Full textThe overall organization of the nuclear pore complexes (NPC) has been evolutionary conserved. Yet variations in organisms likely provide important clues to NPC functions. While most NPC proteins (nucleoporins or Nups) have been identified in S. Cerevisiae and vertebrates, few nucleoporins have been characterized in the yeast Schizosaccharomyces pombe. During my PhD, through sequence homology with S. Cerevisiae, I have identified and further functionally characterized 6 novel S. Pombe nucleoporins. Biochemical approaches have confirmed the organization of these Nups within conserved NPC subcomplexes. While examination of the S. Pombe deletion mutants revealed different viability phenotype as compared to their S. Cerevisiae orthologues, functional studies indicated that the involvement of this complex in NPC distribution and mRNA export has been partly conserved between these highly divergent yeasts. Unexpectedly microscopic analyses of the S. Pombe mutants showed additional cell division defects (spindles, chromosome segregation and septation) reminiscent to some GTPase Ran-cycle mutants. Consistently I further showed a specific functional link between the SpNup107 complex and the Ran pathway. During mitosis, the Human Nup107 complex colocalizes with kinetochores suggesting an as yet unidentified evolutionary conserved role of these Nups in mitosis, even though this feature does not seem to be conserved in S. Pombe. These differences between yeast S. Pombe and vertebrates could be associated to some specificity related to closed versus opened mitosis. My work has thus provided additional clues of the involvement of the Nup107 complex in cell division mechanisms

Full textHierarchical organization is the hallmark of many social vertebrates. Associations between social rank and health are reported within many species, including humans. Yet, the mechanisms underlying these associations are poorly understood. The stress response, specific to the social rank in many animals, is also involved in many psychiatric pathologies. This suggests a role of stress response in the process linking social status and behavior. My PhD work aims to better understand the associations that exist between social rank and individual behavior in tetrad housed mice. My work also seeks to clarify the direction of this association by observing the consequences of social positions on behavior and vulnerability to psychiatric illnesses, and reciprocally, by identifying early individual markers that can participate in shaping social fate. At the physiological level, I am focusing on the stress response and its consequences on the dopamine (DA) system, as potential mediator for such social-related phenotypes. To address these questions, I first assessed adult male wild-type mice for social rank with precedence, territorial and competition tests. I then tested animals of different social ranks for behaviors such as anxiety, sociability and cognitive skills either before or after hierarchy establishment. We recorded the electrophysiological activity of DA cells in the ventral tegmental area. I quantified the DA release throughout the mesocorticolimbic pathway and assessed mice for depressive-like behaviors and addiction. Finally, I addressed the role of the glucocorticoids by assessing mutated mice lacking glucocorticoids receptor in DAceptive cells for social dominance

Full textBased on an ethnography of the Kulung Rai religious life, a small Himalayan society, this work aims at understanding the reasons spirits are conceived and what does such representations involve. The analysis is based on the contexts where the referent "spirit" appears : myth, misfortune and ritual. We then show how spirits are organised in different categories corresponding to different spaces and implying different kinds of rituals, each of them with their own logic and effects. Finally, the work shows that relations with spirits reveal a system of interpretation and actions that structure and legitimise a way to interact in the world that articulate persons and groups, oneself and otherness, order and disorder, nature and society

Full textThe populations of a species with a wide range of distribution can be exposed to various environmental conditions exercising different natura! selection pressures. They can then display genetically based phenotypic particularities giving them an adaptation to the local constraints of their environment. Ln the scope of understanding the mechanisms of species evolution, we set up experimental protocols to assess the variations of sensitivity in the Common frog (Rana temporaria), among populations submitted to two gradients of environmental stressors: the UVb (280-320 nm) gradient and a gradient of contamination by Polycylic Aromatic Hydrocarbons (PAHs). The hypothesis is that the populations submitted to a high level of environmental stressor in the field should show adaptations giving them a lower sensitivity to those environmental stressor. According to a common garden protocol,. The sensitivity of embryos or/and larvae of R. Temporaria collected in different populations experimenting in the field various levels of stress, was assessed in controlled conditions in the laboratory. We were able to show a local adaptation to UVb in high altitude populations at the embryonic stage as weIl as at the larval stage. Yet, we haven't pointed out any variation of sensitivity to PAHs among larvae coming from populations in more or less polluted areas. Besides we have shown that the larvae from high altitude populations, even if virgin of any PAHs contamination show less DNA damages due to genotoxicity of PAHs. We propose the hypothesis of a co-tolerance between UVb and PHAs. Using tools from ecotoxicology and population biology, we propose explicative hypothesis entering the field of evolutionary biology

