Abstract:
The aim of the present study was to investigate the spontaneous motility of spermatozoa and to optimize sperm collection, short-term sperm storage, and fertilization in zebrafish Danio rerio. The movement of spermatozoon in water was propagated along the flagellum at 16 s after sperm activation then damped from the end of the flagellum for 35 s and fully disappeared at 61 s after activation. For artificial fertilization, milt must be added to an immobilizing solution, which stops the movement of sperm and keeps the sperm motionless until fertilization. E400 and Kurokura as isotonic solutions were shown to be suitable extenders to store sperm for fertilization for 6 h. E400 stored sperm for 12 h at 0–2 °C. Sperm motility decreased only to 36% at 12 h post stripping for the E400 extender and to 19% for the Kurokura extender. To achieve an optimal level of fertilization and swim-up larvae rates, a test tube with a well-defined amount of 6,000,000 spermatozoa in E400 extender per 100 eggs and 100 µL of activation solution has proven to be more successful than using a Petri dish. The highest fertilization and swim-up larvae rates reached 80% and 40–60%, respectively, with milt stored for 1.5 h in the E400 extender at 0–2 °C.

Abstract:
The effect of stocking density on growth of zebra fish was examined. Total five different stocking densities (5, 15, 25, 35 and 45 individuals per 2 liters of water) were maintained in triplicate for a period of 60 days. One month old zebrafish were randomly stocked into 15 tanks and fish were fed with commercial diet. The mean weight gain, specific growth rate and length gain for treatment 1 (5 fish/2liter) and treatment 2 (15 fish/2 liter) were significantly (p < 0.05) higher than treatment 3 (25 fish/2 liter), treatment 4 (35 fish/2 liter) and treatment 5 (45 fish/2 liter). The gender weight gain, survival rate and condition factor did not show any significant (p > 0.05) difference among treatments. The result of this study suggests that the stocking density of zebrafish could be 15 fish per 2 litre of water in a laboratory system with aeration.Bangladesh J. Zool. 44(2): 209-218, 2016

Abstract:
A azadiractina consiste no óleo obtido a partir de sementes secas de Azadirachta indica A. Juss. (Família: Meliaceae). Trata-se de um biopesticida para controle de pragas na agricultura e, portanto, um agente "eco-amigável" para a agricultura orgânica. Contudo, os resíduos dos pesticidas, incluindo os biopesticidas, podem causar efeitos deletérios a organismos que não são o seu alvo. Assim, o presente trabalho pretende investigar o efeito genotóxico da azadiractina no meio aquoso em amostras de peixes conhecidos como Paulistinha ou Zebra-fish (Danio rerio - Hamilton 1822). As concentrações da azadiractina de 8, 16 e 32 vezes as taxas mais baixas de produtos comerciais formulados foram testadas durante 7 dias. Os efeitos de genotoxicidade foram avaliados através de Ensaio Cometa obtendo Frequência de Dano (FD) e Índice de Dano (ID) utilizando eritrócitos do D. rerio. A Análise de Sobrevivência avaliou a taxa de mortalidade em exemplares remanescentes do EC. Os resultados sugerem um aumento significativo de FD e ID em relação ao controle negativo (D. rerio não expostos), mesmo após 7 dias de exposição. Além disso, todos os espécimes expostos morreram dentro de uma semana após o final do EC. A utilização do biopesticida azadiractina deve ser realizada com precaução para não aumentar a contaminação ambiental, especialmente nos ecossistemas aquáticos.

This study was conducted to investigate the role of cortisol, a steroid hormone on the gene expression during embryogenesis of zebrafish (Danio rerio). Gene expression analyses were performed using microarray hybridization. A total 80 genes were found to be up-regulated and 36 genes were down-regulated at 24 hrs post fertilization (hpf) when the fertilized eggs were treated with cortisol hormone. Abstract The aim of the present study was to investigate the cellular characteristics of the immune tissues of Zebrafish, Danio rerio (Hamilton, 1822). The fish were fixed in Bouin՚s solution for 24 hours then dehydrated, cleared, paraffinized, embedded and finally sectioned, stained and observed through optical microscopy. Results showed that immune system tissues of Zebrafish include the apical part of the kidney, thymus, and spleen. The apical part of the kidney was composed of hematopoietic tissue containing blood and immune cells. The spleen was a single organ located at the abdominal cavity containing melanomacrophages. The thymus was observed as a paired organ at the posterior part of the branchial cavity. Results showed that the immune system of the Zebrafish was dispersed in several organs of the body and that this species could be used as a laboratory model organism in immune system studies.

