Abstract:
The pyrolysis kinetic behaviors and reaction mechanism of polycarbosilane (PCS) were characterized by means of XRD, SEM and on-line TG-DSC-FTIR-MS coupling technique, which showed that the pyrolysis process accorded with the F2 integral model (300-600°C), and the values of apparent activation energy (Ea) and pre-exponential factor (A) were 56.10KJ/mol and 10.84, respectively. During the pyrolysis process, PCS was converted into amorphous inorganic solid from organic polymer with breakage and rearrangement of chemical bonds, then to well-defined crystal structure of good thermal stability. Furthermore, the weight loss occurred mainly among 300°C and 800°C with CH4, (CH3)4Si,(CH3)3SiH,(CH3)2SiH2and other silane gases releasing from pyrolysis product leading to the conversion from organic polymers to inorganic ceramic. The pyrolysis product was converted into β-SiC crystal completely at 1400°C, and the crystallization of α-SiC phase occurred after 1550°C which might influence the stability of SiC matrix. Abstract:
The paper reports the methods for the synthesis of new cluster compounds of trans-tetrachlorodi--carboxylates of dirhenium(III) with dipeptides of the glycyl series. The compositions and the structures were determined by electron absorption and IR spectroscopies as well as by elemental analysis for the following newly synthesized compounds: trans-[Re2(H3N+–CH2–СO–NH–CH(COO)–CH2–CH(CH3)–CH3)2Cl42СН3CN]Cl2 (І) and trans-[Re2(H3N+–CH2–СO–NH–CH(COO)–CH2–C6H5)2Cl42СН3CN]Cl2 (ІІ). Analysis of the electronic absorption spectra of the solutions of the synthesized substances showed the presence of a doublet pattern (12500 and 16129 сm–1), which corresponds to the *-electronic transition of the Re–Re quadruple bond characteristic of solutions of trans-tetrachlorodi--carboxylates of dirhenium(III). The appearance of oscillations at 1485 сm–1 for І and 1486 сm–1 for ІІ, characteristic of the s(CO) of the coordinated carboxylate group, was detected on the IR spectra, which indicates the bridging coordination of this group to the Re26+ binuclear fragment. The protonation of the amino group is indicated by the appearance of a wide band of valence oscillations (NH3+) in the range of 3400–3350 сm–1 and deformation oscillations (NH3+) at 1557 сm–1 and 1614 сm–1 for I and II, respectively. The stability of the obtained complex compounds in aqueous solutions was determined. It was shown that the hydrolysis of the synthesized substances takes place in 4–5 days, which is accompanied by a decrease in the pH of the reaction solution.

Abstract:
(1) The consistency and magnitude of the prevalence of Clinical High-Risk for Psychosis (CHR-P) individuals are undetermined, limiting efficient detection of cases. We aimed to evaluate the prevalence of CHR-P individuals systematically assessed in the general population or clinical samples. (2) PRISMA/MOOSE-compliant (PROSPERO: CRD42020168672) meta-analysis of multiple databases until 21/01/21: a random-effects model meta-analysis, heterogeneity analysis, publication bias and quality assessment, sensitivity analysis—according to the gold-standard CHR-P and pre-screening instruments—leave-one-study-out analyses, and meta-regressions were conducted. (3) 35 studies were included, with 37,135 individuals tested and 1554 CHR-P individuals identified (median age = 19.3 years, Interquartile range (IQR) = 15.8–22.1; 52.2% females, IQR = 38.7–64.4). In the general population (k = 13, n = 26,835 individuals evaluated), the prevalence of the CHR-P state was 1.7% (95% Confidence Interval (CI) = 1.0–2.9%). In clinical samples (k = 22, n = 10,300 individuals evaluated), the prevalence of the CHR-P state was 19.2% (95% CI = 12.9–27.7%). Using a pre-screening instrument was associated with false negatives (5.6%, 95% CI = 2.2–13.3%) and a lower CHR-P prevalence (11.5%, 95% CI = 6.2–20.5%) compared to using CHR-P instruments only (28.5%, 95% CI = 23.0–34.7%, p = 0.003). (4) The prevalence of the CHR-P state is low in the general population and ten times higher in clinical samples. The prevalence of CHR-P may increase with a higher proportion of females in the general population and with a younger population in clinical samples. The CHR-P state may be unrecognized in routine clinical practice. These findings can refine detection and preventive strategies.

