Abstract:
(Title: Representation of Hero’s Journey on Main Character Chihiro in Hayao Miyazaki’s Spirited Away Anime) This paper is the result of a qualitative descriptive type of literature study. The purpose of this study was to reveal the heroic journey process of main character of Spirited Away anime named Chihiro, based on the Hero’s Journey theory proposed by Vogler. Data about Chihiro's heroic journey are obtained from anime with note taking techniques. The method used in the analysis is the method of content analysis based on the theory of Hero's Journey. From the results of the analysis, it can be seen that the main character Chihiro has undergone a total of the twelve stages of the hero’s journey. Chihiro's adventure has succeeded in changing her personality which was initially timid, spoiled, and whiny to be brave, independent, calm and confident, and full of compassion. Chihiro's heroic journey proves the true definition of a hero according to Vogler, while also proving that to win a fight does not always use physical strength. There are times when thinking intelligence and emotional intelligence are much more needed. In addition, this anime also features a lot of kamisama which was once believed by the Japanese. Miyazaki might want to remind again about the existence of Japanese gods by displaying them in this anime, either those actually found in Japanese mythology or Miyazaki's own creations. Thus, the story of Spirited Away which has a mythical structure as stated by Vogler becomes much more alive and interesting.

Abstract:
This study intends to examine the stages of the hero’s journey based on the mythic structure of Campbell’s monomyth and use the monomyth to analyze how a ten-year-old girl, Chihiro, embarks on a journey to the supernatural world in Spirited Away. The study uses content analysis, a descriptive qualitative method, to analyze the monomyth: a departure–initiation–return journey covering seventeen stages. The monomyth involves metamorphoses and challenges that the heroine Chihiro encounters and explores her evolution during her transition to full autonomy in order to reach transformation and self-individuation. Chihiro, in order to rescue her parents, is destined to take the journey: to sink into darkness, the unconscious, and encounter her natural and primitive essence. By fighting against conflict or opposition, Chihiro, as a mythical heroine, completes her monomyth cycle and attains transformation and regeneration.

INTRODUÇÃO: A adolescência assume diferentes concepções a depender da época vivida, sendo importante refletirmos sobre essas diversas leituras. Entretanto, o foco do presente trabalho está voltado para a abordagem da psicanálise que propõe ler esse momento como uma travessia, o momento de transição para a adultez, marcada pelo surgimento de caracteres sexuais secundários e psíquicos que repercutem no encontro com o desconhecido. O filme A viagem de Chihiro conta a história de Chihiro Ogino, uma menina de dez anos que está mudando com os pais para uma nova cidade. Tomando o filme como uma metáfora do adolescer na contemporaneidade, o presente trabalho busca apreender os impasses da adolescência através da trajetória vivida pela personagem Chihiro. Para o desenvolvimento do estudo, a metodologia escolhida foi a pesquisa documental. METODOLOGIA: Para a articulação entre o filme e o tema da pesquisa foi realizada a construção de três eixos de leitura: os lutos inerentes ao processo de adolescência; a adolescência e o tempo lógico de Lacan e os impactos do discurso capitalista na adolescência contemporânea. DISCUSSÃO E RESULTADOS: A viagem percorrida por Chihiro representa o seu adolescer, o qual é marcado por diferentes lutos. Aos poucos, Chihiro vivencia a perda da posição infantil, também, pelas diversas novas responsabilidades que surgem na sua vida, tais como a necessidade de salvar os seus pais, aprender o funcionamento da casa de banho e lidar com as diferentes criaturas nesse novo dia a dia. Os pais da infância, que eram sinônimo de proteção e segurança, gradativamente e dolorosamente se tornam frágeis, levando Chihiro a elaborar uma outra posição subjetiva, tornando-se adulta. CONSIDERAÇÕES FINAIS: A leitura psicanalítica possibilitou um olhar para a singularidade de tal vivência, mostrando os diversos aspectos em jogo que não podem ser limitados aos parâmetros biológicos.

Las películas de Hayao Miyazaki han recibido reconocimiento mundial, entre otras cosas, por la manera en la que observa los afectos y conductas humanas para después retratarlos en la pantalla. En esta ocasión me centraré únicamente en uno de los personajes más conocidos del cineasta: Kaonashi o Sin-Cara de la película El viaje de Chihiro. Analizaré este personaje pues, aún sin tener los elementos esenciales para representar afecto (rostro y palabras), logra expresar de manera sencilla e inteligible emociones complicadas como el miedo, la ira o la soledad. Además, el éxito en la difusión de este personaje me indica una afinidad afectiva con los espectadores, donde la falta de respuestas dentro de la narración los lleva a buscar interpretaciones dentro de su propia experiencia emocional. El presente ensayo busca establecer que el público de El viaje de Chihiro ve reflejado su lado humano más íntimo y complicado en Kaonashi.

