No fim da década de 90, o mundo assistiu a um boom do romance policial. Na pós-modernidade, com a retomada dos gêneros literários, as narrativas de mistério ganham força e atingem o mercado editorial brasileiro. Mas o policial chega repaginado para o leitor. Não que os elementos da trama dedutiva ou do noir tenham desaparecido nas histórias contemporâneas, mas o formato conhecido e aceito pelo público vem sendo usado para aprofundar questões da sociedade contemporânea. A presente dissertação procura, portanto, mostrar como as narrativas policiais pensam a sociedade, enquanto lêem a cidade. Para isso, são privilegiados os livros escritos por Luiz Alfredo Garcia- Roza, com o delegado Espinosa como protagonista. Teorias sobre a vida nas metrópoles e a forma como o homem reage aos seus efeitos são articuladas na leitura das representações da cidade. O estudo começa ligando o nascimento do romance dedutivo e o surgimento do noir a grandes transformações que a sociedade e os centros urbanos sofriam naqueles momentos. E procura mostrar que a cidade é mais do que um simples cenário para as tramas se desenrolarem. Analisa a retomada do gênero na pós-modernidade e o desenvolvimento do policial no Brasil. E investiga, ainda, a presença do policial na Internet, percorrendo sites dedicados ao gênero. Tão ligada à paisagem urbana, as narrativas de mistério tomaram também o terreno virtual, aparentemente sem fronteiras.
In the end of 90 s the world observe a boom of detective stories. In the post-modernity the literary kinds of narratives were very used again and the mystery stories were a success in the publisher market, even in Brazil, country where this kind of narrative never developed. But the detective stories, in this period, started to present some differences. Not that the classic elements weren t there, but, nowadays, they discuss aspects of the contemporary society. This work will observe how this kind of literature thinks the society and talk about the city life at the same time. The books of Luiz Alfredo Garcia- Roza, with Espinosa detective as the protagonist, were privileged in this work. Theories about the life in the contemporary cities and how people react to the effects of this kind of life were articulated with the reading of the metropolis representations. This study shows how the beginning of the detective stories were related with the big transformations that society were living in those days. The cities never were just a stage setting for the detective and his investigation. This work discuss this kind of narrative in the pos-modernity and the late success in the brazilian publisher market. And, because the detective narrative was always so connected with the city, the study observed too how those stories are in internet.

Abstract:
Walter Mosley reappropriates the detective conventions to represent the American society of the 1950s and 1960s from a marginal perspective. He creates a black private eye, Easy Rawlins, whose profile mirrors that of his white counterparts but also subverts it.This thesis analyzes Raymond Chandler's canonical work "The Simple Art of Murder" and establish those traits that characterize Easy Rawlins. Likewise, it compares Mosley and Chester Himes's black detective heroes and highlight the traits that they have in common. Secondly, focusing on the perspective on identity, consciousness, and subjectivity of black scholars such as Stuart Hall, bell hooks, Cornel West, Henry Louis Gates, Jr. and W.E.B. Du Bois, along with the post-colonial approach of critics such as Bill Ashcroft, Gareth Griffiths, Helen Tiffin, Robert Young, and Homi Bhabha among others, it provide the necessary concepts to discuss Easy's profile from a postcolonial angle. Easy's gradual formation of identity throughout the ten novels can be analyzed from when he is nineteen in Gone Fishin' (1997) until he is forty six years old in Cinammon Kiss (2005). Evidently, his development is affected by the different personal situations he has to go through, and the historical time that he happens to live as a black man through the 50s and 60s. The subject is culturally and socially constructed, and Easy is no exception. Although cultural codes are shared and human beings live together, every individual is an independent self with a unique perspective and a single first-person ontology. In Easy's case, this thesis argues that there are three main aspects that shape the character's subjectivity: his role as a detective, his post-colonial consciousness as a black man raised in a society dominated by whites, and finally, his attachment and defense of a strong African American culture.
Walter Mosley se apropia de la novela negra para mostrar una visión de la sociedad norteamericana desde una perspectiva marginal. Su detective, Easy Rawlins, tan duro, desconfiado, inteligente e intrépido como Phillipe Marlowe o Sam Spade, no solo se dedica a resolver los crímenes y delitos que ocurren en los barrios negros de la ciudad de Los Angeles, sino que también reflexiona sobre su condición de hombre negro en una sociedad caótica dominada a todos los niveles por los blancos. Contextualizada en la década de los 50 y los 60 en Estados Unidos, Easy Rawlins es sujeto de la opresión y el racismo que la mayor parte de los afroamericanos sufrieron durante esa época. La obra de Mosley es parte de lo que diversos autores consideran un discurso postcolonial, un proceso continuo de resistencia y construcción en el que, primero, desmiente los estereotipos sobre la inferioridad de los negros tradicionalmente introducidos por la sociedad dominante; segundo, subvierte y reconstruye los conceptos de la identidad y de la cultura afroamericana; y por último, condena el trato y la falta de justicia a la que la comunidad negra era sometida. Tomando como punto de partida los conceptos de identidad, conciencia y subjetividad de los críticos Stuart Hall, bell hooks, Cornel West, Henry Louis Gates Jr. y W.E.B. Abstract:
This essay is a gender analysis of some of the characters created by Agatha Christie. The aim is to investigate the occurrence of gender stereotypes and if traditional gender norms are challenged by men and/or women characters in Evil Under the Sun and The Body in the Library. Initially, there is an introduction to gender theories which includes the theorists Thomas Laqueur and Simone de Beauvoir among others. Laqueur presents the evolution of gender theories, specifically the two-sex model in which it was believed that sex is pervasive and thus the body rules the mind. Theories presented by him are vastly different from those of de Beauvoir who argues that the mind and body are separate. Other theorists which specifically deal with crime fiction and gender include Susan Rowland, who writes about the reformation of the detective and how the crime genre is gendered, and Gill Plain, who has been able to categorised Christie’s characters into three distinctive groups in which gender roles are clearly distinguishable. The conclusion reached is that male and female characters are portrayed both as adhering to gender roles, contemporary to the time at which the books were written, and as breaking them. However the outcomes and reactions differ depending on the character in question. Marple and Poirot appear to be well aware of the stereotypical way they are viewed and use it to appear less intimidating in order to hide their true nature as two sharp intellects.

