Nowak-Musial, Magdalena. "Evaluation of cellular processes and identification of candidate genes critical to corneal epithelial development." Thesis, Cardiff University, 2009. http://orca.cf.ac.uk/54984/.

Abstract:
The overall aim of this study was to determine factors and mechanisms that underlie the regulation of epithelial patterning and homeostasis during corneal development. Histological staining was performed in chick corneas, embryonic day (ED) 4 to 21, to evaluate changes in the overall epithelial cell morphology, in particular cell shape, cell size and the number of epithelial cell layers. Epithelial differentiation patterns were identified in frozen sections of chicken corneas after immunolocalisation of pan-cytokeratins (Pan-CK) and cytokeratin 3 (CK3). Proliferating Cell Nuclear Antigen (PCNA) and caspase 3 (active) immunolocalisation studies, as well as, TUNEL-labelling (Terminal deoxynucleotidyl transferase dUTP-biotin nick-end labelling) were performed to assess temporal and spatial localisation of cell proliferation and death in the developing corneal epithelium respectively. The expression of PCNA and CK3 were later confirmed by immunoblotting. Total RNA was isolated from epithelia at selected developmental time points and collected for microarray analysis. Gene expression profiles were analysed by appropriate mathematical methods. The sensitivity of arrays in producing data trends was validated by quantitative RT-PCR. Histological findings included changes in stratification an increase in the number of cell layers, change in cell morphology. In this study it was demonstrated that after becoming two layered by ED4, the epithelium underwent further stratification to form intermediate cell layers at about EDM. These changes were accompanied by changes in cell shape commencing at ED10. Cell proliferation appeared high throughout corneal development, with peak proliferation between ED12 and ED14 in the limbal, peripheral and central epithelium, respectively, thereafter the level of proliferation decreased. The above coincided with changes in epithelial morphology (stratification) and changes in expression of cytokeratin (CK) epithelial markers. The appearance of pan-CK labelling was first observed at ED10 and the presence of CK3 immunolabelling appeared in epithelial cells at ED12. TUNEL-labelling and caspase 3 (active) immunolocalisation demonstrated only few TUNEL-positive cells, mostly restricted in the limbal region of the corneal epithelium, in the mid and later developmental stages. Microarray analyses identified gene families and their members (including these involved in stem cell biology) likely to be relevant in the regulation of homeostasis during corneal epithelial development, as well as, differentially expressed genes that reveal changes in biological processes due to the change in time. RT-qPCR confirmed the differential expression patterns of seven genes of interest following analysis of microarray data. Patterns of cell proliferation and differentiation showed changes during the development of the corneal epithelium that reflect the interaction of a complex network of mitogenic, apoptotic and differentiation agents. The changes in gene expression profiles, detected by the microarray analyses, were consistent with the phenotypic changes in the developing chick corneal epithelium. The microarray data provided the first study to present a good overall picture of genes expression in the developing chick corneal epithelium.

Abstract:

Down syndrome is the most frequent cause of mental retardation affecting millions of people worldwide and results from full or partial trisomy of chromosome 21 (HC21). Rare cases of partial trisomy allowed the identification of a small region in HC21 common to all carriers called Down Syndrome Critical Region. The genes DCRA and DSCR8, mapped to this region, encode two proteins of unknown function. With the aim of contributing to a better characterization of these proteins, DSCR8 was subcloned and expressed in fusion with thiorredoxin in Escherichia coli Rosetta (DE3). Recombinant Trx-DSCR8, observed in the soluble fraction, was subjected to the initial purification assays by affinity chromatography in a Ni-NTA column. The gene DCRA was cloned and expressed in fusion with the proteins thiorredoxin and GFP allowing the partial purification of the protein and the achievement of crystallization and immunological assays. The amino acid sequence of DCRA showed 57% and 49% of identity to a protein of unknown function from Anopheles gambiae and Drosophila melanogaster respectively. Also, in silico analyses revealed that DCRA contains a putative Vps26 domain. Subcellular localization of DCRA in fusion with GFP was observed preferentially in the cytoplasm of the cell lines tested. These results contribute to a better characterization of DCRA and DSCR8 and open up possibilities towards the understanding of the cellular role of these proteins and their relationship with the Down syndrome.

Greenlees, Rebecca. "Characterisation of a gene critical in eye and lens development." Thesis, The University of Sydney, 2014. http://hdl.handle.net/2123/12051.

