Abstract:
2053 Background: Poor T cell homing hinders the development of effective cell therapy for central nervous system (CNS) malignancies. Lessons learnt from inflammatory brain diseases can give insight into how to overcome the blood brain barrier (BBB) blockade created by cancer. Activated Leukocyte Cell Adhesion Molecule (ALCAM; CD166) is a pathological adhesion molecule upregulated in the endothelium of a number of inflammatory/infiltrative CNS diseases, such as multiple sclerosis. Antibodies blocking ALCAM decrease leukocyte access to the brain and are currently being tested in a clinical trial for MS. Methods: We studied the difference in the dynamic signature of adhesion molecules in the “anergic” brain tumor endothelium and that of infiltrative brain conditions. Consequently, we mapped the ALCAM minimal binding region to domain 3 (D3) of CD6 and created an artificial molecule with the intent of creating a novel cellular platform to reverse endothelial anergy, through ALCAM specific binding. Results: GBM endothelium fails to launch the second wave of adhesion molecules necessary for firm T leukocyte capture and effective BBB transmigration. Engineered D3 on the T cell crosslinked to ALCAM on endothelial cells in proximity ligation assays (PLA; < 40nm) during TEM. Under shear stress, D3 T cells showed a global improved ALCAM specific trafficking kinetics: higher capture on ALCAM+ endothelium, rolling with slower velocity, and better TEM. In an ex vivo model of BBB, D3 T cells exhibited higher transmigratory ability. We discovered that signaling through the D3 endodomain phosphorylated pZAp70 recruiting Talin that enables LFA-1 (ICAM-ligand) open confirmation, mediating effective TEM. Lastly, in an orthotopic model of GBM, D3 T cells homed more and accumulated at the tumor site compared to NT controls. And, testing Her2 CAR T cells on D3 platform showed an advantageous homing after IV administration which was reflected in tumor control and better survival. Conclusions: We created a cellular platform that enables targeted brain delivery of T cells. This platform serves as a gateway to the effective cellular therapeutics for brain malignancies but potentially as a delivery system for complex biologics for other pathological conditions.

Abstract:
Neurodegenerative diseases, in particular Alzheimer disease (AD), afflict an increasing proportion of the older population with aging. Decreased exposure to sunlight and decreased consumption of fish, fruits, and vegetables, are two epidemiological factors that appear to be related to the pandemic of AD. In addition to replacing simple with complex carbohydrates and avoiding saturated fat, two nutritional components, vitamin D (acting through the endogenous hormonal form 1α,25 dihydroxyvitamin D, 1,25D) and docosahexaenoic acid (DHA) (acting through the docosanoid lipidic modulators resolvins and neuroprotectins) have high potential for prevention of Alzheimer disease. 1,25D is a neuroprotective, it acts both directly and indirectly in neurons by improving the clearance of amyloid-beta by macrophages/microglia. Resolvins and neuroprotectins inhibit amyloidogenic processing of amyloid-precursor protein, inflammatory cytokines, and apoptosis. It is likely that the increased consumption of vitamin D and fish oil could prevent neurodegeneration in some subjects by maintaining adequate endocrine, paracrine, and/or autocrine production of 1,25D and the DHA-derived lipidic modulators. Before firm recommendations of the dosage can be proposed, however, the in vivo effects of vitamin D3 and DHA supplementation should be investigated by prospective studies.Key words: Alzheimer disease, vitamin D3, DHA, fish oil

Abstract:
An adult male cat presented to the Ontario Veterinary College after having a seizure at the humane society. The cat was diagnosed with primary hypoparathyroidism and was treated with calcium and vitamin D3 supplementation. Calcium supplementation included subcutaneous (SC) administration of diluted calcium gluconate according to protocols described in recent literature. Seven days after SC calcium administration, firm SC masses developed in the intrascapular area where the injections were given and in the dependent axillary area. These areas became raised, formed crusts, necrosed, and left scars. Although not histopathologically confirmed, the author postulates that this was a direct reaction to SC calcium gluconate administration.