Abstract:
This thesis examines the eight detective novels produced between 1929 and 1952 by the Scottish writer best known as Josephine Tey. On this side of the Atlantic they are largely out of print and seem almost forgotten. In America, by contrast, her work has grown in reputation and receives respectful academic attention. Critics there examining the novels point to unusual elements in them which they cannot explain. This thesis considers briefly her work as a playwright, novelist and biographer written in the name of Gordon Daviot. Its central concern however, is to address the problems perceived by critics in the detective novels she wrote, by exploring what place the books might occupy in the context of the tradition of Scottish literature. Seen thus, these narratives emerge as selfconsciously literary structures, full of allusion, woven from other texts and underpinned by the use of symbol. Issues of class, gender and sexuality are treated boldly. The detectives created ar e themselves unstable and their own stories and uncertain fates assume as much importance as the cases they solve. Conclusions are bleak and ambiguous rather than reassuring and the prevailing tone is one of irony darkening on occasion to satire. Throughout, Tey's texts interrogate the nature of Scottish experience and identity, engaging with the debate raging between other writers of the time and relevant still. She appropriated techniques of modernism which at the time seemed inimical to the genre of detective fiction. There is an autobiographical subtext. Finally, these sophisticated narratives locate themselves in the tradition of Scottish writing by women, a tradition only now, almost half a century after her death, achieving recognition. Specifically, clear connections are traced between her work and that of Catherine Carswell. This radical reassessment of Tey's detective novels shows them to stake a claim to a position in the mainstream of the tradition of Scottish literature.

This study explores a literary comparative examination of crime between Africa and Scandinavia, with special attention to Botswana and Norway. Smith’s and Nesbo’s selected novels are used as primary texts for analysis. The novels are, therefore, set in two different areas. These writers depict crime from the African and European perspectives. Chapter One deals with a brief introduction, and the aim and objectives of the study. It also expands on the theoretical background and provides definitions of terms that are used in this paper. Chapter Two presents views from various scholars on crime. This study is based on an Afro-Western approach of literary analysis. In other words, there are thoughts by both African and Western writers which assist in determining possible and noticeable similarities and differences, on the issue of crime. Chapter Three analyses crime from an African perspective while Chapter Four discusses crime from a Western point of view. Each of these chapters reflects on crime through character portrayal and depiction within its context. Chapter Five is a comparative analysis of both novels. The chapter identifies possible similarities and differences, mainly of the depiction of crime in different settings – Africa and Scandinavia, committed by blacks and whites. However, the structural and linguistic approaches of both the novels are also reviewed, assisting in discovering the life, in comparison, of the authors. The last chapter (Chapter Six), is a conclusion of the study and future suggestions. Basically, the study argues that blacks only should not be portrayed as perpetrators, but that whites too can be culprits. Again, there should be an equal of measurement on the weight and honour of the two races. Lastly, the moral is that without considering skin colour, financial and social backgrounds, justice must be served equally. Hence, whoever is caught in any form of wrongdoing, they must be given the appropriate punishment – regardless of race, colour, religious creed, gender, financial and social background. This doctoral thesis titled A Foreignising or a Domesticating Approach in Translating dialects? Andrea Camilleri s detective novels in English and The Simpsons in Italian is divided into four parts: 1) an introductory chapter dealing with the most noteworthy translation theories developed in the Western World over the centuries, with a special focus on Translation Studies; 2) a second part analysing two detective novels by Andrea Camilleri, in particular those passages that, because of the high concentration of diatopic, diastratic and diaphatic varieties and the combined use of standard and dialect in the same segment of the text, represent a serious challenge to the translator. This analysis (whose aim is to detect the functions of these linguistic variations) and the skopos the translator wants to maintain in the target text, will be followed by comparisons between the original excerpts and their respective English translations. The basis of my research consists of two detective novels by Andrea Camilleri, one of the first to be translated into English (The Voice of the Violin, first published in Italian in 1997 and in English translation in 2003) and one of the latest (The Wings of the Sphinx, first published in Italian in 2006 and in English translation in 2009). By comparing them to their respective translations, I will be able to say which aspects of the original texts Sartarelli has opted to maintain (precisely which kind of fidelity he has opted for) and whether he has adopted a foreignising approach to preserving the linguistic and cultural peculiarities of the way in which Camilleri uses Sicilian dialect and if over the years, Sartarelli's translating method has changed. Indeed, I consider the foreignising approach the best and perhaps the only way to deal with such culturally-connotated texts. For instance, one of the funniest characters in Camilleri s novels is the semi-illiterate Agatino Catarella whose idiolect merges Sicilian dialect and Italian, a strange mix that linguists define as popular Italian ; 3) A third chapter, specular to the first, which focuses on Screen Translation (dubbing and subtitling), and on the most interesting contributions to the field with a particular focus on Italian dubbing. Both case studies will be exemplified by the contrastive analysis of those excerpts that may represent a challenge to the translator; 4) In the fourth and last part by contrast, the problem of translating culture is treated from a different point of view: the source language (English), the medium conveying the message (the dubbing process imposes various constraints such lip and paralinguistic synchronization) and the translation approach change. The case study analysed in this part of the thesis is represented by some episodes of The Simpsons, an animated American television serial that is broadcast in many countries. The cartoon is ideal for this purpose, since it is extremely humorous and its humour is based on elements of the cultural context and on the exploitation of linguistic varieties. The Simpsons Italian dubbing is in fact one of the best examples of the domesticating method , or rewriting of the dialects, sociolects and idiolects of the famous cartoon. For instance, Italian screen translators had to deal with and to maintain the cultural connotations of for instance, Italian-American spoken by some mafia characters like Fat Tony and his goodfellas, and to totally recreate the connotations of some other original accents. Finally, the contrasting analysis conducted between original and Italian translations of the three episodes selected (namely The Last of the Red Hat Mamas, The Italian Bob and The Color Yellow) will be fundamental to shed light on the translating approach adopted for such a cultural- and linguistic-based humour, so that tendencies in the translation of humour and of cultural references in audiovisual texts can be pinpointed.

