Abstract:
Assimilation of atlas is a rare skeletal malformation causing nerve compression with high risk of fatal. However, the genetic etiology of assimilation of atlas AOA is currently lacking. In this paper, the whole-exome sequencing (WES) analysis was employed to study a Chinese family having a sporadic proband son of assimilation of atlas AOA but other healthy family members. We identified a novel variant in ciliary rootlet coiled-coil gene (NM_014675.5 (CROCC): c.4702C>T (r.4702c>u, p.(Arg1568Cys)). The variant had different genotypes between the proband and healthy family members but with high conservations of “damage” to protein structure based on MutationTaster and SIFT prediction. CROCC gene can be obtained in both healthy (n=220) and non-mutated assimilation of atlas AOA patient samples (n=68) but absented in five sporadic patients with the novel variant. Furthermore, abnormal of cilia was observed after editing the target sequence on CROCC using CRISPR-Cas9. These results suggested that assimilation of atlas AOA might be caused by the mutation of CROCC: c.4702C>T (r.4702c>u, p.(Arg1568Cys)). With strong amino acid conservation and interaction regulation, the variant mutation could cause the signal disorder of skeletal development which may lead to the defective bone formation and finally cause the development of assimilation of atlas AOA.

Abstract:
The purpose of this study is to examine the factors influencing employee turnover in the context of manufacturing industry. The study used existing literature to understand the relationship among these factors by using Interpretive Structural Modeling (ISM). Crocc-Impact Matrix Multiplication Applied to Classification (MICMAC) analysis was also used to classify the factors. The data were collected from primary and secondary sources. The results indicates that work-environment and career growth were critical factors followed by compensation, training and development, reward and recognition, job security, job stress, work-life balance, etc. In order to reduce employee turnover, the manufacturing sectors can mull over employee relations with colleagues and external job opportunities and emphasize job security, job stress, and work-life balance. The fuzzy-MICMAC results showed that the dependent factors were external job opportunities, job stress, job security, and work-life balance. As the study primarily uses experts’ opinion, the probability of collecting views was restricted.

Rhodizonic acid (H2RHOD) was reacted with Co(PF6)2.6H2O in the presence of pyridine modified large macrocyclic ligand (H2Lopy) and triethylamine in N,N-dimethylformamide under argon atmosphere. Instead of [Co4(RHOD)2(Lopy)](PF6)2 (1), the main product of this reaction was a crystalline compound [Co4(CROC)2(Lopy)](PF6)2, which was isolated in low yield (< 25%). Abstract:
In mammals, cilia are critical for development, sensation, cell signaling, sperm motility, and fluid movement. Defects in cilia are causes of several congenital syndromes, providing additional reasons to identify cilia-related genes. We hypothesized that mRNAs selectively abundant in tissues rich in highly ciliated cells encode cilia proteins. Selective abundance in olfactory epithelium, testes, vomeronasal organ, trachea, and lung proved to be an expression pattern uniquely effective in identifying documented cilia-related genes. Known and suspected cilia-related genes were statistically overrepresented among the 99 genes identified, but the majority encoded proteins of unknown function, thereby predicting new cilia-related proteins. Evidence of expression in a highly ciliated cell, the olfactory sensory neuron, exists for 73 of the genes. In situ hybridization for 17 mRNAs confirmed expression of all 17 in olfactory sensory neurons. Most were also detected in vomeronasal sensory neurons and in neighboring tissues rich in ciliated cells such as respiratory epithelium. Immunoreactivity for one of the proteins identified, Spa17, colocalized with acetylated tubulin in the cilia layer of the olfactory epithelium. In contrast, the ciliary rootlet protein, Crocc, was located in discrete structures whose position was consistent with the dendritic knobs of the olfactory sensory neurons. A compilation of >2,000 mouse genes predicted to encode cilia-related proteins revealed a strong correlation ( R = 0.99) between the number of studies predicting a gene's involvement in cilia and documented evidence of such involvement, a fact that simplifies the selection of genes for further study of the physiology of cilia.

