Abstract:
Purebreds, crossbreds and synthetic populations of sheep in North America of divergent genetic types produced 47–103 litres of milk for 121–144 d, consistent with "non dairy-type" breeds; however, this was substantially less than the 200–330 kg of milk per lactation characteristic of "dairy" type breeds in Europe and the Middle East countries. Among established breeds, the Suffolk (Su) breed produced significantly more total and daily milk yield, respectively, for a longer lactation (79 litres and 589 mL for 139 d), followed by the Rambouillet (Ra), Targhee (T) and Dorset (D) breeds (60–62 litres and 422–542 mL for 120–132 d), while the Lincoln (L) breed produced significantly less (48 litres and 285 mL for 128 d). Among fecund-type breeds and their crosses, Finnsheep (F) and Romanov (Ro) breeds produced significantly less (50–55 litres and 425–463 mL, for 122–129 d), while Ro × F produced significantly more milk (91 litres and 744 mL for 126 d), followed by F × Ro (60 litres and 505 mL for 128 d). Among other crossbreds, F × Ra produced significantly more milk (66 litres and 434 mL for 136 d) followed by L × Ra, L × D, F × L, F × Ra (49–60 litres and 296–385 mL for 123–136 d), while Ra × D produced significantly less (47 litres and 245 mL for 121 d). Among multiple breed composite populations, the Outaouais (O) and Rideau (R) Arcott breeds and their crosses had the highest milk production, e.g., R and R × O produced significantly more milk (95–103 litres and 754–820 mL for 131–144 d) than O and O × R (74–80 litres and 621–693 mL for 124–142 d). Concurrently, Synthetic I (½ F, ½ L) produced significantly less (58 litres and 494 mL for 134 d), while Synthetic III (¼ F, ¼ L, ¼ D, ¼ Ra) produced significantly more milk (91 litres and 673 mL, P > 0.05, for 130 d), and Synthetic II (½ D, ½ Ra) was intermediate (74.8 litres and 621 mL for 129 d); however, lactation length was consistent. Sheep breeds in North America exhibited diversity that was substantial for milk yield, but negligible for lactation length, as well as fat, protein, lactose and solid-not-fat content. Lack of inherent potential for milk production continues to be a serious constraint towards development of commercial dairy sheep industry. In the absence of "dairy-type" breeds, superior milk production in the Rideau Arcott breed (with East Friesian lineage) and their crosses demonstrates considerable promise for further improvement, thus increasing the efficiency of sheep production. Key words: Milk yield and composition, sheep breeds, synthetic sheep, Arcott breeds

Goal. To evaluation of the shallot gene pool for bulb yield and its components, resistance to viral diseases and to select the most valuable accessions for breeding programs. Results and Discussion. The article presents the results of evaluating 25 shallot accessions for yield and resistance to viral diseases. Five accessions with very high bulb yields (> 135% related to the standard) were selected from the following regions of Ukraine (from the Dnipropetrovska region - D-135 (13.1 t/ha); from the Poltavska region - D-127 (13.9 t/ha); from the Kharkivska region - D-15 (14.0 t/ha), D-34 (14.9 t/ha)) and from the Netherlands (Bonilla (15.9 t/ha). Accessions with high yields (116 - 135% related to the standard) were from the Dnipropetrovska region (D-136 (11.2 t/ha), varieties Olviia (12.3 t/ha) and Kushchyovka Kharkovskaya (12.6 t/ha). Ten accessions were the best in terms of bulb weight, exceeding the standard by 3.0 - 8.3 g:- D-33 (14.2 g), Olviia (14.5 g), D-4 (14.7 g), D- 133 (15.1 g), D-137 (15.2 g), D-130 (15.7 g), D-127 (15.9 g), D-15 (16.6 g), D- 34 (18.3 g), and Bonilla (19.5 g). Seven accessions were noticeable for the bulb number per bulb stem: from the Dnipropetrovska region (D-136 (6.1 bulbs), D-123 (7.0 bulbs), D-124 (7.4 bulbs), D-135 (7.4 bulbs), D-137 (6.4 bulbs), and D-129 (8.0 bulbs)) and from the Netherlands (Bonilla (6.1 bulbs). The following accessions were distinguished by resistance to viral diseases (onion yellow dwarf and mosaic) as sources for breeding: with very high resistance (9 points) - D-120 from Russia, 12 accessions with high resistance (7 points) from Ukraine (D- 4, D-26, D-136, D-125, D-127, D-133, D-130, D-123, D-126, D-137, and D-140) and Bonilla from the Netherlands. Accessions D-127 (Poltavska region), D-137 (Dnipropetrovska region), D-34 and D-15 (Kharkivska region), Bonilla (Netherlands) with a set of traits are of practical interest for breeding. Conclusions. Having studied 25 shallot collection accessions in 2016 – 2018, we distinguished the following accessions: by bulb yield - 8 accessions (11.7 - 15.9 t/ha), by bulb weight - 10 accessions (14.2 - 18.3 g), by the bulb number per bulb stem - 7 accessions (6.1 – 8.0 bulbs), by resistance to viral diseases - 13 accessions (7 – 9 points), by a set of valuable economic traits – 5 accessions.

