Abstract:
The crystal structure of arabinose-5-phosphate isomerase (API) fromBacteroides fragilis(bfAPI) was determined at 1.7 Å resolution and was found to be a tetramer of a single-domain sugar isomerase (SIS) with an endogenous ligand, CMP-Kdo (cytidine 5′-monophosphate-3-deoxy-D-manno-oct-2-ulosonate), bound at the active site. API catalyzes the reversible isomerization of D-ribulose 5-phosphate to D-arabinose 5-phosphate in the first step of the Kdo biosynthetic pathway. Interestingly, the bound CMP-Kdo is neither the substrate nor the product of the reaction catalyzed by API, but corresponds to the end product in the Kdo biosynthetic pathway and presumably acts as a feedback inhibitor for bfAPI. The active site of each monomer is located in a surface cleft at the tetramer interface between three monomers and consists of His79 and His186 from two different adjacent monomers and a Ser/Thr-rich region, all of which are highly conserved across APIs. Structure and sequence analyses indicate that His79 and His186 may play important catalytic roles in the isomerization reaction. CMP-Kdo mimetics could therefore serve as potent and specific inhibitors of API and provide broad protection against many different bacterial infections.

Kementerian Pekerjaan Umum dan Perumahan Rakyat melalui Direktorat Jenderal Bina Marga pada tahun 2017 membangun beberapa jembatan layang untuk mengatasi kemacetan, salah satunya adalah Fly Over Dermoleng yang terletak di Provinsi Jawa Tengah ditargetkan dapat difungsikan pada Lebaran 2017 dengan masa konstruksi yang hanya sekitar empat bulan. Pembangunan Fly Over Dermoleng menggunakan teknologi Corrugated-Mortar Busa Pusjatan (CMP) yang dikembangkan oleh Pusat Penelitian dan Pengembangan Jalan dan Jembatan (Pusjatan) Badan Penelitian dan Pengembangan (Balitbang) Kementerian Pekerjaan Umum dan Perumahan Rakyat, teknologi andalan sebagai salah satu solusi untuk mengatasi kemacetan di perkotaan. Penggunaan teknologi tersebut dapat berhasil mengefisiensikan masa konstruksi jembatan layang menjadi hanya sekitar empat bulan serta mampu menghemat biaya pembangunan hingga 70 persen dibandingkan teknologi konstruksi jembatan layang konvensional. Teknologi CMP merupakan pengembangan teknologi Corrugated Steel Arch, yang mempunyai banyak kelebihan dibandingkan apabila fly over menggunakan konstruksi konvensional. Kelebihan teknologi CMP antara lain lebih efisien 60-70% serta 50% lebih cepat dalam masa konstruksi dibandingkan konstruksi konvensional, sehingga istilah fly over CMP dapat juga diartikan sebagai “Fly Over Cepat Murah Pusjatan”. Tingkat efisiensi tinggi yang diperoleh merupakan hasil kontribusi penggunaan mortar busa untuk timbunan pendekat jembatan, yang merupakan ciri unik dari teknologi CMP. Potensi penggunaan CMP, dengan perkembangan teknologi yang ada, tidak hanya dapat dipergunakan untuk perlintasan kereta api saja, namun sangat cocok untuk persimpangan jalan yang membutuhkan bentang yang panjang. Guna memperbesar daya tahan rambat api pada kayu lapis, dilakukan pelaburan venir muka (face) dan belakang (back) menggunakan natrium silikat pada konsentrasi 30%, 40% dan 50% sebanyak 2 dan 4 kali ulangan. Dari hasil penelitian diperoleh bahwa pelaburan menggunakan natrium silikat dalam konsentrasi 50% sebanyak 4 kali ulangan menghasilkan nilai daya tahan rambat api yang terbaik atau rata-rata 68,08% dengan nilai retensi 0,13 gr/cm3, sedangkan kadar air dan keteguhan rekatnya adalah 10,60% dan 9,74 kg/cm2. Hasil ini memenuhi persyaratan Standar Nasional Indonesia 01-05.2/1999. Perlakuan tanpa pelaburan (blanko) menghasilkan nilai daya tahan rambat api rata-rata 28,95%, kadar air 10,83% dan keteguhan rekat 13,78 kg/cm2.

