Full textFatigue, along with stress corrosion is the main cause of degradation observed in the components of French nuclear power plants. The justification of components regarding fatigue crack propagation is currently considered in a penalising way without taking into account the loading history. The study of fatigue closure effects makes it possible to determine driving forces representative of crack propagation. Loads inducing low cycle fatigue in primary components are of thermal origin. A campaign of reference tests on standardised specimens was first carried out before controlled total deformation tests on uniaxial specimens. An experimental set-up using Heaviside-DIC and electrical potential difference monitoring techniques was used to monitor crack propagation while evaluating crack opening rates at the surface and averaged over the specimen. It is shown that for the two materials studied, crack opening rates vary as a function of the level of stress applied, the load ratio and, to a lesser extent the crack depth. The combination of experimental results and the development of finite-element models makes it possible to define and evaluate driving forces based on deformation intensity factors and the J integral. A new crack propagation driving force calculation method adapted to cases of confined plasticity at the crack tip and to cases of generalised plasticity has been defined, making it possible to obtain a unified propagation law for each material that does not depend on the load ratio or the stress level

Full textThe behaviour of 2D physically short cracks has been studied for an austenitic stainless steel type A304L. Experimental fatigue tests have shown an increase of the threshold fatigue range when the crack length is increasing from 0. 1 to 0. 8 mm. This effect is explained in term of crack closure variation. The numerical simulation by mean of finite elements supports such an effect of crack closure when computing is performed in plane stress at R=0. 1 and R=0. 7. Such behaviour is typical for a material characterized by an isotropic and cinematic constitutive cyclic hardening law. The local plasticity effect is shown to control the crack growth rate and has been related to an effective plastic intensity factor range

Full textInwardly-rectifying potassium (Kir) channels are transmembrane proteins involved in fundamental physiological processes. Pathologies are directly linked to a mutation of these channels: some mutations in Kir2.1 and Kir6.2 cause the Andersen’s syndrome and hyperinsulinism, respectively. This study has four objectives: i) to understand the gating mechanism of the KirBac3.1, ii) to understand the impact of the mutation W46R on the channel (W68R equivalent in Kir6.2 causing hyperinsulinsm), iii) to understand the impact of the mutation S129R on the channel, iv) to resolve the structure of Kir2.1 at an atomic level. To study the dynamical characteristics of this system, Molecular Dynamics using excited Normal Modes (MDeNM) method was used. The results were confronted to HDX-MS Spectrometry and electrophysiological experiments for validation. To determine the structure of the human Kir2.1 WT, we are using cryo-electron microscopy combined with image analysis. Key motions in the gating mechanism were determined like the involvement of the cytoplasmic domain, the slide-helix and the transmembrane helices during the opening of the channel. Concerning W46R, a modification in these interaction at the level of the residue 46 causes the loss of interactions between the bottoms of the two transmembrane helices that triggers the uprising of the slide-helix which opens the channel. The study of KirBac3.1 S129R mutant in the open state explains how the mutation pushes away the point of constriction and keeps the channel open with a surprising role of the mutation R129. We investigated the structure of Kir2.1 by cryo-EM and obtained an electronic density at 7 Å resolution

Full textDamage tolerance analyses of rotating parts, containing 2D-short crack, in turbomachines of helicopters is the main focus of this thesis.Fatigue crack propagation tests were performed at R=0.1 on two bimodal Titanium alloys at different temperatures (20°C and 400°C for TA6V and 20°C, 200°C and 500°C for Ti6242). Near-threshold effective behaviors (after crack closure correction) were approximately the same for the two alloys attested temperatures. Crack closure was principally due to oxide layer at high temperatures and to fracture surface roughness at ambient temperature.Then the variations of the crack propagation threshold with crack length was studied at R=0.1 and different temperatures. A numerical tool was developed to automatically detect crack closure level more precisely for, as short as, 80μm cracks. Crack closure levels measured at different crack lengths obtained from progressive machining of the plastic wake were quite similar to the ones obtained during propagation of 2D-short cracks near the threshold, except at high temperatures where oxide-induced crack closure developed rapidly. Differences in behavior of long cracks and short cracks were explained by a reduction of crack closure level when crack wake was not fully developed.A non-propagation criterion based on formalism initially proposed by Kitagawa and a modified-Paris propagation law allow to give a better description of physically 2D-short crack propagation resistance

Full textDans ce cadre, nous nous somme intéressés dans cette thèse à la possibilité d'utiliser les pompes à électrons de type R à 3 jonctions de façon métrologique autour de 10 pA. Notre montage expérimental est conçu afin de mesurer le courant quantifié généré par le dispositif en utilisant un comparateur cryogénique de courants (CCC).
Les principaux résultats ont été obtenus avec une pompe fabriquée par la PTB couplée à un CCC connecté en contre-réaction interne, i.e. utilisé comme un amplificateur de courant et ne donnant qu'une valeur relative du courant. Ces expériences ont permis de montrer l'existence de marches de courant jusqu'à environ 16 pA et une stabilité du courant généré par le dispositif sur des temps longs, indépendamment de la fréquence de pompage jusqu'à 100 MHz. Le niveau de bruit blanc est de 10-15 fA/Hz^(1/2) et, après 7 heures de mesure à une fréquence de 100 MHz, une incertitude relative de type A de 4.10^(-6) a été atteinte.
La dernière partie de ce travail présente la première réalisation directe de l'ETM. Dans ce cas, le courant généré par la pompe traverse une résistance étalonnée et la tension aux bornes de celle-ci est comparée à la tension délivrée par un réseau Josephson. Les premiers résultats qui sont présentés semblent prometteurs.

Full textThis work provides an experimental contribution to the study of the Screech phenomenon. Various experimental techniques such as microphones array, Schlieren and Particle Image Velocimetry (PIV) together with advanced post-processing techniques like azimuthal Fourier decomposition and Proper Orthogonal Decomposition (POD) are employed. These techniques enable the evaluation of the lip thickness effects on the jets generated by two different round nozzles. The differences on the flow aerodynamics and acoustics are discussed. Then, we carry out experiments to analyse the effects of the different dominant Screech modes (B and C) on the flow characteristics. No noticeable differences are found in the mean fields. However, the fluctuation fields shows the contrary: B mode has larger fluctuation. In the last part, we investigate the Screech closure mechanism. The signature of upstream jet waves is revealed in the axisymmetric (A2) and helical (C) mode. However, the mode B does not present evidence of this instability in the flow, indicating that its closure mechanism may be bonded to another kind of waves. The conclusion from the results is that the Screech phenomenon seems be driven by different closure mechanisms
