Abstract:
Vigilin, a multidomain hn-ribonucleo-K-homologous protein, is part of a ribonucleoprotein complex with cognate tRNA and is found in both the nucleus and the cytoplasm. In an approach to identify genomic regions involved in regulation of the chicken vigilin gene, we carried out transfection studies with a reporter gene in suitable chicken cells. After including a distantly positioned 5′-sequence in the construct, we observed a 10.5-fold increase in luciferase (EC 1.13.12.7) expression compared with basal promoter activity. Accordingly, chromatin analysis of freshly isolated embryonic tendon fibroblasts with high levels of vigilin mRNA expression shows a DNase-I-hypersensitive site (DHS1) localized 2.2 kb upstream of the transcriptional start site. Similarly, phytohaemagglutinin-stimulated lymphocytes with a 4-fold elevated expression of vigilin mRNA compared with resting lymphocytes also exhibited this unique DHS, having switched from that found at 3.3 kb (DHS2) in resting lymphocytes. Furthermore, using gel-retardation experiments with DNA representing either DHS1 or DHS2, a specific interaction with chicken nuclear extracts was seen.

Abstract:
2,3-Dehydrosilybin (DHS) was previously shown to chelate and reduce both copper and iron ions. In this study, similar experiments with 2,3-dehydrosilychristin (DHSCH) showed that this congener of DHS also chelates and reduces both metals. Statistical analysis pointed to some differences between both compounds: in general, DHS appeared to be a more potent iron and copper chelator, and a copper reducing agent under acidic conditions, while DHSCH was a more potent copper reducing agent under neutral conditions. In the next step, both DHS and DHSCH were tested for metal-based Fenton chemistry in vitro using HPLC with coulometric detection. Neither of these compounds were able to block the iron-based Fenton reaction and, in addition, they mostly intensified hydroxyl radical production. In the copper-based Fenton reaction, the effect of DHSCH was again prooxidant or neutral, while the effect of DHS was profoundly condition-dependent. DHS was even able to attenuate the reaction under some conditions. Interestingly, both compounds were strongly protective against the copper-triggered lysis of red blood cells, with DHSCH being more potent. The results from this study indicated that, notwithstanding the prooxidative effects of both dehydroflavonolignans, their in vivo effect could be protective.

This paper presents an exergy-efficiency analysis of low-temperature district heating systems (DHSs) with different sanitary hot-water (SHW) boosters. The required temperature of the sanitary hot water (SHW) was set to 50 °C. The main objective of this study was to compare the exergy efficiencies of a DHS without a booster to DHSs with three different types of boosters, i.e., electric-, gas-boiler- and heat-pump-based, during the winter and summer seasons. To achieve this, we developed a generalized model for the calculation of the exergy efficiency of a DHS with or without the booster. The results show that during the winter season, for a very low relative share of SHW production, the DHS without the booster exhibits favorable exergy efficiencies compared to the DHSs with boosters. By increasing this share, an intersection point above 45 °C for the supply temperatures, at which the higher exergy efficiency of a DHS with a booster prevails, can be identified. In the summer season the results show that a DHS without a booster at a supply temperature above 70 °C achieves lower exergy efficiencies compared to DHSs with boosters at supply temperatures above 40 °C. The results also show that ultra-low supply and return temperatures should be avoided for the DHSs with boosters, due to higher rates of entropy generation.

Abstract:
District heating systems (DHSs) which utilize excess heat play an important role in energy infrastructure in many European countries. In contrast to Europe, the DHS is not common and excess heat is not reused effectively in Japan. Almost all the DHSs in Japan were designed as first-generation district heating (1GDH) systems or 2GDH systems. No 4GDH systems have been introduced in Japan. The present study designs a 4GDH system utilizing excess heat from a wide area of Northern Japan and evaluates its feasibility. First, available heat amounts from two excess heat resources were calculated: waste incineration plants and thermal power plants. Second, heat demand from both residential and commercial sectors was estimated using a 1 km mesh, and a heat load curve was created for each mesh based on load curve data. Third, the DHS was designed with excess heat plants as a supply-side heat resource, and spatial information of the demand side made use of the geographical information system (GIS). Further analysis was conducted on selected DHSs in three cities in order to evaluate those systems’ feasibility based on energy efficiency, CO2 emissions, and economic aspects. The result shows that 70.5 PJ of heat can be supplied by DHS in Northern Japan, replacing imported fossil fuels such as petroleum and LPG with regional excess heat. The designed DHS could supply heat with equivalent costs compared to European countries.