AimsPrimary cilia play an important role in the regulation of cell signalling pathways and are thought to have a role in cancer but have seldom been studied in human cancer samples.MethodsPrimary cilia were visualised by dual immunofluorescence for anti-CROCC (ciliary rootlet coiled-coil) and anti-tubulin in a range of human cancers (including carcinomas of stomach, pancreas, prostate, lung and colon, lobular and ductal breast cancers and follicular lymphoma) and in matched normal tissue (stomach, pancreas, lung, large and small intestines, breast and reactive lymph nodes) samples using a tissue microarray; their frequency, association with proliferation, was measured by Ki-67 staining and their structure was analysed.ResultsCompared with normal tissues, primary cilia frequency was significantly elevated in adenocarcinoma of the lung (2.75% vs 1.85%, p=0.016), adenocarcinoma of the colon (3.80% vs 2.43%, respectively, p=0.017), follicular lymphoma (1.18% vs 0.83%, p=0.003) and pancreatic adenocarcinoma (7.00% vs 5.26%, p=0.002); there was no statistically significant difference compared with normal control tissue for gastric and prostatic adenocarcinomas or for lobular and ductal breast cancers. Additionally, structural abnormalities of primary cilia were identified in cancer tissues, including elongation of the axoneme, multiple basal bodies and branching of the axoneme. Ki-67 scores ranged from 0.7% to 78.4% and showed no statistically significant correlation with primary cilia frequency across all tissues (p=0.1501).ConclusionsThe results show upregulation of primary cilia and the presence of structural defects in a wide range of human cancer tissue samples demonstrating association of dysregulation of primary cilia with human cancer.

Analysis results feature a recommended vision and mission statement for Crocs, the best strategies for Crocs to pursue, how Crocs can progress toward achieving such strategies (through certain policies, risk considerations, and financial projections), and how the company can successfully evaluate and adapt strategies if necessary. This analysis implies how Crocs may best utilize the strategic management process to achieve success as a company in the footwear industry.

Pada tahun 2017, 36% dari 43,7 juta pemain game di Indonesia adalah anak usia sekolah. Penelitian pengembangan ini bertujuan menghasilkan game edukasi android Mat Croco yang memiliki kualitas baik (valid, praktis dan efektif). Jenis penelitian ini merupakan penelitian pengembangan model ADDIE. Hasil analisis uji kevalidan game Mat Croco oleh dua ahli dinyatakan memiliki kevalidan yang baik. Hal ini ditunjukkan dari skor kevalidan media pembelajaran 36,5 dan skor kevalidan soal penalaran matematika juga 36,5. Hasil analisis kepraktisan oleh dua guru matematika dan dua siswa menunjukkan bahwa game Mat Croco memiliki kepraktisan yang sangat baik. Rata-rata skor kepraktisan oleh guru 37 dan skor kepraktisan dari siswa 36. Hasil analisis uji keefektifan game Mat Croco efektif berorientasi penalaran matematika siswa. Analisis deskriptif mengalami peningkatan rata-rata nilai penalaran matematika (pretes=28,4 dan postes=84,4) dan persentase ketuntasan (pretes=0% dan postes=75,9%). Paired Samples Test menyimpulkan bahwa game Mat Croco efektif berorientasi penalaran matematika (|t| (25,200) > t tabel (t 0,025;28 = 2,368) dan sig.(0,000) < 0,0025). Hasil analisis menunjukkan bahwa game edukasi android Mat Croco memiliki kualitas yang baik (valid, praktis dan efektif) dan layak diaplikasikan dalam pembelajaran matematika materi barisan dan deret.

