Abstract:
Dietary supplementation with inorganic nitrate has been shown to lower the oxygen cost of submaximal exercise and to improve performance during continuous endurance exercise. The objectives of this thesis were: 1) to improve understanding of the supplementation procedures in which nitrate is most likely to benefit exercise physiology and performance, and 2) to explore the ergogenic potential of dietary nitrate in intermittent exercise. Recreationally-active adult participants volunteered to participate in the original investigations presented in this thesis. These participants underwent various oral nitrate supplementation regimes and subsequently provided venous blood samples for the determination of plasma nitrate concentration and plasma nitrite concentration, and completed different exercise tests to assess the potential for nitrate supplementation to improve physiological responses during exercise and exercise performance. Chapter 4: Following the ingestion of 70, 140 and 280 mL concentrated nitrate-rich beetroot juice (containing 4.2, 8.4 and 16.8 mmol nitrate, respectively) plasma nitrite concentration increased dose-dependently, with peak changes occurring at ~2-3 h post ingestion. The oxygen cost of submaximal exercise and exercise tolerance were also influenced dose-dependently 2.5 h post acute BR ingestion when compared to a nitrate-depleted placebo. Specifically, 8.4 mmol and 16.8 mmol lowered the oxygen cost of moderate-intensity exercise by 1.7% (P = 0.06) and 3.0% (P < 0.05), respectively. Exercise tolerance during severe-intensity exercise was significantly improved after ingestion of 8.4 mmol (+14%; P < 0.05), with no further improvement evident following ingestion of 16.8 mmol (+12%; P < 0.05). 4.2 mmol nitrate did not significantly lower the oxygen cost of submaximal exercise or improve exercise tolerance. Chapter 5: Plasma nitrite concentration was dose-dependently elevated 2 h and after 7 days and ~4 weeks of supplementation with 3 mmol and 6 mmol nitrate. Compared to pre-supplementation baseline, and placebo, moderate-intensity exercise oxygen uptake was not lowered with 3 mmol nitrate acutely (2 h) or after ingesting it daily up to ~4 weeks. In contrast, ingestion of 6 mmol nitrate significantly lowered submaximal exercise oxygen uptake by 3% after 2 h (P = 0.06), 7 days and ~4 weeks (both P < 0.05) of supplementation. Another interesting observation was that the reduction in submaximal exercise oxygen uptake after ~4 weeks of supplementation with 6 mmol nitrate was preserved 24 h after consumption of the final nitrate dose despite plasma nitrite concentration having returned to baseline. Chapter 6: In recreational team sport players, 28 mmol dietary nitrate administered over ~30 h improved performance in the Yo-Yo Intermittent Recovery Level 1 (Yo-Yo IR1) test (a test that mimics the high-intensity intermittent exercise bouts typical of team sport games) by 4.2%, compared to placebo (nitrate: 1704 m vs. placebo: 1636 m). The decline (or ‘utilisation’) of plasma nitrite from pre-exercise to post-exercise was greater with BR compared to placebo, and the magnitude of this decline in plasma nitrite was positively correlated with the improvement in Yo-Yo IR1 performance. Chapter 7: Supplementation with dietary nitrate for 3-5 days (8.2 mmol nitrate per day) significantly improved performance during 24 x 6-s all-out cycling sprints interspersed with 24 s recovery, but not during 7 x 30-s all-out sprints interspersed with 240 s of recovery or 6 x 60-s self-paced maximal efforts interspersed with 60 s of recovery, in a group of recreational team sport players. The novel findings presented in this thesis suggest that the supplementation procedure, in particular the nitrate dose administered, should be carefully considered by individuals wishing to elicit ergogenic effects following dietary nitrate supplementation. Specifically, the findings suggest that a low dose (≤4.2 mmol) of nitrate is not sufficient to acutely improve exercise tolerance, and that submaximal exercise oxygen uptake is not lowered following acute or prolonged (~30 day) supplementation with a low nitrate dose. In contrast, results show that higher doses (6-8.4 mmol) of nitrate might be sufficient to acutely lower the oxygen cost of submaximal exercise and improve exercise tolerance. The present thesis also provides evidence to indicate that dietary nitrate might hold potential as an ergogenic aid for individuals participating in intermittent exercise involving brief bouts of high-intensity exercise interspersed with short recovery periods, such as team sport players. These findings are of importance as they will help inform supplementation procedures in future studies assessing the ergogenic efficacy of nitrate supplementation. Moreover, they suggest that nitrate supplementation has the potential to improve performance during intermittent exercise as well as continuous endurance exercise.

Abstract:
The congested fixture schedules in elite soccer leagues around the world has bought the issue of recovery between games and subsequent performance to the fore in soccer related research. Studies have described the time-course of recovery for numerous biochemical and physiological measures of performance, fatigue and recovery from match-play. However, the research also suggests that there is individual variation in the external load both between players and between matches. The external load measured as distance in match-play has been shown to vary by ~30% between games. However it is the internal training load that will determine the magnitude of the physiological responses on an individual basis. Therefore the major aim of this thesis was to examine the dose-response relationships between measures of training load and the physiological and biochemical responses used as markers of recovery from match-play. The thesis also assessed the relationships between these proposed markers of recovery and soccer specific performance. In meeting the aims of the thesis a number of preliminary studies were conducted. The study in section 3 assesses the extent of fixture congestion in the English Premier League. The results showed over 30% of games for the most successful teams are played with 3 days recovery time, justifying the need for investigating recovery from soccer match-play. Given the variation in soccer match-play section 4 examines the reliability and validity of the modified BEAST90 soccer simulation. A measure of performance with less variance would allow changes in soccer specific performance to be identified with greater certainty in section 7. Section 5 assesses the influence of intermittent exercise on the blood lactate response. Given that the new iTRIMP method of measuring internal training load weights exertion with the blood lactate response it was important to assess the influence exercise mode may have on the calculation of internal training load. The results showed that at higher intensities intermittent exercise produced significantly higher blood lactate responses. Section 6 assesses the dose-response relationships between training and fitness using numerous measures of internal training load over a 6 week training period. The results showed only the iTRIMP method showed a significant relationship with changes in fitness. Section 7 assesses the dose-response relationships between exertion in soccer match-play and various physiological measures of fatigue and recovery. The relationships between these measures and changes in soccer specific performance were also assessed. Finally the internal and external load were integrated and the relationships of this ratio assessed with measures of fitness and performance. The results showed that changes in any of the physiological and biochemical measures used to assess recovery did not relate to changes in performance with the exception of testosterone which showed significant positive relationships with changes in distance covered from the 1st trial of the modified BEAST protocol to 2nd. Testosterone also was the only measure to show a significant relationship during the recovery period with any measure of training load (sRPE). Finally, the novel findings of this thesis is the relationships between the integrated ratio’s of internal and external training load with measures of aerobic fitness is also presented in section 7. The studies provided in this thesis have made a major contribution in demonstrating how data that is routinely collected at elite levels of soccer can be used more appropriately. It has also shown limitations of some the methods currently employed to measure training load. Furthermore changes in many of the markers used to assess recovery of soccer players do not seem to relate to changes in soccer specific performance. This may point to a change in paradigm which is required in both research and practice.