Abstract:
Stable isotopes of carbon, nitrogen, and sulfur in organisms can trace nutrient sources and determine trophic interactions. However, time is required for an organism to reflect the isotopic composition of its diet. The rate at which an organism incorporates the isotopic signature of its food has a growth (k) component and a metabolic tissue replacement (m) component. This study shows that metabolic tissue replacement accounts for between 68% and 80% of the observed changes in isotopic signature in zebra danio ( Danio rerio (Hamilton, 1822)) muscle following a dietary shift, with the remainder owing to growth of new tissue. The half-lives of carbon, nitrogen, and sulfur were ~53, ~147 and ~58 days, respectively. This indicates that a significant period of isotopic disequilibria exists before zebra danio resemble dietary isotopic composition. These results show that catabolic replacement of tissue, as well as growth, must be taken into account when predicting the rate of isotope incorporation in warm-water fish. When using isotopic ratios to decipher food webs and trophic interactions, researchers should be aware that the often held assumption of isotopic equilibrium between diet and consumer may be violated.

Effect of rearing density (5, 15, 25, 35 and 45 fish/l2) on reproductive performances such as spawning success, fertilization rate, hatching rate and embryogenesis in terms of per cent survival rate of embryos at cleavage, gastrula, segmentation, pharyngula and hatching were evaluated. Survival, hatching, fertilization rate and per cent survivability of embryos did not differ significantly between the treatments 1, 2 and 3, but showed significant difference (p < 0.05) between 4 and 5. Treatment three (25 fish/l2) had five-folds higher number of successful progeny than treatment one. Results suggest that 25 fish/l2 can be used as optimum rearing density. Dhaka Univ. J. Biol. Abstract:
The present study was focused on the concentration-dependent changes in oral acute toxicity of leaf extracts of E. fluctuans in zebrafish. The study was also aimed at the details of histopathological changes in the gill, liver, brain, and intestine of zebrafish exposed to the leaf extracts of the plant E. fluctuans. Enydra fluctuans Lour is an edible semiaquatic herbaceous plant used widely for the alleviation of the different diseases. Since there were no toxicity studies conducted on this plant, the present study was an attempt to look into the elements of toxicity of the plants. Two types of experiments are conducted in the present study. First, the acute oral toxicity study was conducted as per the OECD guidelines 203. Second, histopathological changes were observed in the fishes exposed to the lethal concentrations of plant extract. The oral acute toxicity studies conducted on Zebrafish have revealed that the leave extracts of E. fluctuans were toxic to the tested fish at the concentration of 200 mg/kg body weight. The histopathological studies conducted on the intestine of treated fishes showed that treatment has induced rupturing of the villi structure and fusion of villi the membrane and detachment of the villi structure from the basal membrane of the intestine. The histology of the liver also showed severe vacuolization in the cells while it is not affected in control. The studies on gills showed the detachment of the basal epithelial membrane in the gills compared to control which might have led to death of the fish. The histopathological observations of brain tissues treated with test samples also revealed the marked impingement in the brain parenchyma while the control is normal without impingement of the brain.

In order to verify the influence of chronic and acute ambient oxygen levels from egg to adult stage of the zebrafish, in vivo oxygen consumption (MO2), critical tensions of oxygen (Pcrit), heart rate (fH) and total body lactate concentration (Lc) were determined for Danio rerio (Hamilton, 1822) raised at 28 °C under normoxic (7.5 mgO2.L-1 or 80 mm.Hg-1) and hypoxic conditions (4.3 mgO2.L-1) and exposed to acute hypoxia during different developmental stages. Our findings confirmed that very early stages do not respond effectively to ambient acute hypoxia. However, after the stage corresponding to the age of 30 days, D. rerio was able to respond to acute hypoxia through effective physiological mechanisms involving aerobic and anaerobic metabolism. Such responses were more efficient for the fishes reared under hypoxia which showed that D. rerio survival capability increased during acclimation to mild hypoxia. Measurements of body mass and length showed that moderate hypoxia did not affect growth significantly until the fish reached the stage of 60 days. Moreover, a growth delay was verified for the hypoxic-reared animals. Also, the D. rerio eggs-to-larvae survival varied from 87.7 to 62.4% in animals reared under normoxia and mild hypoxia, respectively. However, the surviving animals raised under moderated hypoxia showed a better aptitude to regulate aerobic and anaerobic capacities when exposed to acute hypoxia.

