Abstract:
In this essay, Jon Wergin reminds readers of the philosophical and historical foundations of the doctor of education (EdD) degree. He argues that the EdD should be based, in large part, on John Dewey's progressive ideals of democratization and Paulo Freire's concepts of emancipatory education. Drawing on theories of reflective practice, participatory action research, and signature pedagogies, Wergin proposes five principles through which schools of education can "reboot" their EdD degrees, creating programs that represent continued scholarship into practice and a commitment to social action. He then illustrates one potential application of these principles.

Abstract:
The structural analysis of sulfated carbohydrates such as glycosaminoglycans (GAGs) has been a long-standing challenge for the field of mass spectrometry. The dissociation of sulfated carbohydrates by collisionally-activated dissociation (CAD) or infrared multiphoton dissociation (IRMPD), which activate ions via vibrational excitation, typically result in few cleavages and abundant SO3 loss for highly sulfated GAGs such as heparin and heparan sulfate, hampering efforts to determine sites of modification. The recent application of electron activation techniques, specifically electron capture dissociation (ECD) and electron detachment dissociation (EDD), provides a marked improvement for the mass spectrometry characterization of GAGs. In this work, we compare ECD, EDD and IRMPD for the dissociation of the highly sulfated carbohydrate sucrose octasulfate (SOS). Both positive and negative multiply-charged ions are investigated. ECD, EDD and IRMPD of SOS produce abundant and reproducible fragmentation. The product ions produced by ECD are quite different than those produced by IRMPD of SOS positive ions, suggesting different dissociation mechanisms as a result of electronic versus vibrational excitation. The product ions produced by EDD and IRMPD of SOS negative ions also differ from each other. Evidence for SO3 rearrangement exists in the negative ion IRMPD data, complicating the assignment of product ions.

Abstract:
The growth pattern of Pseudomonas putida KT2440 in the presence of glucose and phenylacetic acid (PAA), where the sugar is used in preference to the aromatic compound, suggests that there is carbon catabolite repression (CCR) of PAA metabolism by glucose or gluconate. Furthermore, CCR is regulated at the transcriptional level. However, this CCR phenomenon does not occur in PAA-amended minimal medium containing fructose, pyruvate or succinate. We previously identified 2-keto-3-deoxy-6-phosphogluconate (KDPG) as an inducer of glucose metabolism, and this has led to this investigation into the role of KDPG as a signal compound for CCR. Two mutant strains, the edd mutant (non-KDPG producer) and the eda mutant (KDPG overproducer), grew in the presence of PAA but not in the presence of glucose. The edd mutant utilized PAA even in the presence of glucose, indicating that CCR had been abolished. This observation has additional support from the finding that there is high phenylacetyl-CoA ligase activity in the edd mutant, even in the presence of glucose+PAA, but not in wild-type cells under the same conditions. Unlike the edd mutant, the eda mutant did not grow in the presence of glucose+PAA. Interestingly, there was no uptake and/or metabolism of PAA in the eda mutant cells under the same conditions. Targeted disruption of PaaX, a repressor of the PAA operon, had no effect on CCR of PAA metabolism in the presence of glucose, suggesting that there is another transcriptional repression system associated with the KDPG signal. This is the first study to demonstrate that KDPG is the true CCR signal of PAA metabolism in P. putida KT2440.

In recent times, aerospace, chemical industries and nuclear plant have usually used Inconel 718 alloy because of its excellent mechanical and chemical properties at elevated temperatures. It falls under the category of difficult-to-cut materials due to its high toughness, poor thermal conductivity and high hardness. The set-ups for electric discharge drilling (EDD) and powder-mixed electric discharge drilling (PM-EDD) were developed, and experiments were conducted on them separately. This research shows a comparative study amid producing holes by EDD and PM-EDD in Inconel 718 alloy workpiece with copper tool electrode. SiC was used as an abrasive powder because of its better thermal conductivity in order to get properly mixed with dielectric in a separate tank. Output response was assessed in the form of material removal rate, under the influence of discharge current, duty factor, pulse-on-time and tool speed, as the input parameters.

