Abstract:
In Ying Xian Fogong Si Shijia Ta 應縣佛宮寺釋迦塔 (Sakyamuni Pagoda of Fogong Temple in Ying County), Shanxi, there are statues set on each floor, and 26 exist in total, with six murals painted on the first floor. The pagoda was designed as a vertically rising Buddhist temple, and the interior space of each floor was customized for the statues. Based on previous research and through surveying and mapping of the architecture and statuary (including the murals on the first floor) combined with geometric design analysis, this paper proposes the following: First, there exists a clear geometric proportion among the interior space and statues on each floor of the pagoda. Second, clear proportional relationships also exist among the statues on each floor, and each of the 26 statues has ‘classical’ proportion rules. Third, the height of the giant Buddha statue on the first floor is the module for not only the height of the statues on each floor but also the construction of the whole pagoda such that the height of the statue on the first floor is 1/6 of the total height of the pagoda (excluding the base). And the ratio of the pagoda’s total height to the first floor’s diameter, the ratio of the total height to the top-story height under the column capital, and all the other geometric proportions are closely related to the architectural modeling. And finally, in the construction of the pagoda, statues, and murals, the scale is deduced to be 1 chi 尺 (Chinese foot) = 29.5 cm. These values give clear scale logics not only to the construction but also to the details of the statues. Accordingly, the most frequently used proportions in the architectural space, statues, and murals of the Pagoda of Fogong Temple are 2, 3:2, 5:3 (or 8:5), and 9:5, which are imbued with cultural messages, like Zhou Bi Suan Jing 周髀算經 (The Mathematical classic of the Zhou shadow-gauging instrument), Ying Zao Fa Shi 營造法式 (Treatise on Architectural Methods or State Building Standards), the ancient Chinese world view—tian yuan di fang 天圜地方 (the dome-shaped heaven and the flat, square earth) reflected from “ yuan fang tu 圓方圖 (rounded-square map)” and “fang yuan tu 方圓圖 (squared-circle map)”, ancient Chinese ideas that ”san tian liang di er yi shu 參天兩地而倚數 (‘three’ is the number of the heaven and ‘two’ is the number of the earth, and all numbers are based on them)” and “jiu wu zhi zun 九五之尊 (nine and five are the numbers of the honorable central position)”, and most probably related to the “mandala” of Esoteric Buddhism and to the Western “Golden Ratio”, which all need further research in depth.

Abstract:
Objetivo: Esse estudo objetivou investigar percepções de estudantes de Odontologia quanto ao medo e à ansiedade em relação ao manejo de pacientes e ao risco de infecção por COVID-19. Materiais e métodos: Esse estudo transversal envolveu todos os alunos regularmente matriculados em Odontologia, no primeiro semestre de 2020, da Universidade Federal de Pelotas. Um questionário foi aplicado, coletando dados demográficos, nível de formação e perguntas relacionadas ao medo e ansiedade frente à pandemia de COVID-19. Quatro comparações de acordo com a fase da graduação (fase pré-clínica ou clínica), nível de graduação e pós-graduação e de acordo com os sexos foram feitas. Análises independentes para as comparações entre os sexos foram realizadas para os alunos de graduação e de pós-graduação (α<5%). Resultados: Foram incluídos 408 estudantes. Na graduação, mulheres relataram sentirem-se mais ansiosas ao realizar tratamento em pacientes com suspeita de COVID-19 (54%) e sentem mais medo ao ouvir que a infecção tem causado mortes (92,4%), na pós-graduação, responderam ser mais nervosas para conversar com pacientes em ambientes fechados em comparações com homens (P<0,05). Alunos em fase pré-clínica possuem significativamente menor receio (65,5%), ansiedade (32,3%) e nervosismo (28,3%) do contágio do COVID-19 quando comparados com aqueles na fase clínica. Conclusões: Mulheres e alunos na fase clínica apresentam maior ansiedade e nervosismo. Descritores: Ansiedade; Estudantes de Odontologia; Medo; Infecções por Coronavírus. Abstract:
Devaluation Yuan affect the global economy, this can be seen with the decline in currency exchange rates and falling stock prices in many countries. Based on this background, the authors do research event study. The selected time period is 11 days of exchange as the event period separated to 5 days before the event day (11 August 2015 as event day) and 5 days after the event day. The results showed that (1) by calculating the average abnormal return (AAR) is known to have a positive abnormal return during the event period; (2) different test results with paired sample test there is no significant difference of average abnormal return between before and after devaluation of Chinese Yuan currency; And (3) with different test using paired sample test there is no significant difference of average trading volume activity between before and after devaluation of Yuan Chinese currency.

Peh Cun merupakan salah satu budaya peranakan hasil akulturasi budaya Tionghoa dan Indonesia. Tradisi Peh Cun bertujuan untuk mengenang dan menghormati patriotisme seorang negarawan asal Tiongkok bernama Qu Yuan yakni menteri Negara Chu yang hidup pada Warring States Period. Qu Yuan seorang menteri yang jujur dan setia, namun difitnah oleh pejabat lainnya sehingga diusir dan bunuh diri karena tidak dihargai kesetiaannya oleh raja. Para nelayan mencari mayat Qu Yuan dengan menaiki perahu naga karena kepercayaan adat bahwa sang naga bisa membantu menemukan tubuh Qu Yuan dari dasar sungai. Selain itu, nelayan juga melempar bakcang ke sungai agar tubuh Qu Yuan tidak dimakan hewan dan dianggap untuk membuang sial. Koleksi busana berjudul ZÀI JÌ YÌ terinspirasi dari keunikan tradisi Peh Cun terutama perlombaan mendayung perahu naga dan tradisi memakan bakcang melalui representasi aksen teknik smock, ombre dye spray, dan opnaisel. Project based learning menjadi metode yang digunakan dalam perancangan koleksi ini. Tujuan perancangan ZÀI JÌ YÌ adalah untuk: 1) Menciptakan koleksi busana berkonsep modern dengan inspirasi Peh cun, 2) Menerapkan teknik manipulasi material ke dalam koleksi busana. Hasil akhir yang diperoleh berupa koleksi rancangan busana deluxe siap pakai dengan siluet simple, stylish dan kontemporer sesuai acuan Indonesia Trend Forecasting 2021/2022 bertema Spirituality subtema Modern dengan menampilkan detail manipulasi dan material secara menyeluruh pada busana. Peh Cun, which emerged from the blending of Chinese and Indonesian traditions, is one of the peranakan cultures. The Peh Cun tradition seeks to celebrate and remember the nationalism of a Chinese leader named Qu Yuan, the Minister of State of Chu who lived during the Warring States Period. Although Qu Yuan was a trustworthy and obedient minister, the monarch disregarded his allegiance, and as a result, he banished him and later killed himself. In accordance with the conventional wisdom that the dragon may aid in locating Qu Yuan's body at the river's bottom, the fishermen used a dragon boat to look for his remains. Additionally, fisherman also toss bakcang into the river to prevent animals from eating Qu Yuan's body, which is said to be unlucky. Incorporating smock technique accents, ombre dye spray, and opnaisel, the ZÀI JÌ YÌ apparel line was motivated by the distinctiveness of Peh Cun's traditions, particularly the dragon boat rowing race and the tradition of eating bakcang. This collection was created using a project-based learning approach. The design goals of ZÀI JÌ YÌ are to: 1) Create a fashion collection with a contemporary concept influenced by Peh Cun, and 2) Apply material manipulation techniques to the fashion collection. The ultimate result is a range of ready-to-wear deluxe apparel designs with simple, stylish, and contemporary silhouettes that adhere to the Spirituality theme and Modern sub-theme of the Indonesia Trend Forecasting 2021/2022.

Abstract:
Penelitian ini bertujuan untuk menganalisis model kurva pertumbuhan pada itik Magelang generasi ke-2 dengan menggunakan dua model pertumbuhan yang berbeda. Data yang digunakan adalah pertumbuhan bobot badan hasil penimbangan itik Magelang dari umur 0 sampai 8 minggu yang diambil setiap 3 hari sekali. Itik yang digunakan sebanyak 297 ekor yang terdiri dari 113 ekor itik jantan dan 184 ekor itik betina. Data dianalisis dengan menggunakan model yaitu Gompertz dan Logistic untuk menggambarkan pola pertumbuhan setiap jenis kelamin. Hasil yang diperoleh menunjukkan model kurva pertumbuhan memiliki tingkat akurasi yang berbeda. Persamaan regresi model Gompertz pada itik jantan Y= 2.386,88 exp (-3,89 exp -0,03) dan betina Y= 1.760,02 exp (-3,78 exp -0,05), sedangkan model Logistic pada itik jantan Y= 1.558,17 / (1+19,78 exp -0,08) dan betina Y= 1.248,90 / (1+18,44 exp -0,08). Titik infleksi itik Magelang jantan dicapai pada Yi= 878,08 g dan ti= 42,44 hari, itik betina dicapai Yi= 647,48 g dan ti= 37,98 hari, sedangkan titik infleksi pada model Logistic itik Magelang jantan dicapai Yi=779,09 g terjadi di ti=39,27 hari dan itik betina dicapai di Yi=624,45 g dan ti=36,43 hari. Simpulan penelitian ini adalah kedua model memiliki tingkat akurasi yang tinggi, Model Gompertz memiliki tingkat akurasi yang lebih tinggi dibandingkan dengan Logistic. Abstract:
Метою наших досліджень було вивчення та аналіз посухостійкості сортів озимої пшениці селекції Інституту кліматично орієнтованого сільського господарства НААН та Селекційно-генетичного інституту Національного центру насіннєзнавства та сортовивчення НААН в умовах Південного Степу України. Матеріали і методи досліджень. Реакцію 18 сортів озимої пшениці на різні умови вирощування вивчали на Асканійській державній сільськогосподарській дослідницькій станції у с. Тавричанка, Херсонська область (46°33’12»N; 33°49’13»E; 39 м над рівнем моря) протягом 2015/16–2019/20 рр. Дослідження проводилися за різних умов зволоження: при зрошенні та без зрошення. Аналіз стійкості сортів озимої пшениці до стресу проводили за допомогою 17 індексів посухостійкості. Результати дослідження та їх обговорення. Найбільшу продуктивність при зрошенні сформували сорти озимої пшениці Марія – 7,41 т/га та Щедрість одеська – 7,53 т/га. За стресових умов найбільшою урожайністю характеризувалися сорти Кохана – 5,24 т/га, Кошова – 5,23 т/га, Зиск – 5,25 т/га та Ліра одеська – 5,42 т/га. За більшою кількістю індексів (13), як найбільш посухостійкий, був виділений сорт Ліра одеська, сорти Кошова та Зиск виділилися за шістьома індексами, сорти Кохана та Ледя ‒ за п’ятьма індексами. Урожайність при стресі мала високу позитивну кореляцію (r = 0,748–1,000) з індексами YSI, YI, RDI, DI, M2STI, HMP, ISR та високу від’ємну кореляцію (r = -0,758) з індексом SSI. Натомість урожайність при зрошенні характеризувалася низькою (позитивною, або від’ємною) залежністю (r = -0,246–0,233) з індексами SSI, YSI, RDI, DI, ISR та середньою (r = 0,457–0,691) з індексами YI, M2STI, HMP. Індекси TOL і SSPI характеризувалися середньою позитивною залежністю з врожайністю при зрошенні (r = 0,485) та середньою від’ємною (r = -0,556) ‒ при стресі. Висновки. Виділено сім основних індексів: толерантності до посухи (TOL), сприйнятливості до стресу (SSI), стабільності урожаю (YSI), відносний індекс посухи (RDI), посухостійкості (DI), схильності до стресу (SSPI), стійкості до стресу (ISR) та три допоміжні індекси: урожайності (YI), другий модифікований індекс толерантності до стресу (M2STI) та гармонійної продуктивності (HMP). За індексами посухостійкості та біплот-аналізом, як найбільш посухостійкі виділені сорти Ліра одеська, Кохана, Зиск та Кошова. Сорти Марія, Нива одеська та Щедрість одеська виділилися як найбільш нестійкий до посухи.

