Introduction and Aim: W. coagulans Dunal has been reported to contain an array of bioactive compounds. The present investigation was carried out to investigate MMP inhibitory molecules from W. coagulans Dunal fruit. Materials and Methods: Isolation of active principle compounds and anti-ChC activity from methanol extract of W. coagulans Dunal fruit were carried out using chromatographic techniques and dot-blot assay on X-ray film. Active principle compounds were identified by ESI MS, 1H, and 13C NMR spectroscopy. The Bioefficacy of compounds was assessed by in vitro solution assay and gelatin zymography. Results: Dot blot assay of methanol extract of W. coagulans Dunal fruit exhibited inhibitory activity against ChC (Clostridium histolyticum collagenase). Bioactivity assay guided chromatographic fractionation revealed the presence of five compounds out of which three were tentatively identified as Coagulin-H, Coagulin-L, and Coagulin-O by ESI MS, 1H, and 13C NMR spectroscopy. In vitro solution assay revealed Coagulin-H exhibits the highest MMP-2 and MMP-9 inhibitory activity. Coagulin-L and Coagulin-O exhibit 80.57 ± 2.1 % and 70.96 ± 2.8 % inhibitor activities against MMP-2 and 78.94 ± 3.6% and 63.15 ± 3.8% inhibitor activities against MMP-9 at 150 µg/ml respectively. In gel inhibition assay by gelatin zymography revealed that crude extracted residue of fruit exhibits dose-dependent inhibitory potential against MMPs of NIH3T3 fibroblast and HeLa cervical cells. Conclusion: Our results advocate the anti-MMP potential of purified Coagulin-H, Coagulin-L, and Coagulin-O of W. coagulans Dunal fruits. These bioactive principles could be further investigated in detail for anti-cancer properties.

Cubiu (Solanum sessiliflorum Dunal), a genetic resource from western Amazon, presents a great potential for the modern agriculture, on account of its rusticity, good yield capacity and usage of its fruits in various diversified ways. On account of it being a sexual reproduction plant, it keeps a high genetic variability as to the agronomic importance characteristics which are not interesting in a commercial viewpoint, where the purpose is the evenness of the growing plants as well as their fructification. Micropropagation is an alternative for obtaining clones with desirable characteristics such as high productivity, superior quality or biotic and abiotic stress tolerance, providing a fast multiplication within a small physical space. The aim of the present study was to establish protocols for the in vitro culture of cubiu. Different mg/L concentrations of the BAP (0.0; 0.5; 1.0; 3.0; 5.0) and AIA (0.0; 0.5; 1.0; 2.0; 3.0) growth regulators and all possible combinations between them in MS medium were assessed in the in vitro induction and regeneration of cubiu sproutings and rooting. The experimental design was in random blocks with five replicates and 25 treatments, constituting a 5x5 factorial scheme. The best BAP and AIA mg/L combinations the following respectively: for leaves 1.0 plus 0.5 producing an average number of 8 leaves per explant; for sprountings 3.0 plus 0.5 producing an average number of 3.10 sprouts per explant; for the height of sproutings 3.0 plus 0.5 producing an average height of 4.17 cm; for root 1.0 plus 0.5 producing an average number of 8.45 roots and for the length of root 1.0 and 0.5 producing a 6.4 cm mean length. It was possible to micropropagate the S. sessiliflorum species adding BAP and AIA growth regulators to the MS medium, obtaining a larger number of sproutings as well as complete plants with well developed root system and ideal height for acclimatization.

The objective of this study was to evaluate the effect of different recovery materials on the physiological quality of cubiu seeds. This was evaluated with three binder materials (manioc starch, inert talc and dolomitic limestone) and two binder polymers (polyvinyl acetate and gum arabic) with a 30% concentration, and artificial dye finishing. The experimental design was completely randomized, in factorial scheme 3 (agglomerates) x 2 (binders), and additional treatment (bare seeds), containing four replicates and experimental units of fifty seeds/pellets. The seeds were evaluated for: yield; seed/pellet diameter; thousand seed weight; number of seeds per pellets; germination; germination speed index; mean germination time, seedling length; fresh and dry mass of the seedling and emergence. In general, pelletizing increased the weight and diameter of the propagation unit. The physical barrier applied to the seeds by the pelletization did not influence the germination, but delayed the germination rate in relation to the bare seeds, and this effect was not observed in seedling emergence. The coating with dolomitic limestone and polyvinyl acetate presented the best results of germination and vigor.

