Full textDown syndrome is the most frequent cause of mental retardation affecting millions of people worldwide and results from full or partial trisomy of chromosome 21 (HC21). Rare cases of partial trisomy allowed the identification of a small region in HC21 common to all carriers called Down Syndrome Critical Region. The genes DCRA and DSCR8, mapped to this region, encode two proteins of unknown function. With the aim of contributing to a better characterization of these proteins, DSCR8 was subcloned and expressed in fusion with thiorredoxin in Escherichia coli Rosetta (DE3). Recombinant Trx-DSCR8, observed in the soluble fraction, was subjected to the initial purification assays by affinity chromatography in a Ni-NTA column. The gene DCRA was cloned and expressed in fusion with the proteins thiorredoxin and GFP allowing the partial purification of the protein and the achievement of crystallization and immunological assays. The amino acid sequence of DCRA showed 57% and 49% of identity to a protein of unknown function from Anopheles gambiae and Drosophila melanogaster respectively. Also, in silico analyses revealed that DCRA contains a putative Vps26 domain. Subcellular localization of DCRA in fusion with GFP was observed preferentially in the cytoplasm of the cell lines tested. These results contribute to a better characterization of DCRA and DSCR8 and open up possibilities towards the understanding of the cellular role of these proteins and their relationship with the Down syndrome.
A síndrome de Down é a causa mais freqüente de retardo mental que afeta milhões de pessoas em todo o mundo e resulta de uma trissomia completa ou parcial do cromossomo 21 (HC21). Casos raros de trissomia parcial permitiram identificar uma pequena região do HC21 comum a todos os portadores denominada de Região Crítica da Síndrome de Down. Os genes DCRA e DSCR8 foram mapeados nesta região e codificam proteínas de função desconhecida. Com o objetivo de contribuir para a caracterização destas proteínas DSCR8 foi expresso em fusão com seqüência codificadora da tiorredoxina na linhagem de Escherichia coli Rosetta (DE3) o que possibilitou a obtenção de Trx- DSCR8 na fração solúvel em quantidade suficiente para os ensaios iniciais de purificação em cromatografia de afinidade Ni-NTA. O DCRA foi clonado em fase com seqüências codificadores da tiorredoxina e GFP o que permitiu que a solução protéica fosse parcialmente purificada e que ensaios de cristalização e imunização pudessem ser realizados. A seqüência de aminoácidos da proteína DCRA possui 57% e 49 % de identidade respectivamente com proteínas de Anopheles gambiae e de Drosophila melanogaster, ambas de função desconhecida. Análises in silico da seqüência de aminoácidos demonstram que DCRA possui um provável domínio de Vps26. A localização subcelular da DCRA em fusão com a GFP foi observada preferencialmente no compartimento citoplasmático em todas as linhagens celulares analisadas. Esses resultados contribuem para melhor conhecimento da DCRA e da DSCR8 e estimulam futuras análises para o entendimento da função celular destas proteínas, bem como suas implicações na síndrome de Down.

Full textThere is plenty of fiction written for children with an educational starting point whose intention is to challenge traditional gender patterns. However, there are few studies on how children actually perceive this norm-critical fiction, which is important for the use of literature as a didactic tool in teaching. As fiction forms a large part of the work in preschool class, this study aims to explore how student perceptions of gender can be made visible through critical literacy-inspired preschool work. The norm-critical fairy tale Rödluvan (Jansson & Jensfelt, 2018) is the fiction that forms the basis of the study and the following issues have been formulated: • How do the students name the characters' gender in Rödluvan (Jansson & Jensfelt, 2018)? • How do the students' ways of naming the characters gender differ, depending on whether they participated in a joint conversation about the text or not? The study is qualitative and the data collection is done through semi-structured interviews with 20 students in preschool class. The analysis of the material is based on gender theory and the concept of didactic potential, in accordance with Alkestrand's (2017) definition. The conclusions that can be drawn from the results of the study are that the participating students had difficulty understanding the gender of the fairy characters as described in the story of the character in question, portrayed in a norm-critical manner. Based on the results of the study, it can also be stated that there is a marked difference in the students' ways of perceiving the gender of the norm-critical characters depending on whether they participated in a joint conversation about the text or not.

