Abstract:

Abstract:
Fibrosis is a hallmark of atrial structural remodeling. The main aim of this study was to investigate the role of micro-ribonucleic acids (miRNAs) in the modulation of fibrotic molecular mechanisms in response to hypoxic conditions, which may mediate atrial fibrosis. Under a condition of hypoxia induced by a hypoxia chamber, miRNA arrays were used to identify the specific miRNAs associated with the modulation of fibrotic genes. Luciferase assay, real-time polymerase chain reaction, immunofluorescence and Western blotting were used to investigate the effects of miRNAs on the expressions of the fibrotic markers collagen I and III (COL1A, COL3A) and phosphorylation levels of the stress kinase c-Jun N-terminal kinase (JNK) pathway in a cultured HL-1 atrial cardiomyocytes cell line. COL1A and COL3A were found to be the direct regulatory targets of miR-let-7a, miR-let-7e and miR-133a in hypoxic atrial cardiac cells in vitro. The expressions of COL1A and COL3A were influenced by treatment with miRNA mimic and antagomir while hypoxia-induced collagen expression was inhibited by the delivery of miR-133a, miR-let-7a or miR-let-7e. The JNK pathway was critical in the pathogenesis of atrial fibrosis. The JNK inhibitor SP600125 increased miRNA expressions and repressed the fibrotic markers COL1A and COL3A. In conclusion, MiRNA let-7a, miR-let-7e and miR-133a play important roles in hypoxia-related atrial fibrosis by inhibiting collagen expression and post-transcriptional repression by the JNK pathway. These novel findings may lead to the development of new therapeutic strategies.

Abstract:
By promoting atrial structural remodeling, atrial hypoxia contributes to the development of the atrial fibrillation substrate. Our study aimed to investigate the modulatory effect of hypoxia on profibrotic activity in cultured HL-1 cardiomyocytes and explore the possible signaling transduction mechanisms of profibrotic activity in vitro. Hypoxia (1% O2) significantly and time-dependently increased the expression of hypoxia-inducible factor (HIF)-1α and fibrotic marker proteins collagen I and III (COL1A and COL3A), transforming growth factor (TGF)-β1 and α-smooth muscle actin (SMA). Western blot or immunohistochemistry analysis showed that hypoxia-induced increase in COL1A and COL3A was significantly attenuated by the addition of SP600125 (a specific c-Jun N-terminal kinase [JNK] inhibitor) or expression of dominant-negative JNK before hypoxia treatment. The inhibition of hypoxia-activated phosphorylation of JNK signal components (JNK, MKK4, nuclear c-Jun and ATF-2) by pre-treatment with SP600125 could suppress hypoxia-stimulated HIF-1α upregulation and fibrotic marker proteins expression. Hypoxia significantly increased reactive oxygen species (ROS) production in cultured HL-1 atrial cells. Pre-treatment with N-acetylcysteine significantly abrogated the expression of nuclear HIF-1α, JNK transduction components and fibrotic marker proteins. Taken together, these findings indicated that the hypoxia-induced atrial profibrotic response occurs mainly via the ROS/JNK pathway, its downstream upregulation of HIF-1α and c-Jun/ATF2 phosphorylation and nuclear translocation to up-regulate the expression of fibrosis-related proteins (COL1A, COL3A, TGF-β1 and α-SMA). Our result suggests that suppression of ROS/JNK signaling pathway is a critical mechanism for developing a novel therapeutic strategy against atrial fibrillation.

Abstract:
The discovery of experimentally derived induced pluripotent stem cells (iPSCs) has fostered prospects of patient-specific cell replacement therapies, novel toxicology and drug screening assays, and informative cell models for understanding disease pathogenesis. The latter is particularly valuable to the study of human syndromes caused by gene defects where animal models are lacking or inadequately mimic the human condition. One such vascular variant is Ehlers-Danlos Syndrome (EDS), a heritable disease of connective tissues involving collagen types COL5A, COL3A, tenascin-X, and to a lesser extent COL1A, lysyl hydroxlase and ADAM metallopeptidase. Among the six major types, the vascular form of EDS (vEDS) is the most severe. It results principally from mutations in the collagen III, alpha 1 (COL3A1) gene and is thought to cause aberrant collagen fibrillogenesis. These mutations contribute to catastrophic ruptures of large arteries, strokes, pregnancy-related mortalities, and pre-mature death among young adults. The presentation of this disease is heterogeneous, even among patients with the same gene mutation, and it remains unclear why some individuals exhibit severe phenotypes while others do not. In mouse models, COL3A1 deficiency mostly produces a weak disease variant without vascular ruptures, while a spontaneous COL3A1 mutation that leads to vascular defects lacks important disease features found in humans. Currently, there are no adequate therapeutic treatments. To determine how COL3A1 mutations lead to vEDS, we propose to examine and exploit a human cell model of this disease using iPSC lines derived from fibroblasts obtained from two probands. We will correct the Col3A1 gene mutations with TALENs to ensure against phenotypic iPSC variability and determine the principal source of COL3A1 secretion from iPSC-derived vascular progeny. We will examine basic growth characteristics and functional properties of these cells as well as determine the effects of doxycycline on collagen and matrix metalloproteinase synthesis (RNA and protein), stability or activity. By establishing high quality, experimentally confirmed iPSC vascular progeny, this study should lead directly to the development of reliable human tissue models of vEDS suitable for examining the effects of COL3A1 mutations on vasculogenesis and vascular integrity as well as establish a reliable system to test for possible therapeutic (pharmacological or regenerative) interventions.