The cubiu is a fruit with high moisture, acid and pectin. In osmotic dehydration, the moisture that is associated with the perishability of the process and cost is reduced in the final product. The acidity is a limiting factor for acceptance is masked by the incorporation of sugar. As a bioactive substance, preservation retains the functionality of pectin, however, the partial hydrolysis facilitates the process and ensure the appearance of dehydrated fruit. This study aims to develop technologies for conservation and value addition of cubiu for industrialization in the form of fruit in passes. The ripe fruit of cubiu were subjected to different pretreatments: blanching or not, more natural fermentation or not and rapid or slow osmotic dehydration followed by convective drying at 65 ° C. The natural fermentation process was monitored by the determination of moisture, pH, total acidity, sodium chloride, pectin and texture of the slices of cubiu. After rapid and slow dehydration were determined at equilibrium and initial moisture, pH, acidity, total sugars, mass change, water loss, gain and efficiency of sucrose osmotic process. The experimental data of convective drying models were fitted to the Page, Lewis, simple exponential, with two and three parameters and Wang & Singh. The physic-chemical and sensory characteristics were used to monitor the whole process and final product quality. In relation to fresh fruit, money not increase the moisture and pH, decrease in total acidity and sodium and chloride did not significantly alter the texture. During the process of natural fermentation, the parameters moisture, acidity, texture and pH (control) were reduced from their values for pH (blanched) and sodium chloride was no increase in the values. The highest percentage of mass change, water loss and sucrose gain occurred in SF/DOR, SF/DOL and CF/DOL, respectively. The highest rate of drying was obtained by the treatment before the blanching, with natural fermentation and rapid osmotic dehydration, indicating that fermentation influenced the increase of drying rate and the reduction of process time. The r2 values were above 99% for the settings with the Page and Exponential models with three simple parameters. The natural fermentation process was detected during the immersion of slices in NaCl solutions cubiu, blanched or not. The physical and physic-chemical properties of slices cubiu were modified during the natural fermentation process, being more pronounced in slices pre-treated by blanching. The variation in mass loss of water and process efficiency were higher in the treatments without natural fermentation. The time of osmotic dehydration was reduced rapidly by natural fermentation. The highest and lowest drying time were obtained by treatment before the blanching, natural fermentation, followed by rapid osmotic dehydration. The simple exponential model with three parameters and Page have the best fit to experimental data. Separately, money and natural fermentation improved the organoleptic characteristics of cubiu passes in obtaining the highest percentage of acceptance.

Abstract:
The North Florence Dunal Aquifer is the only feasible source for drinking water for the coastal city of Florence, Oregon and Florence's Urban Growth Boundary. High infiltration rates and a shallow groundwater table leave the aquifer highly susceptible to contamination from septic tank effluent, storm runoff, chemical fertilizers and recreational ATV use throughout the dunes. Public interest in the quality and quantity of the aquifer water has been sparked since the City of Florence received a grant from the Environmental Protection Agency for a watershed protection and restoration project. Delineation of the shallow groundwater surface and its relationship to the surface water bodies within the dunes is crucial in protecting this drinking water source from contamination. This thesis project created a GIS representation of the shallow groundwater elevation and associated prediction error map. Surface water bodies were confirmed as window lakes into the dunal aquifer and no signs of perched aquifer conditions were observed between Holocene and Pleistocene dunes. Ground Penetrating Radar, well data provided by the city of Florence and LiDAR were the primary sources for data collection.

