Abstract:
Background Two ongoing phase II clinical trials (RENEW and SYNERGY) have been developed to test the efficacy of anti-LINGO-1 antibodies in acute optic neuritis and relapsing forms of multiple sclerosis, respectively. Across a range of experimental models, LINGO-1 has been found to inhibit neuron and oligodendrocyte survival, axon regeneration, and (re)myelination. The therapeutic effects of anti-LINGO-1 antibodies on optic nerve axonal loss and regeneration have not yet been investigated. Objective In this series of studies we investigate if LINGO-1 antibodies can prevent acute inflammatory axonal loss, and promote axonal regeneration after injury in rodent optic nerves. Methods The effects of anti-LINGO-1 antibody on optic nerve axonal damage were assessed using rodent myelin oligodendrocyte glycoprotein experimental autoimmune encephalomyelitis (EAE), and its effects on axonal regeneration were assessed in optic nerve crush injury models. Results In the optic nerve, anti-LINGO-1 antibody therapy was associated with improved optic nerve parallel diffusivity measures on MRI in mice with EAE and reduced axonal loss in rat EAE. Both anti-LINGO-1 antibody therapy and the genetic deletion of LINGO-1 reduced nerve crush-induced axonal degeneration and enhanced axonal regeneration. Conclusion These data demonstrate that LINGO-1 blockade is associated with axonal protection and regeneration in the injured optic nerve.

Abstract:
Objective:To evaluate whether the anti-LINGO-1 antibody has immunomodulatory effects.Methods:Human peripheral blood mononuclear cells (hPBMCs), rat splenocytes, and rat CD4+ T cells were assessed to determine whether LINGO-1 was expressed and was inducible. Anti-LINGO-1 Li81 (0.1–30 μg/mL) effect on proliferation/cytokine production was assessed in purified rat CD4+ T cells and hPBMCs stimulated with antibodies to CD3 +/– CD28. In humans, the effect of 2 opicinumab (anti-LINGO-1/BIIB033; 30, 60, and 100 mg/kg) or placebo IV administrations was evaluated in RNA from blood and CSF samples taken before and after administration in phase 1 clinical trials; paired samples were assessed for differentially expressed genes by microarray. RNA from human CSF cell pellets was analyzed by quantitative real-time PCR for changes in transcripts representative of cell types, activation markers, and soluble proteins of the adaptive/innate immune systems. ELISA quantitated the levels of CXCL13 protein in human CSF supernatants.Results:LINGO-1 is not expressed in hPBMCs, rat splenocytes, or rat CD4+ T cells; LINGO-1 blockade with Li81 did not affect T-cell proliferation or cytokine production from purified rat CD4+ T cells or hPBMCs. LINGO-1 blockade with opicinumab resulted in neither significant changes in immune system gene expression in blood and CSF, nor changes in CXCL13 CSF protein levels (clinical studies).Conclusions:These data support the hypothesis that LINGO-1 blockade does not affect immune function.Classification of evidence:This study provides Class II evidence that in patients with MS, opicinumab does not have immunomodulatory effects detected by changes in immune gene transcript expression.

Transhipment problems are in the form of indirect distribution problems because the products sent must go through several stages before reaching the consumer. This study will compare the Vogel Approximation method in previous studies with the completion of linear programming with Lingo software tools. Transportation costs using the Vogel Approximation method in previous studies compared with transportation costs using the Lingo software tool have the same cost of IDR 508,289,000, but there are several different fertilizer distributions. Lingo software simplifies data processing so that the results obtained are more accurate than the Vogel Approximation method. Companies can use lingo software to minimize costs by working effectively and efficiently.

Abstract:
The purpose of doing research is to have the investigation on the students’ belief when Duo lingo application is applied in the class of vocabulary and to describe the justification of the advantages using the Duo lingo to empower the vocabulary enrichment. The students of English Study Program in State Islamic College as Pre Service –teachers have participated in this study. The data were collected by the questionnaire distribution and the interview methods. The data analysis uses the qualitative description. The result of the study exposes the facts that the respondents agree the Duo lingo can facilitate the vocabulary learning achievement. The medium labelled Duo lingo makes the students easy and interesting in learning the English word knowledge. The interview with the sample of informants suggests the testimony that the young learners can use the app overtime as far as they want. They are free to set or make up the schedule of lesson session. One more meaningful thing found in the application is that the English is spoken by the native speaker. By performing the English of the native speaker can be a model or master for the students. They get the chance to exercise repeating the word or the sentence spoken by the native speaker correctly.

