Abstract:
Let Ckn,l be a simple closed k-curves with l elements in Zn and W:=Ckn,l∨⋯∨Ckn,l︷m-times be an m-iterated digital wedges of Ckn,l, and F(Conk(W)) be an alignment of fixed point sets of W. Then, the aim of the paper is devoted to investigating various properties of F(Conk(W)). Furthermore, when proceeding with this work, this paper addresses several unsolved problems. To be specific, we firstly formulate an alignment of fixed point sets of Ckn,l, denoted by F(Conk(Ckn,l)), where l(≥7) is an odd natural number and k≠2n. Secondly, given a digital image (X,k) with X♯=n, we find a certain condition that supports n−1,n−2∈F(Conk(X)). Thirdly, after finding some features of F(Conk(W)), we develop a method of making F(Conk(W)) perfect according to the (even or odd) number l of Ckn,l. Finally, we prove that the perfectness of F(Conk(W)) is equivalent to that of F(Conk(Ckn,l)). This can play an important role in studying fixed point theory and digital curve theory. This paper only deals with k-connected digital images (X,k) such that X♯≥2.

Abstract:
This study deals with a competitive knowledge network (CKN) where participants desire to pass their personally important exams, and henceforth want to contribute and seek knowledge about exam-related issues they need to solve. Besides, CKN members are basically competing with each other since they all aim to pass the same exams. Therefore, from the contributor’s perspective, if the advisor fears that his/her advice can be detrimental to his or her own benefits, knowledge sharing (KS) intention within the CKN may be hindered by their expectation of competitive advantage. However, few studies were done about the effects of such fear and a sense of competition on KS continuance intention in the context of CKN. In this sense, our study aims to elucidate the effects of competition and fear on KS continuance intention in a mobile CKN. By using 296 valid questionnaires, we obtained very meaningful conclusions such that competition is a driving force rather than an obstacle to KS among CKN members, and fear also enhances members’ sustainable KS intention.

Abstract Yeast products have found much favor within companion animal nutrition. A recently developed proprietary process has introduced an enhanced yeast biomass from Candida utilis (merchandised as SylPro®) into the animal food marketplace. Candida utilis has been shown to be a valuable protein in canine, swine and aquaculture diets; however, no previous research has evaluated its use in feline diets. Therefore, the current objective was to determine the nutrient digestibility of diets containing SylPro® yeast biomass (SYL) as the primary protein source relative to soybean meal (SOY), pea protein concentrate (PEA), and chicken meal (CKN) fed to cats. Apparent total tract digestibility (ATTD) of diets were estimated using Titanium dioxide as an indigestible marker. The DM ATTD for SYL was lower (P < 0.0001) than CKN (86.39 vs. 87.43%, respectively) but similar to both SOY (85.66%) and PEA (86.54%). The OM ATTD of SYL, SOY, and PEA were not different (P < 0.0001) and were each lower than CKN (average of 89.60 vs. 91.16%, respectively). The CP ATTD for SYL (89.9%) was not different from the other three treatments (P = 0.0200). Crude fat ATTD of SYL was lower (92.52%; P < 0.0001) than all treatments with PEA being greatest (94.82%) and CKN and SOY intermediate (average of 93.84%). The SYL and CKN had higher TDF ATTD (average of 66.20%; P < 0.0001) relative to PEA and SOY (average of 49.79%). The GE ATTD of SYL was lower than CKN (90.18 vs. 90.97%, respectively; P = 0.0154) but similar to both SOY (90.18%) and PEA (90.30%). In conclusion, all diets were highly digestible but CKN preformed the best. The SYL diet was similar to CKN for ATTD of CP and TDF. The SYL diet was also similar to both SOY and PEA for ATTD of DM, OM, CP and GE.

AbstractThe main purpose of this paper is to establish several general Caffarelli–Kohn–Nirenberg (CKN) inequalities on Carnot groups G (also known as stratified groups). These CKN inequalities are sharp for certain parameter values. In case G is an Iwasawa group, it is shown here that the {L^{2}}-CKN inequalities are sharp for all parameter values except one exceptional case. To show this, generalized Kelvin transforms {K_{\sigma}} are introduced and shown to be isometries for certain weighted Sobolev spaces. An interesting transformation formula for the sub-Laplacian with respect to {K_{\sigma}} is also derived. Lastly, these techniques are shown to be valid for establishing CKN-type inequalities with monomial and horizontal norm weights.