Abstract:
Professional Doctorates in Education (EdD) have been established in many universities in England over the last 25 years, but there is little empirical research on them. The studies that do exist are largely literature reviews comparing professional doctorates to traditional doctorates. The empirical work tends to be student-centred, dealing with issues such as motivation for undertaking a professional doctorate; how professional doctorates impact on professional identities; and, the type of critical skills required to engage successfully with a professional doctorate. There has been one empirical survey of EdDs, but this was only as part of a wider survey on the field of professional doctorates. What is missing is twofold: first a consideration of the landscape of EdD programmes; what is being offered where, and how might we understand the nature of the knowledge acquired. Secondly, there is no description of the pedagogical frameworks of EdDs and how these bridge the various knowledge boundaries between theoretical, academic and professional knowledges. This thesis sets out to investigate these issues. The study draws on the theoretical concepts of Basil Bernstein and, in part, Karl Maton’s development of these concepts. Maton’s extension of Bernstein’s horizontal and vertical discourse, the semantics modality of legitimation code theory, was used to situate the EdD programmes in relation to one another with regards to the type of knowledge transmitted and acquired by students engaged in those EdDs. This was done through a survey of the EdD websites, quantifying the discourse of those websites and relevant online documentation related to the programmes. Having mapped the programmes through this survey of publicly available material, three partial case studies were developed, each demonstrating a different knowledge focus and pedagogical approach. I used Bernstein’s concept of classification and framing to describe how the pedagogical process of the EdD might, or might not, weaken the boundaries between theoretical knowledge and professional knowledge thereby making it possible for students to connect the two. The findings showed that most EdD programmes emphasised context-independent knowledge and context-dependent knowledge in broadly equal measure. Diverse students were accommodated within structured curricula in communities of learners, albeit in slightly different ways, by providing a theoretical and pedagogical framework in which the students found commonalities and by drawing on the shared experiences of different professional practices to enrich students’ learning experience. Despite each programme having a different focus, all programmes allowed students to develop their own research focus relevant to their professional practice. The deeper description of the three partial case studies also indicated that although the programme focus was an important part of enabling students to find an EdD which best suited their needs, the pedagogic framework should carry equal weight with regards to facilitating students’ ability to connect theoretical knowledge to professional knowledge. For this reason, the pedagogic approach should also be explicated clearly as part of the online information. There has not been any recent mapping of professional doctorates such as EdDs in England. This research has developed a new approach to this problem that a) sheds light on EdDs in England and b) could be useful in mapping other professional doctorates and investigating knowledge acquisition and production in these disciplines.

Abstract:
Since first appearing in British universities during the early 1990s, the Doctor of Education (EdD) degree has spread rapidly through the UK higher education sector. However, despite the existence of a single set of Quality Assurance Agency (QAA) descriptors for doctoral level achievement, some in academia have always been willing to describe the EdD, either openly or in private, as inferior to the PhD. This thesis endeavours (through a wide-ranging questionnaire completed anonymously by 27 academics from a total of 16 English universities, and follow-up interviews with seven individuals selected from the original sample) to discover how a sample of those who teach on such programmes (EdD academics) view the EdD, in general terms. For instance, it seeks to ascertain how widespread among EdD academics is the notion that the EdD does not reach the ‘gold standard’ represented by the PhD in Education. As data collection proceeds from the questionnaire to the interviews, the focus narrows to three key topics: specific characteristics of the EdD as compared with the PhD in Education (in terms, for example, of learning experience, programme aims, and modes of assessment); strengths, weaknesses and purposes of the EdD viva voce examination; and the concept of 'originality' as operationalised by EdD academics/examiners in deciding whether or not an EdD candidate/thesis displays 'doctorateness'. A range of informed and sometimes forthright views from EdD academics on these matters is recorded. On the basis of data analysis and interpretation, the thesis concludes with proposals for further, more extensive research, and a call for one of two courses of action: either the abolition of the EdD, or the appointment of a committee to review EdD practices nationally, and to recommend ways of strengthening EdD rigour and reputation.

