Abstract:
The postembryonic development of the gill parasite, Lamproglena clariae, infecting the sharp tooth catfish, Clarias gariepinus was established from egg culture and artificial infection of fish under laboratory conditions. Like most fish parasitic copepods, L. clariae has a direct life cycle utilizing only a single fish host species. Adult post metamorphosis females produce two egg strings. The mean number of eggs in each egg string was 52. Three naupliar and first copepodid stages were obtained in culture while two copepodid stages, cyclopoid and adult specimens were obtained after artificial infection of catfish in aquaria. First stage nauplii were globular in shape and densely filled with yolk. Nauplii lacked a perforation for the mouth and masticatory parts of the appendages, all of which indicate that they do not feed. Body architecture of the first copepodid stage of L. clariae is similar to that of all other copepods in the number and kind of somites: a cephalothorax with five appendages, three thoracic somites, one abdominal somite and furca rami. This seems to be a conserved morphology among the copepods. The three naupliar and three copepodid stages are described and compared to related copepods.

Abstract:
Abstract KRAS is the most frequently mutated oncogene in cancer and drives uncontrolled growth through hyperactivation of the MAPK pathway. Significant progress has been made in the past several years to directly target KRASG12C with the FDA approval of sotorasib and the reported clinical activity of adagrasib (MRTX849). Despite these remarkable breakthroughs, additional therapies that enhance the depth and duration of response to KRASG12C inhibitors provide the opportunity to build upon the initial progress. SOS proteins are guanine nucleotide exchange factors (GEFs) that transduce receptor tyrosine kinase (RTK) signaling from the cell surface and facilitate the activation of RAS family proteins. In addition, SOS1 is a target of negative feedback signaling following RAS-mediated activation of the RAF-MEK-ERK cascade. Thus, SOS proteins represent a significant therapeutic node that maintains RAS pathway equilibrium as well as oncogenic signaling dynamics. Here we highlight the discovery and preclinical evaluation of MRTX0902, a potent, selective, and orally bioavailable inhibitor of SOS1 presently in IND-enabling studies. A structure-based approach was used to identify a novel chemical series that disrupts the protein-protein interaction between SOS1 and KRAS, thereby preventing SOS1-mediated GTP-exchange on GDP-bound KRAS. Considering MRTX849 preferentially binds to inactive GDP-bound KRASG12C, targeting SOS1 in this genetic context increases the ability of MRTX849 to bind and inhibit KRASG12C. The combination of MRTX0902 with MRTX849 enhances the depth and durability of an anti-tumor response when compared to MRTX849 alone in pre-clinical KRASG12C tumor models. MRTX0902 augments additional targeted therapies across a variety of RAS-addicted tumors, indicating that SOS1 inhibition is effective against a broad spectrum of mutations within the MAPK pathway. Furthermore, drug-anchored CRISPR experiments with MRTX0902 and MRTX849 uncovered a previously underappreciated functional role of the SOS1 paralog, SOS2, in KRAS-addicted tumors. In addition to aiding in the understanding of SOS and RAS family signaling dynamics, these studies implicate SOS2 as a potential cancer drug target in the context of SOS1/KRASG12C inhibition. In summary, we have used a structure-based approach to discover a SOS1 inhibitor that augments the anti-tumor activity of MRTX849 and additional targeted MAPK pathway inhibitors. We anticipate our findings to translate into the clinic and make an impact in patients with RAS-addicted tumors. Citation Format: John M. Ketcham, Shilpi Khare, Niranjan Sudhakar, David M. Abstract:
Usher Syndrome (USH) is the first cause of deafness blindness in humans. 3 USH clinical types (USH1-3) are defined. Type III clinical form patients hearing loss is not congenital, but progressive, usually occurring during or after adolescence, and the presence of vestibular defects and age of onset of retinitis pigmentosa is variable. I studied the role of clarin-1, causing USH3A. CLRN1 gene encodes clarin-1. The characterization of Clrn1 mutant mice revealed that clarin-1 is essential for the structural organization and function of the presynaptic channels Cav1.3 Ca2+ at the inner hair cell ribbon synapse and for the distribution of postsynaptic AMPA receptors. The viral-mediated transfer of the intact Clrn1 into the clarin-1 mutant mice in cochlea durably prevented synaptic defects and occurrence of the hearing loss. I also explored the role of clarin-2 another member of the clarin family, the absence of which leads to hearing loss. The clarin-2 mutant mice have a progressive, early-onset hearing loss. Our findings demonstrate a key role for clarin-2 in mammalian hearing, providing insights into the interplay between mechano-electrical transduction and stereocilia maintenance. Finally, I studied the compensatory mechanisms involving the two clarins which might conceal important functions in the inner ear. The inactivation of both Clrn1 and Clrn2 impairs prematurely the vestibular function, total loss of mechano-electrical transduction & extreme disruptions of the hair bundle stereocilia. Further elucidation of the mechanisms through which the two clarins interact, and the importance of such interactions in the vestibular and cochlear systems is underway

