This research considers the potential motivations of using a social app for the realization of donations and probe the symbolic interactions that occur during solidarity exchanges mediated by technology. Smartphones and their applications are inseparable increasingly part of the daily lives of people who spend all their time with their devices: always on, always connected, as close as the shirt itself. Applications for mobile devices -apps- are the software installed in these smartphones that perform or assist in performing tasks for their users. Among the tools that the various types of apps present, there is a category of social apps, which are those facing a social cause. The conducted study noted when addressing the social application Solidarius, created for realization of donations to the group of welfare Sociedade S??o Vicente de Paulo, Vicentinos, which by enabling the realization of solidarity mediated by technology, materializes a new type of solidarity , anonymous, but allows the existence of a link, which excludes the direct involvement with people in common vulnerability in voluntary actions, but allows the user a sense of belonging, even if momentarily, to a larger institution or cause himself, and generate feelings of satisfaction and gratitude. These observations were made possible by the bibliography study published on the subject, the data analysis and information gathered during interviews with the developers of social application on screen and representatives of the institution that it will use, and the above discussion in focus group held with potential users of the application. As a result, to be detailed in subsequent research, it was realized that social applications allow its users an empowerment, regarding the perception that are interfering with reality and contributing to a cause, and that the technology of apps, for convenience, ease and closeness they have with the user, can be instruments of social change.
Esta pesquisa considera as potenciais motiva????es do uso de um aplicativo -app social para a realiza????o de doa????es e sonda as intera????es simb??licas que ocorrem durante as trocas solid??rias mediadas pela tecnologia. Os smartphones e seus indissoci??veis aplicativos fazem cada vez mais parte do cotidiano das pessoas, que passam todo o tempo com seus aparelhos: sempre ligados, sempre conectados, t??o pr??ximos como a pr??pria roupa do corpo. Os aplicativos para dispositivos m??veis -apps- s??o os softwares instalados nestes smartphones que realizam ou auxiliam a realiza????o de tarefas pelos seus usu??rios. Entre as ferramentas que os v??rios tipos de apps apresentam, existe a categoria de apps sociais, que s??o aqueles voltados para uma causa social. O estudo conduzido observou, ao abordar o aplicativo social SOLID??RIUS, criado para realiza????o de doa????es para o grupo de assist??ncia social Sociedade S??o Vicente de Paulo- Vicentinos, que ao possibilitar a realiza????o da solidariedade mediada pela tecnologia, materializa-se um novo tipo de solidariedade, an??nima, mas que possibilita a exist??ncia de um v??nculo, que exclui o envolvimento direto com as pessoas em situa????o de vulnerabilidade comum nas a????es de voluntariado, mas permite ao usu??rio um sentido de pertencimento, ainda que moment??neo, a uma institui????o ou causa maior que ele mesmo, al??m de gerar os sentimentos de satisfa????o e gratid??o. Estas observa????es foram possibilitadas pelo estudo de material bibliogr??fico publicado sobre o assunto, pela an??lise de dados e informa????es colhidas durante a realiza????o de entrevistas com os desenvolvedores do aplicativo social em tela e representantes da institui????o que dele far?? uso e pelo debate exposto em grupo focal realizado com potenciais usu??rios do aplicativo. Como resultados, a serem aprofundados em pesquisas posteriores, foi percebido que os aplicativos sociais permitem aos seus usu??rios um empoderamento, no tocante ?? percep????o que est??o interferindo na realidade e contribuindo com uma causa, e, que a tecnologia dos apps, pela praticidade, facilidade e proximidade que possuem com o usu??rio, podem ser instrumentos de transforma????es sociais.

