Abstract:
A novel gene, Xin, from chick (cXin) and mouse (mXin) embryonic hearts, may be required for cardiac morphogenesis and looping. Both cloned cDNAs have a single open reading frame, encoding proteins with 2,562 and 1,677 amino acids for cXin and mXin, respectively. The derived amino acid sequences share 46% similarity. The overall domain structures of the predicted cXin and mXin proteins, including proline-rich regions, 16 amino acid repeats, DNA-binding domains, SH3-binding motifs and nuclear localization signals, are highly conserved. Northern blot analyses detect a single message of 8.9 and 5.8 kilo base (kb) from both cardiac and skeletal muscle of chick and mouse, respectively. In situ hybridization reveals that the cXin gene is specifically expressed in cardiac progenitor cells of chick embryos as early as stage 8, prior to heart tube formation. cXin continues to be expressed in the myocardium of developing hearts. By stage 15, cXin expression is also detected in the myotomes of developing somites. Immunofluorescence microscopy reveals that the mXin protein is colocalized with N-cadherin and connexin-43 in the intercalated discs of adult mouse hearts. Incubation of stage 6 chick embryos with cXin antisense oligonucleotides results in abnormal cardiac morphogenesis and an alteration of cardiac looping. The myocardium of the affected hearts becomes thickened and tends to form multiple invaginations into the heart cavity. This abnormal cellular process may account in part for the abnormal looping. cXin expression can be induced by bone morphogenetic protein (BMP) in explants of anterior medial mesoendoderm from stage 6 chick embryos, a tissue that is normally non-cardiogenic. This induction occurs following the BMP-mediated induction of two cardiac-restricted transcription factors, Nkx2.5 and MEF2C. Furthermore, either MEF2C or Nkx2.5 can transactivate a luciferase reporter driven by the mXin promoter in mouse fibroblasts. These results suggest that Xin may participate in a BMP-Nkx2.5-MEF2C pathway to control cardiac morphogenesis and looping.

Previous studies have described the effects of zingerone (ZO) on cisplatin (CXP)-induced injury to the kidneys, liver, and other organs but not to the cochlea. This study aimed to investigate the effects of ZO on CXP-induced ototoxicity. Eight-week-old Sprague–Dawley rats were used and divided into a control group, a CXP group, and a CXP + ZO group. Rats in the CXP group received 5 mg/kg/day CXP intraperitoneally for five days. Rats in the CXP + ZO group received 5 mg/kg/day CXP intraperitoneally for five days and 50 mg/kg/day ZO intraperitoneally for seven days. Auditory brainstem response thresholds (ABRTs) were measured before (day 0) and after (day 10) drug administration. Cochlear histology was examined using hematoxylin and eosin (H&E) staining and cochlear whole mounts. The expression levels of cytochrome P450 (CYP)1A1, CYP1B1, inducible nitric oxide synthase (iNOS), nuclear factor kappa B (NFκB), tumor necrosis factor alpha (TNFα), and interleukin 6 (IL6) were estimated using quantitative reverse transcription-polymerase chain reaction. The expression levels of heme oxygenase 1 (HO1) and caspase 3 were analyzed via Western blotting. The auditory thresholds at 4, 8, and 16 kHz were attenuated in the CXP + ZO group compared with the CXP group. The mRNA expression levels of CYP1A1, CYP1B1, iNOS, NFκB, TNFα, and IL6 were lower in the CXP + ZO group than in the CXP group. The protein expression levels of HO1 and caspase 3 were lower in the CXP + ZO group than in the CXP group. Cotreatment with ZO exerted otoprotective effects against CXP-induced cochlear injury via antioxidative and anti-inflammatory activities involving CYPs, iNOS, NFκB, and TNFα.

