Abstract:
5′-Nucleotidases play an important role in the metabolism of nucleosides; for example, the hydrolysis of AMP generates adenosine, which can modulate a variety of cellular functions. We have used the membrane-bound AMPase from chicken gizzard and a secreted form of these enzymes to analyse their modification by the substrate analogue 5′-p-fluorosulphonylbenzoyladenosine (5′-FSBA). 5′-FSBA irreversibly inactivates 5′-nucleotidases by means of covalent modification of the proteins. ATP, a competitive inhibitor of chicken gizzard and snake-venom 5′-nucleotidase, abolished the inactivation by 5′-FSBA, demonstrating that the inactivation was due to the modification of amino acid residues essential for AMPase activity. We have synthesized radioactive 5′-FSBA, which was employed for the radiolabelling of chicken gizzard 5′-nucleotidase. Incorporation of radioactivity was completely abolished in the presence of ATP, which showed that 5′-FSBA acted by the selective modification of amino acid residues at the active site whereas other potential reactive residues of the protein were not attacked. Limited proteolysis of affinity-labelled chicken gizzard 5′-nucleotidase permitted the identification of digestion products containing the catalytic centre. Pseudo-first-order kinetics indicate that modification of a minimum of one amino acid side chain at the active centre is sufficient to result in inactivation of both chicken gizzard and snake-venom 5′-nucleotidases. Incorporation of the radioactive p-sulphonylbenzoyladenosine moiety parallels the inactivation of 5′-nucleotidase by 5′-FSBA and further substantiated the idea that modification of one amino acid residue at the active centre results in loss of the AMPase activity.

Abstract:
These studies centre on the ‘Barcelona’ karyotypic race of the western house mouse (Mus musculus domesticus), first described by Adolph & Klein (1981). This is one of many races within M. m. domesticus characterized by metacentric chromosomes that have originated by repeated Robertsonian fusions, with perhaps further modification by whole-arm reciprocal translocations. Data on 111 mice from 20 sites show that the race is centred 24 km to the west of Barcelona city and has a homozygous metacentric karyotype of 2n = 28 (3·8, 4·14, 5·15, 6·10, 9·11, 12·13). The race has a small range, and mice with the standard 40-acrocentric karyotype were caught only 30 km from the race centre. Throughout the area of occurrence of metacentrics there is polymorphism (i.e. presence of acrocentrics in the population), although all six metacentrics approach fixation close to the race centre. Thus, there is a hybrid zone between the Barcelona and standard races. The centres and widths of all clines (except 3·8) were determined. Likelihood ratio tests showed that most of the cline centres differed significantly in position (i.e. the clines were staggered) and the clines for metacentrics 6·10 and 9·11 were significantly narrower than those for 4·14, 5·15 and 12·13. Overall, the clines tended to be wider the further they were from the race centre. There are various possible explanations for this hybrid zone structure and further data are needed to distinguish between them.

Ces opérations de prépolissage et de polissage sont encore régulièrement réalisées manuellement. En effet, malgré des avantages en termes de répétabilité, de productivité et de qualité géométrique, les méthodes de polissage automatique sont peu utilisées, car elles nécessitent une mise au point importante. Les travaux de recherche présentés dans ce mémoire participent à la maîtrise du procédé de polissage automatique tout en contrôlant la qualité géométrique des pièces. Pour parvenir à cette maîtrise, un outil de simulation de l'enlèvement de matière est mis en place. Cet outil permet de simuler l'enlèvement de matière au cours d'une opération de prépolissage réalisée sur centre d'usinage 5 axes. Il se base sur un modèle du contact obtenu entre l'outil de pré-polissage et la pièce, ainsi que sur un modèle du pouvoir abrasif intégrant l'usure et l'encrassement du disque. Cette simulation permet de vérifier la régularité de l'abrasion sur une surface et d'identifier les zones pouvant faire apparaitre des défauts macro-géométriques importants. Une méthode est également proposée pour compenser les variations du pouvoir abrasif au cours du temps. La compensation s'effectue en optimisant les consignes de vitesse d'avance et/ou de fréquence de broche le long de la trajectoire. Cette méthode de pilotage permet d'obtenir un taux d'enlèvement de matière plus constant et ainsi de réduire les défauts géométriques générés pendant une opération de prépolissage.