Full textAlzheimer’s disease (AD) is defined as a neurodegenerative disorder where synaptic defects lead to neuronal loss and concurrent memory impairments. It is now well-established that synaptic dysfunction in AD is initiated by oligomeric forms of the amyloid-β peptide (Aβ), a proteolytic derivative of Amyloid Precursor Protein (APP). However, the pathway by which Aβ induces its deleterious effects, whether it is due to intra- and/or extracellular Aβ pools, and how these effects are sustained and propagated throughout the brain, are still unclear.In this study, we used several mutated forms of APP which give rise to Aβ peptides with unique molecular signatures, such as: the Swedish mutation (K670M/N671L) (APPswe) which increases secreted (extracellular) Aβ; the Osaka mutation (E693Δ) (APPosa) which causes intraneuronal (intracellular) accumulation of Aβ; and the Icelandic mutation (A673T) (APPice) which has been reported to decrease Aβ production and protect against AD. These mutated forms of APP were overexpressed in cultured mouse cortical neurons in order to: i) study the morphology and function of dendritic spines, the post-synaptic element of synapses, by confocal microscopy, ii) get a better insight into pathology development and propagation and iii) identify a novel interacting partner bringing to light the possible physiologic role of Aβ in neurons.We report that pathological Aβ accumulation, due to APPwt, APPswe and APPosa overexpression but not APPice overexpression induces a significant decrease in spine density especially mushroom spines, accompanied by a significantly increased volume of the remaining mushroom spines, and that intracellular Aβ is sufficient to induce these effects. These enlarged mushroom spines have impaired structural plasticity as they did not increase in volume following synaptic activation seemingly as a result of defective activity-dependent actin dynamics in the spines. This alteration of synaptic morphology, structure and plasticity seems to be due to a newly-identified interaction between actin and Aβ, hinting a possible physiological role for Aβ in activity-dependent synaptic plasticity. We also show that synaptic activity modulates amyloïdogenic APP processing which, in pathological conditions, further exacerbates these synaptic defects. Furthermore, we show that Aβ sequence is as important as Aβ concentration in inducing synaptic alterations since pathological concentrations of Aβ harbouring the Icelandic mutation had no effect on spine density or volume. Lastly, we bring to light that secreted Aβ, not only affects the Aβ-secreting neuron itself, but also affects spine density of nearby neurons in an APP-dependent manner, reminiscent of a prion-like mechanism. Together these results demonstrate that APP processing is a finely tuned equilibrium involved in actin-remodelling during activity-dependent synaptic plasticity and opens a new route for AD therapeutic strategies

Full textAlzheimer’s disease (AD) is the most common neurodegenerative disease in the world. Failures of candidate treatments targeting the beta amyloid (Aβ), a catabolite originating from the amyloid precursor protein (APP), highlight the multifactorial nature of this pathology. Hence, the early defects of mitochondrial structure and functions, and the neuroinflammation are implicated in AD development. Several studies describe the AMPK (AMP-activated protein kinase) as a master regulator of mitochondrial homeostasis, notably via the elimination of damaged mitochondria by mitophagy, and also of neuroinflammation. The central hypothesis of my thesis postulates that, besides the Aβ peptide, other APP catabolites, the C-terminals fragments (APP-CTFs) can contribute to mitochondrial dysfunctions and that the modulation of the AMPK could hold beneficial effects in AD.My thesis is structured into two specific objectives:Axe 1. Study of the APP-CTFs contribution to the alterations of mitochondria structure, functions, and mitophagy in AD. We have observed an accumulation of APP-CTFs in mitochondria of human neuroblastoma cells stably expressing APP with the double Swedish mutation (SH-SY5Y-APPswe), or the APP-βCTF: C99 (SH-SY5Y-C99), causing an alteration of mitochondria morphology, an increase reactive oxygen species (ROSmit) production, and a blockade of mitophagy. These toxic effects were confirmed in presymptomatic 3xTgAD mice (APPswe, TauP301L, PS1 (M146V)) and in mice expressing the C99 fragment via a