Abstract:
To identify Ejiao and its adulterants at the DNA level by using DNA molecula marker. Ejiao (Asini Corii Colla) is a commonly used medicinal material. However, its adulteration is a serious concern. Due to the morphological characteristics of Asini Corii Colla and its adulterants, traditional identification techniques often complex and professional, which is not conducive to the circulation management and safety of the medicinal materials. To improve the distinction between Asini Corii Colla and its adulterants accurately, this study identified and its adulterant samples based on the CytB sequence. Sequence characteristics, Basic Local Alignment Search Tool (BLAST) application, genetic distance, construction of phylogenetic tree showed the CytB sequence to accurately identify Asini Corii Colla from its adulterants. Furthermore, in this study, we designed a specific primer, based on the CytB sequence, and established a PCR detection system for rapid, sensitive, and specific identification of Asini Corii Colla. Compared to DNA barcoding technology, this method has shorter detection time, stronger specificity, and higher sensitivity, which lays the foundation for the rapid identification of Asini Corii Colla.

Abstract:
Abstract Mechanisms by which leptin stimulates collagen α1(I) [Col1a(I)] gene expression are unclear. The purposes of this study were to identify the trans-acting factors and cis-acting elements in Col1a(I) promoter involved in this effect as well as the pathways that are implicated. In primary cultures of rat hepatic stellate cells (HSCs), we measured the effects of leptin on Col1a(I) gene and protein expression and on the binding of nuclear proteins to the Col1a(I) promoter. We found that leptin increased Col1a(I) gene and protein expression in activated HSCs. Transient transfections showed that leptin exerted its effects through elements located between −220 and −112 bp of the Col1a(I) promoter. Gel retardation assays demonstrated that leptin induced the binding of transcription factors specific protein (Sp)-1 and Sp3 to two elements located between −161 and −110 bp of the Col1a(I) promoter. Leptin-induced Sp1/Sp3 phosphorylation, but this effect was suppressed by inhibiting or silencing Janus kinase-2, phosphatidylinositol-3-kinase, nonphagocytic adenine dinucleotide phosphate (NADPH) oxidase, or ERK1/2, by the use of antioxidants or catalase, or by preventing protein-aldehyde adduct formation. Leptin provoked oxidative stress, aldehyde-protein adduct formation, and increased gene expression of some components of the NADPH oxidase complex. In conclusion, in HSCs, leptin up-regulates Col1a(I) gene expression after activating NADPH oxidase, inducing oxidative stress, aldehyde-protein adduct formation, and ERK1/2 phosphorylation, which in turn activates Sp1/Sp3 and provokes the binding of these two factors to regulatory elements located between −161 and −110 bp of the Col1a(I) promoter. These findings may contribute to a better understanding of mechanisms involved in the leptin-induced liver fibrosis.

Abstract:

Abstract Pollen morphology of herbarium specimens of four Centranthus species (C. ruber, C. longiflorus, C. kellereri and C. calcitrapae) was studied using LM and SEM. The research aim was to provide data on their pollen characteristics and to evaluate the taxonomic value of these data for species-specific identification. Pollen grains are tricolpate, suboblate to prolate (P/E = 0.81–1.42); medium- or large-sized (P = 49.21–90.44 µm; E = 43.89–93.10 µm). Colpi are long or medium-length, wide at equator, tapered to acute or obtuse ends. Exine sculpture is echinatemicroechinate-nanoechinate; echini (1.00–1.39 µm high) and microechini (0.55–0.98 µm) are conical, with straight or convex sides and acute apices, nanoechini are 0.22–0.46 µm high. Most important characters of taxa diagnostic at species level for the taxonomy are: size of pollen and colpi, exine structure, size of echini and microechini, and pattern of tectum in areas between echini. Pollen grains of C. calcitrapae and C. macrosiphon (sect. Calcitrapa) are generally smaller in size than grains of C. ruber, C. longiflorus and C. kellereri (sect. Centranthus). Pollen of C. kellereri was analysed for the first time in the current study.

