Abstract:
Lipid nanoparticles (LNP) are the most potent carriers for the delivery of nucleic acid-based therapeutics. The first FDA approved a short interfering RNA (siRNA) drug that uses a cationic LNP system for the delivery of siRNA against human transthyretin (hTTR). However, preparation of such LNP involves tedious multi-step synthesis with relatively low yields. In the present study, we synthesized cationic peptidomimetic functionalized cholesterol (denote Chorn) in straightforward chemical approaches with high yield. When formulated with helper lipids, Chorn LNPs complexed with siRNA to form nanoparticles with an average diameter of 150 nm to 200 nm. Chorn LNP mediated transfection of a green fluorescence protein (GFP) expressing plasmid resulted in 60% GFP positive cells. Moreover, Chorn LNP delivered siRNA against polo-like kinase 1 (Plk1), a disease related gene in cancer cells and efficiently suppressed the expression of the gene, resulting in significant morphological changes in the cell nuclei. Our data suggested that cholesterol based cationic LNP, prepared through a robust chemical strategy, may provide a promising siRNA delivery system.

The Lucky Dragon is a vitamin, Chinese medicine and nutraceutical company located in Causeway Bay in Hong Kong. Although the company was ran for 50 years by the Ng family, after the patriarch Mr. Ng Shu Chorn passed away, his children decided to sell it, as none of the three children were interested in managing the store or sourcing products and selling them to other shops. Increasingly, competition from mainland China had been eroding profit margins of similar businesses in Hong Kong, and each of the siblings had respectable jobs, so cashing in was the best course of action. Anderson Products LLC, a health and wellness company headquartered in Provo, Utah, had hoped to expand to the seemingly lucrative Chinese market, but they were inexperienced with international business and afraid of mainland China regulations. They saw the advertisement for the sale of The Lucky Dragon as a great opportunity to get their feet in the door, under the more auspicious regulations that Hong Kong could offer.

Experiments were carried out in an 82.6-mm-dia column with a perforated distributor plate. Conductivity probes on the axis of the column were used to measure local bubble properties in the developing flow region for superficial air velocities from 0.0018 to 6.8 m/s and superficial water velocities from 0 to 0.4 m/s, corresponding to the discrete bubble, dispersed bubble, coalesced bubble, slug, churn, bridging, and annular flow regimes. Bubble frequency increased linearly with gas velocity in the discrete and dispersed bubble regimes. Bubble frequency also increased with gas velocity in the slug flow regime, but decreased in the churn and bridging regimes. Bubble chord length and its distribution were smaller and narrower in the dispersed than in the discrete bubble regime. Both the average and standard deviation of the bubble chord length increased with gas velocity in the discrete, dispersed, and churn flow regimes. However, the average bubble chord length did not change significantly in the slug flow regime due to the high population of small bubbles in the liquid plugs separating Taylor bubbles. The bubble travel length, defined as the product of local gas holdup and local bubble velocity divided by local bubble/void frequency, is used to correlate bubble characteristics and to characterize the flow regimes. Abstract:
Overlay networks are not a new field or area of study. This domain of computing will someday drive P2P systems in various application areas such as block-chain, energy trading, video multicasting, and distributed file storage. This study highlights the two widely known methods of routing information employed in one of such overlay networks called chord. In this study, simulations of both routing modes (iterative and recursive) and their variations under no-churn (leaving and joining of nodes) and churn conditions was carried out. The routing parameter (successor list size) was varied for each of the routing techniques in a simulation study. The results obtained show that semi recursive routing gives a better routing performance under churn scenarios.

Abstract:
Local government is one of three tiers of government that operate in Australia. This research investigates how marketing can be applied to local government in a holistic manner. To achieve this goal, theory needed to be reviewed and developed with one particular outcome being a model outlining the components of the marketing environments that need understanding prior to the application of marketing practice. Local government was selected as the context of this study as much is written about the marketing of not-for-profit, social and public sector organisations but the literature on the integration of these practices in a holistic approach for local government is very limited. Further local government has a major economic impact on the viability and longevity of many rural and semi-rural areas of Australia. Equally in the metropolitan areas, local government accounts for a considerable proportion of the employment and impact on growth and development of these regions. Traditional marketing theory has been found wanting in the local government area, as traditional marketing practices is being applied in a piecemeal approach. This practice has caused local governments to have conflicting messages and product offerings to the community. However the review of marketing derivatives theory shows that no one derivative addressed the scope of products and activities managed by local government. Reviewing these derivatives show that marketing core concepts are relevant to local government, but the complexity lies in the application of marketing where the components of added complexity were derived from the organisational focus, structure and target market definitions. The theoretical process to develop this understanding of local government marketing and to develop a proposed model for the application of local government marketing was derived from the literature relating to the core traditional marketing concepts and the marketing derivatives of public, not-for-profit and social marketing. This review of the literature aided in defining the complexities of local government marketing and helped found the preliminary local government marketing model. With the use of case analysis three local government cases were explored. The first case, a metropolitan local government, used convergent interviewing to determine the factors relevant to the development of the holistic local government marketing model. Confirmatory case analysis was used for the remaining two cases, one semirural and one rural, to refine the developing model. Ultimately, the model was confirmed in principle but minor changes were required to make the model robust across the three cases. From a theoretical perspective the research identified that the marketing derivatives used in the development of the local government marketing model were relevant and further sets local government apart from but integrated with the three derivatives studied. From a practical perspective the development of the local government marketing model goes some way to developing practices that are coordinated and integrated across the local government organisation, thus providing local government with the advantages of having an integrated local government marketing approach.

