2 g tomatoes was used for MAP storage in which the gas composition equilibrated to 6% O2/ 4% CO2 and a carbon dioxide incubator was used for CA storage in which the CO2 level was monitored and maintained as 5% through the term of storage at 7 and 22°
C. During the research, the effect of ozone treatment (5-30 mg/L ozone gas for 0-20 min) was also considered for surface sanitation. The results demonstrate that S.Enteritidis can survive and/or grow during the storage of tomatoes depending on the location site of the pathogen on fruit, suspension cell density and storage temperature. During MAP, CA and air storage, S.Enteritidis with initial population of 7.0 log10 CFU/tomato survived on tomato surfaces with an approximate decrease of 4.0-5.0 log10 CFU/tomato in population within the storage period
however, in the case of initial population of 3.0 log10 CFU/tomato, cells died completely on day 4 during MAP storage and on day 6 during CA and air storage. The death rate of S.Enteritidis on the surfaces of tomatoes that were stored in MAP was faster than that of stored in air. Storage temperature was effective on the survival of S.Enteritidis for the samples stored at ambient atmosphere

C. Stem scars provided protective environments for Salmonella
an approximate increase of 1.0 log10 CFU/tomato in stem-scar population was observed during MAP, CA and air storage at 22°

C. The development of the microbial association in tomatoes was dominated by lactic acid bacteria (LAB). The pH values of the tomatoes changed approximately from 4.0 to 3.0 during the storage period. LAB grew well under all atmospheric conditions with or without the presence of S.Enteritidis. Gaseous ozone treatment has bactericidal effect on S.Enteritidis, inoculated on the surface of the tomatoes. 5 mg/L ozone gas treatment was not effective. 30 mg/L ozone gas treatment affected surface color.

Full textPorphyrins consist of twenty-atom rings containing four nitrogen atoms and can be used as sensor to detect odours and gases.
This thesis investigates whether or not porphyrins can be used as functional materials on grid gate devices. Drops of PVC embedded porphyrins were deposited on the surface of a grid gate which is a Metal Oxide Semiconductor (MOS) capacitor. In order to detect the gas sensing properties of the porphyrins a light addressable method called Scanning Light Pulse Technique (SLPT) has been used.
Drops of porphyrins were deposited with a stretched capillary tube (1 mm diameter).
The MOS capacitor has been exposed to nitrogen atmosphere as reference environment, while the target gases were carbon monoxide (100 ppm) and ammonia (500 ppm).
The result from the eight porphyrins is that one of them [Pt(II) TPP] has a response for both gases, ammonia induces a change in both the work function and surface resistance, while the carbon monoxide induces only a change in the surface resistance.

Full textIn this research, initial storage studies were done to determine the effect of various atmospheres (air, vacuum, oxygen absorbent and gas packaging) on the microbial changes of packaged chicken thighs followed by challenge studies with a strain of S. enteritidis$ sp{ rm{NAST}}$. Chicken thighs were packaged in Cryovac bags and stored at 4 and 12$ sp circ$C for up to 28d. Changes in headspace gas composition, pH, drip loss, color and odor were monitored at each sampling day.
The effect of various packaging treatments, dipping solutions (chitosan (0.2%w/v) and potassium sorbate (0.2%w/v)) and low dose irradiation (1.5 & 3.0 kGy) on the growth of S. enteritidis$ sp{ rm NAST}$ and on the shelf-life of chicken thighs stored at 4 and 12$ sp circ$C was also investigated. Fucan is a term used to denominate L-fucose rich sulfated polysaccharides. The fucans have been studied due their pharmacological activities like antithrombotic, antiproliferative and antioxidant. We have extracted three fucan fractions from the brown seaweed Spatoglossum schr?ederi. These fucans were denominated Fuc B 1, Fuc B 1.5 and Fuc B 2. The chemical analyzes show that the fucans have very similar composition as demonstrated by agarose electrophoresis gel, sugar and sulfate content. The antiproliferative effect was determined by MTT and BrdU methodologies in CHO cells. The inhibition of proliferation effect of the three fractions was about 40%. Therefore this we proceed just with the Fuc B 2 due the higher yield. There is no apoptosis indication using the anexin V/propidium iodide test. We found a cell cycle phase G1 arrest. The western blotting show that the PKC; pFAK; pERK 1/2 are activated when the cells were treated with fucans. The treatement with inhibitor of MAPK PD98059 extinguished the fucan effect. These results indicates that fucan act by the ERK pathway inducing the cell death.