Abstract:
Moral is what guides human to act humanly. That is why moral education should be taught as early as possible in order to make a society that upholds moral values. This paper focused on moral values of the Japanese that can be seen in Miyazaki’s anime entitled Spirited Away. The moral values discussed in this paper are based on Japanese moral education for elementary school level, which divided into four groups of desirable habits. They are matters belonging to oneself, relationship of self to other persons, matters related chiefly to nature and sublime things, and matters concerning the group and society. The determination of this level is due to the behavior of the main character named Chihiro who was an elementary school student, which is used as a guide to discussing moral values consisted in the anime. The conclusion is that the Japanese government has succeeded in giving moral education in elementary school, at least in this anime. Chihiro's actions are proof that she acts and reacts as what she taught in Japanese elementary school. She did exactly the same with recommended traits in the four groups of desirable habits, although she sometimes broke the rules which was natural for human being, especially for kids. Therefore, we can say that well-structured Japanese moral education can be used in much wider place; not only in Japan but also in other countries.

Le voyage de Chihiro est un film d’animation japonais du réalisateur Hayao Miyazaki, sorti en 2001. La jeune héroïne Chihiro a pénétré dans un monde alternatif, et elle a rencontré Haku, Sans-Visage, ainsi que toute une galerie d’autres personnages. La présentation de ces personnages se constitue autour des métamorphoses fantastiques et des représentations des mythes, des légendes, et des folklores, qui témoignent d’une diversité culturelle. Suite à la métamorphose des parents de l’héroïne en porcidés ainsi que d’autres personnages en formes variées, les questions autour de l’identité, de la mémoire, ainsi que la singularité ou la banalité du sujet en tant qu’être humain se posent. Quand la mémoire du passé s’avère le principe primordial de survie – principe qui formule d’une certaine manière l’identité du personnage - quel mystère renferme-t-il donc le souvenir de soi à travers celui de l’autre ? Ces interrogations sur le nom et la métamorphose problématisent le maintien du nom “véritable”, voire l’identité des personnages. Elles suscitent de nombreuses réflexions sur les influences qui s’exercent en dehors de leurs origines locales. De telles influences ne correspondent pas toujours à un voyage s’effectuant de l’Occident vers l’Orient. Au lieu d’insister sur les catégories opposées entre les deux, cet article explore le rapport entre la mémoire du passé, le nom et l’identité qui font tous référence à une diversité de sources transculturelles et contribuent à un échange transculturel dans cette œuvre, tout en entamant la réflexion sur l’interprétation allégorique de l’œuvre à partir des mondes des différents contextes culturels et sociaux.

Abstract:
This study aims to describe the equivalence of translating Japanese cultural terms into Indonesian in the anime Sen to Chihiro no Kamikakushi and the translation procedures applied in translating SL (source language) into TL (target language). This research is a qualitative descriptive study using data cards. The theory used is the category of cultural terms by Newmark (1988), the theory of equivalence by Nida and Taber (1974), and the theory of translation procedures by Newmark (1988). There are 38 data which are Japanese cultural terms and its translations in Indonesian. The results showed that 30 data reached equivalence and 8 data did not reach equivalence. Then, there are 7 translation procedures applied, including cultural equivalent, synonym, neutralization, additional explanation, modulation, transposition, and reduction and expansion. Cultural equivalence and synonyms are the most frequently applied procedures.

Argumento que la película japonesa El viaje de Chihiro del director Hayao Miyazaki es una alegoría nacional en términos de Jameson. Esto es así porque, en primer lugar, el destino individual de su protagonista representa una aporía entre nacionalismo y globalización; en segundo lugar, la alegoría es consciente y voluntaria; en tercer lugar, la forma de representación constituye una alteridad cultural frente a las formas occidentales; en cuarto lugar, porque la película es síntoma de una oposición entre cultura y Naturaleza. Realizo esta lectura inscribiéndome en el campo de los estudios culturales. Además, el modelo cuaternario de explicación alegórica al que suscribe Jameson me permite explicar qué sentido tiene que una película pensada para niños/as pueda tener estas características.