Background: The lack of consensus criteria of acute on chronic kidney injury (ACKI) affects the judgment for its clinical prognosis. Methods: In this study, we analyzed the data from 711,615 hospitalized adults who had at least 2 serum creatinine (SCr) tests within 30 days. We estimated the reference change value (RCV) of SCr given initial SCr level in adults without known risks of acute kidney injury other than chronic kidney disease (CKD). We proposed a criterion for ACKI based on the RCV of SCr (cROCK), which defined ACKI as a ≥25% increase in SCr in 7 days. We validated cROCK by its association with the risks of in-hospital mortality, death after discharge, and CKD progression in a large cohort of patients with CKD stage 3. Results: In 21,661 patients with CKD stage 3, a total of 3,145 (14.5%), 1,512 (7.0%), and 221 (1.0%) ACKI events were detected by both cROCK and Kidney Disease Improving Global Outcomes (KDIGO), cROCK only, and KDIGO only, respectively. cROCK detected 40% more ACKI events than KDIGO. Compared with patients without ACKI by both definitions, those with cROCK- but not KDIGO-defined ACKI had a significantly increased risk of in-hospital mortality (hazard ratio [HR] 5.53; 95% CI 3.75–8.16), death after discharge (HR 1.51; 95% CI 1.21–1.83), and CKD progression (OR 5.65; 95% CI 3.05–10.48). Conclusions: RCV-based criterion (cROCK) for ACKI is clinically valid in that it has a substantially improved sensitivity in identifying patients with high risk of adverse outcomes.

The relation between velocity and enthalpy in steady boundary layer flow is known as the Crocco relation. It describes that for an adiabatic wall the total enthalpy remains constant throughout the boundary layer, when the Prandtl number (Pr) is one, irrespective of pressure gradient and compressibility. A generalization of the Crocco relation for Pr near one is obtained from a perturbation approach. In the case of constant-property flow an analytic expression is found, representing a first-order extension of the standard Crocco relation and confirming the asymptotic validity of the square-root dependence of the recovery factor on Prandtl number. The particular subject of the present study is the effect of compressibility on the extended Crocco relation and, hence, on the thermal recovery in laminar flows. A perturbation analysis for constant Pr reveals two additional mechanisms of compressibility effects in the extended Crocco relation, which are related to the viscosity law and to the pressure gradient. Numerical solutions for (quasi-)self-similar as well as non-similar boundary layers are presented to evaluate these effects quantitatively.

OBJECTIVE:To investigate if IgG can be expressed in clear cell renal cell carcinoma (cRCC) , and the expression of IgG is involved in the cancer progression. If IgG expression can serve as a potential target in cancer therapies and be used for judging the prognosis.MATERIALS AND METHODS:By immunohistochemistry, we detected IgG in cRCC tissues(75 cRCC tissues and75 adjacent normal kidney tissues). Immunofluorescence and Western blot was used to detect the IgG in cRCC cell lines (786-0, ACHN and CAKI-I). By RT-PCR, the functional transcript of IgG heavy chain was detected. Knockdown of IgG was to analyze the proliferation, migration and invasion ability by CCK8, Transwell and Matrigel and apoptosis in cRCC cell lines.RESULTS:By immunohistochemistry, we found strong staining of IgG in 66 cases of 75 cRCC tissues and 63 cases of 75 adjacent normal kidney tissues. Immunofluorescence and Western blot was found IgG in cRCC cell lines. Knock-down IgG in cRCC cell lines resulted in significant inhibition of cell proliferation, migration and invasion, and the induction of apoptosis of the 786-0 cells. The immunohistochemistry analysis showed that high IgG expression significantly correlated with the poor differentiation and advanced stage of cRCC.CONCLUSION:IgG was over expressed in cRCC and was involved in the proliferation, migration and invasion of cancer cells. IgG expression may serve as a potential target in cancer therapies and could be used for judging the prognosis.

