Abstract:
Sporulation of the yeast Saccharomyces cerevisiae is a process of cellular differentiation that occurs in MATa/MAT alpha diploid cells in response to starvation. The sporulation-specific genes DIT1 and DIT2, which are required for spore wall formation, are activated midway through the sporulation program, with maximal transcript accumulation occurring at the time of prospore enclosure. In this study, we have identified a negative regulatory element, termed NREDIT, that is located between the start sites of transcription of these divergently transcribed genes. This element, which prevents expression of the DIT1 and DIT2 genes during vegetative growth, reduces expression of a CYC1-lacZ reporter gene more than 1,000-fold and acts in an orientation- and position-independent manner. We found that the ability of NREDIT to turn of expression of the reporter gene and the chromosomal DIT1 and DIT2 genes in vegetative cells requires the Ssn6-Tup1 repression complex. Interestingly, NREDIT-mediated repression of the reporter gene is maintained during sporulation. Derepression during sporulation requires complex interactions among several cis-acting elements. These are present on an approximately 350-bp DNA fragment extending from NREDIT to the TATA box and an approximately 125-bp fragment spanning the TATA box of DIT1. Additionally, a region of NREDIT which is very similar in sequence to UASSPS4, an element that activates gene expression midway through sporulation, contributes both to vegetative repression and to sporulation-specific induction of DIT1. We propose a model to explain the requirement for multiple elements in overcoming NREDIT-mediated repression during sporulation.

Background: Non-enhancing black holes (neBHs) are more common in multiple sclerosis (MS) patients with longer disease durations and progressive disease subtypes. Objective: Our aim was to analyse the added value of neBHs in patients with clinically isolated syndromes (CISs) for predicting conversion to clinically definite MS (CDMS). Methods: Patients were classified based on the presence or absence of neBHs and on the number of Barkhof–Tintoré (B-T) criteria fulfilled. Dissemination in space (DIS) was defined as the presence of at least three of the four B-T criteria. Dissemination in time (DIT)1 was defined by simultaneous presence of enhancing and non-enhancing lesions. DIT2 was defined by simultaneous presence of neBHs and T2 lesions not apparent on T1-weighted images. Results: Focal T2-hyperintense brain lesions were identified in 87.7% of the 520 CIS patients, and 41.4% of them presented at least one neBH. Patients meeting DIS, DIT1, and DIT2 had a significantly higher rate of conversion to CDMS. After adjusting for DIS, only patients who fulfilled DIT1 preserved a significant increase in CDMS conversion. Conclusions: Non-enhancing black holes in CIS patients are associated with a higher risk of conversion to CDMS. However, the predictive value of this finding is lost when added to the DIS criteria.

Abstract:
The dityrosine layer is a unique structure present in the spore wall of the budding yeast Saccharomyces cerevisiae. The primary constituent of this layer is bisformyl dityrosine. A sporulation-specific protein, Dit1 is localized in the spore cytosol and produces a precursor of bisformyl dityrosine. Although Dit1 is similar to isocyanide synthases, the loss of Dit1 is not rescued by heterologous expression of the Pseudomonas aeruginosa isocyanide synthase, PvcA, indicating that Dit1 does not mediate isocyanidation. The product of Dit1 is most likely formyl tyrosine. Dit1 can produce its product when it is expressed in vegetative cells; however, formyl tyrosine was not detected in the crude cell lysate. We reasoned that formyl tyrosine is unstable and reacts with some molecule to form formyl tyrosine-containing molecules in the cell lysate. In support of this hypothesis, formyl tyrosine was detected when the lysate was hydrolyzed with a mild acid. The same property was also found for bisformyl dityrosine. Bisformyl dityrosine molecules assemble to form the dityrosine layer by an unknown mechanism. Given that bisformyl dityrosine can be released from the spore wall by mild hydrolysis, the process of formyl tyrosine-containing molecule formation may resemble the assembly of the dityrosine layer.