Essas fucanas, denominadas de Fuc B 1, Fuc B 1.5 e Fuc B 2, apresentam uma composi??o muito similar como demonstrado pela eletroforese em gel de agarose, e conte?do de a??car e sulfato. O efeito antiproliferativo foi determinado pelas metodologias de MTT e BrdU em c?lulas CHO. O efeito na inibi??o da prolifera??o das tr?s fra??es foi de cerca de 40%. Assim, procedemos somente com a Fuc B 2 devido seu maior rendimento. N?o houve indica??o de apoptose usando a marca??o com anexina V-FITC/ Iodeto de Prop?deo. Identificamos uma parada na fase G1 do ciclo celular. Os ensaios de western blotting mostraram que PKC, pFAK e pERK 1/2 s?o ativadas quando as c?lulas s?o tratadas com Fuc B. O tratamento com o inibidor de MAPK PD98059 aboliu o efeito da fucana. Esses efeitos indicam que a fucana age via ERK para inibir a prolifera??o das c?lulas.

Full textEndast en tredjedel av alla IT-projekt lyckas. Detta beror till viss del på att cheferna inte involveras tillräckligt i designprocessen. Användarna som IT-projektet är tänkt för har inte fokuserats på tillräckligt, utan istället har det lagts ned mer tid på de tekniska aspekterna. Effektkartläggaren löserproblemen som uppstår när cheferna och målgrupperna inte är tillräckligt involverade i IT-projekt, genom att samarbeta med båda parterna. I detta forskningsprojekt har metoden effektkartläggningundersökts och hur metoden används av effektkartläggare för att hjälpa verksamheter att identifiera och nå sina mål. Effektkartläggaren samarbetar med verksamheten för att formulera syftet med IT-projektet. Samarbetet med målgrupperna leder till att effektkartläggaren får reda på hur användarna använder IT-produkten. För att de därmed ska kunna bidra till att verksamhetens mål uppnås när användarna interagerar med IT-produkten. Detta resulterar slutligen i att en effektkarta skapas. Denna effektkarta är sedan utgångspunkten för prototyper som effektkartläggaren skapar för uppdragsgivarens nya webbsida. Sedan är det webbutvecklare som utgår från prototyperna när de skapar verksamhetens webbsida. Det är däremot cheferna som ansvarar för alla beslut i projektet. En effektkartläggare rekommenderar enbart vilka målgrupper som kan tänkas prioriteras utifrån hurde kan bidra till att verksamhetens mål nås. Detta begränsar effektkartläggaren i sitt arbete. Sedan läggs även ett stort ansvar på effektkartläggaren gällande att förstå verksamheten och målgruppen. Genom användandet av intervjuer har det även undersökts vilka möjligheter och begränsningar effektkartläggare ser när de samarbetar med chefer och målgrupper. I detta forskningsprojekt har vi också tagit reda på vilka möjligheter och begränsningar effektkartläggning har rörande utvecklingen av en ny webbsida.

