Abstract:
Abstract Aurora kinase inhibitors such as alisertib can destabilize MYC-family oncoproteins and have demonstrated compelling anti-tumor efficacy. In this study, we report 6K465, a novel pyrimidine-based Aurora A (AURKA) inhibitor that reduces levels of c-MYC and N-MYC oncoproteins more potently than alisertib. In an analysis of the antiproliferative effect of 6K465, the sensitivities of small cell lung cancer (SCLC) and breast cancer (BC) cell lines to 6K465 were strongly associated with the protein levels of c-MYC and/or N-MYC. We also report DBPR728, an acyl-based prodrug of 6K465 bearing fewer hydrogen-bond donors that exhibited 10-fold improved oral bioavailability. DBPR728 induced durable tumor regression of c-MYC- and/or N-MYC- overexpressing xenografts including SCLC, triple-negative breast cancer (TNBC), hepatocellular carcinoma and medulloblastoma using a 5-on-2-off or once-a-week dosing regimen on a 21-day cycle. A single oral dose of DBPR728 at 300 mg/kg induced c-MYC reduction and cell apoptosis in the tumor xenografts for more than 7 days. The inhibitory effect of DBPR728 at a reduced dosing frequency was attributed to its uniquely high tumor/plasma ratio (3.6-fold within 7 days) and the long tumor half-life of active moiety 6K465. Furthermore, DBPR728 was found to synergize with the mTOR inhibitor everolimus to suppress c-MYC- or N-MYC- driven SCLC. Collectively, these results suggest DBPR728 has the potential to treat cancers overexpressing c-MYC- and/or N-MYC. Abstract Upregulation of Aurora kinases has been associated with increased tumor progression, and thus they are appealing targets for the development of anti-cancer therapies. In addition to have a critical role in cell cycle regulation, Aurora kinases have been shown to stabilize MYC-family oncoproteins for the maintenance of malignancy. Aurora kinase inhibitors such as Alisertib has demonstrated compelling anti-tumor efficacies; however, the reported side effects including serious haematological disturbances have limited its risk-benefit ratio in clinical use. In this study we discovered a novel pyrimidine-based Aurora kinase inhibitor compound A that degraded MYC- and MYCN- oncoproteins with better potency than Alisertib. The acyl-based prodrug design leads to the discovery of compound B which was able to regress MYC- or MYCN- overexpressing tumor xenografts including small cell lung cancer, neuroblastoma, hepatocellular carcinoma and medulloblastoma using a once-a-week (QW) oral dosing regimen. Pharmacokinetic studies revealed that the tumor/plasma ratio of compound B was about 20 at 24 h post drug administration, and the active compound remained detectable in the tumors after 7 days. No significant haematological or liver/kidney biochemical aberration was observed in mice treated with up to 500 mg/kg of DBPR728 on a QW dosing regimen in a 21-day cycle. The unique pharmacokinetic and molecular properties of compound B hold potential clinical promise for treating MYC- and MYCN- amplified tumors with manageable on-target haematological adverse effects caused by Aurora kinase inhibition. Abstract Recombinant monoclonal antibodies (rAbs) are superior tools for cancer biology research as they offer distinct advantages over standard polyclonal and hybridoma-generated monoclonal antibody reagents. Recombinant technology produces antibodies that can be defined at the primary sequence level and are characterized by consistent performance and supply. These rAb features are in stark contrast to traditional polyclonal and hybridoma-generated monoclonal antibodies, whose lack of dependability has been widely documented as a primary reason for data irreproducibility plaguing biomedical research. In an effort to respond to this challenge, GeneTex has established a recombinant antibody production platform combined with more rigorous validation protocols. The goal is to develop reliable recombinant rabbit monoclonal antibodies for reproducible performance in various experimental applications utilized routinely by cancer biologists. GeneTex’s rAb production protocol is based on the approach described by Starkie et al. (2016). This involves a multi-parameter fluorescence-activated single cell sorting (FACS)-based methodology to identify and isolate antigen-specific IgG+ memory B cells obtained from an immunized rabbit. The heavy and light chain variable region genes from selected cells are PCR-amplified and cloned into plasmids to produce full-length heavy and light chains for a single IgG. These constructs are