ABSTRACT The divergently transcribed DIT1 and DIT2 genes of Saccharomyces cerevisiae, which belong to the mid-late class of sporulation-specific genes, are subject to Ssn6-Tup1-mediated repression in mitotic cells. The Ssn6-Tup1 complex, which is required for repression of diverse sets of coordinately regulated genes, is known to be recruited to target genes by promoter-specific DNA-binding proteins. In this study, we show that a 42-bp negative regulatory element (NRE) present in the DIT1-DIT2 intergenic region consists of two distinct subsites and that a multimer of each subsite supports efficient Ssn6-Tup1-dependent repression of a CYC1-lacZ reporter gene. By genetic screening procedures, we identified DFG16, YGR122w, VPS36, and the DNA-binding proteins Rim101 and Nrg1 as potential mediators of NRE-directed repression. We show that Nrg1 and Rim101 bind simultaneously to adjacent target sites within the NRE in vitro and act as corepressors in vivo. We have found that the ability of Rim101 to be proteolytically processed to its active form and mediate NRE-directed repression not only depends on the previously characterized RIM signaling pathway but also requires Dfg16, Ygr122w, and components of the ESCRT trafficking pathway. Interestingly, Rim101 was processed in bro1 and doa4 strains but was unable to mediate efficient repression.

Abstract We previously identified a transcriptional regulatory element, which we call NREDIT, that is required for repression of the sporulation-specific genes, DIT1 and DIT2, during vegetative growth of Saccharomyces cerevisiae. Repression through this element is dependent on the Ssn6-Tup1 corepressor. In this study, we show that SIN4 contributes to NREDIT-mediated repression, suggesting that changes in chromatin structure are, at least in part, responsible for regulation of DIT gene expression. In a screen for additional genes that function in repression of DIT (FRD genes), we recovered alleles of TUP1, SSN6, SIN4, and ROX3 and identified mutations comprising eight complementation groups of FRD genes. Four of these FRD genes appeared to act specifically in NREDIT-mediated repression, and four appeared to be general regulators of gene expression. We cloned the gene complementing the frd3-1 phenotype and found that it was identical to SPE3, which encodes spermidine synthase. Mutant spe3 cells not only failed to support complete repression through NREDIT but also had modest defects in repression of some other genes. Addition of spermidine to the medium partially restored repression to spe3 cells, indicating that spermidine may play a role in vivo as a modulator of gene expression. We suggest various mechanisms by which spermidine could act to repress gene expression.

ABSTRACT Abietane diterpenoids are defense compounds synthesized by trees that are abundant in natural environments and occur as significant pollutants from pulp and paper production. Burkholderia xenovorans LB400 has diverse catabolic capabilities and represents an important group of heterotrophic bacteria in soil environments. The genome sequence of LB400 revealed homologs of the dit genes of Pseudomonas abietaniphila BKME-9, which encode abietane diterpenoid degradation. LB400 grew on abietic acid (AbA), dehydroabietic acid (DhA), palustric acid (PaA), and 7-oxo-DhA. A Xeotron microarray set, with probes for 8450 of the estimated 9000 LB400 genes, was used to compare the transcriptomes of LB400 growing on DhA versus on succinate. On DhA, 97 genes were upregulated, 43 of which were within an 80-kb cluster located on the 1.47-Mbp megaplasmid of LB400. Upregulated genes in this cluster encode a permease, a ring-hydroxylating dioxygenase system (DitA), a ring-cleavage dioxygenase (DitC), a P450 monooxygenase (DitQ), and enzymes catalyzing beta-oxidation-type reactions. Disruption of the ditA1 gene, encoding the alpha-subunit of DitA, abolished growth on these abietanes. Analyses of the metabolism of abietanes by cell suspensions of wild-type LB400 and the ditA1 mutant indicate a convergent pathway, with 7-oxo-DhA as a common intermediate for ring hydroxylation by DitA. Also, 7-oxo-PaA was identified as a metabolite of both AbA and PaA. Sequence analysis indicates that genes encoding this pathway have been horizontally transferred among Betaproteobacteria and Gammaproteobacteria.