Within the brain, cells (neurons) communicate with each other to pass along information. This communication is important for normal functions of the brain such as learning and memory, muscle movement, etc. Epilepsy is a disease of the brain that is caused by rapid over synchronized communication between cells. This leads to seizures which can include convulsions, loss of attention, and much more. Currently, 30% of patients suffering from epilepsy do not have a treatment option that works for them, it is, therefore, imperative to investigate new targets for treatment in this disease. Connexin36 is a protein in the brain that directly connects cells so they can pass information quickly between them. Connexin36, therefore, might make a good target for treatment. Previous work has aimed to understand this relationship but has been limited in their ability to look at the entire brain at any one time. The goal of this study was to understand the relationship between connexin 36 and brain hyperactivity across the whole brain simultaneously. To understand this relationship, we first determined what happened to brain activity if the protein was missing entirely after exposure to a seizure causing drug. We were asking: How does connexin 36 affect hyperactivity. We found that different regions of the brain responded differently without the connexin 36 protein. This suggests that one size does not fit all, and one must look at the whole brain to understand the effects of the connexin 36 protein. Next, we asked a similar question, but in the opposite direction, how does hyperactivity affect connexin 36? We found, in the short-term, hyperactivity reduced the amount of connexin 36 present in certain regions of the brain. This continued until 3 hours following exposure to the seizure causing drug Pentylenetetrazol (PTZ). Lastly, to determine if this short-term reduction in connexin 36 meant that an individual was more likely to experience hyperactivity. To do this, we used a connexin 36 blocking drug, then introduced the seizure causing drug at different concentrations. We found, at all concentrations, the connexin 36 blocking drug caused significant changes in neuronal activity, depending on the brain regions. Overall, our results showed that connexin 36 plays an important role in hyperactivity and that a short-term reduction in connexin 36 is detrimental, and may contribute to an increase in the possibility of subsequent hyperactivity.

Full textDue to their sessile lifestyle, plants are constantly subjected to a variety of biotic and abiotic stresses causing tremendous yield losses. To survive under these conditions, plants have evolved different sigualing pathways allowing efficient stress responses. The Mitogen Activated Protein Kinase (MAPKs) are key sigual transduction molecules, which respond to varions external stimuli. However, the activity of MAPKs has to be strictly regulated by protein phosphatase such as MAPK Phosphatase (MKPs). ln the present study we describe the regulation ofTMKPI, a wheat MKP, by Calmodulins (CaM) and 14-3-3 proteins. We fust showed that phosphatase activity oTMKP1 is stimulated by K\ Li\ Mg2+ and especially by Mo2+. Using GST pull dawn assays, we demonstrated that TMKP1 binds to CaM in a Ca2+-dependent manner. Moreover, the CaM/Ca2+ complex inhibits the catalytic activity of TMKP1 in a CaM-dose dependent marmer. However, in the presence of Mo2+, this activity is enhanced by CaM/Ca2+ complex. Such effects were not reported so far, and raise a possible role for CaM and Mo 2+ in the regulation of plant MKPs during cellular response to extemal siguals. Moreover, a fine sequence analysis ofTMKPl revealed the presence of a conserved 14-3-3 mode 1 binding site. This finding incited us to perfonn co-immunoprecipitation assays on wheat protein extracts through which we provide proof-of-concept evidence for interaction of TMKP1 with 14-3-3 proteins. Interestingly, the phosphatase activity of TMKP1 was shawn to be enhanced by several plant and yeast 14-3-3 isoforms. Finally, the involvement of TMKP1 in plant stress responses was evaluated. Our data showed that the overexpression of TMKP1 in Arabidopsis resulted in a higher tolerance to salt stresses and to bacterial infection caused by P. syringae. Such findings suggest thal TMKP1 may act as a positive regulator of Arabidopsis responses to salt stresses and P. syringae infection. All together, our results provide novel functional properties for plant MKPs, and should add new knowledge to our understanding of the raie of these phosphatases in the control of MAPK signaling pathways controlling plant responses to various environmental stresses

Hypsometric color is one of the most important cartographic design elements, and it affects regional height and terrains perception to map readers. Hypsometric color combined with hill shading was commonly used to present landform. There are different opinions in choosing hypsometric colors in geographic field and publishing field. What kind of hypsometric colors is more attractive to map readers? And more essentially, which one is more effective in transmitting landform information? Researchers used 15 maps (5 hypsometric colors, including 2 cartographic theory colors, 2 commonly used colors in publishing field, 1 commonly used color in atlas, and 3 different regions) to test readers with different backgrounds. By using visual experiment, this paper attempts to find out the favorite and acceptable color type. Moreover, using statistics analysis to find out the most effective color type for height information transmitting, and to analyze the behavior variation of map reader for different hypsometric color maps. The quantitative results suggested that the color used in common atlas was the favorable color to the subjects. Nevertheless, the color combined with hill shading which was originally assumed better feedback turned out to be in worse preference. In the height information transmitting parts, however, the colors concerned cartography exhibited the better performances, both in the accuracy and value of height. The research results showed the variation between acceptable degree of subjects and efficiency of height information transmitting in different hypsometric colors.