Abstract:
Angiotensin II (AII), found in seminal plasma, has been shown to stimulate capacitation in uncapacitated mammalian spermatozoa. The present study investigated the location of AII receptors on spermatozoa and AII’s mechanism of action. AT1 type receptors for AII are present on the acrosomal cap region and along the whole of the flagellum of both mouse and human spermatozoa. Because combinations of low concentrations of AII and either calcitonin or fertilization-promoting peptide (FPP), both known to regulate the adenylyl cyclase (AC)/cAMP signal transduction pathway, elicited a significant response, this study investigated the hypothesis that these peptides act on the same pathway. AII was shown to significantly stimulate cAMP production in both uncapacitated and capacitated mouse spermatozoa and this was associated with increases in protein tyrosine phosphorylation. Using an anti-phosphotyrosine antibody to visualize the location of tyrosine phosphoproteins within individual cells, AII significantly stimulated phosphorylation within 20 min in both the head, especially in the acrosomal cap region, and the flagellum, especially in the principal piece, of uncapacitated mouse spermatozoa; combined AII + FPP was stimulatory within 5 min. In addition, Western blotting revealed that AII stimulation increased phosphorylation in a number of tyrosine phosphoproteins in both uncapacitated and capacitated mouse spermatozoa, with some being altered only in the latter category of cells. These results support the hypothesis that AII stimulates AC/cAMP in mammalian spermatozoa.

Abstract:
In Sertoli cells, anti-Müllerian hormone (AMH) expression is upregulated by FSH via cyclic AMP (cAMP), although no classical cAMP response elements exist in the AMH promoter. The response to cAMP involves NF-κB and AP2; however, targeted mutagenesis of their binding sites in the AMH promoter do not completely abolish the response. In this work we assessed whether SOX9, SF1, GATA4, and AP1 might represent alternative pathways involved in cAMP-mediated AMH upregulation, using real-time RT-PCR (qPCR), targeted mutagenesis, luciferase assays, and immunocytochemistry in the Sertoli cell line SMAT1. We also explored the signaling cascades potentially involved. In qPCR experiments, Amh, Sox9, Sf1, and Gata4 mRNA levels increased after SMAT1 cells were incubated with cAMP. Blocking PKA abolished the effect of cAMP on Sox9, Sf1, and Gata4 expression, inhibiting PI3K/PKB impaired the effect on Sf1 and Gata4, and reducing MEK1/2 and p38 MAPK activities curtailed Gata4 increase. SOX9 and SF1 translocated to the nucleus after incubation with cAMP. Mutations of the SOX9 or SF1 sites, but not of GAT4 or AP1 sites, precluded the response of a 3,063-bp AMH promoter to cAMP. In conclusion, in the Sertoli cell line SMAT1 cAMP upregulates SOX9, SF1, and GATA4 expression and induces SOX9 and SF1 nuclear translocation mainly through PKA, although other kinases may also participate. SOX9 and SF1 binding to the AMH promoter is essential to increase the activity of the AMH promoter in response to cAMP.

Abstract:
Metabotropic glutamate receptors (mGluRs) include receptors sensitive to L-2-amino-4-phosphonobutyrate (L-AP4) and 1S,3R-1-aminocyclopentane-1,3-dicarboxylic acid (1S,3R-ACPD). To determine whether 1S,3R-ACPD is an agonist at retinal L-AP4 receptors, whole cell voltage clamp recordings were obtained from mudpuppy ON bipolar cells in a superfused retinal slice and L-AP4 and 1S,3R-ACPD were bath applied. Both compounds evoked similar outward currents which reversed near 0 mV and were accompanied by an increased input resistance. Responses to both agonists washed out in parallel suggesting they act through the same second messenger pathway(s). Inhibitors of cGMP-PDE activity suppressed responses to both L-AP4 and 1SR,3RS-ACPD, suggesting that both compounds activate cGMP-PDE. Responses to 1S,3R-ACPD were occluded by prior activation of L-AP4 receptors, but not blocked by the non-AP4, mGluR antagonists, L-aminophosphonopropionic acid (L-AP3) or 4-carboxy-3-hydroxyphenylglycine (4C3H-PG). These results indicate that 1S,3R-ACPD is an agonist at L-AP4 receptors. 1S,3S-ACPD and 4C3H-PG evoked outward currents similar to L-AP4 suggesting they may also be L-AP4 receptor agonists. Using the b-wave of the ERG as an assay for ON bipolar cell responses, concentration/response curves were obtained for ACPD enantiomers. The rank-order potency of ACPD enantiomers at L-AP4 receptors in ON bipolar cells is similar to their rank-order potency at non-AP4, mGluRs in brain which suggests that the receptors possess similar binding sites and may be members of a common receptor family.