Full text
Abstract:
Visual impairment or blindness caused by vision disorders affect over 500,000 Australians, with approximately one-third caused by genetic eye disorders. Previous work identified a cataract and anterior segment abnormality patient with a de novo balanced chromosomal translocation transecting the 5’ UTR of a novel candidate disease gene. An expression vector containing the candidate gene and a GFP tag revealed its subcellular localisation to the plasma membrane domain of two epithelial cell lines. Colocalisation experiments showed colocalisation of the candidate disease gene with proteins of adherens junctions, tight junctions and cytoskeletal F-actin in Caco2 cells. Caco2 cells transfected with a mutation in the candidate disease gene caused clustering of F-actin stress fibres in the basal regions of the cell, suggesting the presence of functional actin-binding domains in this candidate disease gene. Stable knockdown cells when cultured in a 3D-cell assay produced abnormal cyst structures characterised by the presence of additional ectopic lumens, multilayering of cells and misexpression of a polarity marker. Knockdown cells generated a significantly greater number of abnormal cysts. The phenotype exhibited by knockdown cell lines is suggestive of defects affecting the actin cytoskeleton and polarity. To further investigate the role of the candidate disease gene in eye development, gene trap loss-of-function mice were generated and examined. Homozygous loss-of-function mice developed congenital cataracts and demonstrated structural lens abnormalities and abnormalities in the expression of cell adhesion and polarity proteins in the developing lens. The lens abnormalities exhibited by the mutant mice, along with the defects observed in the mutant and knockdown cell assays suggests that this novel candidate disease gene has an important role in the regulation of the actin cytoskeleton and cellular polarity in epithelial cells and lens development.

Abstract:
Recent national and international conservation legislation has led to an increased focus on ‘prioritised species lists’ for the allocation of conservation resources. However, prioritised species lists become problematic when there are difficulties in species delimitation.  One particularly complex group accounting for the majority of these problems is Euphrasia. The goal of this thesis is to use molecular markers to evaluate taxon limits and evolutionary processes in British Euphrasia to clarify the most appropriate approach for conservation. The results show that the major reproductive barrier in the group corresponds to ploidy level, with AFLP and chloroplast data both significantly differentiated between diploid and tetraploid species, although occasional gene flow via inter-ploidal hybridisation appears to contribute towards diversification of the diploid group. The genetic data support the current species level taxonomy for the diploid but not the tetraploid species, where a considerable overlap between taxa was detected. The chloroplast data detected four discrete lineages, the distribution of these among species suggest at least three allopolyploid events in the formation of the tetraploid taxa, within which distinct ecological groups occur. Variation in the breeding system was detected at the intra- and inter-specific levels and estimates of the inbreeding coefficient showed a strong correlation with flower size and support the importance of multiple shifts in breeding system as contributing towards the overall diversification within the group. Given the lack of clear species limits in the tetraploid group and the dynamic nature of Euphrasia evolution, a change from a species- to process-based conservation approach is recommended. This charge include recognising the importance of progenitor taxa, and ecological and morphological diversity, and a decrease in the importance presently given to individual named endemic taxa.

Abstract:
This dissertation wants to understand the church history regarding the divide which resulted in the so called fundamentalist and the liberal movements in the church being birthed in identifying the reasons for the conflict between the aforementioned groups. It then presents principles of conflict transformation and management in order to understand and to help transform and manage potential confrontational situations between the aforementioned groups effectively. It further propose alternative interpretations to potential conflict-generating Bible texts by extrapolating new information from those texts in order to stir the potential of birthing a second naïve love for the discredited texts by offering new meanings to those same texts. Unfortunately, this objective can’t effectively be measured within the scope of this dissertation, but will nevertheless remain as an objective of hope.

Abstract:
Microarray technology enables simultaneous gene expression level monitoring for thousands of genes. While this technology has now been recognized as a powerful and cost-effective tool for large-scale analysis, the many systematic sources of experimental variations introduce inherent errors in the extracted data. Data is gathered by processing scanned images of microarray slides. Therefore robust image processing is particularly important and has a large impact on downstream analysis. The processing of the scanned images can be subdivided in three phases: gridding, segmentation and data extraction. To measure the gene expression levels, the processing of cDNA microarray images must overcome a large set of issues in these three phases that motivates this study. This study presents automatic gridding methods and compares their performances. Two segmentation techniques already used, the Seeded Region Growing Algorithm and the Mann-Whitney Test, are examined. We present limitations of these techniques. Finally, we studied the data extraction method used in MicroArray Suite (MS), a microarray analysis software, via synthetic images and explain its intricacies.