Full textThis case study deals with conceptions and attitudes about gender in a preschool context and the study includes both preschool pedagogues and parents. The frame work of the study is made up by the Social constructive perspective and the study includes two parts. The first part includes interviews analyzed with Critical Discourse analysis and the second part includes an Ethnographical perspective. The reason to include two part studies is to draw attention to the difficulties in the working processes that surrounds the enforcement of a common ground for gender equality work in a preschool context. The study shows that the informants’ conceptions and attitudes about the word gender differ and this aspect effects the implementation of gender equality work at the examined preschool.

Full textThe purpose of this study is to examine media’s depiction of women’s depression, and what consequences this may possibly have from a gender perspective. This study has a certain focus that consists of the group women and on how media constructs and the describes the image of the depressed woman. The study is based on the critical discourse analysis. Our empirical material consists of news articles by the two Swedish evening newspapers Aftonbladet and Expressen, and the newspaper Svenska Dagbladet. The study is limited to articles between the period 2013-2020. Norman Fairclough’s three-dimensional model is used for analysis of the study’s empirical material. At first, we used textual analysis and discursive practice to study our empirical material. Thereby we identified media’s depiction of women’s depression. Then we used the third dimension, social practice, to further examine what consequences media’s depiction possibly may have. We used previous research regarding the increased mediatization, gender and depression as well as media in relation to gender. The theoretical framework of the study consists of gender theory and social constructivism. Our study shows that media creates and maintains discourses on women’s depression. Women are depicted by media as a high-risk group concerning the disease depression. Media’s depiction of women’s depression results in an individualized health responsibility. The over-representation of women in media’s discourses on depression is described as female symptomatology, which is explained through social constructivism and gender theory. Media’s production and maintenance of discourses results in consequences as continued repression of women. It also results in an overdiagnosis of women’s depression, which leads to men’s emotional distress being overlooked. Media’s power through discourses has a strong impact on people’s social conditions and perceptions of the world. Media’s depictions of the depressed women lead to maintenance of structures and gender norms, which also affects clinical work.

Full textThe problem I chose to base my project on is the fact that a big portion of the products we use daily are marketed against either men or women. The result of that is an exclusion of the people who do not fit inside the norm, which states that there are only two genders. My purpose during the project was to investigate this problem and in the end also criticize it in a suitable way. Later on, my focus was shifted towards a specific category of products - menstrual hygiene products. More specifically their packaging. They are marketed only towards women and exclude all who are not cis-women. The subject of menstruation is also a loaded one and at the moment frequently discussed within certain circles. Most people seem to have a lot of feelings and opinions regarding the subject, which will hopefully get my project into the public eye. My process started in collecting a broad range of facts and theories surrounding the subject, which the remaining parts of my project is based on. The information about norms, stereotypes, gender, packaging, transpersons, menstruation and menstrual hygiene products was something I could always go back to whenever I needed help in decision making during the project. The contact I had with the consumers through the surveys were an important part in building my understanding of what creates attention and feelings, which has been valuable in working towards critical design. The packaging is created as a contrast against the packaging for menstrual hygiene products existing today. The period is allowed to take up space in the form of illustrations of the menstrual product as well as the color red, which symbolizes menstrual blood. This is also combined with clear plastic on two sides which show the containing products. The font used on the majority of the packaging is a combination of hard shapes, often considered masculine, and thin round shapes, often considered feminine. Together they create a gender neutral appearance. The name of the company ”har du mens eller?” is chosen after a phrase commonly used by men to disparage women’s feelings and opinions. In this case, it is written in a typically feminine font which is very common in this category of products. With the different parts of the packaging, my hope is to rise attention and bring awareness to how the products surrounding us are marketed and how this affect us.

