The paper deals with the bicycle center infrastructure in a city, which becomes popular every year, but is not yet studied properly. Little attention is devoted to the spatial development for two-wheeled vehicles, so there are many problems to be discussed and solved.The purpose of this paper is to determine the types of bicycle centers in the structure of a city, from the main and central to regional and peripheral that connect all urban areas. Moving along the bicycle paths from home to a large bicycle center and back to home, one can communicate in bicycle centers. Less functional peripheral centers provide repair services, bicycle rental and small recreation. The city of Rostov-on-Don is taken as an example.The following parameters are determined: optimum distance between bicycle centers of different types, their functional content, bicycle center location in the city. In the future, it is possible to arrange such centers in any city due to their unification.The model of the bicycle center network is proposed for the city of Rostov-on-Don, that are characterized by different area and multifunctional purposes for each district to attract people. The main bicycle center includes sports grounds, bicycle track, rock climbers, skate park, gyms, ski jumping hall and other sports facilities. Roof-top bike trails, amphitheater with a street stage, art spaces, go-karting can be arranged on the territory of these centers.

ABSTRACT:Most colonial Hispanic American cities were originally planned around a main plaza, which was a multifunctional square crucial for urban life. This spatial model for the whole city based on a main square is termed thePlaza Mayormodel. Bourbon reforms of the second half of the eighteenth century aimed at transforming this model according to aPlaza de Armasorganization. Here, these two models (Plaza MayorandPlaza de Armas) are characterized, and their contradictions in terms of political projects and quotidian city life are analysed. For late colonial Lima, Bourbon efforts to introduce thePlaza de Armasare shown to have affected both the main function of the central square and the entire urban system.

The aim of the study is to calculate the coefficients of M. Williams asymptotic expansion of stress anddisplacement fields using the data of finite element modeling of a plate with an inclined central crack in a uniaxial tension field. In this work, we also simulated the loading of a half-disk with a vertical and oblique notch under conditions of three-point bending. The simulation was carried out in the multifunctional software SIMULIA Abaqus. The paper proposes an algorithm for calculating the coefficients. The program, written in the MAPLE computer algebra system, allows calculating any predetermined number of M. Williams expansioncoefficients (amplitude or scale factors) and uses the values of the stress tensor components at points in the vicinity of the crack and their coordinates as input. The analysis of the influence of the number of calculated coefficients on the accuracy of their determination is carried out. Recommendations on the choice of points for calculating the coefficients are given.

ABSTRACTHerpesviruses are nuclear-replicating viruses that have successfully evolved to evade the immune system of humans, establishing lifelong infections. ICP27 from herpes simplex virus is a multifunctional regulatory protein that is functionally conserved in all known human herpesviruses. It has the potential to interact with an array of cellular proteins, as well as intronless viral RNAs. ICP27 plays an essential role in viral transcription, nuclear export of intronless RNAs, translation of viral transcripts, and virion host shutoff function. It has also been implicated in several signaling pathways and the prevention of apoptosis. Although much is known about its central role in viral replication and infection, very little is known about the structure and mechanistic properties of ICP27 and its homologs. We present the first crystal structure of ICP27 C-terminal domain at a resolution of 2.0 Å. The structure reveals the C-terminal half of ICP27 to have a novel fold consisting of α-helices and long loops, along with a unique CHCC-type of zinc-binding motif. The two termini of this domain extend from the central core and hint to possibilities of making interactions. ICP27 essential domain is capable of forming self-dimers as seen in the structure, which is confirmed by analytical ultracentrifugation study. Preliminaryin vitrophosphorylation assays reveal that this domain may be regulated by cellular kinases.IMPORTANCEICP27 is a key regulatory protein of the herpes simplex virus and has functional homologs in all known human herpesviruses. Understanding the structure of this protein is a step ahead in deciphering the mechanism by which the virus thrives. In this study, we present the first structure of the C-terminal domain of ICP27 and describe its novel features. We critically analyze the structure and compare our results to the information available form earlier studies. This structure can act as a guide in future experimental designs and can add to a better understanding of mechanism of ICP27, as well as that of its homologs.

