Various KKN-DR activities start from observation, program planning, program consultation in accordance with the planned program, namely the KKN-DR activity held in the form of vidio edting training with the title "training on the use of vidio edting as a medium in ICT-based learning and marketing during the COVID-19 pandemic". The work of this report is shown to provide vidio editing training for educational needs and personal interests and provide an understanding of the importance of interesting learning media during the Covid-19 pandemic. The result of this ilmiyah work is the sumbersari community who participate in vidio edting training in increasing the needs needed and to utilize this increasingly sophisticated HP media.

Abstract:
Abstract Adoptive transfer of regulatory T cells (Treg) is therapeutic in T1D mouse models. Notably, Treg specific for pancreatic islets are more potent than polyclonal Treg in blocking disease. However, the frequency of antigen-specific Treg is extremely low and ex vivo expansion has the potential to destabilize Treg leading to an effector phenotype. Here, we developed methods to generate durable, antigen-specific engineered (ed)Treg from primary human CD4+ T cells using a combination of lentiviral TCR transduction and FOXP3 homology-directed repair (HDR)-editing. Using TCRs derived from clonally expanded CD4+ T cells in T1D, we generated islet-specific edTreg that exhibit a Treg-like phenotype. Islet-specific edTreg effectively suppress proliferation and cytokine production by islet-specific effector T cells (Teff). Notably, edTreg suppress Teff recognizing the identical peptide as well as bystander Teff recognizing alternative islet antigens. Consistent with this, islet-specific edTreg suppress polyclonal islet-specific T cells derived from PBMC. Further, edTreg expressing TCR with high avidity have superior suppressive capacity to those expressing TCR with low avidity. To directly assess the capacity of edTreg to modulate T1D in vivo, we established an identical HDR-editing strategy in islet-specific murine cells. Adoptively transferred islet-specific mu-edTreg homed to the pancreas and blocked diabetes triggered by islet-specific Teff in recipient mice. Collectively, our approach enables the production of edTreg specific to pancreatic islets with the capacity to efficiently suppress islet specific responses. This approach has the capacity to deliver targeted islet specific therapy to treat or prevent T1D.

Abstract:
This research sought to establish an eco-driving training system based on a driving simulator. The eco-driving training system contained five modules: human machine interface, data management, scene management, mode management, and evaluation algorithm management. It was proposed to base the new eco-driving training system on drivers’ individual characteristics. This system first asked drivers to conduct a diagnostic drive on a stretch of roadway in a driving simulator. The data on each driver’s non-ecological driving behaviors under different events were collected. Then each driver was given a customized training course based on an evaluation of his/her driving behaviors during the diagnostic drive. This training process is called eco-driving training based on individual characteristics (EDTIC). Eighty taxi drivers were recruited and divided into two groups for eco-driving training. One group was trained by watching videos, and the other was trained by the EDTIC training. An analysis of results shows that the EDTIC training was significantly more effective than traditional video training. Under the EDTIC training, all driving behaviors improved and emissions and fuel consumption were greatly reduced; the reduction was as great as 8.3–8.4%. The EDTIC training was proven effective at improving the eco-driving behavior of taxi drivers (i.e., professional drivers), and it could also be employed to train other professional drivers (bus and truck drivers) and non-professional drivers.

Abstract:
Thymic regulatory T cells (tTregs) are potent inhibitors of autoreactive immune responses, and loss of tTreg function results in fatal autoimmune disease. Defects in tTreg number or function are also implicated in multiple autoimmune diseases, leading to growing interest in use of Treg as cell therapies to establish immune tolerance. Because tTregs are present at low numbers in circulating blood and may be challenging to purify and expand and also inherently defective in some subjects, we designed an alternative strategy to create autologous Treg-like cells from bulk CD4+ T cells. We used homology-directed repair (HDR)–based gene editing to enforce expression of FOXP3, the master transcription factor for tTreg. Targeted insertion of a robust enhancer/promoter proximal to the first coding exon bypassed epigenetic silencing, permitting stable and robust expression of endogenous FOXP3. HDR-edited T cells, edTregs, manifested a transcriptional program leading to sustained expression of canonical markers and suppressive activity of tTreg. Both human and murine edTregs mediated immunosuppression in vivo in models of inflammatory disease. Further, this engineering strategy permitted generation of antigen-specific edTreg with robust in vitro and in vivo functional activity. Last, edTreg could be enriched and expanded at scale using clinically relevant methods. Together, these findings suggest that edTreg production may permit broad future clinical application.