The article presents data about the content of active metabolite of vitamin D3 – 25OHD3, calcium (total, bounded with protein and ultrafiltrated), inorganic phosphorus, magnesium and alkaline phosphatase activity and its isoenzymes in the blood of cows and their calves in the early postnatal period.Studies were conducted in the cows of the Ukrainian Black-and-White dairy breed and calves obtained from them. The experiment was performed during the winter housing period in pilot farm «Pasichna» of Institute of forage and agricultural Podillya NAAS of Ukraine, located in the natural geographical areas of Podillya. The blood for tests was collected from the jugular vein before morning feeding in the 5th−7th day since calving. The blood from the calves was collected in the following dates: at 5th−7th days old and at 28–30th days old. Vitamin D provision rate of animal organism was evaluated for the content of 25OH D3 concentration in blood. The level of 25-hydroxycholecalciferol is considered as a total reflection of the endogenous formation of cholecalciferol in the skin and its receipts from feed or vitaminpreparations. The concentration of 25ОНD3 in the blood of the examined animals was detected by means of the enzymelinked immunoassay using the test system developed by the Immunodiagnostik. The method is based on the competitive binding of 25OHD3 serum and 25OH D3-biotin with vitamin D3-binding protein (VDBP), that immobilized on 96-well immunological plates. The content of calcium (total, bounded with protein and ultrafiltrated), inorganic phosphorus, magnesium and alkaline phosphatase (ALP) activity were detected usingthe biological test kits produced by the Pliva Lachema firm (the Czech Republic). The performed research reported that the content of 25-hydroxycholecalciferol in serum of calves at 5–7-days age old was in the range of 29 to 34 nmol/l and slightly increased for the 28–30-days. It was established the dynamics of changes in the content of total calcium and its fractions, inorganic phosphorus, magnesium and activity of alkaline phosphatase and its isoenzymes in calves blood from 5–7- to 28–30-day-olds. The concentration of 25-hydroxyvitamin D3 in the blood of calves in the first days after birth was lower, and the concentration of calcium and inorganic phosphorus – higher compared with their values in the blood of mothers in the postpartum period. It was found that the activity of alkaline phosphatase in the calf serum was higher, compared with its value in the serum of mothers. The increase in activity of alkaline phosphatase in the calf serum is expressed to a greater extent due to its bone enzyme.

Abstract:
The study aimed to determine the efficiency of a wastewater treatment plant and the quality of effluent discharged by the plant into the receiving water body (Okochiri River) in Eleme, Rivers State. The study lasted for four months. Wastewater samples were collected at various stages of the treatment unit and tested for major water quality parameter such as Biological Oxygen Demand (BOD), oil/grease, phenol, Dissolved Oxygen (DO), Total Suspended Solids (TSS), and sulphide. In general, the composite wastewater treatment system (comprising dissolve air floatation, Rotary Bio-Disk, and sedimentation unit) was effective in pollutant removal, except for sulphide, which had a negative efficiency (-238%). In line with Pearson Correlation, there are strong positive relationships between several parameters, such as pH and Sulphide (0.93), Phenol and Oil/grease (1.00), and Phosphate and Sulphide (0.91). The Principal Component Analysis yielded four principal components (PC1-4): PC1 parameters with the highest loadings on the first factor (D1) were sulphates and phenols, whereas Chemical Oxygen Demand and Biological Oxygen Demand had the highest loadings on the second factor (D2). Dissolved oxygen was most strongly associated with the third factor (D3), and pH was the parameter with the highest loading on the fourth factor (D4). It is recommended that the wastewater treatment system's overall efficiency be improved for the system to consistently meet regulatory limits for pollutant control. Recommendations for improving system efficiency include (1) ensuring regular maintenance of individual units in the treatment plant; and (2) maintaining optimal operating conditions for the rotary bio-disk unit.

Vitamin D supplementation is proposed as a fall prevention strategy, as it may improve neuromuscular function. We examined whether three years of vitamin D supplementation (400, 4000 or 10,000 IU daily) affects postural sway in older adults. Three hundred and seventy-three non-osteoporotic, vitamin D-sufficient, community-dwelling healthy adults, aged 55–70 years, were randomized to 400 (n = 124), 4000 (n = 125) or 10,000 (n = 124) IU daily vitamin D3 for three years. Sway index was assessed at baseline, 12-, 24- and 36-months using the Biosway machine. We tested participants under four conditions: eyes open or eyes closed with firm (EOFI, ECFI) or foam (EOFO, ECFO) surfaces. Secondary assessments examined sway in the anterior-posterior (AP) and medio-lateral (ML) directions. Linear mixed effects models compared sway between supplementation groups across time. Postural sway under EOFO and ECFO conditions significantly improved in all supplementation groups over time. Postural sway did not differ between supplementation groups at any time under any testing conditions in normal, AP or ML directions (p > 0.05 for all). Our findings suggest that high dose (4000 or 10,000 IU) vitamin D supplementation neither benefit nor impair balance compared with 400 IU daily in non-osteoporotic, vitamin D-sufficient, healthy older adults.

