Abstract:
e11513 Background: Human epidermal growth factor receptors and their ligands are involved in carcinogenesis. Four ERBB receptors have been identified; ERBB1 (EB1), ERBB2 (EB2), ERBB3 (EB3) and ERBB4 (EB4). EB2 is over-expressed/amplified in 20-30% of BCa. Molecularly targeting agents, such as pertuzumab (P), in combination with standard therapy have shown important clinical results in EB2 (+) BCa patients. The aim of the project is to evaluate the expression/localization and dimerization pattern of EB1, EB2, EB3, EB4 in breast cancer cell lines (BCCLs) with or without heregulin-1 (He) and various EB2, EB3 and He inhibitors. The identification of a molecular phenotype of low-expressing and acquired resistant high-expressing EB2 BCa will enhance the targeted use of EB2 inhibitors in certain BCa patients Methods: Using confocal microscopy, localization/expression patterns were studied for EB1, EB2, EB3, EB4 in various human BCCs; here presented in MCF7 and SKBR3. Additionally, the dimerization pattern of EB2, EB3 and/or EB4 was studied with and without stimulation with He plus or minus P (kindly provided by Genentech, US). The dimers formation was studied using proximity ligation assay and immunofluorescence (DUOLINK; OLINK Biosciences, Sweden). Results: In MCF7-He stimulated cells EB2/EB3, EB2/EB4 and EB1/EB3 dimmers showed a 3-fold decrease formation compared to untreated cells. The EB1/EB4 dimmers were not significantly affected. After P addition in MCF7 cells a 3-fold decrease of dimmer formation was noticed compared to untreated cells for EB2/EB3, EB2/EB4 and EB1/EB3. The EB1/EB4 formation seemed only slightly affected by P. SKBR3-He stimulated cells resulted in a similar decrease of EB2/EB3, EB2/EB4 and EB1/EB3 dimmer formation compared to untreated cells. In the same BCLL, EB1/EB4 dimmer did not show any decrease after He treatment compared to untreated control cells. Further results quantified and analyzed for statistical significance will be shown Conclusions: The addition of P seems to block ligand-dependent heterodimerization of EB2 with other ERBB family members, such as EB3 and EB4.

Abstract:
Buah Andaliman (Zanthoxylum acanthopodium) dan Kecombrang (Eltingera elatior) merupakan jenis tanaman rempah lokal yang tergolong kedalam keanekaragaman sumber daya alam di Sumatera Utara. Kedua jenis tanaman rempah ini memiliki kandungan ekstrak heksana, methanol, dan asetat yang berfungsi sebagai senyawa antimikrobial. Pemanfaatan telur asin belum dilakukan secara optimal melalui penggunaan rempah buah andaliman dan kecombrang. Kualitas telur asin yang rendah dapat dipengaruhi oleh aktifitas mikroba yang dapat merusak kualitas protein dalam telur. Sebanyak 60 butir telur itik segar digunakan dalam penelitian. Rancangan yang digunakan adalah Rancangan Acak Lengkap (RAL) dengan 4 perlakuan dan 5 ulangan. Perlakuan yang digunakan meliputi P1 (Kontrol), P2 (telur diasinkan dengan 25% EBA + 25% EBK), P3 (telur diasinkan dengan 50% EBA + 50% EBK) dan P4 (telur diasinkan dengan 75% EBA + 75% EBK). Ekstrak tanaman rempah yang digunakan merupakan ekstrak kasar melalui teknik penyaringan konvensional yang disebut ekstrak kasar buah andaliman (EBA) dan kecombrang (EBK). Hasil penelitian menunjukkan bahwa peningkatan level tanaman rempah menurunkan total populasi bakteri E. Coli sampai 1.5 x 109 cfu/g dan meningkatkan populasi bakteri Bacillus sp sampai 0.1 x 108 cfu/g. Dari hasil penelitian disimpulkan bahwa pemanfaatan ekstrak kasar buah andaliman dan kecombrang dapat memperbaiki kualitas telur asin.