Abstract:
Currently, the World Anti-Doping Agency (WADA) permits asthmatic athletes to use inhaled salbutamol (SAL) to help attenuate compromised lung function during exercise. Although the majority of previous research shows no benefit in non-asthmatic athletes, there lacks an examination of the dose-response effect of SAL on performance using a sport-specific evaluation. Additionally, there lacks a description of how dose affects the concentration of SAL in the urine (cSAL). We hypothesized that salbutamol would have no effect on performance in non-asthmatic athletes and that cSAL would be affected by dose and be highly variable. Three projects were completed. Study 1 established the typical error and reliability of time-trial performance using the Velotron cycle ergometer. Highly trained, male cyclists performed three 20-km time-trials (TT) demonstrating the test to be highly reliable with low coefficients of variance for power and time (1.8-2.0% and 0.8-1.0% respectively). In Study 2, lung function was positively affected by SAL and urine analysis revealed a dose-response relationship with cSAL while at rest, up to a dose 800μg. Peak values were observed at 60min post-inhalation and cSAL was highly variable at each time point. Although several samples approached the WADA limit of 1000 ngml⁻¹, none exceeded this value. Using doses of 200μg, 400μg, and 800μg, Study 3 revealed no effects of SAL on time-trial performance or physiological measures over placebo. Additionally, athlete perception of leg and breathing effort was unaffected across conditions. Similar to Study 2, cSAL was related to dose and highly variable, with no samples resulting in a doping violation. SG was found to be significantly related to cSAL and when corrected to a dehydrated state, several samples exceeded the WADA limit. In summary, these findings allow us to accept the hypothesis that acute inhalation of SAL lacks ergogenic properties in non-asthmatic athletes and does not affect ventilation or metabolic parameters during exercise. Additionally, inhaled SAL does not appear to alter athlete perceptions of effort. The findings further suggest that urine samples will generally fall below the WADA limit following therapeutic doses of SAL, although this may be affected by hydration.

A radioatividade natural presente em solos, rochas e materiais de construção, devida ao 40K e às séries radioativas do 232Th e 238U é a principal contribuição à exposição externa aos seres humanos. Neste trabalho, determinou-se as concentrações de atividade de 226Ra (da série do 238U), 232Th e 40K presentes em 50 amostras de tintas látex de cor branca comercializadas no Brasil, especificamente, 15 do tipo econômico, 15 do tipo standard, 20 do tipo premium e em uma amostra de dióxido de titânio. As amostras foram seladas e armazenadas por um período mínimo de 30 dias para se alcançar o equilíbrio radioativo secular nas séries do 238U e do 232Th e medidas pela técnica analítica de espectrometria gama de alta resolução. As concentrações de atividade foram calculadas utilizando-se as médias ponderadas pelas incertezas do 214Pb e 214Bi para o 226Ra e médias ponderadas pelas incertezas do 228Ac, 212Pb e 212Bi para o 232Th. A concentração de atividade do 40K foi determinada pela sua transição única de 1460,8 keV. Fatores de autoatenuação gama foram calculados e utilizados para correção da concentração de atividade das amostras com densidade maior que 1,0 g.cm-3. Os índices radiológicos equivalente em rádio (Raeq), índice de concentração de atividade (Iγ), índice de risco à exposição gama interna (Hin), o índice de risco à exposição gama externa (Hex) e a taxa de dose (D) e dose efetiva anual (Def) foram calculados a partir das concentrações de atividade do 226Ra, 232Th e 40K. As concentrações de atividade de 226Ra das tintas variaram entre valores abaixo da atividade mínima detectável e 38,7 Bq.kg-1, as de 232Th variaram entre valores abaixo da atividade mínima detectável e 101,2 Bq.kg-1 e as de 40K variaram entre valores abaixo da atividade mínima detectável e 256 Bq.kg-1. O Raeq variou entre 1,41 Bq.kg-1 e 203 Bq.kg-1, o Iγ variou entre 0,0047 e 0,720, o Hin variou entre 0,0076 e 0,653 e o Hex variou entre 0,0038 e 0,549. A taxa de dose variou de 0,170 nGy.h-1 a 21,3 nGy.h-1 e a dose efetiva anual variou entre 0,83 μSv.a-1 e 104,2 μSv.a-1. Estes resultados mostram que as concentrações de atividades das tintas utilizadas neste estudo estão abaixo dos limites recomendados por Hassan et al. para Raeq (370 Bq.kg-1), pela Comissão Européia para o Iγ (limite de 2 para materiais superficiais) e pela Organização para Cooperação Econômica e Desenvolvimento para Hin e para Hex (ambos com limite de 1), para todas as 50 amostras estudadas, mostrando assim a segurança destas tintas com relação a proteção radiológica.