Abstract:
Churn analysis is an important tool for companies as it can reduce the costs that are related to customer churn. Churn prediction is the process of identifying users before they churn, this is done by implementing methods on collected data in order to find patterns that can be helpful when predicting new churners in the future.The objective of this report is to identify churners with the use of surveys collected from different golfclubs, their members and guests. This was accomplished by testing several different supervised machine learning algorithms in order to find the different classes and to see which supervised algorithms are most suitable for this kind of data.The margin of success was to have a greater accuracy than the percentage of major class in the datasetThe data was processed using label encoding, ONE-hot encoding and principal component analysis and was split into 10 folds, 9 training folds and 1 testing fold ensuring cross validation when iterated 10 times rearranging the test and training folds. Each algorithm processed the training data to create a classifier which was tested on the test data.The classifiers used for the project was K nearest neighbours, Support vector machine, multi-layer perceptron, decision trees and random forest.The different classifiers generally had an accuracy of around 72% and the best classifier which was random forest had an accuracy of 75%. All the classifiers had an accuracy above the margin of success.K-folding, confusion-matrices, classification report and other internal crossvalidation techniques were performed on the the data to ensure the quality of the classifier.The project was a success although there is a strong belief that the bottleneck for the project was the quality of the data in terms of new legislation when collecting and storing data that results in redundant and faulty data.
Churn analys är ett viktigt verktyg för företag då det kan reducera kostnaderna som är relaterade till kund churn. Churn prognoser är processen av att identifiera användare innan de churnas, detta är gjort med implementering av metoder på samlad data för att hitta mönster som är hjälpsamma när framtida användare ska prognoseras. Objektivet med denna rapport är att identifiera churnare med användning av enkäter samlade från golfklubbar och deras kunder och gäster. Det är uppnå att igenom att testa flera olika kontrollerade maskinlärnings algoritmer för att jämföra vilken algoritm som passar bäst. Felmarginalen uppgick till att ha en större träffsäkerhet än procenthalten av den dominanta klassen i datasetet. Datan behandlades med label encoding, ONE-hot encoding och principial komponent analys och delades upp i 10 delar, 9 träning och 1 test del för att säkerställa korsvalidering. Varje algoritm behandlade träningsdatan för att skapa att klassifierare som sedan testades på test datan. Klassifierarna som användes för projekted innefattar K nearest neighbours, Support vector machine, multi-layer perceptron, decision trees och random forest. De olika klassifierarna hade en generell träffssäkerhet omkring 72%, där den bästa var random forest med en träffssäkerhet på 75%. Alla klassifierare hade en träffsäkerhet än den felmarginal som st¨alldes. K-folding, confusion matrices, classification report och andra interna korsvaliderings tekniker användes för att säkerställa kvaliteten på klassifieraren. Projektet var lyckat, men det finns misstanke om att flaskhalsen för projektet låg inom kvaliteten på datan med hänsyn på villkor för ny lagstiftning vid insamling och lagring av data som leder till överflödiga och felaktiga uppgifter.

Abstract:
Population churn--the population turnover experienced by a community--can have differential effects on a community. Mass society theory suggests that because the churn rate experienced by communities can contribute to their uprooting, fragmentation, and isolation, churn is a potent threat to the stability of our modern day communities. Social capital theory, to the contrary, suggests otherwise. Social capital theory suggests that churn can have positive effects on communities by bringing new migrants with valuable human capital skills and experiences to communities. These migrants bring to their new communities the potential for creating new jobs, spurring economic development, and for initiating housing starts that expand housing options for the poor and minorities. In so doing, they help create and sustain vibrant, growing modern day communities. Yet in spite of the significant role churn may play in determining the health and viability of modern day communities, it has been overlooked in the migration literature, which is mostly dominated by individual-level research on the causes and effects of migration, particularly the pecuniary benefits to movers. Using county-level data and multivariate analyses, this research seeks to fill this gap in the literature by examining the relationship between the community and churn, from the perspectives provided by social capital and mass society theories.