The 1st step in the posttranslational hypusine [Nε-(4-amino-2-hydroxybutyl)lysine] modification of eukaryotic translation initiation factor 5A (eIF5A) is catalyzed by deoxyhypusine synthase (DHS). The eIF5A intermediate is subsequently hydroxylated by deoxyhypusine hydroxylase (DHH), thereby converting the eIF5A precursor into a biologically active protein. Depletion of eIF5A causes inhibition of cell growth, and the identification of eIF5A as a cofactor of the HIV Rev protein turns this host protein and therefore DHS into an interesting target for drugs against abnormal cell growth and/or HIV replication. The authors developed a 96-well format DHS assay applicable for the screening of DHS inhibitors. Using this assay, they demonstrate DHS inhibition by AXD455 (Semapimod, CNI-1493). This assay represents a powerful tool for the identification of new DHS inhibitors with potency against cancer and HIV.

Abstract Abstract 2535 Poster Board II-512 We used a high throughput approach to determine the chromatin profiles of the human β and α globin loci and their upstream and downstream regions in human undifferentiated ES cells, ES cell-derived erythroid cells, human fetal and adult origin erythroid cells and in primary cells and cell lines of endo-meso and ectodermal origins. All DNase I hypersensitive sites of the b-locus were absent in undifferentiated human ES cells except for HS2 of the b-globin locus control region. The chromatin profiles of the β and α globin loci of ES cell-derived erythroid cells were identical to those of fetal liver erythroid cells except that the hypersensitive site of the embryonic globin gene was more prominent. DNase I hypersensitive site 2 of the b-globin LCR, a potent enhancer, was present in all the cell lines and primary lineages we studied, providing direct evidence that it is ubiquitous. Several new erythroid specific DHSs were detected upstream of 5′HS7 of the β-LCR, raising the possibility that they play a role in the regulation of the β globin locus. The region downstream to 3′HS1 was depleted of DHSs except for the previously identified DHS mapping near the breakpoint of HPFH 1. Since DHSs are absent near the breakpoints of deletional HPFHs and db thalassemias and since enhancers are typically DHS positive, our results argue against the hypothesis of imported enhancers in the pathogenesis of deletional HPFH and db thalassemia mutants. All the previously identified erythroid specific DHSs of the α globin locus were absent in human ES cells. The α globin locus of ES cells, however, displayed three very prominent DHSs, which were located almost symmetrically about 40 Kb apart from each other and they were constitutively formed in all the lineages and cell lines we have studied; the 3′ and 5′ DHSs carried CTCF sites by ChIP-Seq assay raising the possibility that they mark the sites of chromatin insulators. Overall these results demonstrate the power of the new high throughput chromatin profiling approaches and their ability to uncover features of chromatin that may be of regulatory relevance. The cystic fibrosis transmembrane conductance regulator gene (CFTR) exhibits a complex pattern of expression that shows temporal and spatial regulation, although the control mechanisms are not fully known. We have mapped DNase-I-hypersensitive sites (DHSs) flanking the CFTR gene with the aim of identifying potential regulatory elements. We previously characterized DHSs at -79.5 and -20.9 kb with respect to the CFTR translational start site and a regulatory element in the first intron of the gene at 185+10 kb. We have now mapped five DHSs lying 3′ to the CFTR gene at 4574+5.4, +6.8, +7.0, +7.4 and +15.6 kb that show some degree of tissue specificity. The DHSs are seen in chromatin extracted from human primary epithelial cells and cell lines; the presence of the +15.6 kb site is tissue-specific in transgenic mice carrying a human CFTR yeast artificial chromosome. Further analysis of the 4574+15.6 kb DHS implicates the involvement of CCAAT-enhancer-binding protein (C/EBP), cAMP-response-element-binding protein (CREB)/activating transcription factor (ATF) and activator protein 1 (AP-1) family transcription factors at this regulatory element.

A determinação das probabilidades de ocorrência de certos níveis de deficiência hídrica do solo (DHS) é fundamental para o planejamento agropecuário e a gestão ambiental. O objetivo deste trabalho foi determinar as funções de distribuição de probabilidades (fdp) que melhor caracterizam a distribuição dos valores de deficiência hídrica no solo. As DHSs decendiais e mensais foram determinadas por meio de balanço hídrico diário do solo para as diferentes capacidades de armazenamento de água disponível dos solos da região de abrangência da estação meteorológica de Santa Maria. Verificou-se que os valores mensais e decendiais de DHS se ajustam às funções de distribuição de probabilidades lognormal, gama, exponencial e Weibull. Utilizando-se as fdp selecionadas para cada período do ano, é possível determinar a probabilidade de ocorrência de qualquer valor de DHS para os solos da região de abrangência da estação meteorológica de Santa Maria.