Abstract:
There is plenty of fiction written for children with an educational starting point whose intention is to challenge traditional gender patterns. However, there are few studies on how children actually perceive this norm-critical fiction, which is important for the use of literature as a didactic tool in teaching. As fiction forms a large part of the work in preschool class, this study aims to explore how student perceptions of gender can be made visible through critical literacy-inspired preschool work. The norm-critical fairy tale Rödluvan (Jansson & Jensfelt, 2018) is the fiction that forms the basis of the study and the following issues have been formulated: • How do the students name the characters' gender in Rödluvan (Jansson & Jensfelt, 2018)? • How do the students' ways of naming the characters gender differ, depending on whether they participated in a joint conversation about the text or not? The study is qualitative and the data collection is done through semi-structured interviews with 20 students in preschool class. The analysis of the material is based on gender theory and the concept of didactic potential, in accordance with Alkestrand's (2017) definition. The conclusions that can be drawn from the results of the study are that the participating students had difficulty understanding the gender of the fairy characters as described in the story of the character in question, portrayed in a norm-critical manner. Based on the results of the study, it can also be stated that there is a marked difference in the students' ways of perceiving the gender of the norm-critical characters depending on whether they participated in a joint conversation about the text or not.

Abstract:
This case study deals with conceptions and attitudes about gender in a preschool context and the study includes both preschool pedagogues and parents. The frame work of the study is made up by the Social constructive perspective and the study includes two parts. The first part includes interviews analyzed with Critical Discourse analysis and the second part includes an Ethnographical perspective. The reason to include two part studies is to draw attention to the difficulties in the working processes that surrounds the enforcement of a common ground for gender equality work in a preschool context. The study shows that the informants’ conceptions and attitudes about the word gender differ and this aspect effects the implementation of gender equality work at the examined preschool.

Abstract:
The aim with this thesis was to investigate how the Swedish newspaper Aftonbladet construct and reproduce imaginations of gender and gender order. An important premise for our work is the agenda theory, which states that media has great impact on what is on their audience mind. We chose to investigate how Aftonbladet wrote about two Swedish footballers, Lotta Schelin and Zlatan Ibrahimovic, during a European championship with each players national team. By using a method of critical discourse analysis based on Teun van Dijks idea and with gender glasses, we analysed eight articles and two chronicles regarding each player. Our results show that Aftonbladet construct and reproduce gender and gender order by its different ways of portraying Lotta Schelin and Zlatan Ibrahimovic. Articles regarding Lotta Schelin tend to be more audience oriented, focus more on her and her teammates emotions and more often focus on things outside her profession as a footballer. On a contrary, articles regarding Zlatan Ibrahimovic tend to upgrade masculinity, power and more often highlights his performances on the pitch. He is also portrayed as a person of much greater status than Lotta Schelin.

Abstract:

This dissertation aims to trace the origin and evolution of Massa’s thought. The latter is also placed within the current debate on learning theories, and further developed thanks to Fook’s critical reflection and Latour’s actor-network theory. Firstly, Massa’s writings are discussed. The influence of Marxism, Althusser, Foucault, and psychoanalysis on Massa’s socio-materialistic understanding of the learning process is investigated. Particular attention is given to the Foucauldian notions of ‘micro-physic of power’, ‘apparatus’, and ‘clinic’. On that basis, Massa’s ‘educational apparatus’ and ‘Clinica della formazione’ [Foucauldian clinic applied to the learning process] are debated, together with their mutual relationships. Educational apparatus is proved to be the core of Massa’s thought. Thanks to this notion, Massa simultaneously granted agency to materiality, and depicted education as a complex phenomenon underpinned by both human and non-human elements in interactions between each other. The ‘educational apparatus’ also acts as a theoretical grid to be projected on the learning process under scrutiny in order to reveal its hidden complex structure. Clinica della formazione consists in the set of techniques which makes this procedure possible; hence, it is Foucauldian. Clinica della formazione is presented and placed within the Italian educational debate of the 80’s-90’s. Moreover, its goals and methods are debated. Clinica della formazione is proved to be anthropocentric: it does not grant agency to non-human and it is based on a reflective process. In order to highlight the gap between Clinica della formazione and Massa’s original socio-materialistic epistemological background, the notions of reflection and reflexion are discussed, together with the works of Dewey, Kolb, Schön, Mezirow, Sheppard, Brookfield, and Fook. In order to further develop Clinica della formazione as a consultancy and supervision methodology, a contamination between Clinica della Formazione and Fook’s critical reflection is suggested. Latour’s actor-network theory (ANT) is identified as the most suitable research methodology to investigate the learning process according to Massa’s original materialistic epistemology. History and key aspects of ANT are presented; its position within Science and Technology Studies debated. The impact of ANT on educational research is discussed. Massa’s work is lastly compared to Latour’s one: the notion of ‘educational apparatus’ is compared to Latour’s notion of ‘network’ and Massa’s learning theory is reread taking into consideration Latour’s concepts of ‘translation’, ‘mediation’, and ‘assemblage’. Finally, Massa’s ‘theatrical deixis’ is discussed. This notion clearly suggests that Massa was moving away from Clinica della formazione methodological apparatus to fully embrace his original materialistic background. It also shows Massa’s attempt to plan and implement a consultancy process in line with the socio-material understanding of the learning process.