Abstract Protoplanets formed in a marginally gravitationally unstable (MGU) disk by either core accretion or disk instability will be subject to dynamical interactions with massive spiral arms, possibly resulting in inward or outward orbital migration, mergers with each other, or even outright ejection from the protoplanetary system. The latter process has been hypothesized as a possible formation scenario for the unexpectedly high frequency of unbound gas giant exoplanets (free floating planets, FFPs). Previous calculations with the EDTONS fixed grid three-dimensional (3D) hydrodynamics code found that protoplanets with masses from 0.01 M ⊕ to 3 M Jup could undergo chaotic orbital evolutions in MGU disks for ∼1000 yr without undergoing monotonic inward or outward migration. Here the Enzo 2.5 adaptive mesh refinement 3D hydrodynamics code is used to follow the formation and orbital evolution of protoplanets in MGU disks for up to 2000 yr. The Enzo results confirm the basic disk fragmentation results of the EDTONS code, as well as the absence of monotonic inward or outward orbital migration. In addition, Enzo allows protoplanet mergers to occur, unlike EDTONS, resulting in a significant decrease in the number of protoplanets that survive for 1000–2000 yr in the Enzo models. These models also imply that gas giants should be ejected frequently in MGU disks that fragment into large numbers of protoplanets, supporting ejection as a possible source mechanism for the observed FFPs.

Abstract:
The development of the EU’s security and defence profile has been a constant over the last two decades and one of the most dynamic projects of European integration. In this sense, last few years have recorded significant progress, illustrated both by the conduct of a significant number of civilian and military operations in different geographical perimeters, as well as by the launch of cooperation initiatives in the field of defence capabilities and defence research. The reporting framework of these developments has also undergone major changes, integrated into the process of establishment the European Defence Technological and Industrial Base (EDTIB). After an initial stage of conceptual structuring, carried out between 2007-2013, the profile of this construct has matured rapidly in recent years, offering consistent prospects in terms of supporting EU security and defence objectives. To this end, practical projects on defence capabilities and research have been developed through EDTIB in line with the Common Security and Defence Policy agenda. An extremely important role is represented by the consolidation of the financial potential associated with EDTIB to support cooperation projects and initiatives. The use of EU budget resources, stimulated in the context of EDTIB, represents a strategic paradigm shift in which European cooperation has evolved. The results recorded so far indicate the viability of the approach, supported by Member States’ interest in deepening this trend, including by consolidating investment in defence and industrial purposes.

Dezvoltarea profilului UE în domeniul securității și apărării a reprezentat o constantă a ultimelor două decenii, fiind unul dintre proiectele cele mai dinamice ale integrării europene. Pe aceste coordonate, ultimii ani au consemnat progrese semnificative, ilustrate atât prin derularea unui număr semnificativ de operații civile și militare, în diferite perimetre geografice, cât și prin lansarea unor inițiative de cooperare în domeniul capabilităților de apărare, respectiv al cercetării în acest domeniu. Cadrul de raportare al acestor evoluții a cunoscut, de asemenea, schimbări majore, integrate în procesul de realizare a Bazei Industriale și Tehnologice de Apărare Europeană (EDTIB). După o etapă de structurare conceptuală derulată între anii 2007-2013, profilul acestui construct s-a maturizat accelerat în ultimii ani, oferind perspective consistente în ceea ce privește susținerea obiectivelor asumate de UE în domeniul securității și apărării. În acest sens, au fost elaborate proiecte practice privind capabilitățile și cercetarea în domeniul apărării, prin intermediul EDTIB, în acord cu agenda Politicii de Securitate și Apărare Comune. Un rol extrem de important îl reprezintă consolidarea potențialului financiar asociat EDTIB pentru susținerea proiectelor și inițiativelor de cooperare. Utilizarea resurselor bugetului Uniunii Europene, stimulată în contextul EDTIB, reprezintă o schimbare strategică de paradigmă în care cooperarea europeană a evoluat până acum. Rezultatele înregistrate până în prezent indică viabilitatea abordării, susținută prin interesul statelor membre de aprofundare a acestei tendințe, inclusiv prin consolidarea investițiilor în domeniul apărării și finalitate industrială.

