Abstract:
The goal of this thesis is to design an attachment between a duct and a pod. A pod is what could be compared to an outboard engine for larger vessels. The pod is placed on the outside of the ship’s hull and can rotate 360 ̊. A duct is a large steel ring that can be placed around the propeller on the pod to increasethe propulsion force of the ship in lower velocities. The thesis is written for Rolls Royce AB in Kristinehamn.The demanded requirements of the design are: it must be easy to adapt to different sizes and shapes of pod houses, the duct must be able to bemounted and dismounted from the pod.The result from this thesis is that anupper and a lower attachment weredesigned. The upper attachment consists of two parallel steel plates which arebolted together with a screw plate that is welded to the pod house.The lower attachment is basically a wing with a streamlined profile which is bolted to a stay. Both attachments consist of bolted joints, two guide pins and bursting screws.The design is controlled by parameters in CAD, meaning that if you change the size of the duct the attachments automatically adapts themselves to the new dimensions.The total result from this thesis is a design suggestion and not an actual manufactured product.The design is dimensioned by a load analysis and strength calculations. The dimensions of the bolts are determined by basic load calculation and the guide pins aredimensioned to withstand the shear forces that occur.The conclusion of this thesis is that the developed attachment solution is a good way to solve this problem andthe personal recommendation is that it should be developed further.Further design work that needs to be done with the attachment is a more thorough analysis of the strength and especially with fatigue, since no account has been taken considering fatigue.
Målet med detta arbete är att konstruera en infästning mellan en dysa och en pod. En pod är vad som kan liknasen utombordsmotor för stora fartyg som sitter på utsidan av skrovet och kan rotera 360 ̊. En dysa är en stor stålring som kan sättas runt propellern, dysan ger förenklaten ökad framdrivningskraft och är lämplig för användning vid fartygstillämpningar där det handlar om låga hastigheter.Arbetet utförs på uppdrag av Rolls Royce AB i Kristinehamn.Kraven som ställdes på konstruktionen var att den skulle vara lätt att anpassa och att det skulle gå att montera och demontera dysan.Problemformuleringen för arbetet är att en ny infästning behövs för att kunna fästa dysan i poden.Resultatet av arbetet blev en övre och en nedre infästning på dysan. Den övre infästningen består av två parallella plåtar som skruvas ihop med en svetsad infästning på dysan. Den nedre infästningen består av en vinge med en strömningsvänlig profil som skruvas fast på ett stag. Båda infästningarna består av ett skruvförband, två styrpinnar och sprängskruv.Konstruktionen parametriserades i CAD så attomdysans storlekändrasså följer infästningarna med.Hela arbetet resulterade i ett konstruktionsförslag.Konstruktionen har dimensionerats med hjälp av en lastanalys och hållfasthetsberäkningarför att uppskatta infästningarnas nedböjning. Skruvförbanden har dimensionerats med enklare beräkningar och styrpinnarna har dimensioneratför att uppta skjuvspänningar.Slutsatsen som dras av arbetet är att konstruktionen är en lämplig lösning på problemet och rekommendationen är att det är värt att undersöka denna lösning vidare.Vidarearbete som behöver utföras är nogrannare dimensionering av konstruktionen, ingen hänsynhar tagits till utmattning eller vridning då det inte fanns tid inom arbetets tidsram för detta.

Abstract:
Thermal fatigue is a major problem in various mechanical constructions. Details with strict boundary conditions subjected to cyclic thermal loadings experience significant internal stresses. Due to the capability of materials to expand or contract differently, components in separate materials, assembled, behave in their own ways. The cyclic thermal loads result in strains that cause stresses in the exposed materials. The magnitude of the resulted stresses is commonly dependent on the thermal loads and the material properties, and even if the magnitude of the stresses is too low to cause an immediate failure of the material, these stresses applied repeatedly will eventually result in localized plastic deformation. In the heavily deformed locations, stresses might reach the ultimate tensile strength of the material and thus initiate a fatigue crack. Repeated exposure to stresses will lead to crack growth and failure of components. In the present work a stress computational model in Abaqus was built up to calculate the stresses generated when components are subjected to thermal loads. The stress analysis was performed on two different spray nozzles attached to a steam pipe. The nozzles work under cyclic conditions, they start at a rather high temperature which decreases while cold water is passed through. This occurrence is repeated since the water flow runs sporadically. Cold water runs only when the temperature is too high, it is applied to regulate the temperature inside the steam pipe. The nozzle covers are the main differences between them, one is called a Pressured seal nozzle while the other nozzle is sealed with bolts. The results of the stress analysis showed that the highest stresses were found in the welding between the nozzle and the steam pipe. The Pressured sealnozzle’s stresses were more uniformly distributed around the welding compared to the nozzle with the bolts, which experienced stresses a little more varying in its welding. Worth to remark, the nozzle with bolts had one smaller area in the welding where the stresses were significantly higher than what appeared to be the average value of the stress in the welding of the Pressured seal nozzle. These results indicate that the Pressured seal nozzle’s welding should experience a more uniform fatigue while the other nozzle might get larger fatigue symptoms in a smaller area, while the rest of the part is not so heavily stressed.

