Abstract:

This study has been assigned by Bengt Hellman who works at the logistic department at IKEA Torsvik just outside of Jönköping. The task has been to develop a new picking strategy for the Oversize area in the CDC-Warehouse. The reason why IKEA wants a new strategy is because they want to minimize the route length for the forklifts when collecting the orders.

By evaluating the current strategies on other areas in the CDC-storehouse, study literature within this subject and look at restrictions for the storing of goods, we have analyzed how the Oversize area works today. We compared the gathered information, to how it should be done according to the literature to be able to work out a new functional strategy.

Today, IKEA does not have a working strategy for the oversize area because of lack of time and because all the power has been put on other areas were sales rates are currently higher. This have led to lack of organisation at the Oversize area and items are just put were there is space without first analysing were it should be placed.

The strategy that we have worked out and introduced to IKEA builds on easiness of understanding the layout, the employees shall know why an item is placed where it is. We have also analyzed which items that are frequently ordered with each other. We have put weight on trying to keep those items as close as we can to minimize the route length for the forklifts. To help IKEA with reorganizations in the future we have made it as easy as we could to move high- and low frequently items around and also introducing new articles in the storehouse, this by the reason that sales rate can change drastically when a new sales campaign is initiated by IKEA


Detta examensarbete är utfört på önskemål av Bengt Hellman som arbetar på logistikavdelningen på IKEA Torsvik utanför Jönköping. Arbetet har gått ut på att ta fram ett nytt förslag på hur de skall placera sina artiklar på plocknivå (vad IKEA kallar för mappning) inne på Oversize avdelningen på IKEA:s CDC-lager. Anledningen till detta är att IKEA vill minska sina körsträckor för plockarna inne på lagret och därmed öka effektiviteten.

Genom att studera befintliga mappningsstrategier som redan finns på övriga avdelningar på CDC-lagret, litteraturstudier och titta på vilka begränsningar som finns inne på lagret så har vi analyserat nuläget mot teori för att sedan kunna ta fram en ny strategi för hur en fungerande mappning skulle kunna se ut.

I dag så finns det inte någon väl utarbetat strategi för mappningen på Oversize avdelningen, detta på grund av att andra avdelningar prioriterats på grund av att de säljer mycket mer i dagsläget. Detta har även medfört att den huvudsakliga uppdelning som fanns tidigare på Oversize avdelningen med affärsområden har fått stå åt sidan på grund av tidsbrist och artiklar har placerats in där det finns plats istället för att analysera var de kan placeras bäst.

Det nya förslaget som vi presenterar för IKEA bygger på att det skall vara enkelt att förstå layouten, plockarna skall veta varför en artikel finns där den finns. Vi har även tagit stor hänsyn till vad som säljs med vad och försökt placera dessa artiklar i närheten av varandra för att på så sätt minska körsträckorna. Vi har även haft i åtanke att det skall vara enkelt att placera om hög och lågfrekventa artiklar samt nyheter inne på lagret då försäljningsvolym påverkas väldigt mycket utav olika kampanjer som IKEA har.

Abstract:
The thesis mainly focuses on structural study of proteins in membrane attack complex- perforin/cholesterol-dependent cytolysins (MACPF/CDC) superfamily, in particular, Astrotactins from human and perforin-like proteins (PLPs) from Toxoplasma and Plasmodium. Both of these subfamily proteins are implicated in human diseases and structurally uncharacterised before. Astrotactins (ASTNs) have been shown to play a crucial role in enabling neural migration along glial fibres. While ASTN1 directly forms contacts between the neuron and the fibre, ASTN2 is responsible for extracting ASTN1 from contacts at the lagging edge of the cell and recycling them to the leading edge. ASTN2 is associated with endosomes, with the majority of its structure (at the C-terminus) projecting into their lumen, anchored by a pair of transmembrane –-helices. Here we present the structure of this "endodomain" region of ASTN2, and find it to consist of a unique combination of polypeptide folds comprising a membrane attack com- plex/perforin (MACPF) domain, an EGF-like domain, a previously-unobserved form of fibronectin type III (Fn(III)) domain and an annexin-like domain. Taken together, the structural characterisation provides a framework for better understanding the mechanism of the ASTNs and related proteins in neural and other forms of vertebrate development. Perforin-like proteins from Toxoplasma and Plasmodium (TgPLP1 and PPLPs) are critical for normal life cycle progression of these parasites, and knockout out of any of them results in significant defects in their life cycle, entrapping the parasites within the host cells and thus limiting their ability to egress. Here we present the crystal structures of TgPLP1 MACPF domain and C-terminal domain at 2.0 Å and 1.1 Å, respectively. We also presents the MACPF domain assembled in helical and hexameric ring form, which indicates the possibility of pore-formation by 6 subunits. This is the first structure of perforin-like protein from Apicomplexan parasites and provides a structural basis to elucidate the function of PLPs in toxoplasmosis and malaria pathogenesis. The final section is a continuation of a previous study in our lab on pleckstrin homology (PH) domain of kindlins. We determined the crystal structure of kindlin-3 PH domain and characterised its lipid and membrane binding properties. Using nanodiscs incorporated with different lipids as model membrane system to study the interaction between inositol phosphate lipids and kindlin-3 PH domain, together with molecular dynamic simulation studies (in collaboration with Mark Sansom's group, University of Oxford), we propose that a subset of PH domains is able to bind to multiple inositol phosphates simultaneously and so via an avidity effect have its interaction with target membranes strengthened.