Abstract:
The current investigation intended to evaluate the effect of Benzophenone-3 (BP-3) at the environmentally relevant concentration (44 μg/L) in the gills of Danio rerio through evaluating oxidative stress markers and histopathological analysis. The adult Zebra fish was exposed to BP-3 at environmentally relevant concentration for 45 days. During the experimental period of 15, 30 and 45 days, lipid peroxidation markers like thiobarbituric acid reactive substances (TBARS) and Hydrogen peroxide (H2O2), antioxidant enzymes such as superoxide dismutase (SOD), catalase (CAT), Glutathione peroxidase (GPx) and non-enzymatic glutathione (GSH) in the gill and histology of gill were analyzed. The activity of TBARS and H2O2 was found to be significantly higher meanwhile the activities of antioxidant enzymes viz., SOD, CAT, GPx and Glutathione (GSH) level were found to be significantly reduced in the gill of BP-3 treated fish for 30 and 45 days. Additionally, the morphology of gill also showed several abnormal changes in their morphology when compared to control. BP-3 exposure for 15 days elicited only mild alterations in the biochemical and histopathological variables when compared to 30 and 45 days exposure. Further, the values were also non-significant when compared to the control fish. These results demonstrated that the treatment of BP-3 at environmentally relevant concentration could prominently alter the respiratory physiology and metabolism of the gills of Danio rerio.

Kualitas telur merupakan faktor primer dalam keberhasilan pembenihan ikan. Telur yang berkualitas memiliki tingkat pembuahan dan penetasan yang tinggi. Penelitian ini bertujuan menganalisis faktor yang dapat mempengaruhi proses perkembangan embrio dan penetasan telur ikan zebra (Danio rerio). Penelitian berjenis campuran antara deskriptif eksperimental dengan kuantitatif menjelaskan semua tahapan perkembangbiakan dan rasio penetasan telur serta perkembangan embrio. Hasil penelitian mendapatkan bahwa pemberian pakan alami mampu mempercepat pertumbuhan, menangkal bibit penyakit, dan mempercepat pematangan gonad. Waktu pelepasan induk yang baik yaitu pada pagi dan sore karena suhu perairan cenderung rendah. Hasil pengamatan telur di mikroskop didapati suatu serangan infeksi jamur Saprolegnia sp., jamur ini cenderung tumbuh menyebar di permukaan telur dan berbentuk hifa. Larva menetas mulai 48 jam hingga 72 jam pasca fertilisasi. Larva 2-3 dpf tetap dipelihara di media penetasan hingga menjadi benih. Tingkat keselamatan embrio berkisar 50%, sedangkan untuk larva hingga usia 21 dpf berkisar 100%. Hal ini diakibatkan jamur Saprolegnia sp., sehingga nilai indeks rasio penetasan kurang optimal dikarenakan terhambatnya pertumbuhan embrio dan berhujung kematian. Namun nilai indeks sintasan (survival rate) tinggi dikarenakan manajemen pemeliharaan larva yang optimal dengan pemberian pakan yang tepat dari susu bubuk dancow dan pengolahan air yang baik.