L'insufficienza renale cronica (CKD) è associata ad un rischio cardiovascolare più elevato rispetto alla popolazione generale: fattori come uremia, stress ossidativo, età dialitica, infiammazione, alterazioni del metabolismo minerale e presenza di calcificazioni vascolari incidono fortemente sulla morbosità e mortalità per cause cardiovascolari nel paziente uremico. Diversi studi hanno verificato il coinvolgimento dei progenitori endoteliali (EPC) nella malattia aterosclerotica ed è stato dimostrato che esprimono osteocalcina, marcatore di calcificazione. Inoltre, nella CKD è presente una disfunzione in numero e funzionalità delle EPC. Attualmente, il ruolo delle EPC nella formazione delle calcificazioni vascolari nei pazienti in dialisi non è stato ancora chiarito. Lo scopo della tesi è quello di studiare le EPC prelevate da pazienti con CKD, al fine di determinarne numero e fenotipo. È stato anche valutato l'effetto del trattamento in vitro e in vivo con calcitriolo e paracalcitolo sulle EPC, dato il deficit di vitamina D dei pazienti con CKD: il trattamento con vitamina D sembra avere effetti positivi sul sistema cardiovascolare. Sono stati valutati: numero di EPC circolanti e la relativa espressione di osteocalcina e del recettore della vitamina D; morfologia e fenotipo EPC in vitro; effetti di calcitriolo e paracalcitolo sull’espressione di osteocalcina e sui depositi di calcio. I risultati dello studio suggeriscono che il trattamento con vitamina D abbia un effetto positivo sulle EPC, aumentando il numero di EPC circolanti e normalizzandone la morfologia. Sia calcitriolo che paracalcitolo sono in grado di ridurre notevolmente l’espressione di OC, mentre solo il paracalcitolo ha un effetto significativo sulla riduzione dei depositi di calcio in coltura. In conclusione, il trattamento con vitamina D sembra ridurre il potenziale calcifico delle EPC nell’uremia, aprendo nuove strade per la gestione del rischio cardiovascolare nei pazienti affetti da CKD.
Chronic Kidney Disease (CKD) is associated to an increased cardiovascular risk compared to the general population: factors like uremia, oxidative stress, dialytic vintage, inflammation, mineral metabolism dysregulation and vascular calcifications greatly affect cardiovascular morbidity and mortality in CKD. Several papers reported the involvement of endothelial progenitor cells (EPCs) in atherosclerosis: it was also demonstrated their expression of osteocalcin, a marker of calcification. Moreover, a dysfunction in number and functionality of EPCs is also detectable in CKD. Currently, the role of EPCs in the pathogenesis of vascular calcifications in patients with CKD is not clear. The project presented herein aimed to study EPCs from CKD patients and determine their number and phenotype. Moreover, it was evaluated the effect of treatment with calcitriol and paricalcitol on EPCs, both in vivo and in vitro, because CKD patients suffer from vitamin D deficiency and it was reported that vitamin D has beneficial effects on the cardiovascular system. The number of circulating EPCs and their expression of osteocalcin and vitamin D receptor, morphology and phenotype of EPC in vitro, effects of calcitriol and paricalcitol on expression of osteocalcin and calcium deposition where evaluated. The results suggest that treatment with vitamin D has beneficial effects on EPCs, by increasing their number and improving morphology. Both calcitriol and paricalcitol could strongly reduce the expression of osteocalcin by EPCs, whereas only paricalcitol was able to reduce the deposition of calcium in cell cultures. In conclusion, treatment of CKD patients with vitamin D seems to reduce the calcific potential of EPCs in uremia, highlighting a putative protective role of vitamin D therapy against vascular calcifications.