Abstract:
Rex Stout maintained his popular readership despite the often controversial and radical political content expressed in his detective fiction. His political ideals often made him many enemies. Stances such as his ardent opposition to censorship, racism, Nazism, Germany, Fascism, Communism, McCarthyism, and the unfettered FBI were potentially offensive to colleagues and readers from various political backgrounds. Yet Stout attempted to present radical messages via the content of his detective fiction with subtlety. As a literary traditionalist, he resisted using his fiction as a platform for an often extreme political agenda. Where political messages are apparent in his work, Stout employs various techniques to mute potentially offensive messages. First, his hugely successful bantering Archie Goodwin-Nero Wolfe detective duo—a combination of both the lippy American and the tidy, sanitary British detective schools—fosters exploration, contradiction, and conflict between political viewpoints. Archie often rejects or criticizes Wolfe's extreme political viewpoints. Second, Stout utilizes the contradictions between values that occur when the form of detective fiction counters his radical political messages. This suggests that the form of detective fiction (in this case the conventional patterns and attitudes reinforced by the genre) is as important as the content (in this case the muted political message or the lack of overt politics) in reinforcing or shaping political, economic, moral, and social viewpoints. An analysis of the novels The Black Mountain (1954) and The Doorbell Rang (1965) and the novellas "Not Quite Dead Enough" and "Booby Trap" (1944) from Stout's Nero Wolfe series demonstrates his use of detective fiction for both the expression of political viewpoints and the muting of those political messages.

Esta dissertação apresenta um estudo comparativo tendo como objetos de investigação os romances A varanda do frangipani (2007), de Mia Couto, e O filho da mãe (2009), de Bernardo Carvalho. Considerando que os dois romances se apresentam como narrativas ligadas ao gênero literário romance policial, buscou-se analisar de que maneiras tais romances apresentam modalizações e reformulações em relação aos aspectos formais e estruturais deste gênero. A partir destas constatações, a investigação debruça-se sobre os conflitos gerados pelas relações de gênero, enfatizando os encontrados na elaboração das identidades masculinas. Por fim, avalia-se de que maneira a violência se apresenta nas narrativas, tendo como princípio que nelas o crime fora suplantado por outras demandas.
This dissertation presents a comparative study that has as its main subject the novels \"A varanda do frangipani\" (2007), by Mia Couto, and \"O filho da mãe\" (2009), by Bernardo de Carvalho. Taking into account that both novels present themselves as narratives pertaining to the crime romance literary genre, the study analyses in which manner these romances present modalization and reestructuring in respect to the formal and structural aspects of the genre. From these findings, the investigation delves into the conflicts created by gender relations, emphasizing those found in the elaboration of masculine identities. At last, it is assessed in which manners violence is presented in these narratives, having as a principle that in them crime has been superseded by other demands.

Abstract:
This dissertation examines detective novels for children and young adults written and published in the German Democratic Republic (GDR). The aim of the thesis is to study how the genre developed under the conditions of a socialist society. The analysis of the 66 texts included in the corpus is based on a socio-historical approach assuming that dialogical interdependencies between texts and society exist and can be verified. Central to the analytical work with the texts is the thesis that detective novels written for young readers reflect the socio-political development in East-German society. It shows, however, that—because of their strong didactic impetus—the texts did generally not, like detective novels for adults, develop into a forum for socio-critical discussions. In the diachronic development, which extends from the beginning in the Soviet occupation zone to the post-reunification period, it is shown that changing socio-political conditions interact with the texts, which becomes particularly obvious in the changing presentation of the detectives and criminals. Studying the texts, the dissertation presents basic research and an overview of the genre. Ten texts from the corpus are subject to a detailed analysis in order to deepen the general insights with examples. This way, different aspects of detective novels for children and young adults in the GDR are emphasized, e.g. the interaction between text and illustration. The embedding of figures in a socialist community produces further motives frequently occurring in the texts such as: the Heimat motive or the anti-fascist society. Although the majority of the texts do not go beyond stereotypical representations of characters, criminal cases, and locations—and hence demonstrate the close link between (normative) ideas of society and their literary implementation—the body of texts contains some innovative exceptions in which the social development is questioned and even cautiously criticized.

