Abstract:
The Astrin/SKAP complex plays important roles in mitotic chromosome alignment and centrosome integrity, but previous work found conflicting results for SKAP function. Here, we demonstrate that SKAP is expressed as two distinct isoforms in mammals: a longer, testis-specific isoform that was used for the previous studies in mitotic cells and a novel, shorter mitotic isoform. Unlike the long isoform, short SKAP rescues SKAP depletion in mitosis and displays robust microtubule plus-end tracking, including localization to astral microtubules. Eliminating SKAP microtubule binding results in severe chromosome segregation defects. In contrast, SKAP mutants specifically defective for plus-end tracking facilitate proper chromosome segregation but display spindle positioning defects. Cells lacking SKAP plus-end tracking have reduced Clasp1 localization at microtubule plus ends and display increased lateral microtubule contacts with the cell cortex, which we propose results in unbalanced dynein-dependent cortical pulling forces. Our work reveals an unappreciated role for the Astrin/SKAP complex as an astral microtubule mediator of mitotic spindle positioning.

Abstract:
Deciduous holly branches were visually rated over a period of 5 weeks to evaluate differences in display life between various cultivars of winterberry (Ilex verticillata) and japanese winterberry (I. serrata) x winterberry. Holly branches were naturally defoliated and the postharvest performance of the cut branches was therefore based on the quality and longevity of the fruit. Chemical treatments including floral preservative, floral preservative plus silver, and anti-transpirant were also evaluated. `Bonfire' and `Sunset' had the highest ratings for marketability based on the longevity and quality of their fruit. `Bonfire' and `Winter Red' had the highest fruit density per stem. Treatment with floral preservatives significantly increased the display life of holly branches. Preservative plus silver delayed deterioration later in the study, presumably by delaying the senescence of the fruit. Anti-transpirant treatment did not decrease solution uptake by the holly stems. Cold storage of dry branches at 0.00 ± 1.11 °C (32.0 ± 2.0 °F) did not significantly reduce branch display life if held for 23 days or less. Cut branches of all cultivars had a longer display life when stuck in sand and left outdoors in a lath house than when rated in vase solutions indoors. This study indicates that deciduous holly branches provide an attractive alternative cut branch for both interior and outdoor holiday displays.

AimTo determine whether the IntelliVue monitor (ECG plus Masimo pulse oximeter (PO)) displays heart rate (HR) at birth more quickly than Nellcor PO (PO alone) among infants of 29–35 weeks’ gestational age.MethodsUnmasked parallel group randomised (1:1) study.ResultsWe planned to enrol 100 infants; however, the study was terminated due to the COVID-19 pandemic when 39 infants had been enrolled (17 randomised to IntelliVue, 22 to Nellcor). We found no differences between the groups in the time to first HR display (median (IQR) IntelliVue ECG 49 (33, 71) vs Nellcor 47 (37, 86) s, p>0.999), in the proportion who had a face mask applied for breathing support, or in the time at which it was applied. Infants monitored with IntelliVue were handled more frequently and for longer.ConclusionIntelliVue ECG did not display HR more quickly than Nellcor PO in preterm infants. We found no differences in the rate of or time to intervention between groups. Our study was terminated early so these findings should be interpreted with caution.Trial registration numberISRCTN16473881

In multi-view video plus depth (MVD) coding based free viewpoint video applications, a few reference viewpoints’ texture and depth videos should be compressed and transmitted at the server side. At the terminal side, the display view videos could be the decoded reference view videos or the virtual viewpoints’ videos which are synthesized by DIBR technology. The entire video quality of all display views are decided by the number of reference viewpoints and the compression distortion of each reference viewpoint’s texture and depth videos. This paper studies the impact of the reference viewpoints selection on the entire video quality of all display views. The results show that depending on the available network bandwidth, the MVD coding requires different selections of reference viewpoints to maximize the entire video quality of all display views.

