Abstract:
Abstract We sought to determine whether various inhibitors of Oxidative Phosphorylation (OxPhos) could restore tumor infiltrating T cell functionality and longevity while compromising metabolic fitness of tumor cells. Using a combination of in vitro metabolic profiling with the Seahorse XFe96 Analyzer and in vivo tumor immunotherapy in the immunocompetent TRAMP-C2 prostate adenocarcinoma model, we determined the differential impact of multiple OxPhos inhibitors on both tumor immune metabolic fitness and functional capacity. Impact on the tumor immune microenvironment of TRAMP-C2 tumors was assessed through high parameter flow cytometry on a BD X-30. While OxPhos inhibition compromised initial T cell activation, we found that two of the inhibitors increased the glucose uptake, proliferation, and activation status of effector T cells. In contrast, the same two inhibitors decreased the mitochondrial fitness, proliferation and OxPhos metabolic capacity of TRAMP-C2 tumor cells themselves. To determine whether this selective inhibition of tumor versus T cell metabolic function could improve immunotherapy responses in vivo, we implanted TRAMP-C2 tumors and treated them with these OxPhos inhibitors with or without concomitant immune checkpoint blockade (ICB). The combination of ICB and OxPhos inhibition achieved highly significant and durable rates of tumor control and regression suggesting that these approaches are potentially synergistic. Analysis of the tumor microenvironment has identified the cellular mechanisms underlying these therapeutic effects which may provide useful biomarkers as this novel combination is translated to the clinic. Contemporary “flexible capitalism” requires novel forms of legal regulation. In this vein, Joshua Cohen, Michael Dorf, Archon Fung, and Charles Sabel have developed a provocative set of proposals for a new mode of regulatory law, what they describe as “democratic experimentalism” or, alternately, “directly deliberative polyarchy.” Their proposal are criticized: they not only fail to take traditional liberal democratic rule of law virtues seriously enough, but it remains unclear whether they can effectively tame and humanize capitalism. Instead, some evidence suggests that their proposals simply amount to a normatively problematic synchronization of the legal system with contemporary high-speed capitalism.

Nuclear War, edited by Fox and Groarke, is one of five recent anthologies containing new essays by philosophers on the subject of nuclear war. The Blake and Pole volumes, containing essays mainly by British philosophers, are distinguished by unrelenting and comprehensive opposition to British and American policy, and by the fame of the contributors, which include Anthony Kenny, Michael Dummett, and Bernard Williams. The Chicago volume contains a number of excellent papers by philosophers and the added bonus of nine papers by established specialists in national security issues; it attempts, with partial success, to cover the full range of strategic nuclear options. The Cohen and Lee and the Fox and Groarke volumes consist largely of papers by strategic outsiders far from the centres of political power; the debate in these volumes is not between the principal versions of nuclear deterrence but between nuclear deterrence in any form and some scheme that transcends the present arrangement of mutually deterring superpowers. Of these last two books, Cohen and Lee contains relatively longer and more detailed essays.

AbstractWe prove a structure theorem for triangulated Calabi–Yau categories: an algebraic 2-Calabi–Yau triangulated category over an algebraically closed field is a cluster category if and only if it contains a cluster-tilting subcategory whose quiver has no oriented cycles. We prove a similar characterization for higher cluster categories. As an application to commutative algebra, we show that the stable category of maximal Cohen–Macaulay modules over a certain isolated singularity of dimension 3 is a cluster category. This implies the classification of the rigid Cohen–Macaulay modules first obtained by Iyama and Yoshino. As an application to the combinatorics of quiver mutation, we prove the non-acyclicity of the quivers of endomorphism algebras of cluster-tilting objects in the stable categories of representation-infinite preprojective algebras. No direct combinatorial proof is known as yet. In the appendix, Michel Van den Bergh gives an alternative proof of the main theorem by appealing to the universal property of the triangulated orbit category.

O presente artigo parte da afirmação de Michael Fried em "Arte e objetidade" (1967) de que o teatro é o que se encontra entre as artes. Virando a hipótese do crítico – aventada em chave negativa – do avesso, apontamos tal característica fluida e híbrida do teatro como o caminho percorrido por muitos artistas contemporâneos em significativas obras. Partindo dessa premissa, o artigo tem como objetivo reconstituir – a partir de entrevistas, principalmente – um longo processo de criação centrado na figura da brasileira Laura Vinci, artista visual envolvida também em processos teatrais. Partimos, então, da obra Máquina do Mundo (no singular), obra no campo das artes visuais, chegando às artes da cena com Máquinas do Mundo (no plural), criação da artista junto a mundana companhia. Abstract Most Jewish immigrants to America during the early 20th century arrived speaking Yiddish or Ladino and using Hebrew and Aramaic for liturgical purposes. When subsequent generations abandoned the first two languages, Hebrew and Aramaic were retained, used primarily for liturgy and rites of passage. Jewish American writers have often inserted Hebrew into their English texts by either reproducing the original alphabet or transliterating into Latin letters. This essay focuses on diverse strategies for representing liturgical Hebrew with an emphasis on the poetic, thematic, and sociolinguistic aspects of these expressions of both home and the foreign. Hebrew transliteration is discussed for its literary (rather than phonetic) rendering, for its multilingual creative contact with the other languages and cultures of each narrative. Among the authors whose works are discussed are Philip Roth, Michael Chabon, Nathan Englander, Joshua Cohen, Achy Obejas, and Gary Shteyngart.