Abstract:
The main goal of sports energy drinks is to provide athletes with an additional impulse of internal energy. In the composition of the developed sports drink «Inka Cola» as a source of biologically active substances was used an extract of the seeds of the South American shrub - guarana. A specific feature of this plant is the high concentration of caffeine in its seeds, the content of which is about 4-5 times higher than in coffee beans. The inclusion of guarana seed extract in the «Inka Cola» drink composition helps to reduce fatigue of athletes and ensure long-term maintenance of a high level of their physical and cognitive activity. An extract of the gotu-kola herb, which grows in the countries of Southeast Asia, was used as another source of biologically active substances. The constituents of this extract allow to fight emotional disorders, improve memory and restore nerve cells in the human brain. They also have a wide range of anti-inflammatory, immunostimulating, antibacterial and antioxidant properties. In order to increase the energy value and improve the taste perception of the «Inka Cola» drink, isomaltulose, an isomer of sucrose with a reduced glycemic index, was included in its composition; to increase the physiological impact on the athletes were used important biogenic mineral components - potassium and magnesium; to create a harmonious taste perception of the drink - acidity regulator (citric acid) and attractive rheological properties of the drink - thickener (xanthan gum) were included. The formation of the color-aromatic image of the drink «Inka cola» was carried out by using in its composition of the flavoring product «mojito» and congruent food coloring. According to the reviews of athletes who used the «Inka Cola» drink for several weeks, their fatigue decreased, it became easier for them to perceive and assimilate information, as well as an increase in physical and mental endurance was noted.

Abstract:
Plantains and bananas (Musa spp. sect. eumusa) originated from intra- and interspecific hybridization between two wild diploid species, M. acuminata Colla. and M. balbisiana Colla., which contributed the A and B genomes, respectively. Polyploidy and hybridization have given rise to a number of diploid, triploid, and tetraploid clones with different permutations of the A and B genomes. Thus, dessert and highland bananas are classified mainly as AAA, plantains are AAB, and cooking bananas are ABB. Classification of Musa into genomic groups has been based on morphological characteristics. This study aimed to identify RAPD (random amplified polymorphic DNA) markers for the A and B genomes. Eighty 10-mer Operon primers were used to amplify DNA from M. acuminata subsp. burmannicoides clone 'Calcutta 4' (AA genomes) and M. balbisiana clone 'Honduras' (BB genomes). Three primers (A17, A18, and D10) that produced unique genome-specific fragments in the two species were identified. These primers were tested in a sample of 40 genotypes representing various genome combinations. The RAPD markers were able to elucidate the genome composition of all the genotypes. The results showed that RAPD analysis can provide a quick and reliable system for genome identification in Musa that could facilitate genome characterization and manipulations in breeding lines.Key words: banana and plantain, A and B genomes, genomic groups, RAPD markers.

Abstract:
Effects of fractions A and B from enzyme-digested traditional Chinese medicine colla corii asini on mice with 5-fluorouracil-induced anemia were investigated. The purpose of this study was to further understand the hematopoietic activities and mechanisms of colla corii asini. The fractions A and B were administered to anemic mice for 12 days. After confirming the anti-anemic effect of fractions A and B, we examined the effects of fractions A and B on immature granulocyte and erythroid cell activity and plasma cytokine level. Fraction A administration at 2 g/kg and 1 g/kg and fraction B administration at 1.6 g/kg and 0.8 g/kg activated granulocyte and erythrocyte progenitor cells in bone marrow and erythrocyte progenitors in spleen, led to the recovery of white blood cell and red blood cell counts, and increased the percentage of peripheral reticulocytes in red cells. The GM-CSF and EPO production determined by examining GM-CSF mRNA and EPO mRNA in the kidney and liver of the anemic mice were also enhanced. This treatment significantly increased serum GM-CSF and EPO level and lowered serum transforming growth factor ( TGF -β) level. These results suggested that fractions A and B promoted hematopoiesis by activating immature granulocyte and erythroid cells, partly by stimulating GM-CSF and EPO secretion and suppressing TGF -β release. Identification of a specific peptide or protein is still required for the development of a novel medicine for anemia caused by malignancy or chemotherapy.

Abstract:
Pollen morphology of 34 herbarium specimens belonging to 17 taxa of the genus Valeriana was studied using light and scanning electron microscopy. The aim of this research was to evaluate the taxonomical value of palynomorphological data for species-specific identification. Pollen grains are monads, radially symmetrical, isopolar, tricolpate, oblate to prolate (P/E = 0.69–1.59); medium- or large-sized (P = 26.60–66.50 µm; E = 23.94–57.19 µm). Colpi are long, medium-length or short, with acute, obtuse or rounded ends. Four types and eight subtypes of exine sculpture were recognised: type I—echinate-verrucate (5 spp.), type II—echinate-nanoechinate-verrucate (10), type III—echinate-microechinate-nanoechinate-verrucate (1), type IV—echinate-microechinate-nanoechinate (1). Additional characters of taxa of Valeriana diagnostic at species level for the purposes of taxonomy are: size of pollen and colpi, exine sculpture: presence/absence of verrucae, their form, size and form of echini/microechini/nanoechini, their number, pattern of tectum in areas between echini, exine thickness in mesocolpia and apocolpia. Our palynomorphological data support the results of molecular phylogenetic studies. The close phylogenetic relationships of the species V. montana, V. tripteris, V. supina and V. pyrenaica are also supported by their palynomorphological characters, such as exine sculpture (type I). Cluster analysis mainly supports the definition of pollen types and subtypes (based on qualitative data), since species are grouped as five separate branches. The pollen grains of five taxa were analysed for the first time in the current study.