Presented here is research investigating genes that are involved in the development and maintenance of ciliated nerve endings in the nematode Caenorhabditis elegans. C. elegans utilizes a subset of neurons, referred to as ciliated sensory neurons, to sense certain changes in its environment. There are two amphid sensilla (sense organs) that mediate exposure of these ciliated endings to the animal's external environment. Those ciliated endings that penetrate the cuticle are responsible for a myriad of behaviors that range from chemotaxis to osmotic avoidance, but in general function for the reception of environmental cues and stimuli. The intraflagellar transport (IFT) process facilitates the morphogenesis of these ciliated endings, and animals lacking intact ciliated endings may not be able to detect nourishment, hazardous environments, or other worms for mating. Mutant strains used in this study were generated by EMS mutagenesis of wild-type N2 animals and a subsequent screen of those worms displaying significant cilia dysfunction as evidenced by their dye-filling defective (Dyf) phenotype. Cilia-mediated uptake of lipophilic DiI into six pairs of amphid sensory neurons and two pairs of phasmid sensory neurons is expected in wild-type (N2) animals, but in Dyf animals, this dye-filling is disrupted, either through morphological defects, or deleterious mutations in the IFT process. To investigate the morphogenesis of cilia in C. elegans, we analyzed two specific mutant strains, WX737 dyf-3(og022)IV and PK841 dyf-15(pk841)V, that are defective in the uptake of fluorescent dye DiI and abnormal in sensory cilium structure. Through a variety of genetic mapping techniques, we were able to successfully map experimental gene dyf-15(pk841) to an interval of 2.84cM on chromosome V, and identify og022 as an allele of the gene dyf-3. It has been previously shown that dyf-3 expression is detected in 26 chemosensory neurons, including six IL2 neurons, eight pairs of amphid neurons (ASE, ADF, ASG, ASH, ASI, ASJ, ASK and ADL) and two pairs of phasmid neurons (PHA and PHB). Analysis of cilium malformation and the presence of a recognition sequence for the DAF-19 transcription factor suggest that dyf-3 is involved in the intraflagellar transport system complex B

Abstract:
This work presents an error as a literary practice in contemporary prose. The error is understood here as crossing the literary rules and a change that comes from genre deformation showed in analyzed texts. The functioning of the error is depicted via the examples of works by Dorota Masłowska, Jerzy Pilch, Andrzej Stasiuk and Jacek Dehnel. Furthermore I examine the term - error - s a literary practice using foreign articles and research on deception and error from all over the world. In the introductory chapters, I concentrate on the theoretical thesis of the substance of the error. I try to determine the semantics of the main term of this dissertation basing on explored aberrations in composition and genres. I do not attempt to generate any catalogue of norms or to analyze their transgressions. The genre matters are not an issue here. Taking into consideration the norms, however, I notice crossing the rules, yet I do not assume it comes solely from the convention. For me it is the result of the contemporary qualities of writing. The novels mentioned (and novels in general) are the mirror of their times and as such they become metatextual comment to the changes in literature. The immense part of my work is dedicated to the presentation of the difference between being mistaken and misleading somebody. I associate being mistaken with the motive of a labyrinth. Trying to go through it, we often meet blind spots that are the effect of being mistaken. If we apply a certain procedure, we can either go back or go forward breaking the order and the norms. When it comes to misleading, we can notice a peculiar stimulation of the matter of the text and subjective correlation of components. Taking that into account, misleading is presented through the selectivity of description and the special function of representation. Moreover, I analyze the matter of a novel and its status in the contemporary literature. I apply my findings to examine the latest Polish literature. I set together the critical diagnosis and the characteristics of the book market. I consider the phenomena composed of transgressing the rules, for example mosaic character of prose, the end of the great narratives, diversity of text, modifications of realism and innovation, as a normative state for the error. I show such practices as the origin of the new genders in literature. To sum up, my work is an attempt to prove that error as a literary practice is not a new phenomenon – it only requires systematization and theoretical discussion.

Nous nous représentons le cerveau comme un réseau complexe de régions structurellement connectées et fonctionnellement couplées. Les fonctions cognitives découlent de l'activité coordonnée de régions corticales distantes. La connectivité est utilisée pour représenter la coopération de régions cérébrales ségréguées et fonctionnellement spécialisées. Qu'il s'agisse de l'analyse des liens anatomiques, des dépendances statistiques ou des interactions causales, la connectivité révèle des aspects fondamentaux du fonctionnement (dys)cérébral. Cependant, l'estimation et l'application de la connectivité posent encore des problèmes. C'est pourquoi cette thèse est consacrée à surmonter ces défis. Le premier défi provient de l'effet néfaste du bruit systématique (comme les mouvements de la tête) sur les estimations de la connectivité. Nous avons proposé un indice qui décrit la qualité de la connectivité et qui peut refléter différents types d'artefacts, d'erreurs de traitement et de pathologie cérébrale, permettant son utilisation étendue dans le suivi de la qualité des données et les investigations méthodologiques. En outre, les altérations de la connectivité jouent un rôle inestimable dans la compréhension des dysfonctionnements cérébraux. En étudiant certains mécanismes de l'épilepsie, nous montrons que la connectivité peut suivre les changements progressifs de la susceptibilité aux crises et identifier les facteurs déterminants de la génération des crises. L'identification des moments critiques de modification de la connectivité pourrait aider à prédire avec succès les crises. Enfin, on ne comprend pas bien comment le cerveau s'adapte aux exigences des tâches cognitives à une échelle de temps rapide. Nous présentons une combinaison d'EEG intracrâniens et de mesures de pointe épileptiques pour étudier la dynamique des réseaux pendant la mémoire de reconnaissance. Il est essentiel de comprendre comment le cerveau fait face dynamiquement aux changements rapides des demandes cognitives pour comprendre les bases neurales de la cognition. En conclusion, l'objectif modeste de cette thèse est de répondre au moins partiellement à certains des nombreux défis auxquels les neurosciences actuelles sont confrontées