subsequently introduced together into mammalian cells for expression, meaning that natural pairing of the chains is maintained. This manufacturing strategy allows application-specific testing (e.g., for western blot (WB), immunohistochemistry (IHC), and immunocytochemistry (ICC/IF), etc.) of clones during the antibody production process. Validation of antibodies is performed inhouse and, when possible, in relationships with academic or industry/pharma institutions where ideal samples and reagents are frequently more available. This recombinant monoclonal antibody production platform has created a series of well-validated antibodies against many important targets for cancer biology research. This includes reagents to study tumor immunobiology (e.g., PD-L1), hormone receptors (e.g., ER alpha and androgen receptor variants like ARV7), oncogenic RAS protein mutants (e.g., RAS G12D), transcription factors (e.g., NRF2), and epithelial-mesenchymal transition (EMT) (e.g., E-cadherin), among many others. Extensive validation for multiple applications was performed, with knockdown or CRISPR-based knockout cell lysates employed when possible to establish specificity. In summary, GeneTex is utilizing a FACS-based recombinant rabbit monoclonal antibody production approach to generate and thoroughly validate antibody reagents that will hopefully facilitate reproducible cancer biology research. Background: Estrogen receptors (ER) and progesterone receptors (PR) have important prognostic value in endometrial cancer, but there is no recognized positive immunohistochemical threshold for predicting the recurrence of early low-risk endometrial cancer. The purpose of this study was to clarify the optimal positive thresholds of the immunohistochemical parameters the ER and PR in early low-risk endometrial cancer. Methods: A total of 332 patients with stage IA endometrial cancer were enrolled from the First Affiliated Hospital of Chongqing Medical University and People’s Hospital of Sha ping ba District in Chongqing between January 2013 and December 2018. First, univariate and multivariate Cox regression analyses were used to analyze the correlation between various clinical factors and the prognosis of early low-risk endometrial cancer. Then, the receiver operating characteristic curve (ROC curve) and Youden index were used to determine the positive thresholds of ER and PR. Results: The positive thresholds of ER and PR for predicting the recurrence of early low-risk endometrial cancer were 12% and 8%, respectively. Multivariate analysis showed that ER ( P = 0.004), PR ( P = 0.026), and p53 ( P = 0.021) were risk factors for the prognosis of patients with early low-risk endometrial cancer. The recurrence-free survival and the overall survival in the low ER group and PR group were much lower than those in the high ER group and PR group ( P < 0.001 of all). Conclusions: ER and PR positive thresholds of 12% and 8%, respectively, are the most suitable for predicting the recurrence of early low-risk endometrial cancer.

Brugerne tages tilsyne- ladende alvorligt, i modsætning til den kritiske teori (Frankfurterskolen m.v.), der i debatten om receptionsforskningen er blevet beskyldt for at være formynderisk: Den tager stilling til æstetisk og indholdsmæssig kvalitet på brugernes vegne. I denne artikel forsøger Ib Bondebjerg at fastholde en del næsten glemte, men fundamentale nuancer (ikke mindst mht. tekstens status i forhold til oplevelsen) i både den kritiske teori, semiotikken og dele af receptions- forskningen. Hermed bidrages til den diskussion om hvem der skal fast- lægge den kulturelle og æstetiske værdi i et medieprodukt som tages op af både Jensen, Schrøder og Skovmand i dette nummer. Indirekte viser Bondebjerg, at der sker en afpolitisering af medierne, hvis de alene eksisterer i en uproblematiseret relation mellem afsender og modtager. Populært TV konstrueres jo netop så det fremtræder lyst- og spændings- fyldt og kommer herved til at virke som et passivt spejl for modtagerne. Hvis kunsten forråder det smertefulde og tabuiserede, bliver den ufarlig. Mediernes eneste formål bliver da at igangsætte et på forhånd kalkuleret ping-pong-spil mellem den enkeltes erfaringer, oplevelser og fantasier. Heroverfor stiller Bondebjerg den demokratiske radikalisme hvor æstetik- ken udgør et kampområde. Kunsten er med andre ord en politisk agent, hvor både afsenderen, modtageren og samfundet selv aktivt er med i forandringsprocessen. - "Perspektivet er at gøre modtagerne til kvalifi- cerede producenter i videste forstand".

