Abstract:
Abstract The chr 5p15.33 genomic locus encoding telomerase reverse transcriptase (TERT) is structurally complex, with several highly polymorphic intronic variable number tandem repeats (VNTRs). A germline VNTR located in TERT intron 6 (VNTR6-1, 38 bp repeat unit) has previously been associated with TERT alternative splicing but its potential role in regulating cellular functions and disease risk is unclear. To functionally characterize VNTR6-1, we deleted this region (1-2 kb) using CRISPR/Cas9 in UMUC3 (bladder cancer cell line). Cell count growth tracking of parental and VNTR6-1 knockout (VNTR6-1 KO) cells via automated live-cell monitoring revealed different relative growth rates under serum starvation vs. normal conditions, suggesting that VNTR6-1 might be sensitive to growth factors present in culture media. Evaluation of growth in media with full vs. charcoal-stripped (CS) serum, showed significant differences, with higher cell counts in parental vs. VNTR6-1 KO in CS-serum (P=4.1e-3), parental vs. parental in full (P=1.57e-4) or CS-serum (P=4.76e-2), but not in parental vs. VNTR6-1 KO in full serum (P=0.378). After 5-6 days of growth, cells in CS-serum also rapidly began outgrowing cells in full serum. We observed that VNTR6-1 KO dampened the outgrowth response in cells lacking growth factors in CS-serum. Proliferation-specific growth assays tracking the dilution of an incorporated dye in cells by flow cytometry further showed that VNTR6-1 KO leads to more pronounced proliferation differences due to the presence or absence of serum-supplied factors. Overall, cell growth patterns appear to be sensitive to serum in interaction with VNTR6-1, suggesting a potential VNTR6-1-dependent response to extracellular signaling. In human populations, we have identified two VNTR6-1 groups: Short (24-27 copies) and Long (40.5-66.5 copies). In the PLCO dataset (n=100,445 cancer cases and controls), logistic regression showed that VNTR6-1-Long group was associated with a decreased risk in male-prevalent cancers such as bladder and prostate, but an increased risk in female-prevalent cancers such as breast, endometrial and ovarian. These results support a link between the TERT region, and potentially VNTR6-1, with hormone signaling and cancer risk. Motif analysis within VNTR6-1 predicted binding sites for several transcription factors, including those associated with development and cell growth. Together, these results suggest that VNTR6-1 may be involved in regulating cell growth, potentially by facilitating responses to extracellular signaling pathways. Additionally, given the location of VNTR6-1 within a multi-cancer GWAS region, our findings provide functional context for the role of this polymorphic repeat in differential risk and outcomes in certain cancers. Abstract:
The practice of using active shutters in the modern automotive industry is analyzed in this work, and the high efficiency of such systems is emphasized. It is also noted that by using active shutters the aerodynamic drag of the car can be reduced by 6–10 %. The reduction in the engine’s warm-up time provides faster heating of the car interior. All this helps to save fuel and reduce emissions of harmful substances into the atmosphere. The possibility of utilizing the radiator’s active shutters with two autonomously controlled sections is considered. CFD numerical modelling is used to conduct the research, and the potential of the proposed active shutters design is determined. The research has shown that besides the high efficiency, the proposed shutters system has a simpler design and reliability associated with several factors. Due to the vertical arrangement of the slats, the shutters do not reduce the efficiency of the engine’s cooling system and the air conditioning system in the passenger compartment. Unlike in the existing designs, in the proposed system the cooling air supply is regulated by separate opening or closing of two independent sections.

In traffic flow analysis several regimes are distinguished, such as congested and noncongested flow conditions. Indications exist that driving behavior differs by regime and that it may change discontinuously between regimes. In contrast most traffic flow models used today basically assume the same car-following behavior irrespective of the traffic flow regime. It is hypothesized that, because of this deficiency, these models do not always perform satisfactorily. To clarify this issue, differences in car-following between congested and noncongested flow are analyzed with data from two sites on Dutch freeways. It is shown that, at the same speeds, passenger car drivers follow with smaller headways in noncongested than in congested flow. Car-following of truck drivers does not show differences between regimes. Microscopic distance gap-speed models are established for several road-user classes, valid for each of the two flow regimes. To show the improvements resulting from these new microscopic relationships, the latter are implemented in a microscopic simulation model with which macroscopic patterns in traffic flow are modeled. The macroscopic findings produced with the regime-specific car-following rules show a considerable improvement in modeling performance.