Abstract Chronic myeloid leukemia (CML) patients with imatinib failure are usually treated with second generation tyrosine kinase inhibitors (TKI). In case of dasatinib or nilotinib intolerance or resistance patients with HLA matched donor are submitted to allogeneic hematopoietic stem cell transplantation (HSCT) or switch treatment to a different TKI. Aims to evaluate complete hematological response (CHR), cytogenetic and molecular responses, overall, progression free and event free survival (OS, FPS and EFS) in CML patients in third line therapy that previously used 2 TKI. BCR-ABL mutations was also evaluated in this population Patients and methods between July 2008 and August 2012 26 CML patients were evaluated: 10 patients (pts) treated with dasatinib (38,5%) and 16 with nilotinib (61,5%) in third line therapy. OS, PFS and EFS were calculated with Kaplan-Meier method and Log-Rank was used for comparison, from the date of the third TKI start. The events for PFS were progression to accelerated phase (AP) or blast crisis (BC) and death. For EFS, were considered loss of CHR, loss of complete cytogenetic response (CCR), TKI discontinuation for failure or intolerance, progression or death. Patients were censored in HSCT date or TKI discontinuation for cytogenetic and molecular responses evaluation, but not for OS. Mutational analysis was done with Sanger sequencing. Results 13 male (50%) and 13 female patients with median age 54 years (22-75) and median follow-up of 32 months were analyzed (1-59). Previous response to imatinib: only one patient has achieved CCR. Status before third TKI start: 19 (73%) less than partial cytogenetic response (PCyR); 2 (7,7%) PCyR; 4 (15,5%) CCR;and 1 (3,8%) clonal evolution. 8/18 (42%) patients presented the following mutations: F317L (2), E255V (1), Y253H (1), M351T (1), M244V (1) e F359V (2). The median time of the third TKI treatment was 171 days (15-1140). Responses to third line TKI: CHR 17/19 (89%) in CP; 3/3 AF; 0/4 BC; CCR: 3/19 in CP (2 pts at 3 months and one in the 12º month); 1/3 AP; 0/4 BC; MMR: 4/19 in CP; 1/3 AP; 0/4 in BC. Nine lost CHR (45%) (8 in CP and one in AP). At 3 months, 1/21 pts (4.5%) achieved MMR; 2/13 (15%) at 6 months, while at 12 months 1/11 (9%) had MMR. At 3 months 11/21 had a BCR-ABL/ABL(IS) >10% and at 6 months 10/13 >1%. Three patients progressed to advanced phases (11,5%) ; 6 (23%) died (4 due to LMC, 1 acute myocardial infarction and 1 from graft vs host disease after HSCT; one lost follow-up. Third line treatment was discontinued in 12 (46%) pts: 6 for resistance, 2 death; 2 intolerance and resistance; 1 intolerance and one loss of follow-up. OS, PFS and EFS were 69%, 68% e 30% respectively(figure 1). OS was inferior in AP and BC in comparison with CP (0% vs 50% vs 90% - p< 0,0001)(figure 2). In summary responses may be obtained with dasatinib or nilotinib after 2 TKI failure, but they are not durable in advanced phases. Besides the results, that is an alternative for disease stabilization until the identification of a suitable donor or for patients with no performance status for HSCT. Abstract:
Abstract Prostate cancer (PCa) is one of the most common cancers among men, leading to the second cause of death for men with cancers in the US. Aneuploidy, featured by imbalanced chromosome numbers, is a hallmark of cancer. Epidemiology studies on primary PCa cohorts of Physicians' Health Study and Health Professionals Follow-up Study (PHS and HPFS) have shown that high levels of whole-genome aneuploidy correlate with lethal progression in PCa. However, the detail mechanisms of how aneuploidy drives aggressiveness of PCa are still unclear. Here, we used the case of chromosome 8q (chr 8q, the long arm of chr 8) gain to study aneuploidy-associated prostatic malignancies. Chr 8q gains are the most frequently gain events that occur in around 23% of PCa cases. By using the PHS and HPFS cohorts, we modeled the increased expression of each gene located on chr 8q, for predicting the risks for lethal progression, and obtained the odds ratio (OR) for each. Then, we ranked the ORs for lethal progression and identified several important genes highly associated with lethality when overexpressed. Among them, the cohesin subunit gene, RAD21, is one of the top associators. Increased RAD21 mRNA level, per se, is highly correlated with lethality in all PCa cases, and the lethality is synergistically aggravated in the cases with both increased RAD21 expression and chr 8q gains. indicating that RAD21 cooperates with other chr 8q genes to drive cancer progression and chr 8q gains. To determine how RAD21 overexpression promotes PCa, we studied the effect of overexpression of RAD21 in early prostatic oncogenic events. We utilized the isogenic mouse prostate organoid models carrying an inducible the fusion-oncogene, TMPRSS2-ERG (T-ERG), which 50% of PCa cases harbor. We found that induction of T-ERG leads to a strong oncogenic replication stress at an early stage. Such stress results in an increase in apoptosis and growth impairment in these primary organoids. Overexpression of RAD21, mimicking