Full text
Abstract:
The article is devoted to the analysis of music reviews as affected by transformation of the genre and its framework in the convergent media environment. The proposed original structure of analysis is developed and reasoned based on the ideas presented in works by Russian and foreign scholars on the specific features of the review as a genre (Ye. A. Nabiyeva, A. A. Tertychnyi, and others), media convergence (H. Jenkins, T. Rogers, and others), interpretation of music images (T. Adorno, Yu. Strakovich, and others), suggestiveness of media texts (A. Beck, D. Burns, J. Baron and others). The analysis structure includes three basic sections: content, environment of publication, communicative tactics; each section is expanded in several criteria grades. The review is considered as an analytical genre that is traditionally based on suggestion as the leading communicative strategy. However, today this genre is transforming intensively in the mutable deinstitutionalized environment of the network mass media. Distortions, which are inevitable in conversion of an artistic image into a text, become especially identifiable in music reviews and increase subjectivity initially typical for this genre. The structure of analysis elaborated in the article engages particularly a classification of heuristics, or cognitive biases, which are considered to be cognitive distortions turning in opinion journalism into the means of expression and suggestion. These means are regarded as the foundation for the genesis of local media myths. The proposed method is tested on the review published at the Colta web portal. In conclusion, the paper emphasizes the significance of the research done for some general issues of communication studies, and in particular notes the method’s applicability to the analysis of inner cognitive triggers a person feels exposed to in the network society.

Abstract:
The first treatise of the so‑called Passional of Abbess Cunegund was composed by the Dominican lector Colda of Colditz at the abbess’ behest in 1312, and illustrated by an unknown artist. In this study, images of three specific character‑types are analysed: villain, soldier and Jew, who not only act out their part in the treatises’ account of Christ’s Passion, but also appear to provide a window onto certain aspects of contemporary, medieval Czech society. By examining the iconography of the illustrations it is shown how, through characterisation, caricature and dress, the viewer is led to adopt an attitude either of disgust towards the malefactors, or a certain sympathy towards a chosen few.

- Guilt is one of most studied emotions because of its moral implications and its role in helping and reparative behaviours. However, there is a great debate regarding the features that define this particular self-conscious emotion, what differentiate it from other emotions, as well as the processes and contexts which favour a functional and adaptive experience, rather than a dysfunctional and painful one. Many hypotheses have been suggested during time, and then tested through different methodologies and research tools. The present paper aims to discuss some of the implications in the use of a particular methodology rather than another, showing some of the properties of the main research tools in the study of guilt. The aim is to shed light on the theoretical assumptions on which these tools are based, underlining as various emerging concepts of guilt raise from the specific theoretical and methodological kind of "spectacles" used to observe it.

Abstract:
The application of organic fertilizer has been one of the most important agricultural practices to increase tea plant productivity. However, a single organic fertilizer alone could not match the nutrient requirements of tea plant. According to the nutrient demand of tea plant, tea-specific fertilizer with an appropriate nutrient ratio in combination with organic fertilizer would be an appropriate way. A four-year field experiment was conducted to characterize the sensory quality of green tea subjected to four different fertilization treatments, i.e., tea-specific fertilizer (N-P2O5-K2O: 18-8-12) + urea + colza cake, tea-specific fertilizer + urea + livestock waste compost, and tea-specific fertilizer + urea, combined tea-specific fertilizer (referred to as M1, M2, and M3, respectively); nonfertilizing (CK) served as control. The fertilizer formulated specifically for tea could enhance the taste qualities of green tea, such as water extract, caffeine, tea polyphenol, EGCG, and ECG. However, the effect of this fertilizer on enhancing the aroma level of green tea was weaker than that of the application of tea-specific fertilizer combined with organic fertilizer, whereby this combined fertilization significantly improved the concentration of green tea aroma compounds, such as D-limonene, cis-jasmone, nonanal, linalool, cis-3-hexenyl hexanoate, and cis-3-hexenyl benzoate. This study provides a theoretical basis for judicious fertilization by combining tea-specific fertilizer and organic fertilizer in tea orchards.