Van der Waals (vdW) epitaxial growth provides an efficient strategy to prepare heterostructures with atomically and electronically sharp interfaces. Herein, PbI2 was in situ thermally deposited onto exfoliated thin−layered CrOCl nanoflakes in high vacuum to fabricate vdW PbI2/CrOCl heterostructures. Optical microscopy, atomic force microscopy, X−ray diffraction, and temperature−dependent Raman spectroscopy were used to investigate the structural properties and phonon behaviors of the heterostructures. The morphology of PbI2 films on the CrOCl substrate obviously depended on the substrate temperature, changing from hemispherical granules to 2D nanoflakes with flat top surfaces. In addition, anomalous blueshift of the Ag1 and Au2 modes as the temperature increased in PbI2/CrOCl heterostructure was observed for the first time. Our results provide a novel material platform for the vdW heterostructure and a possible method for optimizing heterostructure growth behaviors.

Abstract. This paper describes a geophysical investigation conducted into two important churches, namely the Cathedral of Santa Maria Assunta and the Church of Santa Croce, both in Gravina in Puglia (close to Bari, southern Italy). The Church of Santa Croce, now deconsecrated, lies below the cathedral. Therefore, the two churches constitute a unique building body. Moreover, below the Church of Santa Croce there are several crypts, which are only partially known. The prospecting was performed both with a pulsed commercial ground penetrating radar (GPR) system and with a prototypal reconfigurable stepped frequency system. The aim was twofold, namely to gather information about the monument and to test the prototypal system. The GPR measurements have also been integrated with an infrared thermography (IRT) investigation performed on part of the vaulted ceiling in the Church of Santa Croce, in order to confirm or deny a possible interpretation of certain GPR results.

For these innovative machines, the literature review shows that more publications concerning axial type turbomachines are available than centrifugal ones. This work deals with a design and experimental performance analysis, applied to two counter-rotating impellers of a centrifugal compressor “CRCC”. CRCC was designed with a specifically developed tool based on mean-line approach coupled with optimization algorithms and a stream-curvature through-flow method to satisfy the design criteria. This paper presents an experimental validation of the CRCC design tool and its performances against the baseline “SR”, composed of one centrifugal impeller and a volute for which experimental data are available. CRCC numeric simulations are also validated by experimental data. For a fair comparison between CRCC and SR, the same volute is used for both configurations. The CRCC studied here includes a first conventional impeller with an axial inlet and a radial outlet, while the second impeller is parametrically designed and can be considered a rotating bladed diffuser with a radial inlet and outlet. The obtained results show that CRCC can deliver a pressure rise increase of two compared to SR, along with an increase of isentropic efficiency and also validate the design method of this novel layout. The experimental results also show that the speed ratio of CRCC has a positive effect on the surge and shock margin.

10568 Background: Adrenomedullin (AM) is a 52 amino acid peptide that has an important role on tumor cell proliferation and neoangiogenesis through its receptors CRLR/RAMP2 and CRLR/RAMP3. AM gene expression is stimulated by Hypoxia Inductible Factor-1 (HIF- 1) and 60–80% of the human conventional renal carcinomas (cRCC) display mutations in the tumor suppressor protein von Hippel-Lindau, which leads to constitutively elevated HIF-1. Methods: Tumors and non-malignant kidney tissues were obtained from patients who underwent unilateral nephrectomy. We studied VEGFA, AM and its receptors expression by quantitative RT-PCR in 62 frozen renal tumors including: 40 cRCC; 5 cRCC metastasis; 5 chromophobe carcinomas; 5 Papillary carcinomas; 2 oncocytomas; 2 collecting duct carcinomas; 2 normal adjacent renal tissue; 1 unclassified renal tumor. AM and AM-receptors expression was studied in 42 paraffin-embedded renal tumors by immunohistochemistry using rabbit anti-AM, anti-CRLR, anti-RAMP2 and anti-RAMP3 polyclonal antibodies. Effects of these anti AM or anti AM- receptors antibodies on cell proliferation were examined in vitro on BIZ cell line (cRCC cells). Results: VEGFA, AM and AM-receptors genes were overexpressed in cRCC compared to other renal tumors (Except AM gene in chromophobe carcinoma). Their expressions were independent of classical prognostic factors (such as Fuhrman Grade and pT status). In cRCC, there was a strong positive correlation between VEGF-A gene expression and AM (r=0.7; p=0.01) and AM-receptors genes expressions (RAMP2 r=0.61; p= 0.01 and RAMP3 r=0.58; p= 0.01). Immunohistochemistry confirmed a tumor expression of CRLR, RAMP2 and AM in more than 80% cRCC. RAMP3 was expressed by inflammatory cells but not by tumoral cells. The cell proliferation assay showed a significant inhibition of BIZ cell proliferation by AM antibody or AM-receptors antibodies. Conclusion: AM and its receptor CRLR/RAMP2 are overexpressed in cRCC. In vitro cell proliferation assay results support that AM inhibition may suppress cRCC growth, independently of potential antiangiogenic effects. Further studies on animal models are needed but AM pathway may be a potential therapeutic target in cRCC. No significant financial relationships to disclose.