Currently, the district heating systems (DHSs) are intensively promoted both nationally and globally. The advantages of using these systems in urban areas compared to individual heating systems are practically indisputable. Still, it is essential that the calculation underlying the assessment of the economic profitability of the projects to connect the new heat consumers to DHS be made correctly by taking into account all the necessary investments and the benefits obtained from the additional amount of energy sale. The article presents the methodology for the optimal regression model selection that can be applied to predict the additional electricity produced in cogeneration mode in case of the new heat consumer connection to the DHS, based on the actual data of the thermal and electrical energy supplied to network from a CHP in the Republic of Moldova. At the same time, it is demonstrated that between the thermal energy supplied to a new consumer connected to DHS and the additional electricity produced in the cogeneration mode, there is a direct and linear correlation.

A well performing District Heating Substation (DHS) is crucial for the efficiency of the District Heating (DH), especially with the shift towards low temperature 4th generation DH systems. For this reason, testing and characterization of commercially available DHSs becomes important to estimate their effect on the DH network. Within the thermo-technical laboratory of EnergyVille, a multipurpose test rig has been built for testing DHSs. In this setup, different DH conditions and heat demand profiles for space heating and for Domestic Hot Water (DHW) can be emulated. Independent tests have been performed on 4 DHSs from three different manufacturers, focused on the DHW preparation for low DH supply temperature and on the stand-by/keep-warm operation of the substations. The latter maintains a certain temperature within the heat exchanger to avoid delays in the delivery of DHW. The results showed that improvements are needed on DHW production for lower DH supply temperatures. Also, enhancements are needed to reduce losses from the keep-warm function. Given that DH systems can have thousands of substations, this will reduce the overall losses and improve the performance of the DH network.

Abstract:
The specific aims of this study were determined on a conceptual appreciation that management of information assets and services in some government agencies was deficient in meeting both existing and developing demands. This appreciation manifested itself in a commitment to investigate the principles and processes intrinsic to existing management methodologies, to relate these processes to the needs of users, and to determine a strategy which could more ably meet the information provision requirements of those users. The achievement of these aims predicated the use of the case study research method, selecting as the first case study the Department of Human Services and Health (DHSH), an agency that had recognised that a problem existed in the provision of information services, and had initiated action to address that problem. As a counter, the Department of Defence, an agency which adrmts to a problem, but which had yet to initiate an active, global program for its resolution, was chosen as the second case study. A theoretical model, which reflects extant international thinking and practice, was initially constructed in order to establish a basis on which to ascertain and evaluate the information management circumstances of the two case study departments. This model specified the objectives considered to be fundamental to effective information management in a public service environment. It included studying the foundation repositories of information services from which information in the portfolio domains of government are sought. These services are the traditional records centres or registries, the library services which provide a repository of published and grey material in printed, image and magnetic formats, and the computerised networks holding electronic records at varying levels of development. An analysis of findings was carried out separately on each case study agency before bringing the data together for cross-case analysis. In order to maximise the veracity and validity of the data collected and its subsequent interpretation by the researcher, the draft analysed case study findings were submitted to the respective agencies for review and critique. All matters of substance received have been incorporated in the final version. The findings from the two case studies and the cross-case analysis confm that, despite significant advances in some specific agencies, the initial hypothesis that government agencies are still deficient in providing optimum services to meet the information needs of users, is demonstrated. The advances that have been made, however, similarly support the other thesis hypotheses that the implementation of an integrated information management strategy in any government agency will provide a foundation for improved information provision and the timely delivery of relevant available information to the user. Finally, a model of optimum processes involved in such a strategy, derived from the theory and practical products of this study, is offered. This could be the subject for future evaluation and testing for realistic and functional application.

The Department of Homeland Security (DHS) leverages information technology to increase the effectiveness of first responders during disaster recovery. At the same time, cyber attacks against these information technologies have significantly increased. Unfortunately, cyber attacks have grown faster than the technologies used to defend them. The reliance on technology coupled with the difficulty of defending it makes it unrealistic to assume that communications will always be available when needed. Therefore, it is critical that first responders are prepared to operate when one or some of their communications abilities are lost. Alarmingly, DHS has the responsibility to prepare first responders to operate during disasters; however, they lack the authority to enforce programs to ensure this happens. This lack of authority affects how first responders communicate and provides gaps in DHS efforts to prepare for disasters. Until DHS has the authority to enforce change across all levels of government, communications will not be guaranteed during disaster recovery operations. However, DHS could leverage communication outages during operational exercises to better prepare first responders. This thesis explores DHS exercises on the federal, state and local levels and how they are preparing first responders to operate through cyber attacks.