Abstract:
AbstractLINGO-1 may be involved in the pathogenesis of cerebral ischemia. However, its biological function and underlying molecular mechanism in cerebral ischemia remain to be further defined. In our study, middle cerebral artery occlusion/reperfusion (MACO/R) mice model and HT22 cell oxygen–glucose deprivation/reperfusion (OGD/R) were established to simulate the pathological process of cerebral ischemia in vivo and in vitro and to detect the relevant mechanism. We found that LINGO-1 mRNA and protein were upregulated in mice and cell models. Down-regulation LINGO-1 improved the neurological symptoms and reduced pathological changes and the infarct size of the mice after MACO/R. In addition, LINGO-1 interference alleviated apoptosis and promoted cell proliferation in HT22 of OGD/R. Moreover, down-regulation of LINGO-1 proved to inhibit nuclear translocation of p-NF-κB and reduce the expression level of p-JAK2 and p-STAT3. In conclusion, our data suggest that shLINGO-1 attenuated ischemic injury by negatively regulating NF-KB and JAK2/STAT3 pathways, highlighting a novel therapeutic target for ischemic stroke.

Abstract:
Conversational agents are increasingly becoming digital partners in our everyday computational experiences. Although rich and fresh in content, they are oblivious to users' locality beyond geospatial weather and traffic conditions. We introduce Lingo, a hyper-local conversational agent embedded deeply into the urban infrastructure that provides rich, purposeful, detailed, and in some cases, playful information relevant to a neighbourhood. Drawing lessons from a mixed-method contextual study (online survey, n = 1992 and semi-structured interviews, n = 21), we identify requirements for such a hyper-local conversational agent and a sample set of questions serving urban neighbourhoods of Belgium. Our agent design is manifested into a two-part system. First, a multi-modal reasoning engine serves as a hyper-local information source using automated machine-learning models operating on camera, microphone, and environmental sensor data. Second, a smart conversational speaker and a smartphone application serve as hyper-local information access points. Finally, we introduce a covert communication mechanism over Wi-Fi management frames that bridges the two parts of our Lingo system and enables the privacy-preserving proxemic interactions. We describe the design, implementation, and technical assessment of Lingo together with usability (n = 20) and real-world deployment (n = 5) studies. We reflect on information quality, accessibility benefits, and interaction dynamics and demonstrate the efficacy of Lingo in offering hyper-local information at the finest granularity in urban neighbourhoods while reducing access time up to a factor of 25.

Tujuan penelitian ini adalah untuk mengetahui pelaksanaan dalam proses optimalisasi biaya pengiriman produk baru yang dilakukan oleh UD. Membiri. Metode yang digunakan dalam penelitian kuantitatif dengan menggunakan data primer UD. Membiri, kemudian dihitung menggunakan metode least cost serta penggunaan aplikasi Lingo untuk menemukan biaya dan jumlah pengiriman yang paling optimal. Hasil optimal dari biaya pengiriman baru menggunakan metode biaya paling sedikit, total biaya yang diperoleh adalah Rp. 12.385.000, saat menggunakan perangkat lunak Lingo, total biayanya adalah Rp. 16.030.000.

Aim and Objective: In the past decade, the drug design technologies have been improved enormously. The computer-aided drug design (CADD) has played an important role in analysis and prediction in drug development, which makes the procedure more economical and efficient. However, computation with big data, such as ZINC containing more than 60 million compounds data and GDB-13 with more than 930 million small molecules, is a noticeable issue of time-consuming problem. Therefore, we propose a novel heterogeneous high performance computing method, named as Hadoop-MCC, integrating Hadoop and GPU, to copy with big chemical structure data efficiently. Materials and Methods: Hadoop-MCC gains the high availability and fault tolerance from Hadoop, as Hadoop is used to scatter input data to GPU devices and gather the results from GPU devices. Hadoop framework adopts mapper/reducer computation model. In the proposed method, mappers response for fetching SMILES data segments and perform LINGO method on GPU, then reducers collect all comparison results produced by mappers. Due to the high availability of Hadoop, all of LINGO computational jobs on mappers can be completed, even if some of the mappers encounter problems. Results: A comparison of LINGO is performed on each the GPU device in parallel. According to the experimental results, the proposed method on multiple GPU devices can achieve better computational performance than the CUDA-MCC on a single GPU device. Conclusion: Hadoop-MCC is able to achieve scalability, high availability, and fault tolerance granted by Hadoop, and high performance as well by integrating computational power of both of Hadoop and GPU. It has been shown that using the heterogeneous architecture as Hadoop-MCC effectively can enhance better computational performance than on a single GPU device.