Abstract In ruminants, conceptus interferon-τ (IFNT) production is necessary for maintenance of pregnancy. We examined the role of protein kinase A (PKA) in regulating IFNT expression through the activation of Ets2 in JAr choriocarcinoma cells. Although overexpression of the catalytic subunit of PKA or the addition of 8-bromo-cAMP had little ability to up-regulate boIFNT1 reporter constructs on their own, coexpression with Ets2 led to a large increase in gene expression. Progressive truncation of reporter constructs indicated that the site of PKA/Ets2 responsiveness lay in a region of the promoter between −126 and −67, which lacks a cAMP response element but contains the functional Ets2-binding site and an activator protein 1 (AP1) site. Specific mutation of the former reduced the PKA/Ets2 effects by more than 98%, whereas mutation of an AP1-binding site adjacent to the Ets2 site or pharmacological inhibition of MAPK kinase 2 led to a doubling of the combined Ets2/PKA effects, suggesting there is antagonism between the Ras/MAPK pathway and the PKA signal transduction pathway. Although Ets2 is not a substrate for PKA, lowering the effective concentrations of the coactivators, cAMP response element-binding protein-binding protein (CBP)/p300, known PKA targets, reduced the ability of PKA to synergize with Ets2, suggesting that PKA effects on IFNT regulation might be mediated through CBP/p300 coactivation, particularly as CBP and Ets2 occupy the proximal promoter region of IFNT in bovine trophoblast CT-1 cells. The up-regulation of IFNT in the elongating bovine conceptus is likely due to the combinatorial effects of PKA, Ets2, and CBP/p300 and triggered via growth factors released from maternal endometrium.

The scarcity of LPG gas that often occurs in Indonesia often makes Indonesian citizens uneasy. This scarcity led many researchers to conduct research that could reduce the dependence of Indonesian citizens with LPG gas. In this study, researchers conducted tests related to the use of premium steam as fuel. This test is carried out by varying the air supply, where this supply has been determined at 2 km/cm2 , 3 km/cm2 and 4 km/cm2 , in addition to the air supply this test also performs fire temperature measurements using thermocouples and data logers, where the location of the thermocouple has been located at points 1 cm, 3cm and 5 cm from the mouth of the nozzle. Based on the test results, the shape of the flame from variations in the air supply obtained the best and stable results were in the 3 km/cm2 air supply, while the other for the 2 km/cm2 air supply was found to be unstable fire and for which the 4 km/cm2 fire air supply looked large and unstable. Whereas for the highest temperature of the three placement points the thermocouple is located at the top point with a distance of 5 cm from the mouth of the fire with an average temperature of around 900oC and the lowest temperature is at 1 cm from the nozzle mouth, where the temperature is at 400oC to 550oC, in addition The temperature stability is in the air supply of 3 km/cm2

Abstract The present study deals with the application of X-ray powder diffraction (XRPD) analysis as a technique for identification of the forms of clopidogrel bisulphate (CLP) present in both the active pharmaceutical ingredients (API) and tablets, specifically addressing the question of whether API converts to another form after 12 months of storage. The investigation of the possibility of phase transitions occurring over a temperature range spanning from room temperature to the melting point, both in tablets and pure CLP, were also performed. Tablet samples were observed for the changes in their structure using polarizing optical microscopy, which was also used to determine the melting points of tablet samples. The results gained during this work confirm that XRPD is applicable for API and tablets testing. This is particularly important if we take into account that the method can be used during stability studies, i.e. in order to test the quality of tablets during the validity period.

Abstract:
This thesis deals with the development of multilingual for web browsers Mozilla Firefox and Google Chrome. The purpose of the extension is to enable the automated management of provided consents to the processing of personal data on websites using the Transparency and Consent Framework. Extension was developed on the basis of knowledge about this framework and about legal norms GDPR and ePrivacy Directive, which deal with the protection of personal data. Knowledge of the method of developing extensions for web browsers using WebExtensions was also used during the implementation. During testing, consent was successfully enforced in 96,2 % of tested websites in Mozilla Firefox. In Google Chrome, success has been achieved in 82,1 % of tested websites. The banner requiring consent was not displayed in 33 % of websites in Mozilla Firefox and in 31,1 % of websites in Google Chrome.

O histórico de contaminação por metais no Vale do Rio Ribeira de Iguape é bem conhecido, com evidências de transporte destes contaminantes ao Complexo Estuarino Lagunar de Cananéia-Iguape (CELCI), adsorvidos ao material em suspensão. Assim, os metais podem alcançar o setor sul do CELCI, podendo acumular nos sedimentos e nos organismos filtradores. Neste estudo foi realizada uma avaliação de risco ecológico no CELCI associado à presença de metais oriundos das antigas áreas de mineração do Alto Ribeira, utilizando diferentes linhas de evidência. Os resultados foram avaliados de forma integrada, incluindo análises geoquímicas e biológicas (ensaios de toxicidade aguda, medidas de bioacumulação e biomarcadores bioquímicos em brânquias de bivalves). Os sedimentos apresentaram níveis moderados a altos de metais e toxicidade aguda, a qual foi associada, especialmente, com as concentrações de Cu, Pb e Zn. Da mesma forma estes metais foram encontrados em concentrações altas nos tecidos de bivalves. Os biomarcadores bioquímicos (MT, GSH, GST, GPx, GR, AChE, LPO e danos em DNA) indicaram a ocorrência de efeitos significativos nos bivalves. Portanto, os resultados mostram que os metais estão sendo carreados para a porção sul do estuário, causando bioacumulação e efeitos nos organismos aquáticos e, portanto, gerando riscos ecológicos para o CELCI.
The historic of contamination by metals in the Valley of Ribeira de Iguape River is well known, and there are evidences that these chemicals are carried to the Estuarine Complex of Cananeia-Iguape (ECCI), adsorbed on the suspended particles. Metals can thus reach the south of the ECCI, where they may accumulate in the sediments and filter feeding organisms. This study aimed to evaluate the ecological risks associated to the metals originated from the former mining areas of the upstream Ribeira de Iguape River, by using different lines of evidence, which included geochemical and biological analysis (acute toxicity tests, bioaccumulation and biochemical biomarkers in bivalves\' gills). The sediments presented moderate to high levels of metals and acute toxicity, which was correlated with the concentrations of metals (Cu, Pb and Zn). These elements were found in high concentrations in the bivalves soft tissues. The biochemical biomarkers (MT, GSH, GST, GPx, GR, AChE, LPO, and DNA damage) indicated the occurrence of significant effects in the bivalves. Therefore, the results showed that metals are carried into the South portion of the estuary, accumulating in the biota and causing negative effects to the aquatic organisms, producing thus ecological risks to the ECCI.