ABSTRACT We have cloned and sequenced the dit gene cluster encoding enzymes of the catabolic pathway for abietane diterpenoid degradation by Pseudomonas abietaniphila BKME-9. Thedit gene cluster is located on a 16.7-kb DNA fragment containing 13 complete open reading frames (ORFs) and 1 partial ORF. The genes ditA1A2A3 encode the α and β subunits and the ferredoxin of the dioxygenase which hydroxylates 7-oxodehydroabietic acid to 7-oxo-11,12-dihydroxy-8,13-abietadien acid. The dioxygenase mutant strain BKME-941 (ditA1::Tn5) did not grow on nonaromatic abietanes, and transformed palustric and abietic acids to 7-oxodehydroabietic acid in cell suspension assays. Thus, nonaromatic abietanes are aromatized prior to further degradation. Catechol 2,3-dioxygenase activity of xylEtranscriptional fusion strains showed induction of ditA1and ditA3 by abietic, dehydroabietic, and 7-oxodehydroabietic acids, which support the growth of strain BKME-9, as well as by isopimaric and 12,14-dichlorodehydroabietic acids, which are diterpenoids that do not support the growth of strain BKME-9. In addition to the aromatic-ring-hydroxylating dioxygenase genes, thedit cluster includes ditC, encoding an extradiol ring cleavage dioxygenase, and ditR, encoding an IclR-type transcriptional regulator. Although ditR is not strictly required for the growth of strain BKME-9 on abietanes, aditR::Kmr mutation in aditA3::xylE reporter strain demonstrated that it encodes an inducer-dependent transcriptional activator of ditA3. An ORF with sequence similarity to genes encoding permeases (ditE) is linked with genes involved in abietane degradation.

ABSTRACT The 80-kb dit cluster of Burkholderia xenovorans LB400 encodes the catabolism of abietane diterpenoids. This cluster includes ditQ and ditU, predicted to encode cytochromes P450 (P450s) belonging to the poorly characterized CYP226A subfamily. Using proteomics, we identified 16 dit-encoded proteins that were significantly more abundant in LB400 cells grown on dehydroabietic acid (DhA) or abietic acid (AbA) than in succinate-grown cells. A key difference in the catabolism of DhA and AbA lies in the differential expression of the P450s; DitU was detected only in the AbA-grown cells, whereas DitQ was expressed both during growth on DhA and during growth on AbA. Analyses of insertion mutants showed that ditQ was required for growth on DhA, ditU was required for growth on AbA, and neither gene was required for growth on the central intermediate, 7-oxo-DhA. In cell suspension assays, patterns of substrate removal and metabolite accumulation confirmed the role of DitU in AbA transformation and the role of DitQ in DhA transformation. Spectral assays revealed that DitQ binds both DhA (dissociation constant, 0.98 ± 0.01 μM) and palustric acid. Finally, DitQ transformed DhA to 7-hydroxy-DhA in vitro. These results demonstrate the distinct roles of the P450s DitQ and DitU in the transformation of DhA and AbA, respectively, to 7-oxo-DhA in a convergent degradation pathway.

Abstract This study evaluated the thermal conditions of urban areas in Petrolina-PE, from continuous data collected in urban and rural areas for the year of 2012. The results characterized urban heat islands (UHI) with varying intensity in urban areas, especially UHI = 5.3 °C (high intensity) occurred on April 28, 2012. It was evident that the constituent elements of urban areas contribute to the formation and expansion of UHI bringing thermal discomfort for its inhabitants. An adaptation to Thom’s equation for calculating the Thermal Discomfort Index (DIT), was used to obtain the maximum (DITx) and minimum (DITm) thermal discomfort. In the urban area, the DITm indicated thermal comfort in 23.0% of the days and partial comfort in 77.0% of days surveyed. Already, the DITx characterized 71.6% of days with partial comfort and 28.4% of days with thermal discomfort. In the rural area, The DITm indicated that 41.5% of days were thermally comfortable and 58.5% of days had partial comfort. However, the DITx pointed 87.7% of the days of this environment with partial thermal comfort and 12.3% of thermally uncomfortable days. Finally, the results showed that afforestation of urban area constitutes to an effective and efficient way to mitigate thermal discomfort.