the chr 8q gain situation, mitigates such replication stress and rescues the growth defect caused by T-ERG. These data suggest the role of increased RAD21 in promoting oncogenic growth of PCa cells by reducing oncogenic toxicity at an early stage. In addition, we showed that such role of RAD21 overexpression sustains in the more advanced cancerous organoid (ERG positive and PTEN loss) and promotes the growth of the cancer organoids. Consistently, increased RAD21 expression correlates with increase proliferative markers in human prostate cancer cases. In conclusion, we identified that overexpression of multiple chr 8q genes are correlated with lethal progression in primary PCa, and RAD21 is one of such genes. Increased RAD21 plays a fundamental role in reducing toxic DNA damage caused by prostatic oncogenesis. Abstract The populations of Artemia (or brine shrimp) from the Americas exhibit a wide variation in the amount of interphase heterochromatin. There is interest in understanding how this variation affects different parameters, from the cellular to the organismal levels. This should help to clarify the ability of this organism to tolerate brine habitats regularly subject to strong abiotic changes. In this study, we assessed the amount of interphase heterochromatin per nucleus based on chromocenter number (N-CHR) and relative area of chromocenter (R-CHR) in two species of Artemia, A. franciscana (Kellog, 1906) (n=9 populations) and A. persimilis (Piccinelli and Prosdocimi, 1968) (n=3 populations), to investigate the effect on nuclear size (S-NUC). The relationship of the R-CHR parameter with the ionic composition (IC) of brine habitats was also analysed. Our results indicate a significant variation in the amount of heterochromatin both within and between species (ANOVA, p<0.001). The heterochromatin varied from 0.81 ± 1.17 to 12.58 ± 3.78 and from 0.19 ± 0.34% to 11.78 ± 3.71% across all populations, for N-CHR and R-CHR parameters, respectively. N-CHR showed less variation than R-CHR (variation index 15.5-fold vs. 62-fold). At least five populations showed a significant association (p<0.05) between R-CHR and S-NUC, either with negative (four populations, r= from -0.643 to -0.443), or positive (one population, r= 0.367) values.Within each species, there were no significant associations between both parameters (p>0.05). The R-CHR and IC parameters were associated significantly for the magnesium ion (r= 0.496, p<0.05) and also for the chloride, sodium and calcium ions (r = from -0.705 to -0.478, p<0.05). At species level, a significant association between both parameters was also found in A. franciscana populations, for the sulphate and calcium ions, in contrast to A. persimilis. These findings suggest that the amount of interphase heterochromatin modifies the nuclear size in Artemia. Our data also indicate that change in the amount of interphase heterochromatin is in line with the ionic composition of brines. This would be a species-specific phenomenon, whose occurrence may be involved in the ability of this organism to survive in these environments.

In these modern times, the world is shifting towards automation whether it is automobile industry or some other. In this paper we propose automation for car wiper control. Apart from some high-end luxury vehicles all other uses manual schemes for wiper control during rainfall, which can be deadly for drivers and passengers as well as it requires proper attention to increase or decrease the wiping frequency according to rain intensity. This increases the risk of accidents; therefore, automatic rain sensing wiper system becomes a more appealing feature. Although such systems are available in market, but they are not popular due to their high cost and other limitations. The objective of this paper is to provide a cost-effective system for automatic rain sensing wiper control. This system employs a rain sensing module which senses the rain and sends an analog signal to Arduino nano which analyzes the signal and energizes the servomotor which performs the wiping operation. By implementing this automation in automobiles, we can increase the driver and passengers’ safety by a great margin.

Aim: Real-world treatment data for psoriatic arthritis are limited. We evaluated switch rates, adherence, and costs for patients initiating apremilast versus tumor necrosis factor inhibitor (TNFi) and interleukin inhibitor (ILi) among biologic-naive psoriatic arthritis patients. Materials & methods: This retrospective analysis used IBM MarketScan claims data to assess treatment switches, adherence and costs. Results: Twelve-month switch rates were significantly lower for apremilast versus TNFi (15.5% vs 26.6%; p < 0.0001) and similar to ILi (15.5% vs 14.0%; p = 0.71). Apremilast initiators had lower total costs versus TNFi and ILi (US$39,854 vs US$57,243 and US$65,687; p < 0.05) and adherence was slightly lower versus TNFi and higher versus ILi. Conclusion: Biologic-naive apremilast initiators had lower switch rates versus TNFi initiators and lower total costs versus TNFi or ILi initiators.