La crête neurale (CN) est une population de cellules pluripotentes migrantes spécifique des embryons de vertébrés. Sa formation est contrôlée par un réseau de régulations épistatiques conservées au cours de l’évolution impliquant les facteurs secrétés BMP, FGF et WNT. Ces facteurs induisent l’expression des déterminants de la bordure neurale qui vont, à leur tour, induire l’expression des déterminants de la crête neurale. Au cours de ma thèse, je me suis intéressée aux mécanismes d’induction de la CN et plus particulièrement au rôle joué par les facteurs de transcription AP2a et Hairy2 dans le réseau de régulations épistatiques contrôlant sa formation. Ces deux protéines sont exprimées précocement à la bordure neurale et interviennent dans le contrôle du développement de la CN. Mes travaux montrent qu’AP2a dans un premier temps joue un rôle central dans l’intégration des signaux FGF et WNT et dans l’induction de la bordure neurale. Dans un deuxième temps, il contrôle le développement de la CN indépendamment de son action sur le maintien de la bordure neurale. Ces travaux montrent aussi qu’Hairy2 intervient en aval de la voie FGF et en parallèle d’AP2a pour permettre le maintien de la bordure neurale. Il bloque la spécification des précurseurs de la CN et renforce l’expression des déterminants de la bordure neurale. Ces résultats ont permis d’enrichir le réseau de régulation épistatique contrôlant la formation de la CN, de révéler un nouveau rôle d’AP2 dans le développement de cette structure et de proposer un modèle du contrôle de l’équilibre entre le maintien d’une population de précurseurs des cellules de la crête neurale et l’initiation de leur spécification
The neural crest is a multipotent, migratory cell population of vertebrate embryos that give rise to a variety of cell derivatives. An evolutionary-conserved epistatic network, which involves secreted factor BMP, FGF and WNT; has been proposed to control neural crest formation. This gene network suggests secreted factors induce the expression of neural border specifiers that will in turn induce neural crest specifiers genes. During my doctoral training I have investigated early steps in the induction of neural crest cells, focusing on the role of transcription factors AP2 and Hairy2. AP2 and Hairy2 are both expressed in the presumptive neural crest domain and are required for neural crest induction. My work demostrates that AP2 activity controls sequential steps in the neural crest network. First, AP2 acts as a neural border specifier to integrate FGF and WNT signals and allow neural border formation. Second, AP2 acts as a neural crest specifier in neural crest formation independent from its earlier role in neural border establishment. Additionally, I showed that Hairy2 activity is required in parallel with AP2 and downstream of FGF signaling to allow neural border formation. Hairy2 both blocks neural crest specification and maintains neural border genes expression. Hairy2 also induces Delta1 expression leading to activation of Notch pathway in the adjacent cells and suggest an additional role in specification of this tissue. Taking together my results allowed us to place AP2 and Hairy2 at specific steps in the regulatory network, which controls neural crest formation. This serves to highlight a new role of AP2 in neural crest development. Furthermore, I propose a model to explain how the embryo sets up a balance between neural crest cell specification and maintenance of a progenitor pool