Abstract:
ABSTRACT Occupational exposure to wood dust and biohazards associated with wood dust (endotoxins, (1->3)-b-D-glucans, Gram (-)ve bacteria and fungi), their correlation to respiratory function, and symptoms among woodworkers have been investigated in the present study. Wood dust, endotoxins, and allergenic fungi are the main hazards found in woodworking environments. Relatively very few studies have been done on wood dust exposure. The present study was designed to comprehensively investigate the health effects of wood dust exposure, and in particular provide new information regarding: Exposure to (1->3)-b-D-glucans in an occupational environment; Levels of exposure to wood dust and biohazards associated with wood dust in different woodworking environments; Correlations among personal exposures, especially correlations between (1->3)-b-D-glucans and fungi exposures, and endotoxins and Gram (-)ve bacteria exposures; Effects of personal exposure to biohazards on lung function; Effects of personal exposure to biohazards on work-related symptoms; and Determinants of inhalable exposures (provide which factors in the environment influence the personal inhalable exposures). Workers at four different woodworking processes; two logging sites, four sawmills, one major woodchipping operation and five joineries situated in the state of New South Wales in Australia were studied for personal exposure to inhalable dust (n=182) and respirable dust (n=81), fungi (n=120), Gram (-)ve bacteria (n=120), inhalable endotoxin (n=160), respirable endotoxin (n=79), inhalable (1->3)-b-D-glucan (n=105), and respirable (1->3)-b-D-glucan (n=62). The workers (n=168) were also tested for lung function. A questionnaire study (n=195) was carried out to determine the prevalence of work-related symptoms. The geometric mean inhalable exposure at logging sites was 0.56 mg/m3 (n=7), sawmills 1.59 mg/m3 (n=93), the woodchipping mill 1.86 mg/m3 (n=9) and joineries 3.68 mg/m3 (n=66). Overall, sixty two percent of the exposures exceeded the current standards. Among joineries, 95% of the hardwood exposures and 35% of the softwood exposures were above the relevant standards. Compared with green mills, the percentage of samples, which exceeded the hardwood standard was high for dry mills (70% in dry mills, 50% in green mills). The respirable dust exposures were high at the joineries compared with the other worksites. Exposure levels to fungi at logging sites and sawmills were in the range 103-104 cfu/m3, woodchipping 103-105 cfu/m3 and joineries 102-104 cfu/m3. The predominant fungi found at sawmills were Penicillium spp. High exposure levels of Aureobasidium pullulans were also found at two sawmills. At the woodchipping mill the predominant species were Aspergillus fumigatus, Penicillium spp., and Paecilomyces spp. The sawmills, which employed kiln drying processes, had lower exposure levels of fungi compared with the green mills. Those workplaces which had efficient dust control systems showed less exposure to fungi and bacteria. Although mean endotoxin levels were lower than the suggested threshold value of 20 ng/m3, some personal exposures at sawmills and joineries exceeded the threshold limit value. The mean inhalable (1->3)-b-D-glucan level at the woodchipping mill was 2.32 ng/m3, at sawmills 1.37 ng/m3, at logging sites 2.02 ng/m3, and at joineries 0.43 ng/m3. For the respirable size fraction, mean endotoxin and mean (1->3)-b-D-glucan concentrations were much lower, being similar to observed dust concentrations. Significant correlations were found between mean inhalable endotoxin and Gram (-)ve bacteria levels (p<0.0001), and mean airborne inhalable (1->3)-b-D-glucan and fungi levels (p=0.0003). The correlations between mean respirable endotoxin levels vs Gram (-)ve bacteria exposure levels (p=0.005), and respirable (1->3)-b-D-glucan exposure levels vs total fungi levels (p=0.005) were also significant. Significant correlations were found between lung function and personal exposures. Multivariate analyses showed that the effect of all the personal exposures on cross-shift decrements in lung function was more prominent among sawmill and chip mill workers compared with joinery workers. Woodworkers had markedly high prevalence of cough, phlegm, chronic bronchitis, frequent headaches, throat and eye irritations, and nasal symptoms compared with controls. Among the woodworkers, smokers had a high prevalence of chronic bronchitis (20%) compared with non-smokers (10%). Some workers also reported a variety of allergy problems due to exposure to various types of wood dust. Both joinery workers and sawmill and chip mill workers revealed significant correlations between work-related symptoms and personal exposures. Chronic bronchitis was significantly correlated with personal exposure to wood dust, endotoxin, (1->3)-b-D-glucan, fungi, and Gram (-)ve bacteria among joinery workers. Whereas among sawmill workers chronic bronchitis was significantly correlated with personal exposure to endotoxin, (1->3)-b-D-glucan, and fungi. Woodworkers showed significant positive correlations between percentage cross-shift change (decrease) in lung function and respiratory symptoms. Significant inverse correlations were also found among percentage predicted lung function and respiratory symptoms. The elevated inhalable dust exposures observed in this study can be explained by a combination of factors, including: lack of awareness of potential health effects of wood dust exposure among both management and workers, aging equipment, inadequate and ineffective dust extraction systems or usually none especially for hand held tools, poor maintenance of the ventilation system in some, non-segregation of dusty processes, dry sweeping, and the use of compressed air jets. The determinant-of-exposure analysis confirmed the field observations. The significant determinants of personal inhalable dust exposures (n=163) were found to be: local exhaust ventilation, job title, use of hand-held tools, cleaning method used, use of compressed air, and green or dry wood processed. Type of wood processed was not found to be statistically significant. A majority of workers (~90%) did not wear appropriate respirators approved for wood dust, while the workers who did wear them, used them on average less than 50% of the time. Workers should be protected by controlling dust at its source. When exposure to wood dust cannot be avoided, engineering controls should be supplemented with the use of appropriate personal protective equipment.