En este artículo estudio el problema de la significación del espacio portuario y sus consecuencias en la configuración social y cultural de las ciudades marítimas y fluviales que lo contenían. Mi reflexión se centra en los pequeños espacios situados en los centros históricos que concentraban las labores portuarias y otras actividades que determinaban la totalidad de la vida de esas poblaciones. Esas áreas fueron vitales hasta que por las nuevas actividades, el crecimiento demográfico y urbano de la primera mitad del siglo XX, fueron sacadas a las afueras de las ciudades y aislados del resto de los espacios urbanos y de otras actividades con las que habían permanecido confundidos durante varios siglos. El argumento central de este artículo propone que en comparación con las ciudades mediterráneas organizadas en torno a la plaza central, las ciudades portuarias poseyeron una dualidad entre ese espacio como sitio de residencia de los notables, las autoridades civiles y eclesiásticas, y en consecuencia de control social, y la plaza portuaria que por su polifuncionalidad en cierta medida fue construida de abajo hacia arriba. Ese protagonismo del puerto en la vida urbana se debió a la diversidad de actividades que contenía, pues era espacio de trabajo, intercambio cultural, plaza de mercado, sitio de diversión y ocio. Esto marcó de singular manera a la sociedad de los puertos pues determinó unas dinámicas sociales, culturales, laborales distintas a las escenificadas en las ciudades mediterráneas.Palabras clave: puertos, Gran Caribe, ciudad portuaria, espacio polifuncional, plaza portuaria, plaza central.From port city to the port as a multifunctional space. The Gran Caribe ports in the 19th century. Abstract In this article I study the problem about the port area importance and its impacts on social and cultural arrangement of maritime and river cities. My idea is focused on small areas located in the historic centers that hold port activities and labors that ruled the whole life of these populations. These areas were vital but due to new activities, urban and population growth in the first half of the 20th century, they were taken to the cities’ outskirts and were isolated from other urban areas and activities that had been with for several centuries. The main argument of this paper says that in comparison to Mediterranean cities that were organized around the main square, port cities had duality between the space for important people, civil and ecclesiastical authorities as a residence place, and because of that for social control. The port main square was built from down to up by its somehow multifunctionality. The port importance in urban life was because of the activities diversity it had, as it was a working, cultural exchange, and marketplace place, as well as entertainment and leisure site. This marked the society ports in a unique way as it defined different social, cultural, labor dynamics from Mediterranean cities. The present paper highlights the importance of hedgerows and enclosures in the mountains of Central Spain. Now, these landscapes have suffered profound variations in terms of agroforestry practices, especially in the Mediterranean mountains where the characteristic multifunctional has largely been lost. The article analyzes land uses changes, dynamics, and their morphological features between the first half of the 20th Century (1956) and the second decade of the present time (2019). The paper was divided into three sections. First, the identification of land uses using orthophotograph and aerial photograph; after that the info was checked with fieldwork. Eleven categories were identified according to the dominant use and land use changes and size of land parcels were taken into consideration. Second, the configuration and the information collected through the type and intensity of change in land uses made it possible to recognise and quantify their distribution and trend between these two dates. Also, the kernel density algorithm available in the Arcgis 10.5 software was used to obtain density and changes in land parcels. Finally, an overview is given of the main role that this agroforestry plays due to the social, ecological, and economic benefits that they provide for allowing sustainable development.

The phosphatidylinositol 3-kinase (PI3K) pathway plays a central role in the regulation of cell signaling, proliferation, survival, migration and vesicle trafficking in normal cells and is frequently deregulated in many cancers. The p85α protein is the most characterized regulatory subunit of the class IA PI3Ks, best known for its regulation of the p110-PI3K catalytic subunit. In this review, we will discuss the impact of p85α mutations or alterations in expression levels on the proteins p85α is known to bind and regulate. We will focus on alterations within the N-terminal half of p85α that primarily regulate Rab5 and some members of the Rho-family of GTPases, as well as those that regulate PTEN (phosphatase and tensin homologue deleted on chromosome 10), the enzyme that directly counteracts PI3K signaling. We highlight recent data, mapping the interaction surfaces of the PTEN–p85α breakpoint cluster region homology (BH) domain, which sheds new light on key residues in both proteins. As a multifunctional protein that binds and regulates many different proteins, p85α mutations at different sites have different impacts in cancer and would necessarily require distinct treatment strategies to be effective.