The objectives of this study were to evaluate whether velvet antler (VA) surface temperature gradients, as measured by either a single-spot infrared temperature (SST) sensor (SSTS; exp. 1) or digital infrared temperature (DIT) imaging (DITI; exp. 2), would pattern VA growth. In exp. 1, growth rates and SST were obtained from yearling (n = 8) and mature (n = 17) red deer stags (Cervus elaphus) every 14 d following eruption through day 56 in yearlings and day 112 in mature stags. In exp. 2, growth rates and DIT (main beam VA base, mid and tip temperatures) were obtained from red deer stags (n = 31) every 14 d following eruption through day 126. Background temperatures were recorded in conjunction with thermal antler measurements. In exp. 1, yearling VA base and tip SST were positively correlated with one another (P < 0.01); however, both measurements were also positively correlated with background SST (P < 0.05). In mature stags, VA base SST paralleled (P < 0.05) background SST measures, while tip SST did not change from day 56 through day 112. In exp. 2, VA DIT changed (P < 0.01) over time and differed (P < 0.01) between base, mid and tip. During the early growth period, VA temperatures increased (P < 0.05) from 38.9 ± 0.2°C at the base to 39.3 ± 0.2°C at the tip of the antler. In contrast, during the late growth period, DIT was higher (P < 0.01) at the base (36.8 ± 0.3°C) than at the tip (35.7 ± 0.3°C) of the antler. In conclusion, SSTS did not have the sensitivity to signify changes in antler growth rates. However, in exp. 2 using DITI, VA thermogenesis paralleled VA growth suggesting that DITI may have value in monitoring VA growth. Key words: Velvet antler, red deer, thermography

ABSTRACT Diterpenoids are naturally occurring plant compounds which have pharmaceutical properties. We have sequenced a 10.4-kbp extension of the dit cluster in Pseudomonas abietaniphila BKME-9, containing genes involved in abietane diterpenoid biodegradation. The ditQ gene was found to encode a cytochrome P450 monooxygenase, P450dit, and to be homologous to the tdtD gene of Pseudomonas diterpeniphila A19-6a. Knocking out ditQ had little effect on growth of BKME-9 on abietic acid but severely impaired growth on dehydroabietic acid (DhA) and palustric acid (PaA), increasing doubling times from 3.8 to 15 h on DhA and from 5.6 to 18.5 h on PaA. A xylE transcriptional fusion showed that transcription of ditQ was induced by a range of diterpenoids. Substrate binding assays of P450dit expressed in Escherichia coli revealed that DhA binds to the enzyme and yields a type I binding spectrum with a Kd of 0.4 μM. These results indicate that P450dit is involved in the metabolism of DhA and PaA and are consistent with its putative role in converting DhA to 7-hydroxy-DhA. Finally, an amino acid sequence identity of greater than 72% and conserved gene arrangement support the hypothesis that the dit gene cluster of P. abietaniphila BKME-9 and the tdt cluster of P. diterpeniphila A19-6a contain functional homologues.