Full textThe RAS/RAF/ERK pathway plays a key role in melanoma, with BRAF and NRAS mutations in 50% and 15% of cases, respectively. The development of chemical inhibitors specifically targeting mutated v600EBRAF represents a major breakthrough in the treatment of metastatic melanoma, although resistance often develops quickly following treatment. Furthermore, such compounds cannot be used to treat half of melanoma patients, especially those mutated on NRAS. In this study, we evaluated the contribution of BRAF and its closely related kinase CRAF downstream of NRAS during tumoral progression in mouse models. We developed NRASQ61K-induced mouse melanoma models in which single or compound ablation of BRAF and CRAF genes can be achieved in the melanocyte lineage upon tyrosinase promoter-driven Cre or CreERT2 expression. These models allowed us investigating the role of both RAF kinases at each step of tumoral progression, from tumor initiation to metastatic melanoma. Temporaly-controlled concomitant ablation of BRAF and CRAF abolished nevi formation and melanoma progression and maintenance, showing that RAF signaling is absolutely required for NRAS-induced melanoma development. However, we will present data showing that BRAF and CRAF play specific roles during the different steps of melanoma progression. In addition, using primary cultures of melanomas, we show that ablation of RAF kinases often lead to the emergence of resistant cells showing reactivation of ERK in the absence of BRAF and CRAF. Our results disclose specific and complementary functions for BRAF and CRAF in NRAS-induced mouse melanoma, and pave the road for further studies on treatment and resistance mechanisms

Full textCandida albicans is a harmless constituent of the human microbiota that causes superficial infections as well as life threatening infections in immune compromised individuals. The transition from a budding form to the highly polarized hyphal form is associated with virulence and requires cytoskeleton reorganization and sustained membrane trafficking. In a range of eukaryotes, Ras related protein in the brain (Rab) G proteins and their regulators have been shown to play a central role in membrane traffic. The objective of this work is to understand the role of Rab proteins, in particular Ypt6, the homolog of Human Rab6, in the morphological transition and virulence of C. albicans. To this aim, I generated loss of function mutants and found that YPT6 is not essential for viability, yet was critical for cell wall integrity and invasive hyphal growth, with ypt6 hyphal filaments shorter compared to that of the wild type (WT). Furthermore, YPT6 was important for virulence in two murine candidiasis models. I determined that Ypt6 was localized at the late Golgi compartment during hyphal growth, where it co-localized with Arl1, a small GTPase of the Arf (ADP Ribosylation Factor) family, also required for hyphal growth and virulence. Interestingly, overexpression of YPT6 specifically rescued the hyphal growth defect of the arl1 mutant, but not the converse. Further characterization of the ypt6 deletion mutant showed that the number of Golgi cisternae is increased in this mutant compared to that of WT strain, suggesting an alteration of Golgi integrity. In addition, using live cell imaging I showed that the distribution of Actin binding protein 1 (Abp1), which is a reporter for actin patches, was altered in the ypt6 mutant, in that it was no longer restricted to the tip of the filament, as is observed in WT cells. These data suggest that the defect in hyphal growth maintenance of the ypt6 deletion mutant is at least partly associated with an alteration of the distribution of endocytic sites. Thus, I identified a critical role of Ypt6 during invasive hyphal growth and virulence in the human fungal opportunistic pathogen C. albicans and revealed an interaction between Ypt6 and Arl1 in the hyphal growth process

Unfortunately, although a widespread knowledge, this definition was misused. At intra-specific level, Management Units of conservation (MUs) were usually only based on genetic diversity. Many examples have shown it's limitation. The aims of this work was to describe Rana temporaria intra-specific diversity along an altitudinal in northern Alps, because amphibians are the most endangered vertebrates over the world. Ln behavioural ecology, radiotracking study did not pointed out variation in mobility but in habitat preferences, especially to aestivate and to over winter. Forests were highly used but animaIs tan compensate its absence. At lowland in anthropogenic landscape, connectedness GIS simulation exhibited population fragmentation, unravelled by microsatellite analysis. On the opposite, in natural highland landscape, populations were isolated by distance. Ln life histories diversity, a detailed analysis of life-history traits pointed out at lowland an "explosive" reproductive strategy whereas at highland, a "long breeders" one. Sexual maturation was genetically based and common frog exhibit local adaptations in larval developmental rate