Fibrillins constitute the major backbone of multifunctional microfibrils in elastic and nonelastic extracellular matrices. Proper assembly mechanisms are central to the formation and function of these microfibrils, and their properties are often compromised in pathological circumstances such as in Marfan syndrome and in other fibrillinopathies. Here, we have used human dermal fibroblasts to analyze the assembly of fibrillin-1 in dependence of other matrix-forming proteins. siRNA knockdown experiments demonstrated that the assembly of fibrillin-1 is strictly dependent on the presence of extracellular fibronectin fibrils. Immunolabeling performed at the light and electron microscopic level showed colocalization of fibrillin-1 with fibronectin fibrils at the early stages of the assembly process. Protein-binding assays demonstrated interactions of fibronectin with a C-terminal region of fibrillin-1, -2, and -3 and with an N-terminal region of fibrillin-1. The C-terminal half of fibrillin-2 and -3 had propensities to multimerize, as has been previously shown for fibrillin-1. The C-terminal of all three fibrillins interacted strongly with fibronectin as multimers, but not as monomers. Mapping studies revealed that the major binding interaction between fibrillins and fibronectin involves the collagen/gelatin-binding region between domains FNI6 and FNI9.

Neurons in the Doppler-shifted constant frequency processing (DSCF) area in the primary auditory cortex of mustached bats, Pteronotus parnellii, are multifunctional, responding both to echolocation and communication sounds. Simultaneous presentation of a DSCF neuron's best low and high frequencies (BFlow and BFhigh, respectively) facilitates its response. BFlow corresponds to a frequency in the frequency-modulated (FM) component of the first harmonic in the echolocation pulse, and BFhigh corresponds to the constant frequency (CF) component in the second harmonic of the echo. We systematically varied the slopes, bandwidths, and central frequencies of FMs traversing the BFhigh region to arrive at the “best FM” for single DSCF neurons. We report that nearly half (46%) of DSCF neurons preferred linear FMs to CFs and average response magnitude to FMs was not significantly less ( P = 0.08) than that to CFs at BFhigh when each test stimulus was paired with a CF at BFlow. For linear FMs ranging in slope from 0.04 to 4.0 kHz/ms and in bandwidth from 0.44 to 7.88 kHz, the majority of DSCF neurons preferred upward (55%) to downward (21%) FMs. Central frequencies of the best FMs were typically close to but did not always match a neuron's BFhigh. Neurons exhibited combination-sensitivity to “call fragments” (calls that were band-pass filtered in the BFhigh region) paired with their BFlow. Our data show a close match between the modulation direction of a neuron's best FM and that of its preferred call fragment. These response properties show that DSCF neurons extract multiple parameters of FMs and are specialized for processing both FMs for communication and CFs for echolocation.

Diploma thesis deals with new building sports and relaxation centre. Building is situated near Masaryk university campus in Brno-Bohunice. The building is two-storey, without cellar, set in a slightly sloping terrain at the back of the street Netroufalky. The building has two movement units. One unit contains sports hall and squash court and second unit contains cloakrooms, wellness part and administration. Structural system is cast-in-place concrete with masonry external wall of Porotherm. Internal wall are also from Porotherm. Sports hall with parabolic arch has steel structural system, that isn´t part of project. Curtain wall is covered with lamella Kalzip. Next to the building is parking place for clients.

Diploma thesis Ponava Centre is solving the architectonic study of Leisure centre. Area is situated in locality Ponava – Brno, Královo Pole. Leisure centre should provide its services to wide spectrum of visitors. Those activities does not have to be undertaken just in the building itself, but the idea to develop and utilize for those purposes the public space around it is suggesting itself. The centre is offering the activities for people of all age categories. It can provide the leisure function and also both educational and commercial function. In this spirit the inner space of the object is divided. The basic principle is to group together the three masses according to their most important functions, which are located in the object.

The area is located in Kohoutovice on Libušina trida street. The location of socio-cultural center with townhall is at the site of the largest concentration of people and intensive traffic in the area. The concept is based on the attempt to create an architecture blending into the surrounding landscape , respecting the context of space and favorably affect the environment. The building consists of two spaces and its dispositions forms a clearly defined exterior space - the new square. The main entrance and the main pedestrian axis are oriented to the square. Externally, the timber building is distinguished by the interplay of wood cladding and vertical louvres, variable light and shadow.