Zebrafish (Danio rerio: Hamilton, 1822) is a well-known model organism of biological research. Generally it is found in the fresh water ponds. However, the culture, breeding and embryonic development of wild D. rerio of Bangladesh is scarcely represented in the literature. Therefore, a laboratory experiment was conducted to study these aspects of local zebrafish. Water quality parameters for adult fish husbandry system were recorded. These were temperature (°C) 29.18 ± 1.53, pH 7.55 ± 0.144, TDS (ppm) 225 ± 11.136, (ORP) 48.93 ± 26.04, DO (ppm) 1.24 ± 0.591, conductivity (mS/cm) 0.450 ± 0.022, are resistivity (Mῼcm) 0.0026 ± 0.00049 for adult fish tanks. The parameters obtained from larvae rearing ranks were temperature (°C) 29.56 ± 0.777, pH 7.73 ± 0.421, TDS (ppm) 198.33 ± 39.627, (ORP) 51.37 ± 17.387, DO (ppm) 2.02 ± 0.694, conductivity (mS/cm) 0.397 ± 0.079, resistivity (MΩcm) 0.0026 ± 0.00044. After one month of rearing, adult male and female fishes were selected for natural breeding and spawning and obtained 87.5 ± 7.72% spawning success, 96.83±0.81% fertilization rate and 94.89 ± 0.27% hatching rate. The embryonic development for zygote, cleavage, blastula, gastrula, segmentation and pharyngula periods were observed and obtained proper development for each state at 28.5ºC. The survival rate of hatchlings was 10 - 20% up to 20 days post fertilization. The higher spawning success, fertilization rate, hatching rate and survival rate indicate the possibility and potentiality of culturing zebrafish in laboratory conditions and could be established as a model animal for Bangladeshi researchers. Hypertension is one of the diseases with the highest prevalence in Indonesia and is the main cause of death in patients. Pregnant women have a risk of losing their baby due to hypertension or death. ACEI and ARB-type antihypertensive drugs are dangerous for pregnant women because they can cause fetal death. In ancient times, Indonesians have used plants as herbal medicines for generations. One of the plants that has medicinal properties is mangosteen skin (Garcinia mangostana L.) as an antihypertensive drug through its diuretic effects. However, the drugs consumed can have toxic effects if used in high concentrations. A teratogenic test is a toxic test that is specifically carried out to determine the presence of organ abnormalities in the fetus due to exposure to xenobiotic compounds. The zebrafish (Danio rerio) embryo was chosen as the research object because the chorion in the embryonic phase is transparent, so it is easy to observe. This study aimed to determine the effect of exposure to mangosteen rind decoction on zebrafish embryos' hatchability and spinal curvature. Observations were made using a Leica microscope with ANOVA data analysis. The observations showed that high treatment concentrations decreased heart rate, while high concentrations didn’t affect egg hatching rate, spine curvature, or pigment density.

Abstract Pesticides are designed to kill a very wide range of undesirable organisms, but they simultaneously adversely affect non-target organisms in ecosystems. Dimethoate is an organophosphorous insecticide used widely in agriculture that has an anticholinesterase mode of action. The goal of the study was to assess the effects of dimethoate on catalase, reduced glutathione, and lipid peroxidation in the gills and livers of zebrafish, Danio rerio (Hamilton), exposed to different concentrations of the pesticide for different periods of time. Insecticidal stress caused a reduction in catalase activity and reduced glutathione levels in zebrafish gills and livers. There was also increased lipid peroxidation in both organs. Alterations in all parameters were concentration and time dependent. The probable causes are discussed.

Fucoidan memiliki aktivitas hepatoprotektif yang dapat menstimulasi regenerasi hepatosit. Studi ini bertujuan untuk mengamati struktur histologis hepar ikan zebra (Danio rerio, Hamilto 1822) dewasa yang telah diberi perlakuan parasetamol dan fucoidan. Desain penelitian yang digunakan adalah Rancangan Acak Lengkap (RAL) dengan ikan zebra dibagi menjadi empat kelompok perlakuan yaitu kontrol (K) selama 10 hari, parasetamol 5 mM (P) selama 3 hari dilanjutkan air RO selama 7 hari, parasetamol 5 mM selama 3 hari dilanjutkan fucoidan 100 µg/mL (P + F 100) selama 7 hari dan parasetamol 5 mM selama 3 hari dilanjutkan fucoidan 500 µg/mL (P + F 500) dengan ulangan setiap kelompok 6 ekor ikan. Studi berlangsung selama 10 hari, parameter yang diamati adalah histopatologis hepar, berat badan ikan, keaktifan berenang, dan nafsu makan. Pada semua kelompok perlakuan, hasil menunjukkan bahwa struktur histologis hepar ikan zebra mengalami kerusakan jaringan berupa hemoragi, dan kerusakan sel berupa vakuolisasi, piknosis dan nekrosis. Terdapat penurunan aktivitas berenang dan nafsu makan setelah perlakuan parasetamol. Kelompok yang diberi fucoidan mengalami pemulihan aktivitas renang dan nafsu makan. Terdapat perbedaan nyata (P <0,05) kerusakan hepar antara perlakuan kontrol, parasetamol dan pemberian fucoidan. Kelompok P + F 100 dan P + F 500 mengalami pemulihan hepatosit setelah paparan parasetamol. Kelompok P + F 500 memiliki perbaikan yang lebih baik dibandingkan kelompok P + F 100. The research design used was a completely randomized design (CRD) with zebrafish divided into four treatment groups; control (K) for 10 days, paracetamol 5 mM (P) for 3 days followed by RO water for 7 days, Paracetamol 5 mM for 3 days followed by fucoidan 100 g/mL (P + F 100) for 7 days and Paracetamol 5 mM for 3 days followed by fucoidan 500 g/mL (P + F 500) with replicates for each group of 6 fish. The study lasted for 10 days, data on liver histopathology was evaluated, fish body weight, swimming activity, and appetite was also evaluated. In all treatment groups, the results showed that the histological structure of the zebrafish liver experienced tissue damage in the form of hemorrhage, and cell damage in the form of vacuolization, pyknosis and necrosis. There was a decrease in swimming activity and appetite after paracetamol treatment, the group given fucoidan experienced a recovery in swimming activity and appetite. There was a significant difference (P <0.05) in liver damage between the control, paracetamol and the fucoidan treated groups. The P + F 100 and P + F 500 exhibited hepatocyte recovery after exposure to paracetamol. The P + F 500 group had better improvement than the P + F100 group. The result of this research showed that fucoidan can be used as a protective liver agent after paracetamol high dosage exposure.