Abstract:

Abstract:
This dissertation constitutes a critical examination of Swedish spatial planning for safety. Spatial planning for safety rests on a number of assumptions about the desired order of the world. These assumptions appear as given and unproblematic, making the formulation of alternatives appear unnecessary. This dissertation provides an account of how, and on what basis a spatial planning problem such as ‘fear and insecurity’ is formulated and acted upon. It is an account of how and what conceptions of knowledge operate to legitimise ideological representations of spatial planning problems. And furthermore, what these ideological representations of spatial planning problems substantially entail, so as to allow for a political spatial planning practice that formulates and deliberates alternatives. This is carried out by analysing assumptions of public life and knowledge within Swedish spatial planning for safety.  This dissertation finds that Swedish spatial planning for safety constitutes ‘certainty’ as a hegemonic criterion for participating in public life, which operates to limit the articulation of alternative discourses in spatial planning for safety. The desired for safe public life is organised based on visual certainty, where the urban fabric should be configured in such ways as to allow for stereotypical visual identifications of one another. Such a public life reflects an individualised practice, where perceptions of fear should be governed by individuals themselves, by independently assessing situations and environments in terms of risks. This individualised conduct is coupled with the fostering of active subjects, which encompasses being engaged in the local residential areas as well as in one another. Such substantial content of ‘planning for safety’ brings about tensions in terms of its ideological legitimating basis, by moving from principles of ‘rights’, where the individual constitutes the first ethical planning subject, to unitary principles of ‘collective values’, in which the ‘community’ constitutes the first ethical planning subject. These presuppositions are further enabled through the ways in which knowledge is conceptualised in spatial planning. This dissertation argues that a hegemonic instrumental emphasis on knowledge in spatial planning prevails. Having such a hegemonic emphasis on knowledge has the implication that even though spatial planning adopts different assumptions, or moves between alternative assumptions of knowledge, the knowledge becomes meaningful only in its instrumental implementation. The instrumental emphasis on knowledge should be regarded in light of the rational and goal-oriented nature of project-based planning, which constitutes a logic that constrains the emphasis on knowledge in spatial planning. This dissertation argues further that if spatial planning should be considered a political practice that debates its goals and values, a politicisation of the emphasis on knowledge in spatial planning is imperative.

Full text
Abstract:
The purpose of this study is to examine media’s depiction of women’s depression, and what consequences this may possibly have from a gender perspective. This study has a certain focus that consists of the group women and on how media constructs and the describes the image of the depressed woman. The study is based on the critical discourse analysis. Our empirical material consists of news articles by the two Swedish evening newspapers Aftonbladet and Expressen, and the newspaper Svenska Dagbladet. The study is limited to articles between the period 2013-2020. Norman Fairclough’s three-dimensional model is used for analysis of the study’s empirical material. At first, we used textual analysis and discursive practice to study our empirical material. Thereby we identified media’s depiction of women’s depression. Then we used the third dimension, social practice, to further examine what consequences media’s depiction possibly may have. We used previous research regarding the increased mediatization, gender and depression as well as media in relation to gender. The theoretical framework of the study consists of gender theory and social constructivism. Our study shows that media creates and maintains discourses on women’s depression. Women are depicted by media as a high-risk group concerning the disease depression. Media’s depiction of women’s depression results in an individualized health responsibility. The over-representation of women in media’s discourses on depression is described as female symptomatology, which is explained through social constructivism and gender theory. Media’s production and maintenance of discourses results in consequences as continued repression of women. It also results in an overdiagnosis of women’s depression, which leads to men’s emotional distress being overlooked. Media’s power through discourses has a strong impact on people’s social conditions and perceptions of the world. Media’s depictions of the depressed women lead to maintenance of structures and gender norms, which also affects clinical work.