Abstract:
This dissertation is concerned with various mathematical aspects of Topological Quantum Field Theories (TQFTs) known as Chern-Simons theories. Although this subject has its origins in theoretical physics, the treatment here is in terms of the axiomatic approach due to Segal and Atiyah. A key feature of the thesis is the notion of a 3-tier (axiomatic) TQFT. This involves assigning a category to a closed I-manifold and a functor to a 2-manifold with boundary which is viewed as a cobordism between I-manifolds. To a closed 2-manifold Σ the theory assigns a vector space HΣ , and to a 3-manifold M the theory assigns a numerical invariant (if M is closed), a vector in HδM (if M has closed boundary δM ) or a natural transformation of functors (if the boundary δM of M has a 1-dimensional boundary). After a brief introduction, we introduce in chapter 1 the definition of a TQFT and that of a 3-tier TQFT. We then describe the geometrical set-up for Chern-Simons Theory for a Lie group G and focus on the particular case of G = SU(2). Finally we describe quite concisely how it might fit into a 3-tier TQFT structure. Roughly the next half of the thesis treats the specific case of Chern-Simons theory for the circle group T. In chapter 2 we describe a number of interesting topological aspects of the theory. In chapter 3 we go on to show how the theory fits into 3-tier TQFT framework. In the next two chapters we begin to deal with Chern-Simons theories for G a non-compact group. In chapters 6 and 7 we deal with a rather more algebraic theory which is the abelian version of a theory which is meant to compute the Casson invariant for oriented homology 3-spheres. For this reason we call it the abelian Casson-type theory. From the physics viewpoint, it coincides with the Chern-Simons theory where G is a supergroup. This is rather difficult to motivate mathematically, so we adopt an algebraic-topological definition of the theory and show it satisfies the TQFT axioms. We then go on to show how it fits into the 3-tier TQFT structure. The novelty here is that the category assigned to a 1-manifold is not semisimple.

A presente pesquisa analisou diferentes gravações da peça Nosso Choro de Garoto, com o objetivo de levantar aspectos interpretativos semelhantes entre os intérpretes. A metodologia empregada contempla a escolha das gravações, dos tópicos de análise e as ferramentas para realizar a mesma. As gravações utilizadas foram realizadas por violonistas da tradição do choro, que na maioria dos casos conheceram e conviveram com o autor da peça. Os tópicos analisados nas gravações são: localização e utilização do rubato, articulação e acentuação da figura rítmica semicolcheia – colcheia – semicolcheia, articulação dos baixos, e a utilização de arpejos. Foi realizada uma análise formal da peça, com o intuito de organizar melhor a coleta de dados. A análise de ditas gravações foi feita por meio de softwares especializados para tais fins. As análises revelam aspectos interpretativos semelhantes entre os intérpretes, os quais consideramos como pertencentes ao estilo interpretativo de um gênero musical.
The present research analyzed different recordings of Nosso Choro by Garoto, with the aim of bringing up common interpretive aspects between performers. The methodology includes the choice of the recordings, of the topics of analysis and of the tools to accomplish the same. The recordings were performed by guitarists who belong to the tradition of Choro, who in most cases met the composer. The topics analyzed in the recordings are: location and use of rubato, articulation and accentuation of the rhythmic sixteenth note – eight note – sixteenth note, articulation of the bass line, and the use of arpeggios. A formal analyses of the piece was made with the aim of better organizing the data collected. The analysis of this recording was made through use of specialized software for such purposes. The analyzes reveal similar interpretive aspects between performers, which we consider as belonging to an interpretative style of a musical genre.

Abstract:
The psychological research on music and emotion supports how the two concepts have always been intertwined. The most common line of research in this field is related to the commonly experienced phenomenon that major chords sound happy while minor chords sound sad. This proposed study takes a specific extended major chord (the Vadd11 chord) and tests whether the V chord with a color note, an added 11, has a significantly different emotional impact on listeners as compared to a V chord without an 11. The chord will be played within a I-V-I progression so that the Vadd11 chord is given musical context. Participants are asked to rate the how emotional the progression is and to indicate their preference between the two chords. Both piano and strings timbres will be tested. The chord progression change is hypothesized to cause a significantly increased emotional impact. Multiple confounding variables will be examined within the study (specifically cultural musical background, or what culture’s music participants listened to in their upbringing, and musical sophistication, or how musically adept participants are). Neither of these variables nor the timbre are anticipated to affect the changes in emotional reaction.

The way humans listen to music and perceive its structure isautomatic. In an attempt by Friberg et al. (2011) to model thishuman perception mechanism, a set of nine perceptual features wasselected to describe the overall properties of music. By letting atest group rate the perceptual features in a data set of musicalpieces, they discovered that the factor with most importance fordescribing the emotions happy and sad was the perceptual featuremodality. Modality in music denotes whether the key of a musicalpiece is in major or minor.This thesis aims to predict the modality in a continuous scale (0-10) from chord analysis with multiple linear regression and a NeuralNetwork (NN) in a computational model using a custom set offeatures. The model was able to predict the modality with anexplained variability of 64 % using a NN. The results clearlyindicated that the approach of using chords as features to predictmodality, is appropriate for music data sets that consisted of tonalmusic.