The zebrafish Danio rerio is a popular model species for genetic and early development studies. It is relatively easy to maintain in laboratory, has a high fecundity rate and produces transparent embryos. Here, we characterise muscle development in early life stage zebrafish using paraffin sections of embryos and larvae treated with haematoxylin and eosin staining, and whole mount fluorescent immunohistochemistry. We found variations in the distribution of muscle mass throughout the body, with the greatest proportion of muscle mass found in the tail. Our data also showed for the first time the reaction of antibodies (protein expression) in muscle at early life stages of development. Whole mount fluorescent immunohistochemistry staining with three markers (PAX7, MF20 and F59) suggests that muscle development starts even earlier than previously suggested at the embryonic stage (1 dpf).

With immunofluorescence microscopy, the positions of centromeres and MLH1 (MutL homolog) foci representing the sites of presumptive chiasmata are shown for zebrafish (Danio rerio Hamilton 1822) synaptonemal complexes (SCs) in spermatocyte nuclei at meiotic prophase. Most SCs have a single focus and a few (7 of 140) have 2 chiasmata. MLH1 foci tend to be in the distal regions of SCs, with progressively fewer occurring towards the middle of the SCs. This non-random distribution suggests chiasma interference. Synaptic initiation, as well as replication protein A (RPA) foci at the chromosome ends, correlates with the distal localization of MLH1 foci. These observations may provide the physical basis for the reported limited genetic recombination in the centromeric region of androgenetic offspring of a male.Key words: zebrafish, recombination, chiasmata, interference, MLH1, RPA.

Abstract Microinjection is a micromanipulation technique that has been used in various fields and in a variety of species, including teleost fish, for over a hundred years. Microinjection is mainly used in genetic, molecular, and toxicological research. The paper describes an experimental study of a microinjection technique for the administration of Infectious Pancreatic Necrosis Virus (IPNV) in fertilized eggs of brook trout, Salvelinus fontinalis (Mitchill), and a rainbow trout Oncorhynchus mykiss (Walbaum) hybrid, Salvelinus fontinalis ♂ × Oncorhynchus mykiss ♀. The oocytes and embryos of these salmonids have complex structures that require modifying the method commonly used in zebrafish, Danio rerio (Hamilton), research. A modular apparatus and technique for injecting brook trout and rainbow brook trout fish embryos with IPNV are described. The modified microinjection method can be used successfully to administer to salmonid eggs infectious substances that simulate the vertical transmission of pathogens.

Application of fluorescent protein transgenesis has commercial use for the ornamental aquarium trade by creating new colour phenotypes in various species. To determine the potential for transgenic ornamental aquarium fish to overwinter in Canada, the minimum temperature tolerance of albino rainbow shark (Epalzeorhynchos frenatum (Fowler, 1934)) was estimated to complement a previous study examining cold tolerance of zebrafish (Danio rerio (Hamilton, 1822)), black tetra (Gymnocorymbus ternetzi (Boulenger, 1895)), and tiger barb (Puntius tetrazona (Bleeker, 1855)). Rainbow shark had higher low temperature tolerance limits (LD50 = 10.7 ± 0.1 °C) than surveyed winter water temperatures in Canada. There was a significant negative correlation between condition factor and temperature at loss of equilibrium, suggesting fish with lower social status may be more susceptible to cold temperature than dominant fish. These results indicate transgenic E. frenatum are not expected to persist over winter in Canadian waters.