Abstract:
Carcinomas de mama são heterogêneos e consistem de diversos tipos celulares. Perfis de expressão gênica usando DNA microarrays identificaram quatro subtipos moleculares fundamentais baseados na expressão de receptores hormonais (estrógeno e progesterona) e de fator de crescimento epidérmico (HER2) (luminal tipo A, luminal tipo B, tumores expressando somente HER2 e triplos negativos) refletindo a heterogeneidade molecular dos carcinomas. Sugeriu-se que esta heterogeneidade advém da presença de células tronco tumorais com a capacidade de se diferenciar ao longo de vias divergentes e outros estudos sugeriram que a presença destas células tronco tumorais pode ser evidenciada pela análise fenotípica de CD44 e CD24. Nosso objetivo foi detectar a freqüência de CD24 e CD44 isolados ou combinados, analisados por imunoistoquímica e sua associação com os subtipos moleculares e com diversos marcadores biológicos em 95 casos de carcinoma ductal infiltrativo organizados em um microarranjo tissular (TMA). Realizamos determinações imunoistoquímicas de CD44, CD24, citoqueratinas (CK5, CK6, CK18), claudina 7 e Ki67. Subgrupos moleculares foram definidos pela expressão imunoistoquímica de RE, RP e HER2. Resultados: Os tumores apresentaram uma maior freqüência dos grupos luminais (49,5%) atribuído à alta expressão de RP ou RE (47,4%), e freqüência menor de tumores triplo negativos (21,5%) e HER2 (9,5%). Os fenótipos CD44+CD24- e CD44-/CD24+ estavam respectivamente presentes em 8,4% e 16,8% dos tumores e o fenótipo duplamente positivo foi predominante (45,3%). Ausência de ambas as proteínas foi evidente em 6,3% dos tumores. Tumores com fenótipo CD44+CD24- (definido como um marcador de células tronco tumorais por estudos in vitro) foram mais comuns em tumores triplos negativos mas não demonstraram nenhum tipo de associação com características clinico-patológicas e demais marcadores. Este fenótipo não foi expresso nos tumores HER2 positivos. O fenótipo duplamente positivo CD44+CD24+ mostrou-se mais freqüente nos subtipos luminais ou com alta expressão de HER2. Os fenótipos (CD44-CD24+ e CD44-CD24-) não mostraram associação com os subgrupos. Tumores expressando CD24+ isolado, com grande freqüência deste marcador (74,7%), mostraram significativa associação com positividade do RE, RP e Ki67 e uma significância marginal com marcadores de diferenciação luminal (CK18 e claudina 7, p = 0,14). Nenhuma associação foi observada com tumores CD44+ quando analisado isoladamente. A expressão de claudina 7 e Ki67 não mostrou associação com os subgrupos e a expressão de CK5 apresentou uma tendência a uma maior negatividade nos subtipos luminais e uma freqüência maior de positividade nos tumores HER2 e triplo negativos. De outro lado, associação da freqüência da expressão positiva de CK18 nos subgrupos luminais foi estatisticamente significativa (p = 0,003). Para se determinar se CD24+ e CD44+ e seus subtipos combinados poderiam afetar a sobrevida global e o intervalo livre da doença preparamos curvas de sobrevida de acordo com Kaplan-Meier que foram analisadas estatisticamente (log rank test). A mediana do período de seguimento das pacientes do nosso estudo foi de 4,8 anos (0,36 10,9 anos). Estas análises não demostraram influência dos fenótipos CD44+CD24- ou CD44+ sobre a sobrevida global ou intervalo livre de doença, mas observamos uma tendência a um prognóstico mais favorável. Interessantemente tumores HER2 positivos não expressaram este fenótipo, sugerindo que outros marcadores de células tronco caracterizam estes tumores. O fenótipo CD44-CD24+ mostrou-se mais freqüente nos tumores luminais, mas não apresentou correlação com marcadores clínico-patológicos ou biológicos analisados. Não houve diferenças significativas com respeito a sobrevida global ou intervalo livre de doença . A expressão de CD24+ isolado associou-se a expressão dos marcadores de diferenciação celular e a uma diminuição do intervalo livre de doença. A sobrevida livre de doença (10 anos) indicou uma percentagem de 94,1% para CD24- e 72,1% para os pacientes CD24+ enquanto a sobrevida global foi de 84,2% para os pacientes CD24- e 72,1% para os pacientes CD24+. Citoqueratinas (CK5, CK18) e Ki67 não influenciaram a sobrevida e o intervalo livre de doença. No entanto a expressão positiva de claudina 7, embora não associada à sobrevida global, foi estatisticamente associada ao decréscimo do intervalo livre da doença (p = 0,05). Conclusão: As características dos tumores CD44+CD24- e sua tendência a associação um prognóstico mais favorável parecem não estar de acordo com as propriedades descritas na literatura para células tronco e enfatizam a necessidade de outros marcadores. A determinação da freqüência de CD44+ e claudina 7 positiva pode contribuir para a análise do prognóstico em carcinoma de mama
