Full textWe study some properties of Double Manifold, or D-Manifolds. In particular, we study of deformations of D-structures and of CR D-structures, and we found a condition which is equivalent to the classical Maurer-Cartan equation describing the integrability of the deformations. We also focus on the cohomological properties of D-Manifold, showing that a del-delbar-Lemma can not hold for any compact D-Manifold. We also state some properties of special subgroups of de-Rham cohomology, studing also their behaviour under small deformations. Finally, a result by Harvey and Lawson about the minimal Lagrangian Submanifold of a D-Kahler Ricci-flat manifold is generalized to the case of a special almost D-complex symplectic manifold.

Full textThe vitamin D nuclear receptor (VDR) is a transcription factor activated by the biologically active form of vitamin D3. VDR is a potential candidate to treat neurodegenerative and autoimmune disorders, and cancer. VDR modulates the expression of vitamin D3-regulated genes by selective recruitment of coregulators of transcription which are, in turn, attractive targets in epigenetic-oriented drug discovery. Available structural data for receptor-coregulator complexes are limited; investigation of such complexes is highly important. The present work focuses on the architecture of the complex between VDR and a large part of the coactivator MED1, a subunit of the Mediator complex linking nuclear receptors to the basal transcription machinery. Obtained results revealed important details of the interaction, as well as the overall organization of the complex. This work provides a solid background for the structural investigation of similar complexes involved in the transcriptional control

Full textThe eukaryotic initiation factor 3 (eIF3) complex plays a core role in the interaction network among several eIFs that assemble on the 40S ribosomes and participate in the different reactions throughout the translation initiation pathway. The Saccharomyces cerevisiaea eIF3 complex comprises five subunits, all of which are the core subunits of the mammalian eIF3 complex consisting of 13 subunits. Attempts to decipher its tridimensionnal structure are under way. A first path to study the structure of this complex is to complete the identification of binding regions, few of which are currently known. Recently, the interaction region between eIF3i and extreme C-terminal domain of eIF3b has been obtained through NMR and crystal structure. On the other hand, the interaction region between 3i and 3g, although located to the N-terminal domain of 3g still remains to be defined. Hydrogen/deuterium exchanges (HDX) have been developed for a long time and are widely used for structural studies of proteins and multiprotein complexes. It is commonly analyzed using mass spectrometry. The most classic standard HDX-MS approach consists in making a mass measurement of deuterium-labelled peptides from an enzymatic digestion of the protein of interest to determine the level and rate of deuterium incorporation. In this study, a high performance 7 T FT-ICR mass spectrometer was used in combination with nanoLC separation to acquire highly accurate HDX-MS data. The precision on the mass measurement of FT-ICR MS is by itself not sufficient to unambiguously identify peptides from a pepsin digest due to the lack of pepsin specificity. We therefore developed a statistical approach for peptide identification, based on a probability of occurrence value of a given peptide within a pepsin digest. In combination with high mass accuracy, this method allows efficient identification of the peptides, without additional need of MS/MS verification. This method has been applied on the study of the binding regions in the complexes of eIF3i:bC3 and eIF3i:gC1ΔC. Peptide reference lists with high sequence coverage and rich sequence superposition ensured structure elucidation with high spatial resolution. For the binding of 3i and 3b, the detailed interaction regions were unveiled for proteins in the solution phase which resembled the physiological condition and were coherent with the reported protein structure, thus provided complimentary information to the crystallographic structure in solid phase. For the binding of 3i and 3g, the interaction regions were studied with the absence of any atomic structural information of 3g. This provides significant insights of the complex formation of 3i and 3g, and for the first time the precise binding regions were successfully revealed

Full textLa voie de synthèse par méthode solvothermale a permis de caractériser des complexes originaux à base de macrocycles. Deux complexes dinucléaires à base de CoII et FeIII où les deux centres métalliques sont pris en « sandwich » entre deux unités calixarènes ont été ainsi isolés. Ces dimères ont été utilisés comme brique moléculaire de départ pour l'élaboration de cluster à plus haute nucléarité. Un complexe associant six ions CoII a pu être isolé et caractérisé.
La synthèse de nouveaux de ligands de type base de Schiff résultant de la condensation de l'acetylacétone avec différents acides aminés a été développée et leur complexation avec différents éléments de la première série de transition a été étudiée. Ainsi, un trimère à base de CuII a été caractérisé et ses propriétés magnétiques ont été modélisées par une série de calculs ab initio. Cette étude théorique a été complétée par un ensemble de mesures RPE en solution en fonction de la température.
Enfin, la chimie de coordination de ligands de type β-dicétone, en particulier l'acétylacetonate, avec différents métaux (3d et 4f ) a été revisitée. Ce ligand et certains de ses dérivés permettent de contrôler la nucléarité d'édifices moléculaires à base d'ions 3d et 4f. L'utilisation d'une brique moléculaire préformée de type cubane a permis d'isoler un complexe heptanucléaire de type « double cubane » à base de NiII qui présente les caractéristiques d'une molécule-aimant. En jouant sur l'effet de l'encombrement stérique du ligand, trois familles de complexes polynucléaires Ln5, Ln8 et Ln9 ont été obtenues par auto-assemblage suivant les méthodes de la chimie douce à base d'ions 4f. Des études structurales, magnétiques et une analyse approfondie des caractéristiques spectroscopiques des complexes à base d'europium(III) ont été réalisées. Une étude complète a été menée sur une famille de complexes Y9 dopés en europium (Y9-xEux).