Abstract: Road side emergencies such as tyre punch, is a problem commonly observed in cars. Conventional car jacks uses mechanical advantage to allow a human to lift a vehicle by manual force. This paper is a modification of the current toggle jack by incorporating a servomotor whose motion as well as torque is transmitted via gear trains to the power screw, in order to make load lifting easier for emergency use with the aid of a 12V car battery as power source. Gear ratio is used to increase the lifting power. The significance and purpose of this work is to modify the existing car jack in order to make the operation easier, safer and more reliable in order to save individual internal energy and reduce health risks especially back ache problems associated with doing work in a bent or squatting position for a long period of time. Fabrication work has been done using milling, drilling, grinding, and welding machines. The developed car jack is tested on 1400kg Toyota Camry car. Loads were increased at intervals of 50 Kg using bags of cement and it was observed that, deformation of threads began at loads beyond 1550 Kg. this implies that, the electric powered jack mechanism is capable of liftingvehicles with maximum load of 1550 Kg.

El uso de combustibles renovables provenientes de fuentes distintas a las fósiles toma importancia como alternativa a fuentes energéticas, en este caso el biogás. Sin embargo, motores exclusivos de uso de biogás tienen un alto costo económico, y poca disponibilidad comercial para bajas potencias, lo que hace difícil el aprovechamiento de dicho combustible sin la presencia del combustible diésel. Por lo que en este trabajo un motor de encendido por compresión se modifica a modo encendido provocado, sin cambiar la relación de compresión con el fin de lograr eficiencias efectivas altas. En este trabajo se analiza el comportamiento mecánico y ambiental de un motor estacionario diésel de 1550 cm3, con una potencia nominal de 20 kW@3000 rpm, con una relación de compresión de 15.5:1, adaptado a encendido provocado. Para el desarrollo de las pruebas planteadas en este trabajo se tiene las siguientes condiciones, el motor gira a 1800 rpm a 4 kW que corresponde al 57% de la carga del motor, a una presión atmosférica de 0.852 bar que corresponde a la ciudad de Medellín, trabajando con mezclas de metano (CH4) y dióxido de carbono (CO2) con el objetivo de simular la composición de un biogás común (60% CH4 – 40% CO2), y de biogás enriquecido (70% CH4 – 30% CO2 y 80% CH4 – 20% CO2), donde cada una de las mezclas fueron enriquecidas con adiciones de hidrógeno en porcentajes de 0%, 5%, 10% y 15% en volumen, la adición de hidrógeno presenta ventajas en la combustión ya que extiende los límites de inflamabilidad del biogás, además contribuye a la disminución de gases de efecto invernadero como el CO2, y de contaminantes como CO e hidrocarburos sin quemar, sin embargo la adición de hidrógeno aumenta las emisiones de NOx. Se encontró que la adición de 15% de hidrógeno mejora significativamente las propiedades de combustión cuando se hace uso de una mezcla combustible pobre, equivalente a un dosado relativo de 0.822, para el caso del biogás puro, mientras que para las mezclas de biogás con hidrógeno no se encontraron mejoras significativas en el comportamiento mecánico y ambiental del motor.

Today, video is a common medium for sharing information. Navigating the internet to download a certain form of video, it takes a long time, a lot of bandwidth, and a lot of disk space. Since sending video over the internet is too costly, therefore video summarization has become a critical technology. Monitoring vehicles of people from a security and traffic perspective is a major issue. This monitoring depends on the identification of the license plate of vehicles. The proposed system includes training and testing stages. Training stage comprises: video preprocessing, Viola-Jones training, and Support Vector Machine (SVM) optimization. Testing stage contains: test video preprocessing, car plate (detection, cropping, resizing, and grouping detecting test car plate, feature extraction using HOG feature. The total time of local recorded videos is (19.5 minutes), (15.5 minutes) for training, and (4 minutes) for testing. This means, (79.5%) for training and (20.5%) for testing. The proposed video summarization has got maximum accuracy of (86%) by using Viola-Jones and SVM by reducing the number of original video frames from (7077) frames to (1200) frames. The accuracy of the Viola-Jones object detection process for training 700 images is (97%). The accuracy of the SVM classifier is (99.6%).