AbstractA menu-driven IBM-PC/MS-DOS software system for quantitative XRF analysis was designed combining elements of pattern recognition (spectral “fingerprinting”) with mathematical correction models. The following topics are covered:-pattern recognition method; N - dimension representation-local empirical Lachance-Traill or Lucas-Tooth & Price models-EGA/VGA intelligent driver for a calibration display-dSASE III Plus “data files - accessed from dBXL/Quicksifver”, dBASE III Plus or from compiled BASIC equipped with “call subroutines”-Examples of applications (stainless steels and ceramics).In this approach, experimental alphas are computed utilizing the closest automatically preselected standards from available data files. Alpha values generated in this fashion we term “local alphas”. An intelligent software program is employed which automatically recognizes the resolution of tiie CRT display and delivers the best possible display for the available equipment. This new design also permits the utilization of dBXL/Quicksitver, dBASE III Plus or compiled BASIC resources for additional on site system customization programming. A dBASE III Plus file format is used for XRF data storage. This permits fast data exchange with a local ASCII database and also with all popular spreadsheet formats. Additionally, there are utility subroutines available which allow direct and rapid manipuiation of dBASE III Plus files in compiled BASIC. Application of this software system and graphics to stainless steels and engineered ceramic materials are demonstrated. RIGAKU Smax/PDP-11/73 and TRACOR X-RAY SPECTRACE 5000/Compaq 386 spectrometers are used in these studies.

Movement of mitochondria in Schizosaccharomyces pombe depends on their association with the dynamic, or plus ends, of microtubules, yet the molecular basis for this interaction is poorly understood. We identified mmd4 in a screen of temperature-sensitive S. pombe strains for aberrant mitochondrial morphology and distribution. Cells with the mmd4 mutation display mitochondrial aggregation near the cell ends at elevated temperatures, a phenotype similar to mitochondrial defects observed in wild-type cells after microtubule depolymerization. However, microtubule morphology and function appear normal in the mmd4 mutant. The mmd4 lesion maps to peg1+, which encodes a microtubule-associated protein with homology to cytoplasmic linker protein–associated proteins (mammalian microtubule plus end–binding proteins). Peg1p localizes to the plus end of microtubules and to mitochondria and is recovered with mitochondria during subcellular fractionation. This mitochondrial-associated fraction of Peg1p displays properties of a peripherally associated protein. Peg1p is the first identified microtubule plus end–binding protein required for mitochondrial distribution and likely functions as a molecular link between mitochondria and microtubules.

Abstract:
This present study sought to evaluate the effectiveness of two types of school library displays: a display with books only, called Display, and a display with books, posters, models, copy, and realia, called Display +. The effectiveness of these displays was to be measured in two ways: a) through the observation of the attaction power, holding power and viewer participation in a display and b) through measuring the circulation of displayed books. Before commencing the study a literature search was conducted. The result of the literature search yielded four authors, Goldhor (1972; 1981), Aguilar (1983), Watson (1985), and Baker (1986), who had researched the relationship between circulation and displays. Each of these studies used the measurable effect of circulation increasing, decreasing or remaining constant to determine the effect of the display. This present study used a similar methodology and monitored the circulation of books on both types of displays. However, this study sought to go beyond the effect of circulation in determining the effectiveness of displays because the previous studies had not taken into account the library user who is affected by the display but is unable to borrow a book from the display. The literature search turned towards the area of museum exhibits and displays and the methodology employed to evaluate these exhibits and displays. Shettel (1968), Warren (1972), Screven (1976), Linn (1976), Clowes and Wolfe (1980), and Miles (1982) used attraction and holding power as measures of museum exhibit and display effectiveness. Similarly, library users can be attracted and their attention held by library displays. To the variables of attraction and holding power this study added the variable of participation, picking up books or other items in the display not necessarily with the intent of borrowing the books. In order to construct a Display + and control as far as possible the elements in the display a literature search of books and articles relating to the design elements of displays was conducted, and findings applied in the construction of the display. Shettel's (1968) methodology of unobtrusive observation was employed in this study with the added benefit of videotaping the observations. The hypotheses were set out in three groups, those relating to a single display, those comparing the effectiveness of Display and Display +, and those relating to circulation. The results of this study found in general that the attraction power of Display + exceeded the attraction power of Display but the holding power and participation in Display was greater than that of Display + indicating that the designer of library displays should pay particular attention to the purpose of displays in their libraries. The results of the circulation hypothesis confirmed the results of Goldhor (1972;1981), Aguilar (1983), Watson (1985), and Baker (1986) that more books circulated when they were displayed than when they were on the library shelves.