It also describes the public roles and political ideas of the editor of La Critica, Benedetto Croce, and the editors of the Glasnik, Bogdan Popovic, Pavle Popovic and Jovan Skerlic. It looks at the interpretations of Croce?s political ideas put forward in the Glasnik, recognizing a closeness between the liberal literary and political renewal programmes of Benedetto Croce, on the one hand, and Bogdan Popovic, Pavle Popovic and Jovan Skerlic, on the other. Finally, it points to the Glasnik?s repulsion towards the imperialist ideas of Gabriele D?Annunzio, Croce?s main rival in the Italian culture of the period. But under the editorship of Jovan Skerlic, at the time when Serbia was subjected to Austria-Hungary?s pressure and war threat, the Glasnik published D?Annunzio?s short stories and advocated the ideals of activism, vitalism and heroism.

Abstract SLAM family receptors play a critical role in immune regulation. Their function is controlled primarily by SAP, EAT-2, and ERT adaptors. We have previously described a novel immunostimulatory function for EAT-2 in a vaccine model system. In the current work we sought to examine the mechanism of action of EAT-2 in APCs. Our results demonstrated that Ad vector infection itself of macrophages significantly (p&lt;0.001) induced the transcription of the SLAM family member CRACC, but not other SLAM family members. In dramatic contrast, infection of macrophages with Ad-EAT2 significantly prevented Ad induced CRACC expression (p&lt;0.001). FACS analysis confirmed that Ad mediated expression of EAT-2 also resulted in decreased protein levels of CRACC on APCs. By utilization of a variety of inhibitors, we confirmed that EAT-2 mediated suppression of CRACC required a functional ERK, but not Src-kinase, PI3K, or p38 pathways. In addition, blockade of ERK signaling abolished the ability of Ad-EAT2 to induce other innate immune response genes in APCs. Finally, utilization of immunoblot confirmed that Ad-EAT-2 significantly induced ERK activation in macrophages. Thus, EAT-2 over-expression by Adenovirus activates ERK pathway that inhibits Ad mediated induction of CRACC in APCs, an activity that correlates with increased induction of several innate immune response genes, as well the improved induction of antigen specific adaptive immune responses after Ad mediated transfer of immunogenic antigens.

A scanning electron microscope (SEM) study of G. bakeri is given for the first time. Diastolaimus croca is found to have a circle of six setiform processes anterior to the true labial setae. Detailed descriptions of the female reproductive system in G. backeri, D. croca and Macrolaimus sp. are given. The use of the female reproductive system and structure of the labial sensilla in the systematics of the family Chambersiellidae, together with its taxonomic placement, are discussed. Notes on the systematic position of the family Bicirronematidae and a key to the species of this family are given.