Indonesia merupakan salah satu negara dengan produksi dan ekspor kopi terbesar di dunia. Beberapa negara yang menjadi tujuan utama ekspor kopi Indonesia merupakan importir kopi terbesar di dunia. China sebagai salah satu negara importir kopi terbesar di dunia, belum menjadi negara tujuan utama ekspor kopi Indonesia padahal konsumsi kopi di China semakin meningkat. Selain itu, diketahui bahwa Indonesia dan China memiliki kesepakatan perdagangan bebas yang disebut ASEAN-China Free Trade Area (ACFTA). Penelitian ini bertujuan untuk menganalisis faktor-faktor yang memengaruhi ekspor kopi Indonesia ke China, termasuk adanya penerapan kebijakan ACFTA. Hasil penelitian dengan menggunakan analisis data time series pendekatan Error Correction Mechanism (ECM) menunjukkan bahwa harga teh dunia, harga ekspor kopi Indonesia ke China, dan kurs rupiah terhadap yuan berpengaruh signifikan terhadap volume ekspor kopi Indonesia ke China pada jangka pendek. Pada jangka panjang, variabel nilai ekspor kopi Vietnam ke China, harga teh dunia, harga ekspor kopi Indonesia ke China, kurs rupiah terhadap yuan, dan pemberlakuan ACFTA berpengaruh signifikan terhadap volume ekspor kopi Indonesia ke China. Dibutuhkan waktu 2 triwulan agar ketidakseimbangan terkoreksi menuju keseimbangan jangka panjang.

Sales of business units have decreased due to the implementation of Large -Scale Social Restrictions (PSBB) by the government as an effort to suppress the spread of Covid-19 so that business units including “YUAN” industrial home-owners are required to consider post-pandemic marketing strategies to restore the economic condition of their business. Therefore, the aim of this study is (1) to analyze the existing strategy of YUAN’s home industry marketing; (2) to analyze the marketing strategy of YUAN’s home industry after the covid-19 pandemic; (3) to determine the priority of the marketing mix strategy after the covid-19 pandemic. The data analysis method used this research is STP analysis, marketing mix and Analytical Hierarchy Process. The results showed that (1) the owner has implemented segmenting, targeting, positioning, and marketing mix strategy however it does not reach the optimal; (2) the owner needs to evaluate the previous marketing strategies; (3) the priority of the marketing mix strategy after the covid-19 pandemic is distribution or place strategy.

Hasil kajian yang telah dilakukan mendapati kebanyakan seramik ini berasal dari pelbagai negara dan jangka masa yang berbeza-beza, seperti seramik China dari pelbagai dinasti, seramik dari Asia Tenggara seperti Vietnam, Thailand dan juga Kemboja. Di samping itu, seramik dari Eropah juga turut ditemui. Jangka masa seramik ini adalah dari zaman Dinasti Sung (Abad 10-13 Masihi), Dinasti Yuan (Abad 13-14 Masihi), Dinasti Ming (Abad 14 -17 Masihi), Dinasti Ch'ing (Abad 17-20 Masihi), Eropah (Abad 19-20 Masihi), Thailand (Abad 13- 15 Masihi) dan Vietnam (Abad 14 -15 Masihi). Jumpaan ini menunjukkan berkemungkinan pada suatu masa dahulu kawasan pantai barat Sabah merupakan kawasan perdagangan atau persinggahan yang penting bagi kapal-kapal dagang berikutan penemuan seramik yang banyak dari pelbagai negara dan berlainan jangka masa.

This paper attempts at carrying out this work in this direction. The potential of mackerel fish reaches 5000 tons / year. Mackerel fish is not seasonal, which means it is available throughout the year. This is what makes Yuan Kartika UKM take advantage of the opportunity to process fish products into amplang crackers. Constraints experienced by SMEs so far are; 1) the amount of production that has not been optimal, because of inadequate production equipment, 2) the frying process is less efficient and requires labor skills; 3) marketing of products is still limited in the local area so that the income earned is not maximal. Based on the survey, SMEs need technology improvements for the frying process so that the process can be done semi-mechanically. Mechanical fryers have a capacity of 3-4 kg / process, twice as large as manual fryers which have a capacity of 2 kg of dough / process, so the production process is shorter and meets the SME production target of 30 kg / day. The frying process takes 15 minutes, faster than the manual frying process for 20 minutes. The level of comfort and safety in the frying process is also better, because the equipment is equipped with a protective / frame so that workers are not affected by heat due to the burner stove heating process. Production results are smooth and meet the production targets, and the quality of the amplang cracker products is the same both before and after the semi-mechanical fryer is applied.