AbstractObjectiveTo explore whether initiatives to promote fruit and vegetables in primary schools are associated with changes in children’s diet.DesignCross-sectional dietary survey. Main outcome measures were intakes of fruit, vegetables and key nutrients; and a score for initiatives promoting fruit and vegetables in school.SettingOne hundred and twenty-nine English primary schools.SubjectsYear 2 children (aged 6–7 years,n2530).ResultsIn schools running a gardening club, children ate more vegetables, 120 (95 % CI 111, 129) g/d, compared with those that did not, 99·3 (95 % CI 89·9, 109) g/d; and where parents were actively involved in school initiatives to promote fruit and vegetables, children’s intake of vegetables was higher, 117 (95 % CI 107, 128) g/d, compared with those where parents were not involved, 105 (95 % CI 96·2, 114) g/d. In schools that achieved a high total score (derived from five key types of initiatives to promote fruit and vegetables in school) children ate more vegetables, 123 (95 % CI 114, 132) g/d, compared with those that did not, 97·7 (95 % CI 88·7, 107) g/d.ConclusionsGardening, parental involvement and other activities promoting fruit and vegetables to children in school may be associated with increased intake of vegetables but not fruit. These effects were independent of deprivation status and ethnicity.

Abstract:
The article reviews the literature on the use of vitamin D in endometriosis treatment. Endometriosis, one of the most common health problems in the female population, is an estrogen-dependent chronic inflammatory disease. Endometriosis affects 10-15% of all women of reproductive age and 35–50% of women with chronic pelvic pain and/or infertility. Despite multiple studies in all aspects of endometriosis, the selection of the most effective individualized combination treatment options for each patient is still considered as a pressing issue. Animal experiments and clinical trials have demonstrated multiple immune system changes in endometriosis occurring at different levels, from aberrant cytokine signaling to alterations of the immune system cell populations. There is a growing number of recent reports in the international and domestic literature describing the impact of vitamin D on the clinical course of endometriosis and the options of its use in this disease. There is a pathogenetic rationale for involving vitamin D in combination therapy of endometriosis in view of its immunomodulatory, anti-proliferative and anti-angiogenic properties. However, it is suggested that further studies are needed to determine an optimum therapeutic dose of vitamin D for different endometriosis types. Abstract:
Purpose: Lonidamine (LND) can enhance the activity of anthracyclines in patients with metastatic breast cancer. A multicenter, prospective, randomized trial was designed to determine whether the association of LND with high-dose epirubicin plus cyclophosphamide (EC) could improve disease-free survival (DFS) in patients with early breast cancer (BC) compared with EC alone. Granulocyte colony-stimulating factor (G-CSF) was added to maintain the EC dose-intensity. Patients and Methods: From October 1991 to April 1994, 506 patients with stage I/II BC were randomly assigned to four groups: (A) epirubicin 120 mg/m2 and cyclophosphamide 600 mg/m2 administered intravenously on day 1 every 21 days for four cycles (124 patients); (B) EC plus LND 450 mg/d administered orally (125 patients); (C) EC plus G-CSF administered subcutaneously (129 patients); (D) EC plus LND plus G-CSF (128 patients). Results: Median follow-up was 55 months. Five-year DFS rate was similar for LND (B+D groups; 69.6%) versus non-LND arms (A+C groups; 70.3%) and G-CSF (C+D groups; 67.2%) versus non–G-CSF arms (A+B groups; 72.9%). Five-year overall survival (OS) was comparable in LND (79.1%) versus non-LND arms (81.3%) and in G-CSF (80.6%) versus non–G-CSF arms (79.6%). DFS and OS distributions in LND and G-CSF arms did not change according to tumor size, node, receptor, and menopausal status. G-CSF dramatically reduced hematologic toxicity without having a significant impact on dose-intensity (98.1% v 95.5% for C+D and A+B groups, respectively). Conclusion: EC is active and well tolerated in patients with early breast cancer. The addition of LND or G-CSF does not improve DFS or OS.