Abstract von Willebrand factor (VWF) is a large multimeric protein whose monomers are of the domain structure D’-D3-A1-A2-A3-D4-B1-B2-B3C1-C2. The three tandem A domains mediate several important functions of the protein including supporting platelet adhesion, through an interaction of A1 with the platelet GP Ib-IX-V complex, and binding collagen, through A3 and A1. Further, the A2 domain contains the cleavage site for the plasma metalloprotease ADAMTS-13, which processes the newly synthesized and extremely adhesive ultra-large forms of VWF (ULVWF) to the forms normally found in plasma. Our goal was to compare the functions of the individual A domains as isolated domains to their function in the context of the A1-A2-A3 fragment. All recombinant domains were produced in bacteria. The A1-A2-A3 fragment contained an N-terminal thioredoxin-tag and a C-terminal 6Xhis tag and was purified in 2 steps using nickel and heparin columns. The recombinant protein was correctly folded, as indicated by the fact that it was recognized by a number of conformation-specific antibodies. Using ELISA, we examined the interaction of the fragment with both glycocalicin (the soluble extracellular domain of GP Iba) and collagen. At equivalent concentrations, A1-A2-A3 bound collagen better than did the isolated A1 and A3 domains, but the binding to glycocalicin was approximately 7.5 times lower than the A1 domain. We then tested the ability of the fragment to support platelet adhesion under flow, by coating the fragment onto a coverslip that made up the bottom of a parallel-plate flow chamber. In contrast to the findings with immobilized glycocalicin, immobilized A1-A2-A3 supported the attachment, rolling, and firm adhesion of the platelets to the surface, whereas A1 did not support firm adhesion. Finally, we tested cleavage of the fragment by plasma ADAMTS-13, using both ELISA and western blotting to detect cleavage. The rate of cleavage of the A1-A2-A3 fragment was much slower than that of the recombinant A2 domain, needing a longer incubation time to achieve a similar percentage of cleavage in a 15 fold higher concentration of plasma. These results show clearly that within the triplicate A domains of VWF, adjacent A domains influence each other’s functions, possibly by binding each other or by conformational modulation. The fact that the fragment is able to support firm platelet adhesion in isolation suggests that it may contain a second, previously unidentified binding site for platelet receptors.

We propose a roll-to-roll process for vitamin D3 patch production. A solution of 7-dehydrocholesterol is applied to a plastic film by roll-to-roll slot-die coating and dried by a far-infrared lamp. Upon exposure to ultraviolet B irradiation, these films are converted to previtamin D3 films. After heat-treating the previtamin D3 film, high-performance liquid chromatography measurements are performed using commercial vitamin D3 as a standard sample. The results confirm that vitamin D3 can be produced by large-area coating and post-treatment processes. Specifically, 3.16 ± 0.746 mg of vitamin D3 is obtained through ultraviolet B irradiation and heat-treatment of 24.8 ± 1.44 mg of coated 7-dehydrocholesterol.

Vitamin D3 is available in oral and injectable dosage forms. Interest in the transdermal route as an alternative to the oral and parenteral routes has grown recently. In this study, several film-forming solutions for the transdermal delivery of vitamin D3 were prepared. They contained 6000 IU/mL of vitamin D3 that formed a dry and acceptable film in less than 5 min after application. The formulations consisted of ethanol and acetone 80:20, and one or more of the following ingredients: Eudragit L100-55, PVP, PG, limonene, oleic acid, camphor, and menthol. Vitamin D3 release was studied from both the film-forming solution and pre-dried films using a Franz diffusion cell. The film-forming solution released a significant amount of vitamin D3 compared to the dry film, which is attributed mostly to the saturation driving force due to the evaporation of volatile solvents. In vitro permeation studies through artificial skin Strat M® membrane revealed that the cumulative amount of vitamin D3 permeated after 24 h under the experimental conditions was around 800 IU across 3.14 cm2. The cumulative permeation curve showed faster permeation in earlier stages. Young’s modulus, viscosity, and pH of the formulations were determined. Most of the formulations were stable for 3 weeks.