Abstract While polyADP-ribose polymerase (PARP) inhibitors have made advancements in the treatment of breast cancer, challenges such as chemotherapy resistance and limited application persist. Fanconi AnemiaComplementation Group I (FANCI), a DNA repair protein associated with breast cancer development, represents a potential target for novel combination therapeutic strategies. However, the role of FANCI in breast cancer and its impact on the efficacy of PARP inhibitors require further investigation. In our study, we analyzed FANCI expression in breast cancer tissues and cell lines, and its correlation with clinical parameters and patient prognosis. Lentiviral vectors were utilized and functional assays were performed to evaluate the effects of FANCI modulation on breast cancer cell growth and migration. Co-immunoprecipitation assays and protein interaction analysis were conducted to identify the interaction between FANCI and PARP1 and determine the specific binding region. The functionality and nuclear distribution of PARP1 were assessed upon FANCI modulation. Finally, the sensitivity of breast cancer cells to the PARP inhibitor talazoparib upon FANCI knockdown was evaluated in vitro and in vivo. Our findings demonstrated that FANCI was overexpressed in breast cancer and associated with poor prognosis. FANCI significantly promoted breast cancer cell proliferation both in vitro and in vivo. We identified the interaction between FANCI and PARP1, specifically at the FANCI helical domain 2 binding site. FANCI knockdown led to reduced nuclear localization of PARP1 and decreased PARP1 activity. Importantly, combination treatment with FANCI knockdown and talazoparib significantly inhibited cancer growth in vitro and in vivo. Additionally, we found that the cyclin-dependent kinase (CDK) 4/6 inhibitor palbociclib, which effectively suppresses FANCI protein expression, exhibited a robust synergistic effect with talazoparib both in vitro and in vivo. In conclusion, FANCI is a novel therapeutic target for breast cancer. Suppression of FANCI regulates PARP1 redistribution and activity, making breast cancer cells more responsive to PARP inhibitors. This combination therapeutic strategy shows potential in enhancing the effectiveness of PARP inhibitors for breast cancer treatment, regardless of BRCA mutations. Teknik analisis data menggunakan uji validitas, reliabilitas, regresi linier berganda, pengujian hipotesis melalui uji t dan uji f, serta koefisien determinasi. Hasil penelitian menunjukkan bahwa pengaruh motivasi terhadap keputusan berkunjung di Pantai Menganti adalah signifikan dengan nilai regresi sebagai berikut Yi = 4.152+ 0.093X1 + 0.427 X2 + 0.178 X3. Secara lebih rinci data penelitian menunjukkan: (1) Hasil frekuensi penyebaran data variabel motivasi dan keputusan berkunjung berada pada kategori baik, (2) Nilai pengaruh motivasi fisik terhadap keputusan berkunjung signifikan artinya jika motivasi fisik meningkat maka akan ada peningkatan keputusan berkunjung ke Pantai Menganti. (3) Motivasi sosial meningkat diikuti oleh peningkatan keputusan berkunjung ke Pantai Menganti secara signifikan (4) Motivasi fantasi berpengaruh secara signifikan terhadap keputusan berkunjung artinya peningkatan nilai motivasi fantasi akan diikuti oleh peningkatan nilai keputusan berkunjung di Pantai Menganti.Kata Kunci: motivasi, motivasi fisik, motivasi sosial, motivasi fantasi, keputusan berkunjung. ABSTRACTThe object in this study was about motivation (X) and visiting decision (Y) with the analysis unit of tourists who are visiting Menganti Beach Kabupaten Kebumen. The type of research used is quantitative descriptive. The sample in this study was 110 respondents with sampling techniques accidental sampling. Data analysis techniques using validity tests, reliability, multiple linear regression, hypothesis testing through T-Test and F-test, and coefficient of determination. The results showed that a motivation influence visiting decision in Menganti Beach is significant with the regression value as follows Yi = 4.152+ 0.093X1 + 0.427 X2 + 0.178 X3. In more detail the research data shows: (1) The result of the frequency of variable data dissemination of motivation and visiting decision is in good category, (2) The value of physical motivation influence against a significant visit decision means that if Physical motivation increased, there will be an increase in visiting decision Menganti Beach Kabupaten Kebumen. Abstract Colorectal cancer (CRC) ranks as one of the most lethal cancers worldwide. Given the advances in immunotherapy and targeted therapy, the survival time in CRC has been improving recently. However, the development of novel and effective therapies for CRC still remains an unmet medical need. Guanylyl cyclase C (GUCY2C) is a transmembrane receptor expressed on the surface of intestinal epithelial cells. Recent studies have demonstrated that GUCY2C is causally linked to intestinal inflammation, dysbiosis, and tumorigenesis. To this end, GUCY2C has emerged as a promising target for immunotherapy and a diagnostic marker of primary and metastatic cancer. Here we describe the discovery and optimization of a novel panel of GUCY2C-targeting T cell engagers (TCEs) developed via our streamlined AI-guided TCE development platform. We have identified a number of anti-GUCY2C antibodies with various binding epitopes on GUCY2C, and subsequently generated a panel of novel GUCY2C-targeting multispecific antibodies with favorable developability and functionalities via our AI-driven engineering technology. Our data demonstrate that a biparatopic GUCY2C x CD3 TCE engineered as an asymmetric ‘1+2’ format can recognize two distinct epitopes on GUCY2C. This biparatopic GUCY2C x CD3 TCE shows enhanced tumor cell binding affinity and superior T cell-mediated cytotoxicity compared to the ‘1+1’ GUCY2C x CD3 TCE and ‘1+2’ bivalent GUCY2C x CD3 TCE where the two GUCY2C targeting arms can only bind to an identical epitope on GUCY2C. In addition, we have engineered a novel GUCY2C × CD3 x CD28 trispecific TCE, in which the CD3-targeting arm in tandem with the co-stimulating CD28-targeting arm mediate the interaction with T cells. This anti-GUCY2C trispecific TCE induces favorable synergistic T cell activation and tumor cell killing in vitro compared to the bispecific counterparts. In vivo anti-tumor activity assessments of these multispecific TCE molecules in various tumor xenograft mouse models are currently ongoing. Abstract. Linear regression analysis is one of the statistical methods aimed at determining the influence of the independent variable (X) on the dependent variable (Y). In estimating the parameters of the linear regression model, it can be done using the method of least squares or ordinary least squares (OLS). However, if you want to model with research data with an unknown functional form, then the regression analysis used is nonparametric regression analysis. Local polynomial regression is one of the models used in nonparametric regression approaches. Estimation of parameters in local polynomial regression can be done using the Weighted Least Squares (WLS) estimation method by considering the value of the kernel function. The kernel function commonly used includes the Gaussian kernel and the Epanechnikov kernel. The best local polynomial regression model can be observed from the bandwidth, local points, weighting, kernel function, and the optimal polynomial degree. The data used in this study are non-oil and gas export values in Indonesia from January 2018 to December 2022. Based on the calculation results, the best local polynomial regression model is the local polynomial regression model with the Gaussian kernel function, with a Mean Absolute Percentage Error (MAPE) of 8.33%, and a coefficient of determination (R2) of 83.99%. The best model to predict non-oil and gas export values in Indonesia is the second-order with a local point (t0) of 22 and a bandwidth (h) of 10, i.e., yi^ = 12521.411 + 35.744 (ti-22)+8.475 (ti-22)^2 Abstrak. Analisis Regresi Linier merupakan salah satu metode statistika yang bertujuan untuk mengetahui pengaruh variabel bebas (X) terhadap variabel tak bebas (Y). Dalam melakukan estimasi parameter model regresi linier dapat dilakukan dengan menggunakan metode kuadrat terkecil atau ordinary least square (OLS). Namun apabila ingin melakukan pemodelan dengan data penelitian yang tidak diketahui bentuk fungsinya, maka analisis regresi yang digunakan adalah analisis regresi nonparametrik. Regresi polinomial lokal merupakan salah satu model yang digunakan dalam pendekatan regresi nonparametrik. Penaksiran parameter pada regresi polinomial lokal dapat dilakukan dengan metode estimasi Weighted Least Square (WLS) dengan memperhatikan nilai fungsi kernel. Fungsi kernel yang banyak digunakan adalah kernel gaussian dan kernel epanechnikov. Model regresi polinomial lokal terbaik dapat dilihat dari bandwidth, titik lokal, pembobot, fungsi kernel, serta derajat polinomial yang optimal. Data yang digunakan dalam penelitian ini adalah nilai ekspor non-migas di Indonesia pada Januari 2018 hingga Desember 2022. Berdasarkan hasil perhitungan, diperoleh model regresi polinomial lokal terbaik adalah model regresi polinomial lokal dengan fungsi kernel gaussian dengan nilai MAPE sebesar 8,33% dan nilai koefisien determinasi (R2) sebesar 83,99%. Abstract Background: Dual HER2 targeted therapy with pyrotinib (a tyrosine kinase inhibitor targeting HER1, HER2, and HER4) and trastuzumab plus chemotherapy has been approved as neoadjuvant therapy for patients with HER2-positive breast cancer in China based on the results from phase 3 PHEDRA study. However, the optimal chemotherapy partner still needs exploration. This multicenter phase 2 trial (ChiCTR1900022293) aimed to investigate the efficacy and safety of epirubicin, cyclophosphamide and pyrotinib followed by docetaxel, trastuzumab and pyrotinib (ECP-THP) as neoadjuvant therapy for patients with stage II-III HER2-positive breast cancer. Methods: Patients received intravenous epirubicin (90 mg/m2) and cyclophosphamide (600 mg/m2) on day 1 of each cycle for four 21-day cycles, followed by intravenous docetaxel (75 mg/m2) and trastuzumab (8 mg/kg loading dose, followed by 6 mg/kg) on day 1 for 4 cycles. Pyrotinib 400 mg was given orally once daily throughout the neoadjuvant therapy period. Surgery was performed within 16-20 days after the last neoadjuvant therapy. The primary endpoint was total pathological complete response (tpCR, ypT0/is ypN0) rate. Results: Between May 2020 and May 2022, a total of 175 patients enrolled. As of May 31, 2022, 144 patients had undergone surgery; the median age was 51 years (range, 26-67). Sixty-seven (46.5%) of 144 patients had hormone receptor (HR)-negative disease, and 77 (53.5%) had HR-positive disease. The tpCR rate was 67.4% (97/144; 95%CI, 59.3%-74.5%). Patients with HR-negative disease had numerically higher tpCR rate than those with HR-positive disease (73.1% [95%CI, 61.5%-82.3%] vs. 62.3% [95%CI, 51.2%-72.3%]), but without statistical significance (P=0.230). Miller-Payne grade 4 and 5 pathological responses were found in 22 (15.3%) and 97 (67.4%) of 144 patients, respectively. Regarding clinical response to neoadjuvant therapy before surgery, 31 (21.5%) of 144 patients achieved complete response and 99 (68.8%) achieved partial response, with an objective response rate of 90.3% (95%CI, 84.3%-94.1%). Of 161 patients with available safety data, the most common grade ≥3 adverse events included diarrhea (57.1%), white blood cell count decreased (8.7%), and neutrophil count decreased (5.6%). No treatment-related deaths occurred. Conclusions: Patients with stage II-III HER2-positive breast cancer show favorable clinical and pathological response to this ECP-THP neoadjuvant regimen, with an acceptable safety profile. <p>Bawang merah merupakan jenis sayuran umbi yang potensial secara ekonomi. Produksi dan harganya yang fluktuatif menjadikan komoditas ini perlu mendapat perhatian yang serius, khususnya untuk pengembangan budidayanya ke lahan marjinal yang masih luas di Indonesia. Tujuan penelitian adalah menerapkan bioteknologi pupuk hayati mikoriza spesifik lokasi lahan marjinal berbasis azolla (Mikola) dan pengurangan dosis pupuk anorganik pada budidaya tanaman bawang merah. Metode penelitian berupa percobaan pot di rumah plastik menggunakan rancangan Central Composit Second Order Design dengan 2 faktor. Faktor pertama, dosis pupuk Mikola yaitu 6, 12, 18, 24, 30 g tanaman-1 dan faktor kedua adalah pengurangan dosis pupuk anorganik Urea, ZA, SP 36 dan KCl yaitu 20%, 40%, 60%, 80% dan 100% dari dosis anjuran. Analisis dilakukan dengan metode Respon Surface Methodology (RSM) dengan model persamaan matematika : Yi= β0X0 + β1X1 + β2X2 + β11X1² 1+ β22X2² +β12X1X2 + εij dengan bantuan program minitab16. Kesimpulan dari penelitian ini menunjukkan bahwa aplikasi pupuk Mikola pada budidaya tanaman bawang merah di pot dengan dosis 18 gram per tanaman dapat mengurangi pemakaian pupuk anorganik hingga 40% dari dosis rekomendasi dan meningkatkan hasil hingga 15%.</p><p><strong>Keywords</strong></p><p>Mikoriza, azolla, pupuk hayati, bawang merah, sayuran</p><p><strong>Abstract</strong></p><p>Shallots is a kind of tuber vegetable economic potential. The objective of research was to apply of biofertilizer mycorrhizal marginal land of azolla based (Mikola) and dose reduction of inorganic fertilizers in the cultivation of shallot. The research was pot experiment using the Central Composite Second Order Design with 2 factors. The factors are the dose of Mikola namely 6, 12, 18, 24, 30 g plant-1 and reduction of Urea, ZA, SP 36, KCl i.e. 20%, 40%, 60%, 80% and 100% of recommended doses. The analysis using Response Surface Methodology, a mathematical equation: Yi= β0X0 + β1X1 + β2X2 + β11X1² 1+ β22X2² +β12X1X2 + εij. The conclusion showed that the application of Mikola fertilizers on the shallot planting with 18 grams per plant can eliminate the use of inorganic fertilizers up to 40% of the dose recommendation and increase the yield up to 15%.</p>