Full textIn this essay I examine how the norm-critical material ”BRYT! ett metodmaterial om normer i allmänhet och heteronormen i synnerhet” is used by Hyresgästföreningen in Stockholm in their education of personnel in the perspective of equalisation. I’ve mostly been interested in how the personnel who takes part in workshops about the material react and reflect in relation to the purpose of the material. How do they discuss about norms and the connection to discrimination and how do they look upon the power and responsibility that comes with power in relation to norms? Earlier research in norm critical pedagogy and the critic of tolerans pedagogy perspective helps me to analyse how critical pedagogy stands above non-critical pedagogy in many perspectives.
By observing workshops and by interviewing personnels taking part in these workshops I’ve examined how they react on both the practical and theoretical parts of the material.
To summarise my examination I find the personnel who is educated in working with the material discover new experiences in knowledge and on how norms and discrimination functions. They explain that they have experienced knowledge about the connection between norms and discrimination and thereby how oppression could be counteracted and prevented.

Full textEvery year, approximately 17 500 drunk drivers are sued in Sweden. Of these, only 10 percent are women. The women are therefore considered a minority. In this study, I have taken interest in how the female drunk driver is constructed implemented by the probation service. By using critical discourse analysis I have found that the woman in these investigations is contributed the role of a deviant in two senses. She deviates because she is a woman who has committed a criminal deed and also because she often has problems with alcohol as well. I have found that the woman, because of these deviations, is depicted as a victim, both for the situation she is in and for her own actions.

Full textSwedish school is obliged to actively work with questions concerning gender, equality, rights and opportunities. Furthermore, school shall give the students tools to critically review gender patterns and understand how gender patterns can constrain people’s living conditions (Skolverket, 2019). In school the students read fiction, which in turn contributes to how norms are produced in different ways. Through didactic efforts through critical literacy inspired working processes fiction can be used as a foundation for work with values. It is of thus of relevance how children perceive the norms or norm-breaking aspects as they are produced in fiction. The purpose of the study is therefore to contribute knowledge about students’ different ways of reasoning and relating to a norm-breaking story through a Critical literacy-inspired didactic perspective. As a basis for the study is the norm-breaking fairytale about Cinderella (Jansson & Jensfelt, 2018). The study aims to address the following questions: · Which norm-breaking aspects do the students retell in a dialogue about the norm-breaking fairytale? · Which aspects of a norm-breaking fairytale can the students relate to from their own reality? · How are the students’ reasoning affected by the norm-breaking fairytale after a literacy talk? The study is qualitative and is built on semi-structured interviews with 12 students in year 2 and 3. The result of the study shows which norm-breaking aspects the students’ reason about. Furthermore, the results show that a literacy talk seems to have an impact on the students’ reasoning. The empirical material is analyzed through gender theory together with Alkestrand’s (2017) definition of didactic potential.

Full textThis thesis aims to describe the different historical stages through which the narrative structure of the character of Perceval evolved during the first two centuries of its genesis in Romance and European literature. The character had a significant success from the Middle Ages (Chrétien de Troyes) up to the modern and contemporary age. The philological study about the evolution of the character within the textual galaxy and the forms of his tradition allows us to outline a composite picture, in which it is possible to trace the genesis and development of important historical-literary phenomena, destined to influence European and Western literature in the long term. A second part is instead dedicated to the study of the literary and narrative structure of the character that can be traced within the reading of the different works that hand down the versions of the story of Perceval. Finally, the research will focus on a case study capable of illuminating the persistence of the structures identified above: the Prophecies de Merlin, an arthurian romance from the second half of the 13th century, are in fact an expression of one of the many ways in which Perceval’s life story is staged in texts that do not directly focus on his biography. Within the tradition of the Prophecies de Merlin, the prophet of the Arthurian kingdom whose numerous prophecies are collected, the case of the Livre d’Helias et Perceval is emblematic. The Livre d’Helias et Perceval from the Prophecies de Merlin, accompanied by a philological study that includes a critical edition, a complete study of the tradition and linguistic analysis, is thus proposed as an ideal case study for ana-lysing the fortune and evolution of the character of Perceval in the Romance and European Middle Ages.