Abstract:
Vitamin D plays a crucial and very well-known role in regulation of calcium homeostasis and bone metabolism and mineralization. However, a huge and more recent body of evidence supports the positive influence of vitamin D on the regulation of immune response, ranging from protection against respiratory tract infections to prevention and management of asthma. Nevertheless, vitamin D deficiency is a very common condition and there is an increasing need for suitable products for proper supplementation, allowing good compliance also in specific populations. Orally disintegrating tablets (ODT) were first developed to overcome the difficulty experienced by pediatric and geriatric patients of swallowing traditional oral dosage forms and, recently, orodispersible films (ODF) are gaining popularity as novel dosage form for assuming active pharmaceutical ingredients, vitamins, and ingredients for food supplements. This study describes a 2000 IU Vitamin D3 ODF for daily intake, consisting of hydrophilic polymers and suitable excipients, manufactured by film-casting process. Elongation-at-break (E%), Young’s modulus (Y), and tensile strength (TS) were investigated using a dynamometer. Chemical stability was evaluated assaying the vitamin D3 in the films stored at different environmental conditions. In addition, in vitro disintegration and dissolution studies were performed. Correlation existed between the mechanical properties of the film and the residual water, acting as plasticizer. The stability study showed that vitamin D3 assay was ≥90% also after 3 months at 40 °C. The film disintegrated in less than 1 min and the vitamin D3 released was ≥75% after 15 min. An ODF with suitable properties can be manufactured and used as innovative dosage form for vitamin D3 food supplements.

Stora mängder miljödata samlas hela tiden in och för att använda all data behöver den förstås av användarna så de kan applicera kunskapen inom deras område. Visualisering skapar förståelse om data. Heatmaps kan användas för att visualisera geospatial data och interaktivitet är ett hjälpmedel för att skapa ytterligare grafiska representationer. I detta arbete evalueras JavaScript-teknikerna D3.js, Heatmap.js och Vue.js angående vad som är mest lämpligt för att visualisera geospatial data utifrån effektiviteten vid utritning av heatmaps. Ett experiment genomförs där biblioteken D3.js, Heatmap.js testas i ramverket Vue.js. Detta för att ta reda på vilket bibliotek som föredras vid utritning av heatmaps och om ett ramverk påverkar resultatet. En miljö sätts upp för att genomföra undersökningen och tester för att påvisa detta. Resultatet indikerar att Heatmap.js och mindre datamängder ger en lägre utritningstid i den tillämpning som undersökts. I framtiden är det intressant att undersöka flera bibliotek och flera datamängder.