Barley is widely cultivated in the northern coasts of Egypt and in the newly reclaimed lands. The goal of this study was response of some naked barley cultivars with high yield potential using supplemental irrigation under rain-fed conditions at Al-Kasr area, Marsa Matrouh, North-Western Coast of Egypt. The growth, yield, and drought-tolerance indices of Giza 129, Giza 130, and Giza 131, barley cultivars were estimated under rain-fed and supplemental irrigation regimes (SI0: rainfall only; SI1: two supplementally irrigations at tillering and heading stages; SI2: three supplementally irrigations at tillering, stem elongation, and heading stages). Agronomic traits including yield components and drought indices for the studied barley cultivars were assessed. The results displayed that Giza 131 gave a higher performance in most studied characters under rain-fed and irrigation regimes. Also, it produced the highest tolerance index (TOL), yield index (YI), harmonic mean (HM), stress susceptibility percentage index (SSPI), sensitivity drought index (SDI), mean productivity (MP), and geometric mean productivity (GMP). MP was positively correlated with YSI, GMP, DI, and HM. So, Giza 131 cultivar was identified as a drought-tolerant genotype. Moreover, the drought tolerance indices of TOL, YI, HM, SSPI, SDI, MP, and GMP are suitable for screening cultivars with high yields under stressed and non-stress conditions. The correlation analysis between the studied traits of barley indicated that highly significant positive correlation was obtained between number of spikes m-2 and grain yield. So, these traits are the most important components of the estimated grain yield of naked barley

Results: (1) the most frequently selected points for dysmenorrhea in order were: San Yin Jiao(SP6), Guan Yuan (RN4), Zhong Ji (CV3), Qi Hai (CV6), Ci Liao (BL32), Di Ji (SP8), Shen Shu (BL23). (2) the major characteristics of point selection was combination of local points and distant points which located on Ren meridian and spleen meridian. (3) the major treatment methods were normal acupuncture, then acupuncture combined with moxibustion or auricular therapy, for the latter, were especially stressed and confirmed by many studies. Conclusion: The acupuncture recipes for dysmenorrhea were usually made by consideration of combining local and distant points, utilizing multiple methods especially auricular therapy.

Among abiotic stresses, drought is undoubtedly one of the most important ones, that have great impact on crop growth and productivity worldwide. Therefore, identifying of plants' performance against drought stress and estimating drought tolerance become a necessary part of the breeding phase. The main purpose of the present study was to investigate the effect of several indices that combine drought tolerance and high yield potential in chickpea. The trials were conducted under both stressed and no-stressed environments for two growing seasons (2015/2016-2016/2017) in Southeast Anatolia Region of Turkey. Varyans analysis results showed that there were significant differences among genotypes regarding Yp, Ys, MP, MRP, GMP, REI, STI, MISTIk 1, MISTIk 2, HM, YI, PI, ATI, SNPI and RDY. The genotypes FLIP09-51C, FLIP97-503C and FLIP06-97C had high yield under non-stressed condition, while the genotypes FLIP09-51C, FLIP06-97C and ‘Aksu’ displayed high amount under stressed condition. Thus, the genotypes FLIP09-51C and FLIP06-97C were found as good candidates for commercial recommendation in both conditions. Spearman rank correlation matrix showed that drought indices were significantly related to each other. The yields in stress and no-stress conditions (Yp and Ys) showed a significant and positive correlation with MP, MRP, GMP, REI, STI, MSTIK1, MSTIK2 and HM and showed a negative correlation with PI and RDY. As a result, it has been found that MISTIK2, DI, HM, STI and YI can be used as optimal indicators for screening drought-tolerant genotypes, while FLIP09-51C, FLIP06-97C, EN934 and ‘Aksu’ varieties have been the most tolerant genotypes in terms of these indices examined in study.

Abstract Purpose: First-in-human Phase I study for advanced MUC1* positive breast cancer with autologous T cells engineered to express either a chimeric antigen receptor, huMNC2-CAR44 or huMNC2-CAR22, which specifically bind to a cleaved form of MUC1 (MUC1*); evaluate the safety and preliminary anti-tumor activity. Methods: MUC1* (muk 1 star) is the growth factor receptor form of MUC1, created by cleavage and release of the N-terminal portion of MUC1. The targeting antibody, huMNC2, only recognizes the conformational epitope created when MUC1 is cleaved by specific tumor-associated enzymes that are correlated with poor prognosis. huMNC2 does not bind to full-length MUC1, which is expressed on all normal epithelial cells. huMNC2-scFv was incorporated into huMNC2-CAR44, comprising a CD8 hinge and transmembrane region, 4-1BB costimulatory domain and wild-type CD3z. huMNC2-scFv was also incorporated into huMNC2-CAR22 wherein the hinge, transmembrane and co-stimulatory portions were derived from CD28 and CD3z bears the 1XX mutations to increase in vivo persistence. Inclusion criteria require that the patient’s tumor is at least 30% MUC1* positive and that patient has progressed through 2 or 3 prior therapies, while in the metastatic setting. Patients receive standard Cy/Flu lymphodepletion approximately 3-days before CAR T treatment, administered at 1 of 4 dose levels ranging from 3.3X10^5 up to 1.0X10^7 CAR+ T cells. Results: To date, 8 patients have been treated with huMNC2-CAR44. No patients experienced neuro toxicities. No off-target toxicities were observed. 3 patients experienced CRS Grade 1-3. In 6 of the 8 patients, side effects were non-existent or minimal. However, one patient experienced a Grade 5 SAE that was deemed to be possibly related to the treatment. Best responses include Partial Responses and Stable Disease at a low CAR-T dose. Greatest efficacy was observed for patients whose biopsy showed H Scores &gt;120. Patients are currently being enrolled for treatment with huMNC2-CAR22, where the 1XX mutations are expected to increase in vivo persistence, durability of response and reduced incidence of CRS. Conclusions: These data support a conclusion that the MUC1* antibody, huMNC2, is safe and could have high therapeutic value as a CAR T treatment for solid tumors with moderate to high antigen density. As the huMNC2-CAR22 (1XX) trial proceeds, we will assess if patient responses mirror our animal results that show that the 1XX mutations confer increased persistence, reduced exhaustion and the ability to kill tumors with low antigen density. Abstract There is no fixed categorical term for barbarians in Chinese. Specific groups are identified as “aliens” or “cultural others” through words like Man 蠻, Yi 夷, Rong 戎, and Di 狄, but all four terms can be specific or categorical. It is often said that the us-versus-them formula in Zuozhuan (and early Chinese texts in general) is cultural rather than ethnic, but precise definitions of cultural difference can be elusive. This article begins with a discussion of the historical basis for defining possible differences in socioeconomic and cultural practice and moves to the question of representation. It focuses on three issues: (1) Who is the barbarian? Using Lu's dealings with the eastern Yi domains as case studies, the author explores how representation of cultural others is inseparable from cultural self-definition. Similarities and shared roots seem to have generated the impetus for emphasizing distinctions. (2) Arguments on cultural connections or lack thereof are often built on historical retrospection. Embracing historical ties with barbarians can be a way to resist Zhou dynasty demands, even as using the ancient past to disclaim the status of cultural other can function to assert hegemonic ambition. The author examines the uses of history to manipulate notions of shared roots and radical difference. (3) Since the negative qualities attributed to barbarians come up in speeches, we need to consider the rhetorical context of moralizing otherness. Whether the issue is debates on military strategy, the choice of war or peace, or the etiquette of presenting the spoils of victory, we see how attention to particular motives and circumstances driving historical developments results in a complex and nuanced picture that resists simplistic and moralized formulations of cultural identity and cultural difference.