Atomically thin two-dimensional (2D) materials make it possible to create a variety of van der Waals (vdW) heterostructures with different physical features and attributes, which enables the growth of innovative electronics and optoelectronics applications. The band alignment and charge transfer play a crucial role in the physical and optoelectrical properties of the vdW heterostructure. Here, we design a vdW heterojunction device comprising low-symmetric CrOCl to induce a stable anti-ambipolar behavior and polarization-sensitive photodetection performance. 2D CrOCl exhibits strong in-plane anisotropy and linear dichroism, and an anti-ambipolar transport behavior is observed in a MoTe2 channel due to the gate-tunable band bending and charge transfer at MoTe2/CrOCl interface. The devices also exhibit well photodetection performance with a responsivity of 1.05 A/W and a temporal response of 970 μs. Owing to the anisotropic CrOCl serving as a photosensitizing layer, the device achieves the capability of polarization-sensitive photodetection with a photocurrent dichroic ratio up to ∼6. This work offers a valid device model and design strategy to realize the versatile optoelectronics, including the anti-ambipolar transistor and polarimetric photodetectors.

ABSTRACTPeriodontitis is a highly prevalent, biofilm-mediated chronic inflammatory disease that results in the loss of the tooth-supporting tissues. It features two major clinical entities: chronic periodontitis, which is more common, and aggressive periodontitis, which usually has an early onset and a rapid progression. Natural killer (NK) cells are a distinct subgroup of lymphocytes that play a major role in the ability of the innate immune system to steer immune responses. NK cells are abundant in periodontitis lesions, and NK cell activation has been causally linked to periodontal tissue destruction. However, the exact mechanisms of their activation and their role in the pathophysiology of periodontitis are elusive. Here, we show that the predominant NK cell-activating molecule in periodontitis is CD2-like receptor activating cytotoxic cells (CRACC). We show that CRACC induction was significantly more pronounced in aggressive than chronic periodontitis and correlated positively with periodontal disease severity, subgingival levels of specific periodontal pathogens, and NK cell activationin vivo. We delineate howAggregatibacter actinomycetemcomitans, an oral pathogen that is causally associated with aggressive periodontitis, indirectly induces CRACC on NK cells via activation of dendritic cells and subsequent interleukin 12 (IL-12) signaling. In contrast, we demonstrate that fimbriae fromPorphyromonas gingivalis, a principal pathogen in chronic periodontitis, actively attenuate CRACC induction on NK cells. Our data suggest an involvement of CRACC-mediated NK cell activation in periodontal tissue destruction and point to a plausible distinction in the pathobiology of aggressive and chronic periodontitis that may help explain the accelerated tissue destruction in aggressive periodontitis.

The waste energy recovery and management philosophy represent a great challenge for scientists. This article outlines a scheme to utilize two different source temperature levels in the range of (160–200) °C. Two regenerative organic Rankine cycles (RORC) were implemented to construct a compound regenerative organic Rankine cycle (CRORC) to improve the energy management of the sources. The method of energy management for these cycles was accomplished by extracting a certain amount of energy from the high-temperature cycle and rejecting it to the working fluid in an economizer at the low-temperature level. R-123 was circulated in the high-temperature cycle due to its high critical temperature at evaporation and condensation temperatures of 150 °C and 50 °C respectively. R-123, R-245fa, R-1233zd-E, and the hydrocarbon R-600a were used as working fluids for the low-temperature cycle at evaporation and condensation temperatures of 130 °C and 35 °C respectively. This technique showed that the first law of thermodynamics efficiency was augmented by (3–5)% for the low-temperature mini-cycle of the (CRORC). The energy consumption at the low-temperature cycle was also reduced by (3–5)%. The latter reduction range accounts for 2% for the total extracted energy for the independent system where both high-temperature and low-temperature cycles were utilized separately. The data showed that increasing the superheat degree from 10 °C to 20°C has enhanced the thermal efficiency of the compound (CRORC) system by (2–4)%. The (CRORC) system of R-123/R600a, R-123/R-123, and R-123/R-245fa fluid pairs exhibited higher thermal efficiency than that of R-123/R-1233zd-E pair by (4.5–6)%, (4–6)% and (3–4)% respectively. The net thermal efficiency of the compound (CRORC) system fell in the range (12–13)% and the low-temperature mini-cycle of the (CRORC) system had a range of (12–14)% for all of the examined operating conditions.