Abstract:
Donkey-hide gelatin, also called Ejiao (colla corii asini), is commonly used as a food health supplement and valuable Chinese medicine. Its growing popular demand and short supply make it a target for fraud, and many other animal gelatins can be found as adulterants. Authentication remains a quality concern. Peptide markers were developed by searching the protein database. However, donkeys and horses share the same database, and there is no specific marker for donkeys. Here, solutions are sought following a database-independent strategy. The peptide profiles of authentic samples of different animal gelatins were compared using LC-QTOF-MS/MS. Fourteen specific markers, including four donkey-specific, one horse-specific, three cattle-specific, and six pig-specific peptides, were successfully found. As these donkey-specific peptides are not included in the current proteomics database, their sequences were determined by de novo sequencing. A quantitative LC-QQQ multiple reaction monitoring (MRM) method was further developed to achieve highly sensitive and selective analysis. The specificity and applicability of these markers were confirmed by testing multiple authentic samples and 110 batches of commercial Ejiao products, 57 of which were found to be unqualified. These results suggest that these markers are specific and accurate for authentication purposes.

Abstract:
Abstract This paper presents a characterization of the following dry granular materials: soya, colza and rye seeds. The physical properties of the grains and the materials are useful for characterizing the materials’ behaviour during flow, while the external conditions (consolidation) determine storage and handling conditions. The physical properties of the grains (specific densities) and of the materials as a whole (compacity or porosity, and critical angles) were measured. The flow functions were determined by modified shear box testing. Then the internal friction angles and the flowability index for each granular material were obtained. Indeed, the behaviour of a flowing granular material results from these two groups of factors and is characterized by the flowability, which is the ratio of highest consolidation stress and unconfined yield strength. In practice, the flowability index is used to classify materials, so that the larger the flowability index, the smaller the bulk solids strength will be in relation to the consolidation stress, and therefore the higher the flowability of the bulk solid.

Abstract Pollen morphology of representatives of the genera Succisa and Succisella in the flora of Ukraine was studied using light and scanning electron microscopy. Pollen grains in both taxa were tricolpate, prolate, rarely oblate-spheroidal or spheroidal; large-sized. Their outline in polar view was subcircular, rarely trilobate, in equatorial view elliptical or rarely circular. Colpi short, of variable width, margins irregular with distinct, narrow margo, and blunt or acute ends. Exine sculpture was echinate-microechinate. The revealed characteristics of pollen grains are taxonomically significant at the generic and specific levels, and they can be used in pollen analysis. Palynomorphological data are consistent with the results of recent molecular phylogenetic studies. Data on pollen morphology also confirm taxonomic circumscription of Succiseae V. Mayer & Ehrend. Pollen grains in Succiseae are characterized by a tricolpate aperture type, which was probably ancestral in Dipsacaceae s. str.

Abstract:
Abstract Osteoporosis represents an important cause of morbidity in adult patients with thalassemia. Its pathogenesis is multifactorial, and includes mainly bone marrow expansion, endocrine dysfunction and iron overload. Bone metabolism is altered in thalassemia. Osteoclast function is elevated, while osteoblast activity seems to be reduced and thus the balance of bone remodeling is in favor of bone loss. The exact mechanisms of osteoblast dysfunction have not been fully clarified to-date. Wingless-type (Wnt) signaling is an important pathway for osteoblast differentiation. Dickkopf-1 (Dkk-1) protein is an inhibitor of Wnt pathway and is implicated in the pathogenesis of several bone disorders. Collagen type-I is the main structural protein of the bone. The collagen type-I alpha (COLIA)-1 specific protein (Sp)-1 polymorphism has been related to osteoporosis in thalassemia. The aim of this study was to evaluate the serum levels of Dkk-1 in patients with thalassemia-induced osteoporosis who receive therapy with zoledronic acid (ZOL) and evaluate possible correlations with clinical and laboratory data, including the COLIA-1 Sp1 polymorphism. Sixty-six patients (21M/45F; median age 35.5 years) with thalassemia and osteoporosis were studied. Patients were blindly randomized to receive ZOL at a dose of 4 mg, iv, in 15 min infusion, every 6 months (group A, n=23) or every 3 months (group B, n=21), or to receive placebo every 3 months (group C, n=22) for a period of one year. All patients received oral calcium (500 mg) during the treatment period. Dkk-1 was measured at baseline and after 12 months of therapy using ELISA methodology (Biomedica Medizinprodukte, Wien, Austria) along with a series of serum bone remodeling indices: bone resorption markers [C-telopeptide of type-I collagen (CTX), tartrate-resistant acid phosphatase isoform-5b (TRACP-5b)], bone formation markers [bone-specific alkaline phosphatase (bALP), osteocalcin, and C-terminal propeptide of collagen type-I (CICP)], and osteoclast regulators [receptor activator of nuclear factor-kappa B ligand (RANKL), osteoprotegerin (OPG), and osteopontin]. The above bone markers were also evaluated in 30, age- and gender-matched, healthy controls. The G-->T mutation at base 1 of intron 1 at the binding site of the Sp1 transcription factor of the COLIA-1 gene was detected by polymerase chain reaction using mutagenesis primers followed by restriction enzyme analysis in all patients. BMD of the lumbar spine (L1-L4), femoral neck (FN) and wrist (W) was determined using DEXA, before and 12 months after treatment. At baseline, all patients had increased serum levels of Dkk-1 (mean±SD: 39±17.1 pmol/L) compared to controls (27.4±9.7 pmol/L; p<0.0001). Furthermore, thalassemia patients had increased values of CTX (p<0.0001), bALP (p<0.001), CICP (p=0.003), sRANKL (p=0.02), and OPG (p=0.001) compared to controls. Results for the COLIA-1 Sp1 polymorphism were available for 53 patients. Seventeen patients (32%) were G/T heterozygotes at the polymorphic Sp1 site (Ss), while 3 (5.6%) were T/T homozygotes (ss). Dkk-1 serum levels correlated with L1-L4 BMD (r=−0.290, p=0.022) and W-BMD (r=−0.415. p=0.001), but also with TRACP-5b (r=0.310, p=0.011) and bALP levels (r=−0.289, p=0.018). Ss and ss patients tended to have lower L1-L4 BMD compared with SS patients (p=0.09). No significant correlations were observed between Ss and ss patients with the measured bone markers or the response to ZOL. As reported previously, patients of group B experienced an increase of L1-L4 BMD, while no other alterations in BMD were observed in the 3 studied groups after 12 months of ZOL administration. Interestingly, patients of groups A+B showed a strong reduction of Dkk-1 after 12 months of ZOL (from 39.6±16.6 to 28.9±16.3 pmol/L; p=0.004); indeed they almost normalized Dkk-1 levels (no difference from control values). In contrast, patients of group C showed a borderline increase of Dkk-1 (from 33.1±16.8 to 40.1±23.2 pmol/L, p=0.08). These results show for the first time in the literature that Dkk-1 is increased in the serum of patients with thalassemia and osteoporosis, correlates with their BMD and is reduced post-ZOL therapy. This Dkk-1 elevation may be at least partially responsible for osteoblast dysfunction in thalassemia and reveal a novel possible target for the development of new agents for the management of bone loss in thalassemia patients.

