Abstract:
Five volatile organic compounds were identified throughout 1991 in the hypolimnion of Organic Lake. These were dimethylsulphide (DMS), dimethyldisulphide (DM2S), dimethyltrisulphide (DM3S), dimethyltetrasulphide (DM4S) and phenol. The concentration of these compounds increased with lake depth. The concentration of DMS and DM2S was higher in the sediment than in the water column. Carbon disulphide occurred only in the sediment. DMS was the only volatile organic compound detected in the epilimnion of the lake, where its concentration increased from winter onwards. The source of DMS was not dimethylsulphoniopropionate (DMSP). This was determined by hydroxylation of the sample with NaOH. There was no change in the concentration of DMS but the concentration of DM2S increased dramatically while the concentrations of DM3S and DM4S decreased concomitantly. This has important implications in the estimation of DMS derived from DMSP by hydroxylation when organic polysulphides are also present. The stability of the hypolimnion of Organic Lake was reflected by the lack of change in temperature, density, redox potential and the relatively constant concentration of volatile organic compounds throughout the year. Potential sources of the volatile organic compounds are discussed in relation to the isolated nature of the lake.

Abstract:
Myotonic dystrophy type 1 and 2 (DM1 and DM2) are two multisystemic autosomal dominant disorders with clinical and genetic similarities. The prevailing paradigm for DMs is that they are mediated by an in trans toxic RNA mechanism, triggered by untranslated CTG and CCTG repeat expansions in the DMPK and CNBP genes for DM1 and DM2, respectively. Nevertheless, increasing evidences suggest that epigenetics can also play a role in the pathogenesis of both diseases. In this review, we discuss the available information on epigenetic mechanisms that could contribute to the DMs outcome and progression. Changes in DNA cytosine methylation, chromatin remodeling and expression of regulatory noncoding RNAs are described, with the intent of depicting an epigenetic signature of DMs. Epigenetic biomarkers have a strong potential for clinical application since they could be used as targets for therapeutic interventions avoiding changes in DNA sequences. Moreover, understanding their clinical significance may serve as a diagnostic indicator in genetic counselling in order to improve genotype–phenotype correlations in DM patients.

Die Drive for Muscularity Scale (DMS) ist der am häufigsten verwendete Fragebogen zur Erfassung des Muskulositätsstrebens. Ziel der Studie war die psychometrische Überprüfung der deutschsprachigen Version der DMS. 601 Männer aus muskel- und kraftorientierten Sportarten wurden am Computer oder mit der Papierversion der DMS und mit weiteren Skalen befragt. Interne Konsistenz und Retest-Reliabilität der DMS liegen im sehr guten Bereich (>.88), ebenso die Korrelation mit einem Alternativtest (Male Body Attitudes Scale: .81). Muskulositäts- und Schlankheitsstreben lassen sich eindeutig differenzieren (Eating Disorder Inventory-2-DT: .11). Die durchgeführten Faktorenanalysen stützen die Einteilung in eine kognitive und eine korrelierte behaviorale Subskala. Es zeigten sich Zusammenhänge mit körperbezogenem Selbstwert und körperdysmorphen Sorgen. Die deutschsprachige DMS ist ein reliables und valides Instrument zur Erfassung des Muskulositätsstrebens in Forschung und klinischer Einzelfallanwendung.

The objectives of these studies were to determine the cardiopulmonary effects of a single intramuscular administration of 1,1′-methylenebis[4-[(hydroxyimino)methyl]-pyridinium] dimethanesulfonate (MMB4 DMS) on dogs and on the central nervous system in rats. On days 1, 8, 15, and 22, male and female dogs received either vehicle (water for injection/0.5% benzyl alcohol/methane sulfonic acid) or MMB4 DMS (20, 50, or 100 mg/kg). Pulmonary function was evaluated for the first 5 hours after concurrent dosing with cardiovascular monitoring; then cardiovascular monitoring continued for 72 hours after dosing. Rats were dosed once by intramuscular injection with vehicle (water for injection/0.5% benzyl alcohol/methane sulfonic acid) or MMB4 DMS (60, 170, or 340 mg/kg). In dogs, 100 mg/kg MMB4 DMS resulted in increased blood pressure, slightly increased heart rate, slightly prolonged corrected QT, and moderately increased respiratory rate. There were no toxicological effects of MMB4 DMS on neurobehavioral function in rats administered up to 340 mg/kg MMB4 DMS.