Test theory is an explanation of empirical relationships among examination data. The modern test theory is based on the Item Response Theory (IRT), which considers the parameters of test item and the response of test-receiver (including difficulty, discrimination, ability and so on),and the estimation of its test-receiver’s ability becomes more precise. Concept Effect Relation Map (CERM) constructed by data mining with naïve data preprocessing causes: (1) monotonous concept fuzzification result, (2) the association rules may not reflect the real concept relation and (3) the circulating association rules exit. In this thesis, we apply the IRT as the assessment of students’ concept learning response. With the consideration of the difficulty and the discrimination of test item, we propose an IRT-Based Data Preprocessing Concept Effect Relation Map Construction System. IRT-Based Data Preprocessing Concept Effect Relation Map Construction System includes two modules: the Data Preprocessing Module and the Data Mining Module. The former has four procedures: Test Item Analysis, Learning Response Index (LRI) Generator, Concept Decomposition/Aggregation and Fuzzy ACLR Generator, and the latter has two procedures: Association rule mining and concept map constructor. The experiment results of the proposed Approach show that the CERM construction can be improved and the number of circulated association rules generated can be reduced.

In recent years, the rapid progress in internet dues to huge losses of organizations and enterprises. Therefore, the development of information security is becoming an important issue. According to the research of information security that personal careless mistake leads to almost 50% of information security event. Hence, the information security awareness is very important. The invests in hardware and software cannot improve the information security without improving the personal information security awareness. It points out that knowledge map is useful for learning and the concept of awareness. The research is expected to find the benefit of information security awareness material designed by knowledge map. Otherwise, in order reduce the ratio of information security event and raise the personal information security awareness; we expect to find the best approach to design information security awareness materials.

Full textSketch map is an important way to represent spatial information used in many geospatial reasoning tasks (Forbus, K., Usher, J., & Chapman, V. 2004). Compared with verbal or textual language, sketch map is a more interactive mode that more directly supports human spatial thinking and thus is a more natural way to reflect how people perceive properties of spatial objects and their spatial relations. One challenging application of sketch maps is called Spatial-Query-by-Sketch proposed by Egenhofer. Being a design of query language for geographic information systems (GISs), it allows a user to formulate a spatial query by drawing the desired spatial configuration with a pen on a touch-sensitive computer screen and get it translated into a symbolic representation to be processed against a geographic database (Egenhofer, M. 1997). During the period of sketch map drawing, errors due to human spatial cognition in mind may occur. A ready example is as follows: distance judgments for route are judged longer when the route has many turns or landmarks or intersections (Tversky, B. 2002). Direction get straightened up in memory. When Parisians were asked to sketch maps of their city, the Seine was drawn as a curve, but straighter than it actually is (Milgram, S. and Jodelet, D. 1976). Similarly, buildings and streets with different shapes are often simply depicted as schematic figures like blobs and lines. These errors are neither random nor due solely to ignorance; rather they appear to be a consequence of ordinary perceptual and cognitive processes (Tversky, 2003). Therefore, when processing sketch map analysis and representing it in a formal way, like Egenhofer's analysis approach for Spatial-Query-by-Sketch, the resulting formalization must necessarily be wrong if it does not account for the fact that some spatial information is distorted or omitted by humans. Therefore, when sketch map analysis is processed and represented in a formal way same as Egenhofer’s analytical approach to Spatial-Query-by-Sketch, the resulting formalization is simply erroneous since it never takes into account the fact that some spatial information is distorted or neglected in human perceptions. Though Spatial-Query-by-Sketch overcomes the limitations of conventional spatial query language by taking into consideration those alternative interaction methods between users and data, it is still not always true that accuracy of its query results is reliable.(...)
