Over recent years, a plethora of new genetic tools has transformed conditional engineering of the malaria parasite genome, allowing functional dissection of essential genes in the asexual and sexual blood stages that cause pathology or are required for disease transmission, respectively. Important challenges remain, including the desirability to complement conditional mutants with a correctly regulated second gene copy to confirm that observed phenotypes are due solely to loss of gene function and to analyse structure–function relationships. To meet this challenge, here we combine the dimerisable Cre (DiCre) system with the use of multiple lox sites to simultaneously generate multiple recombination events of the same gene. We focused on the Plasmodium falciparum cGMP-dependent protein kinase (PKG), creating in parallel conditional disruption of the gene plus up to two allelic replacements. We use the approach to demonstrate that PKG has no scaffolding or adaptor role in intraerythrocytic development, acting solely at merozoite egress. We also show that a phosphorylation-deficient PKG is functionally incompetent. Our method provides valuable new tools for analysis of gene function in the malaria parasite.

Abstract:
Plasmodium sporozoites that are transmitted by blood-feeding female Anopheles mosquitoes invade hepatocytes for an initial round of intracellular replication, leading to the release of merozoites that invade and multiply within red blood cells. Sporozoites and merozoites share a number of proteins that are expressed by both stages, including the Apical Membrane Antigen 1 (AMA1) and the Rhoptry Neck Proteins (RONs). Although AMA1 and RONs are essential for merozoite invasion of erythrocytes during asexual blood stage replication of the parasite, their function in sporozoites was still unclear. Here we show that AMA1 interacts with RONs in mature sporozoites. By using DiCre-mediated conditional gene deletion in P. berghei, we demonstrate that loss of AMA1, RON2 or RON4 in sporozoites impairs colonization of the mosquito salivary glands and invasion of mammalian hepatocytes, without affecting transcellular parasite migration. Three-dimensional electron microscopy data showed that sporozoites enter salivary gland cells through a ring-like structure and by forming a transient vacuole. The absence of a functional AMA1-RON complex led to an altered morphology of the entry junction, associated with epithelial cell damage. Our data establish that AMA1 and RONs facilitate host cell invasion across Plasmodium invasive stages, and suggest that sporozoites use the AMA1-RON complex to efficiently and safely enter the mosquito salivary glands to ensure successful parasite transmission. These results open up the possibility of targeting the AMA1-RON complex for transmission-blocking antimalarial strategies.

AbstractGreeks, unlike Romans, gambled with five not four knucklebones (astragaloi) in their dice games. As Pollux explains, the high number of an individual knucklebone was sometimes eight rather than six, and so when a dicer rolled ‘all eights’ they attained a sum of 40. This roll was called the ‘Euripides’. The confusion about this throw is due to a report found in Byzantine sources and attributed to Suetonius, where it is claimed that dicers of Greek antiquity used four knucklebones. Suetonius may have confused the Roman custom with the Greek one, but there are good grounds for questioning whether the report is from ‘Suetonius’ at all. Elsewhere ‘Suetonius’ reports that Greeks played with three rather than two cubic dice, but such a report cannot have been made before the seventh century AD.

Hal ini dikarenakan sistem imun yang kurang sempurna pada bayi yang berusia kurang dari 6 bulan. Sehingga sering terjadi gangguan pada sistem pencernaan dan menyebabkan resiko terjadinya diare lebih besar dibandingkan dengan bayi yang menerima makanan pendamping ASI (MP-ASI) setelah usia 6 bulan. Tujuan Penelitian ini bertujuan untuk menganalisa hubungan pemberian MP-ASI dini dengan kejadian dire pada bayi usia 0-6 bulan berdasarkan literature review. Desain penelitian yang digunakan dalam karya tulis ilmiah ini adalah studi literature revie. Hasil literatur review ini menunjukkan bahwa dari 5 jurnal menunjukkan bahwa terdapat hubungan pemberian MP-ASI dengan kejadian diare pada bayi usia 0-6 bulan. Jurnal 1 menggunakan uji Chi-square, jurnal 2 menggunakan uji Spearman, jurnal 3 menggunakan uji regresi linier sederhana, jurnal 4 menggunakan uji Chi-square, jurnal 5 menggunakan uji Chi-square. Kesimpulan yang diperoleh adalah adanya hubungan antara pemberian Makanan Pendamping ASI (MP-ASI) dini dengan kejadian diare pada bayi usia 0-6 bulan.