Abstract:
EDD (E3 isolated by differential display), located at chromosome 8q22.3, is the human homologue of the Drosophila melanogaster tumour suppressor gene 'hyperplastic discs'. Edd null mice and hyd mutants display embryonic lethality. EDD is also a multifunctional HECT family E3 ubiquitin protein-ligase, with reported roles in both progesterone action and the DNA damage response. To investigate the possible involvement of EDD in human cancer, several cancer types were analysed for allelic gain or loss (allelic imbalance, AI) at the EDD locus. AI of the EDD locus was most frequent in the serous subtype of ovarian cancer (16/22, 73%) and common in other cancers, including breast cancer (31%). AI is likely to represent amplification of the EDD gene locus rather than loss of heterozygosity, as quantitative RT-PCR and immunohistochemistry showed that EDD mRNA and protein are frequently overexpressed in breast and ovarian cancers. These data imply a potential role for EDD in cancer progression. However, depletion of EDD from cells in culture by RNA interference had very little effect on proliferation and cell survival. To identify EDD-regulated pathways, transcript analysis was performed on EDD-depleted cells. The results suggested that EDD modulates cell-cell communication and the actin cytoskeleton. Consistent with transcript analysis, depletion of EDD from two normal breast cell lines (HMEC-184 and MCF-10A) resulted in altered cell morphology, with decreased cell-cell contacts. This was concurrent with altered beta-catenin (an integral component of adherens junctions) at cell-cell contacts, which was also observed in the developing blood vessels of Edd null mice. Interestingly, total cellular beta-catenin levels were not affected. Furthermore, EDD depletion resulted in a decrease in expression of the cytoskeletal regulators twinfilin and R-RAS, with a simultaneous decrease in MAPK (ERK1 and ERK2) activity. Consistent with disruption of adherens junctions, EDD-depleted mammary acini lost tissue coordination and polarity. These data provide a significant advance in our knowledge of EDD, both in its role in regulating the organisation of cells into higher structures and its potential role in the development of cancer. This has relevance to an understanding of embryonic development and the role of tissue homeostasis in cancer progression.

An intact cellular response to DNA damage is important for the maintenance of genomic stability and tumour prevention. EDD, the human orthologue of Drosophila melanogaster ???hyperplastic discs???, is over-expressed or mutated in a number of common human cancers. EDD is a progestin regulated gene that encodes an E3 ubiquitin ligase involved in cell communication and cell adhesion, and although it has also been implicated in the DNA damage response through its association with DNA damage proteins, a definitive role has yet to be demonstrated. The work presented herein shows that EDD is necessary for an adequate cellular response to double-strand DNA breaks. Cells depleted of EDD exhibit reduced survival, radio-resistant DNA synthesis and failure to maintain G2/M arrest following DNA damage induced by phleomycin exposure. Furthermore, EDD-depleted cells display impaired activating phosphorylation and kinase activity of the checkpoint kinase CHK2 after DNA damage. These effects appear to be largely modulated through a phospho-dependent interaction involving the CHK2 FHA domain and a region of EDD spanning a number of putative FHA-binding threonines. These results identify EDD as a novel mediator in DNA damage signal transduction via CHK2 and emphasise the potential importance of EDD in cancer.

A professional doctorate in education degree (EdD) is a degree where students contextualize professional knowledge into a “theoretical academic framework” (Quality Assurance Agency, 2015, p. 8). This study’s context is a 100% online EdD programme in the United Kingdom (UK) comprised of students throughout the world. Because online EdD programmes are relatively new, limited knowledge exists about the online EdD phenomenon from the students’ perspectives. In effect, the student voice about the online EdD is nearly absent in the literature but should be considered. How to fulfil the primary aim of the programme, to interrelate theory and professional practice and evoke social change (Costley, 2014), also has received limited attention. Two main aims of this study are: (1) To investigate the impacts of the online EdD learning experiences and outcomes on the participants’ personal and professional development through a phenomenologically-informed lens; and (2) To advance the student voice about learning experiences in online EdD programmes. This thesis is phenomenologically-informed, which enabled an in-depth investigation and a “Textural-Structural Description” (Moustakas, 1994, p. 121). The methods to collect data were interviews and document collection, which offered response triangulation and further analysis. The study results indicated that the participants experienced emotional and social challenges (e.g., cultural and linguistic disconnections and incremental disorienting dilemmas) that the transnational higher education (TNHE) virtual learning environment (VLE) sometimes exacerbated. Five of the eight participants’ stories contained evidence of each transformative learning stage (explored further in section 4.6) (Mezirow, 1991). The majority of the participants applied new knowledge from the online EdD programme to their professional contexts to effectuate social change, which is discussed in Chapters IV-V. This study draws attention to the online EdD phenomenon and furthers the discourse about the impacts of the online EdD on students’ personal and professional development.