Corruption is one of the most devastating problems in Colombia because it affects the social, political and ethical dimensions. This problem is represented through symbolic prac-tices that question the nature of Colombian culture and its change. The crime novel is one of these forms of representation. But the verbal story is recreated, adapted or rewritten by Colombian films. Those recover the images and imaginary in the background of the cultural universe. The question that guided the research is: What are the imaginary and images of corrup-tion in the novels Perder es cuestión de metodo, by Santiago Gamboa, Satanás, by Mario Mendoza and Paraíso Travel, by Jorge Franco and in the homonymous films by Sergio Cabrera, Andi Baiz and Simon Brand? To answer this question, the research develops three sections. The first deals with the Colombian socio-cultural context recreated and reinterpreted in these novels and films; the second concerns the social, psychological and physiological dimensions of the imaginary and the images of corruption, and the third focuses on the process of rewriting verbal texts into film texts. We adopt a semiotic approach, both theoretical and methodological, because literary and film practices correspond to two different modes of production and to two languages each having its own characteristics. The analyses show that if the text statements on corrup-tion and evaluations differ in the form of the expression they share the contents

The primary objective of this M.A. thesis is to examine the portrayal of Switzerland and the role of the Swiss detective in the modern Swiss crime novel, with special focus on the works of three modern Swiss authors of different social status: Friedrich Glauser, Friedrich Durrenmatt and Hansjorg Schneider. While the crime novel is generally considered trivial entertainment for mass audiences within the realm of German literature, the case is somewhat different in Switzerland, a country with a small state mentality. The forthcoming analysis will demonstrate how these authors employ the crime novel as an educational device to convey a very important message to their fellow countrymen about the society in which they live. In their portrayal of Switzerland, the authors cover a wide range of circumstances relevant to their respective time periods, often dealing with controversial issues. Consequently, the Swiss detective plays a major role, as he must often solve difficult cases while faced with tremendous pressure from society. By focusing exclusively on Switzerland, this analysis will ultimately prove that the modern Swiss crime novel contains not only an entertainment aspect, but also important political, sociological and historical elements that distinguish the phenomenon from its international counterparts.

This thesis constitutes the first attempt to examine formally the use of self-referential forms in the detective genre. By focusing detective fiction’s self-referential invocation of the genre within its narratives, it explores the relation between generic boundaries and the boundaries between reality and fictionality. Because the self-referential moments in detective fiction maintain the realistic representation of the narrative frame, they unselfconsciously indicate the textuality of the detective form, so they never wholly expose the disjuncture associated with metafiction. This creates an impression rather than an awareness of metafictionality. These self-referential moments in detective fiction directly relate to critical explication of metafiction because they negotiate the boundaries of reality and fictionality, particularly as implied in fictional narrative. Since these forms appear throughout detective fiction, my project tracks this self-referential examination of the boundaries of reality and fictionality across subgenre. As this examination continues throughout these forms, self-referentiality in detective fiction suggests that the nature of reality is the one mystery that the detective genre has not— and perhaps cannot—solve. To explore this, Chapter One considers self-referential statements that explicitly acknowledge detective fiction and its tropes, which I call overt self-referentiality. Chapter Two broadens the criteria, examining intrageneric intertextuality, where the texts refer to classic examples of detective conventions. Chapter Three explores the self-referentiality implicit in the figure of the detective protagonist who is a detective writer. The self-referentiality in these moments metafictively engages with the boundaries of text and criticism and of reality and fictionality. By considering how these moments work simultaneously to construct and deconstruct the boundaries of the genre, this study of self-referentiality provides a method for considering deviations as a means of underscoring, rather than simply undermining, our understanding of what constitutes a novel. As it exposes the critical analysis of literary construction embedded within the detective genre, this thesis challenges both the division between the popular and the literary and the dominant association of metafictionality with experimental art, revealing the philosophical debates about the nature of reality in literary realms not traditionally considered as metafictional.

The use of surface-enhanced Raman spectroscopy (SERS) as an analytical tool has gained increased interest in recent years due to its greater sensitivity and “fingerprinting” ability when compared to other spectroscopic techniques. This thesis discusses approaches used to detect aggregated and single metallic nanoparticles in a high throughput fashion using SERRS. This was accomplished by building a sensitive optical detection platform with high temporal resolution detectors. Initially, nanoparticle aggregates compartmentalized within microdroplets were detected. Each microdroplet can be considered an individual microreactor. Microdroplet flow rate was changed to characterize the effect on microdroplet throughput. Multiple full spectra acquisitions were taken within each microdroplet which was used to compare the different aggregate geometries within each droplet. In addition to segmented flow, single particles in a continuous flow environment are also detected and characterized. A flow based system significantly increases nanoparticle throughput through the probe volume reducing acquisition time. Using correlation spectroscopy, the size of the nanoparticle can be determined and the effect of flow rate on the nanoparticle flow time can be seen. As another detection strategy, single metallic nanoparticles are also detected optically as they translocate through a solid state nanopore. The high electric field generated at the nanopore electrophoretically drives the charged nanoparticles through the nanopore. The nanopore confines the particles to a single channel ensuring single particle detection. Besides the vibrational information that SERS is able to extract, coupling metallic nanoparticles with metallic nanoparticles creates an additional electromagnetic enhancement with increases the SERS signal.

Abstract:
The animal-vehicle collision has been a topic of concern for years, especially in North America. To mitigate the problem, this thesis focuses on animal detection based on the onboard camera for intelligent vehicles. In the domain of image classification and object detection, the methods of shape matching and local feature crafting have reached the technical plateau for decades. The development of Convolutional Neural Network (CNN) brings a new breakthrough. The evolution of CNN architectures has dramatically improved the performance of image classification. Effective frameworks on object detection through CNN structures are thus boosted. Notably, the family of Region-based Convolutional Neural Networks (R-CNN) perform well by combining region proposal with CNN. In this thesis, we propose to apply a new region proposal method|Maximally Stable Extremal Regions (MSER) in Fast R-CNN to construct the animal detection framework. MSER algorithm detects stable regions which are invariant to scale, rotation and viewpoint changes. We generate regions of interest by dealing with the result of MSER algorithm in two ways: by enclosing all the pixels from the resulted pixel-list with a minimum enclosing rectangle (the PL MSER) and by fitting the resulted elliptical region to an approximate box (the EL MSER). We then preprocess the bounding boxes of PL MSER and EL MSER to improve the recall of detection. The preprocessing steps consist of filtering out undesirable regions by aspect ratio model, clustering bounding boxes to merge the overlapping regions, modifying and then enlarging the regions to cover the entire animal. We evaluate the two region proposal methods by the measurement of recall over IOU-threshold curve. The proposed MSER method can cover the expected regions better than Edge Boxes and Region Proposal Network (RPN) in Faster R-CNN. We apply the MSER region proposal method to the framework of R-CNN and Fast R-CNN. The experiments on the animal database with moose, deer, elk, and horses show that Fast R-CNN with MSER achieves better accuracy and faster speed than R-CNN with MSER. Concerning the two ways of MSER, the experimental results show that PL MSER is faster than EL MSER and EL MSER gains higher precision than PL MSER. Also, by altering the structure of network used in Fast R-CNN, we verify that network stacking more layers achieves higher accuracy and recall. In addition, we compare the Fast R-CNN framework using MSER region proposal with the state-of-the-art Faster R-CNN by evaluating the experimental results of on our animal database. Using the same CNN structure, the proposed Fast R-CNN with MSER gains a higher average accuracy of the animal detection 0.73, compared to 0.42 of Faster R-CNN. In terms of detection quality, the proposed Fast R-CNN with MSER achieves better IoU histogram than that of Faster R-CNN.