We picture the brain as a complex network of structurally connected regions that are functionally coupled. Brain functions arise from the coordinated activity of distant cortical regions. Connectivity is used to represent the cooperation of segregated and functionally specialized brain regions. Whether it is the analysis of anatomical links, statistical dependencies, or causal interactions, connectivity reveals fundamental aspects of brain (dys)function. However, estimating and applying connectivity still faces many challenges; therefore, this work is devoted to tackling them. The first challenge stems from the detrimental effect of systematic noise (such as head movements) on connectivity estimates. We proposed an index that depicts connectivity quality and can reflect various artifacts, processing errors, and brain pathology, allowing extensive use in data quality screening and methodological investigations. Furthermore, connectivity alterations play an invaluable role in understanding brain dysfunction. Investigating the mechanisms of epilepsy, we show that connectivity can track gradual changes of seizure susceptibility and identify driving factors of seizure generation. Identifying critical times of connectivity changes could help in successful seizure prediction. Finally, how the brain adapts to task demands on fast timescales is not well understood. We present a combination of intracranial EEG and state-of-art measures to investigate network dynamics during recognition memory. Understanding how the brain dynamically faces rapid changes in cognitive demands is vital to our comprehension of the neural basis of cognition. In conclusion, the modest goal of this thesis is to at least partially answer some of the many challenges that current neuroscience is facing

With the growth in number of international schools from the 50 identified in 1964 to the current 5,000 international schools worldwide with an associated teaching staff of 250,000 predicted to grow to 10,000 and 500,000 respectively by 2020, the international school is an increasingly prominent sector of education. Yet despite this standing what is known about how international schools discipline and silence their dissenting teachers is negligible. Metaphorically speaking, the problem is that the "black box" of teacher control and correction remains firmly closed in the international school domain. The aim of this study is to open up the "black box" as it were, to use my experiences as an educator to shed some light on how organisational structures and the people working within them can disempower, silence and discipline a dissenting teacher. Although I have used autoethnography as methodology the study is not purely autoethnographic but rather a bricolage of methods that through conceptual enquiries of a philosophical nature (on the emotions, ethics etc.) deepen not only my narrative research but also my understanding of the issues. Engaging with these diverse philosophical analyses has taken me from questions of essence that focused on a Cartesian understanding of difference and conflict ( us/them; cause/effect ; guilt/innocence; darkness/light) to a posthumanist stance on both and questions of how conflict can be understood as emergent from intra-actions between apparatuses of material-discursive practices, conditions both social and material. For conflicts are collisions head on, to the accompaniment of squealing brakes and breaking glass. And it is important to understand not why but how such collisions occur if we are to avoid, prevent, or resolve them ethically. As such I hope that this study contributes to studies on conflict by showing how discourses, material conditions, affect, and power can converge to produce situations with serious consequences for those involved.

Abstract:
The purpose of this thesis was to extend our understanding of the assessment and interpretation of aerobic exercise function of paediatric patients with cystic fibrosis (CF). The first investigation sought to establish (1) the validity of traditional criteria to verify maximal oxygen (V ̇O2max) during a maximal cardiopulmonary exercise test (CPET); and (2) the utility of supramaximal verification (Smax) to confirm V ̇O2max. Traditional criteria significantly underreported V ̇O2max, whilst Smax was shown to provide a valid measurement in this patient group. The reproducibility of this CPET protocol, over the short- (48 h) and medium- (4-6 weeks) term, was then established in study two. V ̇O2max was repeatedly determined with no learning effect over 48 h (typical error (TE): ∆150 mL; ∆9.3%) and 4-6 weeks (TE: ∆160 mL; ∆13.3%). Supplementary maximal and submaximal CPET parameters should be incorporated for a comprehensive evaluation of a patient, however they are characterised by greater variability over time. The influence of mild-to-moderate CF on aerobic exercise function and the matching of muscle O2 delivery-to-O2 utilisation during ramp incremental exercise to exhaustion were then examined in study three. Aerobic function was impaired in CF, indicated by very likely reduced fat-free mass normalised V ̇O2max (mean difference, ±90% CI: -7.9 mL∙kg-1∙min-1, ±6.1), very likely lower V ̇O2 gain (-1.44 mL∙min-1∙W-1, ±1.12) and a likely slower V ̇O2 mean response time (MRT) (11 s, ±13). Arterial oxygen saturation was lower in CF, supporting the notion that centrally mediated O2 delivery may be impaired during ramp incremental exercise. Although a faster rate of fractional O2 extraction would be expected in the face of reduced O2 delivery, this was not observed, suggesting additional impairment in O2 extraction and utilisation at the periphery in CF. The fourth study then demonstrated the clinical utility of CPET to assess the response to 12 weeks treatment with Ivacaftor, using a case-based design. Whilst one patient with relatively mild disease demonstrated no meaningful change in V ̇O2max, the second demonstrated a 30% improvement in V ̇O2max, due to increased O2 delivery and extraction. Furthermore, changes in aerobic function were detected earlier than spirometric indices of pulmonary function. This study demonstrated that CPET represents an important and comprehensive clinical assessment tool and its use as an outcome measure in the functional assessment of patients is encouraged. Study five investigated the V ̇O2 kinetics in this patient group. During moderate intensity cycling, the phase II V ̇O2 time constant (τ) (p = 0.84, effect size (ES) = 0.11) and overall MRT (p = 0.52, ES=0.33) were not slower in CF. However, both were slowed during very heavy intensity cycling (p = 0.02, ES = 1.28 and p = 0.01, ES = 1.40, respectively) in CF. Cardiac output and muscle deoxygenation dynamics were unaltered in CF, however, the arterial-venous O2 content difference (C(a-v ̅)O2) was reduced (p=0.03) during VH and ∆C(a-v ̅)O2 correlated with the phase II τ (r= -0.85; p=0.02) and MRT (r = -0.79; p=0.03) in CF. This study showed that impaired oxidative muscle metabolism in this group is exercise intensity-dependent and mechanistically linked to an intrinsic intramuscular impairment, which limits O2 extraction and utilisation. In conclusion, this thesis has provided guidelines for a valid and reproducible CPET protocol for children and adolescents with mild-to-moderate CF, demonstrated the utility of CPET as clinical outcome measure and furthered our understanding of the factors responsible for impaired aerobic exercise function in this patient group.