Penelitian bertujuan untuk mengkaji pengaruh pemberian prebiotik soybean oligosakarida (SOS) dari ekstrak bungkil kedelai (EBK) dan ekstrak kulit kedelai (EKK) terhadap massa lemak daging, persentase lemak abdominal, dan bobot daging pada ayam broiler. Materi penelitian adalah 160 ekor ayam broiler strain Lohman unsex yang dipelihara selama 6 minggu. Penelitian menggunakan rancangan acak lengkap (RAL) dengan 5 perlakuan dan 4 ulangan, setiap ulangan menggunakan 8 ekor ayam broiler. Ransum perlakuan yang diberikan, yaitu T0 (ransum basal/RB), T1(RB+0,15% EBK), T2 (RB+0,3% EBK), T3 (RB+0,15% EKK), T4 (RB+0,3% EKK). Data hasil penelitian dianalisis ragam dan dilanjutkan dengan duncan multiple range test. Hasil penelitian menunjukkan bahwa pemberian prebiotik SOS dari EBK dan EKK pada semua level berpengaruh nyata (P<0,05) terhadap persentase lemak abdominal dan bobot daging ayam broiler, namun tidak berpengaruh nyata (P>0,05) terhadap massa lemak daging.

Penelitian menggunakan rancangan acak lengkap dengan 5 perlakuan dan 4 ulangan (masing-masing 8 ekor ayam). Perlakuan terdiri dari : T0 (ransum basal), T1 (ransum basal+0,15% EBK), T2 (ransum basal+0,3% EBK), T3 (ransum basal+0,15% EKK), T4 (ransum basal+0,3% EKK). Data dianalisis ragam dan dilanjutkan dengan uji wilayah ganda Duncan. Hasil penelitian menunjukkan bahwa pemberian soybean oligosakarida dari EBK dan EKK nyata (P<0,05) meningkatkan bobot karkas, namun tidak berpengaruh nyata terhadap kandungan protein dan REP. Simpulan penelitian adalah pemberian soybean oligosakarida dari ekstrak bungkil dan kulit kedelai sampai level 0,3% dapat meningkatkan bobot karkas ayam broiler.

This article explores the various dimensions of early recruitment activities (ERAs) such as publicity, sponsorship, word of mouth and advertisement, and its impact on employer brand knowledge (EBK) such as employer familiarity, employer image or job association and employer reputation. It further explores the impact of ERAs and EBK on organization attractiveness (OA) and firm performance (FP). The study shows that advertisement, publicity and word of mouth of ERAs impact all aspects of EBK such as employer familiarity, employer image or job association, and employer reputation. Employer reputation and job association are significant for most organization and does impact OA, while brand awareness and job association impact FP. The prominent sources of employment information were the Internet and networking.

With the increasing application of global navigation satellite system (GNSS) technology in the field of meteorology, satellite-derived zenith tropospheric delay (ZTD) and precipitable water vapor (PWV) data have been used to explore the spatial coverage pattern of PM2.5 concentrations. In this study, the PM2.5 concentration data obtained from 340 PM2.5 ground stations in south-central China were used to analyze the variation patterns of PM2.5 in south-central China at different time periods, and six PM2.5 interpolation models were developed in the region. The spatial and temporal PM2.5 variation patterns in central and southern China were analyzed from the perspectives of time series variations and spatial distribution characteristics, and six types of interpolation models were established in central and southern China. (1) Through correlation analysis, and exploratory regression and geographical detector methods, the correlation analysis of PM2.5-related variables showed that the GNSS-derived PWV and ZTD were negatively correlated with PM2.5, and that their significances and contributions to the spatial analysis were good. (2) Three types of suitable variable combinations were selected for modeling through a collinearity diagnosis, and six types of models (geographically weighted regression (GWR), geographically weighted regression kriging (GWRK), geographically weighted regression—empirical bayesian kriging (GWR-EBK), multiscale geographically weighted regression (MGWR), multiscale geographically weighted regression kriging (MGWRK), and multiscale geographically weighted regression—empirical bayesian kriging (MGWR-EBK)) were constructed. The overall R2 of the GWR-EBK model construction was the best (annual: 0.962, winter: 0.966, spring: 0.926, summer: 0.873, and autumn: 0.908), and the interpolation accuracy of the GWR-EBK model constructed by inputting ZTD was the best overall, with an average RMSE of 3.22 μg/m3 recorded, while the GWR-EBK model constructed by inputting PWV had the highest interpolation accuracy in winter, with an RMSE of 4.5 μg/m3 recorded; these values were 2.17% and 4.26% higher than the RMSE values of the other two types of models (ZTD and temperature) in winter, respectively. (3) The introduction of the empirical Bayesian kriging method to interpolate the residuals of the models (GWR and MGWR) and to then correct the original interpolation results of the models was the most effective, and the accuracy improvement percentage was better than that of the ordinary kriging method. The average improvement ratios of the GWRK and GWR-EBK models compared with that of the GWR model were 5.04% and 14.74%, respectively, and the average improvement ratios of the MGWRK and MGWR-EBK models compared with that of the MGWR model were 2.79% and 12.66%, respectively. (4) Elevation intervals and provinces were classified, and the influence of the elevation and the spatial distribution of the plane on the accuracy of the PM2.5 regional model was discussed. The experiments showed that the accuracy of the constructed regional model decreased as the elevation increased. The accuracies of the models in representing Henan, Hubei and Hunan provinces were lower than those of the models in representing Guangdong and Guangxi provinces.

