Since 30 years ago, the growing use of Lanthanides in new technologies has contributed to important releases of these metals into aquatic ecosystems. In a global sustainable development policy aimed at preserving the quality of ecosystems, the impact of Lanthanides on aquatic organisms has naturally been questioned. However, studies on the aquatic ecotoxicology of Lanthanides are incomplete, and no consensus is established yet. In this context, we studied the cellular toxicity of Lanthanides individually and in mixtures. To determine these toxic effects, cell viability was measured on Danio rerio fibroblast-like cells (ZF4; ATCC®, CRL-2050™), Danio rerio hepatic cells (ZFL; ATCC®, CRL-2643™), Oncorhynchus mykiss epithelial cells (RTgill-W1; ATCC®, CRL-2523™), and primary culture of Corbicula fluminea digestive glands exposed to Lanthanides. Direct toxicity of Lanthanides has been observed on all cellular models. Concerning the toxicity of Lanthanides in mixtures, synergistic effects have been underlined on the three fish cell lines. In this research, we focused on the mechanisms of the detoxification of Lanthanides in the case of ZF4 cells from Danio rerio. The effects of Lanthanides were assessed in the presence of specific inhibitors of glutathione-S-transferases (ethacrynic acid) and MRP-like (MK571 and probenecid), by cell viability measurements. We decided to study these actors of the cellular detoxification due to their respective roles in phases II and III of the cellular detoxification of metals in fishes and bivalves. Regarding the results, MRP-like proteins are effectively involved in the detoxification of Lanthanides in ZF4 cells. Overall, our results highlighted the relevance of the toxic effects of Lanthanides at the cellular level for the risk assessment of these metals

Our thesis examines the professional culture of primary school teachers by looking the coulisses at primary school. Behind the scenes represent all the places outside the classroom and they are studied in a methodological approach of ethnographic type. Our research is based on fieldwork composed of participant observations in two schools with contrasting social backgrounds and 44 semi-directive interviews with primary school teachers. We will dealing with giving outlines and meaning to this professional culture that unfolds in the margins of institutional curricula. Our investigation thus opens up a knowledge of the profession from a less studied point of view because it is more distant from the ordinary image of the school teacher and the elementary school. The interest of the concept of professional culture lies in its ability to encompass and bring unity to a diverse norms and practices within the profession. More broadly, professional culture is a set of positioning and solutions shared by members of the same profession. It is also a discussion space marked by tensions, cleavages around themes that may include an ethical dimension. It is also a vector of transmission and professional influence, therefore of horizontal socialization and at the same time of local socialization. Finally, it is a space in which professional autonomy is deployed through the interpretation play, relaxation or reinforcement of institutional and hierarchical injunctions. This informal professional culture is a set of norms, practices and values. It holds its unity in its ability to socialize and train teachers on the job. By so doing, it provides ways to answer to questions or professional difficulties. Informal professional culture is a particular form of socialization that appears as a kind of "informal continuing training" that complements institutional training and, above all, answers concretely the daily professional questions that it does not take into account. The dynamics of this professional culture also strengthens the professional group in its specificity of identity. This informal professional culture has the particularity of being created in an interaction system that counts colleagues and users (pupils and their parents). Interactions with co-workers on the one hand and parents of pupils and their children on the other, thus constitute for primary school teachers two regulation figures in the sense that they constitute a sphere of socialization which contributes to the professionalization of primary school teachers. The context in which the interactions unfold is decisive and modifies the norms and practices. Our study will be developed the contours of this informal professional culture through the norms that define the good teacher, the relations between colleagues and those with the users of the primary school