AbstractThis paper presents initial steps towards an auto-mated analysis for pain-related evoked potentials (PREP) to achieve a higher objectivity and non-biased examination as well as a reduction in the time expended during clinical daily routines. While manually examining, each epoch of an en-semble of stimulus-locked EEG signals, elicited by electrical stimulation of predominantly intra-epidermal small nerve fibers and recorded over the central electrode (Cz), is in-spected for artifacts before calculating the PREP by averag-ing the artifact-free epochs. Afterwards, specific peak-latencies (like the P0-, N1 and P1-latency) are identified as certain extrema in the PREP’s waveform. The proposed automated analysis uses Pearson’s correlation and low-pass differentiation to perform these tasks. To evaluate the auto-mated analysis’ accuracy its results of 232 datasets were compared to the results of the manually performed examina-tion. Results of the automated artifact rejection were compa-rable to the manual examination. Detection of peak-latencies was more heterogeneous, indicating some sensitivity of the detected events upon the criteria used during data examina-tion.

Abstract:
The Collagen family is one of the most extensively researched protein families. However, because an evolutionary study has focused mostly on superfamilies, there is a paucity of information about subfamilies. To promote a healthy circulatory system, healthy skin through enhancing elasticity, and a healthy joint system, it is crucial that the subfamily of collagen evolve. Additionally, these kinds of studies support bone health, development against various developmental problems, and placenta growth in expectant women. The current research sought to comprehend collagen evolution on both a comprehensive and reductive level. The full family of human collagen proteins was exposed to BLAST techniques to identify orthologs and paralogs with various criteria, followed by the creation of phylogenetic trees. Our findings unambiguously demonstrated that the most archaic collagen isoforms in higher eukaryotes are COL20A1HP and COL12A1HPa, which later diverged into other collagen family members, possibly as a result of various gene modification processes. Additionally, a site-specific selection pressure study utilizing the selection server revealed that COL4A isoforms are evolving more quickly than other types of collagen.

Abstract:

Abstract:
Indonesia has been attractive investment destination to many multinational companies since a long time ago, due to its big population, plenty natural resources and cheap labors, including to Coca Cola. This study aims to analyze international market entry and business strategy of Coca Cola when entering Indonesia market. Moreover, this study also aims to identify internal and external factor that affects Coca Cola business in Indonesia by SWOT analysis, resulting in growth strategy recommendation. The result reveals out that Coca Cola came to Indonesia by foreign direct investment at first time, but then decided to expand their distribution all over Indonesia. Therefore, they opened opportunity for franchise. By keeping their secret formula, they can provide differentiation that makes them remains competitive. The strategy adopted by Coca Cola Amatil Indonesia to conquer Indonesia market is transnational strategy, which has characteristics of high in local responsiveness and also high in global integration. Meanwhile, the SWOT analysis suggests that Coca Cola should put priority on market penetration first instead of market development for the time being. Company should focus on sales and distribution in specific market first then explore more market for their product. There are still many possible markets to exploit in Indonesia as well as abroad.