Munoz, Marcia Medicine UNSW. "The EDD protein is a critical mediator in the DNA damage response." Awarded by:University of New South Wales. Medicine, 2006. http://handle.unsw.edu.au/1959.4/25977.

Full text
Abstract:
An intact cellular response to DNA damage is important for the maintenance of genomic stability and tumour prevention. EDD, the human orthologue of Drosophila melanogaster ???hyperplastic discs???, is over-expressed or mutated in a number of common human cancers. EDD is a progestin regulated gene that encodes an E3 ubiquitin ligase involved in cell communication and cell adhesion, and although it has also been implicated in the DNA damage response through its association with DNA damage proteins, a definitive role has yet to be demonstrated. The work presented herein shows that EDD is necessary for an adequate cellular response to double-strand DNA breaks. Cells depleted of EDD exhibit reduced survival, radio-resistant DNA synthesis and failure to maintain G2/M arrest following DNA damage induced by phleomycin exposure. Furthermore, EDD-depleted cells display impaired activating phosphorylation and kinase activity of the checkpoint kinase CHK2 after DNA damage. These effects appear to be largely modulated through a phospho-dependent interaction involving the CHK2 FHA domain and a region of EDD spanning a number of putative FHA-binding threonines. These results identify EDD as a novel mediator in DNA damage signal transduction via CHK2 and emphasise the potential importance of EDD in cancer.

Abstract:
The problem I chose to base my project on is the fact that a big portion of the products we use daily are marketed against either men or women. The result of that is an exclusion of the people who do not fit inside the norm, which states that there are only two genders. My purpose during the project was to investigate this problem and in the end also criticize it in a suitable way. Later on, my focus was shifted towards a specific category of products - menstrual hygiene products. More specifically their packaging. They are marketed only towards women and exclude all who are not cis-women. The subject of menstruation is also a loaded one and at the moment frequently discussed within certain circles. Most people seem to have a lot of feelings and opinions regarding the subject, which will hopefully get my project into the public eye. My process started in collecting a broad range of facts and theories surrounding the subject, which the remaining parts of my project is based on. The information about norms, stereotypes, gender, packaging, transpersons, menstruation and menstrual hygiene products was something I could always go back to whenever I needed help in decision making during the project. The contact I had with the consumers through the surveys were an important part in building my understanding of what creates attention and feelings, which has been valuable in working towards critical design. The packaging is created as a contrast against the packaging for menstrual hygiene products existing today. The period is allowed to take up space in the form of illustrations of the menstrual product as well as the color red, which symbolizes menstrual blood. This is also combined with clear plastic on two sides which show the containing products. The font used on the majority of the packaging is a combination of hard shapes, often considered masculine, and thin round shapes, often considered feminine. Together they create a gender neutral appearance. The name of the company ”har du mens eller?” is chosen after a phrase commonly used by men to disparage women’s feelings and opinions. In this case, it is written in a typically feminine font which is very common in this category of products. With the different parts of the packaging, my hope is to rise attention and bring awareness to how the products surrounding us are marketed and how this affect us.

Abstract:
The biotechnology industry has recently been demanding nanoparticulate products (20-200 nm) such as viruses, plasmids, virus-like particles and drug delivery assemblies. These products are mainly used as gene delivery systems in gene therapy protocols. Adenoviral vectors {Ad) are the most exploited gene therapy vectors in industry worldwide. The :ma.nu.fu.cture of Ad fuces several problems including {i) small volumes of product per batch of cell culture, {ii) inaccurate, time consuming and multifuctor-dependant methods for infectivity determination and (iii) downstream processing. The downstream processing requires methods to purify infective particles :from the contaminants including non-infective particles, partially assembled particles, :free adenoviral components, host cell components and medium components. The solvent extraction method, aqueous two-phase systems {ATPS) and selective chromatography can potentially circumvent such downstream processing problems. ATPS has been applied to the purification of soluble macromolecular products. However, the recovery of nanoparticles represents a practical constraint due to their partition in the interphase. Selective chromatography has been also a method for high performance purification of bioproducts. However, in the context of nanoparticulates, product and contaminants adsorb and desorb (i.e. copurify) under similar mobile phase conditions due to sheared chemistry. In this study, infective Ad were concentrated in the top phase of a polymer-salt ATPS. Additionally, the potential ATPS miniaturisation for scouting experiments was evaluated. The infective Ad were also partially purified exploiting a fluidised bed. In this process, an anionexchange commercial adsorbent laminated with electrophoretic grade neutral agarose was utilised. The porosity of the pellicle was designed to exclude Ad yet fucilitate diffusion of macromolecular components to adsorption sites within the interior of the adsorbent core (subtractive adsorption). Furthermore, a method for monitoring infective Ad with rapid and easy handling by means of flow cytometric analysis was developed. In order to perform the experiments, the Ad production was scaled-up to a 2 L bioreactor. The recovery processes evaluated in this study and conclusion drown will contribute to the recovery of Ad but also could be applied to recover other nanoparticulate bioproducts

