Full textThis PhD treats a multidisciplinary subject based on the design of a biological micro-sensor prototype for the detection of antibodies of patients susceptible to be allergic.The goal has been the miniaturization of the ELISA "Enzyme Linked Immune Sorbent Assay" method, designing in integrated full-custom colorimetric sensor with a CMOS APS technology. This sensor is installed below a microfluidic system containing the biological test sample, the whole is illuminated via an optical fiber. The sensor detects the light passed through the micro reservoir containing the sample, the induced photo-currents is related to the concentration of the solution. The color change of the enzyme reaction due to the presence of antibodies in the serum, and the quantitative evaluation of the concentration will be determined by the measurement of the induced photo-currents. The sensor may contain a matrix of 20x20 color detector pixels and their reading and control electronics. For cost reasons, we validated the method using a matrix of 4 x 4 pixels of color detectors.The design of electrical device was followed by a colorimetric and electrical characterizations. The latter was used to validate the operation of the control block of the circuit as well as that of the pixel (readout electronics, BDJ). The results brought by measurements are in good agreement with those obtained through simulations. The colorimetric characterization consists in measuring the intensity of the color of two different solutions of different colors. These measurements have shown that our sensor is more sensitive than a spectrometer. Therefore, this research work has contributed to the miniaturization of a colorimetric sensor and its electronic part for the Immunoassay based on the ELISA method

Full textDuring the last twenty year, indoor swimming-pool air quality measurement campaigns confirmed the presence of toxic compounds such as chloroform (carcinogenic) at higher level than those advised by French authorities. Hence, the ability to easily perform direct detection of chloroform at concentration above the advised threshold is crucial to ensure the safety of swimmers.At the moment, no devices seems to meet such a need. This PhD work aims at preparing a colorimetric sensor suited to this application, thanks to a nanoporous material incorporating probe-molecules to specifically react with chloroform and yield a coloured product.The optimization of the Fujiwara colorimetric reaction was carried out and the best mode to be used within the nanopores in the gas phase was identified. A new type of silica-zirconia materials had to be specifically developed to resist to the harsh chemical conditions of the Fujiwara reaction.Fine-tuning of the differential reactivity of silica (slow) and zirconia (fast) precursors led to a good control of the homogeneity, absorbance and porosity of the materials which was absolutely essential to meet the specifications of transparency, porosity, mechanical stability, absorbance and ability to host the Fujiwara reaction.Such materials were successfully doped with Fujiwara reagents, and the functional material obtained was validated as a chloroform sensor

Full textThe goal of this thesis, in the frame of the ANR TRICETOX (CESA, 2013-2017), was to develop some alternative methods to conventional chromatographic methods for the monitoring of the β-triketone herbicides. These new analytical tools have to be easy to handle, fast, low-cost, and to respond to the requirements for the detection of these herbicides (critical value of 0.1 μg.L-1). The β-triketones specifically target the 4-hydroxyphenylpyruvate dioxygenase (HPPD) enzyme, involved in the carotenoids biosynthesis in plants ; its inhibition results in foliage bleaching of the treated plant. Two strategies have been considered, both based on the HPPD inhibition : a whole-cell colorimetric bioassay involving a recombinant E. coli expressing the HPPD from A. thaliana, and an electrochemical enzymatic biosensor. The principle of the bioassay is based on the ability of the recombinant E. coli clone to produce a soluble brown pigment from tyrosine catabolism. The addition of β-triketone induces the decrease of the pigment production in the medium. After optimization of the operational conditions, the LODs obtained for mesotrione, tembotrione, sulcotrione, and leptospermone were 0.069, 0.051, 0.038, and 20 μM, respectively. In the biosensor format, the purified HPPD enzyme was immobilized on the surface of a screen-printed graphite electrode. Using the free enzyme biosensor, LODs as low as 1.36-10 M, 1.25.10-11 M and 1.08.10-11 M were obtained for sulcotrione, mesotrione and tembotrione, respectively. Up to date, real samples have been analyzed using the bioassay and the results were validated by HPLC

Full textAs cyanide is one of the most toxic agents on our planet, it is crucial to detect it accurately, especially in the food industry. The work presented in this thesis concerns the development of new colorimetric cyanide detectors. First, benzoquinonemonoimines were synthesized and their optical properties investigated (in free ligand and complex version). We then focused on the synthesis of azacalixarene-type macrocycles, for which the addition of cyanide in solution gives stable Meisenheimer complexes. The work then focused on the synthesis of m-dinitro-benzenes derivatives incorporating different donor and acceptor groups whose optical properties were studied for the colorimetric detection of cyanide. Full textThe work reported in this thesis focuses on the development of new multiplex analytical devices on biochip or electrode microarray format, dedicated to diagnosis and environmental monitoring. The objective of the first research axis is diagnosis, thanks to the detection in patients’ serum of a panel of antibodies, biomarkers of a pathological state. For that purpose, two immunotests have been developed, enabling the multiparametric detection of specific antibodies by automated and high-throughput analysis of serum samples. This approach is based on the antibodies capture by antigens probes immobilized in a matrix of spots on a membrane surface composing the wells bottom of a micro-titer plate. Enzyme-labeled antibodies have been used, providing a colorimetric detection. This device enabled the achievement of the analysis of 96 samples in less than three hours and has been applied to different applications. The first one consists of allergy diagnosis, and the second focuses on cancer diagnosis. The second part of this work is applied to environmental monitoring, through water analysis. Different types of pollutants have been defined as targets: pesticides, toxins and explosives. In order to integrate them in a matrix of probes, different conjugates have been synthesized with these haptens. After screening and optimization of the conjugates through their reactivity and cross-reactivity with the specific antibodies, the developed device demonstrated his analytical performances in terms of sensitivity and selectivity. Finally, for the European Project BONAS, a last sensor based on water analysis has also been developed. This electrochemical microarray aims to detect explosives precursors, used in improvised explosive devices, for the localization of hidden bomb factory. The chip was designed as a screen-printed electrode network, which was modified by different metals electrodeposition