SBT yang merupakan perusahaan yang bergerak dalam bidang pendistribusian barang bangunan ini juga tidak terlepas dari kegiatan transportasi yang menerapkan sistem transhipment dengan kepemilikan 3 gudang sebagai tempat transit, dimana ketiga gudang ini memiliki lokasi yang berbeda-beda sehingga perlu dilakukan penelitian lebih lanjut apakah dengan adanya ketiga gudang tersebut biaya transportasi yang dikeluarkan sudah optimal atau belum. Penelitian dilakukan menggunakan data sekunder dari penelitian terdahulu dengan tujuan membandingkan hasil metode yang digunakan penelitian terdahulu dengan hasil penelitian ini, sehingga dapat memberikan masukan terbaik bagi perusahaan dalam aspek transportasi guna mencapai tujuan perusahaan. Pendekatan yang digunakan dalam menyelesaikan masalah transhipment ini yaitu menggunakan software Lingo yang tentunya dengan harapan memberikan hasil optimal. Hasil penelitian menunjukkan adanya penurunan total biaya transportasi yang awalnya Rp 3.275.000 menjadi Rp 1.413.000 dengan selisih biaya sebesar Rp 1.862.000 atau sebesar 56,85%.

Diploma thesis deals with the evaluation of the efficiency using DEA models focusing on the use of dedicated software tools. There are four chapters in the thesis where different approaches to efficiency are briefly presented. The method of data envelopment analysis (DEA) with its modifications is described in detail -- basic models, CCR, BCC and additive model. The main part of the work is presented in the second part of this thesis where I describe how to work with each program and summarize its advantages and disadvantages. The last chapter applies prior knowledge on real situation -- the evaluation of the efficiency of the regional police stations in the Czech Republic.

This thesis presents the results of the STAMPAS all-sky search for long transient gravitational waves in the 2005-2007 LIGO-Virgo data. Gravitational waves are perturbations of the space-time metric. The Virgo and LIGO experiments are designed to detect such waves. They are Michelson interferometers with 3 km and 4 km long arms, whose light output is altered during the passage of a gravitational wave.Until very recently, transient gravitational wave search pipelines were focused on short transients, lasting less than 1 second, and on binary coalescence signals. STAMPAS is one of the very first pipelines entirely dedicated to the search of long transient gravitational wave signals, lasting from 1s to O(100s).These signals originate, among other sources, from instabilities in protoneutron stars as a result of their violent birth. The standing accretion shock instability in core collapse supernovae or instabilities in accretion disks are also possible mechanisms for gravitational wave long transients. Eccentric black hole binary coalescences are also expected to emit powerful gravitational waves for several seconds before the final plunge.STAMPAS is based on the correlation of data from two interferometers. Time-frequency maps of the data are extracted, and significant pixels are clustered to form triggers. No assumption on the direction, the time or the form of the signals is made.The first STAMPAS search has been performed on the data from the two LIGO detectors, between 2005 and 2007. After a rigorous trigger selection, the analysis revealed that their rate is close to Gaussian noise expectation, which is a significant achievement. No gravitational wave candidate has been detected, and upper limits on the astrophysical rates of several models of accretion disk instability sources and eccentric black holes binary coalescences have been set. Cette thèse présente les résultats de la recherche de signaux d’ondes gravitationnelles produits par des cordes cosmiques dans les données des deux premières périodes d’observation des détecteurs de seconde génération Advanced LIGO et Advanced Virgo. Les cordes cosmiques sont des défauts topologiques unidimensionnels créés dans l’Univers primordial. Ces objets sont prédits par de nombreux modèles de physiques de très hautes énergies. La forme attendue du signal d’ondes gravitationnelles émis par des cordes cosmiques est prédite par la théorie. On utilise une technique de filtrage optimal pour rechercher la présence d’un signal dans les données des détecteurs Advanced LIGO et Advanced Virgo. Cette technique est optimale si le bruit du détecteur est gaussien et stationnaire, ce qui n’est pas le cas. Une partie essentielle du travail de thèse a consisté à comprendre et réduire le bruit de fond de l’analyse. Ce travail sur la qualité des données a permis d’augmenter la sensibilité de la recherche. De plus on montre que la recherche de signaux transitoires d‘ondes gravitationnelles produits par des cordes cosmiques est limitée par la présence d‘une famille particulière de bruits qui miment le signal prédit pour les cordes cosmiques. En l'absence de détection, nous avons placé des limites sur les paramètres qui caractérisent un réseau de cordes cosmiques pour différents modèles théoriques, afin de préciser les conditions d’existence de ces dernières
This thesis shows the results of the search for gravitational-wave signals produced by cosmic strings using the data from the first two observation runs of the second generation detectors Advanced LIGO and Advanced Virgo. Cosmic strings are one-dimensional topological defects created in the primordial Universe. These objects are predicted by many models of very high energy physics. The gravitationalwave signal emitted by cosmic strings is predicted by theory. A matched-filter analysis is used to search for the presence of a signal in Advanced LIGO and Advanced Virgo data. This technique is optimal if the detector noise is Gaussian and stationary, which is not the case. An essential part of the thesis work was to understand and reduce the background noise analysis. This work on data quality has increased the sensitivity of the search. Furthermore, we have shown that the search for transient signals of gravitational waves produced by cosmic strings is limited by the presence of a particular family of noises that mimic the predicted signal for cosmic strings. In the absence of detection, we have placed constraints on the parameters which characterize a cosmic string network for different theoretical models, in order to specify the conditions of their existence