Full textThe objective of this research is to exam the existence of an attachment bond of the younger towards his elder and to show that the elder can be qualified tutor for his younger in a situation of tutoring interaction and lastly, characterize the sibling relationships. We evaluated on a population of 40 siblings (elder : 4-9 years ; younger : 3-4 years) the quality of the sibling attachment bond using an adaptation of story completion task procedure. The elder representation of their sibling relationships is measured with the Sibling Relationships Questionnaire. Lastly, the tutorial interactions within the sibling were observed using a categorization test. Main results highlight that the elder is a "relay attachment figure" for his younger and establishes with him a "relation of proximity-exploratory". The sibling tutorial is characterized by direct assistances and sister use a "guiding supervision" whereas brother use a "freedom supervision". Lastly, there is a modulating tendency of elder representation on the sibling relationships on the existing sibling attachment bond and sibling tutorial interactions

Full textNontyphoidal salmonellae are accountable for :most foodborne infections in Europe and other developped countries. Infection with salmonella is often associated with consumption of foods of animal origin. Antibiotic treatment is not requiered for salmonella gastroenteritis but is essential for patients at high risk of extra-intestinal disease. For many years chloramphenicol, ampicillin, strepromycin, sulfonamides and tetracycline were drugs of choice. The emergence of Salmonella gastroenteritis resistant to these antibiotics has limited theirs therapeutics value. Althouth, fluoroquinolones have proven effective alternatives to treat multidrug resistant straint particularly S.Typhimurium DT104 et 204, résistance to these agents has also emerged. This has been a serious problem both in human and veterinary medicine. In salmonella, fluoroquinolones resistance has been attributed to mutations in the genes coding for gyrase, and topoisomerase IV and also to active efflux pumps. The efflux pump AcrAB is known to play a major role in multidrug rresistance. In E.Coli, acrAB expression is regulated at the lowest level by the local repressor AcrR and at a more glogal level it is regulated by stress conditions and by global regulators like MarA, SoxS or Rob. Overexpression pump AcrAB can confer resistance to wide range of antimicrobial agents and organic solvents which occurs as a result of mutations in one of the genes coding for the regulators. In contrast to E. Coli, mutations in the regulotry regions were not found in multidrug resistant S.Typhimurium overexpressing acrAB genes. Overexpression of ramA also confers multidrug resistance through the activation of acrAB expression in Salmonella entrica. Nonetheless, little knowledge is available with regard to the involvement of mar, sox and ram regions in multidrug and fluoroquinolones resistance in Salmonella entrica

Full textBy way of an experimental model of human African trypanosomiasis (HAT or sleeping sickness), the rat infected by Trypanosoma brucei brucei, we examined the involvement of nitric oxide (NO) in the development of this pathology. In the infected animals, opposite variations in NO concentration were observedeither at peripheral or brain compartments: NO decreases in blood but increases in brain. These changes are dependent on the activity of the inducible NO-synthase (iNOS). In periphery, the decrease observed in NO concentration favors the parasite entrance because the trypanocidal pressure exerted by NO is decreased. In such a situation, L-arginine, the substrate conducing to the synthesis of NO, is employed for the synthesis of polyamines, a category of compounds necessary for the parasite growth. It is likely that above mechanisms might be triggered by parasites. In brain, NO synthesis is submitted to additive complex regulatory processes implying arginase and NG, NG-dimethylarginine dimethylaminohydrolase (DDAH). While the arginase activity remains constant, that of DDAH increases throughout the infection process in keeping with western-blot and amino acids data. This increase, depending mainly on DDAH-2 isoform, lasts in a brain NO increase which enhance the trypanocidal pressure. Above changes, opposite to those observed in periphery, are not favorable to the parasite survival in brain. They might constitute an additive protection against the parasite entry in this organ