Abstract:
Zara Larsson and the male haters. A critical discourse analysis of the medias depiction of the singer Zara Larsson’s commentary on Bråvallafestivalen 2015 Zara Larsson, born in 1997, is a famous, young swedish singer. Besides her career in music she is also famous for her feminist thoughts in social media. In 2015, she critizised the swedish festival Bråvallafestivalen for its lack of female artists. This critizism was first published in the swedish newspaper Dagens Nyheter and afterwards there was a huge reaction in social media where lots of people critizised and threatened Zara Larsson because of her thougths. Therefore, the purpose of this study is to explore how swedish newspapers reports about this situation and how Zara Larsson is portrayed, from a genderperspective. We are using a thematic analysis and a critical discourse analysis as a method. The analysis is based on articles and opinion journalism in swedish newspapers from all over the country. Theories we have used for the study is gendersystems and ideology. The main conclusion of this study is that the debate ended up to be very polarized, and that the supporting side of Zara Larsson dominated the debate, while the opposition tended to be some sort of hopeless contenders. We could also se that the main ideology was feminism, both visible and hidden in the texts. Keywords: Critical discourse analysis, Thematic analysis, gender, Zara Larsson, Bråvalla festival.

Abstract:
This thesis examines if there is any differences in how gender is produced in the Administrative Court’s application of care of young persons (special provisions) act. To do that, we’ve studied 40 judgements that all has the requirement “social destructive behavior”. A social destructive behavior means that the young’s behavior is considered to differ from the basic norms in the society. The deviant behavior may be that the young person socialize in inappropriate environments or through the behavior exposes the young’s health and development. The judgements we’ve been studying are from 2017 and 1996, because we want to see if the discourse has possibly changed over the past twenty years. We’ve used a qualitative method, the critical discursive analyze, to visualize power structures regarding the construction of gender. The thesis is based on the theories social constructionism, discourse analysis and gender theory. By using these method, theories and data, we can note that differences are made between gender and sexuality.

Abstract:

Abstract:

In this essay I examine how the norm-critical material ”BRYT! ett metodmaterial om normer i allmänhet och heteronormen i synnerhet” is used by Hyresgästföreningen in Stockholm in their education of personnel in the perspective of equalisation. I’ve mostly been interested in how the personnel who takes part in workshops about the material react and reflect in relation to the purpose of the material. How do they discuss about norms and the connection to discrimination and how do they look upon the power and responsibility that comes with power in relation to norms?  Earlier research in norm critical pedagogy and the critic of tolerans pedagogy perspective helps me to analyse how critical pedagogy stands above non-critical pedagogy in many perspectives.

By observing workshops and by interviewing personnels taking part in these workshops I’ve examined how they react on both the practical and theoretical parts of the material.

To summarise my examination I find the personnel who is educated in working with the material discover new experiences in knowledge and on how norms and discrimination functions. They explain that they have experienced knowledge about the connection between norms and discrimination and thereby how oppression could be counteracted and prevented.

Every year, approximately 17 500 drunk drivers are sued in Sweden. Of these, only 10 percent are women. The women are therefore considered a minority. In this study, I have taken interest in how the female drunk driver is constructed implemented by the probation service. By using critical discourse analysis I have found that the woman in these investigations is contributed the role of a deviant in two senses. She deviates because she is a woman who has committed a criminal deed and also because she often has problems with alcohol as well. I have found that the woman, because of these deviations, is depicted as a victim, both for the situation she is in and for her own actions.