Abstract:
Purpose Risk-related information is prevalently used in the decision-making process by various counterparties. Therefore, this study investigates how compa-nies conduct their risk-disclosure practice after the new Italian Legislative Decree No. 254 of December 30, 2016. In particular, we draw attention to three aspects: (1) the interaction relationship among risk or risk management (RRM), industry, type of risk, and level of specific disclosure; (2) the variation of specific level of disclosing risk-related information across the industries and types of risk; and (3) the different behavior between risk and risk-management disclosure in the after-math of the regulation's issuance. Design/methodology/approach The study is based on a sample of large un-dertakings and groups that are subject to the Legislative Decree. Two phases of content analysis were executed to analyze the risk and risk-management disclosure. The research questions were investigated with the row effects loglinear model. Findings Our result shows that there are interaction relationships among RRM, type of risk, industry, and level of specific disclosure. Companies provide risk-related information at different levels of specificity depending on whether the information is risk description or risk management, the firms are operating in manu-facturing or nonmanufacturing, and the type of risk that the firms disclosed in their reports. Practical implications The paper provides evidence of inconsistent company behavior in disclosing company-specific information in favor of internal and ex-ternal stakeholders, particularly by balancing company-specific disclosure be-tween risk descriptions and risk-management policies. Policymakers might also consider this current phenomenon to decide to what extent disclosure requirements should be detailed and, instead, what room should be left for management discre-tion with respect to users' needs. Originality/value This paper is an up-to-date assessment of Italian firms' compliance with Legislative Decree No. 254 of December 30, 2016.

Abstract:
This millennium is marked by a new trend: efficien-cy. In the actual economical environment, business sustainability requires high-efficiency technological processes. The efficiency concept has to be present at all levels of industrial activities. However, as com-mon practice, the efficiency concept is still regarded equivalent to ‘energy efficiency’ as mentioned in the Intergovernmental Panel on Climate Change (IPCC 1996), and specifically re-defined by the Federal Energy Management Plan (Department of Energy Federal Register 1994.By analysing specific industrial processes, the authors have defined a global concept of efficiency presenting results of sound research in this field, with reference to electric motors and drives. Typical examples supporting the theoretical background reveal general impacts on the South African econo-my by implementing this new concept:• Technical and economical performance improvement and competitiveness of South African companies to international standards;• Defusing an incipient energy crisis in the sector;• Improving environmental conditions (less ema-nations of carbon dioxide at the power plants); and• Creating new job opportunities in the sector. The global concept of efficiency proposed in this paper can be further developed in assessing effi-ciency of various processes, thus improving compa-nies’ corporate energy policy.

Abstract:
The association between collagen type I alpha (COL1A) and chemoresistance has been verified in cancers. However, the specific role of COL1A2 in gastric cancer (GC) cell resistance to apatinib, a highly selective small-molecule inhibitor of vascular endothelial growth factor receptor 2, has not been investigated before. The purpose of this study was to explore the potential factors associated with COL1A2 regulation on GC cell apatinib resistance in vitro. With the aid of the Oncomine database and integrated bioinformatics methods, we identified COL1A2 overexpression in GC and its prognostic value. Mechanistically, the COL1A2 promoter has a distinct H3K27ac modification site and that E1A binding protein p300 (EP300) and twist family bHLH transcription factor 1 (TWIST1) can bind to the COL1A2 promoter, which in turn transcriptionally activated COL1A2 expression. In addition, overexpression of COL1A2 significantly promoted resistance to apatinib in GC cells, but knockdown of EP300 or TWIST1 remarkably inhibited COL1A2 expression and promoted sensitivity of GC cells to apatinib. Our findings demonstrated that the combination of EP300 and TWIST1 has a synergistically regulatory effect on COL1A2 expression, thus contributing to apatinib resistance in GC cells.

Abstract:
Organoids are 3D-culture systems composed of tissue-specific primary cells that self-organize and self-renew, creating structures similar to those of their tissue of origin. Testicular organoids (TOs) may recreate conditions of the testicular niche in domestic and wild cattle; however, no previous TO studies have been reported in the bovine species. Thus, in the present study, we sought to generate and characterize bovine TOs derived from primary testicular cell populations including Leydig, Sertoli and peritubular myoid cells. Testicular cells were isolated from bovine testes and cultured in ultra-low attachment (ULA) plates and Matrigel. TOs were cultured in media supplemented from day 3 with 100 ng/mL of BMP4 and 10 ng/mL of FGF2 and from day 7 with 15 ng/mL of GDNF. Testicular cells were able to generate TOs after 3 days of culture. The cells positive for STAR (Leydig) and COL1A (peritubular myoid) decreased (p < 0.05), whereas cells positive for WT1 (Sertoli) increased (p < 0.05) in TOs during a 28-day culture period. The levels of testosterone in media increased (p < 0.05) at day 28 of culture. Thus, testicular cells isolated from bovine testes were able to generate TOs under in vitro conditions. These bovine TOs have steroidogenic activity characterized by the production of testosterone.