Nous avons souhaité, dans ce travail de Thèse, préciser l'impact de l'invalidation d'AIP sur la fonction sécrétoire et la prolifération des cellules somatotropes in vivo et explorer in vitro les voies de signalisation potentiellement impliquées dans la tumorigenèse hypophysaire AIP-dépendante. L'analyse du phénotype des souris Aip+/-, en particulier de la sécrétion pulsatile de GH, montre que, contrairement à l'Homme, les animaux mutés ne développent pas de gigantisme ni d'hypersécrétion de GH et que la pénétrance de la pathologie tumorale hypophysaire est beaucoup plus faible qu'initialement décrit. Les études réalisées sur les fibroblastes de patients mutés pour AIP et porteurs d'un adénome hypophysaire ainsi que sur les cellules somatolactotropes de rat GH3 révèlent que les mutations d'AIP altèrent l'activité transcriptionnelle d'AhR (Aryl hydrocarbon Receptor), mais affectent également la voie de signalisation de l'AMPc. Enfin, des mutations germinales du gène GPR101, récemment identifiées dans des cas d'acromégalie sporadique, ont aussi été trouvées chez des patients porteurs d'un adénome hypophysaire sporadique non somatotrope, sans association avec les mutations d'AIP. Ce travail a ainsi permis de préciser les conséquences des mutations d'AIP sur la fonction somatotrope et la signalisation d'AhR. Le rôle de l'AMPc dans la tumorigenèse hypophysaire AIP-dépendante sera évalué dans un nouveau modèle de souris transgénique
In this work, we investigated the effects of AIP deficiency in vivo on somatotroph cells, both at the secretory and proliferative levels and explored in vitro the signaling pathways potentially involved in AIP-dependent pituitary tumorigenesis. Phenotype analyzes of Aip+/- mice, especially of GH pulsatility, show that, unlike humans, mutant mice do not develop gigantism nor GH hypersecretion and present with a much lower penetrance of pituitary adenomas than initially described. In vitro studies in fibroblasts of AIP-mutation positive patients with pituitary adenomas and in somatolactotroph GH3 cells demonstrate that AIP mutations alter AhR (Aryl hydrocarbon Receptor) transcriptional activity and modify the cAMP pathway. Finally, GPR101 mutations, recently reported in patients with sporadic acromegaly, have also been identified in a small portion of patients with sporadic non-somatotroph pituitary adenomas, without any association with AIP mutations. This research work defines the consequences of AIP mutations on somatotroph cell function and AhR transcriptional activity. The role of cAMP signaling in AIP-related pituitary tumorigenesis will be further evaluated in a new transgenic mouse model

Oil well cementing materials consist of slurries of Special class Portland cement dispersed in water. Admixtures can be used to provide the necessary fluidity, so the material can be efficiently pumped down as well as penetrate porous rocks with controlled filter loss. Construction admixtures can be used to modify the properties of oil well cements provided they can withstand and hold their properties at the higher than ambient temperatures usually encountered in oil fields. In civil construction, superplasticizer play the role of dispersants that reduce the facto r of water cement improve mechanical properties and fluidity of the cement, whereas anti-segregation agents improve the workability of the slurry. In the present study, oil well cement slurries were produced adding both a dispersant and an anti-segregation agent conventionally used in Portland CPII-Z-32 RS cement aiming at materials for primary cementing and squeeze operations. Three basic aspects were evaluated: fluidity, filter loss and the synergetic effect of the admixtures at two temperatures, i.e., 27?C and 56?C, following API RP 10B practical recommendations. The slurries were prepared using admixture concentrations varying from 2.60 Kgf/m3 (0.02 gallft3) to 5.82 Kgf/m3 (0.045 galJft3) BWOC. The density of the slurries was set to 1.89 g/cm3 (15.8 Ib/gal). 0.30 to 0.60% BWOC of a CMC-based anti-segregation agent was added to the cement to control the filter loss. The results showed that the addition of anti-segregation at concentrations above 0.55% by weight of cement resulted in the increased viscosity of the folders in temperatures evaluated. The increasing the temperature of the tests led to a reduction in the performance of anti-segregation. At concentrations of 5.20 kgf/m3 (0,040 gallft3) and 5.82 Kgf/m3 (0,045 gal/ft 3) observed a better performance of the properties evaluated in the proposed system. At low temperature was observed instability in the readings of rheology for all concentrations of anti-segregation. Contents that increasing the concentration of anti?-segregation is limited concentrations greater than 0.55 % BWOC of the CMC in temperature analyzed. The use of the system with CMC promoted a good performance against the properties evaluated. The principal function of anti?-segregation was optimized with increasing concentration of superplasticizer, at temperatures above the 2rC. Nas concentra??es de 5,20 kgf/m3 (0,040 gal/p?3) e 5,82 Kgf/m3 (0,045 gal/p?3) observou-se um melhor desempenho das propriedades avaliadas no sistema proposto. Na temperatura ambiente foi verificado instabilidade nas leituras de reologia para todas as concentra??es do anti-segregante. Foi verificado que o aumento da concentra??o de anti-segregante ? limitada as concentra??es superior a 0,55 % em peso de cimento nas temperaturas analisadas. O uso do sistema contendo superplastificante, aliado ao anti-segregante promoveu um bom desempenho face ?s propriedades reol?gicas e de controle de filtrado das pastas. A fun??o principal do anti-segregante foi otimizada com o aumento da concentra??o do superplastificante, a temperaturas acima da ambiente. Por?m o aumento da concentra??o do agente dispersante nas pastas de cimento pode provocar o retardamento das pastas em temperaturas pr?ximas da ambiente. Abstract:
Chronic myeloid leukemia is a myeloproliferative disorder characterized by the presence of the Philadelphia chromosome, encoding a unique fusion gene BCR-ABL. The current first line treatment for patients diagnosed with CML involves administration of the ABL kinase inhibitor imatinib mesylate (IM). However, early relapses and acquired drug resistance remain a current impediment to successful treatment for many patients. This suggests the necessity for alternate treatment options which may include combination therapy targeting multiple vital proteins involved in the malignancy of the leukemia. AHI-1 (Abelson helper integration site 1) is a recently discovered oncogene that is highly deregulated in murine lymphomas and leukemias. AHI-1 displays a significant pattern of overexpression in a Philadelphia chromosome positive cell line K562 cells. To investigate AHI-1’s involvement in CML, AHI-1 was either stably overexpressed or suppressed in K562 cells. Interestingly, an increase in cellular proliferation and colony formation and a decrease in apoptosis were observed in the presence of IM when AHI-1 was overexpressed, while suppression of AHI-1 had the opposite effects. Phosphorylation and total protein expression levels of several proteins known to be involved in BCR-ABL signalling were quantified. Interestingly, elevated phosphorylation and total gene/protein expression levels of several of these proteins were observed when AHI-1 was overexpresessed, in particular NF-κB and JAK2/STAT5 displayed increased expression. Due to the strong effects AHI-1 had on the JAK2/STAT5 signalling cascade, we then inhibited JAK2 activity using a new JAK2 inhibitor, TG101209. AHI-1 overexpression led to a reduction in the cellular response to the inhibitor while suppression of AHI-1 caused an increase in sensitivity in viability, apoptosis, and colony forming cell assays. Finally, a combination of IM and TG101209 was examined in the same K562 cells lines. Results from suggest that using a combination treatment approach was more effective at inhibiting cellular viability and colony formation than either treatment alone. These findings together suggest that AHI-1 may play an important role in mediating cellular resistance to IM and TG101209 and activates several BCR-ABL signalling pathways, and that it may be a vital target in eradicating the malignant leukemic cells arising in CML.