The purpose of this study was to examine social physique anxiety (SPA) and disordered eating in female aesthetic athletes. Participants included 144 female competitive athletes (M = 19.0 years, SD = 2.08) from the four aesthetic sports of synchronized swimming, figure skating, jazz dance, and gymnastics. The participants completed the SPA scale (SPAS) and the Questionnaire for Eating Disorder Diagnoses (Q-EDD) (which is a new more accurate measure to assess disordered eating) that has not previously been used with female aesthetic athletes. Results revealed that 59.7% of the participants were classified in the asymptomatic category, 29.9% in the symptomatic category, and 10.4% in the eating disordered category. For the two Q-EDD subcategories, there were no participants classified in the chew/spitting category, and three participants classified in the exercise bulimia nervosa category. Also, it was shown that the eating disordered group and the symptomatic group had significantly higher SPA than the asymptomatic group. Thus, these results indicate that in this subpopulation of athletes, there are many who display eating disorder symptomatology, and suggest that interventions that target SPA and disordered eating in these athletes are needed.

Abstract:

A complex molecular network is put into place at specific phases of the cell cycle to prevent unscheduled cell division that could result in malignant cell growth. Emerging evidence shows that still uncharacterized proteins play crucial functions at those cell cycle transition points. Nuclear protein Msp58 and EDD E3 ubiquitin ligase have been implicated in different aspects of cell proliferation and reported to be abnormally expressed in numerous types of cancers. The molecular mechanisms underlying Msp58 and EDD functions, however, are not well understood. The work presented here shows that Msp58 and EDD form a stable protein complex that regulates cell viability and proliferation. Interestingly, knockdown of EDD by RNA interference leads to a significant accumulation of Msp58 protein, which suggests that EDD serves as a negative regulator of Msp58. In addition, our in vivo ubiquitination assays and analyses of various cell lines treated with translational and proteasomal inhibitors demonstrate that Msp58 is regulated post-translationally by the ubiquitin-proteasome pathway. These results imply that EDD ligase activity is involved in this regulatory process. Using flow cytometry analyses and biochemical characterization of Msp58 and/or EDD depleted cells, we show that the Msp58-EDD complex plays important roles in cell cycle progression via the control of cyclin gene expression. In particular, silencing Msp58 and/or EDD alters the protein levels of cyclins B, D and E. Taken together, our data suggest that a set of the biological roles attributed to Msp58 and EDD may be executed in the context of the complex that they form, thereby revealing a novel molecular mechanism for these two proteins to accomplish their functions.

Abstract:
The demand for multifunctional micro parts is an ever increasing need for product miniaturization. Amongst the spectrum of micro manufacturing processes, Electro-Discharge Deposition (EDD) is a newly developing additive process to produce parts in micro scale. In EDD the tool and workpiece electrodes are connected to reverse polarity in order to remove material from anode which deposits on cathode surface. The advantage of EDD is, any conducting tool material can be deposited irrespective of its hardness on the specified conducting substrate. So far, limited work has been carried out to develop micro parts like micro cylinders, micro spiral structures, etc., by EDD. In this work an attempt has been made to study the effect of various process parameters like current, duty cycle, pulse on time, voltage and table feed rate in EDD using central composite rotatable design (CCRD). It is found from the experimental results that, current plays a significant role in deposition of tool material.

Abstract:
In this work, investigation into a two-stage hybrid and integrated incremental sheet metal processing of EDD sheet has been carried out. EDD steel was chosen for its excellent formability. The hybrid process consists of two stages, the first stage is the conventional spinning process and the second stage is the asymmetric incremental sheet metal forming (AISF) process. The objectives of the proposed hybrid process are reduction of manufacturing time and improved surface finish, in comparison to AISF process alone. In the proposed hybrid process, the geometry of sheet metal part was sub-divided into two groups of features. The first group of axisymmetric features was obtained by spinning process, on top of which, in the second stage, the second group of asymmetric features was superimposed by AISF process. The hybrid process for studying the manufacturing time consisted of negative AISF in the second stage. A substantial overall reduction in manufacturing time was observed. The hybrid process for studying improved surface finish consisted of, on the other hand, positive AISF in the second stage, in which the experiments were carried out using four different lubricants, namely plain canola oil and three different additives in canola oil, namely, molybdenum disulphide, boric acid and maleic anhydride. The surface topography of the sheet in the two stages of hybrid process was determined using a profilometer. It is found that canola oil with molybdenum disulphide gives the best surface quality.