A linear acceleration car-following model has been developed for realistic simulation of traffic flow in intelligent transportation systems (ITS) applications. The new model provides continuous acceleration profiles instead of the stepwise profiles that are currently used. The brake reaction times of the drivers are simulated effectively and are independent of the simulation time steps. Chain-reaction times of the drivers are also simulated and perception thresholds are incorporated in the model. The preferred time headways are utilized to determine the simulated drivers’ separation during car-following. The features of the model and the realistic vehicle simulation in car-following and in stop-and-go conditions make this model suitable to ITS, especially to autonomous intelligent cruise-control systems. The car-following algorithm is validated at microscopic and macroscopic levels by using field data. Simulated versus field trajectories and statistical tests show very strong agreement between simulation results and field data.

Abstract This study retraces the development of the Later Jin/Qing artillery corps between 1631 and 1642, examines the factors that led to its success, and evaluates its military and socio-political impact. The newly established artillery corps, under the direction of the talented Hong Taiji, learned effectively from its participation in sieges and developed the relevant military knowledge. By 1642, it had turned from an auxiliary force into a full-fledged unit that played a decisive role in siege warfare. The success of the Qing artillery corps greatly facilitated the Qing conquest of China (1644–1683). Moreover, the military performance of the artillery corps in the time period led to the multiple expansions and the ultimate institutionalization of the Han Eight Banner Army (baqi Hanjun).

Purpose: This study is aimed at measuring the efficiency of 37 OECD countries for 2020 using the Data Envelopment Analysis (DEA) method. Besides, it is aimed at ranking the efficient decision making units by using the super-efficiency DEA model. Design/methodology/approach: In the study, analyses were carried out with input-oriented Charnes, Cooper and Rhodes (CCR), input-oriented Banker, Charnes and Cooper (BCC) models and super-efficiency models of these models by using 4 inputs and 3 outputs. Findings: As a result of the analysis, 14 countries in the CCR model and 20 countries in the BCC model were efficient. According to the results of the super-efficiency models, the efficient countries were ranked. Research limitations/implications: The limitations of the study are the analyses are based on input-oriented DEA models and the research was conducted in OECD countries. Originality/value: Performance evaluation of health systems has gained importance in recent years. Many countries are making efforts to improve their health systems. Due to epidemics such as COVID-19, OECD countries, like many countries around the world, have increased the share of health expenditures in GDP. Because of this situation, the evaluation of the performance of OECD countries in the field of health has emerged as a very important research topic.

Urbarz cieszyńskiego klucza dóbr książęcych, funkcjonujący w literaturze jako „urbarz cieszyński z 1621 r.”, nie zachował się w oryginale, ale jedynie w odpisie z 1653 r. W urbarzu został odnotowany Adam Matloch, który nabył grunt w Marklowicach w 1644 r., co każe postawić pytanie, czy zawarte tam dane oddają stan z 1621 r. Szczegółowa analiza odsłania złożoność informacji zamieszczonych w tym urbarzu, a także metodę pracy nad sporządzaniem odpisów tego rodzaju źródeł – gdzie uwzględniano aktualizacje i pomijano fragmenty skreślone. Okazuje się, że wbrew utartej nazwie urbarz stanowi kompilację zapisów z lat 1621–1643, a większość wpisów oddaje stan z ok. 1640 r.

INTEGRATION, TSIS/CORSIM, and WATSim can simulate traffic operations on mixed arterial and freeway networks. Comparisons are presented here using three small networks in Honolulu for which detailed, simultaneous, and contemporaneous flow conditions are known. The models produced reasonable and comparable results on most network links. Of the three software programs, only INTEGRATION can simulate U-turns, but it also is least able to model complex signal operations. TSIS/CORSIM is best at replicating lane-changing behavior, but its percentile input for off-ramps is both inconvenient and inaccurate. WATSim needed the least calibration for producing good results, but its animation is inferior and its capacity-based car-following parameters are undesirable. The default parameters off all three software programs never offered satisfactory results.

Abstract. Natural methane (CH4) emissions from wet ecosystems are an important part of today's global CH4 budget. Climate affects the exchange of CH4 between ecosystems and the atmosphere by influencing CH4 production, oxidation, and transport in the soil. The net CH4 exchange depends on ecosystem hydrology, soil and vegetation characteristics. Here, the LPJ-WHyMe global dynamical vegetation model is used to simulate global net CH4 emissions for different ecosystems: northern peatlands (45°–90° N), naturally inundated wetlands (60° S–45° N), rice agriculture and wet mineral soils. Mineral soils are a potential CH4 sink, but can also be a source with the direction of the net exchange depending on soil moisture content. The geographical and seasonal distributions are evaluated against multi-dimensional atmospheric inversions for 2003–2005, using two independent four-dimensional variational assimilation systems. The atmospheric inversions are constrained by the atmospheric CH4 observations of the SCIAMACHY satellite instrument and global surface networks. Compared to LPJ-WHyMe the inversions result in a~significant reduction in the emissions from northern peatlands and suggest that LPJ-WHyMe maximum annual emissions peak about one month late. The inversions do not put strong constraints on the division of sources between inundated wetlands and wet mineral soils in the tropics. Based on the inversion results we diagnose model parameters in LPJ-WHyMe and simulate the surface exchange of CH4 over the period 1990–2008. Over the whole period we infer an increase of global ecosystem CH4 emissions of +1.11 Tg CH4 yr−1, not considering potential additional changes in wetland extent. The increase in simulated CH4 emissions is attributed to enhanced soil respiration resulting from the observed rise in land temperature and in atmospheric carbon dioxide that were used as input. The long-term decline of the atmospheric CH4 growth rate from 1990 to 2006 cannot be fully explained with the simulated ecosystem emissions. However, these emissions show an increasing trend of +3.62 Tg CH4 yr−1 over 2005–2008 which can partly explain the renewed increase in atmospheric CH4 concentration during recent years.