Abstract:
In this paper, we briefly describe the automatic model selection which is provided by Autometrics in the PcGive program. The modeler only needs to specify the initial model and the significance level at which to reduce the model. Then, the algorithm does the rest. The properties of Autometrics are discussed. We also explain its background concepts and try to see whether the model selected by the Autometrics can perform well. For a given data set, we use Autometrics to find a “new” model, and then compare the “new” model with a previously selected one by another modeler. It is an interesting issue to see whether Autometrics can also find models which fit better to the given data. As an illustration, we choose three examples. It is true that Autometrics is labor saving and always gives us a parsimonious model. It is really an invaluable instrument for social science. But, we still need more examples to strongly support the idea that Autometrics can find a model which fits the data better, just a few examples in this paper is far from enough.

The Department of Homeland Security (DHS) continues to be scrutinized for its inability to meet its mission mandate as outlined by the Homeland Security Act of 2002 This study looked at two of DHSs most important functionsintelligence and emergency management. Problems that constrain DHSs ability to play a relevant role in the intelligence community stem from limitations imposed on the organization in the early phase of creation; the missed opportunity of adding the Federal Bureau of Investigation to its organizational structure; and the poor relationship it has with state and local fusion centers. FEMA presents a similar set of challenges whereas the agency has served as an independent organization for much of its existence. Including FEMA in the DHS merger has downgraded the agencys ability to prepare and respond to all-hazards. The call is to reestablish the organization as a stand-alone agency with direct links to the president. DHSs inability to effectively perform two of its most important tasks requires lawmakers to review their 2002 decision and decide if an organizational change is in order. They may find that the way forward for DHS is to downsize and refocus its mission on border security.

Background: Sexual and reproductive health education and services are often of poor quality in Indonesia and although sexual intercourse among adolescents is not socially accepted, a change of norms is observable, putting adolescents at risk to acquire unwanted health outcomes. Aim: To analyse associations of behavioural, sociodemographic and informational factors with engaging in sexual intercourse among never-married adolescents aged 15 - 24 in Indonesia and to analyse potential differences in age groups. Methods: Data from 19,649 never-married male and female adolescents who ever went to school, obtained from the 2012 Indonesian Demographic and Health Survey, was used for analysis. Multiple logistic regression was applied to test for significance separately and after adjusting for variables. Results: Among all adolescents, factors associated with engaging in sexual intercourse were: older age, male sex, ever drank alcohol, smoking and ever used drugs. After stratifying for age, the same associations were found for older and younger adolescents. Additionally, among younger adolescents (15 - 19 years), those from poorer households, with a higher educational level and those currently attending school were more likely to engage in sexual intercourse. Among older adolescents (20 - 24 years), those from poorer households and rural areas were more likely to engage in sexual intercourse. Conclusions: Interventions are needed to equip male adolescents, those who ever drank alcohol, were currently smoking or ever used drugs with sufficient knowledge and tools to make informed decisions. Further qualitative research is needed to investigate the behaviour of adolescents towards sexual intercourse more in depth.