Metode penelitian ini menggunakan metode kuantitatif. Populasi dalam penelitian ini adalah konsumen yang berkunjung dan membeli di Starbucks Galaxy Mall minimal 2 kali pembelian dan berusia 20-50 tahun. Sedangkan sampel yang diambil berjumlah 100 orang/responden. Teknik analisis data yang digunakan adalah regresi linier berganda. Penelitian ini menggunakan Importance Performance Analysis dan Potential Gain in Customer Value dengan menggunakan alat bantu MS. Office Excel Versi 2010. Hasil penelitian menunjukkan bahwa total dari nilai tingkat kinerja (Xi) yaitu, 8111. Total dari nilai tingkat kepentingan (Yi), yaitu 9209. Dan total dari nilai tingkat kesusaian responden (Tki) sebesar 88,06%. Berdasarkan hasil analisa dapat diketahui bahwa atribut P19 merupakan prioritas ke-1 dengan indeks PGCV sebesar 7.55, atribut P13 merupakan prioritas ke-2 dengan indeks PGCV sebesar 6.65, dan atribut P21 merupakan prioritas ke-3 dengan indeks PGCV sebesar 6.63. Penelitian ini bertujuan untuk mengetahui dan menganalisa kinerja kualitas pelayanan dengan tingkat kepentingan (importance) Kualitas Pelayanan pelanggan dan tingkat kepuasan (satisfaction) konsumen Starbucks Coffe Surabaya serta untuk mengetahui dan menganalisa atribut-atribut yang menjadi prioritas perbaikan peningkatan kualitas pelayanan. Metode penelitian ini menggunakan metode kuantitatif. Populasi dalam penelitian ini adalah konsumen yang berkunjung dan membeli di Starbucks Galaxy Mall minimal 2 kali pembelian dan berusia 20-50 tahun. Sedangkan sampel yang diambil berjumlah 100 orang/responden. Teknik analisis data yang digunakan adalah regresi linier berganda. Penelitian ini menggunakan Importance Performance Analysis dan Potential Gain in Customer Value dengan menggunakan alat bantu MS. Office Excel Versi 2010. Hasil penelitian menunjukkan bahwa total dari nilai tingkat kinerja (Xi) yaitu, 8111. Total dari nilai tingkat kepentingan (Yi), yaitu 9209. Dan total dari nilai tingkat kesusaian responden (Tki) sebesar 88,06%. Berdasarkan hasil analisa dapat diketahui bahwa atribut P19 merupakan prioritas ke-1 dengan indeks PGCV sebesar 7.55, atribut P13 merupakan prioritas ke-2 dengan indeks PGCV sebesar 6.65, dan atribut P21 merupakan prioritas ke-3 dengan indeks PGCV sebesar 6.63. Abstract Background: Previous studies have demonstrated that proliferation- and ER-associated gene expression shows a time-dependent interaction with risk of recurrence. Breast Cancer Index (BCI) is a gene expression-based test that integrates biomarker panels of both proliferation (Molecular Grade Index, MGI), and endocrine response (HOXB13/IL17BR, H/I). BCI has been validated as a significant and independent prognostic factor both for risk of overall (0-10y) and late (&gt;5y) distant recurrence and is predictive of extended endocrine benefit in patients with early-stage, hormone receptor-positive (HR+) breast cancer. In this study, correlative analyses were performed to evaluate molecular measures of proliferation and ER signaling by MGI and H/I that underly recurrence risk across clinical categories in patients receiving BCI testing as part of routine care. Methods: Analysis was performed using the BCI Clinical Database for Correlative Studies, an IRB-approved de-identified database which contains &gt;50 data elements including clinicopathologic and molecular variables from 19,126 clinical cases submitted for BCI testing. Descriptive analyses assessed the distribution of MGI by H/I categories based on time of testing or BCI intervention point (time of diagnosis &lt;1y for early recurrence vs 4-7y post diagnosis for late recurrence), lymph node status (N0 vs. N+), age (&lt;50y vs. ≥50y), tumor size (T1-T3), and grade (1-3) to understand the molecular features that may be associated with recurrence in HR+ disease. Results: Analysis included 18,853 patients (88% ≥50y, 73% N0, 70% pT1, 51% grade 2). Comparative analysis of patients tested with BCI at &lt;1y vs. those recurrence-free and tested at 4-7y post diagnosis showed a similar proportion of patients in the H/I categories (H/I-High: 44%1y, 43% 4-7y, P=0.510). An increased proportion of MGI-High was observed in 4-7y (61%) vs. 1y (53%) (P&lt;0.001). The distribution of MGI categories within H/I-Low and High subsets was also similar based nodal status (N0 vs. N+), age (&lt;50y vs. ≥50y), and tumor size. However, H/I status was inversely proportional to tumor grade (P&lt;0.001): the proportion of H/I-Low decreased with increasing grade (Grade 1: 70%, 2: 59%, 3: 36%), whereas the proportion of H/I-High patients increased (Grade 1: 30%, 2: 41%, 3: 64%). In the H/I-Low subset, as grade increased, the proportion of MGI-Low patients decreased (1: 45%, 2: 23%, 3: 5%), while MGI-High remained relatively consistent across grade (1: 25%, 2: 36%, 3: 31%). In H/I-High patients, MGI-High increased as grade increased (1: 11%, 2: 26%, 3: 59%). Conclusion: BCI functional characterization using MGI for tumor proliferation and H/I for estrogen signaling in this large-scale analysis showed that over half (59%,1y vs. 67%, 4-7y) of endocrine responsive tumors (H/I-High) were also highly proliferative (MGI-High) irrespective of time of testing. Results showed that tumor grade correlated with MGI and H/I status, indicating that a subset of high-grade tumors were highly proliferative as well as endocrine responsive. These findings suggest that proliferation and endocrine signaling are combinatorial molecular drivers of recurrences in HR+ breast cancer. Abstract:
Abstract Background: huMNC2-CAR44 and huMNC2-CAR22 are autologous CAR T cell therapies under study in an ongoing 1st-in-human trial for metastatic breast cancers (NCT04020575), being performed at City of Hope. Both CARs are targeted to the tumor by an antibody, huMNC2, that recognizes a cryptic binding site on MUC1*, which is the transmembrane cleavage product of MUC1. The antibody binds to an epitope that is only unmasked when MUC1 is cleaved to MUC1* by enzymes in the tumor microenvironment. huMNC2 strongly reacts with over 90% of breast cancers. No therapeutic that targets MUC1* had ever been tested in humans before this trial. We note that neither 5E5 nor antibodies that bind to a MUC1 “heterodimer” recognize MUC1*. Eight patients have already been treated with huMNC2-CAR44. The next 8 patients will be treated with huMNC2-CAR22 to enable comparison and inform decision as to which CAR to bring forward for completion of Phase 1 and entry into Phase 2. huMNC2-CAR22 differs from huMNC2-CAR44 in that it is resistant to exhaustion. CAR22 achieves greater in vivo persistence due to Sadelain’s “1XX” mutations of Tyr to Phe in 2 of the 3 ITAMs, which prevent Tyr phosphorylation and signaling, leaving signaling through ITAM 1 alone. Trial Design: Dose escalation or de-escalation is tested in cohorts of 3 patients each using standard “3+3” dose-finding, with the starting dose of 3.3x105 CAR+ T cells/kg up to a maximum of 1.0x107 CAR+ T cells/kg. Patients receive cyclophosphamide (300 mg/m2/day) and fludarabine (30 mg/m2/day) for 3 days prior to CAR T cell infusion. Safety will be evaluated by CTCAE version 5.0 and Lee criteria. Anti-tumor activity will be assessed by imaging studies completed between 1 and 3 months after huMNC2-CAR T cell infusion for determination of response by RECIST 1.1 or by FDG PET modified PERCIST for patients with predominant bone disease. Inclusion Criteria: Patients with confirmed diagnosis of breast cancer, with documented ER, PR, and HER2 status per ASCO/CAP guidelines. Patients with MUC1* expression of at least 30% by IHC. Patients must have received standard metastatic systemic therapy per NCCN guidelines which are known to confer benefit. No maximum on number of prior treatments. Patients must have received at least 2 or 3 prior lines of chemotherapy in the metastatic setting. Exclusion Criteria: Patients requiring &gt;15 mg of prednisone per day or immunosuppressives; patients with major organ dysfunction; Serum creatinine &gt; 2 mg/dL; Bilirubin ≥ 1.5 mg/dL; AST/ALT ≥ 2.5 x upper limit normal; 3x upper limit for patients with known liver metastasis; significant pulmonary dysfunction; significant cardiovascular abnormalities; ANC &lt; 1000/mm3. Abstract:
Abstract Poly(ADP-ribose) polymerase (PARP) inhibitors (PARPi), which induce DNA damage by inhibiting PARP1 enzymatic activity and trapping PARP on the damaged DNA, are used to eliminate BRCA1/2-mutated (BRCAm) cancer. However, clinical observations suggest that BRCAm tumors develop PARPi resistance. Current strategies to overcome PARPi resistance include impeding multiple DNA repair pathways to induce excessive DNA damage. Here, we propose a novel strategy targeting oncogenic receptor tyrosine kinases to enhance PARP trapping. By developing triple-negative breast cancer (TNBC) cells with acquired talazoparib resistance, we observed a high prevalence of activated fibroblast growth factor receptor 3 (FGFR3) kinase in these cells through kinase antibody array analysis. Mass spectrometry analysis and in vitro kinase assay suggested that FGFR3 phosphorylated PARP1 at tyrosine residues 158 and 176. Biochemistry studies suggested that only PARP1 tyrosine 158 phosphorylation contributes to PARPi resistance in the cells we developed. We then developed a monoclonal antibody against tyrosine 158 phosphorylated PARP1, and found that high-level PARP1 tyrosine 158 phosphorylation positively correlated with PARPi resistance in breast cancer patient-derived xenograft models. We further demonstrated that the combination of FGFR inhibitor and PARPi delayed DNA repair with prolonged PARP trapping. Moreover, synergy between PARPi and FGFR inhibition was observed in multiple TNBC cell lines with PARPi resistance in vitro. The combination of PARPi and FGFR inhibitor also showed synergism in vivo, and treatment with the combination of PARPi and FGFR inhibitor was tolerated in mouse models. These findings reveal that PARP1 tyrosine 158 phosphorylation facilitates resolving of the PARPi-induced PARP-trapping, and that the tyrosine 158 phosphorylated PARP1 may be an effective biomarker to indicate FGFR3 mediated PARPi resistance. Determination of location of research conducted by the method of "Purposive Sampling". Sampling conducted by judgment sampling, while the determination of merchant that made the sample was done by snowball sampling method. The research method used in this research is survey method by conducted interviews and observation of fishermen and traders. The results of this study are 1) The cost of farming cover of labor, fertilizers and pesticides. (2) The result of B/C and R/C ratio is greater than one, so farming can be cultivated because it gives benefits for farmers. (3) The cost of cocoa farming is divided into two, namely: a) Fixed costs and non-fixed costs. The fixed cost (TFC) consists of labor, while the variable cost (VC) consists of fertilizers and pesticides. (4) Average income of cocoa farmers is Rp.13.280.000 per year or equivalent to Rp.1.101.500 per month. (5) Cocoa beans marketing in Juhar Subdistrict there are three types of marketing channels. Marketing channel 1 is Farmers à Village traders à Sub-district traders à Traders at provincial level. The types of marketing channel II is Farmers à Sub-district traders à Traders at the provincial level. The marketing channel III is Farmers à Village traders à Traders at provincial level. (6) The largest costs, benefits and margins obtained marketing by tipe of one marketing is Rp.2.338; Rp.3.586 dan Rp.5.924 The marketing efficiency is tipe of three marketing yi 4,2%. The bigger the share of the price received by farmers, it can be said that the marketing channel is more efficient. Thus the most efficient marketing channel is the three marketing channels.