Purpose: The aim of the article was to obtain high-quality connectors for the automotive industry and antenna structures. Design/methodology/approach: For the welding joint the duplex steel and parameters of TIG welding process were chosen. The mechanical properties of the joint were carefully checked. Findings: Relations between process parameters and the quality of welds. Research limitations/implications: In the future, it can be suggested to investigate the effect of micro addition of nitrogen in gaseous shielding mixtures of the welding process. Practical implications: The development allows to obtain high-quality welded joints made of duplex steel. The solution can be implemented in the automotive industry or for the production of antenna structures. Social implications: Modifying the welding method will not affect the environment and production management methods. Originality/value: The article is addressed to manufacturers of stainless steel for automotive industry and to manufacturers of antenna instrumentation. AbstractWe present here two letters of Voetius to Maarten Schoock, professor of philosophy at the university of Groningen. They almost exclusively treat both men's dispute with Descartes, and especially the proceedings the French philosopher instituted against Schoock because of his anti-Cartesian work Admiranda methodus (Utrecht 1643). The first letter (April 1644, published here for the first time), is of particular interest, for Voetius described in detail the reasons for publishing Admiranda methodus in it. The second one (21 January 1645 o.s., only partly known) is a letter of advice, or, as Descartes saw it, a shameless attempt to entice Schoock to give false evidence before his judges (AT VIIIB 248-249). The last document published here is Schoock's Corte memorie. The margins of this manuscript show Voetius's own comments.

Résumé Dans son « petit traité en Anglais » sur La nature humaine (1640) et, ensuite dans Leviathan (1647-1650), Thomas Hobbes examine et redéfinit les passions. En prenant appui sur le concept de motion, mouvement, il les conçoit comme pensées : les passions sont des perceptions qui passent par le cerveau, mais pour se diriger vers le coeur. Une fois dans le coeur, elles provoquent du plaisir en favorisant le « mouvement vital », vital motion, ou bien de la peine, en lui faisant obstacle. La colère (anger), affirme-t-il, est le dépassement d’un obstacle, non la réponse à une insulte (contempt), comme le disaient, à tort, les Anciens. Cette critique est significative. Car orge, comprise comme la peine pour une offense injustifiée, accompagnée du désir de prendre sa revanche, était pour Aristote la passion politique par excellence. C’est parce que nous nous fâchons que nous nous révoltons et que notre révolte se justifie. C’est en colère que nous nous mobilisons dans une guerre juste. Qu’elle se présente comme une explosion tumultueuse ou comme un sourd ressentiment, orge est pour les Anciens la réaction raisonnée d’un acteur social, qui souffre dans son corps aussi bien que dans l’image sociale qu’il s’est donné et au nom d’une attente de justice : on ne reconnaît pas cette personne pour celui qu’il veut être dans le monde. Dans la transition d’une théorie de l’animal politique, naturellement enclin à la sociabilité (Aristote), à une vision mécaniste de la nature humaine, faite de mouvement et de compétition (Hobbes), c’est le fondement anthropologique du politique qui se trouve repensé. L’entrelacs de droits, habitudes et corps qui donnait forme à des émotions complexes se défait. Une comparaison entre la colère des Anciens et la colère de Modernes illustre cette dislocation.

The paper presents a theory of body collision with pit corner. The theory can be successfully applied in approaching tasks coping with the orientation of details in technological processes and control systems, the motion of elements in mechanisms having a gap, collisions in motion of different sport objects, the analysis of car accidents etc. The applied theory deals with car motion after collision with a specific obstacle i.e. pit corner. The study is based on linear and angular momentum theorems and two restitution coefficients of normal impulse from both sides of the obstacle. The obtained results can be used for assessing road accidents in case of car collision with one or more obstacles. The object of contact may involve hitting a plane foundation, the ditch and edging of a highway or a rock in the field.