Le syndrome de l’X fragile (FXS) est la première cause de déficience intellectuelle monogénique héréditaire liée à l’X (1/3000 garçons). Outre la déficience mentale, les patients présentent des troubles du spectre autistique et des perturbations sensorielles, parmi lesquelles une altération des fonctions visuelles. L’origine moléculaire du FXS se trouve dans la mise en silence du gène FMR1, ce dernier ne pouvant plus, par conséquent, exprimer la protéine FMRP. Aux niveaux moléculaire et cellulaire, la perte de FMRP engendre des anomalies structurelles et fonctionnelles des synapses, aux niveaux cérébral mais aussi rétinien. Cette thèse tend à approfondir les connaissances des phénotypes rétinien et visuel en absence de protéine FMRP, ainsi que l’étude de la part d’implication de ce phénotype rétinien dans les phénotypes visuel et global du syndrome de l’X fragile.Nous avons révélé la précocité et la stabilité des anomalies rétiniennes en absence de la protéine FMRP, ce qui confirme la force du phénotype rétinien en conditions FXS, et soulève la question de son influence sur d’autres structures. En parallèle, nous avons montré que l’absence de protéine FMRP engendre des perturbations significatives de plusieurs traits visuels d’un point de vue comportemental. Fragile X syndrome (FXS) is the leading cause of X-linked hereditary monogenic intellectual disability (1/3000 boys). In addition to mental retardation, patients present autism spectrum disorders and sensory disturbances, including impaired visual functions. The molecular origin of the FXS is the silencing of the FMR1 gene, which can no longer express the FMRP protein. At the molecular and cellular levels, the loss of FMRP results in structural and functional abnormalities of the synapses, at the brain as well as at the retinal level. This PhD thesis project aims to deepen knowledge of retinal and visual phenotypes in the absence of FMRP protein, as well as the study of the involvement of this retinal phenotype in the visual and global phenotypes of fragile X syndrome.We have revealed the precocity and stability of retinal abnormalities in the absence of the FMRP protein, which confirms the strength of the retinal phenotype under FXS conditions, and raises the question of its influence on other structures. In parallel, we have shown that the absence of FMRP protein causes significant disturbances of several visual traits in a behavioral standpoint. Finally, a strategy was developed to create an animal model "FXS-retina-specific", the study of which would provide information on the consequences of the absence of the FMRP protein only in the retina.This PhD thesis project highlights the importance of visual and retinal phenotypes of FXS, and provides new insights supporting the centrality of sensory disturbances in this pathology

Abstract:
A comprehensive population study in the Middle East was conducted using different genetic markers in order to establish a wider genetic profile of the Middle Eastern populations. The main goal of this study was to analyze DNA from samples collected from different locations, and produce genetic motifs and patterns that could be used to identify and distinguish the target populations. This information will allow us to analyze the ancestry of these populations, their interactions through time and space, and the effects these interactions have on the populations' structure. We have collected around 1300 individual samples from different populations in the Middle East ranging from Oman in southern Arabia, to the West Bank and Gaza in Palestine. Our samples can be divided into two primary groups: 320 samples come from Oman; this region is important because of its geographical proximity to Yemen which is perceived as the historical area where the Arabs originated and 800 samples came from Palestine, a central region in the Middle East that connects Asia and Africa and was a passageway between the two continents through history. The samples collected from Oman have genealogy charts that were provided by the participants, while the samples from Gaza lack the genealogy charts. DNA was extracted from the samples, amplified using PCR technology, sequenced and genotyped using non-recombining genetic markers. Short Tandem Repeats(STR) were screened in the samples.A specific STR marker, DYS458, exhibited an alternative allele at repeat number 18 which was a 2 base pair shift identified as microvariant 18.2. The samples showed an unusually high frequency of microvariant 18.2.When microvariant 18.2 haplotype was combined with Modal Cohen Haplotype (MCH) motifs, we were able to infer some genetic characteristics about our samples. The Cohen Modal haplotype was used in combination with the results of the 18.2 DYS458 analyses to construct a snapshot of the Middle East, using NETWORK software to analyze the relationship within and between the samples from Oman, which is located in Southern Arabia, and the areas North of Arabia (Jordan, Syria, and Palestine), which is also known as the Levant.