Abstract:
In recent years, there has been increasing talk about reverse acquisitions in capital concentration process- es. These situations are associated with the use of certain economic, tax, and other privileges by compa- nies that are often the initiator of such undertakings. The purpose of the article is, among others, to pre- sent the premises that guide economic entities when making decisions about reverse acquisitions. It is a process in which there is a specific type of acquisition of control of one entity by another. In these cases, there is a reversal of typical relationships between entities involved in the merger, because the entities that have control over the entities that acquire them are taken over. The regulations of the Interna- tional Financial Reporting Standard (IFRS) No. 3 "Business Combinations" have a very important role in accounting for these processes. They allow for the proper recognition of these relationships. Based on numerical examples, the authors present the rules for accounting for such a combination. Thus, they confirm the role of accounting, which implements the principle of faithful and reliable presentation of this process. A critical analysis of the literature and legal regulations, as well as deductive reasoning, made it possible to determine the most important motives of this type of connection.

Abstract:
Despite the importance of and current demand for abaca (Musa textilis Nee) fiber, there has been limited study that capitalizes on RNA-seq to identify candidate genes associated with high fiber quality and bunchy top virus (AbBTV) resistance. Three varieties (Abuab, Inosa, and Tangongon), one wild banana variety (Musa balbisiana Colla) Pacol, and two developed backcrosses (Abuab × Pacol BC2 and BC3) were grown at the Institute of Plant Breeding (IPB), Laguna, Philippines. The pseudostems of 3-month-old suckers of each genotype were sampled for RNA-seq. Datasets were analyzed for differential expression (DE) implementing various model frameworks, including pairwise, genotypic and non-DE models. Results indicate that Abuab and BC3 induce the highest proportion (70%) of abaca-specific genes. Gene ontology (GO) enrichment analysis showed several genes associated with cellulose synthase activity, callose synthase, ß-glucosidase activity, glucan biosynthetic process, etc. KEGG pathway analysis showed several genes encoding for enzymes involved in the lignin biosynthetic pathway. Analysis using genotypic DE (GDE) between abaca bunchy top virus (AbBTV)-resistant and -susceptible groups revealed genes such as pathogenesis-related protein and NBS-LRR. As the genotypes were not infected with the pathogen, these genes are yet to be confirmed for their roles in disease resistance and are an interesting subject for further investigation.

Abstract:
COVID-19 has had a direct impact on the physical and mental health of millions of people worldwide. Therefore, a Mental Health Crisis Helpline (MHCH) was set up and offered free of charge by the Professional College of Psychology of Aragon (COPPA) during the lockdown period. This research aims to study the reasons for the calls, to describe the population segments that used it, and to analyse the possible relationships between the variables studied. A total of 1411 calls were answered and 598 were registered. The main reasons for the calls were: anxiety symptoms, concern for a relative, previous mental health problems, conflicts, and depressive symptoms. Significantly more men called for anxiety symptoms (60.8% vs. 49.5%) versus more women calling regarding a chronic physical illness (3.5% vs. 0.7%), concern about a relative (22.7% vs. 12.4%), care guidelines (6% vs. 1.3%), and bereavement (6.2% vs. 2%). Calls regarding conflict increased slightly as the lockdown period progressed (p < 0.001; r = 0.15), in contrast with calls regarding previous psychological conditions and anxiety symptoms (p = 0.035; r = −0.09; p = 0.005; r = −0.12). These results highlight the intensive use of the MHCH, confirming the need for the implementation of specific psychological care resources in times of crisis.

Abstract:
The research is devoted to possibility of mineral diesel fuel with mixed biodiesel fuel partial replacement, which contains the vegetable oil as a biological component (such as: rapeseed, camelina, radish, mustard, colza, soybean, etc.). Constructive versions of bi-fuel supply system and mixers for diesel engines adaptation for their operation by biodiesel fuel are offered. The advantages of the developed bi-fuel systems and mixers are: universality in relation to different types of diesel engines; polyfuelness; availability of components; competitiveness; possibility of production in the plant conditions, no requirement of big capital investments, the small term of payback. The application of the offered system by the diesel-powered machinery provides the necessary percentage ratio of mixed fuel components depending on the load-speed mode without its essential constructive changes. The using of the mixed biodiesel fuel containing for example 50% of mineral fuel and 50% of camelina oil allows to save 50% of petroleum diesel fuel, to reduce smoke of the exaust gases by 17-20%, to reduce the content of carbon oxide by 35-40% in comparison with the work of the diesel engine on mineral diesel fuel, by insignificant decreasing of the effective power (no more than 6%) and by some increasing of specific fuel consumption of mixed fuel (to 14%).