Full textCoordination chemistry of transition metals gives an access to numerous molecular magnetic materials that exhibit varied properties. During this work, several ligands with intrinsic properties have been chosen and their chemistry has been developed in order to get specific magnetic behavior. Solvothermal syntheses allowed the characterization of several complexes based on macrocycles. Two dinuclear CoII and FeIII complexes were isolated. Their molecular structures are built from metal ions located like in a "sandwich" between two calixarenes units. These complexes considered as molecular starting blocks were used for the synthesis of clusters with higher nuclearity. An original complex associating six CoII ions has been then isolated. The complexation of Schiff base ligands resulting from the condensation of the acetylacetone with various amino acids has been performed. From one of these original ligands, an almost perfect triangular trinuclear CuII complex has been then isolated and fully characterized. Ab initio calculations have been carried out in order to explain its magnetic properties. Finally, the complexation of β-diketone based ligands with various metals was explored. It has been then possible to control the nuclearity of molecular buildings block. The use of a preformed brick (cubane type) gave an access to a heptanuclear NiII complex. This latter presents the characteristics of a single molecule magnet. The last stage of these researches was to modify the steric effect of these β-diketone ligands in order to control the final cluster size. Three families of polynuclear 4f complexes (Ln5, Ln8 and Ln9) were obtained. Structural, magnetic studies and detailed analysis of the spectroscopic characteristics of these europium(III) complexes were achieved. A completely original work has been carried out on Y9 clusters doped with Eu (Y9-xEux) by linking molecular structures with luminescence properties

Full textDans un premier temps, nous étudions l'apparition de modes localisés en champ proche de structures complexes. Nous nous intéressons à deux différents types de structures: des nanoantennes d'or et des films d'or désordonnés. Nos résultats nous permettent de discerner les modes radiatifs et non-radiatifs. Nous introduisons le concept de Cross Density Of States (CDOS) pour décrire quantitativement la cohérence spatiale intrinsèque associée à la structure modale d'un milieu complexe. Nous démontrons ainsi une réduction de l'extention spatiale des modes au voisinage de la percolation électrique des films d'or désordonnés.
Nous nous intéressons ensuite à des milieux fortement diffusants. En éclairant de telles structures par une source cohérente, on obtient une figure d'intensité complexe appelée speckle. Nous utilisons une méthode diagrammatique pour démontrer une corrélation négative entre les figures de speckle réfléchie et transmise à travers une tranche dans le régime mésoscopique. Nous nous intéressons ensuite à la corrélation C0, qui apparait lorsque la source est enfouie dans le milieu. Nous proposons une démonstration générale de l'égalité entre la corrélation C0 et les fluctuations normalisées de la LDOS, et soulignons le rôle fondamental des interactions de champ proche. Finalement, nous observons numériquement le régime de couplage fort entre un diffuseur résonnant et un mode localisé d'Anderson au sein d'un milieu désordonné 2D.

Full textThe L7Ae family proteins are essential components of many RNPs. In vertebrates, C/D and H/ACA snoRNPs are involved in ribosome biogenesis, the U4 snRNP in pre-mRNA splicing, the telomerase complex in telomeres replication, and mRNP SECIS in selenoproteins translation. Like most eukaryotic RNPs, assembly in functional entities is not an autonomous process and requires the intervention of specialized factors. Basing our study on the assembly of C/D snoRNP in the model organism Saccharomyces cerevisiae, and using approaches of molecular biology, biochemistry and genetics, we undertook to decipher these mechanisms. Our work has helped to identify a set of proteins, acting in a coordinated manner within a machinery conserved between yeast and human. This machinery consists of two major subunits: (i) Rsa1p/NUFIP, a platform protein that interacts with some proteins of the L7Ae family and facilitates the RNPs assembly, (ii) the R2TP complex (Rvb1p/TIP49, Rvb2p/TIP48, Pih1p/PIH1, Tah1p/SPAGH), which could induce conformational remodeling necessary for the formation of mature RNPs. In addition to these key players, other factors appeared closely linked to this mechanism. The Hit1p/TRIP3 protein interacts with Rsa1p/NUFIP and is required to ensure its stability in yeast. HSP90 chaperone, whose role is predominant in human, operates on some components of the RNPs. Finally, the Bcd1p/BCD1 protein is associated specifically with this machinery during C/D snoRNPs assembly