Com o intuito de depreender o estilo autoral do japonês Hayao Miyazaki, são analisados, neste trabalho, seus seguintes filmes: Porco Rosso (1992), A Viagem de Chihiro (2001) e Vidas ao Vento (2013). Tais obras são examinadas à luz do conceito de estilo, conforme propõe Discini (2014), articulando-se tal concepção à teoria do autor de Stam (2003), além de debater os alicerces em torno da caracterização de personagens centrais de acordo com Brait (2005) e Brecht (2002). Por meio das análises realizadas, pôde-se concluir que o estilo autoral de Miyazaki se constrói a partir de recorrências observadas tanto no plano do conteúdo, quanto no plano da expressão.

The research method in this research was qualitative and descriptive analytical method. The results of this research will provide the new insights into the animation appreciation and the understanding of the implicit context in the story. People can understand the politic, economic, culture of Japanese after watching the work of Studio Ghibli. The author hopes that the results of this article will produce useful conclusions and will be used for the next study.

Abstract:
The object of the study is the work of the Japanese animator, director Hayao Miyazaki. The subject of the research is the relationship between parents and children in Japanese society on the material of the animated fantasy film, written and directed by Hayao Miyazaki. The empirical material of the article is not only the film itself, but also interviews of the director, participants of the film creation process, journalists, viewers, and posts in social networks. The research focuses on the images of the parents of the main character Chihiro and her parents from the point of view of Japanese viewers. The goals of Miyazaki in creating the image of «inadequate parents» are defined and analyzed. Particular attention is paid to the description of the beliefs associated with spiriting away (hidden by kami) in Japan. The authors describe cases of missing children in medieval Japan, including such phenomenon as 間引き mabiki – «getting rid of children». The basis of the research methodology is an interdisciplinary socio-cultural approach, methods of cultural and phonological interpretation and linguocultural commentary. The relevance of the research topic is determined by the expanding space and forms of intercultural communication, the popularity of Hayao Miyazaki's works all over the world, the humanistic nature of his works, the increased interest of researchers in the subculture of anime, which transfers cultural values and behavior models and serves as a tool of youth socialization. Scientific novelty of the study is determined by the fact that for the first time the film «Spirited Away» is analysed in terms of the relationship between parents and children, taking into account the broad context of national and cultural attitudes. Original materials in Japanese and their interpretation are introduced into the scientific turnover. In the course of the research, the authors conclude that the main task that the director was to help unleash the inner life force of the main character Chihiro, and for this purpose he sent the girl on a journey full of trials.

O presente artigo é referente a uma prática extensionista desenvolvida junto aos familiares de crianças com altas habilidades/superdotação participantes do Projeto de Extensão “Enriquecimento da Aprendizagem para o Desenvolvimento de Habilidades”, da Pontifícia Universidade Católica de Minas Gerais, em Belo Horizonte. Esse estudo tem como foco principal contribuir para reflexões a respeito da utilização e vantagens de grupos focais em moldes virtuais, como forma adaptativa ao contexto de pandemia. A intervenção aqui explicitada foi realizada no Grupo de Pais do projeto supracitado e refere-se a um texto dissertativo construído a partir da análise do filme “A Viagem de Chihiro”, lançado pelo estúdio Ghibli, em 2003 no Brasil. O trabalho realizado resultou em uma produção textual repleta de analogias entre as cenas do filme e as expectativas, anseios e medos paternos referentes ao comportamento e habilidades de seus filhos.

Curated by socio-cultural anthropologist Fuyubi Nakamura, the exhibition entitled A Future for Memory: Art and Life after the Great Japan Earthquake at the Museum of Anthropology at UBC in British Columbia addresses the sociocultural role of art produced in situ in the aftermath of the triple disaster which occurred in the Tōhoku region of northeast Japan in 2011. The exhibition’s curatorial project was born in the affected regions through anthropological research, and the selections of works brought to British Columbia are by The center for remembering 3.11; Lost & Found Project; Lost Homes Scale Model Restoration Project; Chihiro Minato; Atsunobu Katagiri; Masao Okabe; Rias Ark Museum of Art; Tsunami Ladies film project team. This article engages with the conversations that the curator, artists, and collaborators wove through the exhibition. The construction of social memory building on the experiences of a drastically changing environment is its main theme.