Abstract:
Abstract Background NTMLD is a rare mycobacterial infection characterized by worsening lung function and increased healthcare resource burden. Many patients with NTMLD have comorbid respiratory diseases such as COPD. Management of NTMLD in patients with COPD may not get adequate attention as clinical management of COPD is often prioritized. There is a lack of data quantifying the incremental burden of NTMLD among patients with COPD. We assessed the incremental burden of NTMLD in patients with comorbid COPD by comparing their healthcare resource utilization (HCRU), in terms of hospitalizations and emergency room (ER) visits, to matched COPD patients without NTMLD. Methods A retrospective cohort study was conducted using the US Medicare claims database (2010-2017). COPD patients with NTMLD were matched 1:3 to COPD patients without NTMLD (controls) by age and gender. HCRU was assessed in terms of hospitalizations (all-cause, respiratory related and COPD related) as well as ER visits (with and without subsequent hospitalization) over a 12-month follow-up period. Incremental burden of NTMLD was assessed by comparing the HCRU between COPD patients with NTMLD and controls using univariate and multivariate analyses adjusting for baseline comorbidities. Results A total of 4,010 COPD patients with NTMLD were matched to 12,030 controls. In univariate analyses, a significantly higher proportion of COPD patients with NTMLD had hospitalizations and ER visits compared to controls (Fig 1(A)); the number of hospitalizations and ER visits were also higher among patients with NTMLD (Fig 2(A)). This incremental burden was also reflected in subsequent multivariate analyses after adjusting for baseline comorbidities. As an example, respiratory-related hospitalizations in COPD patients with NTMLD had Odds Ratio of 2.5 (95%CI 2.2-2.7) and Incident Rate Ratio (IRR) of 2.3 (95%CI 2.1-2.4) compared to controls (Fig 1(B), 2(B)). Abstract:
Abstract Cyclin-dependent kinase 4/6 inhibitors (CDK4/6i) are approved for the treatment of advanced estrogen receptor-positive, HER2-negative (ER+ve) breast cancer when combined with endocrine therapy. However, the emergence of resistance to CDK4/6 inhibitors poses a significant clinical challenge. This study investigates the mechanisms underlying CDK4/6 inhibitor resistance and identifies a common occurrence of high replication stress in triple-negative breast cancer (TNBC) and CDK4/6 inhibitor-resistant ER+ve breast cancers. While ER+ve breast cancer patients have benefited from targeted therapy, TNBC patients, particularly those without germline BRCA1/2 or PALB2 mutations, lack effective treatment options. TNBC and CDK4/6 inhibitor-resistant ER+ve breast cancer cells exhibit an increased dependence on cell cycle arrest to repair replication stress-induced DNA damage. Failure to exit the cell cycle results in excessive DNA damage, thereby limiting their proliferative potential. Based on these findings, we hypothesize that dual inhibition of WEE1 and PKMYT1, two kinases involved in G2/M cell cycle transition and crucial for cell cycle arrest and DNA damage repair, could synergistically inhibit tumor growth. Although WEE1 and PKMYT1 share a common function in promoting G2/M cell cycle transition through CDK1 phosphorylation, their functions are not entirely redundant, suggesting that targeting both kinases may elicit a synergistic response. Recent clinical trials have shown resistance to WEE1-targeted monotherapies, emphasizing the need for combining WEE1 inhibitors with other agents, such as PKMYT1 inhibitors. We demonstrate the synergistic effects of WEE1 inhibitor (MK1775/AZD1775) and PKMYT1 inhibitor (RP6306) in a panel of TNBC and ER+ve breast cancer cells with acquired palbociclib resistance (PalboR). Combination treatment with both inhibitors leads to excessive unrepaired DNA damage, as indicated by increased γH2AX foci following 48 hours of combination treatment, whereas single-agent treatments