La detección precisa y la dosimetría de neutrones en campos de radiación mixtos y pulsados es un tema instrumental demandado con gran interés por las comunidades médica e industrial. Estudios recientes de la contaminación de neutrones alrededor de los aceleradores lineales médicos han aumentado la preocupación sobre el riesgo de cáncer secundario en pacientes sometidos a tratamiento de radioterapia en las modalidades de fotones con energías superiores a 8 MeV. En respuesta a esa necesidad, en esta tesis se ha desarrollado una innovadora alternativa a los detectores estándares con un método activo para medir los neutrones alrededor de un acelerador lineal médico. Para tal fin, se han fabricado y optimizado nuevos detectores de silicio ultra delgados con electrodos 3D. El volumen activo de estos sensores tiene sólo 10 μm de espesor, lo que permite un alto rechazo a rayos gamma, lo cual es necesario para discriminar la señal de neutrones en el campo de la radiación periférica en radioterapia (con un alto fondo gamma). Estos detectores de neutrones son una solución prometedora para estimar el riesgo del paciente, puesto que pueden proporcionar al personal médico una respuesta rápida para una planificación óptima del tratamiento. También pueden ser utilizados en otras áreas con campos de radiación mixtos de neutrones/gamma tales como entornos nucleares y aeroespaciales, o microdosimetría. Además, las características intrínsecas de los dispositivos de silicio como robustez, pequeño tamaño, peso ligero y bajo consumo, los hacen ideales para ser empleados como sistemas de detección de neutrones portátiles. La investigación presentada en este trabajo describe las simulaciones Monte Carlo llevadas a cabo para optimizar el diseño de los prototipos, los procesos de fabricación de los detectores y su caracterización con fuentes radiactivas. Finalmente, se muestra el buen funcionamiento de estos nuevos detectores 3D ultra–delgados de silicio para la detección de neutrones en salas de radioterapia.
The accurate detection and dosimetry of neutrons in mixed and pulsed radiation fields is a demanding instrumental issue with great interest both for the industrial and medical communities. Recent studies of the neutron contamination around medical linear accelerators have increased the concern about the secondary cancer risk for radiotherapy patients undergoing treatment in photon modalities at energies greater than 8 MeV. In this thesis, an innovative alternative to standard detectors with an active method to measure neutrons around a medical linac has been developed in response to that need. Novel ultra–thin silicon detectors with 3D electrodes adapted for neutron detection have been fabricated and optimized for such purpose. The active volume of these sensors is only 10 μm thick, allowing a high gamma rejection, which is necessary to discriminate the neutron signal in the radiotherapy peripheral radiation field with a high gamma background. These neutron detectors are not only a promising solution to estimate patient risk since they may provide medical staff a fast feedback for optimal treatment planning, but expand the functional applications of current neutron detectors for other environments with mixed gamma–neutron radiation fields such as nuclear and aerospace environments or microdosimetry. Moreover, the intrinsic features of the silicon devices like robustness, small size, consumption and weight, make them ideal for portable systems. The research presented in this work describes first the Monte Carlo simulations to optimize the design of the prototypes, secondly the fabrication processes of the detectors, and third the electrical characterization and calibration with radioactive sources of these sensors. Finally, it is shown the good performance of the novel ultra–thin 3D silicon detectors for neutron detection inside a radiotherapy room.

Although DNA microarray can detect multiple DNA samples simultaneously, current detection techniques involve PCR and other traditional procedures. In this study, a sensitive, specific and rapid detection method, which eliminates PCR and other lengthy processes, for pathogenic DNA is presented. This technology is based on the hybridization of target DNA to the immobilized probe, extension of probe DNAs using the target-DNA as a template and signal generation by streptavidin-horseradish peroxidase and substrate. This method is highly specific and sensitive, allowing single-nucleotide-base mismatches discrimination and the detection at femtomole level. The experiments are designed to achieve short hybridization time. Therefore, satisfactory signal can be detected within minutes, allowing the rapid detection of multiple pathogenic DNA. Most importantly, the E. coli genomic DNA can be detected using this technology. In conclusion, this detection method is useful for applications including on-site pathogenic disease detection, crime scene investigation, and pathogen inspection in the environment.