Abstract:
ABSTRACT An understanding of the determinants of measles virus (MV) virulence has been hampered by the lack of an experimental model of infection. We have previously demonstrated that virulence phenotypes in human infections are faithfully reproduced by infection of human thymus/liver (thy/liv) implants engrafted into SCID mice, where the virus grows primarily in stromal cells but induces thymocyte apoptosis (P. G. Auwaerter et al., J. Virol. 70:3734–3740, 1996). To begin to elucidate the roles of the C protein, V protein, and the 5′ untranslated region of the F gene (F 5′UTR) in MV infection in vivo, the replication of strains bearing mutations of these genes was compared to that of the parent sequence-tagged Edmonston strain (EdTag). Growth curves show that mutants fall into two phenotypic classes. One class of mutants demonstrated kinetics of growth similar to that of EdTag, with decreased peak titers. The second class of mutants manifested peak titers similar to that of EdTag but had different replication kinetics. Abrogation of V expression led to delayed and markedly prolonged replication. Additionally, thymocyte survival was prolonged and implant architecture was preserved throughout the course of infection. In contrast, massive bystander thymocyte death occurred after infection with EdTag and all other mutants. A mutant which overexpressed V in Vero cells (V+) had the opposite phenotype of the A mutant not expressing V (V−). V+ grew more rapidly than EdTag with 100-fold-greater levels of virus production 3 days after infection. These results suggest that C, V, and the F 5′UTR are accessory factors required for efficient virus replication in vivo. In addition, thymocyte survival after V− infection suggests this protein may play multiple roles in pathogenesis of MV infection of thymus. Since these recombinant mutant viruses grew identically to the parent virus in Vero cells, the data show that thy/liv implants are an excellent model for investigating the determinants of MV virulence.

Educazione terapeutica e competenze tecniche educative specifiche per gli adulti hanno caratterizzato la formazione che EDTNA/ERCA ha proposto, non supportando la tesi della marginalità della metodica e delle competenze professionali che la sua pratica richiede. Proprio quest'ultimo aspetto può essere una delle criticità per il suo sviluppo.

Abstract:
ABSTRACT Molecular determinants of neuropathogenesis have been shown to be present in the hemagglutinin (H) protein of measles virus (MV). An H gene insertion vector has been generated from the Edmonston B vaccine full-length infectious clone of MV. Using this vector, it is possible to insert complete H open reading frames into the parental (Edtag) background. The H gene from a rodent brain-adapted MV strain (CAM/RB) was inserted into this vector, and a recombinant virus (EdtagCAMH) was rescued by using a modified vaccinia virus which expresses T7 RNA polymerase (MVA-T7). The recombinant virus grew at an equivalent rate and to similar titers as the CAM/RB and Edtag parental viruses. Neurovirulence was assayed in a mouse model for MV encephalitis. Viruses were injected intracerebrally into the right cortex of C57/BL/6 suckling mice. After infection mice inoculated with the CAM/RB strain developed hind limb paralysis and ataxia. Clinical symptoms were never observed with an equivalent dose of Edtag virus or in sham infections. Immunohistochemistry (IHC) was used to detect viral antigen in formalin-fixed brain sections. Measles antigen was observed in neurons and neuronal processes of the hippocampus, frontal, temporal, and olfactory cortices and neostriatum on both sides of symmetrical structures. Viral antigen was not detected in mice infected with Edtag virus. Mice infected with the recombinant virus, EdtagCAMH, became clinically ill, and virus was detected by IHC in regions of the brain similar to those in which it was detected in animals infected with CAM/RB. The EdtagCAMH infection had, however, progressed much less than the CAM/RB virus at 4 days postinfection. It therefore appears that additional determinants are encoded in other regions of the MV genome which are required for full neurovirulence equivalent to CAM/RB. Nevertheless, replacement of the H gene alone is sufficient to cause neuropathology.

Abstract:
IPEX (immunedysregulation, polyendocrinopathy, enteropathy, X-linked) syndrome is a severe congenital autoimmune disorder in males resulting from hemizygous inheritance of a mutant FOXP3 allele. FOXP3 encodes a transcription factor that governs the development, maintenance, and function of regulatory T cells (Treg). We have developed a cell therapy strategy for treatment of IPEX using a gene-editing approach in which CRISPR/Cas9 RNPs are co-delivered with an AAV6 donor template designed to integrate into the FOXP3 locus an expression cassette containing the MND promoter driving expression of a functional FOXP3 cDNA and a surface LNGFR tag linked by a 2A ribosomal skip peptide. This approach enforces heterologous FOXP3 expression in IPEX CD4 effector T cells (Teff), while simultaneously eliminating expression of the endogenous FOXP3 allele. The resultant high level and stable expression of functional FOXP3 converts Teff to Treg-like cells with immunosuppressive activity. Using an optimized protocol, we obtained efficient HDR rates across multiple healthy donors. Edited cells were consistently enriched to >95% purity by a magnetic LNGFR antibody selection and expanded 50-fold in a week. Expression of FOXP3 cDNA in edited cells was sufficient to enforce Treg-like phenotypes including the up-regulation of Treg-associated markers (CD25, CTLA-4, and ICOS), and down-regulation of CD127 and inflammatory cytokines (IL2, IFNgamma, TNFalpha). Importantly, we demonstrate sustained in vivo suppressive activity of these edited Treg-like cells (edTreg) in a xeno-GvHD mouse model. edTreg (as well as expanded natural Treg) limited effector T cell expansion and tissue infiltration and significantly protected mice from xeno-GvHD induced by co-transferred autologous effector T cells. Along with preliminary data showing successful editing in CD4 T cells from IPEX patients, our data provide key pre-clinical proof-of-concept and safety data supporting use of edTreg in a clinical trial for IPEX and, potentially, for use in other autoimmune diseases. Abstract:
This article introduces an extended data set of 760 terrorist groups that engaged in attacks during 1970 to 2016. Unlike most extant group data sets, the extended data on terrorist groups (EDTG) is not tied to terrorist groups and attacks listed in the RAND terrorism data; rather, EDTG is linked to terrorist groups and attacks given in the Global Terrorism Database. Terrorist groups’ variables in EDTG include ideology, main goals, start date, duration, base country, attack diversity, peak size, alternative endings (if relevant), and others. We display interesting features of EDTG through a series of tables and figures. Our EDTG-based survival analysis is at odds with some of the literature: for example, the demise of a leader and a larger share of transnational terrorist attacks increase the group’s odds of failure. After 2001, religious terrorist groups are more resilient than those with other ideologies. We also analyze terrorist group lethality and productivity.