Abstract:
Usher syndrome (USH) causes a combined deafness-blindness in humans. At least nine causative genes are known. While the analysis of USH knockout mice has shed light on the origin of the auditory deficit, the causes of vision loss are still unclear. Nevertheless, USH1B protein, myosin VIIa, appears to contribute to intracellular traffic in photoreceptor cells. To better understand the role of this myosin in the retina, I studied the functions of its interacting partner, spectrin βV. We found that spectrin V, along with USH1 proteins, participates in intracellular transport by coupling motor proteins (myosin VIIa, kinesin II, dynein/dynactin complex) to the cargoes en route towards the outer segment of photoreceptor cells. Evidence from comparative studies in frog and mouse inner ear, biochemical assays and phylogenetic analyses point to cargo trafficking to and from the apical cell region, as the likely ancestral function of this spectrin. Our analyses also suggest that evolutionary pressures in the mammalian lineage drove the recruitment of spectrin βV to the lateral wall of auditory outer hair cells, probably to support a new function: electromotility. Finally, I explored the origin of hearing loss in Usher syndrome of type III (USH3). So far, the only causal gene known is CLRN1, which codes for clarin-1. The comparative characterization of two Clrn1 mouse mutants revealed that clarin-1 is required for the maturation and maintenance of the hair bundle in the hair cells. Moreover, our results indicate that clarin-1 is also essential to cluster the voltage-gated Ca2+ channels in close proximity to the exocytotic machinery of the ribbon synapse of inner hair cells

The present study aimed to demonstrate the relevance of the technical restructuring of a work, that is, a musical reelaboration to the particularities of new media, focusing on the bass clarinet as a central instrument, triggering a future improvement and adaptation to the new resources of the instrument and its language. Besides this, the purpose of the work was to point out the proposition of an experiment of this instrument to an original non-idiomatic repertoire. In order to do so, the methodological procedure used observed references as reworkings already performed and understood the ways in which one was carried out. The work contributed to the theoretical and practical development of the research in reelaborations for bass clarinet, in this way, favoring the arrangement of a new experimental vector, based on the reelaborations made. Thus, it is concluded that this research, in contrast to the rigor and fidelity to which the reelaboration is usually applied, emphasized the process as a creative and dialogical tool.

