Abstract:
The aim of the present study was to investigate the spontaneous motility of spermatozoa and to optimize sperm collection, short-term sperm storage, and fertilization in zebrafish Danio rerio. The movement of spermatozoon in water was propagated along the flagellum at 16 s after sperm activation then damped from the end of the flagellum for 35 s and fully disappeared at 61 s after activation. For artificial fertilization, milt must be added to an immobilizing solution, which stops the movement of sperm and keeps the sperm motionless until fertilization. E400 and Kurokura as isotonic solutions were shown to be suitable extenders to store sperm for fertilization for 6 h. E400 stored sperm for 12 h at 0–2 °C. Sperm motility decreased only to 36% at 12 h post stripping for the E400 extender and to 19% for the Kurokura extender. To achieve an optimal level of fertilization and swim-up larvae rates, a test tube with a well-defined amount of 6,000,000 spermatozoa in E400 extender per 100 eggs and 100 µL of activation solution has proven to be more successful than using a Petri dish. The highest fertilization and swim-up larvae rates reached 80% and 40–60%, respectively, with milt stored for 1.5 h in the E400 extender at 0–2 °C.

Abstract:
The effect of stocking density on growth of zebra fish was examined. Total five different stocking densities (5, 15, 25, 35 and 45 individuals per 2 liters of water) were maintained in triplicate for a period of 60 days. One month old zebrafish were randomly stocked into 15 tanks and fish were fed with commercial diet. The mean weight gain, specific growth rate and length gain for treatment 1 (5 fish/2liter) and treatment 2 (15 fish/2 liter) were significantly (p < 0.05) higher than treatment 3 (25 fish/2 liter), treatment 4 (35 fish/2 liter) and treatment 5 (45 fish/2 liter). The gender weight gain, survival rate and condition factor did not show any significant (p > 0.05) difference among treatments. The result of this study suggests that the stocking density of zebrafish could be 15 fish per 2 litre of water in a laboratory system with aeration.Bangladesh J. Zool. 44(2): 209-218, 2016

Abstract:
A azadiractina consiste no óleo obtido a partir de sementes secas de Azadirachta indica A. Juss. (Família: Meliaceae). Trata-se de um biopesticida para controle de pragas na agricultura e, portanto, um agente "eco-amigável" para a agricultura orgânica. Contudo, os resíduos dos pesticidas, incluindo os biopesticidas, podem causar efeitos deletérios a organismos que não são o seu alvo. Assim, o presente trabalho pretende investigar o efeito genotóxico da azadiractina no meio aquoso em amostras de peixes conhecidos como Paulistinha ou Zebra-fish (Danio rerio - Hamilton 1822). As concentrações da azadiractina de 8, 16 e 32 vezes as taxas mais baixas de produtos comerciais formulados foram testadas durante 7 dias. Os efeitos de genotoxicidade foram avaliados através de Ensaio Cometa obtendo Frequência de Dano (FD) e Índice de Dano (ID) utilizando eritrócitos do D. rerio. A Análise de Sobrevivência avaliou a taxa de mortalidade em exemplares remanescentes do EC. Os resultados sugerem um aumento significativo de FD e ID em relação ao controle negativo (D. rerio não expostos), mesmo após 7 dias de exposição. Além disso, todos os espécimes expostos morreram dentro de uma semana após o final do EC. A utilização do biopesticida azadiractina deve ser realizada com precaução para não aumentar a contaminação ambiental, especialmente nos ecossistemas aquáticos.