Abstract:
BACKGROUND: Multi drug resistant Tuberculosis (MDR-TB) is a serious public health concern in many parts of the world. As per the WHO- 2010 global report on Surveillance and response 3.6% of all incident TB cases globally are multidrug resistant. In this regard, there is an increasing demand for timely, reliable and comprehensive drug susceptibility testing (DST) as MDR-TB surveillance is being geared up. The intent of this analysis is to determine whether there is a need to continue routine confirmatory DST testing at CDC in addition to just sending the isolates for genotyping. Analysis is done by measuring the discordance between the results of laboratory DST at CDC and the local labs drug type, drug testing concentrations, and study sites. METHODS: The data for this analysis was provided by the Tuberculosis Trials Consortium (TBTC), CDC. Data for this analysis was collected over nearly two decades (1993-2011), gathered from 7 clinical trials. Discordance between the local and CDC lab DST results was measured using Kappa statistic. Sensitivity and specificity analysis was done by taking the CDC DST lab results as the gold standard. Discordance levels were calculated by local sites and baseline drug resistance for each antibiotic in each study was measured. RESULTS: Average Kappa values for inter rater agreement for all the studies was 0.6444 whereas the overall level of discordance across all studies is 7.786%. Drug resistance at baseline was highest for Isoniazid and Streptomycin (except Study 23 and 22). CONCLUSION: Though the current results show few DST result discordances between local and CDC labs, it is better to continue to send isolates to the centralized lab (CDC) in view of the worldwide threat of drug resistant TB epidemic, the recommendations of the current literature and the benefits of reliable confirmatory testing services and availability of other molecular diagnostic methods.

CDC Group IVc-2 est un bacille à Gram négatif, aérobie strict, mobile grâce à une ciliature péritriche, oxydase et catalase positives et non fermentant. Connu depuis les années 80, il a d'abord été rapproché de Bordetella bronchiseptica puis parfaitement différencié biochimiquement de cette espèce en 1986 par Pickett (87). Sa position taxonomique longtemps indéterminée a été précisée en 1999 (74, 83, 116) et nous y avons contribué (74). Bactérie de l'environnement hydrique (7, 63, 82). .
Our study of CDC Group IVc-2 began in 1995 with the report of five bacteriema at Trousseau hospital (APHP). Genotyping with RAPD, used for the first time for this species, showed a single pattern. Pulsed field gel electrophoresis, applied to Trousseau isolates and other French isolates, was unsuccessful since DNA was degraded. These isolates remained undistinguished after genotyping with PCR-ribotyping, PCR-RFLP, IRS-PCR. Surprisingly, all the strains obtained from American (ATCC) and Belgian (LMG) collections were easily distinguished by RAPD. We contributed to the taxonomic study of CDC Group IVc-2 named Ralstonia paucula in. .

Abstract:
RNA splicing is a process by which introns are excised from precursor mRNA. Variations in the segments removed — and the resulting mRNA molecule — may result in gene transcripts with differing and even opposing functions. The mechanisms involved in RNA splicing are tightly regulated, the disruption of which has been implicated in several human diseases including cancer. This presents the RNA splicing machinery as a potential therapeutic target. However, the effects of systematic splicing modulation through pharmaceutical intervention remain under explored. A thorough understanding of splicing can be investigated through controlled disruption of the molecular machinery. The Takeda Pharmaceutical Company Limited (Osaka, Japan) has recently developed a novel compound that inhibits the CDC-like family of kinases, which regulate key splicing factors. Although splicing inhibitors have already been published, their effects on the RNA splicing landscape have not been systematically described. The creation of a novel splicing inhibitor presents the opportunity to perform a methodical analysis of transcriptomic response to RNA processing inhibition using modern RNA sequencing and analysis methods. It is demonstrated, using the Takeda compound, that restricting the function of CDC-like kinases perturbs RNA splicing in both malignant and normal cells in a dose dependent manner. Post-treatment changes in splicing patterns revealed that these changes are mainly due to inefficient recognition of RNA splice sites. Splicing factors were among the earliest responders to treatment, indicating splicing autoregulatory mechanisms are sensitive to changes in splicing efficiency. Downstream effects were seen as dose-dependent changes in gene expression regulation, and down-regulated genes were enriched for splicing factors. Treatment also resulted in increased generation of conjoined gene transcripts — RNA molecules transcribed from at least two different genes, likely caused by transcriptional read-through. This revelation points to a previously undescribed role for CDC-like kinases in RNA processing.