Anomaly detection is a fundamental research topic that has been widely investigated. From critical industrial systems, e.g., network intrusion detection systems, to people’s daily activities, e.g., mobile fraud detection, anomaly detection has become the very first vital resort to protect and secure public and personal properties. Although anomaly detection methods have been under consistent development over the years, the explosive growth of data volume and the continued dramatic variation of data patterns pose great challenges on the anomaly detection systems and are fuelling the great demand of introducing more intelligent anomaly detection methods with distinct characteristics to cope with various needs. To this end, this thesis starts with presenting a thorough review of existing anomaly detection strategies and methods. The advantageous and disadvantageous of the strategies and methods are elaborated. Afterward, four distinctive anomaly detection methods, especially for time series, are proposed in this work aiming at resolving specific needs of anomaly detection under different scenarios, e.g., enhanced accuracy, interpretable results, and self-evolving models. Experiments are presented and analysed to offer a better understanding of the performance of the methods and their distinct features. To be more specific, the abstracts of the key contents in this thesis are listed as follows: 1) Support Vector Data Description (SVDD) is investigated as a primary method to fulfill accurate anomaly detection. The applicability of SVDD over noisy time series datasets is carefully examined and it is demonstrated that relaxing the decision boundary of SVDD always results in better accuracy in network time series anomaly detection. Theoretical analysis of the parameter utilised in the model is also presented to ensure the validity of the relaxation of the decision boundary. 2) To support a clear explanation of the detected time series anomalies, i.e., anomaly interpretation, the periodic pattern of time series data is considered as the contextual information to be integrated into SVDD for anomaly detection. The formulation of SVDD with contextual information maintains multiple discriminants which help in distinguishing the root causes of the anomalies. 3) In an attempt to further analyse a dataset for anomaly detection and interpretation, Convex Hull Data Description (CHDD) is developed for realising one-class classification together with data clustering. CHDD approximates the convex hull of a given dataset with the extreme points which constitute a dictionary of data representatives. According to the dictionary, CHDD is capable of representing and clustering all the normal data instances so that anomaly detection is realised with certain interpretation. 4) Besides better anomaly detection accuracy and interpretability, better solutions for anomaly detection over streaming data with evolving patterns are also researched. Under the framework of Reinforcement Learning (RL), a time series anomaly detector that is consistently trained to cope with the evolving patterns is designed. Due to the fact that the anomaly detector is trained with labeled time series, it avoids the cumbersome work of threshold setting and the uncertain definitions of anomalies in time series anomaly detection tasks.

The ability to detect and interrogate DNA sequences allows further understanding and diagnosis of genetic disease. The ability to perform such analysis of genetic material requires highly selective and reliable technologies. Furthermore techniques which can use simple and cheap equipment allow the use of such technologies for point of care analysis. Described in this thesis are two novel DNA probe systems designed for mutation discrimination and sequence recognition of PCR products. A homogenous PCR system using HyBeacons® which utilise FRET to produce a three probe multiplex system and surface enhanced Raman detection method. Both of these systems allow multiplex detection of PCR products and mutation discrimination by melting temperature analysis. The research reported includes investigations into the effects of different modifications to improve the performance of HyBeacon® probes as well as the effect of different dyes in a FRET system, including unique changes in the optical properties of such dyes. Also a novel method of performing melting temperature analysis using an electrochemical potential is reported. Microfluidic devices are attracting interest for point-of-care diagnostics due to their low unit cost, low reagent and sample usage, fast analysis times and portability. Fluorescence is the most widely used detection method in microfluidics due to its high sensitivity, excellent dynamic range, ease of implementation and non-invasive nature. Fluorescence detection has accordingly been widely used for the interrogation of microfluidic devices, with the majority of reports to date having used non-integrated laser excitation sources coupled with off-chip optics and photodetectors. For point-of-care applications, there is a growing need for self-contained systems which incorporate the microfluidic chip and all associated optical components into a compact, low-cost package. Highly compact systems of this kind would eliminate the need for expensive dedicated bench-top instrumentation. As such they would find multiple uses in the home, ambulance and GP’s surgery, where their ability to provide immediate and/or frequent testing would enable faster, more responsive and ultimately more successful treatment. Work undertaken in this thesis focuses on developing highly sensitive analyte detection systems that would enable the use of cheap optical and electronic components. To accomplish this, two separate methods, based on optical filters and time-gated detection of phosphorescent polymer beads, were investigated. We describe a simple technique for fabricating non-emissive colour filters based on the sensitisation of a highly porous nanostructured metal-oxide film with a monolayer of dye molecules. The resultant filters exhibit much less autofluorescence than conventional colour filters, and are a viable low cost alternative to interference filters. In separate work Ru(dpp)3-doped polystyrene/bisphenol A diglycidyl ether polymer phosphorescent beads developed by Dr. Edwards (Imperial College London) were used to demonstrate the feasibility of highly sensitive of low-cost time-gated detection. This method was applied to a simple biotin assay to show their bio-assay applicability.

Abstract:
Purpose: To evaluate the impact of adding digital breast tomosynthesis (DBT) to digital mammography (DM) on radiologists’ performance, confidence and identification of lesions. Also, to evaluate the radiation dose from DBT and DM and to assess image quality at different dose levels. Methods: Twenty six radiologists examined 50 cases in two modes, DM and DM+DBT. The radiologists were classified into three groups as having no DBT experience, workshop DBT and clinical DBT. Radiologists were asked to localize breast lesions, report their type and give a score of 1-5. The first study examined radiologists’ performance by: sensitivity, location sensitivity, specificity, ROC AUC and JAFROC FOM. The second study, using the same case set, looked at the radiologists’ confidence and their ability to identify lesion type. The third study was a phantom-based experiment to evaluate the mean glandular dose using DBT and DM. Eleven readers reported the visibility of lesions for all phantom images on both modalities. Results: All performance measures were significantly higher for DM+DBT compared with DM AUCs (0.788 vs 0.681, p< 0.0001) and JAFROC (0.745 vs 0.621, p< 0.0001). Similar results were obtained for readers with no DBT and with clinical DBT experience. The confidence of radiologists using DM+DBT was significantly higher than DM in scoring cancer (p<0.0001) and normal cases (p= 0.018). The number of stellate lesions correctly reported on DM+DBT was significantly higher than with DM (p< 0.0001). The radiation dose at a thickness of 50 mm was 13% higher for DBT than for DM. The visibility of lesions was acceptable at 50% mAs for DBT and for DM. Conclusions: Addition of DBT to DM significantly improved: radiologists’ performance whether or not they have DBT experience; radiologists’ confidence and the number of stellate lesions compared with DM. DBT radiation dose is slightly higher than DM dose. However, the exposure may be reduced by 50% mAs with no change in image quality.