Abstract:
Thesis (D.M.A.)--Boston University. This item includes the dissertation paper, handouts, as well as a video of the February 26, 2017 lecture and clarinet performance by Karen Luttik.
Géza Frid (1904-1989) was a significant Hungarian-born Dutch composer and pianist of Jewish descent. His compositional style was highly regarded in the Dutch musical scene of the 20th century; his music has been programmed on multiple occasions by the Concertgebouw Orchestra and in 1949 and 1954 won the City of Amsterdam Music Award. Major influences on Frid’s musical development started in his native Hungary where he studied with Béla Bartók and Zoltán Kodály before his move to the Netherlands. In 1972, he composed an unusually lovely concerto for Bb, A, Eb and bass clarinets (Op. 82) and dedicated it to George Pieterson, principle clarinet with the Concertgebouw Orchestra. Scholarly discussions of Géza Frid’s clarinet music are not to be found in either the US based International Clarinet Society’s The Clarinet, or in the Dutch based De Klarinet. Intensive World Cat library searches have yielded no recordings of this piece, and currently there is not even one recording of Frid’s Concerto for Clarinets on YouTube or in the Naxos Music Library. The Concerto for Clarinets is a significant addition to the standard clarinet repertoire and needs to be introduced to the world wide clarinet community. Furthermore, save for some short selections on the Géza Frid website set up by his son, Arthur Frid and a Wikipedia article, no translations exist of Géza Frid’s autobiographical material. His life story was exceedingly interesting, having been a Jewish composer during the World War II years who was not allowed to perform or compose for years during the German occupation of the Netherlands. Luckily he survived the war years by going into hiding; he managed as part of the musician’s resistance to find ways to perform and work illegally giving clandestine concerts and falsifying documents. These were exceedingly dangerous risks, yet ones Géza Frid and his fellow musicians were willing to take because of their music. Frid published two autobiographical books and numerous articles for the Dutch music magazine Mens en Melodie, (People and Melody) revealing his deep musical insights; especially those relating to the music of the Concerto need to be summarized and translated to English from Dutch. This paper provides a general overview of the historical aspects of Géza Frid’s life, his WWII experiences, and his position as one of many persecuted Jewish musicians at the time. Géza Frid’s autobiographical information relating to his personal friendships with Bartók and Kodály is of interest when considering his music. Summaries and some translations are made of his two Dutch language autobiographical books, In 80 Jaar de Wereld Rond (Around the World in 80 Years) and Oog in Oog Met… (Eye to Eye With…). A basic analysis of Frid’s Concerto for Clarinets is provided regarding form, the various instrument appearances, and a special feature invented by George Pieterson called the ‘tremolo special’. Since this concerto was specifically composed for the Reformed Boehm system clarinets which George Pieterson used, a discussion of the differences between the French, German and Reformed Boehm clarinet systems is included. The performance portion of this project is a historically informed performance of this piece on the specific models of clarinets for which it was written. George Pieterson passed on in April 2016, and this project is a fitting tribute of his teaching to a generation of professional clarinetists in the Netherlands including the author.

This work is a proposed interpretative work for clarinet and Brazilian music band concert entitled "A Vida pela Flor" written by the conductor and composer Antônio Joaquim Langsdorf Naegele. Naegele had three of his compositions published by FUNARTE. The music "A Vida pela Flor" is atypical for making use of a fantasy within the context of repertoire for band. Its structure is divided into: Introduction, 1st theme with two variations, 2nd theme with five variations and codetta, differing from the simple structure of most works focused for band, like march, polkas, mazurkas, waltzes, among others. "A Vida pela Flor" stands out for its virtuosity, if taken into account that at the time was run by an instrument with fewer technical resources (Müller System - 13 keys), a remarkable fact. The piece has simple sentences, but with great richness of technique: scales, arpeggios, leaps between different registers of the clarinet, and variation of articulation. Such sentences, or fragments of them, are comparable to those found in the exercises in the "Complete Method for Clarinet" by Elenore Hyacinthe Klosé (1808-1880) which makes us believe that divulging this work will enrich the clarinet repertoire. Knowing that this work is not well divulged among teachers, composers and clarinetists, this study may arise interest to demonstrate the cultural richness that it may bring to our context.

Abstract:
The makers, design, construction, and origin of the Baroque clarinet are discussed in this chapter. Two tables list chronologically all extant two- and three-key clarinets by country, and include maker, and city when known, maker’s dates, date of clarinet, number of clarinets, nominal pitch, and collection. There are 36 two-key and 22 three-key clarinets, made in Germany, Belgium, the Netherlands, Austria, Czech Republic, France, Sweden, and Finland. The earliest documentation for the invention of the clarinet is reviewed, and how and why Johann Christoph Denner improved the chalumeau and invented the clarinet is proposed, on the basis of previous studies of mechanical inventions. Biographies are provided for each clarinet maker when information was found.

Abstract:
The Data Governance Act was proposed in late 2020 as part of the European Strategy for Data, and adopted on 30 May 2022 (as Regulation 2022/868). It will enter into application on 24 September 2023. The Data governance Act is a major development in the legal framework affecting CLARIN and the whole language community. With its new rules on the re-use of data held by the public sector bodies and on the provision of data sharing services, and especially its encouragement of data altruism, the Data Governance Act creates new opportunities and new challenges for CLARIN ERIC. This paper analyses the provisions of the Data Governance Act, and aims at initiating the debate on how they will impact CLARIN and the whole language community.