Introducción. Hayao Miyazaki y Satoshi Kon son dos de los directores mejor valorados por la crítica y el público en el género anime contemporáneo. A pesar de tener estilos diferentes, coinciden en que ambos sus protagonistas suelen ser mujeres en la mayor parte de su filmografía. Objetivo. Este estudio analiza cuantitativamente la popularidad de estos personajes femeninos en las búsquedas de Google desde 2004 hasta 2021. Metodología. A partir de estos datos, realizaremos un análisis cualitativo sobre la evolución narrativa y el tratamiento estético de cada uno de estos personajes. Resultados y conclusiones. Concluimos que los personajes que experimentan un paso de la inocencia a una primera madurez fueron los más populares entre los aficionados a estas películas: El viaje de Chihiro (29,95) y Mi vecino Totoro (25,01). Asimismo, la apariencia infantil de las heroínas anime fue más popular que las de aspecto adulto.

This collection of readings, based upon recent research into various German, Japanese, Italian, Chinese, French, British, and American documents that were still classified during the 1960s, presents the basis for Finney's observation that the origins of World War II were very much more complex than the traditional Eurocentric or nationalistic scholarship written largely by the victors. The readings come from a selection of works by such historians as David Dilks, Sidney Aster, Anthony Adamthwaite, Tim Mason, and R.J. Overy in the section entitled "Interpretations and Debates." Hayao Miyazaki&rsquo;s (born on January 5, 1941 in Tokyo), is a Japanese animator, animation producer, cartoonist, animation director, and animation screenwriter. His movies have lasting impact. Based on the conceptual metaphor theory of cognitive linguistics, this study analyzes the multi-modal metaphors in his animated movies to explore his cognitive needs behind his creation and find out how these metaphors are generated. This paper uses the method of qualitative analysis and comprehensively analyzes the images, aiming to answer two questions: 1) What are the multi-modal metaphors in his animated movies? 2) What cognitive needs and creative motivations do these multi-modal metaphors reflect in his movies? In this paper, five of Miyazaki&rsquo;s animated movies are employed to analyze the images in it. The secondary data was collected from the website of movies. The results of the study showed that there is a large number of non-textual metaphors in his movies, such as: Pig, Totoro, Oil house, Chihiro, Animals, Fire, Bridge, Tain, and so on. These multimodal metaphors reflect the cognitive needs and psychological motivations behind his creations. Through the analysis of Hayao Miyazaki himself, basic color, emotions, space, daily habits, national culture and other metaphorical cognition in his movies, we have a full understanding of metaphorical cognition, language and culture, national psychology and thinking habits in the process of his creation. The findings are of great importance to the cognitive study of metaphors in the field of animated movies.