Ivermectin has been a very useful drug for the control of diseases caused by helminths. However, the occurrence of drug resistant parasites is a major concern. In an attempt to mimic the conditions under which ivermectin resistance is selected for in the field, James and Davey (2009) generated the IVR6 and IVR10 Caenorhabditis elegans strains from a wild-type strain by growing them in the presence of sub-lethal doses of ivermectin for several generations. To better understand the mechanisms by which ivermectin resistance might arise, I have investigated the IVR6 and IVR10 strains. I found that the IVR6 and IVR10 strains are dye-filling defective (Dyf), a phenotype previously associated with ivermectin resistance. Our results indicate that IVR6 and IVR10 have the same level of ivermectin resistance. We discovered a frame shift mutation in the dyf-7 gene of both strains. The location of dyf-7 on the X-chromosome is consistent with the results of our resistance mapping experiment. IVR6 and IVR10's dye-filling phenotype is roughly 80% penetrant and we show that ivermectin can select for the phenotype. Only dye-filling defective worms can grow at 10 ng/ml ivermectin. We have tested four other dye-filling defective strains, including a strain carrying a mutant dyf-7 allele and all were ivermectin resistant. Preliminary results indicate that dyf-7 confers levamisole resistance, suggesting an alternate mechanism for multidrug resistance. Taken together, my results show that an allele of the dyf-7 gene is the cause of ivermectin resistance in the IVR6 and IVR10 strains and that the dendrite morphology phenotype of Dyf genes is essential for their ability to confer resistance.
L'Ivermectine est un médicament qui a été très utile pour contrôler des maladies causées par les nématodes parasitiques. Cependant, le développement chez les parasites d'une résistance à ce médicament reste inquiétant. Ainsi, en reproduisant les conditions dans lesquelles les parasites développent une résistance à l'ivermectine, James et Davey (2009) ont généré deux souches de Caenorhabditis elegans, l'IVR6 et l'IVR10, en les cultivant sur une dose non-létale pour quelques générations. Afin de mieux comprendre les mécanismes impliqués dans le développement de la résistance à l'ivermectine, j'ai étudié les souches d'IVR6 et d'IVR10. J'ai découvert que celles-ci sont déficientes en absorption de teinture, soit un phénotype associé à la résistance à l'ivermectine. Les résultats obtenus présentaient le même niveau de résistance pour les deux souches. De plus, nous avons découvert une mutation dans le gène dyf-7, aussi chez les deux souches. L'usage de la cartographie génétique qui utilise les polymorphismes pour un nucléotide (SNPs), m'a permis de déterminer que le locus de résistance est en accord avec le locus de dyf-7. Le phénotype de déficience d'absorption de teinture est pénétrant à 80% chez l'IVR6 et l'IVR10. Quant à l'ivermectine, celui-ci peut sélectionner pour ce phénotype. Seulement les vers déficients en absorption de teinture peuvent survivre sur 10 ng/ml d'ivermectine. J'ai examiné quatre autres souches qui sont déficientes en absorption de teinte, incluant une souche avec un allèle différent de dyf-7; elles démontrent toutes une résistance à l'ivermectine. Des expériences préliminaires indiquent que le dyf-7 induit une résistance à un autre médicament, lévamisole. Ce qui me suggère un nouveau moyen de développer la multirésistance. Pris dans leur ensemble, mes résultats montrent que l'allèle du gène dyf-7 est la cause de la résistance à l'ivermectine chez les souches d'IVR6 et d'IVR10 et que la malformation des dendrites chez les souches Dyf est essentielle au développement de la résistance.

Molecular biology and genetics, single nucleotide polymorphism genetic mapping, phenotypic assays including behavioral assessment, and fluorescent microscopy of GFP-tagged proteins were used to study ciliary defects in the nematode Caenorhabditis elegans. Mammalian cilia are multifunctional. Some of the physiological roles in which they are involved include sensing developmental signaling molecules and ligands as well as creating flows of mucus and cerebrospinal fluid that function as flow meters and mechanosensors. Due to the multifunctional nature of cilia, it is not surprising that many human diseases can be caused by ciliary defects. Bardet-Biedl Syndrome is a rare genetic ciliopathy characterized by retinal degeneration, polydactyly, obesity, cystic kidneys, mental retardation, and many other ailments. We have identified osm- 12/bbs-7 to be a C. elegans homologue of human BBS7, a gene known to cause Bardet-Biedl Syndrome when mutated. With the help of Michel Leroux’s group, I showed the BBS-7 protein to be localized to the base of cilia and to undergo intraflagellar transport along the ciliary axoneme. Our findings suggest that BBS- 7 plays a role in the assembly and/or functioning of the IFT complex. I also performed a mutagenesis and phenotypic screen for animals defective in the uptake of DiI into a subset of their ciliated neurons in order to identify new components involved in ciliogenesis and IFT. I describe an extended bulked segregant analysis (BSA) mapping methodology, which can save time and resources by filtering out alleles of previously known genes without performing time-consuming interval mapping. In addition, I identified one of the 11 dyefilling defective alleles from the screen to be a novel allele of dyf-3, which encodes a protein required for sensory cilia formation