In order to investigate the cellular compartmentation of the central nervous system, we first defined the metabolic properties of neurons and astrocytes in homogenous primary culture. The metabolic fate of [1-13C]glucose in cerebellar neurons and astrocytes, as well as in cortical astrocytes, was characterized by NMR spectroscopy. The astrocytes, contrary to neurons, synthesized glutamine. The maturation of the glutamine synthesis pathway was delayed in cerebellar astrocytes, as compared to cortical astrocytes. The fluxes involved in exogenous glucose utilization were quantified. The results demonstrated that if neurons used exclusively glucose as carbon source to fuel the Krebs cycle, the carbon sources for astrocytes were diversified (glucose, exogenous amino acids, endogenous carbon sources). In the same way, the pyruvate carboxylase activity was of minor importance in neurons, that implied the need for these cells of exogenous carbon substrates. We evidenced that alanine and citrate were also synthesized by astrocytes and exported to their extracellular medium. These metabolites may play a role as carbon and/or nitrogen shuttles betwen neurons and astrocytes. 31P NMR data showed similar energy charges in cerebellar neurons, astrocytes and in the cerebellum. Differences in the content of metabolites linked to membrane metabolism were observed. The postnatal development of the cerebellum was studied using 31P and 1H NMR spectroscopy. A large content of acetate was evidenced at birth, that decreased during the first postnatal days whereas the NAA content increased

In the context of the fundamental and comprehensive current reform of education in Vietnam, the need of determining the basic theories of education as well as the teaching and learning models has significantly grown. At the beginning of the 21st century, constructivism-based pedagogies have been carried into Vietnamese education through a model entitled Vietnamese Escuela Nueva (VNEN). Accordingly, a profound deployment of the constructivism-based pedagogical reform was required with the aim to establish learners’ competences rather than merely providing knowledge to them as was done in the traditional education. Whereas the competence-based curriculum aligned with the constructivist pedagogy has become a hectic preparation of the education reform in Vietnam, teachers’ beliefs toward the constructivism-based teaching and learning approach have attracted educators’ attentions. This research comprises four studies as follows.The first study is to explore the Vietnamese primary-school teachers’ beliefs regarding self-study textbooks of the VNEN project, in order to determine to what extent the teachers think the textbooks support them to implement the constructivist pedagogies in Vietnamese language instruction. The results revealed inconsistence in the teachers’ beliefs toward roles of the targeted textbooks in directing teaching and learning activities. The mismatches between the VNEN textbooks and their original versions were also explored. Finally, the strengths and shortcomings of contents and physical quality of the textbooks were collected and used as bases for improving their quality.The second study aims at investigating Vietnamese primary teachers’ beliefs on the dialectical constructivist pedagogy that is approved by Vietnamese Ministry of Education and Training (MoET) through the VNEN-based reform. The study found out a tendency of a simplified interpretation, even far difference from the original pedagogical intentions of the dialectical constructivist approaches. In addition, an inconsistence was unfolded between what the teachers thought about the constructivism-based instruction and what they actually implemented in their classroom. The follow-up interviews about the classroom observations provided the reasons and understandings that are responsible for the mismatch between their expressed beliefs and their teaching practices.In the third study, we a step forward in designing a constructivism-aligned instructional model in order to accommodate the dialectical constructivist features into the Vietnamese language instruction. This study focuses on describing the design of a formal model in which core features of dialectical constructivist pedagogies are synthesised with essential aspects of Vietnamese education and culture. The designed model was deployed in six lessons in Reading and used in the following experimental study.Through a fourth study, the feasibility of the dialectical constructivist-aligned model of teaching primary Vietnamese language was validated and evaluated through an experimental study. The teachers’ teaching practices showed meaningful changes for a traditional way of learning to a more dialectical constructivist way. Concurrently, the experiment had a positive impact on the pupils’ reading competences and their high-level thinking. In addition, the teachers’ perceptions about the dialectical constructivist-aligned lessons as well as their proposition for adjusting the designed model were collected. The challenges of the model implementation in Vietnamese culture and education were perceived and discussed by the teachers. Finally, the pupils also showed their positive attitudes toward the designed lessons. This experimental study brought opportunities to improve the constructivist-aligned model and enhance the quality of teaching Vietnamese language at primary education in order to meet the new requirements of the current pedagogical reform in Vietnam