This thesis presents a critical discourse analysis of how the sociology of gender is constructed in the sociology syllabus and two textbooks aimed for use at upper secondary school level. I use Bernstein’s theory of framing and classification to understand how power over knowledge is distributed through this construction. I take as my starting point that sociology, as framed in the syllabus, allocates a lot of power over knowledge to individual teachers. I outline the short history of sociology as a Swedish school subject and problematise this based on the fact that the majority of sociology teachers have limited education in sociology. I argue further that sociology as a discipline is characterised by an ambiguous canon and lack of pedagogic traditions. This increases the demands on sociology teachers. Based on previous research, I identify the sociology of gender as a potentially contentious area that warrants investigation.  Critical Discourse Analysis  (herein - CDA) takes as its starting point that discourses are a vehicle of ideology, and that ideology is capable of oppression or marginalization. Through critical discourse analysis it can be revealed that the construction of knowledge of gender marginalises queer voices and sociological perspectives on gender from outside the global North whilst simultaenously segregating feminist theory from mainstream sociology. “Mainstream sociology” in this context can be taken to include theories and perspectives which are used to understand society and social processes as a whole, such as the class, crime and deviance, the state and media. The construction of gender also selectively draws upon dominant socio-cultural discourses on gender, such as gender equality politics and sociological research. Violence against women, and masculinity are two topics identified as important in gender equality politics, for example, but are not part of the construction of the knowledge on gender. This is in itself not surprising, the analysed course is only 100 points in total and cannot possibly cover everything, but it does raise questions about which perspectives are included and which are excluded.   Importantly, the purpose of this thesis is not to discredit the knowledge of gender as constructed by the syllabus and textbooks, and to supplant it with other perspectives. This would be unconscionably arrogant and simply replacing one set of ideologies with other ideologies. This would be  counterproductive to the aims of CDA. The purpose is rather to reveal the ideologies behind the discourse and enable an informed discussion from multiple perspectives. I conclude this thesis by arguing for the benefits of introducing norm critical pedagogy into sociology teaching as a means of countering bias in knowledge creation and selection, and as a pedagogical tool in a  field where no identifiable pedagogic approach exists as yet.

Abstract:
Swedish school is obliged to actively work with questions concerning gender, equality, rights and opportunities. Furthermore, school shall give the students tools to critically review gender patterns and understand how gender patterns can constrain people’s living conditions (Skolverket, 2019). In school the students read fiction, which in turn contributes to how norms are produced in different ways. Through didactic efforts through critical literacy inspired working processes fiction can be used as a foundation for work with values. It is of thus of relevance how children perceive the norms or norm-breaking aspects as they are produced in fiction. The purpose of the study is therefore to contribute knowledge about students’ different ways of reasoning and relating to a norm-breaking story through a Critical literacy-inspired didactic perspective. As a basis for the study is the norm-breaking fairytale about Cinderella (Jansson & Jensfelt, 2018). The study aims to address the following questions: ·       Which norm-breaking aspects do the students retell in a dialogue about the norm-breaking fairytale? ·       Which aspects of a norm-breaking fairytale can the students relate to from their own reality? ·       How are the students’ reasoning affected by the norm-breaking fairytale after a literacy talk?  The study is qualitative and is built on semi-structured interviews with 12 students in year 2 and 3. The result of the study shows which norm-breaking aspects the students’ reason about. Furthermore, the results show that a literacy talk seems to have an impact on the students’ reasoning. The empirical material is analyzed through gender theory together with Alkestrand’s (2017) definition of didactic potential.

Abstract:
My research explores the contingency of key discourses of masculinity, race and violence as they are filtered through particular film genres in which it is the very masculinist dimension of their cultural narratives which is emphasized. To that end the two genres which I focus upon are the 'spaghetti' western and the 'gangster' film. I explore the formative role of cultural production in the circulation and critique of masculinist, or in the case of the screen 'outlaw' or 'gangster', hypermasculinist discourses, particularly as they intersect with issues of racial or ethnic difference. This in tum raises important questions about the transnational component of the urban political economy which is the favoured stomping ground of both the screen, and reallife, gangster. Subsequently the research tests the notion of a global register against which the iconographic figure of the male outlaw is constructed and the implications of this for broader cultural mythologies of race, nation and frontier, for instance as a prototypical narrative of American, English or Indian modernity. My argument is that film, as a culturally promiscuous yet tactical resource, is both the instance and the critique of 'the public enemy' or of the violence of our own self-constitution. It (film) produces and sustains the media gangster as a solipsistic medium through which burgeoning, and increasingly violent, neuroses about the role of masculinity are filtered. By engaging the psychotic underbelly of both visual culture and the foundational myths it indexes, my research synthesises otherwise discrete anxieties around race, desire and violence. It makes the case for how the outsider art of gangster films or spaghetti westerns recalibrates the level of violence we are prepared to live with. It ultimately reveals how it is outlaws in the guise of a domestic citizenry who convert life into myth and image by projecting society's fears back to it as style. Weare the frontier and psychosis is our signature.

