Abstract:
La cutine est un polymère d'acides gras oxydés et de glycérol propre aux plantes. Elle forme la matrice structurale de la cuticule qui recouvre l'épiderme des parties aériennes et joue un rôle vital pour les plantes en empêchant la dessiccation. La biosynthèse du polymère de cutine a été étudiée chez la plante modèle Arabidopsis thaliana. Des mutants perte-de-fonction pour la monooxygénase de type cytochrome P450 CYP77A4 ont été isolés. L'analyse de lignées transgéniques exprimant le gène rapporteur GUS sous le contrôle du promoteur de CYP77A4 a montré que le gène était exprimé essentiellement dans les organes floraux et les graines. L'analyse de la cutine dans divers organes a permis de démontrer que le gène était essentiel pour la synthèse d'un acide gras trihydroxylé en C18 présent dans les polyesters des embryons. Une méthode permettant la séparation de l'embryon et des téguments des graines en quantité suffisante pour analyser la cutine a été mise au point. Le profil de monomères des embryons mutants a montré que CYP77A4 est une époxygénase de la voie de biosynthèse des monomères de cutine en C18. L'étude de la physiologie des mutants a par ailleurs permis de démontrer que les acides gras trihydroxylés de la cuticule de l'embryon jouent un rôle important dans la germination de la graine en conditions de stress salin. Dans une deuxième étude, des mutants perte-de-fonction et des suexpresseurs pour le gène d'Arabidopsis BODYGUARD (BDG) codant une protéine de la superfamille des hydrolases à repliement α/β ont été caractérisés. L'analyse des polyesters dans ces lignées a permis de montrer que cette protéine jouait en fait un rôle dans la biosynthèse de la cutine
Cutin is a polymer of oxidized fatty acids and glycerol specific to plants. It forms the structural matrix of the waxy cuticle covering the epidermis of the aerial parts and plays a vital role in plants by preventing desiccation. Biosynthesis of cutin polymer was studied in the model plant Arabidopsis thaliana. Mutant loss-of-function monooxygenase type cytochrome P450 CYP77A4 were isolated. The analysis of transgenic lines expressing the GUS reporter gene under the control of the promoter of CYP77A4 showed that the gene was expressed mainly in floral organs and seeds. The analysis of various organs in the cutin demonstrated that the gene was essential for the synthesis of a C18 trihydroxy polyesters present in seed polyesters (9,10,18-trihydroxyoctadecenoic acid). A method for the separation of the embryo and seed coat allowing to analyze embryo polyesters was developed. The trihydroxy C18 fatty acid was found to be the major cutin embryo monomer. Profile of cutin monomers in mutant embryos showed that CYP77A4 is an epoxygenase in the biosynthetic pathway of C18 cutin monomers. The study of the physiology of the mutants also showed that the trihydroxy- fatty acids of the embryo cuticle play an important role in the germination of the seed under conditions of salt stress. In a second study, mutant loss-of-function and overexpressors for the Arabidopsis gene BODYGUARD encoding a protein of the α / β hydrolase fold superfamily have been characterized. Analysis of polyesters in these lines showed that this protein, whose role in the formation of the cuticle was not understood, plays in fact a role in cutin biosynthesis