The network of detectors comprising the Laser Interferometer Gravitational-wave Observatory (LIGO) are among a new generation of detectors that seek to make the first direct observation of gravitational waves. While providing strong support for the General Theory of Relativity, such observations will also permit new tests of physical theory in regions of strong space-time curvature and high matter-energy density. However, the observed signals are expected to occur near the limit of detector sensitivity. The problem of identifying such small signals is the primary focus of this work. This work presents a novel method for the identification of astrophysically unmodeled bursts of gravitational radiation in data from networks of interferometric detectors. The method is based on the Q transform, a multiresolution time-frequency transform that efficiently targets waveforms within a finite region of time, frequency, and Q space. The method is also based on a modification of linear prediction that greatly simplifies the resulting statistical analysis by whitening interferometric detector data prior to Q transform analysis. Together, these techniques form the basis of a complete analysis pipeline that is equivalent to a template-based matched filter search for minimum uncertainty waveforms in the whitened data stream. This method is then applied to search for gravitational-wave bursts with duration less than 1 second and frequency content between 64 and 1024 Hz in coincident data from two detectors during second LIGO science run. Although no gravitational-wave bursts are identified,
(cont.) upper bounds are reported for the rate of gravitational-wave bursts as a function of signal strength for isotropic and galactic populations of sources with both abstract and astrophysically motivated waveform. The results indicate a maximum of 0.09 events per day at the 90% confidence level for bursts with characteristic strain amplitude in excess of 10-20 to 10-19 strain Hz-l/2 depending on waveform. Introduction: Core collapse supernova are one of the most intriguing astrophysical phenomena. The dying stage of a supergiant star, they occur when the star collapses into a protoneutron star, causing a shock wave and a gamma ray burst. High energy neutrinos are released in this process and offer the possibility of detecting these elusive cataclysms. The number of neutrinos emitted is large but at best only a few will be detected. With a multi-messenger search, we can combine the neutrino signal with another clue to the presence of a supernova: gravitational waves. During the proto-neutron star stage, a fast-rotating star can produce gravitational waves via its asymmetric and rapidly shifting mass. By combining the signals from neutrinos and gravitational waves, we can attempt to detect supernova signals that are too faint to detect alone. Joint searches have already been attempted by several neutrino experiments with high-energy thresholds, including ANTARES and IceCube. This thesis explores the possibility of a joint search with a new set of neutrino data. KamLAND (Kamioka Liquid scintillator Anti-Neutrino Detector) is a large particle detector located in Kamioka, Japan. KamLAND is well-shielded, with an low (~ 1 MeV) energy threshold and has more than ten years of data to explore, making it a good candidate for a joint search. A recent search of KamLAND's data for clustered events indicative of supernova found no clear clusters. A new search is needed to identify single-neutrino events that may have originated in supernovae. A joint search will help KamLAND more carefully examine the possible sources of its single-neutrino events. The gravitational wave data comes from LIGO (Laser Interferometer Gravitational wave Observatory). Located in Hanford, WA and Livingston, LA, LIGO consists of two four-kilometer interferometer arms. Analysis of LIGO data from 2005 to 2010 did not produce any clear gravitational wave events, leading to a need for a more sensitive search. A multimessenger search in conjunction with KamLAND provides this opportunity. We can examine both KamLAND and LIGO's data in order to search for possible supernova signals observed by both experiments. Because a joint data-sharing agreement has not been reached between KamLAND and LIGO, this thesis looks at the potential of a joint analysis and the opportunity for such a study to produce promising results.