Abstract:
The main outcomes of this research are the design of a foreground detection algorithm, which is more accurate and less time consuming than existing algorithms. By the term accuracy we mean an exact mask (which satisfies the respective ground truth value) of the foreground object(s). Motion detection being the prior component of foreground detection process can be achieved via pixel based and block based methods, both of which have their own merits and disadvantages. Pixel based methods are efficient in terms of accuracy but a time consuming process, so cannot be recommended for real time applications. On the other hand block based motion estimation has relatively less accuracy but consumes less time and is thus ideal for real-time applications. In the first proposed algorithm, block based motion estimation technique is opted for timely execution. To overcome the issue of accuracy another morphological based technique was adopted called opening-and-closing by reconstruction, which is a pixel based operation so produces higher accuracy and requires lesser time in execution. Morphological operation opening-and-closing by reconstruction finds the maxima and minima inside the foreground object(s). Thus this novel simultaneous process compensates for the lower accuracy of block based motion estimation. To verify the efficiency of this algorithm a complex video consisting of multiple colours, and fast and slow motions at various places was selected. Based on 11 different performance measures the proposed algorithm achieved an average accuracy of more than 24.73% than four of the well-established algorithms. Background subtraction, being the most cited algorithm for foreground detection, encounters the major problem of proper threshold value at run time. For effective value of the threshold at run time in background subtraction algorithm, the primary component of the foreground detection process, motion is used, in this next proposed algorithm. For the said purpose the smooth histogram peaks and valley of the motion were analyzed, which reflects the high and slow motion areas of the moving object(s) in the given frame and generates the threshold value at run time by exploiting the values of peaks and valley. This proposed algorithm was tested using four recommended video sequences including indoor and outdoor shoots, and were compared with five high ranked algorithms. Based on the values of standard performance measures, the proposed algorithm achieved an average of more than 12.30% higher accuracy results.

La novela Los detectives salvajes, publicada en noviembre del año 1998 es la obra de Roberto Bolaño sobre la que centraremos la investigación del presente informe. En este estudio se pretende leer esta novela desde los conceptos de “sujeto” y de “mediatización” a partir del análisis del tipo de sujeto que se configura en la obra de Bolaño y las relaciones de éste con el “poder” en la sociedad posmoderna. Nos situamos ante la obra de Bolaño como si ésta fuera un espejo contradictorio en el que nos vemos reflejados por personajes tan fascinantes en su caracterización que sólo reconoceríamos como sujetos novelescos pero que, en algunos momentos, viven experiencias tan cotidianas que bien podrían ser las propias. Este primer acercamiento a la variedad y complejidad de personajes que se movilizan entre el Distrito Federal de México y otros lugares geográficos en la novela, propicia la intención de leer esta obra desde el concepto de sujeto -tratando de desentrañar las particularidades de su construcción- y, a partir de su análisis, determinar su lugar en la novela actual.

This study analyzes and accounts for the mixed emotional responses to three Wilkie Collins novels: The Dead Secret (1857), The Woman in White (1860), and The Law and the Lady (1875). Contemporary reviews of these novels, in addition to Collins’s comments on the reception of his works, suggest that these novels elicited both positive and negative emotions in readers who took a “masochistic” pleasure in reading frustrating and often terrifying texts. In the role of detective, Collins’s reader is placed in the uncomfortable and untenable position of disciplinarian (ferreting out secrets and restoring moral order) and of transgressor (reading secret diaries, adopting disguises, eavesdropping, and spying). Collins’s novels portray scenes of masochistic reading: faced with the impossibilities of detection or interpretation resulting from the instability of text, Collins’s characters mimic the reader’s own frustrating interpretive activities. Detection that relies on the text as dependable evidence becomes a psychologically painful activity when texts do not open themselves up to easy interpretation. In addition to the mixed emotional responses produced by acts of detection, the act of reading novels generally (and the sensation novel in particular) has produced strong emotional responses in both literary critics and non-literary observers. The historical context of sensation novel adds an additionally masochistic “layer” or dimension to the reading of these novels. If sensation novels were considered guilty pleasures in of themselves, separate from the guilty pleasures of detection and interpretation of private secrets and criminal activities within the narratives, the guilty pleasure of reading sensation novels would reinforce the reader’s position as transgressor, in addition to his or her position as snooping amateur detective. My project engages existing scholarly works while approaching the subject of emotional responses to literary interpretation (mimicked by the detection in which the novels’ characters are engaged), and while looking at the affective combination of pleasure, terror, anticipation, and guilt, and its role in the popularity of Collins’s novels. This study employs contemporary and historical theories of emotion and masochism as well as scholarship on detective fiction to explain why detection in Collins’s novels is masochistically pleasurable for both readers and characters.