Abstract:
A study on cuticle-degrading enzymes (CDE) of three hyphomycete entomopathogens has produced information on enzyme types, levels, characteristics, mode of action, regulation, sequence of production, cellular localisation and production during host penetration. This is the first critical work on CDE of any entomopathogen. Several pathogenic isolates of Metarhizium anisopliae, Beauveria bassiana and Verticillium lecanii when grown in buffered liquid cultures containing comminuted locust cuticle as sole carbon source (good growth occurred on most monomeric and polymeric cuticular constituents), produced a variety of extracellular and bound enzymes corresponding to the major components of insect cuticle e.g. 3 endo-proteases, aminopeptidase, carboxypeptidase A, lipase, esterase, chitinase and N-acetylglucosaminidase. Considerable variations occurred in levels of production between spp. and even within a sp., but endo-proteases were exceptional in being produced in large amounts by all the isolates. CDE were produced rapidly and sequentially in culture. The first activities to appear (< 24 h) were those of the proteolytic complex, chitinases were always produced substantially later. Properties of CDE were investigated in terms of pH and tem- erature optima, substrate specificity, molecular weight, iso-electric point, mechanism of substrate degradation and the effect of specific inhibitors. Studies with culture filtrates and purified CDE revealed that substrates in intact cuticles are amenable to degradation but the prior action of protease is necessary for significant degradation of the chitin. Staining of chitin by a fluorescent lectin (FITC-WGA) and calcofluor only in cuticles from which protein has been removed (by protease or KOH) also suggests initial masking of chitin. This and determination of amino acid composition of peptides solubilised by endo-protease revealed the potential of CDE in studying the physicochemical structure of insect cuticles. The apparently localised action of CDE during host penetration may result from molecular sieving, binding to fungal walls or binding to cuticle. The first possibility is lessened by the small size of the endo-enzymes (<34 K daltons) which could allow diffusion via the various canals which traverse cuticle. However, cuticle effectively binds (ionically) CDE, and also activities of several CDE remain partly bound in various ways to hyphae and conidia (by ionic binding to walls, by disulphide bonds, and on or within membrane structures). The involvement of proteolytic enzymes in infection was suggested by their presence in conidia, penetration structures, and infected cuticle (detected histochemically and following extraction from cuticles). Also the constitutive production of endo- and exo-proteases lends weight to their possible significance in parasitism as synthesis will be subject only to catabolite repression. Chitinase is induced by N-acetylglucosamine and was not detected in infected cuticle. Possible mechanisms and significance of enzymic degradation of cuticle during infection are discussed, particularly in comparison to host penetration by phytopathogenic fungi.

Abstract:
This essay examines YouTube comments to videos with male and female streaming players of the online game League of Legends. The research carries out a critical discourse analysis with the aim to find gender relevant language in order to analyze ideologies about gender stereotypes among viewers of streamers. The results showed that comments concerning players’ bodies and appearance were more common in comments on female than male players. There was also a higher expectation for female players to match sexual stereotypes close to the imagery of avatars than for male players to do so. Female players received more comments with sexual implications. Both male and female players received offers of sexual acts, but violent acts were only aimed against women. Written comments defending female players showed that there are ideological power struggles between viewers in this specific genre and that some viewers defend the female players’ place in the scene of streamed gaming.
Denna studie undersöker kommentarer på YouTube-videos med spelare av ett online-spel vid namn League of Legends. Undersökningen för en kritisk diskursanalys med syfte att hitta diskurs relevant för kön för att utreda vilka ideologier om kön och sexuella stereotyper som återfinns bland konsumenter av sådana videos. Resultatet visar att kommentarer som nämner spelarens kropp och utseende är betydligt vanligare för kvinnliga än manliga spelare. Resultatet visade också att det fanns en högre förväntan att kvinnliga spelare skulle matcha sexuella stereotyper som liknar bilden av kvinnliga avatarer än att manliga spelare skulle göra det. Kvinnliga spelare fick fler kommentarer av sexuell karaktär. Både manliga och kvinnliga spelare fick erbjudanden om sexuella handlingar, men våldsamma sexuella handlingar var bara riktade mot kvinnor. Svar på kommentarer som försvarar kvinnliga spelare visar på en kraftmätning mellan de som tittar på manliga och kvinnliga spelare inom denna specifika genre och att kvinnliga spelares plats inom denna sfär försvaras av vissa tittare.

Abstract:
In insects, the cuticle provides protection against physical injury and water loss, rigidness for muscle attachment and mechanical support, and flexibility in inter-segmental and joint areas for mobility. As most insects undergo metamorphosis, they need to shred off old cuticle and synthesize new cuticle to fit the body shape and size throughout their life cycles. The newly formed cuticle, mainly composed of cuticular proteins, chitin, and sclerotizing reagents, needs to be hardened through the crosslinks between cuticular proteins and sclerotizing reagents. This dissertation concerns the biochemical activities of several pyridoxal 5-phosphate (PLP)-dependent decarboxylases with most of them involved in insect cuticle hardening. Herein, we first present a detailed overview of topics in reactions and enzymes involved in insect cuticle hardening. Aspartate 1-decarboxylase (ADC) is at the center of this dissertation. beta-alanine, the product of ADC-catalyzed reaction from aspartate, is the component of an important sclerotizing reagent, N-beta-alanyldopamine; the levels of beta-alanine in insects regulate the concentrations of dopamine, therefore affecting insect sclerotization and tanning (collectively referred as cuticle hardening in this dissertation).