Wood dust, endotoxins, and allergenic fungi are the main hazards found in woodworking environments. Relatively very few studies have been done on wood dust exposure. The present study was designed to comprehensively investigate the health effects of wood dust exposure, and in particular provide new information regarding: Exposure to (1->3)-b-D-glucans in an occupational environment; Levels of exposure to wood dust and biohazards associated with wood dust in different woodworking environments; Correlations among personal exposures, especially correlations between (1->3)-b-D-glucans and fungi exposures, and endotoxins and Gram (-)ve bacteria exposures; Effects of personal exposure to biohazards on lung function; Effects of personal exposure to biohazards on work-related symptoms; and Determinants of inhalable exposures (provide which factors in the environment influence the personal inhalable exposures). Workers at four different woodworking processes; two logging sites, four sawmills, one major woodchipping operation and five joineries situated in the state of New South Wales in Australia were studied for personal exposure to inhalable dust (n=182) and respirable dust (n=81), fungi (n=120), Gram (-)ve bacteria (n=120), inhalable endotoxin (n=160), respirable endotoxin (n=79), inhalable (1->3)-b-D-glucan (n=105), and respirable (1->3)-b-D-glucan (n=62). The workers (n=168) were also tested for lung function. A questionnaire study (n=195) was carried out to determine the prevalence of work-related symptoms. The geometric mean inhalable exposure at logging sites was 0.56 mg/m3 (n=7), sawmills 1.59 mg/m3 (n=93), the woodchipping mill 1.86 mg/m3 (n=9) and joineries 3.68 mg/m3 (n=66). Overall, sixty two percent of the exposures exceeded the current standards. Among joineries, 95% of the hardwood exposures and 35% of the softwood exposures were above the relevant standards. Compared with green mills, the percentage of samples, which exceeded the hardwood standard was high for dry mills (70% in dry mills, 50% in green mills). The respirable dust exposures were high at the joineries compared with the other worksites. Exposure levels to fungi at logging sites and sawmills were in the range 103-104 cfu/m3, woodchipping 103-105 cfu/m3 and joineries 102-104 cfu/m3. The predominant fungi found at sawmills were Penicillium spp. High exposure levels of Aureobasidium pullulans were also found at two sawmills. At the woodchipping mill the predominant species were Aspergillus fumigatus, Penicillium spp., and Paecilomyces spp. The sawmills, which employed kiln drying processes, had lower exposure levels of fungi compared with the green mills. Those workplaces which had efficient dust control systems showed less exposure to fungi and bacteria. Although mean endotoxin levels were lower than the suggested threshold value of 20 ng/m3, some personal exposures at sawmills and joineries exceeded the threshold limit value. The mean inhalable (1->3)-b-D-glucan level at the woodchipping mill was 2.32 ng/m3, at sawmills 1.37 ng/m3, at logging sites 2.02 ng/m3, and at joineries 0.43 ng/m3. For the respirable size fraction, mean endotoxin and mean (1->3)-b-D-glucan concentrations were much lower, being similar to observed dust concentrations. Significant correlations were found between mean inhalable endotoxin and Gram (-)ve bacteria levels (p<0.0001), and mean airborne inhalable (1->3)-b-D-glucan and fungi levels (p=0.0003). The correlations between mean respirable endotoxin levels vs Gram (-)ve bacteria exposure levels (p=0.005), and respirable (1->3)-b-D-glucan exposure levels vs total fungi levels (p=0.005) were also significant. Significant correlations were found between lung function and personal exposures. Multivariate analyses showed that the effect of all the personal exposures on cross-shift decrements in lung function was more prominent among sawmill and chip mill workers compared with joinery workers. Woodworkers had markedly high prevalence of cough, phlegm, chronic bronchitis, frequent headaches, throat and eye irritations, and nasal symptoms compared with controls. Among the woodworkers, smokers had a high prevalence of chronic bronchitis (20%) compared with non-smokers (10%). Some workers also reported a variety of allergy problems due to exposure to various types of wood dust. Both joinery workers and sawmill and chip mill workers revealed significant correlations between work-related symptoms and personal exposures. Chronic bronchitis was significantly correlated with personal exposure to wood dust, endotoxin, (1->3)-b-D-glucan, fungi, and Gram (-)ve bacteria among joinery workers. Whereas among sawmill workers chronic bronchitis was significantly correlated with personal exposure to endotoxin, (1->3)-b-D-glucan, and fungi. Woodworkers showed significant positive correlations between percentage cross-shift change (decrease) in lung function and respiratory symptoms. Significant inverse correlations were also found among percentage predicted lung function and respiratory symptoms. The elevated inhalable dust exposures observed in this study can be explained by a combination of factors, including: lack of awareness of potential health effects of wood dust exposure among both management and workers, aging equipment, inadequate and ineffective dust extraction systems or usually none especially for hand held tools, poor maintenance of the ventilation system in some, non-segregation of dusty processes, dry sweeping, and the use of compressed air jets. The determinant-of-exposure analysis confirmed the field observations. The significant determinants of personal inhalable dust exposures (n=163) were found to be: local exhaust ventilation, job title, use of hand-held tools, cleaning method used, use of compressed air, and green or dry wood processed. Type of wood processed was not found to be statistically significant. A majority of workers (~90%) did not wear appropriate respirators approved for wood dust, while the workers who did wear them, used them on average less than 50% of the time. Workers should be protected by controlling dust at its source. When exposure to wood dust cannot be avoided, engineering controls should be supplemented with the use of appropriate personal protective equipment.

O Ipen/Cnen-SP possui um Reator de Pesquisa(IEA-R1) em operação desde 1957. Ele utiliza água leve como blindagem, moderador e como fluido refrigerante, o volume desta piscina é de 273m3. Até 1995 a operação do Reator era descontinua, ou seja, operava diariamente sendo desligado no final do dia, a uma potência de 2,0 MW. A partir daquele ano, após algumas modificações de segurança, o Reator passou a operar de forma continua, ou seja, de segunda-feira a quarta-feira sem ser desligado, totalizando 64 horas semanais. A potência também foi aumentando até 4,5 MW em 2012. Em virtude dessas alterações, a saber, operação contínua e do aumento da potência, as doses dos trabalhadores aumentaram e por isso foram realizados vários estudos para diminui-las. Estudos demonstraram que uma das principais limitações para operação de um reator em potência elevada, provém das radiações gama emitidas pelo sódio-24. Outros elementos como magnésio-27, Alumínio-28, Argônio-51, contribuem de forma considerável para a atividade da água da piscina. A introdução de uma camada de água quente em sua superfície, estável e isenta de elementos radioativos com 1,5m a 2m de espessura constituiria uma blindagem às radiações provenientes dos elementos radioativos dissolvidos na água. Estudos de otimização provaram que a instalação da camada quente não era necessária para o regime e potência atual de operação do Reator, pois outros procedimentos adotados eram mais eficazes. A partir desta decisão o serviço de Proteção Radiológica do Reator IEA-R1, montou um programa de avaliação das doses para certificar-se de que elas se mantinham em valores razoáveis baseados em princípios estabelecidos em normas nacionais e internacionais. O intuito deste trabalho é realizar uma análise das doses individuais dos IOE (Individuo Ocupacionalmente Expostos), considerando as mudanças no regime de operação do Reator e sugerir opções de proteção e segurança, viáveis em primeira instância, para reduzir as doses analisadas, visando se chegar aos níveis de referencia de 3 mSv/ano adotados pela instalação em apreço.