Latar belakang : Oksidan yang terkandung dalam asap rokok dapat menyebabkan terjadinya stres oksidatif. Buah kersen (Muntingia calabura) mengandung antioksidan diantaranya flavonoid. Penelitian ini bertujuan mengetahui efek proteksi ekstrak buah kersen terhadap kerusakan histologis alveolus paru tikus Sprague dawley yang terpapar asap rokokMetode : Tikus sejumlah dibagi 4 kelompok, kemudian di papar asap rokok 7 batang perhari dan ekstrak buah kersen (EBK) selama 4 minggu. Kelompok pertama sebagai kontrol (K1) diberi plasebo, kelompok kedua (K2) dipapar asap rokok saja, sedangkan sisanya merupakankelompok Perlakuan 1 (P1) diberi EBK 100mg/kgBB/hari dan Perlakuan 2 (P2) diberi EBK 200mg/kgBB/hari. Perbedaan derajat kerusakan alveolus di analisis dengan Kruskal-Wallis Test.Hasil : Rerata derajat kerusakan alveolus pada kelompok kontrol (K1)sejumlah (69,44) kelompok perlakuan 1 (P1) sejumlah (80,00) dan kelompok perlakuan 2 (P2) sejumlah (79,63) sedangkan yang terendah yaitu pada kelompok kontrol paparan asap rokok (K2) sejumlah (61,11). Nilai P = 0.464, menunjukkan bahwa tidak terdapat perbedaaan yangbermakna derajat kerusakan alveolus antar kelompok, dimana rerata derajat kerusakan alveolus paru > 60% (kerusakan berat).Simpulan : Pemberian ekstrak buah kersen tidak berpengaruh terhadap gambaran mikroskopis paru tikus yang di papar asap rokok. Ternak yang digunakan dalam penelitian adalah ayam broiler strain Lohman sebanyak 160 ekor umur 7 hari dengan bobot badan awal 112, 24 ± 6,70 g. Penelitian 48 Pemanfaatan Soybean Oligosaccharides dari Bungkil dan Kulit Kedelai Terhadap Ketahanan TubuhAyam Broiler menggunakan rancangan acak lengkap dengan 5 perlakuan dan 4 ulangan. Perlakuan menggunakan soybean oligosaccharides dalam bentuk ekstrak dari bungkil kedelai(EBK) dan ekstrak kulit kedelai(EKK) meliputi T0 (kontrol), T1 (EBK 0,15%), T2 (EBK 0,3%), T3 (EKK 0,15%), dan T4 (EKK 0,3%). Parameter yang diukur meliputi bobot relatif bursa fabricius, bobot relatif limpa, nilai rasio heterofil limfosit, dan pertambahan bobot badan. Data dianalisis varians dan dilanjutkan dengan uji Wilayah Ganda Duncan. Hasil penelitianmenunjukan bahwa penambahan soybean oligosaccharide dalam bentuk ekstrak kulit kedelai maupun bungkil kedelai tidak berpengaruh nyata terhadap bobot relatif organ limfoid, tetapi berpengaruh nyata (P<0,05) terhadap rasio heterofil limfosit dan pertambahan bobot badan ayam broiler. Berdasarkan hasil penelitian dapat disimpulkan bahwa pemanfaatan soybean oligosaccharides dari bungkil kedelai maupun kulit kedelai mampu mempertahankan kondisi kesehatan dilihat dari bobot organ limfoid yang normal dan menurunkan rasio heterofil limfosit. Soybean oligosaccharides dari bungkil kedelai taraf 0,3% menghasilkan pertambahan bobot badan yang terpenuhi.