Hepatitis C virus (HCV) is a human pathogen responsible for liver diseases including acute and chronic hepatitis, cirrhosis and hepatocellular carcinoma. Its high prevalence, the absence of a prophylactic vaccine and the poor efficiency of current therapies raises huge medical problems. In order to control HCV infection and to develop direct or indirect antivirals, our knowledge of the virus has to be improved. Hepatitis C virus has been identified in 1989 and is an developed, positive-stranded virus classified in the Flaviridae family. The development of HCV pseudoparticles in 2003 and, most recently, in 2005 of cell culture grown HCV allowed the study of the early event occurring during infection of hepatoma cell lines. The originality of this work is to make use of an in vitro infection model base on the infection of human hepatocytes in primary culture by HCV particles contained in the sera of infected individuals. This model is closed to the physiological situation of infection and reproduces the entire life cycle of HCV. In this infection system, three different research projects have been developed to decipher the early event of infection

The aim of this work is to describe and understand the elements involved in the construction of the "culture of action" in the case of primary school teachers in Chile. By basing our thinking on the concepts “culture of action” (Barber, 2010) and own-culture (Theureau, 2011), we have made the hypothesis that Culture "shows" itself to the participants in situations where culture is questioned. Based on this premise, four situations seen as " unsettling " were studied: (1) the designation of a teacher in a class level with which the teacher had little (or no) experience; (2) Working in a classroom with another professional; (3) the implementation of a program to improve teaching practices and; (4) the influence of the organization of space on educational strategies. These situations have been analyzed in relation to the theoretical and methodological framework of the empirical program “course of action” (Theureau, 2004, 2006). By favoring the qualitative approach, three types of data were collected from six teachers working in public schools: audiovisual records of the classes, audiovisual records during sessions in self-confrontation with the teachers, and classroom observation notes. The results reveal that the "weight" of cultural components vary depending on the context, the history of institutions and the personal history of the participants. At the same time, these results add to the works that reveal a cultural registration of the objects (Adé et de De Saint-Geoges, 2004) and the collective activity configurations (Veyrunes, 2011). Finally, these results contribute to the development of enactive anthropology, while providing a definition of Culture that reflects the dynamic and complex nature of the human activity of teaching
La presente tesis tiene como objetivo describir y comprender los elementos que participan en la construcción de la "culture de la acción" de los docentes de primaria en Chile. Fundando nuestra reflexión en la nociones de « culture de la acción » Barbier, 2010) et de cultura-propia (Thereau, 2011), hemos hecho la hipótesis que la cultura « se muestra » a los actores en las situaciones en que ella es « cuestionada » . De esta forma, fueron estudiadas cuatro situaciones entendidas como "perturbadoras": (1) la asignación en una nueva clase en que los profesores no tienen (o tienen muy poca) experiencia previa ; (2) el trabajo en clases con otro profesional; (3) la aplicación de una prescripción que busca mejorar las practicas docentes y (4) la influencia de la organización del espacio sobre las estrategias pedagógicas. Estas situaciones fueron analizadas en referencia al cuadro teórico y metodológico del programa empírico del « curso de la acción » (Theureau, 2004, 2006). Privilegiando una entrada cualitativa, tres tipos de datos fueron recogidos durante las clases de seis profesoras de escuelas publicas: registros audiovisuales de las clases, registros de entrevistas en auto confrontación y notas de observación de clases. Los resultados muestran que los componentes culturales "pesan" de maneras diferentes según los contextos, la historia de las instituciones y la historia personal de los actores. Al mismo tiempo, estos resultados prolongan los trabajos que dan cuenta de una inscripción cultural de los objetos (Adé y De Saint Georges, 2004) y de las configuraciones de actividad colectiva (Veyrunes, 2011). Finalmente, nuestros resultados contribuyen al desarrollo de una antropología enactiva al proponer una definición de cultura que pone en evidencia el carácter dinámico y complejo del trabajo docente como actividad humana

In fact, the discourse reveals that some teachers use art at school rather as a means of discipline in their class, instead of using it to develop expression, creation and the development of children