This project examines the concept of computer hubs as a tool to increase technological inclusion in rural areas in lowincome countries. The viability of computer hubs as a concept was evaluated by looking at three factors among people living in such areas: acceptance of computers, the financial viability of computer hubs, and actual ease of use of computers. The study was conducted in Linga Linga, a small village located in the province of Morrumbene in southern Mozambique. Linga Linga is characterized by low technological inclusion and low income, which is typical for villages in rural areas in lowincome countries. Interviews, questionnaires, workshops and focus groups were conducted to examine the viability of computer hubs. The study indicates that the acceptance of computers, measured through attitude towards computers as well as perceived usefulness and perceived ease of use of computers, was sufficient for implementation. The study also concludes that the actual ease is sufficiently high, and even in cases where it is not due to a lack of previous experience with computers, the time it takes to learn is low. In addition, it was found that computer hubs are financially viable from a business perspective. In conclusion, computer hubs can be considered a viable approach to increase technological inclusion in rural areas in low-income countries.
Projektet undersöker konceptet computer hubs och hur det kan användas som ett verktyg för att öka teknologisk inkludering på landsbygden i låginkomstländer. Konceptet utvärderades genom att undersöka tre faktorer: användaracceptansen för datorer, computer hubs finansiella hållbarhet och den faktiska användbarheten hos datorer. Studien genomfördes i Linga Linga, en by belägen i provinsen Morrumbene i södra Moçambique. Linga Linga kännetecknas av låg teknologisk inkludering och invånare med låga eller inga inkomster. Intervjuer, frågeformulär, seminarium och fokusgrupper genomfördes för att undersöka konceptens genomförbarhet. Studien indikerar att användaracceptansen hos datorer, mätt genom attityd gentemot datorer såväl som upplevd användbarhet och upplevd användarvänlighet för datorer, var tillräcklig för implementering. Studien visar även att den faktiska användbarheten hos datorer är tillräckligt hög för implementering samt att den snabbt kan förbättras trots lite eller ingen tidigare erfarenhet av datorer. Studien visar även att en implementering av computer hubs är ekonomiskt hållbar ur ett affärsperspektiv. Sammanfattningsvis kan implementeringen av computer hubs som koncept betraktas som ett  hållbart tillvägagångssätt för att öka teknologisk inkludering i landsbygden i låginkomstländer.