"Prefață: Consiliere și mentorare cu impact Călăuzirea spirituală: de la „avva” Antichității la mentorul de azi Sfântul apostol Pavel le scria romanilor: „Dar cum vor chema pe Acela în care n-au crezut? Și cum vor crede în Acela despre care n-au auzit? Și cum vor auzi despre El, fără ca cineva să predice? Și cum vor predica, dacă n-au fost trimiși? Astfel, credința vine din cele auzite, iar cele auzite, prin Cuvântul Lui Dumnezeu.” (Romani 10: 14-15). Nimeni n-a plecat la propovăduire de capul lui. 1. Inițial, Isus „a chemat pe cine a vrut” (chemarea; vocația divină: gr. kaleo; lat. voco/are = a chema); 2. apoi, „ei au venit la El și a rânduit dintre ei 12, ca să-i aibă cu Sine” (răspunsul personal: voluntar și prompt [lat. protinus = îndată]; asumarea vocației apostolice și „ucenicia” alături de Isus timp de trei ani; gr. mathitevo = a fi discipol; a învăța; lat. discipulus); și 3. în final, Isus „i-a trimis să predice” (misiunea apostolică; gr. apo/stello; lat. mitto/ere = a trimite; missio/onis = trimitere; gr. kirysso = a propovădui; „a face o proclamație în calitate de herald”; un „mesager al Domnului”; gr. kyr = domn) [Marcu 3:13-14]. Chemare, ucenicie, trimitere..., toate acestea s-au petrecut „la împlinirea timpului” (Galateni 4:4), într-un moment de cumpănă al omenirii, cum ar zice E. Cioran: „Pe culmile disperării”, când „Poporul care stătea în întuneric, a văzut o mare Lumină și, celor care zăceau în ținutul și în umbra morții, le-a răsărit Lumina” (Matei 4:16) „transfigurării cosmice” (Cioran) mesianice. Isus n-a venit într-o lume pregătită să-L primească, ci într-o lume bulversată, disperată și în așteptare... Nu era o lume mai dreaptă, mai bună, mai credincioasă, mai morală, mai cultă, mai catehizată, mai primitoare decât cea de azi. Dar Isus n-a ținut cont de nimic din toate astea, ci pur și simplu a venit să-și îndeplinească „misiunea”: a învățat, a vindecat boli incurabile, a înviat persoane trecute în „viața de apoi”, a iertat, a exorcizat „demonii” multor patimi, a dat sens multor vieți, a provocat, a contestat formalismele Templului, cărturarilor și fariseilor, a declanșat furia mai marilor vremii și nu a fugit de supliciul și „rușinea” crucii. Într-un fel, la (răs)crucea vremurilor de azi, lucrurile par a se suprapune cu cele din timpul lui Isus. Trăim într-o lume complet debusolată, secularizată, nihilistă, agnostică, sceptică, indiferentă, relativistă, dezumanizată, lacomă și nesătulă, buimacă, parcă „fără istorie spirituală și fără viitor”, ostilă Celui de Sus și refractară oricărei morale, o epocă a indiscreției și lipsei de pudoare, a pornografiei, traficului de carne vie, vânzărilor de armament, droguri etc., fără sentimentul păcatului (Morale sans péché, dr. Hesnard, 1954), fără pic de rușine, fără valori, direcție, sens și destinație spirituală. Dumnezeu ne-a adus de la haos (abis, „tohu wa bohu” = o lume „fără formă și goală”; „fără cap și fără coadă”) la kosmos (ordine, viață, „căpătâi”), dar noi, parcă tributari „vocației entropiei”, mergem ireversibil către neant, nonsens și autodistrugere. Dumnezeu ne-a dăruit Viața, dar noi, incapabili să-i descifrăm farmecul, bucuria, valoarea și sensul, ne-o suprimăm sau ne-o irosim în nimicnicie. Ne-a dăruit Iubirea, dar ura, intoleranța și resentimentele ne stăpânesc. „Lumina a venit în lume, dar oamenii au iubit mai mult întunericul decât lumina, pentru că faptele lor erau rele.” (Ioan 3:19). Nu suntem cu nimic mai buni peste 2000 de ani decât atunci, dimpotrivă! Dar, în loc să stăm și să ne lamentăm continuu, mai bine căutăm soluții. Omul sfințește locul! Tuturor acestor provocări vor trebui să le facă față duhovnicii, mentorii, cateheţii și toți învățătorii spirituali de azi. O teologie de manual, scolastică, teoretică, stearpă și polemicile noastre confesionaliste sunt de mult depășite. Cum „imputa” ironic un student profesorului de dogmatică: „Dom’ profesor, cred că nici Dumnezeu nu știe despre El atâtea câte ați scris dumneavoastră în manualul acesta!” Termeni, dogme, erezii, speculații filosofice, dispute – la ce servesc toate astea? „Nimic nu e mai sărac decât cugetarea care, stând afară de Dumnezeu, filosofează despre Dumnezeu.” (Diadoh, episcop al Foticeei, sec. V). Azi, e nevoie de creștini autentici și mărturisitori adevărați (gr. martirevo = a mărturisi; de aici și termenul de „martir”) într-o relație vie cu Dumnezeu, „din interiorul Lui”, în Duhul Lui, nu doar de niște transmițători de cunoștințe teologice exterioare, mereu puși pe harță pentru monopolul (exclusivismul) și „drepturile de