O presente estudo tem por objetivo demonstrar a necessidade de se tutelar a desigualdade substancial existente nas relações contratuais celebradas entre empresários, quando uma das partes, ainda que profissional, encontrar-se em situação de dependência econômica, favorecendo o abuso da parte contrária na situação concreta. O cerne do trabalho gira em torno do problema das cláusulas abusivas no âmbito dos contratos interempresariais. Busca-se demonstrar que, nas hipóteses em que não for possível repreendê-las por meio da aplicação do Código Civil ou da Lei Antitruste, seria possível equiparar o contratante vulnerável, sujeito a um abuso por parte de seu parceiro contratual, aos consumidores, para fins de aplicação dos dispositivos do Código de Defesa do Consumidor CDC ao contrato em questão. Para tanto, são analisados os conceitos de empresário, de consumidor tendo em vista as três principais correntes doutrinárias existentes no ordenamento pátrio e de dependência econômica. Especificamente em relação à definição de consumidor, tenta-se demonstrar que, nos dias de hoje, a Teoria Finalista Aprofundada parece ser a que melhor atende à necessidade de se buscar a solução mais justa no caso concreto, sem, contudo, banalizar a aplicação do CDC. Por fim, faz-se uma análise da jurisprudência brasileira sobre o tema, com o objetivo de delimitar os critérios para a incidência do CDC em contratos interempresariais, bem como os principais casos em que o conceito de consumidor-equiparado tem prevalecido nos litígios decorrentes de tais contratos.
The objective of the present study is to demonstrate the need of instructing the substantial inequality existing in contractual relations signed between businessmen, when one of the parties, although still professional, finds itself in a situation of economic dependence, favoring the abuse of the counterpart in the real situation. The core of this work involves the problem regarding abusive clauses in the scope of inter-business contracts. It seeks to demonstrate that, in hypothesis where it is not possible to reprehend them by means of application of the Civil Code or Antitrust Act, it would be possible to match the vulnerable contracting party, subject to an abuse by its contractual partner, to the consumers, for purposes of application of the provisions in the Consumer Defense Code CDC to the contract in question. To do so, it analyzes the concepts of businessman and consumers considering the three main doctrinaire schools of thought existing in the country system and economic dependence. Specially in relation to the definition of consumers, it attempts to demonstrate that, nowadays, the In-depth Finalist Theory seems to be the best theory that meets the need of searching for the more righteous solution in the real case, without, however, trivializing the application of the CDC. At last, an analysis of Brazilian jurisprudence is made on the subject, aiming to delimitate the criteria for the incidence of CDC in inter-business contracts as well as the main cases in which the concept of consumer has prevailed in such contracts for one of the parties.

Abstract:
Polymer-based carbide-derived carbons (CDCs) with combined micro- and mesopores are prepared by an advantageous sacrificial templating approach using poly(methylmethacrylate) (PMMA) spheres as the pore forming material. Resulting CDCs reveal uniform pore size and pore shape with a specific surface area of 2434 m2 g−1 and a total pore volume as high as 2.64 cm3 g−1. The bimodal CDC material is a highly attractive host structure for the active material in lithium–sulfur (Li–S) battery cathodes. It facilitates the utilization of high molarity electrolytes and therefore the cells exhibit good rate performance and stability. The cathodes in the 5 M lithium bis(trifluoromethanesulfonyl)imide (LiTFSI) electrolyte show the highest discharge capacities (up to 1404 mA h gs−1) and capacity retention (72% after 50 cycles at C/5). The unique network structure of the carbon host enables uniform distribution of sulfur through the conductive media and at the same time it facilitates rapid access for the electrolyte to the active material.