Handheld devices have a limited amount of screen real estate. If a handheld device could take advantage of larger screens, it would create a more powerful user interface and environment. As time progresses, Moore's law predicts that the computational power of handheld devices will increase dramatically in the future, promoting the interaction with a larger screen. Users can then use their peripheral vision to recognize spatial relationships between objects and solve problems more easily with this integrated system. In the spillable environment, the handheld device uses a DiamondTouch Table, a large, touch-sensitive horizontal table, to enhance the viewing environment. When the user moves the handheld device on the DiamondTouch, the orientation of the application changes accordingly. A user can let another person see the application by rotating the handheld device in that person's direction. A user could conveniently use this system in a public area. In a business meeting, a user can easily show documents and presentations to other users around the DiamondTouch table. In an academic setting, a tutor could easily explain a concept to a student. A user could effortlessly do all of this while having all of his/her information on the handheld device. A wide range of applications could be used in these types of settings.

Abstract:
Vaccines have saved more lives than any other medical intervention. Recombinant vaccines provide unmatched safety profiles, but at the expense of reduced immunogenicity. Virus-like particles (VLPs) resemble viruses in size, shape and repetitive arrangement but are devoid of pathogenic genetic material and therefore safe. Poor immunogens can be rendered immunogenic by display on VLPs. Successfully decorating VLPs is still a major challenge. Genetic fusion or chemical modification is often time-consuming and can lead to misassembly or misfolding, which obstructs generation of the desired immune response. SpyCatcher is a genetically encodable protein, previously engineered to form a covalent isopeptide bond to its peptide-partner SpyTag. Presented in this thesis are SpyCatcher-VLPs, based on the fusion of SpyCatcher to the bacteriophage VLP AP205. SpyCatcher- VLPs can be conveniently conjugated with SpyTag fused antigens, simply by mixing. I demonstrate the modularity of this approach by covalently linking several complex, cysteine-rich malarial antigens to SpyCatcher-VLPs, such as the transmission-blocking antigen Pfs25 and the blood-stage antigen CIDR. A single administration of Pfs25-SpyTag conjugated to SpyCatcher-VLPs induced potent antibody generation against Pfs25, even in the absence of adjuvant. Anti-Pfs25 antibodies induced by this platform conveyed potent transmission-blocking activity in the mosquito vector. The thesis further demonstrates the feasibility of more complex Catcher-nanoparticle architectures. The previously engineered SnoopCatcher covalently reacts with SnoopTag peptide and is orthogonal to the SpyCatcher / SpyTag pair. IMX313 is an engineered chimera of the multimerization domain of chicken complement inhibitor C4-binding protein. This work describes fusion of SnoopCatcher and SpyCatcher to IMX313, which yields independently addressable Catcher-moieties on a single IMX313 nanoparticle. Display of two antigens on one particle may enable single-particle, multi-disease vaccines as well as multi-stage vaccines to tackle immune evasion of parasites. The platforms presented should accelerate and enhance vaccine development and may create opportunities for imaging and metabolic engineering.

This chapter discusses how seizing the opportunities to remove cruelties depends on enthusiasm for a revived democratic politics, plus facility in language to reinterpret human rights, advocate UN peace-keeping responsibilities, and promote the principles of humane governance. A language for humanity includes questions of identity, ideals of humane governance, and determination not to be cruel to future generations. Politicians have been curtailing freedoms in the interests of 'security', allegedly to make nation-states more efficient and to combat terrorist threats. These developments, coupled to a burning planet, display immediate crises. Instead of undue reverence for the integrity of nation-states, humane governance would champion the principle of the Responsibility to Protect. Advocacy of humane governance rests on indictments of the worst effects of capitalism: worldwide inequity, the invisible powers of corporations and appropriation of resources, impoverishment, huge incidence of mental illness, and vast numbers of people feeling worthless.