L’oggetto della presente tesi è la proposta di traduzione in italiano dell’intero romanzo francese “jeunesse” Dys sur Dix: Dylan a un secret…il est dyslexique, scritto dall’autrice francese Delphine Pessin ed edito da Pocket Jeunesse nel 2018. L’opera narra la storia di Dylan, un ragazzino di quattordici anni che soffre di dislessia, disturbo che lo ha condizionato da quando è bambino, che influisce negativamente sui suoi risultati scolastici e sui rapporti con genitori e amici. L’opera è in grado di affrontare un tema complesso come i disturbi specifici dell’apprendimento (DSA), raccontandolo in maniera semplice, ironica e accattivante, ma mai banale; nello specifico, è un esempio di narrativa per ragazzi sui DSA, genere di romanzo ancora poco presente nel panorama editoriale italiano. Il primo capitolo dell’elaborato esplora diacronicamente il mercato editoriale francese e italiano, dalla loro nascita ad oggi, focalizzandosi poi sulla letteratura per ragazzi a tema DSA in Italia. I tre capitoli successivi sono rispettivamente dedicati agli aspetti seguenti: analisi testuale del romanzo in funzione del processo di traduzione dello stesso, proposta di traduzione integrale dell’opera e, infine, commento alla traduzione con riferimento alle teorie dei translation studies. Abstract Purpose: The issue that this report addresses revolves around sweep-salting of footways and cycleways. More specifically, it focuses on the fact that large differences in elevation can lead to varying land surface temperatures, which, in turn, complicates winter maintenance when using the sweep-salting method. With Jönköping as basis of study, the aim of this report has been to examine the level of knowledge regarding sweepsalting of footways and cycleways in Sweden, this to show optimization possibilities to a developed sweepsalting in Jönköping, taken into account the height differences prevailing in the city.   Method: To obtain data, structured and semi-structured interviews have been carried out. All the interviews have been done and recorded via telephone calls and the interview questions have been emailed to the respondents in advance. The structured interviews have been targeted at four municipalities and the semi-structured interviews have been targeted at two concerned governmental bodies. Parallel to the interviews, three documents have been subject of analysis: Karlstadslaken by Tomas Stomberg, GCM-handboken published by Trafikverket, and Vinterväghållning och expertsystem – en kunskapsöversikt by Magnus Ljungberg. Findings: Sweep-salting as high winter maintenance method is a complicated task. It should be comprehensive, reliable, and conducted hastily over long distances and fairly large areas. It is a relatively new method and knowledge is, as of yet, limited. In the interviews, it became evident that municipalities largely are self-managing in this matter, but that a desire for more intermunicipal collaboration exists. Primarily, municipalities see the need of a governmental framework on how to conduct sweep-salting most efficiently.  De-icing is not carried out the same way in the north of Sweden as it is in the southern parts of the country. In the north of Sweden, the primary focus is snow clearance, and sweep-salting is only carried out when temperatures allow for it. In the south of Sweden, on the other hand, where temperatures often fluctuate around zero degrees Celsius, and snow quantities are manageable, sweep-salting is more prioritized. Implications: Besides keeping track of aerial and land surface temperatures, it is important to control the dew point, in order to apply the appropriate maintenance methods on the most crucial areas of footways and cycleways, at the right time. It is also important to stay updated on municipalities’ development of sweep-salting. Limitations: The economic aspect of sweepsalting is not considered in this study, neither is the perspective from cyclists nor pedestrians of the results of sweepsalting. The study would probably have reached a better result if more municipalities would have been included. Au départ modèle politique occidental, la décentralisation est érigée, dans les années 1990, en modèle de développement puis, à partir de 2000, en outil de la lutte contre la pauvreté. Le modèle est, depuis, financé et transféré dans les pays en développement, censé être levier d’une bonne gouvernance et d’une paix sociale. Cependant, malgré 25 ans d’efforts pour l’instauration du modèle, à Madagascar et Haïti, la décentralisation n’est toujours pas effective. Plus globalement, les deux pays se caractérisent par une instabilité chronique, un faux-semblant de démocratie, une aggravation de la pauvreté et un maintien ou creusement des inégalités. Comment expliquer ce paradoxe? Si le modèle de décentralisation continue d’être transféré et reçu, malgré les objectifs manqués, c’est qu’il sert des intérêts officieux d’acteurs du Sud… et du Nord. De plus, nous considérons que c’est le transfert du modèle lui-même qui est source de dysfonctionnement. En donnant l’opportunité à de nouveaux acteurs d’accéder au pouvoir, il multiplie les sources de conflits et stimule des jeux et enjeux de pouvoir locaux. Nous proposons de décrire les profils des acteurs du Sud, visibles et invisibles, qui s’approprient le modèle et de décoder leurs stratégies de captation de pouvoir. Qui sont-ils? Comment arrivent-ils à entretenir le système tout en le freinant? Comment instrumentalisent-ils le modèle pour en tirer parti? Quels sont leurs objectifs? Quelles sont leurs stratégies d’alliance, et qui est en conflit avec qui? Plus largement, il s’agira de comprendre comment ces jeux s’articulent avec ceux des acteurs du Nord pour conduire au maintien du transfert d’un modèle dysfonctionnel, à l’entretien de l’hégémonie de certains, et à (re)créer des tensions menant aux crises. Originally a western political model, decentralization became a development model and a tool in the fight against poverty between 1990 and 2000. The model has since been massively financed and transferred to developing countries, as it was expected to be a lever of good governance and social peace. However, despite 25 years of efforts to establish the model in Haiti and Madagascar, decentralization is still not efficient. More generally, the two countries are characterized by chronic instability, a false pretence of democracy, an aggravation of poverty and maintained or increased inequalities. How can we explain this paradox? Decentralization continues to be transferred and received despite consistently missing the mark because it serves unofficial interests of actors from the South…and the North. Moreover, we consider that the model’s transfer itself is at the source of its malfunctioning. As it facilitates access to power for new stakeholders, it multiplies the sources of conflicts and foments local power games. We offer a profile of Southern stakeholders, visible and invisible, who appropriate the model and aim to decode theirs strategies of power capture. Who are they? How can they maintain the system and simultaneously sabotage it? How do they turn the model into their own tool? What are their goals? What are their strategies in terms of alliances and conflicts? More broadly, we would like to show how these power games relate to Northern realities and lead to the persistent transfer of a dysfunctional model, the preservation of a small hegemony, and the (re)creation of tensions which can lead to crises. This system is ongoing: the crises justify external aid intervention and the input of a new model. In this vicious circle something does functions, but at the expense of the target population. This paradox offers an opportunity to question the aid approach based on the transfer of a standardized model