Highlights Fish hatchery RAS thickened wastewater was successfully treated by anaerobic digestion. Traditional complete mix digester (CSTR) configuration was compared to the UAFF, an upflow anaerobic floating filter. UAFF start-up resulted faster and more productive than in conventional reactors (188 compared to 100 NmL CH4/g VS). At steady conditions, yields of 190.5 and 180.9 NmL CH4/g VS were recorded, for UAFF and CSTR respectively. Abstract. Thickened wastewater from a fish hatchery recirculating water system (RAS) was successfully treated by anaerobic digestion, showing a high methane potential in brackish and marine conditions. To optimize the process and establish operational criteria for a full-scale transfer, pilot-scale tests were carried out in a BioReactor Simulator System (BRS, Bioprocesscontrol, Sweden). The BRS consists of 6 reactors with a volume of 1.8 L each: 3 reactors operated in conventional configuration (CSTR, continuous stirred tank reactor), while the other 3 were modified with an up-flow fixed bed layout, with floating plastic filling elements (UAFF, upflow anaerobic floating filter). The objective of the present paper was the comparison of the performance of the UAFF and the CSTR anaerobic systems both in the initial colonization batch process, and in the following continuous process. The batch test was performed in 72 days by treating aquaculture sludge inoculated with brackish digestate, for the CSTRs, and with the biofilm of the plastic media for the UAFFs. The following continuous loading phase was characterized by start-up and steady steps. The tests were performed in mesophilic conditions. The start-up in the UAFF reactors resulted significantly faster and more productive than in conventional reactors, with specific yields reaching 187.9 NmL CH4/g VS compared to 100.0 NmL CH4/g VS. During the following steady phase, with the retention time of 20 days, the specific yields increased in all digesters but the differences between the 2 types of reactors were not significant, even if favorable for the UAFF (190.5 and 180.9 NmL CH4/g VS, respectively for UAFF and CSTR). Air Conditioner is part in a car to achieve safety and security in driving. The purpose of this study was to determine the current of the air conditioner electrical system, identify the electrical circuit and analyze the electrical circuit of the air conditioner system. This research used experimental method. The air conditioner system simulator show current and voltage generated at blower speed 1: current 15.1 A, produces a temperature of 26 ℃, blower speed 2: current 15.2 A, produces a temperature of 25 ℃, blower speed 3: current 15.5 A, yields a temperature of 24 ℃. Whereas in the car the current and voltage generated by blower speed 1: current 17.4 A, produces a temperature of 21 ℃, blower speed 2: current 17.7 A, produces a temperature of 20 ℃, blower speed 3: current 17.9 A, produces a temperature of 19 ℃. So, the higher the current has an effect on the resulting performance of the air conditioner system. And the lower the current generated, the blower, magnetic clutch and extra fan performance is not optimal when the AC system is running. Sistem Air Conditioner merupakan bagian dari mobil untuk mencapai keyamanan dan keamanan dalam berkendara. Tujuan dari penelitian ini adalah untuk mengetahui arus sistem kelistrikan air conditioner, mengidentifikasi rangkaian kelistrikan dan menganalisis rangkaian kelistrikan sistem air conditioner. Metode penelitian yang digunakan dalam penelitian ini adalah metode eksperimental. Hasil pengujian arus dan tegangan yang dihasilkan pada simulator blower speed 1: arus 15,1 A, menghasilkan suhu 26℃, blower speed 2: arus 15,2 A, menghasilkan suhu 25℃, blower speed 3: arus 15,5 A, menghasilkan suhu 24℃. Sedangan pada mobil arus dan tegangan yang dihasilkan blower speed 1: arus 17,4 A, menghasilkan suhu 21℃, blower speed 2: arus 17,7 A, menghasilkan suhu 20℃, blower speed 3: arus 17,9 A, menghasilkan suhu 19℃. Jadi, semakin tinggi arus berpengaruh pada kinerja yang dihasilkan sistem air conditioner. Dan semakin rendah arus yang dihasilkan membuat kinerja blower, magnetic clutch dan extra fan menjadi tidak maksimal pada saat sistem AC bekerja.

The heterogeneity in traffic flow composition increases the complexity of road traffic noise analysis for mid-sized in India. This study aims to determine a passenger car noise equivalent (PCNE) with respect to the average traffic stream speed that represents the number of a particular vehicle category with reference to an identified vehicle based on their noise emission characteristics. In the present study, vehicles are classified as bus, truck, light commercial vehicles (minibus, minitruck), three-wheelers (vikram-rickshaw), two-wheelers (bike/scooter), car, e-rickshaw and auto-rickshaw, and tractor-trailer. Car is taken as a reference vehicle for estimation of PCNE in our study due to its high percentage in traffic stream. Data has been collected on both bituminous and concrete pavement in Kanpur city, India, to analyze the differential effect of pavement on the noise level. As per this study, tractors-trailers, trucks, three-wheelers, and buses had a higher PCNE value, while two-wheelers and cars had almost similar PCNE value. A comparative analysis of PCNE value at concrete pavement is also conducted by considering car running on the bituminous pavement as reference vehicle. The study suggests to employ PCNE value in traffic noise analysis as it converts the divergent traffic volume in terms of the car.