Abstract:
Latar belakang : Reactive Oxygen Species (ROS) merupakan salah satu mediator terjadinya infertilitas. Produksi ROS akan meningkat dengan adanya polusi udara dan merokok. Polynuclear Aromatic Hydrogen (PAH) yang terkandung dalam asap rokok dapat menyebabkan atrofi testis, menghambat spermatogenesis, dan merusak morfologi spermatozoa. Apabila terjadi kerusakan atau atrofi sel-sel penyusun tubulus seminiferus akan terjadi penurunan berat testis. Buah kersen mengandung senyawa antioksidan yang dapat meningkatkan kualitas sperma. Tujuan penelitian ini adalah untuk mengetahui efek proteksi ekstrak buah kersen terhadap kerusakan testis tikus Sprague dawley yang terpapar asap rokok.Metode : Tikus sejumlah dibagi 4 kelompok, kemudian di papar asap rokok 7 batang perhari dan ekstrak buah kersen (EBK) selama 4 minggu. Kelompok pertama sebagai kontrol (K1) diberi plasebo, kelompok kedua (K2) dipapar asap rokok saja, sedangkan sisanya merupakan kelompok Perlakuan 1 (P1) diberi EBK 100mg/kgBB/hari dan Perlakuan 2 (P2) diberi EBK 200mg/kgBB/hari. Rasio berat testis di analisis dengan uji statistik parametrik One Way ANOVA Test dilanjutkan analisa Post Hoc Multiple Comparison Test.Hasil : Rasio berat testis pada kelompok kontrol (K1) sejumlah (0,31) kelompok perlakuan 1 (P1) sejumlah (0,24) dan kelompok perlakuan 2 (P2) sejumlah (0,28) sedangkan yang terendah yaitu pada kelompok kontrol paparan asap rokok (K2) sejumlah (0,19). Nilai P = 0,013, menunjukkan bahwa terdapat perbedaaan yang bermakna berat testis antar kelompok.Simpulan : Pemberian ekstrak buah kersen dapat meningkatkan rasio berat testis pada tikus yang di papar asap rokok.

Basin-based water management strategy is one of the necessary instruments for the protection and sustainable use of water resources against climate change. In this paper, the monthly precipitation distributions of the 25 major basins in Türkiye were produced, and amounts and volumes were computed and analyzed. Only annual modeling and assessments of precipitation may hide months with precipitation shortages. Empirical Bayesian Kriging (EBK), Ordinary Kriging (OK), and Inverse Distance Weighting (IDW) were implemented in interpolation. EBK outperformed in all months and calculations were based on the EBK. The month with the highest precipitation potential in Türkiye is December (77.9 mm, 60.77 billion m3), and the month with the lowest precipitation potential is August (13.8 mm, 10.76 billion m3). In the basins, the monthly precipitation amounts range between 2.7 mm and 185.2 mm, and the volumes range between 0.02 billion m3 and 13.24 billion m3. The basins with the highest precipitation depth were determined as the East Black Sea, Antalya, Asi, and Ceyhan, and the lowest as the Small Menderes, Konya, and Tigris-Euphrates in different months. The monthly precipitation patterns and potentials of the basins vary widely. In May, June, July, August, and September, when water, particularly agricultural irrigation, is required the most, the 20 basins, except for the 5 located in Northern Türkiye precipitation shortage was determined.