Abstract:
This thesis examines how women perform as leaders within male-dominated professions, including law, business, politics, the military, and the academy. In studying women's performances in terms of the corporeal and spectacular, the investigation seeks to understand how particular women enact leadership through their materiality within specific times and places. Drawing on Gilles Deleuze and Felix Guattari's (1988) theorising of the processes of 'becoming', woman-as-leader is studied as an entity that passes from one incomplete and multiple assemblage to another, rather than as a singular 'developing' identity. The research is located within and between the paradoxes that complicate the performances of leadership for women. One key paradox serving as a rationale for this investigation is that, while 'equity' has become a truism of contemporary leadership, it is clear from formal reports (for example, the 2002 Equal Opportunity for Women in the Workplace Agency (EOWA) report), that many women continue to be marginalised and under-represented as leaders and senior managers. Moreover, those few women who have achieved success often acknowledge themselves as both legitimately and differently - and sometimes awkwardly, located as leaders in the everyday enactments of their work. The investigation of leadership within and between such paradoxes is problematic for a neo-liberal order of thinking, and even for socially critical theory, because of the assumptions that modernist literature makes about women's struggle for political legitimacy (ie, a narrative of progress, emancipation, and/or linear cumulative historical development). It is for this reason that the conceptual tools used in this study are drawn from post-feminist and post-structuralist theory. Such theorising refuses literal categories in favour of 'ironic categories' (Rorty, 1989) where two apparently oppositional ideas are understood to be both necessary and true. To explore women 'becoming' leaders (Deleuze & Guattari, 1988), 'woman-as-leader' is interrogated using Jean François Lyotard's (1984) notion of 'performativity,' Mary Russo's (1994) theorising of the embodied spectacle of 'the female grotesque', and Richard Rorty (1989) and Donna Haraway's (1991) insistence on partiality, doubt, and the importance of 'undoing' the fixity of modernist categories - in this instance, for women. One ironic category of importance to the study is Haraway's theorising of a 'cyborgian identity', a technological assemblage that is part-human/part-machine. This allows acknowledgement that women leaders inhabit realms beyond the boundaries imposed by the same/difference, human/machine, present/past, and real/virtue binaries. Using these tools, the performances of a number of women leaders is examined in an empirical study that focuses on a few individual women located in male-defined and male dominated settings. The empirical work has two key components. First, it provides a reading of three moments in time where a female individual dys-appears (Leder, 1990) in the public gaze, erupting as a unique spectacle in spaces that are both enabling and constraining. It foregrounds the unique complexities of three public performances in which women made a spectacle of themselves, while the analysis refuses to either celebrate the individuals involved, or to bemoan the conditions under which they did so. The analysis demonstrates the value of re-thinking leadership in terms of its complexity for the female as embodied public 'performer'. It then moves on to focus specifically on the (embodied, spectacular) tactics being deployed by women leaders in contemporary professional work. This analysis is located in the professions of law, business, politics, the military, and the academy. The data-as-evidence emerging from the analysis show women leaders to be both and neither enacting and troubling 'proper' (ie, traditional and/or known) leadership conventions. The analysis provides a reading of how, through certain tactical shifts, women work to 'de-territorialise' both the 'forms of content' and 'forms of expression' (Deleuze & Guattari, 1988) constituting leadership performances. It makes visible the tactical assemblages these women deploy, and the ways in which such tactics separate, combine, and compound the same/difference, equality/inequality, either/or binaries. The specific tactical manoeuvres for achieving legitimacy in the public gaze cluster around four identifiable ironic categories: (i) legitimate cross-dressing (ii) assertive defence (iii) proper blasphemy, and (iv) humanly-machinic. When taken together, the two components of the empirical study compel a re-theorising of 'woman-as-leader' as both insider and outsider, an entity engaging in the on-going work of diss-assembling and re-assembling a leaderly self. This thesis examines how women perform as leaders within male-dominated professions, including law, business, politics, the military, and the academy. In studying women's performances in terms of the corporeal and spectacular, the investigation seeks to understand how particular women enact leadership through their materiality within specific times and places. Drawing on Gilles Deleuze and Felix Guattari's (1988) theorising of the processes of 'becoming', woman-as-leader is studied as an entity that passes from one incomplete and multiple assemblage to another, rather than as a singular 'developing' identity. The research is located within and between the paradoxes that complicate the performances of leadership for women. One key paradox serving as a rationale for this investigation is that, while 'equity' has become a truism of contemporary leadership, it is clear from formal reports (for example, the 2002 Equal Opportunity for Women in the Workplace Agency (EOWA) report), that