Background: Although patients with MPN may achieve complete hematological remission (CHR) with cytoreductive therapy (CRT), lifelong treatment represents a major burden for these chronically ill patients. IFN has emerged as a major MPN therapy thought to positively alter the natural history of MPN and appears to be the only drug that can provide long-term CHR after discontinuation in some patients (Hasselbalch HC et al., Seminars in Immunopathology, 2019). In this study, we aimed to identify clinical and molecular factors associated with long-term CHR after IFN treatment discontinuation and to compare clinical outcome of patients who discontinued therapy, to patients who continued IFN treatment despite achieving a CHR. Methods: All MPN patients followed between January 2000 and May 2020 in our institution who received at least 3 months of IFN were included. Logistic regression analysis was performed to identify variables associated with long-term CHR after discontinuation. Cumulative incidence of relapse was calculated from time of IFN discontinuation, considering death without relapse as a competitive event. Overall survival (OS) and thrombotic/hemorrhagic events free survival (EFS) curves were estimated using the Kaplan-Meier method and compared by cause-specific hazard Cox models. Results: 381 patients treated with IFN were included in the study. Median age at MPN diagnosis was 44 years [interquartile range: 33-54]. 171 had polycythemia vera (PV), 169 essential thrombocythemia (ET) and 34 primary myelofibrosis (PMF). JAK2V617F was the most frequent driver mutation (78.8% of patients), while CALR and MPL were mutated in 15.5%, and 2.9% of patients respectively. Median driver variant allele frequency (VAF) at time of IFN therapy initiation was 34% [12; 51]. Reasons to start IFN included age younger than 50 years in 44.9%, intolerance/resistance to previous therapies in 17.8% or pregnancy in 1.8% patients. CHR was achieved with IFN in 77.2% of patients. After a median follow-up of 72.4 months [28.4; 119.7] from IFN initiation, 131 patients were still on IFN treatment, while 250 patients had discontinued therapy. No significant difference was observed between continued and discontinued IFN patients in terms of MPN subtype, initial clinical, biological or molecular characteristics. Reasons for discontinuation were toxicity in 128 (50.4%), prolonged hematological CHR in 76 (29.9%), failure in 16 (6.3%) and other in 30 (11.8%) patients. At time of IFN discontinuation, 170 (66.9%) patients were in CHR and the median driver mutation VAF was 12% [3; 35]. Of note, IFN was re-introduced in 61 patients who lost CHR with a second CHR rate of 83.6%, arguing for the absence of development of IFN resistance in post-discontinuation relapses. In multivariate logistic regression analysis, stopping for prolonged CHR (OR 3.52, 95%CI [1.18; 10.56], p=0.024) was associated with a higher CHR without CRT rate, while VAF ≥ 10% at time of IFN discontinuation (OR 0.26, 95%CI [0.1; 0.65], p=0.004) was associated with lower long-term CHR without CRT. Accordingly, VAF ≥ 10% at time of IFN discontinuation (HR 3.06 [1.59; 5.90], p=0.001) was independently associated with a higher cumulative incidence of relapse after IFN discontinuation (Figure A). Driver VAF at IFN discontinuation time was available for 117 patients. 53 patients (45.3%) had a driver VAF &lt; 10% at IFN discontinuation. Finally, we compared discontinued IFN patients who stopped IFN for CHR (n=76) to patients who obtained CHR and are still on IFN therapy (n=116). Hematological transformation could not be evaluated due to very low incidence of events: 2 patients from each group progressed to myelofibrosis while leukemic progression was only observed in 1 patient. Importantly, OS (HR 0.23, 95%CI [0.5; 1.14], p=0.07) and EFS (HR 0.53, 95%CI [0.19; 1.45], p=0.217) were not significantly different between patients who discontinued and those who continued IFN. Conclusion: Overall, our study demonstrates that IFN discontinuation represents a safe strategy in MPN patients who achieved CHR, and particularly in patients with a driver VAF lower than 10% at time of discontinuation. Importantly, relapsed patients did not develop IFN resistance. Our data contributes to set the frame for future clinical trials evaluating the possibility of IFN treatment holidays in good responding MPN patients. Variasi pada penelitian ini adalah campuran kotoran sapi dengan penambahan gliserol mentah sebanyak 0, 4, 8, dan 12%. Semua