Biochemical characterization of insect ADC has revealed that this enzyme has typical mammalian cysteine sulfinic acid decarboxylase (CSADC) activity, able to generate hypotaurine and taurine. The result throws lights on research in the physiological roles of insect ADC and the pathway of insect taurine biosynthesis. Cysteine was found to be  an inactivator of several PLP-dependent decarboxylases, such as ADC, glutamate decarboxylase (GAD) and CSADC. This study helps to understand symptoms associated with the abnormal cysteine concentrations in several neurodegenerative diseases. A mammalian enzyme, glutamate decarboxylase like-1 (GADL1), has been shown to have the same substrate usage as insect ADC does, potentially contributing to the biosynthesis of taurine and/or beta-alanine in mammalian species. Finally, the metabolic engineering work of L-3, 4-dihydroxyphenylalanine decarboxylase (DDC) and 3, 4-dihydroxylphenylacetaldehyde (DHPAA) synthase has revealed that the reactions of these enzymes could be determined by a few conserved residues at their active site. As both enzymes have been implicated in the biosynthesis of sclerotizing reagents, it is of great scientific and practical importance to understand the similarity and difference in their reaction mechanisms. The results of this dissertation provide valuable biochemical information of ADC, DDC, DHPAA synthase, and GADL1, all of which are PLP-dependent decarboxylases. ADC, DDC, DHPAA synthase are important enzymes in insect cuticle hardening by contributing to the biosynthesis of sclerotizing reagents. Knowledge towards understanding of these enzymes will promote the comprehension of insect cuticle hardening and help scientists to search for ideal insecticide targets. The characterization of GADL1 lays groundwork for future research of its potential role in taurine and beta-alanine metabolism.

Abstract:
Several types of insect cuticle contain enzymes catalyzing the formation ofof adducts between N-acetyldopamine (NADA) and N-acetylhistidine (NAH). Two such adducts, NAH-NADA-I and NAH NADA-II, have been isolated and their structures determined. In one of the adducts the link connecting the two residues occurs between the I-position (ß-position) in the NADA side chain and the 1-N atom (τ-N) in the imidazole ring of histidine. Diphenoloxidase activity alone is not sufficient for formation of this adduct, whereas extracts containing both diphenoloxidase and o-quinone-p-quinone methide isomerase activities catalyze the coupling reaction. The adduct consists of a mixture of two diastereomers and they are presumably formed by spontaneous reaction between enzymatically produced NADA-p-quinone methide and N-acetylhistidine. The other adduct has been identified as a ring addition product of N-acetylhistidine and NADA. In contrast to the former adduct it can be formed by incubation of the two substrates with mushroom tyrosinase alone. An adduct between N-acetylhistidine and the benzodioxan-type NADA-dimer is produced in vitro, when the N-acetylhistidine-NADA adduct is incubated with NADA and locust cuticle containing a 1,2-dehydro-NADA generating enzyme system. Trimeric NADA-polymerization products of the substituted benzodioxan-type have been obtained from in vivo sclerotized locust cuticle, confirming the ability of cuticle to produce NADA-oligomers. The results indicate that some insect cuticles contain enzymes promoting linkage of oxidized NADA to histidine residues. It is suggested that histidine residues in the cuticular proteins can serve as acceptors for oxidized NADA and that further addition of NADA-residues to the phenolic groups of bound NADA can occur, resulting in formation of protein-linked NADA-oligomers. The coupling reactions identified may be an important step in natural cuticular sclerotization.