Previous preclinical studies have demonstrated the otoprotective effects of resveratrol (RV) at low doses. This study aimed to investigate the dose-dependent effects of RV in rats with cisplatin (CXP)-induced hearing loss. Sprague-Dawley rats (8-weeks old) were divided into six treatment groups (n = 12/group) and treated as follows: control, 0.5 mg/kg RV, 50 mg/kg RV, CXP, 0.5 mg/kg RV + CXP), and 50 mg/kg RV + CXP groups. CXP (3 mg/kg) was intraperitoneally injected for 5 days. RV (0.5 or 50 mg/kg) was intraperitoneally injected for 10 days from the first day of CXP administration. Auditory brainstem response (ABR) thresholds were measured before and within 3 days at the end of the drug administration. Cochlear tissues were harvested, and the outer hair cells were examined using cochlear whole mounts. The mRNA expression of NFκB, IL6, IL1β, and CYP1A1, and protein levels of aryl hydrocarbon receptor (AhR) and cytosolic and nuclear receptor for advanced glycation endproducts (RAGE) were evaluated. The ABR threshold increased in the 50 mg/kg RV and CXP groups at 4, 8, 16, and 32 kHz. The 0.5 mg/kg RV + CXP group demonstrated decreased hearing thresholds at 4 and 32 kHz compared to the CXP group. Cochlear whole-mount analysis revealed loss of outer hair cells in the 50 mg/kg RV and CXP groups and partial prevention of these cells in the 0.5 mg/kg RV + CXP group. The mRNA expressions of NFκB, IL6, and IL1β were increased in the 50 mg/kg RV and CXP groups compared to the control group. In contrast, these levels were decreased in the 0.5 mg/kg RV + CXP group compared to the CXP group. The mRNA expression of CYP1A1 was increased in the CXP group, while it was decreased in the 0.5 mg/kg RV + CXP group compared to the control group. The protein levels of AhR and cytosolic RAGE decreased in the 0.5 mg/kg RV group. Low-dose RV had partial otoprotective effects on CXP ototoxicity. The otoprotective effects of RV may be mediated through anti-oxidative (CYP1A1 and RAGE) and anti-inflammatory (NFκB, IL6, and IL1β) responses. High-dose RV exerted an inflammatory response and did not ameliorate CXP-induced ototoxicity.

Abstract:
To enhance Java programming educations, we have proposed a Web-based Java Programming Learning Assistant System (JPLAS) that provides a variety of programming assignments to cover different learning levels. As fundamental programming exercises for novice students, JPLAS offers the Code Fill-in-blank Problem (CFP) and the Code Fixing Problem (CXP), to learn Java grammar and basic programming skills through code reading. A CFP instance requires filling in the blank elements in the problem code generated by applying the coding rule check function and the blank element selection algorithm. A CXP instance involves correcting the error elements made by the error injection algorithm. In both problems, all answers from the students will be marked through string matching with the stored correct one. In this paper, we propose the Code Amendment Problem (CAP) as a practical problem for learning the debugging process by combining CFP and CXP in JPLAS. As a mixture of CFP and CXP instances, a CAP instance is generated by randomly selecting either blank or error for each element with a given blank probability BP. For evaluations, we apply 12 CAP instances to 21 students in Japan and Myanmar, where the results show that BP = 50% offers the highest difficulty level, and CAP is harder than CFP and CXP.

Abstract Introduction The carcinoma ex pleomorphic adenoma (CXPA) arises from a pleomorphic adenoma (PA) and is supposed to gain lipogenesis and glycogenesis during its malignant transformation. Increased lipogenesis is a characteristic of cancer cells, consisting of increased fatty acid synthesis by the enzyme fatty acid synthase (FASN) and by the accumulation of cytoplasmic lipid droplets, which express adipophilin. The increase in glycolytic metabolism has been associated with the activity of glycolytic enzymes and glucose transporters (GLUTs), whose expression is induced by HIF-1 (inducing factor of hypoxia 1). Objectives To analyze the gene expression of FASN, adipophilin, HIF-1α, and GLUT-1 in PA and CXPA samples. Methodology Gene expression analysis of the FASN, adipophilin, HIF1-α, and GLUT-1 genes was performed by the real-time PCR (qPCR) method. Fourteen cases of PA and 14 cases of CXAP were evaluated. Results FASN, adipophilin, HIF-1α, and GLUT-1 were more expressed in CXAP than in AP, although there was no significant difference between levels of adipophilin expression in CXPA and PA. Conclusion Increased expression of FASN, HIF1-α, and GLUT-1 in CXAP may be associated with malignant transformation of PA, while additional studies are needed to understand the role of adipophilin in PA carcinogenesis. Moreover, this is the first study to investigate the role of these genes in the malignant transformation of PA.