lentiviral approach and were exacerbated (in vitro and in vivo) following the inhibition of the γ-secretase, blocking the Aβ production and accumulating APP-CTFs. Finally, we have reported a mitochondrial accumulation of APP-CTFs in the temporal lobes necropsies of sporadic AD patients correlating with a defective mitophagic phenotype. Hence, targeting the early APP-CTFs accumulation and mitochondrial dysfunctions could constitute promising novel therapeutic targets in the AD context.Axe 2. Study of the impact of AMPK signaling cascade modulation in AD. We have observed a defective AMPK-ULK1 cascade in the temporal lobes necropsies of sporadic AD patients, in in vitro AD study models (SH-SY5Y-APPswe and -C99) and in symptomatic 3xTgAD mice. Afterward, we have demonstrated that the pharmacological inhibition of AMPK enhances APP-CTFs accumulation, worsen mitochondria structure alterations, triggers mitochondria membrane hyperpolarization, increases ROSmit production, and inhibits mitophagy in vitro. Pharmacological AMPK blockade also alters the dendritic spine maturation ex vivo (organotypic hippocampal slices lentivirally expressing APPswe). Oppositely, the pharmacological activation of AMPK alleviates mitochondria dysfunctions in vitro and favors the dendritic spine maturation ex vivo. We confirmed these observations using a genetic approach by expressing mutated AMPK constructs (constitutive active and dominant negative forms). Importantly, pharmacological activation of AMPK reduces Aβ and Tau pathologies, as well as neuroinflammation and learning impairments in symptomatic 3xTgAD mice. Overall, the stimulation of AMPK could stand as a new therapeutic avenue to alleviate AD pathogenesis

The purpose of this paper by performing a bibliographic review and case study, in São Carlos (SP), was to study the application of the Code Forest in Permanent Preservation Areas to urban water courses approaching legal aspects and the others laws corresponding. Like this to reach the problems and conflicts in the application of the Code Forest the use and occupation of the soil. Some subsidies were proposed in order to contribute to the definition of new technical and legal references for a more practical and adequate application of the urban APP, according to the context in which it is inserted on the search for a harmonic acquaintance among Man and Nature.
O presente trabalho através de revisão bibliográfica e estudo de caso, na cidade de São Carlos (SP), estudou a aplicação do Código Florestal em Áreas de Preservação Permanente de corpos d água urbanos, levantando seus aspectos legais e de outras leis correlatas. Desta maneira chegou-se aos problemas e conflitos existentes na aplicação do Código Florestal na área urbana no uso e ocupação do solo. Foram propostos alguns subsídios que possam contribuir na busca de novas referências técnicas e legais para uma aplicação prática mais adequada do instituto da APP urbana, conforme os contextos nas quais está inserida e na busca de uma convivência harmônica entre Homem e Natureza.

Full textThe amyloid precursor protein (APP) is the key element in the appearance and the development of Alzheimer's disease (AD). Basically, APP can be processed following two maturation pathways. The fist one, named the amyloidogenic pathway, involves a secretase, occurs mostly in the endosomes and leads to the release ofbeta-amyloïd peptides that are neurotoxics if overproduced. The second one, named the non-amyloidogenic pathway, involves analpha-secretase, occurs mostly at the plasma membrane and leads to the release of soluble APP alpha fragments that are neuroprotective. First, our work demonstrated that a serotonin receptor, the 5-HT4 receptor (5-HT4R), is able to form a protein complex with the alpha-secretase ADAM10 and APP and that this receptor could modulate ADAM10 and APP trafficking and maturation. By this way, the 5-HT4 receptor is able to increase ADAM10 and APP cell surface localization and to constitutively promote sAPP alpha release. In a second time, using an unbiased proteomic approach, we focused on the quest of protein partners able to interact with this complex and to promote the non-amyloïdogenic pathway. This work allowed us to propose original hypothesis about shared mechanisms able to regulate 5-HT4R, APP and ADAM10 endocytosis and trafficking. These mechanisms involve the AP-2 complex and another protein complex involved in the cellular trafficking of the amyloid precursor protein