Abstract Background and Purpose: Gastric adenocarcinoma and proximal polyposis (GAPPS) is an autosomal dominant genetic disorder that causes gastric carcinoma from fundus gland polyposis. Although germline mutation of APC promoter 1B was reported as a pathogenic variant in 2016, there are still many unknowns about the carcinogenesis. In this study, we performed a comprehensive Next generation sequencing (NGS) analysis of GAPPS clinical specimens to investigate the molecular profile characteristic of carcinogenesis. Patients and Methods: Seven cases from three GAPPS families experienced in our department were included in this study. In each case, tissues were collected from normal mucosa, polyps, and carcinoma, and FFPE and fresh-frozen samples were prepared. After pathological diagnosis was performed on each sample, RNA and DNA were extracted and analyzed based on data obtained by next generation sequence (NGS). Results: In the RNA sequence, gene expression analysis was performed on 45 pathologically diagnosed samples, which were classified into four clusters by K-means clustering. Gene Set Enrichment Analysis (GSEA) of the four clusters showed that cancer-related genes such as p53 and KRAS were significantly enriched in the cluster of cancer, and genes related to immune response were significantly enriched in the cluster of cancer compared to the and clusters of polyp and normal. Gene mutation analysis using whole-exome sequence (read depth/coverage&gt;20) confirmed mutation of APC promoter 1B in all samples. On the other hand, in tumor samples (N=10), the percentage of mutation of genes previously suggested to be associated with GAPPS (TP53, FBXW7, GNAS, and KRAS) were 50%, 0%, 30%, and 60%, respectively. Conclusion: Comprehensive gene expression analysis by RNA sequence and whole-exome sequence was performed using clinical samples of GAPPS. "Successful academic writing from sources requires a broad range of competencies. When writing from sources, students are expected to mine source texts for relevant ideas, present these ideas with precision and in necessary depth, have efficient paraphrasing skills and the knowledge of proper source attribution. In order to assess the combination of these skills in writing and to provide diagnostic feedback to the learners, there is a need to design a rating scale where the required skills are operationalized in separate criteria (Knoch 2011). However, this endeavour may be challenging due to the complex nature of the academic integrated writing construct. This article describes the process of analytic rating scale development in the context of German higher education (HE). We address the issues of construct complexity and the operationalization of the construct elements in rating scale criteria by a combination of theory-based, descriptor-based, empirical, and intuitive approaches to scale development (e.g., Chan, Chihiro and Taylor 2015; Kuiken and Vedder 2021), with a particular focus on the usability of relevant scales from the CEFR Companion Volume (CEFR/CV; Council of Europe 2020). Besides the CEFR scales, we also explore the usability of existing scales for integrated writing and relevant taxonomies (e.g., Keck 2006; Shi 2004). Finally, we present qualitative insights of intuitive expert judgement from a workshop with four content experts who trialled and refined the first draft of the rating scale. The ensuing validation of the rating scale is, however, beyond the scope of this paper and the mixed-methods validation study will be reported elsewhere. The rating scale development reported here was part of the DFG-funded research project Modelling of academic integrated linguistic competencies, conducted at the University of Bremen and the Leibniz Institute for Research and Information in Education in Frankfurt. The project aim was to evaluate the academic-linguistic preparedness of students taking up English-medium studies in Germany by employing authentic integrated writing tasks and valid assessment procedures. Abstract Objective. The presence of intestinal metaplasia (IM) is a risk factor for gastric cancer. However, it is still controversial whether IM itself is a precancerous or paracancerous lesion. Here, we aimed to explore the precancerous nature of IM in terms of epigenetic alterations. Design. Genome-wide DNA methylation analysis was conducted by EPIC BeadArray using IM and non-IM crypts isolated by Alcian blue staining. Organoid establishment, ChIP-seq for H3K27ac, and single-cell ATAC-seq were conducted using IM mucosa. NOS2 was induced using a Tet-on gene expression system in normal cells. Results. IM crypts had a methylation profile unique from that of non-IM crypts, showing extensive DNA hypermethylation in promoter regions with CpG islands, including those of tumor suppressor genes. Also, the IM-specific methylation profile was carried into a fraction of gastric cancers (GC), and the frequency of the IM-type GCs was higher than the fraction of IM cells in the adjacent mucosa, indicating that IM cells have a higher chance of developing into cancer cells than non-IM cells. Moreover, the patients with IM-type GCs showed better overall survival than those without, and IM-type GCs showed good response to chemotherapy. As for the mechanism of accelerated methylation induction, IM organoids had remarkably high endogenous NOS2 expression, and NOS2 induction in normal cells was shown to accelerate aberrant DNA methylation by increasing DNMT activity. IM mucosa underwent dynamic