Abstract:
Abstract Background: Immune checkpoint inhibitor (ICI) combined with chemotherapy as a neoadjuvant therapy has been applied to the treatment of ESCC. However, the optimal regimen needs to be further explored, and the mechanisms by which the ICI combined with chemotherapy modulates the evolution of ESCC are unknown. Methods: In this single-arm phase 2 trial, patients received nab-paclitaxel 260 mg/m2 for 2 cycles and oral S-1 (d1-14, bid) for two weeks, combined with 240 mg intravenous toripalimab for 2 cycles before surgery. The primary outcome was the major pathological response (MPR). Biopsies and plasma before and after neoadjuvant therapy were analyzed for whole-exome sequencing, transcriptome sequencing, PD-L1 expression and 92 proteins using proximity extension assay technology. Results: After neoadjuvant therapy, 26 patients (48.15%) achieved MPR, 16 (29.63%) achieved pCR. 18.3% of the patients experienced grade ≥3 AEs. Compared to patients PD-L1-low, patients with PD-L1-high had a significant higher ratio of PR. During therapy, TMB and TNB were significantly decreased in PR patients, and the ratio of gained and increased clones were significantly lower in MPR patients than nonMPR patients, while the lost and decreased clones were significantly higher in MPR patients than nonMPR patients. Transcriptome analyses before and during therapy revealed CD8 and CD4 T cells were increased in all patients, however, the exhausted cells, nTreg and iTreg were significantly increased in nonMPR patients. Proteins analyses revealed the levels of TRAIL and MUC16 were significantly increased in MPR and PR patients, respectively, while the levels of IFN-γ and IL18 were significantly decreased in MPR and PR patients, respectively. And the expression of ADA, ARG1, CD83, TNFRSF4 and VEGFR2 in plasma were found to be related with serious adverse events. Conclusion: In this study, neoadjuvant therapy with toripalimab, nab-paclitaxel and S-1 was well tolerated and showed promising antitumor activity in patients with resectable ESCC, and the changes of genomic, transcriptome, PD-L1 expression and proteins in plasma were comprehensively analyzed and correlated with clinical outcomes, which provided insight into the mechanism of action of Toripalimab combined with nab-Paclitaxel and S-1 in ESCC patients. Abstract Background: Colorectal cancer (CRC) is a commonly diagnosed subtype of cancer with a high mortality rate. Potassium is a necessary trace element in the human boy and is derived mainly from diet. Prior studies found high intakes of potassium was inversely associated with colorectal cancer occurrence in Western population. No such effort has been made in Asian population. Methods: We examined the associations for dietary potassium with the risk of CRC in the Singapore Chinese Health Study (SCHS), a population-based prospective cohort study that includes 61,321 Chinese men and women in Singapore who were 45-74 years of age at baseline. Daily intake of potassium was derived from the self-reported intake of pre-defined food items using a validated semi-quantitative food frequency questionnaire (FFQ) at baseline according to the Singapore Food Composition Table. Cox proportional hazard regression model was used to estimate the hazard ratios (HRs) and respective 95% confidence intervals (CIs) for CRC associated with higher quartile of potassium compared with the lowest quartile with adjustment for potential confounders. Results: After an average of 17.5 years of follow-up, 2,140 participants developed CRC. Dietary potassium was inversely associated with CRC risk. The multivariable-adjusted HR of CRC for the highest relative to the lowest quarter of daily potassium intake was 0.85 (95% CI 0.75-0.96, Ptrend=0.03). The inverse association between high intake of potassium and CRC risk was more apparent among participants with body mass index (BMI) ≥ 23 kg/m2 (HRQ4vsQ1=0.75, 95% CI: 0.63-0.89; Ptrend=0.003) and those with history of type 2 diabetes (HRQ4vsQ1=0.65, 95% CI: 0.42-1.00; Ptrend=0.36). Conclusions: In a large population-based prospective cohort study of more than 61,000 Chinese Singaporeans, we found inverse associations between dietary potassium intake and CRC risk. Future studies are warranted to elucidate the biological mechanism for the protective effect of potassium on the development of CRC. Citation Format: Hung N. Abstract Introduction: Microsatellite instability-high (MSI-H) colorectal cancer (CRC) consists of 15% of all CRC but only 2-4% of metastatic CRC. The favorable prognosis of non-metastatic (early-stage) MSI-H CRC may be related to vigorous immune reaction in tumor microenvironment. However, the mechanisms of immune evasion in metastatic (late stage) MSI-H CRC are largely unknown. Materials and Methods: We retrospectively collected patients with MSI-H CRC from National Taiwan University Hospital and Kaohsiung Chang Gung Memorial Hospital. The archival tumor specimens were used for immune cells analysis by Nanostring PanCancer Immune Panel, and spatial immune characterization by multiplex immunofluorescence stains. Only surgical resected primary tumors and metastatic tumors were included. Gene expression and immune cells composition were compared between non-metastatic and de novo metastatic tumors. Results: We enrolled 59 and 27 patients with early-stage and late-stage MSI-H CRC, respectively. Twelve metastatic site tumors were also collected. The clinicopathologic characteristics between patients who have early- and late-stage MSI-H CRC were not significantly different. There are heterogeneous immune cell compositions in these 98 MSI-H tumors. Compared to early-stage MSI-H tumors, there are significantly fewer cytotoxic (difference -0.497, p = 0.001) and dendritic cells (difference -0.528, p = 0.021) in late-stage MSI-H tumors. Gene set enrichment analysis disclosed significantly decrease in the expression of JAK-STAT-IFN-γ- and antigen processing-related pathways in late-stage MSI-H tumors. The top 5 genes with significantly differential expression between late-stage and early-stage tumors are C7, TNFRSF12A, MFGE8(up-regulated) and CXCL5, IFNL1, GZMH, HMGB1 and CXCR1 (down-regulated). Conclusions: There are significantly fewer cytotoxic and dendritic cells in late-stage MSI-H CRC, compared to early-stage MSI-H CRC, which is associated with decrease expression in JAK-STAT-IFN-γ- and antigen processing-related pathways. Abstract CRISPR-based functional genomics screening can be designed to identify novel cancer cell intrinsic targets that increase tumor immunogenicity. Using a FACS-based CRISPR sorting screen for PD-L1 expression, we identified Euchromatic histone-lysine-N-methyltransferase 1 and 2 (EHMT1/2) as negative modulators of the interferon signaling pathways. EHMT1 and EHMT2 are histone methyltransferases that mono- and di-methylate lysine 9 of histone H3 to repress gene transcription of defined target genes. Gene knockout or pharmacological inhibition of EHMT1/2 in cancer cells resulted in de-repression of gene promoters, upregulation of interferon-stimulated genes (ISGs), and secretion of pro-inflammatory cytokines. Here, we present the preclinical characterization of TNG917 - an oral and highly selective EHMT1/2 inhibitor with low nanomolar cellular potency, and favorable pharmacodynamic and pharmacokinetic properties. In humanized and syngeneic mouse models, treatment with TNG917 in combination with anti-PD1 promoted a T-cell-infiltrated tumor microenvironment, led to significant anti-tumor activity, and resulted in survival benefit. In summary, our in vitro and in vivo studies provide a rationale for the clinical development path of TNG917, in combination with checkpoint inhibition, in patients with immune cold tumors. Abstract Background: Isocitrate dehydrogenase 1/2 (IDH1/2) is mutated in a subset of cholangiocarcinoma (CCA), gliomas, and other solid tumors. LY3410738 is a potent, selective, covalent, dual inhibitor of IDH1/2 mutations (IDH1/2m). LY3410738 binds covalently at a novel binding site, enabling continued potency in preclinical models in the setting of second site IDH resistance mutations. We present initial results from the first-in-human phase 1 study of oral LY3410738 in patients (pts) with IDH1/2m CCA, and IDH1m glioma or other solid tumors. Methods: Dose escalation (3+3 design) evaluated LY3410738 monotherapy in advanced IDHm CCA and other solid tumors (NCT04521686). Key objectives included determining the RP2D, safety, PK, PD (inhibition of plasma D-2-HG), and preliminary antitumor activity. Results: As of 28 July 2022, 80 pts including 42 with CCA (33 IDH1m, 9 IDH2m), 27 with glioma (IDH1m), and 11 other tumor types (IDH1m) received LY3410738 dosed at 25-600 mg QD or 300 mg BID. Pts were median 52 years of age (range, 23-80) with a median of 2 prior therapies (range, 1-7). 19% of CCA pts had received prior IDH1 inhibitor. Median time on treatment was 3.7 months (range, 0.1-19). No DLTs or treatment related deaths were observed; the MTD was not reached. Treatment emergent adverse events (TEAEs) ≥15% included nausea (35%), vomiting (21%), and decreased appetite (19%) and were mostly grade 1-2. Most frequent grade ≥3 TEAEs &gt;2% were anemia (4%), cholangitis (3%), headache (3%), decreased lymphocyte count (3%), and hyponatremia (3%). LY3410738 exposure was dose proportional. In pts with IDH1m cancers, LY3410738 achieved sustained D-2-HG inhibition at all dose levels, including in pts who received prior IDH1 inhibitor. In pts with IDH2m cancers, a higher dose (≥150 mg daily dose) was required for D-2-HG inhibition. Among the 42 pts with R/R CCA, the best response included 1 PR and 22 SD. Of the 22 glioma pts with contrast enhancing tumors, best response included 3 PR and 9 SD. Conclusions: LY3410738 demonstrated a favorable safety profile with potent and sustained D-2-HG inhibition in pts with IDH1/2m advanced solid tumors. Consistent with the expectations for IDH inhibitor monotherapy in this setting, CCA and glioma pts exhibited prolonged stable disease. RP2D evaluation is ongoing. Penelitian dilaksanakan di IP2TP Bajeng Balai Penelitian Tanaman Serealia Kabupaten Gowa bulan April sampai dengan September 2021. Sebanyak 8 hibrida dan 2 varietas pembanding ditanam pada rancangan tersarang tiga ulangan. Genotipe tersarang pada taraf pupuk N. Taraf pupuk N meliputi 100 kg N ha-1 dan 200 kg N ha-1. Indeks toleransi meliputi Tolerance (TOL), Mean Productivity (MP), Geometric Mean Productivity (GMP), Harmonic Mean (HM), Stress Tolerant Index (STI), Relative Tolerant Index (RTI), Stress Susceptibility Index (SSI), Yield Index (YI), Stress Relative Index (SI), Yield Stability Index (YSI), Stress Susceptibility Percentage Index (SSPI). Hasil penelitian menunjukkan bahwa indeks toleransi menunjukkan bahwa MP, GMP, HM dan STI merupakan yang paling sesuai untuk menyeleksi hibrida toleran N rendah. Eight promising hybrid and two check varieties were planted in a nested design with three replications. The genotypes were nested at the N fertilizer level. The N fertilizer levels were 100 kg N ha-1 and 200 kg N ha-1. The tolerance index used was Tolerance (TOL), Mean Productivity (MP), Geometric Mean Productivity (GMP), Harmonic Mean (HM), Stress Tolerant Index (STI), Relative Tolerant Index (RTI), Stress Susceptibility Index (SSI), Yield Index (YI), Stress Relative Index (SI), Yield Stability Index (YSI), Stress Susceptibility Percentage Index (SSPI). The results showed that tolerance index MP, GMP, HM, and STI were the most suitable for electing low N tolerant hybrids maize. hybrids HLN 02 and HLN 06 were low N tolerant; meanwhile, HLN 03 and HLN 07 and ADV 777 were susceptible based on yields at 100 kg N ha-1 and 200 kg N ha-1 fertilization, tolerant indexes, and clustering.