The writing of texts of rhetoric genres is essential to the literate practices in one democratic society. In this case, is very important to the school formation mediation the development of pedagogical interferences about the teaching this genres. This search constructs events of written that objective to the teaching of the written how social practice. The general objective this search is to investigate the factors that contribute to amplification of the discourses competences of the students, in works with rhetoric kinds that realize the type of dissertation. For to obtain this objective, we realized a search, whose actions supported themselves in Bakhtin’s position, in that the language has been considered in your conception character and about uses the speaking in the verbal complementation (BAKHTIN, 1992), (BAKHTIN, 1997; BAWARSHI and REIFF, 2013), of the argumentation and the new rhetoric (PERELMAN and OLBRECHTS-TYTECA, 2005; BAZERMAN, 2015a) and of the writing and various writing (GOULART, 2011; ROJO, 2013; RIOS, 2014); and we use discussions about the critic think (GIROUX, 1997; NOBRE, 2004; CHAUÍ, 1982). Position of Geraldi (2010) and Bawarshi and Reiff (2013), and position of Geraldi (2010), and Bawarshi and Reiff (2013), the know of the model forms of then kinds don’t transforms, necessarily, in domain of the writing production of this kinds. We verify that, still, there’s a deep wish from the students become proficient in the written of this rhetoric kinds. This wish is revealed in the training needs that valorize the voice, the listening and the dialogue, with the students, in interactions, whose analysis to that show about the students project about relation to the pragmatic questions of the speech. The search was evident that is in the ideological of the sign – that where, the object is inexhaustible – that is the questions the students. In this case, pedagogic interventions founded in the Backtin’s categories of the chronotope, of the thematization and of the dialogism show educationally powerful to contribute with the enlargement of the discursive competence of the students.
A escrita de textos de gêneros retóricos é essencial às práticas letradas de uma sociedade democrática. Diante disso, torna-se importante às mediações formativas escolares o desenvolvimento de intervenções pedagógicas voltadas para o ensino desses gêneros. Esta pesquisa constrói eventos de letramento que visam ao ensino da escrita como prática social. O objetivo geral de pesquisa é investigar os fatores que contribuem para a ampliação das competências discursivas dos educandos, por meio do trabalho com gêneros retóricos que realizam o tipo dissertativo. Para alcançar esse objetivo, realizamos uma pesquisa qualitativa aplicada, cujas ações se apoiaram em uma posição bakhtiniana, em que a linguagem deve ser considerada por seu caráter ideológico e pelos usos e formas reais de comunicação construídas e utilizadas pelos falantes na interação verbal (BAKHTIN, 1992). Recorremos especialmente às teorias de gêneros (BAKHTIN, 1997; BAWARSHI e REIFF, 2013), da argumentação e da nova retórica (PERELMAN e OLBRECHTS-TYTECA, 2005; BAZERMAN, 2015a) e do letramento e multiletramento (GOULART, 2011; ROJO, 2013; RIOS, 2014); e empregamos discussões relativas ao pensamento crítico (GIROUX, 1997; NOBRE, 2004; CHAUÍ, 1982). Ancorados nessas teorias e na reflexão construída por meio da práxis educacional, pudemos evidenciar que – conforme posição de Geraldi (2010) e Bawarshi e Reiff (2013) – o conhecimento das formas tipificadas do gênero não se transforma, necessariamente, em domínio da produção escrita desse gênero. Constatamos, ainda, que há uma profunda vontade dos estudantes de se tornarem proficientes na escrita de gêneros retóricos. Essa vontade é revelada por meio de mediações formativas que valorizem a voz, a escuta e o diálogo com os educandos, em interações cuja análise esclarece sobre aquilo que os estudantes projetam em relação às questões pragmáticas do discurso. A pesquisa evidenciou que é no aspecto ideológico do signo – ali onde o objeto é inesgotável – que se situam as dúvidas dos educandos. Nesse caso, intervenções pedagógicas baseadas nas categorias bakhtinianas do cronotopo, da tematização e do dialogismo se mostraram pedagogicamente poderosas para contribuir com a ampliação da competência discursiva dos estudantes.