enhancer reprogramming, including the regions involved in abnormal NOS2 expression. Single-cell ATAC-seq also showed NOS2 open chromatin in IM cells but not in gastric cells, and frequent closed chromatin of tumor-suppressor genes in IM cells. NOS2 expression in IM-derived organoids was up-regulated by IL-17A, a cytokine secreted by extracellular bacterial infection. Conclusions. IM cells were considered to have a strong precancerous nature with an increased chance of converting into cancer cells in terms of the specific methylation profile and the accelerated DNA methylation induction due to abnormal NOS2 expression. Abstract Various heterogeneities in response of metastatic tumors to systemic anti-cancer therapy are major source of therapeutic resistance. Cancer cells from bulk tumors and cancer cell lines are not clonal. A highly heterogeneous tumor microenvironment is known to develop by subclones. Furthermore, metastasis can evolve during the disease progression as well as in response to systemic therapies. These spatial and temporal variations of heterogeneity in tumors can affect blood perfusion and vascular permeability which can determine the level of mass/anti-cancer drugs transport to tumors (we refer to as “transport phenotype”). Insufficient drug transport to tumor in vivo can generate transport-based therapeutic resistance even for agents with proven efficacy against the target cancer cell lines in vitro. Our goal is to understand spatial and temporal heterogeneity in transport of mass/anti-cancer drugs to metastatic tumors to overcome the therapeutic resistance. The study of bulk tumors will provide a mixture of different functions from heterogeneous sets of various clones. We sought to establish single cell-derived clones from a 4T1 parental murine breast cancer cell line in order to dissect and simplify the heterogeneity in polyclonal cell populations in tumors. First, we found heterogeneous accumulation of systemically injected PEGylated liposomal doxorubicin (PLD) among multiple, experimental liver metastases of 4T1 parental (polyclonal) breast cancer in mouse. Intravital Microscopy imaging of liver metastases through a window chamber over time revealed different transport phenotypes, in terms of circulation and diffusion of systemically injected fluorescent tracers around/inside parental (polyclonal) tumors. Furthermore, transport phenotype can be modulated from one to another after PLD therapy, creating possible mechanisms of transport based on drug resistance. When single cell clone-derived metastases were established, a clone dependent difference in transport phenotype was found. Interestingly transport phenotype even in clonal tumors was temporally evolved and increased its heterogeneity. These data suggest that transport phenotype cannot be determined solely by genotype of tumor and can be evolved and more heterogeneous as a function of tumor growth and systemic chemotherapy. Clinical implication of our findings is that transport of therapeutics to tumor and its efficacy may not be predicted by DNA-based assay for clinical decision-making and transport-based therapeutics resistance will eventually occur in every tumor types. Abstract [Objective] Uterine leiomyosarcoma (LMS) has a poor prognosis, and no effective treatment other than surgery has been established. Kinesin family member4A (KIF4A) is a member of the kinesin4 subfamily and plays an important role in cell division. KIF4A has been revealed to play significant roles in some cancers with tumor proliferation. The aim of this study is to clarify the relationship between KIF4A and tumor growth of LMS. [Methods] One normal human uterine smooth muscle cell line and three LMS cell lines(SK-UT1, SK-LMS and SKN) were used to identify tumor-specific elevated proteins with iTRAQ(isobaric tags for relative and absolute quantitation). SiRNA and ShRNA were used to downregulate KIF4A in vitro and in in vivo. Cell variability was evaluated by MST-8 assay. Cell cycle analysis by fluorescence-activated cell sorting (FACS) and western blotting (WB) were performed to analyze the signal pathway. Immunostaining of 26 clinical specimen from patients operated at our university hospital since 1999 to 2021. [Results] A total of 2084 proteins were identified by iTRAQ. KIF4A was identified as a protein with more than twice the expression level of normal smooth muscle cells. WB confirmed high expression of KIF4A in all three LMS cell lines. And in immunostaining of clinical specimens, KIF4A was positive in 20/26(76.9%) cases. A trend toward worse prognosis was suggested in the group with high KIF4A expression (no significant difference due to the small number of cases). Downregulation with SiRNA significantly suppressed the growth of LMS cell lines in vitro (-37.2±4.73% in SK-LMS, -87.7±4.02% in SKN and -28.1±3.00% in SK-UT1, p&lt;0.05). By FACS, G2/M phase cells was significantly increased in KIF4A suppressed cell lines and by WB, p-cdc2 was upregulated in KIF4A suppressed cells. In vivo, downregulation of KIF4A was found to significantly suppress tumor size in genograft mice. (1186±118mm³ vs 461±84mm³ in SK-LMS and 1704±441mm³ vs 514±230mm³ in SK-UT1, p&lt;0.05, f). Abstract The presence of plasticity in the epigenomes of young individuals is suggested by the aggressive phenotypes observed in cancers among adolescent and young adults (AYAs), including gastric cancers. To demonstrate the presence of this plasticity and identify the underlying epigenomic characteristics in