Background: Breast carcinomas consist phenotypically of diverse cells and exhibit intra tumoral heterogeneity being stratified in several subgroups based in gene expression profiles or histochemical biomarkers. It was suggested that this heterogeneity is derived in part from the transformation of different subsets of cancer stem cells (CSC) in each intrinsic subgroup. The presence of CSC can be evidenced by phenotypic analysis of CD44 e CD24. This study aimed to identify the CD24 and CD44 immunophenotype within invasive ductal breast carcinoma (IDC) subtypes and determine its influence on prognosis as well as its association with the expression of Ki67, citokeratins (CK5, CK6 and CK18) and claudin-7. Methods: Immuno expression of CD44 and CD24 alone or in combination was investigated in 95 IDC cases arranged in a tissue microarray (TMA). The association with intrinsic subgroups defined as luminal A (ER+, PR+, HER2-), luminal B (ER and or PR+, HER2+), HER2 subtype (ER-, PR-, HER2+) and triple negative (ER-, PR-, HER2-), and the other markers and prognosis was analyzed. Results: CD44+CD24- and CD44-CD24+ were respectively presents in 8.4% and 16.8% of the tumors, a lack of both proteins was detected in 6.3%, while CD44+CD24+ was determined in 45.3% of the tumors. Although there was no significant correlation between subgroups and different phenotypes, the CD44+CD24- phenotype was more common in the basal subgroups but the frequency of this subtype has not been associated with clinical characteristic or biological markers. The phenotype was absent in HER2 tumors whereas luminal tumors are enriched in CD44-CD24+ and CD44+CD24+ cells which did not show associations with clinical/biological markers features. There was also no significant association of the subtypes with the event free (DFS) and overall survival (OS) but the CD44+CD24- phenotype showed a more favorable prognostic as compared to CD44-CD44+ phenotype that showed a worse prognosis (p = 0.26) (median follow up, 4.8 years) CD44+ alone was evident in 57.9%, while CD24+ was positive in 74.7% of the tumors, the latter showing a significant association with ER, PR and Ki67 and a marginal association with CK18 and claudin-7. Expression of claudin-7 and Ki67 did not associate with the cancer subgroups, while a positive association between CK18 and the luminal subgroups was found. CD44+ was not significantly associated with OS (p = 0.684) and DFS (p = 0.386) whereas CD24+ expression was also no significantly associated with OS (p = 0.32) but was associated with a decrease in DFS (p = 0.07). CK5, CK18 and Ki67 expression had no influence in OS or DFS, however claudin-7 positive although not statistically associated with OS, was associated with reduced DFS (p = 0.05). Conclusions: The heterogeneity of cells with several CD44CD24 expression may indicate the presence of different stem cell populations. Ocurrence of CD44+CD24- phenotype is more common in triple negative tumors and lower in tumors of luminal type and absent in HER2 tumors. Although not associated significantly with patho-biological markers or OS and DFS, the CD44+CD24- phenotype has a tendency to be a favorable prognostic marker in breast cancer raising the possibilty that the putative tumorigenic ability may no be restricted to cells of this phenotype. The presence of CD44-CD24+ may indicat a worse prognosis. CD24+ was associated with ER, PR, Ki67and showed a marginal association with CK18 and claudin-7. CD24 and Claudin-7 positivity were the only biological markers associated with reduced DFS. These two investigated markers can be used to improve the assessement of prognosis in breast cancer

Abstract:
Este trabalho tem por objetivo desenvolver o processo de retificação a alta velocidade de superligas utilizando rebolos de CBN vitrificados. Para tanto, foram estudadas diversas condições de dressagem, ou seja, as influências dos parâmetros de dressagem no desempenho do rebolo CBN de liga vitrificada. A partir da escolha desses parâmetros, foi também determinado o desgaste do rebolo em diversas condições de velocidade de avanço, e a qualidade superficial das peças retificadas. Foi utilizado um sistema de mapeamento topográfico do rebolo, baseado na emissão acústica emanada da interação entre rebolo-peça, para o monitoramento do processo. Como resultado foi determinada a influência da velocidade de dressagem e da relação de velocidades na rugosidade da peça retificada. Ficou determinado também, o desgaste do rebolo de CBN em função do volume de material retificado para os diferentes materiais das superligas testadas. Através do sistema de mapeamento topográfico, determinou-se os padrões gráficos do comportamento do rebolo de CBN vitrificado. Sendo assim, verificou-se que o sucesso da aplicação de rebolos CBN vitrificados na retificação de superligas depende da qualidade superficial do rebolo após a dressagem e da escolha certa dos parâmetros de retificação, os quais dependem da constituição das ligas testadas.
The aim of this work is to develop a high speed grinding process for super alloys using vitrified CBN wheels. For that, dressing conditions were studied in order to determine its influence on the CBN wheel performance. From the chosen parameters, the wheel wear and the ground surface quality were measured for different infeed speeds. A topographical mapping system based on acoustic emission was used to monitor the grinding process. As a result, the best dressing speed and speed ratio which generated the smallest surface roughness values could be found. The CBN wheel wear was measured and its ratio to the ground volume was evaluated. Through the mapping system, some visual patterns were obtained defining the grinding wheel behavior when dressing and grinding. Thus, the success vitrified CBN wheel application for super alloys depends basically on the wheel topography after dressing and the grinding parameters, which are dose related to the work material composition.