Recently, a new statistical procedure was developed that enables fast, accurate, and robust estimation of composite headway distributions, such as Branston’s generalized queueing model (GQM). Until now, the new procedure had only been applied to aggregate vehicular flow. In this paper, the estimation procedure is extended to headway observations segregated according to vehicle type and period of the day. Consequently, the parameters of a new mixed-vehicle-type headway distribution model based on Branston’s headway model can be estimated. Distinction of vehicle type and sample periods provides additional insight into the plausibility of the headway distributions and parameter values, as well as into the car-following behavior of the distinct vehicle classes varying across the different periods. The estimation procedure was applied to traffic data collected on a two-lane rural road in the Netherlands. Comparison of the estimated headway distributions with real-life data shows that headway distributions can be realistically replicated with the Pearson-III-based mixed-vehicle-type GQM. Inter-pretable differences between the morning, noon, and evening sample periods and between passenger cars, unarticulated trucks, and articulated trucks are found. In addition, passenger-car equivalents for both articulated trucks and unarticulated trucks were determined from the parameter estimates.

For examination of road accidents often calculations are being performed according to mathematical equations having many variables, real values of which are of stochastic nature and they depend on many accident factors. Therefore it is important to know limits of determination of final values of every being analysed parameter of car movement and their probabilistic characteristics. This paper presents some cases of simulation of some road situations by mathematical methods of calculations, which now are widely used for evaluation of accuracy of values of the calculated parameter (in this paper ‐ the braking distance). Also here is presented some analysis of these methods. The paper presents the following cases of car braking: braking with making attempts to prevent a collision with another car, braking through impact‐to‐pedestrian prevention and conventional (rectilinear) car braking. The main attention in the analysis of the mentioned road situations is paid to veracity of the car braking distance, i.e. to limits of variation of values of this parameter and also to influence of accidental factors on the final result which is being looked for. Some conclusions on the expediency of application of the described methods of calculations to analysis of road accidents are present.

AbstractThis article examines how the handling of Irish affairs at the civil-war court was observed, commented upon, and probably influenced, by a man who had been at the heart of Irish government during the previous decade. It argues that the presence of Sir George Radcliffe, arguably Thomas Wentworth, earl of Strafford's closest advisor, at the royalist court in Oxford was significant in a number of respects. His surviving correspondence reveals the extent to which he was able to advise and support the newly-appointed lord lieutenant of Ireland, the marquis of Ormond. His letters also cast light on the politics of the civil-war court, in particular on how he, as an essentially second-level figure, worked flexibly with both secretaries of state. This article argues that Radcliffe's discussions with secretary of state, George Lord Digby, provided an important means by which Strafford's influence could make itself felt at the Oxford court, particularly during the 1644 negotiations with the Confederate commissioners. The pursuit of forceful, contentious policies by the former Irish administration and its contemptuous attitude towards critics, however, ensured that Radcliffe's presence at Oxford was probably unwelcome to many at court and failed to advance his career.

Background:The diagnosis for adult-onset Still’s disease (AOSD) is still based on nonspecific symptoms and laboratory data, and had necessity to rule out several infectious, autoimmune or malignant diseases.Objectives:This study aimed to elucidate the efficiency of inflammatory markers, including systemic immune-inflammation index (SII); C-reactive protein (CRP)-to-albumin ratio (CAR), albumin-to- glubulin ratio (AGR), prognostic nutritional index (PNI), and ferritin-to-ESR ratio (FER) for evaluation of diagnostic or prognostic factors in AOSD.Methods:The medical records of patients with suspected AOSD between January 1999 and June 2019 were collected and retrospectively analyzed. Among 225 patients, 61 patients received another diagnosis, such as infection and malignancy, and 164 patients were newly diagnosed to AOSD. The values of SII, CAR, AGR, PNI, and FER were compared with AOSD and non-AOSD groups. Receiver operating characteristic (ROC) curve analysis was performed to evaluate the diagnostic significance of inflammatory markers. Correlations between inflammatory markers and disease activity index were analyzed.Results:A total of 164 patients who diagnosed AOSD had higher values of SII, CAR, and FER as well as lower values of AGR and PNI. For AOSD diagnosis, the area under the curve (AUC) obtained from the ROC curve were 0.859 (95% CI=0.806-0.911) for SII, 0.769 (95% CI=0.702-0.837) for CAR, 0.749 (95% CI=0.615-0.782) for AGR, 0.699 (95% CI=0.675-0.823) for PNI, and 0.764 (95% CI=0.693-0.834) for FER with cut-off value of 2195.7, 1.80, 1.38, 48.8 and 17.0, respectively. The SII had the largest AUC, and FER and SII each had the highest sensitivity (70.9%) and specificity (91.5%).