Abstract:
The conserved metabolites, oleic acid (18:1), a major monounsaturated fatty acid (FA), and glycerol-3-phosphate (G3P) are obligatory precursors of glycerolipid biosynthesis in plants. In Arabidopsis, the SSI2-encoded SACPD is the major isoform that contributes to 18:1 biosynthesis. Signaling induced upon reduction in oleic acid (18:1) levels not only upregulates salicylic acid (SA)-mediated responses but also inhibits jasmonic acid (JA)- inducible defenses. I examined the transcription profile of ssi2 plants and identified two transcription factors, WRKY50 and WRKY51. Although the ssi2 wrky50 and ssi2 wrky51 plants were constitutively upregulated in SA-derived signaling, they were restored in JAdependent defense signaling. Not only did these plants show JA-inducible PDF1.2 expression, but they were also restored for basal resistance to the necrotrophic pathogen, Botrytis cinerea. Overall, my results show that the WRKY50 and WRKY51 proteins mediate both SA- and low 18:1-dependent repression of JA signaling in Arabidopsis plants. My studies also show that cellular G3P levels are important for plant defense to necrotrophic pathogens. I showed that G3P levels are induced in Arabidopsis in response to the necrotrophic fungal pathogen B. cinerea. G3P-dependant induction of basal defense is not via the activities of other defense-related hormones such as SA, JA or the phytoalexin camalexin. Arabidopsis mutants unable to accumulate G3P (gly1, gli1) showed enhanced susceptibility to B. cinerea. Previous studies in our lab identified acyl-carrier protein 4 (ACP4), a component of FA and lipid biosynthesis, as an important regulator of plant systemic immunity. ACP4 mutant plants were defective in systemic acquired resistance (SAR) because they contained a defective cuticle. I further investigated the role of the plant cuticle in SAR by studying the involvement of long-chain acyl-CoA synthetases (LACS), a gene family involved in long-chain FA and cuticle biosynthesis, in SAR. In all, eight lacs mutants (lacs1, lacs2, lacs3, lacs4, lacs6, lacs7, lacs8, lacs9) were isolated and characterized. Six mutants were compromised in SAR. Together, my studies show that the various LACS isoforms contribute differentially to both cuticle formation and systemic immunity in Arabidopsis.

Abstract:
Omniphobic surfaces found in nature have great potential for enabling novel and emerging products and technologies to facilitate the daily life of human societies. One example is the water and even oilrepellent cuticle of springtails (Collembola). The wingless arthropods evolved a highly textured, hierarchically arranged surface pattern that affords mechanical robustness and wetting resistance even at elevated hydrostatic pressures. Springtail cuticle-derived surfaces therefore promise to overcome limitations of lotus-inspired surfaces (low durability, insufficient repellence of low surface tension liquids). In this review, we report on the liquid-repellent natural surfaces of arthropods living in aqueous or temporarily flooded habitats including water-walking insects or water spiders. In particular, we focus on springtails presenting an overview on the cuticular morphology and chemistry and their biological relevance. Based on the obtained liquid repellence of a variety of liquids with remarkable efficiency, the review provides general design criteria for robust omniphobic surfaces. In particular, the resistance against complete wetting and the mechanical stability strongly both depend on the topographical features of the nano- and micropatterned surface. The current understanding of the underlying principles and approaches to their technological implementation are summarized and discussed.

L’elaborato inizia con un excursus storico sulla figura di Antonio Morassi, celebre personaggio di cultura del XX secolo. In particolare l’analisi si focalizzerà sui suoi insegnamenti e su quello che è stato il suo percorso lavorativo che lo ha poi portato a fare una scelta importante: dedicarsi esclusivamente alla professione di critico e storico dell’arte. Durante il periodo preso in considerazione Morassi aveva rilasciato un’intervista inedita su quello che è secondo lui un falso nell’arte. Questa ha fornito un punto di partenza per cercare di ricostruire e capire, tramite definizioni ed esempi, che cosa si intende per falso. Nell’ultima parte della tesi si è cercato di ricreare un catalogo di fotografie che Morassi aveva analizzato. Si vuole cercare di dare una lettura storico-critica di queste opere che sono custodite nella Fototeca Antonio Morassi, presso il Dipartimento di Filosofia e Beni Culturali di Ca' Foscari. L’intento è quello di ripercorrere le analisi che Antonio Morassi aveva iniziato con lo scopo verificarne la loro autenticità.