gastric stem cells during youth, we examined organoids established from murine stomach tissue and conducted single-cell RNA-seq and ATAC-seq using murine antrum-pyloric tissues at ages of 3 weeks and 50 weeks, corresponding to human infancy and middle-age adult, respectively. Organoids established from infant mice showed higher dependence to niche factors, and stronger response to cytokine signals. Ligand-receptor analysis using single-cell RNA-seq data of tissues showed that infant epithelial stem cells and stromal cells had stronger interactions. Both supported the plastic nature of infant gastric epithelial cells. Single-cell ATAC-seq enabled us to identify epithelial stem cell population in infant and middle-aged mice, and 71 and 118 chromatin peaks were specific to infant and adult stem cells, respectively. Motif enrichment analysis showed an enrichment of binding sites for bZIP family transcription factors in infants, and an enrichment of binding sites of a chromatin remodeling factor and a family of transcription factors in adults. Additionally, comparison between differentiated epithelial cells, marked by the pit cell marker Muc5ac, and stem cells, marked by Lgr5, showed more pronounced differences: 46,772 peaks specific to pit cells and 39,076 peaks specific to stem cells. Interestingly, regions specific to pit cells were enriched in binding sites of the chromatin remodeling factor and the family of transcription factors, identical to the open regions specific to adult stem cells. This suggested that the chromatin accessibility profile of aged stem cells approaches to that of differentiated cells, leading to a loss of stemness. The single-cell RNA-seq data also revealed an increase in interactions between differentiated epithelial cells and fibroblasts in aged mice, as opposed to infant, suggesting the emergence of aberrant interactions in aged stomach tissue. Collectively, our data suggested that chromatin accessibility of gastric stem cells undergo an epigenomic shift during aging, approaching to the profiles of differentiated cells and losing their plasticity, and that a specific chromatin remodeling factor and a family of transcription factors could be involved in the process. Abstract Background: Antibody-drug conjugates (ADCs) are a new class of therapeutics that combine a monoclonal antibody with a cytotoxic agent (known as their payloads) via a synthetic linker. While ADCs are one of game changer and are the fastest growing classes with novel clinical benefits recently, interstitial pneumonia (ILD) is a clinical problem as a fatal adverse event. However, the mechanism of ADC-induced ILD is under investigation. Purpose: We clarify ADC-ILD mechanism by analyzing released payload distribution in the lung using a novel Trophoblast cell surface antigen 2 (TROP2)-targeting ADC. Material and Methods: TROP2-eribulin is a novel TROP2-targeting ADC composed of the TROP2 antibody and cytotoxic payload eribulin conjugated by a cleavable linker. Four human cancer cell lines (A549, HeLa, NCI-H2110 and SKOV3) and one human- leukemia monocytic cell line (THP-1) were used in this study. Cell surface TROP2 antigen expression was evaluated by flow cytometry. We investigated the concentration and distribution of the TROP2-eribulin and the payload released from the TROP2-eribulin in tumor and lung tissue, using TROP2 low and high expression human cancer xenograft model that were treated with various doses of TROP2-eribulin. The concentration of erbulin was analyzed by liquid chromatograph mass spectrometry, and tissue distribution of ADCs was assessed by immunofluorescence (IF) assay. Results: In vitro analysis showed that the concentration of intracellularly released eribulin was significantly higher in NCI-H2110 with high TROP2 expression compared to A549 with low TROP2 expression (P &lt; 0.05). In vivo analysis, the concentration of released eribulin in tumor tissue was approximately 10-fold higher in the NIC-H2110 xenograft model than the A549 xenograft model, while analysis of lung tissue showed that TROP2-eribulin was distributed in lung tissue in a dose-dependent manner of TROP2-eribulin regardless of TROP2 expression, with significantly more eribulin released in the high-dose group of TROP2-eribulin than in the other dose groups (P &lt; 0.05). IF assay showed that TROP2-eribuilin localized to alveolar macrophages. In the analysis using THP-1 cell, which mimic macrophage function, the concentration of eribulin released from TROP2-eribuilin was significantly reduced by the use of an Fc receptor inhibitor (P &lt; 0.05). Conclusions: Nonspecific uptake of ADC by alveolar macrophages via Fcγ receptors releases cytotoxic payload into lung tissue, regardless of tumor antigen expression levels. This understanding can help elucidate the pathogenesis of ADC-induced ILD. Abstract Background: Interstitial pneumonia (IP) is a risk factor for lung cancer (LC). LC, a significant contributor to mortality, shows improved prognosis with molecular targeted drugs. However, LC combined with IP (LC with IP) has a poor prognosis because of acute exacerbations. Therefore, developing effective drugs for LC with IP is necessary. Cancer-associated fibroblasts (CAFs), influencing cancer progression through interactions with the microenvironment are anticipated as a new therapeutic target. Therefore, we hypothesized that CAFs in LC with IP, having undergone repeated destruction and regeneration, exhibit distinct characteristics from those in LC without