variasi dengan volume total 350 mL. Pengukuran volume biogas dilakukan setiap hari. Pengukuran konsentrasi CH4 dan CO2 diukur menggunakan alat Geotech Biogas 5000 analyzer. Parameter yang diukur adalah COD, BOD, TS dan VS dan untuk pH diukur di awal serta di akhir proses. Suhu lingkungan diukur setiap hari dengan interval waktu selama 30 menit menggunakan alat Weather Station model PCE-FWS 20. Hasil penelitian menunjukkan lama waktu yang dibutuhkan untuk memproduksi biogas adalah 14 hari. Volume biogas tertinggi adalah 836 mL, pada variasi penambahan gliserol mentah 12%. Gliserol mentah yang ditambahkan memiliki COD 475,2 mg/L, BOD 133,22 mg/L, TS 20% dan VS 14,8%. Konsentrasi CH4 tertinggi juga didapatkan dari variasi penambahan gliserol mentah 12% yaitu 44,1%. Sedangkan volume biogas terendah adalah 292 mL pada digester tanpa penambahan gliserol mentah. Konsentrasi CH4 terendah didapatkan pada variasi penambahan gliserol mentah 8% yaitu 15,5%. Identifikasi bakteri yang berperan dalam proses produksi biogas berdasarkan uji biokimia dengan Bergey’s manual adalah genus Bacillus. Bakteri ini berperan dalam proses pendegradasi bahan organik yang ada di dalam digester. ABSTRACTBiogas is a gas produced by anaerobic activity in decomposing organic matter with the main content of methane (CH4) and carbon dioxide (CO2). This study aims to analyze the effect of crude glycerol with a mixture of cow dung in producing biogas. The digester used in this research was a laboratory-scale batch system. The variation in this study was a mixture of cow dung with the addition of 0, 4, 8, and 12% crude glycerol. All variations with a total volume of 350 mL. The measurement of the biogas volume was carried out every day. Measurements of CH4 and CO2 concentrations were measured using a Geotech Biogas 5000 analyzer. The COD, BOD, TS, and VS, and pH parameters were measured at the beginning and the end of the process. With parameters COD 475.2 mg/L, BOD 133.22 mg/L, TS 20%, and VS 14.8%. The ambient temperature was measured every 30 minutes using a PCE-FWS 20 Weather Station model. The results showed that the length of time needed to produce biogas was 14 days. The highest biogas volume was 836 mL, when 12% cured glycerol was added. The highest CH4 concentration of 44,1% was also obtained from variations in the addition of 12% crude glycerol. The lowest biogas volume of 292 mL was obtained from the absence of crude glycerol in the reactor. The lowest CH4 concentration was found in the variation of the addition of 8% crude glycerol, namely 15.5%. Identification of bacteria that play a role in the biogas production process based on biochemical tests using Bergey's manual is the Bacillus genus. These bacteria contribute in these bacteria contribute in organic matter degradation inside the digester.

Abstract Background: We present the development of NECTIN4-CAR T cells targeting bladder cancer and evaluate the ability of thiazolidinedione, a class of anti-diabetic drugs, to specifically upregulate NECTIN4 expression and expand the therapeutic window of NECTIN4-CAR T cells. In addition, we assess efficacy of NECTIN4-CAR T cell therapy against a model of bladder cancer with acquired resistance to enfortumab vedotin (EV). Methods: NECTIN4-targeting chimeric antigen receptor (CAR) constructs were designed and cloned into a second-generation EQ-28ζ CAR backbone construct. Human primary T cells were transduced with lentiviral NECTIN4-CAR constructs and co-cultured with a panel of bladder cancer cell lines with a range of NECTIN4 expression and against NECTIN4 knockout cells to assess specificity. Tumor cell count over time was monitored in vitro using an IncuCyte S3. Bladder cancer cell lines (RT112, UMUC-1, HT1197) were treated with PPARγ agonists (rosiglitazone) and antagonists (T0070907), and total and surface NECTIN4 protein levels were assessed. Anti-tumor effects of the NECTIN4-CAR T were evaluated in vitro and in vivo, either with or without rosiglitazone pre-treatment. Finally, RT112 bladder cancer cells were exposed to “cycles” of escalating doses of EV to generate EV-resistant cell line models, and anti-tumor efficacy against EV-resistant lines was evaluated. Results: NECTIN4-targeting CAR T cells displayed potent and NECTIN4-specific cytotoxic activity against bladder cancer cell lines. The efficacy of NECTIN4-CAR T cells was strongly correlated with NECTIN4 expression. Treatment with PPARγ agonists and antagonists was shown to upregulate and downregulate NECTIN4 