Abstract In order to achieve optimal outcomes in an ever-changing environment, humans and animals generally manage their action control via either goal-directed action or habitual action. These two action strategies are thought to be encoded in distinct parallel circuits in the dorsal striatum, specifically, the posterior dorsomedial striatum (DMS) and the dorsolateral striatum (DLS), respectively. The striatum is primarily composed of two subtypes of medium spiny neurons (MSNs): the direct-pathway striatonigral and the indirect-pathway striatopallidal MSNs. MSN-subtype-specific synaptic plasticity in the DMS and the DLS has been revealed to underlie goal-directed action and habitual action, respectively. However, whether any MSN-subtype-specific synaptic plasticity in the DMS is associated with habitual action, and if so, whether the synaptic plasticity affects the formation of habitual action, are not known. This study demonstrates that postsynaptic depression in the excitatory synapses of the direct-pathway striatonigral MSNs in the DMS is formed after habit learning. Moreover, chemogenetically rescuing this depression compromises the acquisition, but not the expression, of habitual action. These findings reveal that an MSN-subtype-specific synaptic plasticity in the DMS affects habitual action and suggest that plasticity in the DMS as well as in the DLS contributes to the formation of habitual action.

Abstract While discourse markers (DMs) and (dis)fluency have been extensively studied in the past as separate phenomena, corpus-based research combining large-scale yet fine-grained annotations of both categories has, however, never been carried out before. Integrating these two levels of analysis, while methodologically challenging, is not only innovative but also highly relevant to the investigation of spoken discourse in general and form-meaning patterns in particular. The aim of this paper is to provide corpus-based evidence of the register-sensitivity of DMs and other disfluencies (e.g. pauses, repetitions) and of their tendency to combine in recurrent clusters. These claims are supported by quantitative findings on the variation and combination of DMs with other (dis)fluency devices in DisFrEn, a richly annotated and comparable English-French corpus representative of eight different interaction settings. The analysis uncovers the prominent place of DMs within (dis)fluency and meaningful association patterns between forms and functions, in a usage-based approach to meaning-in-context.

Protein kinases exert physiological functions through phosphorylating their specific substrates; however, the mode of kinase–substrate recognition is not fully understood. Rho-kinase is a Ser/Thr protein kinase that regulates cytoskeletal reorganization through phosphorylating myosin light chain (MLC) and the myosin phosphatase targeting subunit 1 (MYPT1) of MLC phosphatase (MLCP) and is involved in various diseases, due to its aberrant cellular contraction, morphology, and movement. Despite the importance of the prediction and identification of substrates and phosphorylation sites, understanding of the precise regularity in phosphorylation preference of Rho-kinase remains far from satisfactory. Here we analyzed the Rho-kinase–MYPT1 interaction, to understand the mode of Rho-kinase substrate recognition and found that the three short regions of MYPT1 close to phosphorylation sites (referred to as docking motifs (DMs); DM1 (DLQEAEKTIGRS), DM2 (KSQPKSIRERRRPR), and DM3 (RKARSRQAR)) are important for interactions with Rho-kinase. The phosphorylation levels of MYPT1 without DMs were reduced, and the effects were limited to the neighboring phosphorylation sites. We further demonstrated that the combination of pseudosubstrate (PS) and DM of MYPT1 (PS1 + DM3 and PS2 + DM2) serves as a potent inhibitor of Rho-kinase. The present information will be useful in identifying new substrates and developing selective Rho-kinase inhibitors.

AbstractThis paper reviews hyphenated differential mobility spectrometry (DMS) technology. DMS is a type of ion mobility spectrometry (IMS) also called high-field asymmetric waveform IMS. It is widely used in the detection of chemical warfare agents, explosives, drugs, and volatile organic compounds. Stand-alone DMS analysis of complex mixtures in real-field applications is challenging. Hyphenated DMS can improve resolution for rapid separation and detection. This review focuses on hyphenated DMS, including gas chromatography-DMS, DMS-mass spectrometry (MS), DMS-IMS, IMS-DMS, and DMS-DMS, as well as their associated principles, applications, and research procedures. Key problems in hyphenated DMS are considered.