O sistema microPET/CT é um importante equipamento utilizado nas pesquisas de imagem diagnóstica em pequenos animais. O radiofármaco mais usado nesta tecnologia é o fluordeoxiglicose marcado com flúor-18. Este estudo tem como objetivo efetuar o controle radiológico no laboratório de pesquisa microPET/CT do Centro de Radiofarmácia do IPEN-CNEN/SP, de forma a satisfazer tanto as normas nacionais como as recomendações internacionais. O laboratório está classificado pela equipe de radioproteção da instalação como área supervisionada, nas quais embora não seja obrigatória a adoção de medidas específicas de proteção e segurança, devem ser submetidas reavaliações regulares das condições do ambiente de trabalho. Visando assegurar a proteção radiológica dos trabalhadores diretamente envolvidos no manuseio do equipamento, realizou-se o monitoramento do local de trabalho e a avaliação do controle de dose individual. Inicialmente foi feito o monitoramento pré-operacional, isto é, o levantamento radiométrico no laboratório. Além disso, mediu-se nível de radiação externa nas instalações do laboratório e suas adjacências, por meio da colocação de nove dosímetros termoluminescentes (TL) de CaSO4:Dy, em locais previamente selecionados. Os indivíduos ocupacionalmente expostos foram avaliados mensalmente por meio do uso de dosímetros TL posicionados no tórax e por medidas de corpo inteiro, tomadas a cada seis meses. O período do estudo foi de dois anos, com início em abril de 2014. Para o controle do microPET/CT realizou-se testes de desempenho de acordo com o protocolo padrão do equipamento e em conformidade com a norma desenvolvida pela força tarefa para estudos com PET em animais Animal PET Standard Task Force. O presente estudo permitiu demonstrar que os níveis de radiação das áreas (estimativas de dose ambiente e dose efetiva), assim como a blindagem do equipamento estão adequados de acordo com os limites da exposição ocupacional. Ressalta-se a importância de se seguir rigorosamente os princípios de radioproteção, já que se trata de pesquisas com fontes radioativas não seladas.

A Doença de Chagas é um dos maiores problemas de saúde pública na América do Sul causando um elevado prejuízo à população. A despeito dos inúmeros esforços para o seu controle, a doença não tem cura e apresenta problemas científicos ainda não esclarecidos. Considerando-se que vários pesquisadores têm usado a radiação ionizante para modificar protozoários ou propriedades imunológicas de biomoléculas, neste trabalho foram estudados aspectos da resposta imunológica induzida em camundongos, resistentes e suscetíveis ao T. cruzi, utilizando formas irradiadas deste parasita. Doses baixas de radiação preservaram a capacidade reprodutiva e de invasão celular. Animais resistentes e suscetíveis, imunizados com os parasitas tratados por radiação, produziram anticorpos específicos. Após o desafio, os animais apresentaram baixa parasitemia, com exceção dos grupos imunizados com parasitas que receberam apenas altas doses de radiação. A seleção de formas tripomastigotas foi obtida irradiando-se os parasitas com baixas doses, o que promoveu aprimoramento da qualidade da resposta imune, a exemplo do que se observa quando da utilização de complemento. Estes dados evidenciam a importância da seleção das formas tripomastigotas para a imunização contra o T. cruzi e apontam a radiação ionizante como alternativa para este fim, uma vez que quando a seleção é feita utilizando-se complemento, depara-se com a dificuldade de sua remoção, colocando em risco o processo de imunização por introduzir substancias estranhas ao organismo.
Chagas\'s disease is one of the major public health problems in South America, promoting high prejudice to the local population. Despite the massive efforts to control it, this disease has no cure and presents puzzling unsolved questions. Considering that many researchers have used ionizing radiation to modify protozoans or biomolecules, we investigated the immunological response aspects of susceptible and resistant mice using irradiated parasites. Low radiation doses preserved the reproductive and invasive capacities of the parasite. Both susceptible and resistant animals, after immunization with irradiated parasites produced specific antibodies. After a challenge, the animals presented low parasitaemia, excepting those immunized with the antigen irradiated with higher doses. Using low radiation doses, we were able to selectively isolate trypomastigotes, leading to an improvement in the quality of the immune response, as previously reported when performing complement system assays. These data highlight the importance of selecting trypomastigote forms for immunization against T. cruz; and point towards ionizing radiation as an alternative to achieve this selection, since when this procedure is performed using complement, the subsequent steps are impaired by the difficulties to remove this component from the system.

