all timepoints except two weeks postoperatively. Accelerometry data were used to quantify the intensity (sedentary, light, moderate, and vigorous activities) and frequency (bouts) of activity during the day and sleep efficiency. The analysis investigated changes with time and differences between Charnley class. Results Patients slept or were sedentary for a mean of 19.5 hours/day preoperatively and the 24-hour activity pattern did not improve significantly postoperatively. Outside of sleep, the patients spent their time in sedentary activities for a mean of 620 minutes/day (sd 143) preoperatively and 641 minutes/day (sd 133) six months postoperatively. No significant improvements were observed for light, moderate, and vigorous intensity activities (p = 0.140, p = 0.531, and p = 0.407, respectively). Sleep efficiency was poor (< 85%) at all timepoints. There was no postoperative improvement in sleep efficiency when adjusted for medications (p > 0.05). Patient-reported outcome measures showed a significant improvement with time in all domains when compared with preoperative levels. There were no differences with Charnley class at six months postoperatively. However, Charnley class C patients were more sedentary at two weeks postoperatively when compared with Charnley class A patients (p < 0.05). There were no further differences between Charnley classifications. Conclusion This study describes the 24-hour activity profile of THA patients for the first time. Prior to THA, patients in this cohort were inactive and slept poorly. This cohort shows no improvement in 24-hour activity profiles at six months postoperative. Abstract:
ABSTRACTChikungunya virus (CHIKV) is a mosquito-borne arthralgic alphavirus that has garnered international attention as an important emerging pathogen since 2005. More recently, it invaded the Caribbean islands and the Western Hemisphere. Intriguingly, the current CHIKV outbreak in the Caribbean is caused by the Asian CHIKV genotype, which differs from the La Réunion LR2006 OPY1 isolate belonging to the Indian Ocean lineage. Here, we adopted a systematic and comparative approach against LR2006 OPY1 to characterize the pathogenicity of the Caribbean CNR20235 isolate and consequential host immune responses in mice.Ex vivoinfection using primary mouse tail fibroblasts revealed a weaker replication efficiency by CNR20235 isolate. In the CHIKV mouse model, CNR20235 infection induced an enervated joint pathology characterized by moderate edema and swelling, independent of mononuclear cell infiltration. Based on systemic cytokine analysis, localized immunophenotyping, and gene expression profiles in the popliteal lymph node and inflamed joints, two pathogenic phases were defined for CHIKV infection: early acute (2 to 3 days postinfection [dpi]) and late acute (6 to 8 dpi). Reduced joint pathology during early acute phase of CNR20235 infection was associated with a weaker proinflammatory Th1 response and natural killer (NK) cell activity. The pathological role of NK cells was further demonstrated as depletion of NK cells reduced joint pathology in LR2006 OPY1. Taken together, this study provides evidence that the Caribbean CNR20235 isolate has an enfeebled replication and induces a less pathogenic response in the mammalian host.IMPORTANCEThe introduction of CHIKV in the Americas has heightened the risk of large-scale outbreaks due to the close proximity between the United States and the Caribbean. The immunopathogenicity of the circulating Caribbean CHIKV isolate was explored, where it was demonstrated to exhibit reduced infectivity resulting in a weakened joint pathology. Analysis of serum cytokine levels, localized immunophenotyping, and gene expression profiles in the organs revealed that a limited Th1 response and reduced NK cells activity could underlie the reduced pathology in the host. Interestingly, higher asymptomatic infections were observed in the Caribbean compared to the La Réunion outbreaks in 2005 and 2006. This is the first study that showed an association between key proinflammatory factors and pathology-mediating leukocytes with a less severe pathological outcome in Caribbean CHIKV infection. Given the limited information regarding the sequela of Caribbean CHIKV infection, our study is timely and will aid the understanding of this increasingly important disease.