Abstract:
General Abstract There has been a steady increase in the use of medicinal plants in the last two decades in both developed and developing countries for prevention, management and treatment of diseases. This increase has been due to reasons such as ease of access, better cultural acceptability and compatibility, cost effectiveness and also the bid to “go natural”. Unfortunately, the notion that herbal medicines are relatively safe because they are natural has led to serious and potentially fatal consequences in phytotherapy. The lack of rigorous research to prove the effectiveness and safety of many medicinal plants is of great concern to the health care system. This thesis therefore addresses not just the efficacy, but also the safety of the extracts of the berry of Solanum aculeastrum - a medicinal plant used, among other things, for the treatment of breast cancer in the Eastern Cape Province of South Africa. Particular attention was paid to the possible effect of different processing methods of the berry extracts on inflammation, cytotoxicity, and toxicity. In studying the comparative effects of various processing methods, four different preparations of the extracts were investigated. These include fresh, dried, boiled fresh, and boiled dried berries. While the effect of processing on the anti-inflammatory properties of the extracts was not dose dependent, the percentage reduction in inflammation was highly significant and more prominent in both concentrations of the boiled fresh berries than the reference drug, indomethacin. Furthermore, the four extracts varied in their ability to act either centrally or peripherally in their effect on pain. Assessment of the analgesic response using the formalin test showed that, at both concentrations tested, none of the extracts inhibited the first phase of the formalin test. Furthermore, it was observed that boiling had differing results on the activity of the iii fresh and dried extracts. While boiling of the dried berries reduced pain in the rats, the opposite trend was observed with the boiled fresh berries. Results of the influence of processing of the berries on cytotoxicity indicated that the extracts are potent inhibitors of human breast, cervical and colonic carcinoma cells and the non-cancerous cells (both the actively dividing and confluent Chang liver cells). Although, in terms of relative potency, the fresh berries appeared to be the most active of the extracts, processing of the berries caused an increase in apoptotic cells and a subsequent decrease in the necrotic cells. The effect of processing on the safety of the berry of S. aculeastrum on the rats fed for 28 days was also investigated. The various doses (1, 10 and 25 mg/kg body weight) of all the four extracts did not alter the activity and the weight of the animals throughout the period of treatment. A reduction in organ to body weight ratio of the heart, kidney, liver and spleen was observed in all the extracts. Regarding the haematological parameters, different patterns of effect were observed between the extracts and within the treated doses. The observed alterations in the biochemical parameters by the various extract of Solanum aculeastrum berries at all the doses may have consequential effects on the normal functioning of these organs. In conclusion, this study has shown that there is some justification for the traditional use of the berries of Solanum aculeastrum in the treatment of inflammation related ailments and cancer. However, the medicinal use of the plant also poses considerable health risks. Investigation conducted into the plant’s safety revealed that the berry extracts were nephrotoxic, hepatotoxic, haematotoxic and at higher doses, fatal. Another concern with regard to the plant’s safety is the non-selectivity of its extracts in the inhibition of carcinoma, actively dividing and un-dividing cells. Assessment of the effect of the processing on the berry’s efficacy and safety as herbal iv remedy produced mixed results. On the one hand, processing seemed to improve the extract’s anti-inflammatory and analgesic activity, while reducing its cytotoxic potential. On the other hand, a reduction in the toxicity was observed on the processed extracts compared to the fresh ones. This may be an indication that processing has an overall beneficial effect on the medicinal properties of the plant and should thus be considered as a method of making the berries of Solanum aculeastrum safer for use as a herbal remedy.

The cubiu (Solanum sessiliflorum Dunal) is a typical Amazon fruit featuring pulp with high-yielding, acid flavor, yellow color and rich in pectin. Thus, the objective of this work was to evaluate the use of the mesocarp of cubiu to produce pickles testing the effect of different formulations of liquid medium on the sensory attributes of the products. The ripe fruits from the experimental station of the Ariau belonging to the National Research Institute of the Amazon (INPA) went through the wash, Peel, cut and remove the core. The sliced mesocarp was packaged in glass, added the covering liquid, subjected to the heat treatment by soaking for 15 minutes in boiling water (± 95° C). In the experiment used the liquid medium composed of water, vinegar, sucrose, sodium chloride and condiments, which presented various concentrations corresponding to four treatments. Sensory analysis was employed a group of 33 untrained tasters, who through the hedonic scale structured seven points assessed the attributes color, aroma, flavor, texture and overall acceptance. No significant difference was observed among treatments. The attribute that received highest score was the color and the opposite occurred with aroma and flavor. All counts measured in sensory analysis obtained approval above 70% and the formulation with lower concentration of vinegar received higher grades.

Abstract:
This paper highlights the design challenges encountered during the project execution phase of HMPL’s Mundra - Bathinda Pipeline (MBPL), a crude import pipeline for feeding Guru Gobind Singh Refinery of HMEL at Bathinda in Punjab State. For the first time in India, a cross-country pipeline was planned through a terrain wherein section of more than 270 km length runs through the eastern fringe of Thar desert in the state of Rajasthan. MBPL routed in newer and exotic expanse of Great Indian Thar desert posed greater challenges in respect of pipeline routing, design and construction. The pipeline route across the bygone desert is characterized by severe climate and remote locations. The summer daytime temperature reaches upto 50 deg C while in winter it drops to near freezing temperature. Sand dunes at some places along the route were more than 40 m high and were of different kinds and separated by inter-dunal sandy plains of varying stretches.