induce significantly lower levels of DNA damage. Furthermore, we observed the AZD1775/RP6306 combination decreases the expression of the DNA repair protein Rad51 in PalboR cells. To evaluate the clinical potential of dual WEE1 and PKMYT1 inhibition, we generated organoids from metastatic breast cancer lesions of patients who had progressed on CDK4/6 inhibitors for ex vivo studies. Our results demonstrate that dual inhibition of WEE1 and PKMYT1 is more effective than single-agent treatments in eliminating CDK4/6 inhibitor-resistant organoids. Additionally, using patient-derived xenograft mouse models from patients who had progressed on palbociclib and endocrine treatment, we show that the AZD1775/RP6306 combination exhibits superior tumor suppression effects compared to single agents. Overall, this study highlights the potential therapeutic benefits of dual inhibition of WEE1 and PKMYT1 in overcoming CDK4/6 inhibitor resistance in TNBC and ER+ve breast cancer patients. These findings provide a rationale for future clinical trials aimed at exploring the clinical utility of this combination therapy. Er Pai (“Two Amazing Tales”), the collective name of Chuke Paianjingqi (“Amazing Tales - First Series”) and Erke Paianjingqi (“Amazing Tales - Second Series”), is best of the representative novels in the China and is on a par with Feng Menglung’s (“Three Volumes of Words”). The author of “Two Amazing Tales”, Ling Mengchu, showed his admiration to Feng in the preface of “First Series” and implied that he wanted to borrow Feng’s writing technique. With totally 40 volumes . “Two Amazing Tales” are mainly based on these materiales: Taiping Guangji (“Anthology of Tales from Records of The Taiping Era”),,Yi Jian Zhi (“Chinese Supernatural stories”), Jian Deng Xin Hua (“New stories told while trimming the wick”), Jian Deng Yu Hua (“Other stories told while trimming the wick”) and other books. There have been many subjects on the research of “Two Amazing Tales”. The relationship between “Two Amazing Tales” and “Anthology of Tales from Records of the Taiping Era” was illustrated by Xu Yongbin in his “An investigation on the sources and relationship of ‘Two Amazing Tales’ and ’Anthology of Tales from Records of the Taiping Era’”. However, in addition to borrowing the content of “Anthology of Tales from Records of the Taiping Era”, Ling adopted a new perspective of his own and modified the story for the plot’s sake, so as to introduce new elements in the original theme, which gives a makeover to “Two Amazing Tales” and make it a work of his innovation. This research focuses on the rewriting skills that Ling utilized in his borrowing “Anthology of Tales from Records of the Taiping Era”, and intends to demonstrate the nature of rewriting in the translation literature with the discussion of Ling’s originality. The first part is to define “rewriting”, to introduce the background of “Two Amazing Tales” and “Three Volumes of Words” as well as the author’s life, and to present the development of Huaben (“script for story-telling”). And from the content, form, description, delay the passage of four areas to illustrate " Er Pai " rewrite " Taiping Guangji " the show features. In content,When " Er Pai " be written, the main consideration behind the many factors when compared with "Taiping" search recorded unofficial novels, " Er Pai " had more purposeful. There are stylistic differences in form and content of institutional differences, describing the modus operandi is able to see the evolution of novel techniques, as well as "Er Pa" is different from " Three Volumes of Words " For the theme of the deal, has the innovative ; The last part is the extension of the passage " Taiping Guangji " thinking and the main story, to show cultural heritage. The conclusion is to presents the real ideas of Ling from his selection of material. This research supposes to help the study of “Two Amazing Tales” or become a reference for those who study rewritten texts.