La composante vocale donne à l’énonciateur la capacité de faire avancer très rapidement le développement de son thème, et elle lui offre de nombreuses facilités pour des stratégies discursives, comme : gauchir le sens dénoté de ses mots, le renforcer, le contester, l’infirmer, en réduire la portée, ménager sa face et celle de l’allocutaire. Il transmet alors à la dérobée jugement, moquerie, insinuation, réserve, surprise, suggestion, espoir, réprobation, conseil, appréciation, remontrance, colère, conviction, demande de connivence, compassion... Dystrophin is absent in muscle fibers of patients with Duchenne muscular dystrophy (DMD) and in muscle fibers from the mdx mouse, an animal model of DMD. Disrupted excitation-contraction (E-C) coupling has been postulated to be a functional consequence of the lack of dystrophin, although the evidence for this is not entirely clear. We used mechanically skinned fibers (with a sealed transverse tubular system) prepared from fast extensor digitorum longus muscles of wild-type control and dystrophic mdx mice to test the hypothesis that dystrophin deficiency would affect the depolarization-induced contractile response (DICR) and sarcoplasmic reticulum (SR) function. DICR was similar in muscle fibers from mdx and control mice, indicating normal voltage regulation of Ca2+ release. Nevertheless, rundown of DICR (<50% of initial) was reached more rapidly in fibers from mdx than control mice [control: 32 ± 5 depolarizations ( n = 14 fibers) vs. mdx: 18 ± 1 depolarizations ( n = 7) before rundown, P < 0.05]. The repriming rate for DICRs was decreased in fibers from mdx mice, with lower submaximal DICR observed after 5, 10, and 20 s of repriming compared with fibers from control mice ( P < 0.05). SR Ca2+ reloading was not different in fibers from control and mdx mice, and no difference was observed in SR Ca2+ leak. Caffeine (2–7 mM)-induced contraction was diminished in fibers from mdx mice compared with control ( P < 0.05), indicating depressed SR Ca2+ release channel activity. Our findings indicate that fast fibers from mdx mice exhibit some impairment in the events mediating E-C coupling and SR Ca2+ release channel activity.

Abstract MicroRNAs (miRNAs) regulate hematopoietic cell fate and their global down-regulation by Dicer1 deletion promotes tumorigenesis in a cancer-cell-autonomous manner (Kumar M.S. et al, 2007). Raajimakers MH et al. (2010) using neonatal Osterix specific dicer deletion showed altered hematopoiesis and developed myelodysplasia. However, there is no study illustrating the role of the ablation of bone marrow (BM) niche specific miRNA processing machinery in the adult mice. Since expression and functions of different mesenchymal and osteoprogenitors vary from embryonic development to adulthood, studying the dicer ablation in adult mice may provide more insight about the role of miRNA processing in adult mice niche. Here we investigate whether adult Osterix expressing cells play a similar role in the HSC niche compared to fetal Osterix expressing cells. We crossed Osx-GFP-tTA-Cre recombinase mice with mice with floxed Dicer1 allele. Crossing generated Osx- GFP-tTA-Cre+Dicerfl/+ (OCDfl/+control) and Osx-GFP- tTA-Cre+ Dicerfl/fl (OCDfl/fl mutant) mice. Osx-GFP-tTA-Cre expression was either activated during embryonic development (young dicer KO) or suppressed using tetracycline until mice were 6 weeks of age (adult dicer KO). We found young dicer KO mice had reduced weight (p=0.0031), leukopenia, anemia, reduced mature CD19+B220- B lymphocytes (p=0.0034) and increased CD11b+Gr- monocytes and CD11b+Gr+ neutrophils (p=0.02 and p=0.04 respectively) in peripheral blood compared to OCDfl/+ control aged littler mates. The leucocytes and platelets showed dysplastic changes suggestive of myelodysplasia and had extra-medullary hematopoiesis. Adult dice KO, on the other hand, show no leukemia development 6 months after Cre activation. The number of BM hematopoietic progenitors (Lin-Sca1+ c-Kit+ cells, LSK) and long term hematopoietic stem cells (LT-HSCs, LSK CD150+CD48+ cells) in young dicer KO mice were significantly reduced compared to age matched control (OCDfl/+ control) mice. We observed increased Annexin V positive LSK, LT-HSCs and megakaryocytes erythroid progenitors (MEP) in the young dicer KO mice indicating increased apoptosis. Adult dicer KO mice didn't have significant changes in apoptosis in different hematopoietic progenitors. In young dicer KO mice, BM derived LSK and LT-HSCs showed increased cycling (SG2M phase, p=0.0133) and less quiescenece (Go phase, p=0.013). However LT-HSC from adult dicer KO didn't show any difference in cell cycling (p=0.18 and 0.09 respectively). Together these results indicate that while Osterix expressing cells in fetal and young mice give rise to a variety of HSC niche supporting cells the adult expression is limited to more mature osteoblast that are not absolutely essential for HSC maintenance. Our study provides the rationale for further exploration of the complexity in hierarchy of activity within niche constituting mesenchymal stroma progenitors and their role in different developmental stages to maintain hematopoiesis. Abstract:
The Punishment of Dirce was a theme that intrigued both artists and patrons of the Roman period. It appeared in diverse locations and media, notably as a wall painting in the House of the Vettii in Pompeii and the Toro Farnese once displayed in the Baths of Caracalla in Rome. In all representations, Dirce struggles with the bull that will trample her to death. Traditional studies of this imagery have focused on the formal characteristics of these representations, studying issues of workshop practice and the relationship between originals and copies. Scholars seldom analyze the meaning of the myth in depth. While most studies note that Dirce often appears in the guise of a maenad, they dismiss this observation. Additionally, it is rarely noted that Dirce's semi-nudity has any role in this story. In fact, her nudity is highly significant, for it was not part of the literary accounts. This study offers a fresh interpretation of Dirce's punishment considering the function of gender. Using literary sources such as Euripides, Plautus, Lucian, and Petronius, as well as visual images in a variety of media including wall painting, sculpture, gems, medals, and lamps, it is argued that artists and patrons combined maenadism and nudity to portray Dirce as a specifically female social transgressor. As a semi-nude maenad, the queen abandons accepted female decorum. These attributes allow the viewer readily to identify Dirce as guilty of hubris against family and society.