Several works study the performance of Content Delivery Networks (CDNs) under various network infrastructure and demand conditions. Many strategies have been proposed to deal with aspects inherent to the CDN distribution model. Though mostly very effective, a traditional CDN approach of statically positioned elements often fails to meet quality of experience (QoE) requirements when network conditions suddenly change. CDN adaptation is a key feature in this process and some studies go even further and try to also deal with demand elasticity by providing an elastic infrastructure (cloud computing) to such CDNs. Each Content Provider (CP) gets served only the amount of storage space and network throughput that it needs and pays only for what has been used. Some IaaS providers offer simple CDN services on top of their infrastructure. However, in general, there is a lack of PaaS tools to create rapidly a CDN. There is no standard or open source software able to deliver CDN as a service for each tenant through well-known managers. A PaaS CDN should be able to implement content delivery service in a cloud environment, provision and orchestrate each tenant, monitor usage and make decisions on planning and dimensioning of resources. This work introduces a framework for the allocation of resources of a CDN in a multi-tenant environment. The framework is able to provision and orchestrate multi-tenant virtual CDNs and can be seen as a step towards a PaaS CDN. A simple dot product based module for network change detection is presented and a more elaborate multi-tenant resource manager model is defined. We solve the resulting ILP problem using both branch and bound as well as an efficient cache slicing algorithm that employs a three phase heuristic for orchestration of multi-tenant virtual CDNs. We finally show that a distributed algorithm with limited local information may be also offer reasonable resource allocation while using limited coordination among the different nodes. A self-organization behavior emerges when some of the nodes reach consensus.
Vários trabalhos estudam o desempenho de Redes de Distribuição de Conteúdo (CDN) em diferentes condições e demanda e de infraestrutura. Muitas estratégias têm sido propostas para lidar com aspectos inerentes ao modelo de distribuição de CDN. Embora essas técnicas sejam bastante eficazes, uma abordagem tradicional de elementos estaticamente posicionados numa CDN muitas vezes não consegue atender os requisitos de qualidade de experiência (QoE) quando as condições da rede mudam repentinamente. Adaptação CDN é uma característica fundamental neste processo e alguns estudos vão ainda mais longe e tentam lidar com a elasticidade da demanda, proporcionando uma infraestrutura elástica (computação em nuvem) para a CDN. Cada provedor de conteúdo obtém apenas a quantidade de armazenamento e de rede necessários, pagando apenas pelo efetivo uso. Alguns provedores IaaS oferecem serviços de CDN sobre suas estruturas. No entanto, em geral, não existe padrão ou softwares de código aberto capazes de entregar serviços de CDN por meio de gerenciadores. Uma CDN PaaS deve ser capaz de fornecer um serviço de entrega de conteúdo em um ambiente de nuvem, provisionar e orquestrar cada tenant, monitorar uso e tomar decisões de planejamento e dimensionamento de recursos. Este trabalho apresenta um framework para alocação de recursos de uma CDN em ambiente multi-tenant. O framework é capaz de provisionar e orquestrar CDNs virtuais e pode ser visto como um passo em direção a uma PaaS CDN. Um módulo baseado em simples produto escalar para detecção de mudanças na rede é apresentado, bem como um modelo mais elaborado de gerenciamento de recursos. Resolvemos o problema ILP resultante dessa abordagem por meio de um algoritmo de divisão de cache que emprega uma heurística em três fases para a orquestração de CDN virtuais. Por fim, mostramos uma outra abordagem com algoritmo distribuído que usa informação local e que também oferece uma alocação razoável usando coordenação limitada entre os diferentes nós. Um comportamento de auto-organização surge quando alguns desses nós chegam a um consenso.

Abstract:
The receiver operating characteristic (ROC) curve is one of the most commonly used methods to compare the diagnostic performances of two or more laboratory or diagnostic tests. In this thesis, we propose semi-empirical likelihood based confidence intervals for ROC curves of two populations, where one population is parametric while the other one is non-parametric and both populations have missing data. After imputing missing values, we derive the semi-empirical likelihood ratio statistic and the corresponding likelihood equations. It has been shown that the log-semi-empirical likelihood ratio statistic is asymptotically chi-square distributed. The estimating equations are solved simultaneously to obtain the estimated lower and upper bounds of semi-empirical likelihood confidence intervals. Simulation studies are conducted to evaluate the finite sample performance of the proposed empirical likelihood confidence intervals with various sample sizes and different missing rates.