A radiação ionizante é amplamente utilizada em diversas áreas na indústria e na medicina. No campo da medicina, a radiação ionizante é utilizada tanto para fins terapêuticos como para fins diagnósticos, abrangendo assim um grande intervalo de doses de diferentes tipos de radiações. Para certificar que a finalidade da prática esteja sendo alcançada, são necessários estudos detalhados de detectores e dispositivos que atendam aos diferentes tipos de radiações. Neste trabalho foi realizado um estudo das características dosimétricas de amostras de BeO em feixes padronizados de radiodiagnóstico convencional, mamografia e tomografia computadorizada, utilizando as técnicas de termoluminescência (TL) e luminescência opticamente estimulada (OSL) e propondo um sistema adequado para sua utilização em dosimetria de feixes. Os principais resultados obtidos foram: alta sensibilidade à luz branca durante a irradiação, boa reprodutibilidade das respostas TL e OSL (coeficientes de variação inferiores a 5%), parâmetros cinéticos correspondentes aos valores obtidos em todos os métodos testados, sem fading dentro do período estudado para ambas as técnicas. As características dosimétricas obtidas neste trabalho mostram a possibilidade de aplicação das amostras de BeO em dosimetria da radiação X, considerando os intervalos de dose empregados, pelas técnicas de TL e OSL. Assim, as amostras de BeO apresentaram sua utilização potencial para dosimetria de feixes de radiodiagnóstico convencional, mamografia e tomografia computadorizada.

A irradiação de alimentos é um meio de preservação de alimentos onde se utiliza uma técnica de processamento que expõe os alimentos a uma radiação ionizante controlada de alta energia. O tratamento realizado com o uso das radiações ionizantes em alimentos tem muitas aplicações tecnologicamente e tecnicamente viáveis incluindo a capacidade de melhorar a segurança microbiológica, reduzir os níveis de bactérias patogénicas, inibir a germinação de tubérculos, aplicação fitossanitária, conservar alimentos armazenados ou a estabilidade de armazenamento e também é utilizada para aumentar a vida de prateleira de determinados produtos devido à redução da contaminação por microrganismos. Em função do aumento do comércio internacional de alimentos e das crescentes exigências regulatórias dos mercados consumidores, cada vez mais países importadores e exportadores têm demonstrando interesse na irradiação de alimentos e desenvolvido pesquisas na aplicação prática desta tecnologia e de métodos de detecção do tratamento. Inúmeras pesquisas foram realizadas mundialmente, resultando em eficientes protocolos para identificar quais alimentos foram ou não irradiados. Até então, o \"mito\" que alimentos irradiados não poderiam ser detectados e que não eram formados quaisquer produtos únicos da radiação foi substituído pelo conhecimento que muitas mudanças podem ocorrer nos alimentos irradiados e essas mudanças poderiam ser utilizadas como ferramentas para identificar essa tecnologia. O processamento por radiação resulta em formações de padrões característicos de hidrocarbonetos saturados, aldeídos, metil e etil ésteres e 2-alcilciclobutanonas, dependendo da composição do ácido graxo do lipídio que compõe o alimento. Com isso a proposta do trabalho foi levantar dados para comparar os efeitos das diferentes doses de radiação gama e de elétrons em alimentos que possuem gordura para verificar as possíveis alterações decorrentes do uso da irradiação, como a presença de 2-Alcilciclobutanonas e também, mostrar os principais equipamentos utilizados para a irradiação de alimentos e suas categorias, com o objetivo de informar o publico em geral.

The dose-effect relationship in psychotherapy has been examined extensively, but few studies have included session frequency as a component of psychotherapy "dose." Those studies that have examined the effects of session frequency have indicated that it may affect both the total amount of recovery and the speed of recovery. No studies were found examining the clinical significance of this construct in a naturalistic setting. The change trajectories of 16,003 clients were examined using multi-level modeling and including session frequency as a fixed effect. Of these clients, subgroups were identified that were scheduled approximately once a week or approximately once every two weeks. These groups were compared to each other for differences in speed of recovery and clinically significant change. Results indicated that more frequent therapy was associated with steeper recovery curves. When comparing groups scheduled once a week to those scheduled once every two weeks, more clinically significant gains were identified in those attending once a week, and more significant deterioration was identified in those attending once every two weeks. These findings are discussed in light of the existing literature and the implications for future psychotherapy research and clinical practice.

Abstract:
Type 2 diabetes has become one of the most common human diseases. The safety and efficacy of antidiabetic medicines is an important part of diabetes management in the community. This thesis investigates a number of factors that may potentially cause variability in response to the antidiabetic drug gliclazide. This information will help to improve the use of these medicines in diabetic patients. Patients have free access to, and commonly take, complementary medicines for a variety of reasons. Many take these in conjunction with conventional drugs without clear evidence of safety or risk of herb—drug interactions. St John’s wort is one of the most widely used herbal medicines in Western societies. The aim of the first clinical study in this thesis was to investigate potential pharmacokinetic (PK) and pharmacodynamic (PD) interactions between St John’s wort and gliclazide in healthy subjects with different cytochrome P450 2C9 (CYP2C9) and CYP2C19 genotypes.

No ensaio II foram alojados 567 pintos de corte de um dia de idade e avaliou-se o turnover do 13C no músculo peitoral e fígado. Todos os animais foram submetidos à dieta a base de arroz e farelo de soja do primeiro aos 21 dias de idade. No ensaio I foi verificado melhora no desempenho de 1 a 7 dias de idade nos tratamentos 100% e 120% para peso corporal e ganho de peso e de 1 a 21 dias de idade nos tratamentos 100%, 110%, 120% para ganho de peso e peso corporal. Observou-se melhora na conversão alimentar com os tratamentos 100%, 110%, 120% e 130%. The objectives of this dissertation include 1) to review athlete monitoring strategies and the physical performance demands of female soccer match-play and to provide practical application of athlete monitoring programs, 2) to examine individual and group relationship between training load (TL) and subjective recovery and stress state and neuromuscular performance, and 3) to investigate acute effects of match-play on neuromuscular and subjective recovery and stress state in National Collegiate Association Athlete (NCAA) division I collegiate female soccer. TL was assessed using 10Hz Global Navigation Satellite System units. Subjective recovery and stress state was measured using the Short Recovery and Stress Scale (SRSS) consisting of 8 subscales including Physical Performance Capability (PPC) Mental Performance Capability (MPC), Emotional Balance (EB), Overall Recovery (OR), Muscular Stress (MS), Lack of Activation (LA), Negative Emotional State (NES), and Overall Stress (OS). Neuromuscular performance was assessed using countermovement jump (CMJ) with a polyvinyl chloride pipe (CMJ0) and 20kgs bar (CMJ20). CMJ variables included body mass (BdM), jump height (JH), modified reactive strength index (RSI), peak force (PF), relative peak force (RPP), eccentric impulse (EI), concentric impulse (CI), peak power (PP), relative peak power (RPP), eccentric average peak power (EAP), and concentric average power (CAP). Results of this dissertation showed that 12 individual players demonstrated negative correlations between total distance and MPC (p≤0.05, r=-0.78 to -0.34, number of significant individual correlations [N]=3) and OR (p≤0.05, r=-0.91 to -0.08, N=3). Positive correlations were observed between MS and total distance among all individual players (p≤0.05, r=0.21 to 0.82, N=3) while the group correlations were moderate to large (p≤0.001, r=0.55). Results of this dissertation also demonstrated that significant moderate to large decreases were observed at 12 hours post-match in JH, RSI, CI, PP, RPP, and CPA in CMJ0 and CMJ20 (p