Zone expropriation, based on user fees concept, is an important methods to urban development. However, what is the benefit of the new community development under zone expropriation? This study applied the Net Present Value method (NPV) to calculate both direct and indirect profits on developing new community in Tai-Ping Hsin-Kuang Zone, Taichung County through zone expropriation. It is found that there is a direct profit increase in the net income of NT$889.35 million dollars, a positive NPV figure. It apparently indicates that zone expropriation in this case could not only achieve the goal of self-liquidation, but also have a surplus to pay other public expenditures. Nevertheless, there is an indirect benefit loss of NT$267.94 million dollars, that is, public expenditures needed for said development plan is far less than financial revenue can afford. Consequently, this decision has made the Taichung County Government in more financial difficulties. Besides, various factors including exploitation and construction costs, anticipated selling land price, land value increment tax, ratio of applying for cash compensation and the growth rate of population influence the profitability on new community development. In this case, it can be found that the increase tax revenue from the supposed growth of population is insufficient for handling additional expenditures. In addition, under the assumption of all lands can be successfully sold out on schedule, the higher ratio of applying for cash compensation gives advantage to the overall financial revenue. During the slump in the real estate market these years, anticipated land price sold should be carefully estimated to prevent the great loss from the downward trend.

In 2009, the asymmetric precipitation pattern of Typhoon Morakot associated with the southwest monsoon produced the extreme rainfall accumulation in 48 hrs over southern Taiwan and caused a large scale landslide at Er-Wan-Ping in Ali-Shan watershed. From aerial photo interpretation, there is no obvious formation of sediment deposition within the landslide boundary. Therefore, the large amount of sediment is assumed to still accumulate in the watershed. How to delineate the deposition area and the potential enlargement landslide area is an important issue for treatment work. In order to extract the sediment deposition area, the multi-temporal satellite imageries were applied to assess vegetation recovery rate at Er-Wan-Ping landslide. In addition, the 1-m resolution of digital elevation model (DEM) was used to extract gully path. Elevation and slope longitudinal profile through gully path were explored the potential enlarge landslide at the headwater area. According to the results, the potential map of secondary disaster at Er-Wan-Ping landslide would be delineated as a reference for landslide treatment. The results show that one year after landslide, the areas of well-growth vegetation recovery was gradually stable. The deposition area was verified using pre- and post- landslide DEM with the accuracy of 52.2%. The deposition area was misinterpreted due to the tree island effect, whereas the collapse area was misinterpreted as a result of artificial treatment. Most of extracted gully path shows a rather gentle slope profile which could be implied the stability of sediment. However, the site where the slope changes steeply could indicate the potential enlargement of landslide at the headward area. The site should be strengthened the treatment of disaster prevention.

Chinese composer Zhao, Ji- Ping was born and raised during the golden years of Er-hu. During these decades, a vast amount of new pieces were composed for and new techniques were applied to this traditional instrument. The music of Zhao is largely inspired by Chinese folk music and shows a great variety, including symphony, symphonic poem, Chinese orchestra, concerto, chamber music, dance drama, and film scoring. Among his compositions, the film scores are frequently rearranged to instrumental pieces by composer himself. For example, the film score for Yellow Earth was adapted to Chinese orchestra, the music for TV drama Qiao's Grand Courtyard was adapted to symphonic pieces, and the film score for The True Hearted was adapted to Er-hu concerto. My research would first focus on the reason why Zhao did such adaption of The True Hearted, followed by the analysis and interpretation of this piece. The body of this interpretational research is mainly based on case and bibliography study, including periodical articles, concert reviews and interviews. These materials provide crucial clues for the approach of analysis and interpretation, which are supported by music narratives and extract of music scores. Under this process, I would explain the music concepts and characteristics of Zhao in detail; for example, on Zhao’s culture and humanity aspects, the unique music atmosphere and dexterous instrumentation. Furthermore, I would connect the analysis of music components to the scenes of movie. The closer examination of the two would unfold a clearer map of the Erhu concerto adaption, which would contribute to an overall understanding, moreover, a more convincing and comprehensive interpretation of this piece.