The chicken eggshell possesses physical and chemical barriers to protect the embryo from pathogens. The avian eggshell cuticle is the outmost layer of the eggshell whose protein constituents remain largely unknown. Since eggs with incomplete or absent cuticle are more susceptible to bacterial contamination, we hypothesize that cuticle protein components play an important role in microbial resistance. In our study, at least 47 proteins were identified by LC/MS/MS in the non-calcified cuticle layer. Similar to Kunitz-like protease inhibitor (also annotated as ovocalyxin-25, OCX-25) and ovocalyxin-32 (OCX-32) were two of most abundant proteins of the cuticle proteins. Some proteins that have antimicrobial activity were also detected in the proteomic results, such as lysozyme C, ovotransferrin, ovocalyxin-32, cystatin, ovoinhibitor. This study represents the first comprehensive report of the cuticle proteome. Since the sequence similarity of the kunitz motif in OCX-25 is similar to that of BPTI, it is predicted that it will have the same trypsin inhibitory and antimicrobial activity against Gram-positive and/or Gram-negative bacteria. In order to test the antimicrobial property and trypsin inhibitor activity of OCX-25, cuticle proteins were extracted by 1N HCl. Antimicrobial activity was monitored using the Bioscreen C instrument; and antimicrobial activity was identified primarily against Staphylococcus aureus. Trypsin inhibitor activity was studied by using a specific trypsin assay, and the assay indicated that the cuticle proteins could inhibit the reaction of trypsin and substrate. Therefore, the current research has provided some insight into the antimicrobial and enzymatic aspects of the cuticle proteins, and its function for egg protection. Eggshell membranes are another important component of the chicken eggshell.Due to its insoluble and stable properties, there are still many questions regarding formation and constituents of the eggshell membranes. The purpose of our study was to identify eggshell membrane proteins, particularly these responsible for its structural features, by examining the transcriptome of the white isthmus during its formation. Bioinformatics tools were applied to analyze the differentially expressed genes as well as their encoded proteins. Some interesting proteins were encoded by the over-expressed genes in the white isthmus during the formation of eggshell membranes, such as Collagen X, and similar to spore coat protein SP75. These proteins may have potential applications. Our study provides a detailed description of the chicken white isthmus transcriptome during formation of the eggshell membranes; it could lead to develop the strategies to improve food safety of the table egg.

Abstract:
La cuticule, une matrice lipidique extracellulaire constituée de cires et d'un squelette de cutine, est la barrière de défense la plus externe des plantes face à leur environnement. Elle intervient dans de nombreuses propriétés agronomiques comme la conservation post récolte, les propriétés mécaniques ou bien l'aspect du fruit, dont la brillance. Afin d'isoler des mutants de cuticule, le criblage d'une collection de mutants EMS de tomate a été entrepris, en se basant sur la brillance des fruits, conduisant à la sélection de 24 mutants. Chez ceux-ci, des analyses biochimiques ont montré de fortes variations de charge et de composition de la cuticule, notamment chez les mutants de cutine. La caractérisation de 4 mutants remarquables a été entreprise afin d'identifier les mutations responsables des phénotypes de brillance. Le mutant le plus affecté, présentant une charge en cutine réduite de 85% par rapport au type sauvage, a révélé une mutation du gène SlGDSL2 codant pour une acylhydrolase à motif GDSL, responsable de la polymérisation de la cutine. Afin d'étudier la formation et la régulation de la cutine, la suite du travail a consisté à obtenir et à caractériser des simples et des doubles mutants affectés dans la synthèse des monomères de cutine, le transport apoplastique et la polymérisation de la cutine.

Abstract The surface of multicellular organisms is covered with epithelial cells that provide a barrier to the external environment. As part of this barrier function, most epithelia produce apical extracellular matrices (aECMs). The generation of such chemical and physical barriers requires specialized deposition of macromolecules and is likely to involve a spatial and temporal coordination of biochemical activities at the apical surface. A challenging task is thus to characterize key proteins that underlie apical cell surface organization and correct aECM assembly. The Drosophila trachea provides an excellent system to study aECM formation, as they produce an ordered aECM, called the cuticle. The tracheal cuticle is unique by its presence of cuticular ridges, called taenidial folds, which prevent collapse of tracheal tubes while allowing them to expand and contract along their length. A gene called filzig encodes a transmembrane serine protease and is required for taenidial organization. The aim of this research was to further understand Filzig function through characterization of filzig mutants and Filzig protein expression. The results showed that Filzig is expressed in cuticle-producing epithelia as cuticle deposition begins. Moreover, Flz localized to the apical epithelial surface, as well as to the aECM. The apical Flz localization does not reflect the pattern of cuticle ridges, indicating that Flz-localization is not a determinant for taneidial patterning. Instead, Flz might act on extracellular targets that localize to the future taneidial folds. Alternatively, Filzig is involved in a cascade of self-organizing activity of cuticular components to form the regular taenidial folds.