Abstract:
Aflatoxin B1 (AFB1), a secondary metabolite of Aspergillus flavus and Aspergillus parasiticus, is known for its potent carcinogenicity and immunosuppressive effects. It is also known that AFB1 toxicity appears in different degrees in different animal species and strains. The present study was performed to reveal the involvement of the hypothalamus-pituitary-adrenal gland (HPA) axis in the immunosuppressive effects of AFB1 on C57B/6 mice. Splenic lymphocy1es were assayed to investigate their phenotyping using flow cy1ometry, proliferative response against mitogen and allogenic lymphocy1es, cy1oly1ic cell activity, and IL-2 production. In addition, antibody-mediated immunocompetence was checked using sheep red blood cell (SRBC)-challenged animals by plaque-forming cell (PFC) assay and enzyme-linked immunosorbent assay (ELISA). Corticotropin releasing factor (CRF) in brain hypothalamus and cerebral cortex, plasma adrenocorticotropic hormone (ACTH), and corticosterone were determined by radioimmunoassay (RIA). Hypothalamic catecholamine and its metabolites were assayed by high-performance liquid chromatography (HPLC). The adrenalectomized animals and their respective control animals were used to evaluate corticosterone involvement in AFB1 immunosuppressive effects. A relatively higher dose was applied in the present study, compared to the previous studies that used different strains of mice. Immunosuppressive effects were observed in blastogenic response, IL-2 production, and primary antibody production of splenic cells. The amount of circulating anti-SRBC antibody was also affected. Decreases were observed in the helper-T cell and B cell percentage in phenotyping splenic lymphocyte. No significant changes were observed in natural killer cell activity, mixed lymphocyte response, brain biogenic amine concentrations, concentration of CRF in the hypothalamus, and those of ACTH and corticosterone in plasma. However, the expected effect of adrenalectomy to compensate for the immunosuppression of AFB1 was not observed. The results indicate that the HPA axis does not appear to have a major role in AFB1-induced immunotoxicity.

Abstract:
Recently developed potent and selective CDK4/6 inhibitors fall into two classes based on structure and toxicity profiles in clinical studies. One class, exemplified by palbociclib and ribociclib, exhibits neutropenia as a dose-limiting toxicity and requires discontinuous dosing. In contrast, the structurally distinct CDK4/6 inhibitor abemaciclib is dosed continuously, and has diarrhea and fatigue as dose-limiting toxicities. In preclinical models, palbociclib has been extensively studied and induces cell cycle inhibition in an RB-dependent manner. Thus far, abemaciclib has been less extensively evaluated. We found that abemaciclib cell cycle inhibitory activity is RB-dependent at clinically achievable concentrations. Abemaciclib elicited potent suppression of RB/E2F regulated genes associated with prognosis in ER-positive breast cancer. However, unlike palbociclib, at 250nM-1 mu M doses abemaciclib induced cell death in RB-deficient cell lines. This response was associated with a rapidlyinduced multi-vacuolar phenotype indicative of lysosomal membrane permeabilization that could be ameliorated with chloroquine. This event was not a reflection of inhibition of other CDK family members, but could be recapitulated with CBX4945 that inhibits casein and DYRK/HIPK kinases. To determine if these "off-target" features of abemaciclib were observed in vivo, mice harboring matched RB-positive and negative xenografts were treated with palbociclib and abemaciclib. In vivo, all of the apparent activity of abemaciclib was RB-dependent and strongly elicited suppression of cell cycle regulatory genes in a fashion markedly similar to palbociclib. Using gene expression data from cell lines and tumors treated with abemaciclib and palbociclib a composite signature of response to CDK4/6 inhibition was developed that included many genes that are individually required for tumor cell proliferation or viability. These data indicate that while abemaciclib and palbociclib can exert distinct biological and molecular effects, there are common gene expression features that could be broadly utilized in measuring the response to CDK4/6 inhibition.

Micro-irradiation systems are used to analyse the effect of ionizing radiation at the cellular and tissue level, targeting individual cells within a population with a controlled low dose. Cell survival experiments using micro-irradiation systems are limited by factors including: 1) the radiation attenuation and optical properties of the chosen cell dish substrate, 2) the registration of the cell dish before and after irradiation or between multiple imaging modalities and 3) the analysis of the cell or colony growth after irradiation. In this thesis, a set of tools have been developed to improve micro-irradiation experiments and to increase the accuracy of information provided by the cell survival data. The first contribution, the substrate cell dish evaluation, provides a set of characteristics defining the substrates used for micro-irradiation experiments based on minimal energy loss and optical clarity using unstained cell imaging. The second contribution was the development of a novel and low cost fiducial marking device for micro-irradiation experiments using an 808 nm laser and providing marks suitable for imaging with multiple modalities. The minimum focused spot diameter was calculated as 22.9 urn and the device was used to create fiducial marks with diameters ranging from 20 urn to 130 urn. The third contribution, the development of a cell counting methodology for use with a lens-free imaging device, has been shown to accurately count thousands of cells suitable for immediate analysis. Approximately 1000 cell colonies, containing 17 729 cells on 11 cell dishes were used for testing and training for automatic cell counting. Validation of the cell counting method showed that 76% and 89% of the cell colonies were counted within a ± 20% and ± 30% error of the ground truth, respectively. Further development of the fiducial marking device, by modifying the choice of laser and making it suitable for multiple types of cell dish substrates, would increase the applications of the device. Development of the cell counting methodology for different cells line, and for cells grown on multiple types of substrates, would make the system suitable for analysis of a wide variety of cell survival studies. The cell counting methodology, applied to the CyMap lens-free imaging device, has the potential to be an extremely useful and cost effective tool for cell survival studies.