Full textChronic pain is an incapacitating and long lasting pathology mainly characterized by threesymptoms: allodynia (a non painful stimulus is perceived as painful), hyperalgesia (a painfulstimulus is perceived as more painful) and ambulatory pains. When chronic pain is due to alesion or dysfunction of nervous system it is called neuropathic pain. In both patients and animalmodels of neuropathic pain, researchers found that thalamic neurons are hyperexcitable. Glialcells, astrocytes and microglia, are strong synaptic partners involved in synaptic transmissionand plasticity and therefore could be involved in this phenomenon. Indeed, these cells canmodify their phenotype when nervous system is damaged. They become reactive: theirmorphology is hypertrophied, mRNA and protein expression of iba-1 (ionized binding-adaptormolecule 1) and CD11b/c (cluster of differentiation 11b/c) for microglia and GFAP (glialfibrillary acidic protein) and S100β (S100 calcium binding protein β) for astrocytes is increased.They could also release pro-inflammatory molecules. All of these could contribute to generate oramplify the thalamic neuronal hyperexcitability.In my PhD work I studied thalamic astrocytes and microglia in a rat neuropathic pain model ofL5-L6 spinal nerves ligation (SNL). Mechanical allodynia and hyperalgesia were characterizedwith von Frey filament test and ambulatory pain with dynamic weight bearing apparatus. mRNAexpression of glial markers were studied with qRT-PCR technique on thalamic punches andlaser-microdissected nuclei. Neurochemical expressions of iba-1, CD11b/c, cathepsin S, GFAPand S100β markers were quantified using an immunohistofluorescence approach to count thenumber of immunopositive cells and surface stained by these markers. All these experimentswere done at D14 and D28 after surgery.At D14, SNL animals develop mechanical allodynia and hyperalgesia as well as ambulatory pain..For these animals, thalamic microglial cells showed signs of reactivity with the increase mRNAexpression of CTSS and CX3CR1, fractalkine receptor, well known markers involved in spinalneuronal hyperexcitability under neuropathic pain conditions. In addition, the number ofimmunopositive cells for the glial markers is decreased in SNL animals. At D28, the neuropathicpain symptoms are still present. Furthermore, thalamic microglial reactivity found at D14 withqRT-PCRm method is still present with the increased mRNA expression of fractalkine (CX3CL1),partner of CTSS/CX3CR1/CX3CL1 pathway. The decreased neurochemical expression of glialmarkers found at D14 was transient as I didn’t find this result at D28. However, thalamicastrocytic reactivity was found at D28 in SNL animals.So, this work reveal a new glial process at thalamic level in this SNL model of neuropathic pain :an early decreased expression of glial markers and then a later thalamic astrocytic reactivityconcomitant with signs of thalamic microglial reactivity. Numerous studies are required toexplore the role of such novel ambivalent glial alterations in the context of neuropathic pain

Full textPain is a major public health issue. Tramadol prescriptions and their consequent poisonings have increased recently. This atypical opioid has various adverse effects including respiratory depression, seizures and serotonin syndrome. In our rat study, we demonstrated that tramadol-induced respiratory toxicity was moderate and worsened by diazepam co-administration. Tramadol-induced seizures were of rapid onset, generalized and accompanied by status epilepticus. Seizures were not related to serotonin syndrome since not induced by serotonin. We suggested tramadol-induced allosteric modulation of GABAA receptors resulting in its inhibition to explain tramadol-attributed seizures. Finally, tramadol-induced serotonin syndrome was responsible for well-characterized clinical symptoms accompanied by mild encephalopathy on the EEG. Interestingly, our results suggested that EEG study was required for an accurate diagnosis of serotonin syndrome in addition to the clinical observations. In order to treat tramadol-induced toxicity, diazepam/naloxone combination appears to be the best treatment to abolish tramadol-induced electro-clinic seizures, to reduce its deleterious respiratory effects, and to reverse its serotonin toxicity. KGNOP1, a bi-functional opioid/anti-nociceptin hybrid suggested as alternative compound to tramadol in the treatment of neuropathic and nociceptive pains, showed advantages with improved efficacy and safety in comparison to tramadol and morphine, despite important deleterious respiratory effects. However, the onset of rapid tolerance to its analgesic effects questioned the possible administration of this hybrid to humans

Full textPACAP is a neuropeptide that exerts important biological activities in vertebrates. In order to gain insight into the phylogeny, the structure/activity relationships, the mechanisms of action and the function of PACAP and its receptors, we have first characterized the precursor of this neuropeptide in a non-mammalian vertebrate, the frog Rana ridibunda. We have shown that the frog PACAP precursor encodes a second peptide termed GHRH-like peptide. These two peptides stimulate GH secretion from frog pituitary cells. Moreover, we have cloned two PACAP receptors in frog : a VPAC1 receptor which exhibits characteristics of the two mammalian VIP/PACAP receptors, and a PAC1 receptor which only recognizes PACAP. Finally, we have identified three PAC1-R isoforms, some of which display particular properties. These results contribute to the understanding of the PACAP-ergic system in vertebrates