expression, respectively. Treatment with the PPARγ agonist rosiglitazone induced NECTIN4 expression in a PPARγ-dependent and dose responsive manner. NECTIN4 expression was increased in bladder cancer xenografts established in immunodeficient NSG mice that were treated with systemic rosiglitazone. Priming with rosiglitazone enhanced sensitivity to NECTIN4-CAR T cells in vitro, and the combination of rosiglitazone pre-treatment and NECTIN4-CAR T cell therapy showed enhanced tumor control in bladder cancer xenograft models. We generated RT112 cells that exhibited resistance to EV in vitro and in vivo, which retained NECTIN4 expression. Interestingly, EV-resistant RT112 cells remained sensitive to NECTIN4-CAR T cell therapy both in vitro and in vivo. Conclusions: We engineered a NECTIN4-targeting CAR T cell therapy that is highly antigen-specific and demonstrates antitumor activity against multiple bladder cancer models, including EV resistant cells. Modulating the PPARγ pathway increases NECTIN4 expression, which we leveraged by repurposing an FDA-approved anti-diabetes drug rosiglitazone to increase targeting and anti-tumor efficacy of NECTIN4-CAR T cells. These preclinical results lay the groundwork for CAR T therapy development in bladder cancer and suggest rational drug combinations that can expand the therapeutic window of NECTIN4-targeting therapies. Abstract From August 1973 to December 2003, 1651 patients (median age: 28yr., range: 1–74 yr.; M: 982, F: 669) received stem cell transplants for haematological malignancies (Ac. Leuk: 1246, Chr. Leuk: 182, other: 223). Donor was allogeneic (n=1088), autologous (n=537) or syngeneic (n=26) and conditioning was with (n=1243) or without TBI (n=408). Primary disease was classed as high risk in 714 and low risk in 937 patients. Dose of TBI was ≤9.5 Gy (n=342), 10.5 Gy (n=613) or ≥ 10.5 Gy (n=282) delivered in single fraction (n=1015) or multiple fractions (n=222). Donor was sibling (n=900), matched unrelated (n=135) or mismatched family member (n=53). Cranial top-up radiation was used in 357 patients. Source of stem cell was marrow (n=1312), PBSC (n=320) or both (n=19). GVH prophylaxis was CyA alone (n=527), CyA with other agent (n=516) or other measures (n=45). Of all the patients 1515 received single transplant but 136 received more than one transplant. Data was analysed as of 01/07/2006. Of all the patients 897 (54.3%) survived for at least 1 yr. post transplant and the median follow-up in this group was 27yr. Second cancers developed in 50 cases with the incidence of 2.6% at 5yr (95% CI: 1.3–3.9), 7.5% at 10 yr (95% CI: 4.9–10.1) and 11.5% at 15yr (95%CI: 7.6–15.4). Site of second malignancy was brain (n=10), breast (n=6), cervix (n=3), GIT (n=2), lung (n=1), skin (n=9), sarcoma (n=3), thyroid (n=1), oral cavity (n=4), MDS (n=6), CML (n=1), ovary (n=1) and non EBV related lymphoma (n=3). In univariate analysis 10 yr. probability of developing SMN was significantly higher with chronic GVHD (5% vs. 10%, p=0.008, high risk disease (5% vs. 11%, p=0.0007), cranial RT (4% vs. 18%, p=0.015) and ALL as primary diagnosis (4% vs. 10%, p=0.06). In multi-variate analysis advanced stage disease (RR: 2.4, 95% CI: 1.3–4.2, p=0.004), Chr. GVHD (RR: 2.5, 95% CI: 1.4–4.4, p=0.003) and cranial RT (RR: 2.3, 95% CI: 1.2–4.4, p=0.01) were independently associated with increased risk of SMN. There was strong interdependence between cranial RT and ALL as primary diagnosis. It was noted that the multi-variate risk factors were different for site specific SMNs. For brain tumours cranial RT (RR: 12.8, 95% CI:2–75.5, p=0.006) and age <16 yr at the time of transplant (RR:15.7, 95% CI:1.9–130.6, p=0.011) were associated with increased risk. For epithelial malignancies Chr. GVHD (RR: 5.8, 95% CI: 2.2–15.5, p=0.0005), female sex (RR: 3.0, 95% CI:1.1–8.3, p=0.033) and advanced stage disease (RR: 3.6, 95% CI: 1.4–9.4, p=0.01) were associated with increased risk. No independent risk factors could be identified for development of skin or haematological malignancies. 25 patients have died due to SMN (49%) at a median of 31 months. Survival was significantly better in skin SMNs (85%) as compared to haematological SMNs (20%), brain tumours (30%) and epithelial SMNs (56%)(p=0.02). This single centre analysis shows that the risk of developing SMN does not relate to the type of conditioning and increases with longer follow-up. The risk factors vary for the different sites of SMNs. These patients need long term cancer screening.