many women continue to be marginalised and under-represented as leaders and senior managers. Moreover, those few women who have achieved success often acknowledge themselves as both legitimately and differently - and sometimes awkwardly, located as leaders in the everyday enactments of their work. The investigation of leadership within and between such paradoxes is problematic for a neo-liberal order of thinking, and even for socially critical theory, because of the assumptions that modernist literature makes about women's struggle for political legitimacy (ie, a narrative of progress, emancipation, and/or linear cumulative historical development). It is for this reason that the conceptual tools used in this study are drawn from post-feminist and post-structuralist theory. Such theorising refuses literal categories in favour of 'ironic categories' (Rorty, 1989) where two apparently oppositional ideas are understood to be both necessary and true. To explore women 'becoming' leaders (Deleuze & Guattari, 1988), 'woman-as-leader' is interrogated using Jean François Lyotard's (1984) notion of 'performativity,' Mary Russo's (1994) theorising of the embodied spectacle of 'the female grotesque', and Richard Rorty (1989) and Donna Haraway's (1991) insistence on partiality, doubt, and the importance of 'undoing' the fixity of modernist categories - in this instance, for women. One ironic category of importance to the study is Haraway's theorising of a 'cyborgian identity', a technological assemblage that is part-human/part-machine. This allows acknowledgement that women leaders inhabit realms beyond the boundaries imposed by the same/difference, human/machine, present/past, and real/virtue binaries. Using these tools, the performances of a number of women leaders is examined in an empirical study that focuses on a few individual women located in male-defined and male dominated settings. The empirical work has two key components. First, it provides a reading of three moments in time where a female individual dys-appears (Leder, 1990) in the public gaze, erupting as a unique spectacle in spaces that are both enabling and constraining. It foregrounds the unique complexities of three public performances in which women made a spectacle of themselves, while the analysis refuses to either celebrate the individuals involved, or to bemoan the conditions under which they did so. The analysis demonstrates the value of re-thinking leadership in terms of its complexity for the female as embodied public 'performer'. It then moves on to focus specifically on the (embodied, spectacular) tactics being deployed by women leaders in contemporary professional work. This analysis is located in the professions of law, business, politics, the military, and the academy. The data-as-evidence emerging from the analysis show women leaders to be both and neither enacting and troubling 'proper' (ie, traditional and/or known) leadership conventions. The analysis provides a reading of how, through certain tactical shifts, women work to 'de-territorialise' both the 'forms of content' and 'forms of expression' (Deleuze & Guattari, 1988) constituting leadership performances. It makes visible the tactical assemblages these women deploy, and the ways in which such tactics separate, combine, and compound the same/difference, equality/inequality, either/or binaries. The specific tactical manoeuvres for achieving legitimacy in the public gaze cluster around four identifiable ironic categories: (i) legitimate cross-dressing (ii) assertive defence (iii) proper blasphemy, and (iv) humanly-machinic. When taken together, the two components of the empirical study compel a re-theorising of 'woman-as-leader' as both insider and outsider, an entity engaging in the on-going work of diss-assembling and re-assembling a leaderly self. Dysferlin(DYSF) gene encoding dysferlin is mutated in Miyoshi myopathy and Limb-Girdle Muscular Dystrophy type 2B(LGMD 2B), the two main phenotypes recognized in dysferlinopathies. Dysferlin deficiency in muscle is the most relevant feature for the diagnosis of dysferlinopathy and prompts the search for mutations in DYSF gene. DYSF, located on chromosome 2p13, contains 55 coding exons and spans 150 kb of genomic DNA. We performed a genomic analysis of the DYSF coding sequence in 11 unrelated LGMD 2B patients, including 7 suspected LGMD 2B Taiwanese patients, and 4 Japanese patients with 6 confirmed mutations in DYSF gene. All patients showed an absence or drastic decrease of dysferlin expression in muscle. A primary screening of DYSF using denaturing high performance liquid chromatography(DHPLC)of PCR products of each of 55 exons of the gene was followed by sequencing whenever a variation was detected. In 7 Taiwanese patients, 17 sequence variations were identified in DYSF, 3 of which predict single amino-acid substitution and are novel, while the other 14 changes are known SNPs (Single nucleotide polymorphisms). The three novel sequence variations, G486、A3472G、C334T, was not detected in 150 normal individuals and was identified as potential pathogenic mutation (SNP<1%). These three mutations were widely spread in the coding sequence of the gene.

A series of metal free organic dyes (DA) with two anchoring groups were synthesized for DSSCs application. Different arylamines and 2-cyanoacrylic acid were used as the electron donor and the electron acceptor, respectively. The dyes absorb intensely (ε = 41900−54600 M-1cm-1) in the range of 350−450 nm. The HOMO (0.97 to 1.24 V vs NHE) and LUMO (-0.99 V to -1.64 vs NHE) levels calculated from electrochemical and optical data assure favorable electron injection and regeneration of the dye after electron injection. The power conversion efficiencies of DSSCs based on these dyes are in the range of 2.60-7.28 %, which are 1.2−1.7 times heigher than that of S1 dye with only one anachoring group. The power conversion efficiency of DSSC based on DA5 has the best power conversion efficiency (7.28%) among all. The efficiency reaches 88 % of the standaed cell based on N719 fabricated and measured under the same condicitions. The presence of two anchoring groups in DA dyes increases electron injection efficiency and results in higher short circuit currents. Compared the S1 congener, DA1−DA3 can more effectively suppress charge recombination, leading to higher open circuit voltage.