Abstract Located in the sub-tropical area, Taiwan features hot and humid climate, long and intensified rainy season, and seasonally uneven-distributed rainfall. Due to such climate conditions, run-off water on the road pavement, if not drained timely, causes lingering water particle to reduces the anti-skid capacity of the road and to greatly reduces driving safety. In view of these, the Directorate General of Highways (DGH) ,Ministry of Transportation and Communications (MOTC) in October 1999 introduced pervious asphalt, with the hope of improving highway safety. The MOTC-DGH’s Material Lab. conducted four separate observations and assessments of the road ’s effectiveness within the first, third, sixth, and twelfth month after pavement of pervious asphalt complete. The assessment includes road flatness, damage survey, vehicle tracks, anti-skid values, traffic noise, flexibility, drainage capacity (including in-lab and in situ drainage tests). This study also attempts to develop a in-situ pressure-style drainage measuring device, which uses air and nitrogen as permeable media. Results are compared with those obtained at the MOTC-DGH’s Material Lab. The findings suggest that use of air as permeability media can expedite the experiment and speed up the procedures. Use of gas as permeability media are also expected to minimizes the effects of capillary and gravity effects while using water as the permeability media. The permeability coefficient must be converted to that with water as permeability media at 15℃(K15℃). Compared to the permeability coefficient obtained based on the theory of Radial Parallel Flow (K15=2㎝/sec),which is simillar to the formular of puming tests, the permeability coefficient obtained based on the theory of Hemispherical Flow (K15=1×10-2㎝/sec) is closer to that obtained at MOTC-DGH’s Material Lab. (K15=1.4×10-2㎝/sec). Thus, field air pressure-style device, after some further studies and adjustments, could be another way to obtain field permeability coefficient. The average values of the permeability coefficient obtained in this study and that obtained at MOTC-DGH’s Material Lab. both exceed the standard set in Japan (K15=1×10-2cm/sec). Service industry and commercial activities in Taiwan are flourishing. The idea of commercial hub is gradually being accepted. Through years of effort by the government, several towns and cities are working hard to establish their own commercial hubs. How to create or improve the image of a commercial hub is becoming an important issue. The study focuses on the rebuilding projects of retail centers in rural areas in Taiwan. We investigate the cooperation among public sector (central and local governments), private sector (businesses and residents), and local organizations and how the political power, capital and human resource work together. We compare two such projects: Main Street project from the USA in 1980’s and revival project from Japan in 1960’s. We create a model of how the different organizations work together and come up with some guidelines. Then, we use the model and guidelines to examine the projects in Taiwan. Through face-to-face interviews, we come to a clear understanding of the Ku-guan commercial hub revival project. We also present our suggestions on how to integrate different mechanisms. Finally, we analyze the differences between Ku-guan and Wu-lai projects and offer solutions to sustainable development. In conclusion, we found: 1. It is important to fight for more resources on local level. 2. It is essential to have coordination between public sector and local organizations. 3. It is necessary to create an integrating mechanism. 4. It is vital for the local organizations to have good decision-making abilities.

The An Ping Wei Zhen Tang is the first temple which consecrating the Kuang Tse Tsun Wang in An Ping, Tainan. The main god of this temple are Kuang Tse Tsun Wang, Miao Ying Sian Fei, and Shih San Tai Bao. The administrator of An Ping Wei Zhen Tang decide to create the temple since 1997, and the opening ceremony was the 24th day of 12th lunar month. In An-Ping area, there are many temples built in 100 years ago.Even the temple only have 20 years history, the young temple try to follow the traditional concepts and join the cultural creative idea, developed the particular style of itself. The research is aim to design a special symbol named Wo-Jyuan which matches the temple’s traditional value. And this paper also discuss the relationship between the Coming-of-age Ceremony and the Kuang Tse Tsun Wang. The Wo-Jyuan is the most important connection of the god and the ceremony. According to the reference, this research point out the kernel concept, and create the Wo-Jyuan with auspicious symbol, try to analyze the original design, traditional culture, and the differences of them.