IP and exert varied influences on carcinogenesis and cancer progression. Methods: Six cases of lung adenocarcinoma, the predominant type of LC, were enrolled in this study. Spatial profiling was performed on FFPE specimens from three LC with IP and three LC without IP cases, using GeoMx whole-transcriptome atlas assay. Immunostaining for α-SMA, a major CAFs-marker, and pan-cytokeratin (CK), an epithelial marker, were employed to identify regions of interest (ROIs). In LC with IP, two ROIs were designated for cancerous and non-cancerous areas, respectively, whereas in LC without IP, two ROIs were selected for cancerous areas. Each ROI was segmented for epithelium and cancer (CK+/α-SMA -) and α-SMA-positive stroma (CK-/α-SMA +) areas of illumination. NGS data were analyzed using the edgeR to compare mRNA expressions in cancerous (LC-IP) and non-cancerous (NC) areas of LC with IP. Additionally, comparisons were made between LC-IP and cancerous areas of LC without IP (LC-nonIP) for mRNA expression. Results: Analysis of α-SMA-positive stroma revealed 86 differentially expressed genes (DEGs) in LC-IP vs. NC. Notably, COTL1 and NFKB2 increased, whereas TNC and MMP7 were decreased in LC-IP α-SMA-positive stroma. Pathway Enrichment analysis indicated activation of the response to cytokines. Between LC-IP and LC-nonIP, 151 DEGs were detected. CXCL8 and NFKB2 were elevated, whereas ACAN and ELN decreased in LC-IP α-SMA-positive stroma. KEGG pathway analysis demonstrated inactivated ECM-receptor interactions. Analysis of CK-positive regions showed 710 DEGs in LC-IP vs. NC, and KEGG pathway analysis indicated inactivated ECM-receptor interaction. In LC-IP vs. LC-nonIP, 321 DEGs were detected, with KEGG pathway analysis suggesting inactivation of ECM-receptor interaction and activation of a chemokine signaling pathway and cytokine-cytokine receptor interaction. Conclusion: These findings suggest that inflammatory factors may play a more critical role than extracellular matrix in the interaction between α-SMA-positive stroma and CK-positive cancer cells in LC with IP. Future plans include conducting in vitro experiments, as well as immunohistological studies. Abstract Background: CBA-1205 is a novel humanized anti-delta-like 1 homolog (DLK-1) antibody. It is a glycoengineered antibody by GlymaxX® technology to potentiate antibody-dependent cellular cytotoxicity activity. DLK-1 is a membrane protein with 6 tandem EGF-like motifs in extracellular region. Overexpression of DLK-1 has been reported in variety of cancer types including hepatocellular carcinoma (HCC), suggesting that DLK-1 could be an ideal target for cancer therapy. Methods: This is a first in human, multicenter, open-label Phase I study. The purpose of part 1 is to evaluate the safety and tolerability in patients with advanced solid tumors who had no standard therapy or were refractory or intolerant to standard therapies and to determine the starting dose for part 2 in HCC patients. The standard 3+3 dose escalation design was utilized in seven cohorts (0.1, 0.3, 1, 3, 10, 20, 30 mg/kg). CBA-1205 was administered at 2-week intervals in a 28-day cycle. If the patient had no Dose Limiting Toxicity (DLT) during 28 days after the first two doses, CBA-1205 administration will be continued for further cycles unless any of the criteria for discontinuation. Serum DLK-1 concentration before the start of CBA-1205 administration was analyzed using the Human Pref-1/DLK1/FA1 DuoSet ELISA (R&D Systems, Inc.). Immunohistochemical analysis of DLK-1 expression in the archival tumor specimen was determined in FFPE tissue sections using mouse anti-hDLK-1 antibody (clone DI-2-20, Chiome Bioscience). Results: 22 patients with advanced solid tumors were enrolled. CBA-1205 was well tolerated in these patients and no DLT was observed even at the highest dose tested. No ≥ Grade 3 treatment related adverse events was observed throughout the study periods. The serum concentration of CBA-1205 increased dose-proportionally. The exposure to CBA-1205 exceeded the effective concentrations observed in a mouse xenograft model. No serum anti-CBA-1205 antibody was detected. The disease control was seen in 8 patients (38.1%, all with SD). Especially, long SD was observed in 3 patients with invasive thymoma, malignant melanoma, and pancreatic carcinoma, where progression free survival (PFS) was 39, 72(ongoing, at data cutoff), and 41 weeks, respectively. The serum concentration of soluble DLK-1 was detected in 6 out of 22 patients. Patients with SD by the best overall response had a higher rate of detectable serum DLK-1 concentration and a higher mean concentration than patients with PD. No DLK-1 expression in tumor was detected by IHC method, in 14 patients who consented to use the archival tumor specimens. Conclusions: CBA-1205 was well tolerated without any severe toxicity in patients with advanced solid tumors.Part 2 of this Phase I study with two cohorts (20, 30 mg/kg) in patients with unresectable advanced/recurrent HCC refractory or intolerable to the standard therapy has been initiated. Um diese Lücke zu schliessen, setzen die Japaner verstärkt auf Roboter, welche Dank verbesserter Feinmotorik und emotionaler Intelligenz auch für anspruchsvollere Pflegeaufgaben voll einsatzfähig sein werden. Vor einem Jahr wurde in Japan die Roboterdame Aiko Chihira vorgestellt. Sie ist lebensgross und hat eine menschliche Anmutung und soll künftig als Kommunikationsroboter am Empfang oder im sozialen Bereich eingesetzt werden.