Abstract:

Abstract: The ASTM D1209-00 method describes the procedure for the visual measurement of the color of essentially light colored liquids, which involves the visual comparation between the color of the samples with the color of Pt-Co standards solutions, with concentration from 1 to 500 unities of Pt-Co, the ASTM D 1209-00 scale (solutions obtained by mixing potassium chloroplatinate, from 1,5x10 to 3,6x10 mol L and cobalt chloride in the concentrations of 1,7x10 to 4,2x10 mol L). It is kown and was confirmed in tests with volunteers that there and serious difficulties with this method for very diluted solutions (concentration from 1 to 25 unities of Pt-Co), that are extremely clear, hardly distinguished by visual observation, whose in many cases represent the samples of most interest in some application sectors. In an attempt to overcome these difficulties, a robust and low cost device was developed for a rapid analysis. A blue light emitting diode (LED) was used as a radiation source and a Photo IC Diode S7183 (Hamamatsu) as the detector, in a set of black PVC cells with different options of optical path and with the option of using or not an internal glass tube to load the solutions. Analytical curves were obtained with second degree fitting for 340 and 170 mm cells. An 85 mm cell gives linear analytical curve with correlation coefficient = 0.9978, from 25 to 500 units of Pt-Co. With the use of the glass tube, curves with second order polynomial fitting were obtained for 170 and 85 mm cells. The performance of the device developed were tested with samples of epoxy bisfenol, cumene hidroperoxide, salicilic acid in ethanol and in aqueous solution of sodium carbonate 10 % w/v and cyclohexanol. The results presented deviations of the order of ± 1 units of Pt-Co, more precises than the ASTM method, which presents deviations up to ± 4 units of Pt-Co; with respect to the accuracy, there was statistic difference at the level of 95 %, which can be related to problems with the reference method

Abstract:
This paper explores the differential marking of (human) objects (DOM) in a corpus of old Sardinian texts from two different areas, Logudoro and Arborea. We investigate the constraints on the marking of objects, whether semantic, reflecting the Individuation Hierarchy (Silverstein, 1976, p. 122), syntactic, i.e., determined by verbal valency and/or the syntactic position of the O argument in relation to the verb and in the clause, or pragmatic, functioning as a marking device for topics. The data investigated reveal the role played by the notion of Individuation in the grammaticalization of the preposition a(d) as a DOM marker, which appears to spread progressively from the core to peripheral points along the hierarchy, namely from human proper names, to human, kinship and then common nouns, initially definite, subsequently indefinite. Already in 11th–13th century texts, a(d) does not appear to have only/mainly an identificational function, differentiating the A and O arguments when they are both high on the Individuation hierarchy, but has become a marker of high Individuation, its use being sensitive to the notions of definiteness and humanness.

Abstract:

In 10th International Conference on Nuclear Engineering. ASMEDC, 2002. http://dx.doi.org/10.1115/icone10-22324.

Full text
Abstract:
The critical heat fluxes (CHFs) of subcooled water flow boiling are systematically measured for the flow velocities (u = 4.0 to 13.3 m/s), the outlet subcoolings (ΔTsub, out = 3 to 129 K) and the outlet pressure (Pout = 800 kPa). The SUS304 test tubes of 3, 6, 9 and 12 mm in inner-diameter, d, and 33, 66, 99 and 133 mm in length, L, respectively for L/d = 11 are used. The CHFs first become lower and then become higher with the increase in subcooling. The CHFs for four different inner-diameters with L/d = 11 measured here become higher with the decrease in the diameter. CHF correlation for the latter increasing regime was given in non-dimensional form against average outlet subcoolings based on the experimental data. The correlation can describe not only the CHFs obtained in this work at the outlet pressure of 800 kPa but also the authors’ published CHFs (1284 points) for the wide range of Pout = 159 kPa to 1 MPa, d = 6, 9 and 12 mm, L = 49, 99 and 149 mm, ΔTsub, out = −4 to 130 K and u = 4.0 to 13.3 m/s within 15% difference for 50 K≤ΔTsub,out≤130 K and within +30 to −10% for 30 K<ΔTsub,out<50 K.