Abstract:
This thesis aims to describe the different historical stages through which the narrative structure of the character of Perceval evolved during the first two centuries of its genesis in Romance and European literature. The character had a significant success from the Middle Ages (Chrétien de Troyes) up to the modern and contemporary age. The philological study about the evolution of the character within the textual galaxy and the forms of his tradition allows us to outline a composite picture, in which it is possible to trace the genesis and development of important historical-literary phenomena, destined to influence European and Western literature in the long term. A second part is instead dedicated to the study of the literary and narrative structure of the character that can be traced within the reading of the different works that hand down the versions of the story of Perceval. Finally, the research will focus on a case study capable of illuminating the persistence of the structures identified above: the Prophecies de Merlin, an arthurian romance from the second half of the 13th century, are in fact an expression of one of the many ways in which Perceval’s life story is staged in texts that do not directly focus on his biography. Within the tradition of the Prophecies de Merlin, the prophet of the Arthurian kingdom whose numerous prophecies are collected, the case of the Livre d’Helias et Perceval is emblematic. The Livre d’Helias et Perceval from the Prophecies de Merlin, accompanied by a philological study that includes a critical edition, a complete study of the tradition and linguistic analysis, is thus proposed as an ideal case study for ana-lysing the fortune and evolution of the character of Perceval in the Romance and European Middle Ages.

Abstract:
The study aims to examine how three books written for teenagers and young adults address topics that are generally considered as deviant to social norms in order to conclude if they can contribute to a norm critical pedagogy in school. Norm critical pedagogy is foremost based on gender- and queer theory and tends to discuss how social norms and power structures are constructed and reproduced in different social contexts. To work in a norm critical fashion the teacher must be aware of the present norms and work to make the pupils conscious of them. Thereby the focus is on existing norms, rather on the deviances. To use norm critical pedagogy correct it must be incorporated in all subjects in school. The books that have been analysed in this essay are Syskonkärlek by Katarina von Bredow, Henrietta är min hemlighet by Maja Hjertzell and Tusen gånger starkare by Christina Herrström. This study shows that all books include and discuss subjects that are deviant to social norms, such as: non-conformal gender roles, homosexuality and incest. Tusen gånger starkare and Henrietta är min hemlighet are suitable to teach in upper school and upper secondary school, because they discuss subjects and themes that pupils and students in that age can relate to and they are written in ways that open up to discussions.  However, all books are not suitable to use in a norm critical pedagogy. Syskonkärlek’s theme is the incestuous relationship between a brother and a sister. Incestuous love is not only deviant to social norms, it is also against the law. To teach that book in the purpose to make the pupils aware of norms and start thinking critical about them, can have the opposite effect, though incest is seen as wrong in so many ways.

The writing of texts of rhetoric genres is essential to the literate practices in one democratic society. In this case, is very important to the school formation mediation the development of pedagogical interferences about the teaching this genres. This search constructs events of written that objective to the teaching of the written how social practice. The general objective this search is to investigate the factors that contribute to amplification of the discourses competences of the students, in works with rhetoric kinds that realize the type of dissertation. For to obtain this objective, we realized a search, whose actions supported themselves in Bakhtin’s position, in that the language has been considered in your conception character and about uses the speaking in the verbal complementation (BAKHTIN, 1992), (BAKHTIN, 1997; BAWARSHI and REIFF, 2013), of the argumentation and the new rhetoric (PERELMAN and OLBRECHTS-TYTECA, 2005; BAZERMAN, 2015a) and of the writing and various writing (GOULART, 2011; ROJO, 2013; RIOS, 2014); and we use discussions about the critic think (GIROUX, 1997; NOBRE, 2004; CHAUÍ, 1982). Position of Geraldi (2010) and Bawarshi and Reiff (2013), and position of Geraldi (2010), and Bawarshi and Reiff (2013), the know of the model forms of then kinds don’t transforms, necessarily, in domain of the writing production of this kinds. We verify that, still, there’s a deep wish from the students become proficient in the written of this rhetoric kinds. This wish is revealed in the training needs that valorize the voice, the listening and the dialogue, with the students, in interactions, whose analysis to that show about the students project about relation to the pragmatic questions of the speech. The search was evident that is in the ideological of the sign – that where, the object is inexhaustible – that is the questions the students. In this case, pedagogic interventions founded in the Backtin’s categories of the chronotope, of the thematization and of the dialogism show educationally powerful to contribute with the enlargement of the discursive competence of the students.