Zusammenfassung Hintergrund Die vertikale Blickparese (vBP) ist ein typisches Symptom bei Läsionen des dorsalen Mittelhirns. Die klinische Ausprägung der vBP kann dabei stark variieren und im Rahmen eines dorsalen Mittelhirnsyndroms (DMS) mit zusätzlichen Symptomen wie einer Konvergenzparese, einem Konvergenzretraktionsnystagmus, einer verminderten Pupillenreaktion oder einer Skew Deviation vergesellschaftet sein. Je nach Lokalisation und Größe der Läsion kann auch eine Kombination mit weiteren zentralen Motilitätsstörungen vorliegen. Material und Methode Im Rahmen einer retrospektiven Studie wurden die Patientendaten unserer neuroophthalmologischen Ambulanz über einen Zeitraum von 25 Jahren nach Fällen mit Einschränkungen konjugierter vertikaler Augenbewegungen durchsucht und hinsichtlich ihrer neuroorthoptischen Befunde und Ätiologien ausgewertet. Resultate Es wurden 202 Patienten mit vBP identifiziert. Das Befundspektrum reichte von einer isolierten Sakkadenstörung nur nach oben bis hin zum kompletten Ausfall aller willkürlichen und reflektorischen vertikalen Augenbewegungen. Nur 12 vBP lagen isoliert vor. Ursächlich war bei 155 Patienten ein DMS, wovon aber nur 18 Fälle dem von Parinaud geprägten Syndrombild (vBP + Konvergenzparese ± Pupillenstörung ohne weitere Symptome) entsprachen; 42 Patienten mit DMS zeigten lediglich eine Sakkadenparese. Der Aufblick war bei DMS insgesamt mehr eingeschränkt als der Abblick. Die häufigsten Begleitsymptome waren eine Konvergenzparese (49 %), Konvergenznystagmus (40 %), Pupillenstörung (32 %), Skew Deviation (21 %) und Blickrichtungsnystagmus (15 %). In 58 Fällen traten weitere zentrale Okulomotorikstörungen auf. Bis zum 18. Lebensjahr war eine vBP bei 58 % der Patienten durch Gehirntumoren bedingt, wohingegen nach dem 60. Lebensjahr neben einem DMS durch Infarkte (41 %) v. a. degenerative Erkrankungen (44 %) ursächlich waren. Schlussfolgerungen Langsame und schnelle Augenbewegungen nach oben und unten werden im Hirnstamm unterschiedlich generiert, geleitet und integriert und können daher bei Läsionen des Mittelhirns verschieden stark gestört sein. Die Ergebnisse verdeutlichen zudem die enge Lagebeziehung der Zentren für vertikale Augenbewegungen, Konvergenz und Pupillenreaktion im dorsalen Mittelhirn. Die Kombination einer vBP mit weiteren neuroophthalmologischen Diagnosen kann daher eine topografische und ätiologische Zuordnung erleichtern.

Neste estudo, trata-se de um ensaio teórico cujo objetivo é discutir o papel das delegacias especializadas de atendimento à mulher (DMs), a partir do olhar da Psicologia com enfoque de gênero, reflexivo e sistêmico. As DMs desenvolvem procedimentos relativos à apuração de infrações cometidas mediante violência doméstica ou familiar, nos termos da Lei 11.340/2006. No entanto, neste artigo buscase enfatizar as DMs também como um espaço de fortalecimento das medidas integradas de prevenção a esse tipo de violência, trazendo foco para o eixo preventivo instituído pela referida lei. Para tanto, foi discutida a complexidade do fenômeno da violência conjugal, bem como apresentadas algumas possibilidades de atuação policial, baseadas na segurança cidadã, que promovam discussões sobre a complexidade da violência conjugal e os processos culturais que a naturalizam. Entende-se, ainda, que estes serviços só podem ser oferecidos a partir da capacitação continuada dos/as policiais que atuam nas DMs.

During the Arctic summer when the anthropogenic influence is limited, the natural marine source of sulfur in the form of gas-phase dimethyl sulfide viz. DMS(g), is of great importance for cloud formation. The harsh environment of the Arctic makes it difficult to perform in situ measurements of DMS(g) and hence regional model simulations can serve as a complement to increase our understanding of DMS related processes in the Arctic. In this thesis a regional scale meteorological forecast model, extended with DMS(g) calculations, is used to provide a consistent three-dimensional time evolving picture of DMS(g) over the pack-ice region. The analysis focus on meteorological aspects on the horizontal and vertical distribution of DMS(g). Our results show that the amount of DMS(g) over an oceanic source region alone does not determine concentration found over the pack-ice, the prevailing wind also exerts a large influence on the horizontal DMS(g) distribution. The modeled DMS(g) concentrations are advected in plumes in over the pack-ice, which, in combination with the photo-chemical decay, explain the large observed temporal variability of DMS(g) over the pack-ice. The modeled vertical structure show episodes with DMS(g) maxima well above the local boundary layer. Also shown is that DMS(g) maxima can be formed adjacent to frontal zones. In the presence of turbulence DMS(g) can be mixed downwards into the local boundary layer and aid growth to local particles and hence contribute to cloud formation in the boundary layer.