Full textThe aim of this thesis is to provide the French Metrology with a reference instrument that can be used for measuring the appearance of objects. Several physical properties of object appearance are used not only to identify the object itself but also determine its quality. These include color, gloss, texture and translucency. This work describes the design, characterization and the implementation of an instrument that has been built particularly to measure the bidirectional reflectance distribution function (BRDF) of planar surfaces. BRDF is a physical quantity that enables measuring the surface color and gloss. Accordingly it is important to ensure accurate and traceable measurements of such a quantity in order for a comprehensive understanding of surface properties to be fulfilled.Some surfaces show pigmented, metallic, sparkling or pearlescent appearance. These surfaces, called goniochromatic surfaces, have the ability to change their appearance depending on the viewing angle. To characterize them, it is necessary to deliver surface reflectance measurements at several directions and under different illumination conditions. The instrument that enables these measurements is called gonioreflectometer.The experimental setup we have designed consists of two separate detection lines. The first line, called "color line", is used to study the color. It consists of a stationary illumination source that uses a xenon lamp spectrally resolved by a monochromator. The light coming out from the source is focused on the surface under investigation. The reflected light by the surface is collected by using a lens and directed onto a photodiode by a mirror. This detection system can be moved over a wide angular range so as to measure the BRDF.The second line, called "gloss line", is used to measure the gloss of the surface. In order to be very close to the average human observer, high angular resolution is required. In this line the detection part is stationary. It consists of a Charged Coupled Device (CCD) camera of 512 x 512 pixels. The source used to illuminate the surface is an incandescent tungsten lamp. Angular measurements in this line are provided by the movement of the illumination source itself. Both color and gloss lines share a robot sample-holder that provides six degrees of freedom. Full textResearchers have found a strong correlation between hot summers and the frequency and intensity of forestfires. Global warming due to greenhouse gases such as carbon dioxide is increasing the temperature in someparts of the world. Fires release large amounts of greenhouse gases, causing an increase in the earth'saverage temperature, which in turn causes an increase in forest fires... Fires destroy millions of hectares offorest areas, ecosystems sheltering numerous species and have a significant cost for our societies. Theprevention and control of fires must be a priority to stop this infernal spiral.In this context, smoke detection is very important because it is the first clue of an incipient fire. Fire andespecially smoke are difficult objects to detect in visible images due to their complexity in terms of shape, colorand texture. However, deep learning coupled with video surveillance can achieve this goal. Convolutionalneural network (CNN) architecture is able to detect smoke and fire in RGB images with very good accuracy.Moreover, these structures can segment smoke as well as fire in real time. The richness of the deep networklearning database is a very important element allowing a good generalization.This manuscript presents different deep architectures based on convolutional networks to detect and localizesmoke and fire in video images in the visible domain

Full textThe selective and sensitive detection of heavy metals, such as transition metals, is ofparamount importance for health and safety an environmental monitoring. Current referencemethods, due to their lack of portability, are limiting factors to obtain high-resolution spatialand temporal data. Optical sensors offer an attractive and convenient way to overcome theselimitations of cost and time per analysis by offering real time, on-site measurementcapabilities.In order to demonstrate this potential, this thesis is focused on the detection and quantificationof hexavalent chromium Cr(VI) in water samples by a colorimetric reaction based on areaction with the 1,5-diphenylcarbazide (DPC), that produces a complex possessing anabsorption maximum in the visible range. This works endorse the goal of creating a true labon-chip, integrating both the fluidic function based on ElectroWetting on Dielectric (EWOD)to create the colorimetric reaction, and the sensing function based on the integration of anoptical sensor able to measure absorption variations in micro-volumes (< μL). In order toobtain sufficient sensitivity on such small volumes, optical microring resonators are used inthis work, due to their ability to enhance the effective optical path length by constructiveinterferences.This thesis describes the conception and fabrication of the EWOD microfluidic platform, aswell as the conception, simulation and fabrication of submicronic microring resonators usingstepper lithography. Polymer materials and glass substrates are selected, due to their greatoptical properties in the visible range, their compatibility with the EWOD platform, and theirintegrability at a reasonable cost