Introduction Cancer cachexia affects many advanced non-small-cell lung cancer (NSCLC) patients. Cachexia index (CXI) was developed to assess the degree of cachexia in these patients. Methods Patients with metastatic NSCLC diagnosed between January 1, 2000, and June 30, 2011, at our institution were retrospectively studied. Abdominal computed tomography scans done within 1 month of diagnosis were reviewed to estimate skeletal muscle area (SMA) and skeletal muscle index (SMI) at the L3 level. CXI was developed as follows: [Formula: see text] where SMI is the skeletal muscle index, Alb is the serum albumin, and NLR is the NLR neutrophil-to-lymphocyte ratio. Progression-free survival (PFS) and overall survival (OS) were estimated using the Kaplan–Meier method. Survival among various factors was calculated using the log-rank test. Multivariate Cox regression was used to perform survival analysis in order to estimate the effects of various factors. Results Patients were divided into two groups around the median into stage I cachexia (CXI ≥35, n = 56) and stage II cachexia (CXI <35, n = 56). Groups did not differ in age, gender, ethnicity, or histology of cancer. Patients with stage II cachexia had significantly worse PFS (2.45 vs 5.43 months, P < 0.0001) and OS (3.45 vs 8.8 months, P = 0.0001) than those with stage I cachexia. On multivariate analysis adjusting for gender, race, and histology, patients with stage II cachexia were found to have worse PFS (hazard ratio [HR] 1.94, 95% confidence interval [CI] 1.27–2.95) and OS (HR 1.53, 95% CI 1.0009–2.34). Conclusion The CXI is a novel index for estimating cachexia that also correlates with prognosis in both men and women with advanced NSCLC.

Nonhomologous end-joining (NHEJ) repairs DNA double-strand breaks (DSBs) during V(D)J recombination in developing lymphocytes and during immunoglobulin (Ig) heavy chain (IgH) class switch recombination (CSR) in peripheral B lymphocytes. We now show that CD21-cre–mediated deletion of the Xrcc4 NHEJ gene in p53-deficient peripheral B cells leads to recurrent surface Ig-negative B lymphomas (“CXP lymphomas”). Remarkably, CXP lymphomas arise from peripheral B cells that had attempted both receptor editing (secondary V[D]J recombination of Igκ and Igλ light chain genes) and IgH CSR subsequent to Xrcc4 deletion. Correspondingly, CXP tumors frequently harbored a CSR-based reciprocal chromosomal translocation that fused IgH to c-myc, as well as large chromosomal deletions or translocations involving Igκ or Igλ, with the latter fusing Igλ to oncogenes or to IgH. Our findings reveal peripheral B cells that have undergone both editing and CSR and show them to be common progenitors of CXP tumors. Our studies also reveal developmental stage-specific mechanisms of c-myc activation via IgH locus translocations. Thus, Xrcc4/p53-deficient pro–B lymphomas routinely activate c-myc by gene amplification, whereas Xrcc4/p53-deficient peripheral B cell lymphomas routinely ectopically activate a single c-myc copy.

Abstract:
A large number of patients with coronavirus disease 2019 (COVID-19) present at hospitals. There are a limited number of isolation rooms open, and patients must often wait a long time to get a reverse transcription-polymerase chain reaction (RT-PCR) test done. This necessitates the introduction of effective triage plans. A patient with suspicions is referred to an emergency room (ED) depending on their medical record for a simple physical assessment, blood test findings, and chest imaging.A retrospective study design was conduct at Prince Sultan Medical Military City (PSMMC). Ethical approval was obtained from the institutional board to wave the consent forms since it is a retrospective study. Only the primary investigator has had the data access to the patients’ medical records. The collected patient records were under specific categories, including symptoms score starts from 5 and above, RT-PCR test result done after CXRP imaging, the patient admitted to the emergency department (ED). Excluding all CXRP done after RT-PCR TEST, positive Covid 19 admitted to the intensive care unit (ICU), pediatric patients, and patients with score symptoms were less than five. Two experienced radiologists reviewed the images blindly, and the inter-observer reliability of observations noted by the radiologists was calculated. As for the relationship between the x-ray reading and the RT-PCR test result, our results showed a high correlation between the variables (chi-square χ² = 12.44, with df =1, and p<0.001). The sensitivity of x-ray diagnosing covid19 was 65.52 %, while the specificity was 54.51 %, and the accuracy of radiologists reading was 58.17 %. Furthermore, the positive predictive value (PPV) was 41.76 %, and the negative predictive value (NPV) was 76.05%. Finally, the false positive rate (type-i error (alpha) was 45.49%, and the false-negative rate (type-ii error (beta) was 34.48% Our research findings show that CXRP imaging can detect COVID-19 infection in symptomatic patients and can be a valuable addition to RT-PCR testing. In an inpatient ED environment where availability of test kits, laboratory equipment, and laboratory personnel is compromised and risks delaying patient treatment and hospital workflow, serial CXRP could theoretically be used as an adjunct diagnostic function and monitoring in patients suspected of having COVID-19.