L'objectif de ce travail est la détection conjointe d'ondes gravitationnelles et de neutrinos cosmique de haute énergie à travers une approche multi-messagers. Les astronomies "neutrinos" et "ondes gravitationnelles" sont encore en phase de développement, mais elles sont appelées à jouer un rôle fondamental dans le futur. En effet, ces "messagers" peuvent parcourir de grandes distances grâce à leur faible interaction avec la matière (contrairement aux photons qui, à haute énergie, sont rapidement absorbés), sans être affectés par les champs magnétiques (contrairement aux rayons cosmiques chargés). Ils peuvent également s' échapper de milieux denses et fournir des informations sur les processus qui ont lieu au coeur des sources astrophysique (les photons s' échappent des couches périphériques des objets célestes). En un mot, ces astronomies sont susceptibles d'ouvrir une nouvelle fenêtre d'observation sur le cosmos. La collaboration ANTARES a construit en Méditerranée un télescope sous-marin de neutrino de haute énergie d'une surface de détection proche de 0.1 km². C'est le télescope le plus sensible pour la partie du ciel observée. Les interféromètres VIRGO et LIGO sont des détecteurs terrestres pour l'observation directe d'ondes gravitationnelles, instalés en Europe et aux états-Unis d'Amérique respectivement. Les instruments ANTARES, VIRGO et LIGO offrent une sensibilité inégalée dans la zone de recherche commune. Le premier chapitre de cette thèse introduit les motivations théoriques pour une recherche jointe d'ondes gravitationnelles et de neutrinos de haute énergies en développant les différents scénarios d'émission. Le deuxième et troisième chapitres sont consacrés à l'étude des techniques de détection avec les interféromètres VIRGO-LIGO et le télescope à neutrinos ANTARES. Les quatriéme et cinquième chapitres de ce travail présentent les résultats d'analyses de données combinées d'ANTARES, VIRGO et LIGO prises séparement pendant les années 2007 et 2009-2010.

We propose a “smart” language learning system for students to acquire domain-specific vocabulary while taking an online course. F-Lingo, a browser plugin, works on top of the FutureLearn MOOC platform to provide learners with opportunities to study the words, phrases, and concepts that are important to the course topic. F-Lingo comprises three components. The Material Gathering component crawls the web pages of the MOOC course the student has chosen, collecting the entire textual content (with some exceptions). The Vocabulary Extraction component identifies domain-specific words, phrases, and concepts, and hyperlinks in the MOOC page to draw the student’s attention to them. Clicking a link displays a dialog window in which lexico-grammatical features, and definitions, of the extracted items can be studied, including illustrations in example sentences retrieved from external resources such as Wikipedia and FLAX. The Progress Tracking component records the clicks that students make on hyperlinks and the time spent in the dialog windows. This allows us to build the student’s vocabulary learning profile under the assumption that the more time the student pays attention to an item, the more worthy the item to be included in a follow-up language activity. These statistical data provide evidence and reasoning in our current and ongoing work on automatically generating personalized language activities and vocabulary tests at the end of the MOOC course. F-Lingo has been made available in three Data Mining courses on the FutureLearn MOOC platform and has been used by 109 learners. This research is ongoing. Future work focuses on automatically generating personalized vocabulary tests and activities based on the student’s click statistics.

Data objects having low value like insurance or data-entry forms are shared between a client and rural business process outsourcing (RBPO) organisations for tasks like translation, proofreading, and data entry. These data objects are first decomposed into smaller parts and then assigned to RBPO users. Each user in a RBPO has access to only a few parts of a complete data object which he can leak to unauthorised users. But since the value of these parts is low, there is not enough incentive for the user to leak them. Such scenarios need good-enough security models that can provide reasonable security to an aggregate number of parts of low value data objects. In this chapter, the authors study the secure data assignment and leakage in RBPO by modeling it in the form of an optimisation problem. They discuss different scenarios of object decomposition and sharing, penalty assignment, and data leakage in the context of RBPO. They use LINGO toolbox to run their model and present insights.

Abstract Efficient modeling strategies are developed to study thermomechanical durability of high I/O Ball Grid Array (BGA) packages, in order to facilitate virtual qualification and accelerated testing of component designs. A viscoplastic stress analysis technique is developed where the critical solder joint(s) (joint(s) at which failure first occurs) are modeled in detail with a multi-domain Rayleigh-Ritz (MDRR) methodology (Ling and Dasgupta, 1996; Ling and Dasgupta 1997; Ling 1997; and Rassian and Lee, 1998) while the load-sharing offered by non-critical joints is modeled with a simplified compact model. This hybrid technique is used to study the behavior of solder interconnects in selected Ball Grid Array (BGA) package under thermal cycling environments. Parametric studies are conducted to determine the optimal scheme for allocating a critical number of solder joints to the MDRR model, and the remaining non-critical joints to the compact models. Damage calculations are made with the Energy Partitioning Solder Durability model (Dasgupta et al., 1992) and cycles-to-failure predictions are compared with both finite element model predictions as well as experimental failure data provided by CALCE EPSC sponsors. Parametric studies on change in solder joint durability with interconnect volume are also discussed in this paper.