Introdução: A hidradenite supurativa (HS) é uma doença folicular inflamatória crônica causada pela oclusão e ruptura de unidades foliculares, com acometimento secundário de glândulas apócrinas, principalmente nas regiões axilar, inframamária, inguinal e anogenital. Afeta 1 a 4% da população, com maior incidência em mulheres (3:1). A obesidade e o tabagismo são importantes agravos à doença. Objetivo: Divulgar conhecimento científico sobre a HS, a fim de expor e elucidar os desafios diagnósticos e, principalmente, de seu manejo. Material e Métodos: Realizada revisão da literatura científica disponível nas principais bases de dados, incluindo análise do mais recente editorial publicado sobre HS pela The International Society for the Study of Vulvovaginal Disease (ISSVD). Resultados e Conclusão: Geneticamente, há fraqueza de parede folicular, com facilidade em sua ruptura. De acordo com a severidade, é classificada em Estágio I de Hurley — formação de abscesso, único ou múltiplo, sem fistulização ou cicatriz, geralmente de resolução espontânea; Estágio II — lesões coalescidas, fibrose cutânea e fístulas, por abscessos recorrentes, persistente; Estágio III — envolvimento difuso, conexões de tratos fistulosos, abscessos e cicatrização hipertrófica fibrosa. Diagnosticada pela característica típica das lesões, por sua topografia e pela presença de recorrência e cronicidade. O tratamento objetiva prevenir o aparecimento de novas lesões e reduzir a extensão e a progressão das existentes; prevenir a cicatrização e a fistulização das lesões extensas; estimular a cessação do tabagismo, a prática de depilação a laser; e reduzir traumas adicionais, evitando-se calor, umidade, fricção e suor. Em todos os estádios, considerar o uso de metformina, principalmente em sobrepeso. No geral, sugere-se o uso de medicações tópicas, com clindamicina loção 1%, e corticoides intralesionais, como acetato de triancinolona, e antibioticoterapia sistêmica por 7‒10 dias, com amoxicilina + clavulanato, doxiciclina ou clindamicina. Terapia antiandrogênica com drospirenona e etinilestradiol, e uso de espironolactona ou finasterida mostraram-se benéficos em alguns estudos. O uso de gluconato de zinco, vitamina C e vitamina D3 foi favorável na prevenção de novas lesões. O tratamento cirúrgico envolve incisão ampla quando de forma persistente. No Estágio III, a terapia principal é cirúrgica, sendo as outras medidas farmacológicas também importantes, mas de menor impacto. Recomenda-se o uso de imunobiológicos, como infliximab ou adalimumab, por serem grandes aliados na redução de sinais flogísticos no pré-operatório, a fim de simplificar a cirurgia. A HS é uma enfermidade de difícil diagnóstico e manejo. Não se dispõe de um tratamento ideal que forneça bons resultados funcionais e estéticos para todos os pacientes. A abordagem cirúrgica vem despontando como alternativa com bons resultados. A informação e o apoio são essenciais, haja vista o impacto na qualidade de vida gerado pela doença.

Abstract:
Devido à popularização de Geckos como pets e a falta de informação dos tutores, a incidência de problemas relacionados ao manejo sanitário e alimentar tem aumentado. Poucos relatos foram descritos sobre a ingestão de corpos estranhos e os seus devidos tratamentos em Geckos, o que torna relevante a descrição deste caso para um maior conhecimento acerca do manejo e das técnicas terapêuticas nessa espécie. Um Gecko Leopardo, sem sexo definido, com 7 meses de idade, pesando 29 gramas, vivia em um terrário, com papel toalha como substrato, condições de temperatura (25-30ºC), sem fonte de luz e umidade (50%), alimentado com presas vivas suplementadas com cálcio e vitamina D3. Segundo a tutora, o animal encontrava-se sem comer e com ausência de fezes a 2 dias. No exame físico apresentava-se apático e cavidade celómica encontrava-se dilatada com presença de conteúdo nodular firme palpável na região caudolateral. O paciente foi internado e submetido a tratamento médico com dose inicial de Lactulona de 0,5 ml/kg SID, Metroclopramida oral 0,7ml/kg SID, banho morno por 15 minutos TID. No quarto dia de internação foi adicionado a massagem com vibrador TID, aumentado para 230 ml/kg a dose da Lactulona e realizado fluidoterapia com Ringer Lactato na dose de 10% do peso do paciente. Em menos de 24 horas, após as mudanças no tratamento, o paciente defecou. No dia seguinte foi realizado exame radiográfico e foi adicionado cálcio oral na dose de 37ml/kg. No sétimo dia foi realizado a lactulona BID. No oitavo dia o paciente defecou, foi identificado papel toalha como corpo estranho. O paciente alimentou-se sozinho e recebeu alta no décimo dia após a realização de outro exame radiográfico. Os exames radiograficos foram realizados para diagnóstico e acompanhamento clínico do paciente no qual o primeiro havia conteúdo heterogênio em região de estômago e intestino. Nos dias seguintes houve diminuição significativa do conteúdo intraluminal de alças intestinais. A impactação por corpo estranho é um problema comum na clínica de répteis por estar diretamente relacionada ao manejo ambiental inadequado e deficiência nutricional destes animais. A ingestão de substrato utilizado no terrário é considerada uma das principais causas de estase no trato gastrointestinal, e consequentemente ocasionar problemas mais graves de quadros de obstrução (1). O tratamento medicamentoso juntamente com banho de água morna e a realização de massagem com vibrador foram eficazes para a eliminação do corpo estranho sem intervenção cirúrgica.