Abstract:
Abstract Glioblastoma (GBM) is a common CNS tumor with poor prognosis. The median progression-free survival (mPFS) of standard Stupp regimen was about 6.9 months. The 2-THE-TOP study suggested that a triple regimen of temozolomide (TMZ) combined with pembrolizumab and Tumor treating fields (TTFields) could further prolong the mPFS of newly diagnosed GBM to 12.1 months. Here we report a case showing long PFS from this triple regimen. In October 2019, a 60-year-old female patient was admitted to the hospital due to intermittent dizziness and headache with rapid progression of left limb weakness for more than 10 days. MRI showed a space-occupying lesion in the basal ganglia of the right temporal lobe, with obvious peritumoral edema involving the basal ganglia, posterolateral thalamus, and cerebral foot, with midline shift, demonstrating a high-grade glioma. Total resection of the tumor was performed on October 30, 2019. Pathological diagnosis was glioblastoma, IDH wild type, WHO grade 4. Next generation sequencing showed negative MGMT promoter methylation, TERT promoter mutation, PTEN mutation, CDK4 and EGFR amplification. TMZ concurrent chemoradiotherapy was initiated on November 19, 2019, followed by triple regimen of TMZ adjuvant chemotherapy (200mg/m2), pembrolizumab (200mg VD q3w) and TTFields in March 2020. During this period, the compliance of TTFields remained above 92%, which means the daily duration of TTFields was above 22.08 hours. Regular MRI review showed that the intracranial condition was stable, and there was no interruption of treatment due to treatment-related adverse reactions. No progression was found in the MRI reexamination in April 2022 but TTFields and TMZ chemotherapy were subsequently discontinued for the patient’s personal reasons, however, pembrolizumab monotherapy was maintained. In July 2022 she was hospitalized due to hemiplegia and the tumor recurrence was revealed in MRI, indicating a PFS of 33 months. She underwent tumor resection again and was histologically diagnosed as recurrent GBM on August 2,2022. After surgery, electric field therapy and immunotherapy were resumed. The patient was well recovered with a stable disease at the time of submitting this abstract. This patient had multiple negative prognostic factors of GBM. But under the combined regimen of electric field therapy, immunotherapy and chemotherapy, the disease control was maintained for up to 33 months, which is significantly prolonged compared to historical data of 12.1 months. The patient maintained TTFields for nearly 2 years, and tumor recurrence occurred 3 months after discontinuation of TTFields, indicating that continuous use of TTFields take great importance on disease control. Overall, our case of long-term PFS provides an important reference for the treatment of GBM patients with TTFields combined with immunotherapy and TMZ chemotherapy. There are significant differences in the dominant driving factors of rocky desertification evolution in different historical periods in southwest karst mountainous areas. However, previous studies were mostly conducted in specific periods. In this study, taking Bijie City as an example, the spatial and temporal evolution pattern of rocky desertification in Bijie City in the recent 35 years was analyzed by introducing the feature space model and the gravity center model, and then the dominant driving factors of rocky desertification in the study area in different historical periods were clarified based on GeoDetector. The results were as follows: (1) The point-to-point B (bare land index)-DI (dryness index) feature space model has high applicability for rocky desertification monitoring, and its inversion accuracy was 91.3%. (2) During the past 35 years, the rocky desertification in Bijie belonged to the moderate rocky desertification on the whole, and zones of intensive and severe rocky desertification were mainly distributed in the Weining Yi, Hui, and Miao Autonomous Region. (3) During 1985–2020, the rocky desertification in Bijie City showed an overall weakening trend (‘weakening–aggravating–weakening’). (4) From 1985 to 2020, the gravity center of rocky desertification in Bijie City moved westward, indicating that the aggravating degree of rocky desertification in the western region of the study area was higher than that in the eastern region. (5) The dominant factors affecting the evolution of rocky desertification in the past 35 years shifted from natural factor (vegetation coverage) to human activity factor (population density). The research results could provide decision supports for the prevention and control of rocky desertification in Bijie City and even the southwest karst mountainous area.