autor” asupra „adevărului” divin. Vremea polemicilor sterile a apus. Lumea nu mai are nevoie să afle „sexul îngerilor”! E nevoie de o Întâlnire adevărată cu Dumnezeu, nu doar la nivelul minții și speculațiilor teologice, ci și la nivelul inimii și al spiritului. Un consilier sau mentor spiritual asta va trebui să facă, să-l conducă pe omul zilelor de azi la Marea Întâlnire existențială și spirituală cu Dumnezeu. „Oare poate un orb să călăuzească pe un alt orb? Nu vor cădea amândoi în groapă?” (Luca 6:39). Avem nevoie de duhovnici adevărați, de consilieri adevărați, de mentori spirituali adevărați, cu pregătire umană, teologică și viață spirituală pe măsură, care să înțeleagă omul de azi cu toate problemele, slăbiciunile și ezitările lui, nu de triumfaliști „îmbelferiţi ai spiritului”, de legaliști, formaliști, moraliști de mucava care să te trateze de pe poziții de superioritate, suficiență și omnisciență. Avem nevoie de călăuze spirituale umane, calde, autentice, vii care să „nu ne dea lecții de morală ieftină” și de „auto-izbutită soteriologie”. Nu e nevoie de „experți” care să ni se insinueze drept „modele” și „păstori” ai unei turme paraplegice și oarbe, ci de oameni adevărați cu inimă caldă și mare, cu care să putem intra într-un dialog real, de la om la om, de la suflet la suflet, cu Dumnezeul cel Viu prezent în mijlocul nostru. Predica publică e una, relația de îndrumare spirituală, de consiliere sau de mentorat este alta. Este o relație particulară, de la om la om, cu o forță de impact soteriologic imensă: o adevărată „chirurgie spirituală” în care omul își oferă mentorului spiritual inima deschisă ca „pe tavă” cu toată încrederea. De măiestria și responsabilitatea duhovnicului, consilierului sau mentorului spiritual depinde „reușita operației” și felul în care este „suturată” incizia. Din păcate, mulți au rămas profund dezamăgiți și debusolați după „întâlnirea” cu unele „pseudo-călăuze” sau „mercenari” implicați în acest câmp psihologic și spiritual extrem de fragil. Predica te poate atinge parțial, dar cuvântul adresat direct, de la inimă la inimă, n-are cum să nu-ţi trezească sentimente, întrebări, idei, aspirații puternice. Aceasta și era relația directă dintre avva (maestrul spiritual) și ucenic, încă din antichitatea creștină, și către asta tindem și acum, spre o neo-evanghelizare sau re-încreştinare autentică (nu îndoctrinare sau prozelitism) a omului, prin comunicarea față în față cu mentorul, un om mai experimentat, pregătit (psihologic și spiritual), cu învățăcelul, în curs de formare, pentru a-l smulge din marasmul și pericolele societății atee despre care vorbeam mai sus. Mai trebuie doar să înțelegem că, azi, această relație spirituală de călăuzire este cumva „pe picior de egalitate”. Nimeni nu se consideră superior nimănui, doar „maestrul” spiritual este dispus și disponibil să (se) investească în „discipol”, în beneficiul orientării lui existențiale și mântuirii. Aceasta înseamnă „artă” și autenticitate spirituală! Volumul de față, Consiliere și mentorare cu impact, ca, de altfel, întreaga colecție Autentic, urmăreşte obținerea unor mărturii adevărate ale unor persoane autentice, care să le fie de folos în orientarea și formarea oamenilor dispuși să-și caute calea și rostul lor spiritual. Așa cum, deja, ne-a obișnuit apariția primului volum, Copilărie și parentalitate cu impact, structura celui de față este la fel de coerentă, variată și interesantă cum, nu mă îndoiesc nicio clipă, vor fi și cele ce vor urma. Os aditivos são utilizados em produtos cárneos com diversas finalidades e, dentre os principais, estão o sal (NaCl), que apresenta ação antimicrobiana, realça o sabor dos produtos, extrai e solubiliza proteínas; e o fosfato, que atua sinergicamente com o NaCl, para reter a água do produto. Entretanto, o consumo em excesso de ambas substâncias está atrelado a diversos problemas de saúde, o que gera um problema de saúde pública e grande demanda por produtos com maior saudabilidade. A redução pode ocorrer de várias formas, mas pincipalmente por substituição parcial por outros sais, ou através de tecnologias verdes, tendo como principais a alta pressão hidrostática (APH), campo elétrico pulsado (CEP), ultrassom (US) e água eletrolisada básica (AEB). O trabalho objetivou destacar os principais e mais recentes estudos voltados à aplicação de APH, CEP, US e AEB como estratégia na redução de NaCl e fosfato em diferentes produtos cárneos, bem como explicar os princípios de cada uma das tecnologias e os possíveis fenômenos que possibilitaram a redução desses aditivos.