Morceaux in&eacute;dits de la charte et quelques remarques pr&eacute;liminaires </p><p>&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp; </p><p>Au cours de ses recherches dans les archives des M&eacute;t&eacute;ores, l'auteur a rep&eacute;r&eacute; (dans le couvent de Varlaam) quatre morceaux (en papier) tr&egrave;s endommag&eacute;s d'une charte byzantine ayant trait &agrave; l'&eacute;v&ecirc;ch&eacute; de Stagi (lignes conserv&eacute;es: 56+12+34=102).- L'&eacute;tude du document l'a men&eacute;e &agrave; la conclusion que ce dernier constitue la premi&egrave;re partie manquante (mais toujours ac&eacute;phale) d'un acte de 1163 dont la derni&egrave;re partie (lignes: 55) est conserv&eacute;e &agrave; Paris (Suppl. grec 1371). Cette derni&egrave;re partie a &eacute;t&eacute; &eacute;dit&eacute;e et richement comment&eacute;e par Ch. Astruc (Bull. Corr. Hell. 83, 1959, p. 206-256).- Description et analyse de la partie in&eacute;dite; essai de reconstitution du document entier. Suivent quelques remarques pr&eacute;liminaires. Generally, it is difficult to generate a high-performance pure blue emission organic light-emitting diode (OLED). That is because the intrinsically wide band-gap makes it hard to inject charges into the emitting layer in such devices. To solve the problem, carbazole derivatives have been widely used because they have more thermal stability, a good hole transporting property, more electron rich (p-type) material, and higher photoconductivity. In the present work, novel copolymers containing donor-acceptor-acceptor-donor (D-A-A-D) blue compounds used for OLEDs were investigated. The theory of the geometrical and electronic properties of N-ethylcarbazole (ECz) as donor molecule (D) coupled to a series of 6 acceptor molecules (A) for advanced OLEDs were investigated. The acceptors were thiazole (TZ), thiadiazole (TD), thienopyrazine (TPZ), thienothiadiazole (TTD), benzothiadiazole (BTD), and thiadiazolothienopyrazine (TDTP). The ground state structure of the copolymers were studied using Density Functional Theory (DFT) at B3LYP/6-31G(d) level. Molecular orbital analysis study indicated 3 investigated copolymers (ECz-diTZ-ECz, ECz-diTD-ECz, ECz-diBTD-ECz) have efficient bipolar charge transport properties for both electron and hole injection to the TiO2 conduction band (4.8 eV). In addition, the excited states electronic properties were calculated using Time-Dependent Density Functional Theory (TD-DFT) at the same level. Among these investigated copolymer ECz-diTZ-ECz and ECz-diTD-ECz showed the maximum absorption wavelengths (λabs) with blue emitting at 429 and 431 nm, respectively. The results suggested that selected D-A-A-D copolymers can improve the electron- and hole- transporting abilities of the devices. Therefore, the designed copolymers would be a promising material for future development of light-emitting diodes, electrochromic windows, photovoltaic cells, and photorefractive materials.

Notre article tentera de montrer qu’al-Ǧāḥiẓ utilise l’ironie de façon pragmatique pour décrier l’attitude d’un de ses contemporains atteint du mal de l’avarice. Abstract This paper reports on an explorative sociophonetic study of the phoneme /s/ in the Dutch language area. Our aim is to investigate the regional variation in the realisation of this phoneme, and to test experimentally the observation of Collins & Mees (2003) that /s/ is sometimes pronounced more like [ ], especially in the Randstad area (called s-retraction). One hundred native speakers of Dutch produced nineteen monosyllabic words containing /s/ in different syllabic contexts. The speakers were born and raised in one of five regions of the Dutch language area (West Flanders, Flemish Brabant, Netherlands Limburg, South Holland and Groningen). Spectral centre of gravity (CoG) and duration were used to measure the degree of s-retraction. CoG values turned out to be significantly lower (consistent with more retraction) in the regions in The Netherlands than in the Flemish regions. Speakers from South Holland produced significantly shorter /s/ than the other speakers. In conclusion, /s/ shows patterns of regional variation that are not fully in line with the observation forwarded by Collins & Mees (2003). The difference between the Flemish and Dutch regions shows that s-retraction is found in an area larger than the Randstad, possibly pointing towards a North-South pattern of variation.