Abstract:
Kasus diare di Indonesia masih sangat besar, menurut data 10,2% penduduk mengalami diare. Kematian anak balita akibat diare di dunia masih sangat tinggi. Pemahaman masyarakat tentang keberbahayaan diare terutama pada balita masih sangat kurang, demikian pula pola hidup bersih dan sehat masih menjadi permasalahan bagi sebagian besar masyarakat Indonesia. Kegiatan ini bertujuan untuk meningkatkan pengetahuan masyarakat tentang diare, penyebab, keberbahayaannya, serta cara penanganannya menggunakan bahan alam maupun obat-obatan. Metode yang dilakukan adalah dengan penyuluhan secara online dalam bentuk webinar menggunakan media zoom. Peserta webinar ini adalah masyarakat umum tanpa batas usia, gender, dan wilayah geografis. Jumlah peserta yang mengikuti kegiatan penyuluhan sebanyak 48 orang dari berbagai wilayah di Indonesia dan sebagian besar dari usia remaja sampai dewasa. Hasil yang dicapai dalam kegiatan penyuluhan adalah terjadi peningkatan pengetahuan masyarakat terhadap diare dan cara penanganannya dengan bahan alam maupu obat-obatan. Evaluasi pengetahuan masyarakat didapatkan dengan melihat kenaikan nilai-rata-rata terhadap uji sebelum dan sesudah kegiatan.

Abstract:
In order to conduct analytical tasks, data scientists often need to find relevant data from an avalanche of sources (e.g., data lakes, large organizational databases). This effort is typically made in an ad hoc, non-systematic manner, which makes it a daunting endeavour. Current data discovery systems typically require the users to find relevant tables manually, usually by issuing multiple queries (e.g., using SQL). However, expressing such queries is nontrivial, as it requires knowledge of the underlying structure (schema) of the data organization in advance. This issue is further exacerbated when data resides in data lakes, where there is no predefined schema that data must conform to. On the other hand, data scientists can often come up with a few example records of interest quickly. Motivated by this observation, we developed DICE---a human-in-the-loop system for <u>D</u>ata d<u>I</u>s<u>C</u>overy by <u>E</u>xample---that takes user-provided example records as input and returns more records that satisfy the user intent. DICE's key idea is to synthesize a SQL query that captures the user intent, specified via examples. To this end, DICE follows a three-step process: (1) DICE first discovers a few candidate queries by finding join paths across tables within the data lake. (2) Then DICE consults with the user for validation by presenting a few records to them, and, thus, eliminating spurious queries. (3) Based on the user feedback, DICE refines the search and repeats the process until the user is satisfied with the results. We will demonstrate how DICE can help in data discovery through an interactive, example-based interaction.