This chapter begins with the introduction of Robert Kafin and his law partner, Ed Needleman who had been talking to Ted Hullar from the Sierra Club, David Newhouse from the Adirondack Mountain Club (ADK), and Courtney Jones, a philanthropist from the Lake Champlain town of Westport at Harold Hochschild's Great Camp in October 1971. The chapter describes Kafin's career shift and how he learned what was going on in the Adirondacks. Kafin saw that the National Environmental Policy Act (NEPA) and other new federal laws were broadly written and not widely understood, especially by law firms north of Albany. The chapter reviews how the Adirondack Pack Agency Act added a big new state law to that pile. These laws could be used to shape or stop development. The chapter then shifts to describe how the Adirondack Project established Kafin & Needleman as the people to talk to if you wanted to block development in the Adirondacks. It also reviews the implications of Horizon and Ton-Da-Lay development for the Adirondack Park Agency (APA). Ultimately, the chapter examines the power and influence of some lawyers in relation to the APA.

Consumers are largely unaware regarding the use being made to the data that they generate through smart devices, or their GDPR-compliance, since such information is typically hidden behind vague privacy policy documents, which are often lengthy, difficult to read (containing legal terms and definitions) and frequently changing. This paper describes the activities of the CAP-A project, whose aim is to apply crowdsourcing techniques to evaluate the privacy friendliness of apps, and to allow users to better understand the content of Privacy Policy documents and, consequently, the privacy implications of using any given mobile app. To achieve this, we developed a set of tools that aim at assisting users to express their own privacy concerns and expectations and assess the mobile apps’ privacy properties through collective intelligence.

In Fall 2020, the American Talent Initiative (ATI), an alliance of high-graduation-rate colleges and universities committed to expanding access and opportunity for low- and middle-income students, established its newest community of practice (CoP) focused on academic equity. Together, the 37 CoP members explore topics related to creating equitable academic communities. One such area of focus is how institutions can more effectively utilize data to enhance equity-related projects. In January 2021, members participated in a webinar discussion on this topic, during which CoP representatives presented on how they have leveraged data in their academic equity work. This case study builds on a presentation given by Dr. Jill Sible, Associate Vice Provost for Undergraduate Education and Professor of Biological Sciences at Virginia Tech, titled, “Using data to fuel inclusive excellence at Virginia Tech.”