Advanced Driver Assistance System (ADAS) was widely learned nowadays. As crucial parts of ADAS, lane markings detection, as well as other objects detection, have become more popular than before. However, most methods implemented in such areas cannot perfectly balance the performance of accuracy versus efficiency, and the mainstream methods (e.g. Machine Learning) suffer from several limitations which can hardly break the wall between partial autonomous and fully autonomous driving. This thesis proposed a real-time lane marking detection framework for ADAS, which included 4-extreme points set descriptor and a rule-based cascade classifier. By analyzing the behavior of lane markings on the road surface, a characteristic of markings was discovered, i.e., standard markings can sustain their shape in the perpendicular plane of the driving direction. By employing this feature, a 4-extreme points set descriptor was applied to describe the shape of each marking first. Specifically, after processing Maximally Stable Extremal Region (MSER) and Hough transforms on a 2-D image, several contours of interest are obtained. A bounding box, with borders parallel to the image coordinate, intersected with each contour at 4 points in the edge, which was named 4-extreme points set. Afterward, to verify consistency of each contour and standard marking, some rules abstracted from construction manual are employed such as Area Filter, Colour Filter, Relative Location Filter, Convex Filter, etc. To reduce the errors caused by changes in driving direction, an enhanced module was then introduced. By tracking the vanishing point as well as other key points of the road net, a method for 3-D reconstruction, with respect to the optical axis between vanishing point and camera center, is possible. The principle of such algorithm was exhibited, and a description about how to obtain the depth information from this model was also provided. Among all of these processes, a key-point based classification method is the main contribution of this paper because of its function in eliminating the deformation of the object caused by inverse perspective mapping. Several experiments were conducted in highway and urban roads in Ottawa. The detection rate of the markings by the proposed algorithm reached an average accuracy rate of 96.77% while F1 Score (harmonic mean of precision and recall) also attained a rate of 90.57%. In summary, the proposed method exhibited a state-of-the-art performance and represents a significant advancement of understanding.

Early detection and screening have reduced mortality from many cancers, but there remains a need for improved biomarkers of risk. Cytometric DNA ploidy analysis has been used for the detection, treatment, and management of many cancers, but greater clinical utility would come with increased accuracy. Improvements to ploidy-based screening might come from adding complementary biological information. The first aim combined ploidy with the additional biological information provided by malignancy associated changes as detected by automated nuclear morphometry. In 2249 sputum samples, the resultant biomarker, the Combined Score (CS), correlated with lung cancer risk factors like dysplasia grade, age, smoking status, and p53 and Ki-67 immunostaining. CS is a minimally invasive tool for risk assessment for the presence of precancerous lung lesions and could enrich chemoprevention trials with subjects likely to have high-risk dysplasias. The second aim complemented ploidy with biological information provided by immunocytochemistry in a double staining procedure. Testing 49 cervical cytology brushings showed addition of Ki-67 immunostaining to distinguish abnormal cells from normal cycling cells did not improve ploidy’s ability to separate high- and low-grade dysplasias. Nevertheless, double staining with Feulgen thionin and immunocytochemistry was shown to be technically feasible, even with antigen retrieval, and might be applicable to other immunocytochemical stains. Motivated by the ability to combine ploidy with immunocytochemistry, the third aim investigated techniques for biomarker discovery pertinent to cervical dysplasia development. Cervical squamous epithelium consists of a continuum of differentiating cells and carcinogenesis disrupts this cell maturation program. Gene expression differences between the basal and superficial epithelial layers and across various grades of dysplasia could catalyze the discovery of novel biomarkers through a better understanding of carcinogenesis. Microdissection and expression microarray analysis of molecular fixative preserved cervical biopsies resulted in the immunohistochemistry validation of four candidate targets showing correlation with dysplasia grade. This work underscores the importance and potential of accounting for heterogeneity within stratified squamous epithelium and constitutes the first report of successful gene expression microarray analysis of microdissected epithelial layers from molecular fixative preserved paraffin-embedded cervical specimens. Ploidy combined with digital morphometry and immunocytochemistry can generate useful biomarkers of early squamous cell carcinomas.

The occurrence of beer spoilage bacteria belonging to the genera Pectinatus and Megasphaera in ten major UK breweries was investigated. The sampling points were selected from fermentation areas, beer conditioning areas and beer bottling and canning sites. Multiplex PCR methodology was used for detection of three Pectinatus and three Megasphaera species using species specific primers. The presence of six Lactobacillus and three beer spoilage Pediococcus species were also examined. Overall, 117 samples were analysed from ten breweries; six samples were positive for the presence of Pectinatus species and three samples were positive for the presence of Megasphaera species, while 34 samples were positive for the presence of Lactobacillus species and 23 samples were positive for Pediococcus species. Lactobacillus and Pediococcus species appeared to be the major potential spoilage microorganisms. Although none of the actual beer samples were found to be positive for Pectinatus and Megasphaera species, their occurrence in aerobic brewery environments indicates sanitation problems and revealed the presence of highly established biofilms in some breweries. The morphological and physiological characterisation of the presumed Pectinatus and Megasphaera isolates along with NCBI nucleotide blast search assignments and maximum parsimony phylogenetic analysis showed good agreement on their identity. All the Pectinatus and Megasphaera isolates showed ability to spoil low alcohol beer, and the presence of these microorganisms in the brewery environments could be a potential threat to low alcohol and unpasteurised beer. Novel implementation of Hybridisation Protection Assay for detection of beer spoilage microorganisms, Pectinatus and Megasphaera was demonstrated. DNA probes specific for 16S ribosomal RNA were utilised for the detection of beer spoilage bacteria of genera Pectinatus and Megasphaera using a Hybridization Protection Assay. All the probes were modified during synthesis by inserting an amino linker arm at the 5′ end during synthesis and also internally modified by replacing thymidine base with amine modified thymidine base; synthesised probes were labelled with (AE) and purified using reverse phase HPLC. The designed internally AE-labelled probes were able to detect target RNA within the range of 0.016-0.0032 pmol. All the designed probes showed high specificity towards target RNA and could detect bacterial contamination within the range of ca. 5x102 – 1x103 CFU using HPA assay. The developed assay was also compatible with MRS, NBB and SMMP beer enrichment media.