II: Nach einer Definition von Inter- bzw. Palintextualität (Stocker) werden dann die Beziehungen des Psalm zu anderen Texten innerhalb der Hebräischen Bibel untersucht. Ergebnis ist u. a., dass der Psalm zum Genre der präsentischen JhwhKönigs-Psalmen zu rechnen ist und dass es thematisch um den Antritt der Königsherrschaft Jhwhs sowohl über Israel als auch über die ganze Erde geht (Partikularismus → Universalismus).

Abstract Seedlings of 60 loblolly pine (Pinus taeda L.) polycross families representing four populations, Atlantic Coastal (CxC), Piedmont (PxP), Coastal × Piedmont (CxP), and Piedmont × Coastal (PxC) were evaluated for first-year seedling height growth in an outdoor site in Raleigh, North Carolina. The CxP hybrids grew as well as the pure Coastal loblolly source, and the PxC hybrids were not significantly taller than the pure Piedmont population. The hybrid populations were intermediate between the parental populations and did not exhibit heterosis for height. However, the hybrids were 4–10% taller than the PxP population, which is the population currently used for forestation in the Piedmont region. There was an advantage for early seedling height growth in having the female parent from the Coastal plain, which appeared to be related to length of the growing season as measured by bud set time. There were strong family differences for seedling height within populations, indicating the potential for selecting the best families within the best populations. The implication is that the hybrid populations of CxP and PxC with improved growth may be better planting stock for the Piedmont than the pure Piedmont populations on some sites. Before implementing such a plan, cold-hardiness and long-term growth and adaptability should be evaluated. South. J. Appl. For. 28(2):83–90.

Abstract:
(1) Background: The preparation and characterization of novel fully injectable enzymatically hardened tetracalcium phosphate/monetite cements (CXI cements) using phytic acid/phytase (PHYT/F3P) hardening liquid with a small addition of polyacrylic acid/carboxymethyl cellulose anionic polyelectrolyte (PAA/CMC) and enhanced bioactivity. (2) Methods: Composite cements were prepared by mixing of calcium phosphate powder mixture with hardening liquid containing anionic polyelectrolyte. Phase and microstructural analysis, compressive strength, release of ions and in vitro testing were used for the evaluation of cement properties. (3) Results: The simple possibility to control the setting time of self-setting CXI cements was shown (7–28 min) by the change in P/L ratio or PHYT/F3P reaction time. The wet compressive strength of cements (up to 15 MPa) was close to cancellous bone. The increase in PAA content to 1 wt% caused refinement and change in the morphology of hydroxyapatite particles. Cement pastes had a high resistance to wash-out in a short time after cement mixing. The noncytotoxic character of CX cement extracts was verified. Moreover, PHYT supported the formation of Ca deposits, and the additional synergistic effect of PAA and CMC on enhanced ALP activity was found, along with the strong up-regulation of osteogenic gene expressions for osteopontin, osteocalcin and IGF1 growth factor evaluated by the RT-qPCR analysis in osteogenic αMEM 50% CXI extracts. (4) Conclusions: The fully injectable composite calcium phosphate bicements with anionic polyelectrolyte addition showed good mechanical and physico-chemical properties and enhanced osteogenic bioactivity which is a promising assumption for their application in bone defect regeneration.

The article examines the VII and X ‘Odessan Sonnets’ by Adam Miсkiewicz, which are translations of the СХІІ and LXI sonnets by Petrarch from the ‘Canzoniere’. A comparative analysis of Polish and Italian texts is carried out. The problem of the loss of VII and X ‘Odessan Sonnets’ by Adam Mickiewicz when translating this cycle into other languages is noted. The study of the translation strategy of Adam Mickiewicz suggests that these sonnets can be considered as original author’s works having intertextual connections with CXII and LXI sonnets of Francesca Petrarch. This circumstance opens up the possibility for translations VII and X ‘Odessan Sonnets’ by Adam Mickiewicz into foreign languages.