Ein gutes, gesundes, erfolgreiches Jahr 2021 wünsche ich Ihnen allen! 2020 war das Jahr der Pflegenden und Hebammen – und was für eines. Aufmerksamkeit erregten unsere Berufsgruppen nicht durch sorgfältig geplante Aktionen. Vielmehr machten Bilder von uns in kompletter PSA (persönlicher Schutzausrüstung) und Schildern mit „Wir bleiben für euch da. Bleibt ihr zu Hause.“ die Medienrunde. In der Öffentlichkeit wurde für uns geklatscht und Versprechungen gemacht. Der Arbeitsalltag jedoch war geprägt von hohem Arbeitsaufkommen mit PSA-Engpässen, täglich neuen Erkenntnissen, Vorschriften und Hygieneplänen sowie vielen Unsicherheiten bei Patienten/-innen und Kollegen/-innen. Das Jahr hat uns viel abverlangt – und COVID-19 (Coronavirus Disease 2019) wird unseren Alltag noch eine Weile begleiten. Mit meinen guten Wünschen für das neue Jahr möchte ich Ihnen Helen Noble‘s Webinar über Achtsamkeit ans Herz legen. Es ist auf der EDTNA/ERCA-Homepage abrufbar.

<p>En el presente trabajo se muestra la forma en la que se evalúa la invarianza utilizando los modelos de análisis factorial confirmatorio para medias y covarianzas (AFC-MACS) para datos categóricos y los modelos de Teoría de Respuesta al Ítem (TRI). Se ejemplifica el análisis de la invarianza en el estudio de la Escala de Detección del Trastorno de Ansiedad Generalizada (EDTAG) comparando hombres y mujeres. La EDTAG es una escala ampliamente utilizada en las instituciones de salud y por sus características (escala breve de 12 ítems dicotómicos) cualquier error de medida puede tener un impacto importante. En los resultados se muestra que la escala tiene la misma configuración, sin embargo, el ítem 9 muestra funcionamiento diferencial siendo los hombres quienes mayor probabilidad tienen de responder de manera afirmativa comparado con mujeres del mismo nivel de rasgo. Se discute la necesidad de éste tipo de análisis en las escalas.</p>

In this paper, the statistical analysis determined a representative of the state of reverberation in theSerbian Orthodox Church of "St. Prokopius" in Katun. The first part of the paper explains the experiment based onthe process of recording acoustic impulse responses and determines the value of the early reverberation time EDTand the reverberation time RT10, RT20, and RT30 at the central frequencies fc = {125, 250, 500, 1000, 2000, 4000} Hzusing ARTA software package. The experiment results (tabular and graphical) obtained by statistical analysis usingthe Matlab software package are also presented. In the second part of the paper, the results of the experimentanalysis were carried out. By comparing them, it was concluded which acoustic parameter is representative of thestate of reverberation in the analyses of the Serbian Orthodox church.

In this study, we investigate the causal relationships between international tourism growth and regional economic expansion in China, and more importantly, disclose the factors determining the occurrence of these relationships. The empirical results reveal that 10 of 29 regions experienced tourism-led growth (TLG) during 1978 to 2013, whereas nine regions experienced economy-driven tourism growth (EDTG). Different from the past literature, this study uses Bayesian probit models to unveil the factors influencing these different growth patterns. Our results suggest that regions with less-developed economies, larger economic sizes, and covering larger geographic areas are more likely to experience TLG, and regions with less-developed economies are more likely to experience EDTG as well. Lastly, practical implications are provided.