Resumo: Panicum maximun está entre as plantas daninhas mais importantes nas culturas tropicais, justificando plenamente a necessidade de controlá-la, o que é feito basicamente com o emprego de herbicidas. Diante disso, objetivou-se avaliar o efeito de subdoses de glyphosate nas trocas gasosas, crescimento e morfologia de P. maximun. O experimento foi realizado em casa-de-vegetação, sendo as plantas cultivadas em vasos, em delineamento inteiramente casualizado, com quatro repetições. Os tratamentos experimentais foram constituídos por uma testemunha (sem aplicação do produto) e nove doses (3,78; 8,10; 16,64; 33,48; 67,70; 135,00; 270,00; 540,00 e 1080,00 g e.a. ha-1). Foram avaliados os parâmetros fisiológicos: fotossíntese líquida, condutância estomática, transpiração, temperatura foliar, concentração interna de CO2. Também foram avaliados o número de perfilhos e intoxicação das plantas e, ao final do experimento determinou-se o acúmulo de massa seca da parte aérea e área foliar. Os dados obtidos foram submetidos à análise de variância e as médias comparadas pelo teste de Tukey a 5% de probabilidade. Quando significativos, realizou-se análise de regressão usando os modelos para as curvas de dose-resposta. Os parâmetros fisiológicos avaliados, foram influenciados positivamente pelas subdoses de glyphosate, apresentando maiores valores que a testemunha. Abstract: Panicum maximum is among most important weed specie in tropical crops, justifying the need to control it, basically with herbicides. Therefore this work aimed to evaluate glyphosate sub dose in the gas exchange, dry mass and morphological in P.maximum. The experiment was conducted in a green house, plants were grown in plots, with a casually randomized design, with four replications. The experimental treatments were constituted with one control ( without application of the product) and nine doses of glyphoste(3.78: 8.1: 16.64: 33.48: 67.7: 135: 270: 540: 1080 g a.e. ha-1 ). It was evaluated: Liquid photosintesys, estomatic conductance, transpiration, leaf temperature, intern CO2 concentration. Besides it was evaluated tillering and intoxication, at the end of the experiment it was determined the aerial dry mass and leaf area. The data obtained was submitted to a variance analysis test and means submitted to a Tukey test at 5 % probability and when significative was used regression using the models for dose response curves. The physiological parameters evaluated where positive influenciated by the glyphosate sub dose showing higher values than the control, furthermore the intern CO2 concentration was negative influenced, beyond it was observed increment in the leaf temperature progressively, also promote an increment for aerial dry mass accumulation and leaf area besides. Further more, concluding that in the Panicum maximun plants submitted to the glyphosate doses of 3,78 to 270 g a.e. ha-1 showed and hormetic effect.

Abstract:

Objective: The objective of the current investigation was to attempt to contribute to the limited body of literature regarding the role and importance of social context, in particular social support, to the experience of pain. Specifically, the current study proposed that some of the previously observed effects of pain catastrophizing on pain experience operate through a social mechanism of action by reducing available social resources to help people cope with the stress of pain. Thus, it was hypothesized that perceived social support would at least partially mediate the relationship between pain catastrophizing and pain experience or response variables (perceptions of pain intensity and unpleasantness and tolerance time).

Method: A sample of 285 college students enrolled in introductory psychology courses answered baseline questionnaires about dispositional variables before completing a fixed-time cold pressor task (60s) followed by an open-ended cold pressor task (to test pain tolerance). Questionnaires assessed perceived general social support (the Social Provisions Scale), dispositional and situational pain catastrophizing (Pain Catastrophizing Scale), and perceptions of pain intensity and unpleasantness (using visual analog scales).

Results: The final sample consisted of 285 college students (Age: M = 19.62, SD = 1.73), was primarily White (77.9%) and female (58.2%), and the majority denied having either chronic or recurrent pain (n = 160, 56.1%). Regression and SEM analyses demonstrated that the study mediation hypothesis was not supported.

Conclusion: Results of the current study suggest important revisions and considerations for future replications and similar studies.

Abstract:
The purpose of this dissertation was to determine if a there was a dose-response relationship across an NCAA Division I men’s soccer season. Specifically, this dissertation serves to: 1.) assess the validity of duration measurements that have previously been used to calculate session RPE during competitive matches, 2.) determine the degree and magnitude of change in intermittent endurance performance across a season and to observe how change in endurance relates with training load, 3.) determine the degree and magnitude of change in strength and explosiveness across a season, and to see how these strength changes relate to training load. 1.) Minutes played were found to be the most accurate duration for calculating session RPE during men’s NCAA Division I soccer matches. 2.) Endurance performance, measured using the Yo-Yo IR1, increased by an average of 14 percent from pre to post season. Furthermore, a large, negative relationship was observed between training load and change endurance performance. These relationships suggest that excessive accumulation of training load can impair endurance performance. The congested NCAA DI match schedule may make it difficult to prevent excessive training loads. 3.) When players were grouped by amount of college soccer experience (upper and underclassmen), upperclassmen jumped significantly higher than underclassmen during the preseason. Moreover, the upperclassmen maintained or increased jump height from pre to post season, while underclassmen experienced a general decrease in jump height from pre to post season. Moderate relationships indicate a positive relationship between training load from resistance training and changes in strength. Likewise, these same relationships strengthened in the players who received more playing time during matches. Taken together, the high numbers of moderate relationships indicate a high level of individual variability. Dose-response relationships with strength variables were not sufficiently established. The overall findings of this dissertation provide evidence that subjectively and objectively monitoring training load and soccer-related performance variables can assist coaches in making decisions that will promote the welfare of their team.