The browser war is far from over, and the HTML5 <video> tag has not improved video access despite its promises to work without specifying a plug-in. This chapter discusses m-learning with respect to video. It outlines the m-learning paradigm in conjunction with the technical aspects of video display in browsers, when varying media formats are used. The <video> tag used in this work renders videos from two sources with different MIME types. Feeds from the video sources, namely YouTube and UCT Matterhorn, are pulled by a Website that acts as a content aggregator. The content aggregator presents the various user-generated contents and lectures from the two repositories to both lecturers and students as a single source. By leveraging on HTML5, CSS3, and JavaScript, the application should render uniform video display and gather comments among various students, who use various personal mobile devices. Results show that a uniform display cannot be achieved when developing an application for personal mobile devices.

Web-based applications (Web services and service-oriented architectures) can be run via a Web-based browser. There are several approaches to writing such Web-based applications. A lightweight approach is suitable for hand-held devices. In this approach, a Java servlet or a JSP page (Java 2 Platform, JSP), or an ASP application (Microsoft .NET, ASP) generates HTML (Hypertext Markup Language), XHTML, or XML documents (W3C Semantic Web Activity, XHTML, XML) to be displayed by the browser. Most browsers use an anchored URLs extension (e.g., .doc, .jpg, .xml, etc.) to choose an appropriate plug-in to display the URL when it is clicked. Besides displaying text and multimedia, Web servers and/or browsers can also execute Java applets or scripting languages to read and/or change persistent data. Previously, about 98% of these data were stored in relational or object-relational databases. However, recently more of these data are being stored in XML-based documents. Often these documents have an associated “schema” declaring the nesting of tags and the types of primitive values, or an “ontology” (Everett et al., 2002, Hunter, 2003) declaring classes, attributes, and relations that are used in the document.

The present chapter provides insights about the play specialist-based intervention for children with a wide range of pathologies. Telematic adaptations of pedagogical interventions are needed in order to overcome the obstacles given by pandemic emergency and social isolation. The chapter offers a literature review around the role of the play specialist, providing definitions, historical evolution, and outcomes on children's wellbeing. Plus, results from a research around parental wellbeing after the telematic play specialist-based adaptation are displayed and discussed. Finally, qualitative outcomes from parent and educator interviews over the telematic intervention are presented.

Abstract:
This chapter examines the CBBE of two different product category PLBs of the same retailer, holding the retailer constant. Specifically, the study presented examines and compares the CBBE and its underlying dimensions and their relationships for Walmart's Great Value (grocery, consumable household goods) and Equate (beauty, health, pharmacy) PLBs. Based on a total of 421 surveys (270 for the Great Value brand and 151 for the Equate brand), the study found that 1) while consumers seem to have accepted PLBs in general, Great Value and Equate have lower levels of acceptance among consumers and 2) the comparison of models for Great Value and Equate brands displayed similarities and a few differences in significant relationships among the CBBE dimensions. The findings of this study provide important insights to the management of the retail stores to identify the areas for improvements and develop strategies based on similarities and differences of the two brands.

This chapter considers the American wars in Vietnam and Iraq as remembered and curated at the Vietnam Veterans Memorial in Washington, DC, and its traveling Wall That Heals, plus Section 60 of Arlington National Cemetery. The following chapter describes a methodological instrument for managing data, namely the Interactive Visualisation Tool (InViTo). InViTo aims at supporting spatial decision-making processes by proposing a framework for data knowledge. Principally based on Grasshopper, a free plug-in for McNeel's Rhinoceros, InViTo combines GIS data with CAD drawings and raster images for generating interactive spatial visualisations. It is conceived to display in real time the relationships between the territory and planning choices; thus, it is particularly indicated for stimulating discussions and sharing information in collaborative processes. Its high flexibility allows its use in different case studies with a variety of purposes and scales. Innovative elements in approaching spatial decision processes are discussed.