Abstract:
In 2020, the University of Dundee initiated the development of an Open Research strategy. As part of this initiative, in February 2021 the University’s Library and Learning Centre together with Open Research Champions from the Schools of Health Sciences and Dentistry, formed an Open Research Working group. To build on the University’s open research policy and infrastructure, the purpose of the group was to facilitate ongoing research and development of best practice approaches for our interdisciplinary environment to make outputs, data and other products of our research publicly available, building on University of Dundee’s Open Research policy and infrastructure. Through informal consultations with academic staff and students, the Open Research Working Group found that: → access and reach of research findings can be amplified through effective knowledge mobilisation, and stakeholder and patient and public involvement; and → there was a need for guidance and resources on how-to implement knowledge mobilisation activities with and for stakeholders throughout the entire research process – from proposal development to project completion. In June 2021, the Open Research working group, in partnership with Simon Fraser University’s Knowledge Mobilization Hub began the development of an Integrated Knowledge Translation (IKT) Toolkit, with funding support from the University of Dundee’s Doctoral Academy and Organisational Professional Development. IKT is an approach to knowledge translation that emphasises working in an engaged and collaborative partnership with stakeholders throughout the research cycle in order to have positive impact. The aim was to co-produce evidence-informed, best practice learning materials on how-to: → maintain ongoing relationships between researchers, community stakeholders and decision-makers in research development and implementation; and → facilitate an integrated, participatory way of knowledge production whereby researchers, practitioners and other knowledge users can collaborate to co-generate new and accessible knowledge that can be utilised in contexts ranging from supporting community development to policy guidance for practice. The IKT Toolkit was informed by a focused evidence review and synthesis of published peerreviewed and grey literature and consists of 8 knowledge briefs and a slide deck co-produced for use in any discipline or sector. Each knowledge brief provides practical guidance and resources to support an IKT process in each of eight key research stages: (i) Partnership Building; (ii) Generating Priorities and Ideas; (iii) Proposal development; (iv) Study Design; (v) Data Collection; (vi) Data Analysis; (vii) Reporting and (viii) Dissemination. The current knowledge brief provides IKT guidance on Research Stage 5: Data Collection.

Abstract:
Under the new Fiscal Framework and devolution of some tax powers, from April 2019, the Welsh Government and local authorities will control nearly £5 billion of tax revenues, equivalent to roughly 30 per cent of their combined current spending. In this report we have worked with the Wales Governance Centre at Cardiff University to examine the key characteristics of the Welsh tax base, the risks and opportunities to the Welsh tax base after fiscal devolution, and some of the implications for Welsh Government policy. It concludes that tax policy reform should be pursued in an integrated way, with all devolved and local taxes considered in the round. It also highlights that given the influence that wider policy areas like education and housing have on the economy, there are cross-departmental challenges for the Welsh Government to ensure it successfully manages the increased risk inherent to fiscal devolution.

The Geological Survey of Canada (GSC)mp;gt;'s Public Safety Geoscience Program (PSGP) has collaborated with the Canada Centre for Remote Sensing (CCRS) to assess the performance of new RCM data for monitoring slope movements. The PSGP has the mandate to study natural hazards and provide baseline geoscience information to help stakeholders and decision-makers mitigate against potential risk. This report provides preliminary results observed from new RCM InSAR data acquired over 21 sites in British Columbia (BC), Alberta (AB), and Nunavut (NU) from April 2020 to September 2021. , In some cases, comparisons with RCM imagery were made with RADARSAT-2 and Sentinel-1 observations. A total of 13 sites in BC, two sites in AB, and six sites in NU that are located close to communities and/or infrastructure were investigated. From these, we acquired a total of 1235 RCM single look complex (SLC) images of HH polarization (ascending: 514, descending: 721) from April 2020 to September 2021. Most were acquired with 3 m very-high-resolution and/or 5 m high-resolution modes. Based on the preliminary observations, the advantages and limitations of RCM InSAR for landslide monitoring are highlighted.