Abstract:
The paper presents the work in progress for a PhD thesis about preposition incorporation in the Bulgarian BulTreeBank WordNet. Being one of the most polysemous parts of speech, prepositions are still relatively challenging for NLP and are usually missing in wordnets. A preposition semantic classification, a model for preposition synsets and synset relations are proposed. The planned applications of the prepositions and the directions for future processing are introduced.

Abstract:
This paper describes a project aimed at creating a new resource family of multilingual and multimodal resources centered around the concept of “Linguistics of self”, that is personal re-flections on the role of languages in shaping one’s identity. Language portrait silhouettes, drawing bilingualism, and linguistic autobiographies are different types of resources that share this common feature. We describe the resources and the criteria for their metadata annotation, focusing in particular on linguistic autobiographies, where the writer explicitly reflects on the relationship between him/herself and language. These genres are fruitfully used in different educational settings, and research has shown that they help to uncover the social, affective, and psychological dimensions of language learning. The potential of a multilingual and mul-timodal collection is discussed starting from data collected in Italy and Norway.

Abstract:
This article presents a work-in-progress doctoral project that explores measuring text readability in Sesotho, a Bantu language spoken by more than 10 million speakers across Southern Africa. The main project adopts a classical readability formulas approach to text readability analysis. We aim to adapt nine existing readability metrics into Sesotho using English as a higher-resourced helper language. So far, five resources have been developed as part of the study. The rule-based and the TeX-based syllabification systems, the syllable annotated word list, and the grade 12 exam reading comprehension and summary writing corpus have been published on the South African Centre for Digital Language Resources' (SADiLaR) online repository. The machine-translated corpus is still under development. This article describes the progress of the PhD project by overviewing the basic digital language resources developed for the project. The metrics under consideration for adaptation into Sesotho are also briefly discussed.

Full text
Abstract:
This paper discusses the encoding, validation and development of language resources of the completed ParlaMint I and on-going ParlaMint II CLARIN projects, which centre on the collaborative development of a large set of interoperable corpora of parliamentary proceedings. It focuses on the ParlaMint encoding and the GitHub development platform and the evaluation of their use by project partners. We introduce the use of TEI ODD for the encoding guidelines and validation schemas. We motivate and explain using Git to develop and maintain the encoding schemas, validation and conversion scripts and valid samples of the corpora. Apart from revision control, issues, and publishing documentation, the GitHub platform also supports integrating code execution with pull requests, which significantly automates the data submission process. The paper also presents the results of a survey on the use of TEI and Git in the ParlaMint projects among the project participants. Overall, participants were mostly positive about their experiencewith TEI and Git, although some difficulties were reported. Some partners also expressed doubts about whether the current scheme is flexible enough to support encoding the unique features of sometimes drastically different parliamentary systems. On the other hand, reactions to Git were very positive in terms of information and feedback received via GitHub Issues, effectiveness in the communication process, and plans to use Git in the future. However, there was less agreement on whether the requirements and workflows were adequately explained. The reported difficulties will serve as a basis for further Git workflow optimisation in ParlaMint.

Abstract:
We investigate changes in the use of four Swedish words from the fields of design and archi- tecture. It has been suggested that their meanings have been blurred, especially in governmental reports and policy documents, so that distinctions between them that are important to stakeholders in the respective fields are lost. Specifically, we compare usage in two governmental public reports on design, one from 1999 and the other from 2015, and additionally in opinion responses to the 2015 report. Our approach is to contextualise occurrences of the words in different representations of the texts using word embeddings, topic modelling and sentiment analysis. Tools and language resources developed within the SweClarin infrastructure have been crucial for the implementation of the study.

Abstract:
WebLicht is a workflow engine that gives researchers access to a well-inhabited space of natural language processing tools that can be combined into tool chains to perform complex natural language analyses. In this paper, we present WebLicht-Batch, a web-based interface to WebLicht's chainer back-end. WebLicht-Batch helps users to automatically feed large input data, or input data of multiple files into WebLicht. It disassembles large input into smaller, more digestible sizes, feeds the resulting parts into WebLicht's pipelining and execution engine, and then assembles the results of such processing into files that preserve the usual input-output dichotomy.