The efficiencies of raw and modified kaolinite mineral in removing selected heavy metal ions from their respective aqueous solutions were investigated. The mineral was modified through two different methods; i) activation with HNO3, H2SO4, H3PO4, CH3COOH and C2H2O4 acids to form NK, SK, PK, AK and OK acid activated clays respectively and ii) preparations of 3:1 and 1:1 Kaolinite: Bentonite blends to form UBK and EBK composites respectively through manual blending. The adsorbents were characterized by X-ray Diffraction (XRD), Scanning Electron Microscopy (SEM), Fourier Transform Infra-Red Spectroscopy (FTIR) and Brunauer Emmett Teller (BET) analysis for surface area determination. The surface area increased in some of the modified clays from 114.9457 m2/g (RK) to 288.685 m2/g (EBK), 205.92 m2/g (UBK), 162.227 m2/g (NK), 151.335 m2/g (SK), and 115.837 m2/g (OK) but reduced to 113.872 m2/g (PK) and 112.865 m2/g (AK) after modification. Adsorption studies were subsequently conducted out to remove Pb2+, Cd2+ and Ni2+ ions from synthetic solutions. Pb2+ was found to be most removed (383.5 mg g-1 (RK), 591.13 mg g-1 (EBK), 576.61 mg g-1 (UBK), 475 mg g-1 (NK), 450 mg g-1 (SK), and 425 mg g-1 (PK), 375 mg g-1 (OK) and 375 mg g-1 (AK)) with highest removals on the composites.

Accurately estimating forest aboveground biomass (AGB) is imperative for comprehending carbon cycling, calculating carbon budgets, and formulating sustainable forest management plans. Currently, random forest (RF) and other machine learning models are widely used to estimate forest AGB, as they can effectively handle nonlinear relationships. However, by constructing a global model using all the samples collected from a study area, these models fail to account for the spatial heterogeneity in the AGB and cannot correct the prediction biases, thereby constraining the estimation accuracy. To overcome these limitations, we proposed a novel approach termed geographical random forest and empirical Bayesian kriging (GRFEBK). This hybrid model combines the localized modeling capability of geographical random forest (GRF) with the bias correction strength of empirical Bayesian kriging (EBK). GRF adapts RF to account for the spatial heterogeneity of the AGB, while EBK utilizes the spatial autocorrelation of residuals to correct the prediction deviations. This study was conducted in Hainan Island, utilizing spectral bands, vegetation indices, tasseled cap components derived from Landsat-8 imagery, backscattering coefficients from ALOS-2 synthetic aperture radar, topographic features, and the forest canopy height as the explanatory variables. A total of 195 forest aboveground biomass (AGB) samples were collected for modeling and assessing the predictive accuracy. The results demonstrate that, among the tested models, including GRFEBK, RF, support vector machine (SVM), k-nearest neighbor (KNN), geographically weighted regression (GWR), GRF, and EBK, GRFEBK attains the highest R2 (0.78) and the lowest RMSE (36.04 Mg/ha) and RRMSE (22.87%), significantly outperforming the conventional models and using GRF or EBK alone. These results demonstrate that by accounting for local non-stationarity in AGB and correcting prediction biases, GRFEBK achieves significantly higher accuracy than conventional RF and other models. While the results are promising, the computational cost of GRFEBK and its performance under varying geographical conditions warrant further investigation at larger scales to assess its broader applicability. Nevertheless, GRFEBK provides an innovative and more reliable approach for accurate forest AGB estimation with great potential to support global forest resource monitoring.

Abstract In grey models, GM(1, 1) is an important prediction model. The grey model GM(1, 1) has good prediction results in the case original data change exponentially at a low speed. However in practical cases sometimes, original data show exponential changes or approximately exponential changes change at a high speed. In these cases, the grey model GM(1, 1) has poor prediction results because the data fail to meet the laws of traditional model. Therefore, the paper proposes an extended grey model GM(1, 1, ebk) and its modeling method. In the final section, the paper builds grey models of GM(1, 1, ebk) for a practical problem and the results show the grey model proposed has greatly improved simulation and prediction accuracy compared with the traditional model.