BackgroundRapidly progressive interstitial lung disease (RP-ILD) is often seen in dermatomyositis patients with anti-melanoma differentiation-associated gene 5 (anti-MDA-5) antibody. They often have a poor prognosis with rapid decline in pulmonary function, leading to respiratory failure (1). Aggressive immunosuppressive therapy has been reported with improved prognosis, however; it may lead to opportunistic infections, including cytomegalovirus (CMV) or Pneumocystis pneumonia (PCP) infection (2, 3).ObjectivesThis study aimed to evaluate the effectiveness of tofacitinib (TOF) in combination with CMV and PCP prophylaxis in anti-MDA-5-positive patients.MethodsMedical records of 17 anti-MDA-5-positive RP-ILD patients enrolled during Mar 2017 to May 2021 were reviewed. RP-ILD was defined by the presence of deteriorated dyspnea, with a decrease in PaO2 levels and emerging radiographic anomalies within 4 weeks without evidence of infection (4). Chest CT was scored using Ichikado score (5). Clinical parameters including ferritin levels, white counts (WBC), Lactate dehydrogenase (LDH) levels, GAP scores (Gender, Age, and Physiology score for idiopathic pulmonary fibrosis) were recorded. Medications included cyclophosphamide (CyP), intravenous immunoglobulin (IVIG), mycophenolic acid derivatives (MPA), rituximab (RTX), and calcineurin inhibitor (CNI). Kaplan-Meier survival analysis and Log-rank test were used to evaluate one-year mortality differences (MedCalc version 19.6). The Ethics Committee approved our study (CE17038B).ResultsSix anti-MDA-5-positive RP-ILD patients were treated with tofacitinib; five had concomitant CMV prophylaxis with valganciclovir (VGCV); 4 had PCP prophylaxis with trimethoprim/sulfamethoxazole (TMP-SMX). Patients’ demographic data are shown in Table 1. The median age, clinical manifestations, laboratory data, and chest CT scores were comparable between tofacitinib and non-tofacitinib groups. Prevalence of MPA use was higher in the non-TOF group. Abstract Background. Colorectal cancer (CRC) is a commonly diagnosed cancer. Glucose intake enhances the secretion of insulin, may contribute to cell growth and proliferation, leading to CRC development. Epidemiologic studies have shown that individuals with diabetes history are at increased risk of CRC. A low-carbohydrate diet, represented by a low-carbohydrate diet (LCD) score, may reduce the risk of diabetes. Yet, little is known about the association between LCD scores and the risk of CRC. Methods. We evaluated the associations between LCD and CRC risk in the Singapore Chinese Health Study, a population-based prospective cohort study that includes 61,321 Chinese men and women in Singapore who were 45-74 years old at enrollment. Food and dietary supplement consumption were assessed by a validated semi-quantitative food frequency questionnaire. Participants were assigned a score from 0 to 10 according to their rankings by daily intake of calories from carbohydrates, fat and protein, respectively. A score of 0 was assigned to a subject ranked in the highest one-eleventh of calorie intake from carbohydrates or in the lowest one-eleventh of calories from fat or protein, respectively. In contrast, a score of 10 was given to each subject in the lowest one-eleventh of carbohydrate intake or the highest one-eleventh of fat or protein intake. The LCD score for each subject was the sum of the 3 individual ranking scores. The highest possible value was 30, which represented the lowest carbohydrate intake and the highest fat and protein. In addition, animal-based and plant-based LCDs were similarly created from the daily consumption of calories from animal or plant protein and fat, respectively, as well as carbohydrates. Cox proportional hazard regression models were used to estimate hazard ratios (HRs) and their 95% confidence intervals (CIs) for CRC according to higher quartiles of LCD scores with adjustment for potential confounders. Results. After an average of 17.5 years of follow-up, 2,140 participants developed CRC. Overall, there was a null association between total LCD and CRC risk. A moderate positive association was found for animal-based LCD with CRC risk. The multivariable-adjusted HRs(95%CIs) for CRC in quartiles 2, 3, and 4 of animal-based LCD were 1.12(0.99-1.26), 1.18(1.04-1.33), and 1.11(0.98-1.25)(Ptrend=0.06) compared with the lowest quartile. There were no statistically significant differences in the animal-based LCD and CRC risk in subgroups stratified by sex, body mass index, smoking status, and history of type 2 diabetes. Conclusions. This large population-based prospective cohort study shows that a high animal-based LCD score, which represented low intake of carbohydrates and high intake of animal protein and fat, was associated with ~10% increased risk of CRC. Our findings suggest that replacing low-carbohydrate food with animal-based food may increase the risk of developing CRC. Abstract Background: IBI351 (GFH925) is an irreversibly covalent inhibitor of KRASG12C. Data from prior data cutoffs (primary: Feb, 2022; initial update: July, 2022) showed that IBI351 (GFH925) was well-tolerated and demonstrated promising efficacy in patients (pts) with advanced solid tumors harboring KRAS p.G12C mutation. Here, we update results of this phase I study evaluating IBI351 (GFH925) in pts with advanced solid tumors again. Methods: Pts with locally advanced, recurrent or metastatic solid tumors with KRASG12C mutation for whom standard therapy had failed were enrolled. Phase I dose escalation had an accelerated titration design for dose level 250mg once daily (QD) and a Bayesian optimal interval (BOIN) design with 450-900mg QD and 450-750mg twice daily (BID). The primary endpoints were safety and tolerability. The secondary endpoints were pharmacokinetics (PK), anti-tumor activity of IBI351 (GFH925) monotherapy per RECIST v1.1, and overall survival. Results: As of November 30th, 2022, 74 pts (1 at 250mg QD, 3 at 450mg QD, 9 at 700mg QD, 5 at 900mg QD, 21 at 450mg BID, 31 at 600mg BID and 4 at 750mg BID; 62 men, 12 women; median age: 64 yrs, range: 42-76 yrs) were enrolled, among whom 67 pts had non-small cell lung cancer (NSCLC). Among 67 NSCLC pts, 44.8% pts received ≥2 prior lines of treatment (tx), 38.8% pts had brain metastases; adenocarcinoma was the most common histology (n=66, 98.5%). All 74 pts were included for safety analysis. No dose-limiting toxicity (DLT) were observed in any dose cohorts. The overall safety profile was consistent with the latest previous report, with no new safety signals identified. As of December 15th, 2022, among 67 response-evaluable NSCLC pts across all dose levels, the ORR (by investigator assessment) was 58.2% (95% CI, 45.5-70.2), and the confirmed ORR was 44.8% (95% CI, 32.6-57.4); the disease control rate (DCR) was 92.5% (95% CI, 83.4-97.5). At the 600mg BID dose level (RP2D), the ORR was 63.3% (95% CI, 43.9-80.1), and the confirmed ORR was 50.0% (95% CI, 31.3-68.7), the DCR was 96.7% (95% CI, 82.8-99.9). With a median progression-free survival (PFS) follow-up of 5.5 months (95% CI, 5.3-6.8) for NSCLC pts at the 600mg BID dose level, 21 (70%) pts were continuing treatments, and the median duration of response (DoR) and PFS were not reached. Thirteen out of 15 confirmed responders were still in response. Conclusion: IBI351 (GFH925) was well-tolerated across all doses explored in patients with advanced solid tumors harboring KRAS p.G12C mutation. The data also demonstrated promising efficacy and durable response of IBI351 (GFH925) in previously treated advanced NSCLC. Clinical trial information: NCT05005234. Abstract Background Iron chelators are used in the treatment of iron overload related diseases and are currently receiving a major attention as potential antitumor drugs. In recent studies, the antitumor activity of thiosemicarbazones-class of iron chelator, including Di-2-pyridilketone-4,4- dimethyl-3-thiosemicarbazone (Dp44mT) has been investigated in over 20 phase I and II clinical trials [1, 2,3]. Iron chelators were also considered as anti-HIV-1 agents. However, the main obstacle to using iron chelators in vivois the deleterious side effect of methemoglobinemia induced by some iron chelators that are able to scavenge electrons from the heme-bound iron in hemoglobin. In our previous studies, we developed novel phenyl-1-pyridin-2yl-ethanone (PPY)-based iron chelators that we showed to increase IKBα expression, modulate CDK2 and CDK9 activities and inhibit HIV-1 [4]. Objective Our objective was to test the effect of PPYeT iron chelator for methemoglobin induction. The methemoglobin induction effect was compared with several additional iron chelators including Di-2-pyridylketone-4,4-dimethyl-3-thiosemicarbazone (Dp44mT) and PPY analogues. Methods Fluorometric analysis was carried out in promonocytic THP-1 cells to evaluate the ability of our novel PPYeT iron chelator to reduce labile iron pool (LIP). The effect of PPYet on LIP was compared to the effect to SIH. Subsequently, spectrophotometric analysis was used to measure and quantify the production of methemoglobin in human red blood cells lysates and in isolated intact human red blood cells treated with PPYeT and various other iron chelators including DP44mT and DP4mT. Results PPYeT significantly reduced LIP in THP-1 cells overloaded with iron comparing to the cells treated with SIH. In RBC lysates and in intact RBC, PPYeT treatment showed notably lesser production of methemoglobin in comparison to DP44mT and DP4mT chelators. In RBC lysates, PPYeT produced about four-fold less methemoglobin than Dp44mT and ten-fold less than Dp4mT. Conclusion The novel compound, PPYeT, shows a remarkably low ability to catalyze the formation of methemoglobin in human RBC lysates and also in intact RBCs as compared to Dp44mT. These findings indicate that PPYeT may be useful for future in vivo studies as it produces less methemoglobinemia. Further studies will evaluate the effect PPYeT as anti-cancer or anti HIV-1 inhibitor in vivo. Steel is a cheap renewable material with many interesting properties but its presence in the electroplating sector is very limited due to adhesion difficulties. Stainless steel contains approximately 10.5% chromium, which forms a surface oxide layer that passivates the steel, making it resistant to corrosion [1]. It is the presence of this surface oxide film that makes it difficult to electroplate steel. The aim of electroplating steel is to refine it by depositing nobler metals of the desired color, such as ruthenium and gold. The activation of steel involves the remotion of the oxide layer. The main method of electroplating stainless steel is generally through the deposition of nickel, but the intensive use of nickel baths makes these processes inappropriate for the sustainability paths taken by the electroplating industry. Even when nickel-based galvanizing processes are not used, it is possible that steel objects do not pass the nickel release test. This is because the steel itself contains nickel, so the removal of the surface oxide layer during the electroplating process may facilitate the release of nickel. The nickel release mechanism is not yet fully clear but bring to allergenic issues in the final product [2]. We performed a systematic study electroplating various metals and alloys on steel to evaluate which one performed better as barrier against the release of nickel and shed light on this issue. The steel substrate was characterized microscopically and spectroscopically before and after the release test. The amount of nickel was evaluated in the solution that simulates human sweat to evaluate its release in µm·cm-2·week-1 according to the standard EN 1811:2015. Berdasarkan masalah yang ada maka tujuan dari penelitian adalah untuk merancang prototipe Sistem Pendukung Keputusan berbasis website untuk Pemilihan Penerima Bantuan Langsung Tunai Menggunakan Metode Multi Objective Optimization on The Basic of Ratio Analysis (MOORA). Sistem ini nantinya dapat membantu aparat desa untuk melakukan seleksi penerima bantuan BLT-DD secara cepat dan objektif. Prototipe Sistem Ini menjadi akan landasan untuk dibangunnya SPK berbasis web untuk penentuan penerima Bantuan Langsung Tunai yang bersumber dari Dana Desa. Sistem yang akan dibangun nantinya mengunakan metode pengambilan keuputusan MOORA (Multi-Objective Optimization on The Basic of Ratio Analysis). Metode MOORA dipilih karena dapat menghasilkan tingkat fleksibilitas dan kemudahan untuk dipahami dalam memisahkan bagian subjektif dari proses evaluasi ke dalam kriteria bobot keputusan dengan beberapa atribut pengambilan keputusan. Prototipe sistem yang dibangun akan menjadi dasar pembangunan sistem kedepannya. Dari hasil perhitungan dapat disimpulkan bahwa Metode Moora dapat merekomendasikan alternatif yang paling sesuai untuk mendapatkan BLT-DD. Penelitian ini mencontohkan 5 alternatif perhitungan. Dari hasil perhitungan didapatkan hasil bahwa alternatif 1 dan 2 merupakan rekomendasi alternatif terbaik untuk penerima BLT-DD.

Abstract:
Abstract Homologous recombination deficiency (HRD) assays determine patient eligibility for treatment with PARP inhibitors and other DNA repair targeting drugs; therefore, understanding variability in how these assays measure and report HRD is critical for patients and providers. HRD assays measure various factors to determine HRD status including causes (i.e., inactivation in HR pathway genes) and consequences (i.e., genomic scarring). Methodological variability across HRD assays has led to a suggestion that the assays may not agree on a per patient basis. An empirical assessment of assay variability may guide our understanding of how to implement “HRD status” as a biomarker. Friends of Cancer Research (Friends) initiated a unique research partnership to assess levels of agreement and variability across HRD assays. We previously presented an analysis of the HRD status of 348 TCGA ovarian cancer samples across 11 assays. Concordance across assays was analyzed by measuring all possible pairings of samples and assays leading to a median (and IQR) positive percent agreement (PPA) of 74% (51-89%) and negative percent agreement (NPA) of 81% (64-92%). The median percent positivity (percent of patients with HRD) was 49% (range 9-67%). However, some groups modified their HRD pipelines to analyze the in-silico data and we were unable to interrogate the influence of patient and sample characteristics on HRD calls. To establish a more comprehensive dataset, we identified 142 archival specimens from the University of Alabama at Birmingham from patients with stage III-IV high grade serous ovarian cancer diagnosed between 2011 and 2022. Full clinical information is available including response to platinum therapy. FFPE tumor from debulking surgery was sectioned for the 99 samples with adequate tissue and underwent DNA and RNA extraction by the Molecular Characterization Laboratory (MoCha) at the NCI Frederick National Laboratory. MoCha shipped identical aliquots of DNA and/or RNA from the 90 samples that passed QC for independent sequencing and HRD measurement by the 13 different assays. Statisticians from the NCI performed statistical analyses to assess concordance across assays. Three of the 13 assays considered mutations in non-BRCA1/2 HR pathway genes and 7 measured gLOH as determinants for HRD status among other factors. HRD calls resulted in a median pairwise PPA of 81% (69-91%) and a median pairwise NPA of 74% (61-89%). The median percent positivity was 53% (range 23-74%). Ongoing analyses will consider how each of the HRD assays predict responsiveness to platinum-based chemotherapy. Additional analyses will consider assay, patient, and sample characteristics that may drive variation in HRD calls. Preliminary findings demonstrate variability in HRD calls across HRD assays, similar to the in-silico analysis.