The geomagnetic field in the Brazilian sector is influenced by the South American Magnetic Anomaly (SAMA) that causes a decrease in the magnitude of the local geomagnetic field when compared to other regions in the world. Thus, the magnetometer network and data set of space weather over Brazil led by Embrace are important tools for promoting the understanding of geomagnetic fields over Brazil. In this sense, in this work we used the H component of geomagnetic fields obtained at different sites in South America in order to compare results from the phase coherence obtained from wavelet transform (WT). Results from comparison between Cachoeira Paulista (CXP) and Eusébio (EUS), and Cachoeira Paulista and São Luis (SLZ), indicated that there exist some phenomena that occur simultaneously in both locations, putting them in the same phase coherence. However, there are other phenomena putting both locations in a strong phase difference as observed between CXP and Rio Grande, Argentina (RGA). This study was done for a specific moderate geomagnetic storm that occurred in March 2003. The results are explained in terms of nonlinear interaction between physical phenomena acting in distinct geographic locations and at different times and scales. Keywords. The newly synthesized heterocyles were characterized on the basis of their chemical properties and spectroscopic data. In his account of the contents of the second volume of London, British Library, Cotton Otho C. i, Kenneth Sisam drew attention to an unpublished sermon on the sins of the tongue to which he gave the title Evil Tongues. The sermon is written in the same hand as the two texts which precede it in the manuscript – an Old English version of St Jerome's Vita Malchi monachi captivi (henceforth Malchus) and a translation of a letter by Boniface (Wynfrith) on an anonymous priest's vision of the next world (henceforth Wynfrith's Letter) Sisam was unable to identify a source for the Evil Tongues sermon, but close parallels for most of the Old English text are found in anther work by Jerome, the exposition of Ps. CXIX in his Tractatus in Psalmos.

In this article, 4 methods for calculating the induced drag coefficient Cxi of a simple airfoil with an end plate at different relative heights were considered. Four methods of induced drag coefficient determination were considered for different relative flight’s heights. Calculations were performed according to the Phillips, Wieselsberge, Panchenkov-Surzhik, Mantle methods for the TsAGI-876 wing profile. The calculated values of induced drag coefficients were compared with the experimental wind tunnel’s data at the Central Aerohydrodynamic Institute.

Magda B. Arnold's phenomenological and cognitive theory on emotions is subjected to a critical review. The theory is well-known and has inspired other theorists in the field of emotion research. However, it has generated hardly any research. At first sight this is astonishing because of its obvious qualities. Arnold does justice to the subjective nature of emotions, integrates psychological with physiological aspects, and offers a relatively simple and parsimonious theoretical model. Salient points are the conceptualization of emotions as action-tendencies, the role of appraisal in generating emotions and in emotional action, the classification of basic emotions, the explanation of increase of strength in emotions, and their organizing function or disturbing effects. A closer look into the theory, however, shows serious flaws in logical consistency and difficulties for operationalization. The authors conclude that the lack of research is partly due to the theory.

X-ray free-electron lasers (FELs) are being recognized as a powerful tool in an ever-increasing number of research fields, but are very limited as to the number of experiments that they can support. This work shows that more beamtime could be made available by using `parasitic' geometries, where a secondary experiment uses the X-ray beam that the primary experiment does not utilize. The first successful ptychography experiment, a scanning coherent diffractive imaging technique, in a parasitic geometry at an X-ray FEL is demonstrated. Utilizing the CXI hutch at the Linac Coherent Light Source (LCLS), it is shown that the obtained data are of high quality and that characterizing the beam using ptychography can be much faster than traditional imprinting methods.

Whereas the activity of enzymes produced by avirulent isolate of pathogens did not decrease and these enzyme activities increased with the increase in the age of culture. These pathogens also produced nonspecific toxic metabolites in culture filtrate which reduced seed germination, root length, shoot length and vigour index of the seedlings of chilli, rice, mungbean, maize, cotton, groundnut, okra, egg plant, cucumber and tomato. The toxins of the pathogens reduced seed germination and caused mortality of chilli seedlings in pot culture. The toxins also produced phytotoxic symptoms in the treated ripe and green chilli fruits and leaves.

Glass formation from the melt of organic monomers was studied for a variety of different organic molecular structures with Tg near ambient temperature. Crystallization is suppressed by one or more of the molecular properties, hydrogen bonding, interlocking, dipolar, and hydrogen bonding, combined with hindered rotational isomerism. Examples of materials in each category are presented for illustration. The viscosity of undercooled liquids was characterized by the Vogel-Tammon-Fulcher (VTF) equation, η = A cxp[DT0/(T - T0)], where A, D, and T0 are experimentally determined parameters. Our experimental D values are discussed in relation to the molecular structure and glass formation mechanism. The insight provided by our interpretation is intended to assist in the design of new molecular structures with controlled viscosity-temperature characteristics, as well as glass-forming ability by cooling from melts.