Full text
Abstract:
A number of the key inhibitors of coagulation and fibrinolysis have recently been shown to be members of the same superfamily of serine protease inhibitors, the serpins. The archetypes of the group are alpha-l-antitrypsin and antithrombin and it includes antiplasmin, C1-inhibitor, heparin cofactor II and the newly recognised inhibitors of plasminogen activators and activated Protein C. Alignment of their structures shows that they have the same skeletal three-dimensional conformation and, by inference, the same general function mechanisms. The serpins have a reactive centre, primarily dependent on a single amino acid, exteriorly placed on a stressed peptide loop. This functions by offering the cognate protease a high-affinity substrate that resists complete cleavage to form a tight 1:1 complex of inhibitor and protease that is subsequently removed from the circulation. The loop is vulnerable to cleavage with resulting loss of inhibitory activity. This irreversible switch is utilised: pathologically by venom and invasive bacterial proteases; and physiologically by the neutrophil leucocyte to modify local inflammatory responses. These mechanisms contribute to the changes seen in DIC and the shock syndromes.Modelling of antithrombin indicates the likely topological features involved in the binding of heparin, namely a sphere of positive charge centred on the A and D helices and involving Arg 47, Lys 125, Arg 129 and probably Arg 132 and Lys 133.Because the serpins are largely dependent for their specificityon a single amino acid it is now possible to precisely tailor inhibitory activity by site specific mutation. This has been used to produce recombinant antitrypsins that function as an improved inhibitor of neutrophil proteases (valine or leucine reactive centre), or as an analogue of antithrombin (arginine reactive centre). An elegant application of this approach is the engineered mutants of antiplasmin recently described by Holmes, Collen and colleagues (Leuven).

Abstract:
Factor XII clotting activity (FXII), Plasma Pre-kallikrein amidolytic activity (PK), 2-Macroglobulin ( α2-M) and cT-Inhibitor (cT-Inh) antigen have been measured in 17 patients immediately before and sequentially up to four months after kidney transplantation. Based on suspected Cyclosporin A (CyA) induced endothelial damage, activation of the contact system with resulting consumption of the contact activation factors was^evaluated. Before transplantation, FXII, PK, α2-M, Cl-Inh levels were 99±27%, 102±21%, 115±55%, and 129±32%, respectively. In the first two weeks after transplantation FXII decreased to 65±27%, PK to 67±20% and α2-M to 88±42%; Cl-Inh rose to a maximum of 201±44% (mean ± S. D.)(2p<0.001). Mean FXII levels correlated positively with PK, α2-M and albumin and negatively with CyA level and dose and serum bilirubin. PK and α2-M correlated positively with each other and with albumin and negatively with creatinine, bilirubin and CyA (p<0.01). The changes of FXII, PK and α2-M after transplantation suggest an influence of CyA on production or consumption of these factors. The behaviour of the cT-Inh may be unspecific and related to its action of an acute phase reactant.

"DEEP LEARNING NEURAL NETWORK IMAGE ANALYSIS OF IMMUNOHISTOCHEMICAL PROTEIN EXPRESSION REVEALS A SIGNIFICANTLY REDUCED EXPRESSION OF BIGLYCAN IN BREAST CANCER." In Brazilian Breast Cancer Symposium 2022. Mastology, 2022. http://dx.doi.org/10.29289/259453942022v32s2014.

Abstract:
Objective: The aim of this study was to compare the protein expression of biglycan (BGN) in normal breast tissue and in breast cancer using deep learning and digital HScore techniques. Methods: In this case-control study, 24 formalin-fixed, paraffin-embedded tissues were obtained from pathological archives for analysis. Normal breast (n=9) and breast cancer (n=15) tissue sections were analyzed by immunohistochemistry using BGN monoclonal antibody (M01 – Abnova), clone 4E1-1G7 at dilution 1:300 at pH 6, and 3,3’-diaminobenzidine (DAB) as the chromogen. Photomicrographs of the slides were analyzed using the ImageJ software with “color deconvolution”. After selecting the regions of interest (ROI), deconvoluted panels with DAB only were quantified using arbitrary DAB units. Another set, with higher magnification without ROI selection, was submitted to the inception V3 deep neural network image embedding recognition model. Next, supervised neural network analysis, using stratified 20-fold cross-validation, with 200 hidden layers, ReLu activation, and regularization at α=0.0001 were applied for SDLNN. The sample size was calculated for a minimum of seven cases and seven controls, having a power of 90%, an α error=5%, and a standard deviation of 20, to identify a decrease from the average of 40 DAB units (control) to 4 DAB units in cancer. Ethical approval was obtained from the Hospital de Clínicas de Porto Alegre Ethical Review Board (2019/0337). CAAE 15329119.9.0000.5327. Results: BGN expression (mean±SD) was 6.1±3.9 in breast cancer tissue, while in normal breast tissue, it was 39.6±21.9, using D-HScore (p=0.0017, student t-test, Welch corrected). SDLNN was able to correctly classify 110 out of 129 photomicrographs of the dataset using DAB panels only, with a classification accuracy of 85.3% (95%CI 78.1–90.3%) and the area under the curve of 94.3%. Conclusion: D-HScore and SDLNN revealed that BGN protein expression is reduced in breast cancer tissue, compared to normal tissue. The use of SDLNN seems to be a potential tool for image analysis in histological samples.