The assignment is an architecture and urban planning study of a sports and recreation center. The solved area is located in the town of Vsetín, in the part called Ohrada. Nowdays, the area is obsolete, unfunctional and also unmodern. My project is mainly focused to create a concept that responds to the environment, offers outdoor and indoor spaces for all generations. It allows better connection with nature, variable use, and division of area by function. The design is defined by the river Bečva on the west side and a busy 1st class road on the east side. It was important to separate pedestrians and cyclists from motor vehicles. The response to this restriction is to direct traffic in the area to the west, towards the river. This creates a terrain works that optically deflects the design in a quieter direction. Buildings are placed in the line and oriented in the western direction. After creating the terrain, I divided the area into several segments according to functionality. In the heart of the area, a multifunctional central hall with an inner climbing wall is set in the terrain. In front of it is a public space for meetings, sports and recreation. On the other side, there is a basketball court, a workout playground, a children's playground in front of the hall and a rest area of ??greenery closer to the river. If we continue southerly down, another cube set into the terrain is a building with a restaurant and a cafe. The southernmost building is a tennis club with facilities for athletes and squash. In front of the building there is a 9x tennis court with a grandstand. Closer to the river and the bike path there is an outdoor climbing wall. From the newly created embankment, the area is divided by a cycle path around the river. Behind the bridge is a green belt separated area which, unlike the previous ones, is tuned in a calmer spirit. We will find a building of a yoga and meditation center.

The main idea in dealing with my thesis was to design a high quality public space where people will feel comfortable, safely and have a reason to come back. Furthermore, this is the place to be full of life and will serve the general public. The cultural center, that was designed, to become part of Královo Pole cultural campus, so-called K2-center, which will create a basis for free time activities, relaxation and education. A significant part of the proposed building is made up of multi-functional hall, library, restaurant, teen center and spaces for leisure time activities. On the ground floor will be located business parter and coffee bar. The aim of my project was to take into account what is most needed for inhabitants of Královo Pole for the purpose to meet the demands for quality leisure time for the widest spectrum of people. There is an effort to place the object itself as the most sensitive, given the current surrounding areas. The aim was not to design a huge object but especially a pleasant environment functioning as a center of leisure time activities.

The main aim of the diploma thesis was to design a building that meets the requirements provided by the announcer and at the same time has a positive impact on the area in both closer and larger context. This may include increasing the number of ecclesiastical buildings, improving the quality of local architecture, expanding the range of social and leisure activities, and cultivating terrain and greenery. All of above on the basis of the analysis of the locality in its context and further coherences (cultural, historical). The object is not only to provide background to enlisted activities, but also to serve as a representative building attracting new visitors. It is an active space connecting the urban population with culture, nature and above all God - with the help of its location on a visible, attractive place with good access. It will be a dominant building expressing its purpose, related to its surroundings and functionally cooperating with the cultural and social happening in Líšeň with a transition to a city-scale dimension.

Chosen territory is located on the Libušina street, Brno - Kohoutovice. Socio-kultural center with townhall will serve as the dominant of district. Due to its shape, which is growing up, it reflects the character of the surrounding high-rise buildings. Entrance to the building is concentrated in one space, the communication part, which connects two masses - the town hall and socio-cultural center. Office space is structured around the perimeter of the building and also organized as a contact point with the society, due to glass partitions. The interior space of the town hall is lighted by glass atrium that allows sunlight penetrates to the whole building. Socio-kultural center with townhall is an expressive architectural element and includes three functions: culture, recreation and sport. The main part of the building is the multifunctional hall, that is used for such purposes as big celebrations, performances, music and dance events and film screenings. The idea to create the function of sport is based on interview and wishes of the local inhabitants. The restaurant, library and a recreation space have large glass windows, which allows visual connection between the interior and exterior and creates a pleasant aesthetic impression.

The submission of this thesis was preceded by a specialised studio „Brněnské nábřeží a kulturní čtvrť Ponava“. This work focused on a complex urban architectural solution to the area around the former “Jaselská kasárna”, the urban context, transport services, the field configuration etc. The thesis follows this project in a section defined by streets U Červeného mlýna, Staňkova and Střední. The goal of the thesis lies in the creation of an intensive goal – a cultural area, which will offer a high-quality space for intergenerational communication and new social experiences and activities. It newly connects the urban part Ponava with the town center of Brno by a high-quality axis for pedestrian and cycling transport. The project of this cultural area arose after a thorough analysis of surrounding, intercity and global relations as a combination of aims: for children – the elementary art school with graphic and dancing fields, for seniors – leisure time center, and for all together - a multifunctional concert hall and a revitalized area of the former industrial zone of furniture production to a multipurpose intergenerational area with a modern art gallery, offices for young businessmen, architects and artists, and a music club in the basement of the gallery.