Abstract:
O estudo fez comparações entre a intensidade e a distribuição das deformações a que fica submetido o fêmur proximal intacto e o fêmur proximal fraturado e implantado com o sistema parafuso deslizante Dynamic Hip Screw – DHS, em fraturas transtrocantéricas do tipo 31 - A2 – 2. Para isso, foram produzidas deformações, mediante carregamentos cíclicos, em fadiga. As amostras constituíram-se de ossos de fêmur sintéticos, intactos, e de ossos de fêmur sintéticos onde foram feitos cortes em laboratório produzindo situações de fratura do tipo 31 –A2.2. Nas amostras fraturadas foram colocadas as placas DHS simulando situações reais de osteossíntese. Os conjuntos de ossos mais placas foram então submetidos a ensaios de fadiga. Os resultados demonstraram que as presenças da fratura e do implante DHS alteraram a amplitude das deformações em todas as regiões do fêmur. Houve absorção da carga pelo implante DHS em todas as regiões de medição. Na região do colo femoral, o DHS absorveu 98% da carga, na região lateral 97%, no centro medial transversal 48%, no centro medial longitudinal 92%, no centro medial a 45 graus 94%, no calcar, antes da linha da fratura, 80%, na região do calcar, depois da linha da fratura, 81%, na região do segundo parafuso cortical transversal 81%, na região do segundo parafuso cortical longitudinal 73% e na região do segundo parafuso cortical a 45 graus 92%. A região do centro do pino guia e a região do segundo parafuso cortical foram as regiões de maior instabilidade mecânica. Para as condições de fratura do tipo 31 – A2.2 o implante do tipo DHS não absorve totalmente as deformações geradas, apresentando mobilidade do foco da fratura. Pequenas oscilações entre os ensaios causaram modificações importantes nos resultados, indicando que a relação ponto de medição ou a posição exata do implante podem implicar num desgaste ou afrouxamento prematuro do mesmo devido a uma sobrecarga.
The study compared the intensity and distribution of the deformations to the intact proximal femur and the proximal femur fractured and implanted with the slide screw system, Dynamic Hip Screw – DHS, in transtrochanteric fractures, 31 – A2 – 2 type. In this way, deformations were produced, by means of cyclic loads, in fatigue. The samples consisted of synthetic femoral bones intact and synthetic femoral bones that were cut in the laboratory producing fracture situations of type 31 – A2.2 type. In the fractured samples the DHS plates were placed simulating real osteosynthesis situations. The bone sets and plaques were then subjected to fatigue testing. The results showed that the presence of fracture and DHS implant altered the amplitude of deformations in all regions of the femur. The load was absorbed by the DHS implant in all measurement regions. In the femoral neck region, DHS absorbed 98% of the load, in the lateral region 97%, in the medial center – transverse – 48%, in the medial center – longitudinal – 92%, in the medial center – 45 degrees – 94%, in the calcar region, before the fracture line 80%, in the calcar region, after the fracture line, 81%, in the region of the second cortical screw – transverse – 81%, in the region of the second cortical screw – longitudinal – 73% and in the region of the second cortical screw – 45 degrees – 92% . The region of the center of the guide pin and the region of the second cortical screw were the regions of greater mechanical instability. For fracture conditions of type 31 - A2.2, the DHS type implant does not totally absorb the generated deformations, presenting mobility of the fracture focus. Small oscillations between the tests caused significant changes in the results, indicating that the relationship between the point of measurement and the exact position of the implant may lead to premature wear or loosening of the implant due to an overload.

Abstract:
Recent experimental investigations demonstrate that the level of noise generated by double helical synchronous belt (DHSB) is lower than that by straight-toothed synchronous belt (STSB). The present study is to theoretically elucidate the mechanisms whereby DHSB produces lower noise. In the theoretical analysis, a model DHSB is divided into several parallel narrow DHSBs with equal width. Let each narrow DHSB’s helical angle be 0° so each narrow DHSB becomes a narrow STSB. The theory of the one-dimensional sound field is then applied to obtain the standing wave solution of impact sound pressure. The sum of impact sound pressures by the narrow STSBs derived from one pair (left- and right-handed) of helical teeth of the model DHSB represents transmission impact noise. Computational results reveal that the power of impact sound at the damping-frequency decreases as the helical tooth’s angle increases. In addition, the impact sound power decreases as the degree of tooth’s offset of a double helical offset-toothed synchronous belt (DHOTSB) increases. Impact sound power is in the following order from high to low: STSB » DHSB > DHOTSB. In experimental investigations, the noise was in the following order from high to low: STSB » DHSB > DHOTSB, consistent with theoretical prediction. Also, the sound power at high frequency was markedly attenuated in DHSB or DHOTSB, the impact noise is the major sources of noise. Thus, the reduced noise in DHSB and DHOTSB can be explained by the one-dimensional standing wave sound field theory.

In response to increased interest in risk-informed decision making regarding terrorism, EPRI was selected by U.S. DHS and ASME to develop and demonstrate a nuclear sector specific methodology for owner / operators to utilize in performing a Risk Analysis and Management for Critical Asset Protection (RAMCAP) assessment for the transportation of spent nuclear fuel (SNF). The objective is to characterize SNF transportation risk for risk management opportunities and to provide consistent information for DHS decision making. The method uses a characterization of risk as a function of Consequence, Vulnerability, and Threat. Worst reasonable case scenarios characterize risk for a benchmark set of threats and consequence types. A trial application was successfully performed and implementation is underway by one utility.