Abstract:
A chitinolytic organism was cultured from a lotic habitat in central Indiana. It was isolated on the basis of its hydrolytic activity on chitin agar. It was subsequently identified as EF-4a. The chitinases of the EF-4a were isolated, purified and concentrated. A colloidal chitin suspension was then incubated with the enzyme. Samplings were taken at one minute, one hour and 24 hours. The samplings were filtered through a Swinney filtration unit and immediately after applied to a Beckman Ultrasil Amino column. It was eluted with acetonitrile: water (75:25) at a flow rate of 2 ml/min. The absorbance at 214 nm was monitored using the Beckman 160 UV detector. The chromatographs were analyzed for product elaboration. It was observed that EF-4a produced soluble, extracellular enzymes. The enzymes were a chitinase-chitobiase complex. The enzymes' action on chitin was immediate. There were varying reaction products including G1cN, GlcNAc, chitobiose, chitotriose and higher order oligomers. There was also chromatographic evidence suggesting a and B anomers of these saccharides.

This thesis presents an analysis of the Siochain Rental Rehabilitation Project to identify areas of concern for the development procedures of the Neighborhood of Affordable Housing (NOAH), a Community Development Corporation (CDC). Development practices in acquisition, financing, budget, construction, and the use of the CDC development team were evaluated and specific processes within each area identified as contributing to the overall marginal success of the Siochain project. Case studies were then created outlining the development practices of three Boston-area CDCs: the Jamaica Plain Neighborhood Development Corporation, Urban Edge, and Codman Square Neighborhood Development Corporation. Best practices from these CDCs in each of the areas of concern were profiled, leading to a set of recommendations for the improvement of affordable housing development practices at NOAH.

Large text streams are commonplace; news organisations are constantly producing stories and people are constantly writing social media posts. These streams should be analysed in real-time so useful information can be extracted and acted upon instantly. When natural disasters occur people want to be informed, when companies announce new products financial institutions want to know and when celebrities do things their legions of fans want to feel involved. In all these examples people care about getting information in real-time (low latency). These streams are massively varied, people’s interests are typically classified by the entities they are interested in. Organising a stream by the entity being referred to would help people extract the information useful to them. This is a difficult task: fans of ‘Captain America’ films will not want to be incorrectly told that ‘Chris Evans’ (the main actor) was appointed to host ‘Top Gear’ when it was a different ‘Chris Evans’. People who use local idiosyncrasies such as referring to their home county (‘Cornwall’) as ‘Kernow’ (the Cornish for ‘Cornwall’ that has entered the local lexicon) should not be forced to change their language when finding out information about their home. This thesis addresses a core problem for real-time entity-specific NLP: Streaming cross document coreference resolution (CDC), how to automatically identify all the entities mentioned in a stream in real-time. This thesis address two significant problems for streaming CDC: There is no representative dataset and existing systems consume more resources over time. A new technique to create datasets is introduced and it was applied to social media (Twitter) to create a large (6M mentions) and challenging new CDC dataset that contains a much more variend range of entities than typical newswire streams. Existing systems are not able to keep up with large data streams. This problem is addressed with a streaming CDC system that stores a constant sized set of mentions. New techniques to maintain the sample are introduced significantly out-performing existing ones maintaining 95% of the performance of a non-streaming system while only using 20% of the memory.

Poröse Kohlenstoffe stellen aufgrund ihrer hohen chemischen und physikalischen Belastbarkeit, hohen spezifischen Oberfläche und einstellbaren Porengrößen eine wichtige Materialklasse in der chemischen Industrie dar. Dabei finden Kohlenstoffe sowohl in der Katalyse, in Adsorptions- und Separationsprozessen und in der Abwasserbehandlung, wie auch in elektrochemischen Energiespeichern Anwendung. In all diesen Applikationen ist eine hohe spezifische Oberfläche des Materials, welche durch das Vorhandensein von Mikroporen erreicht wird, essentiell für eine gute Performance. Rein mikroporöse Systeme weisen allerdings aufgrund der sehr langsamen Diffusion in den kleinen Poren große Probleme im Stofftransport auf, welche zu erheblichem Druckverlust, Verlust an Kapazität und Selektivitätsänderungen führen können. In vielen Anwendungen ist daher die Kombination einer hohen spezifischen Oberfläche und eines guten Stofftransports unabdingbar, woraus das besondere Interesse an der Synthese von hierarchisch strukturierten Kohlenstoffen mit einem hohen Mikroporenanteil für eine hohe spezifische Oberfläche und großen Transportporen (Meso- und/oder Makroporen) resultiert. Carbidabgeleitete Kohlenstoffe (carbide-derived carbons - CDC) [1, 2], welche einen vorrangig mikroporösen Charakter besitzen, werden durch selektive Ätzung von Metall- oder Halbmetallatome aus Carbiden dargestellt. Die Einführung von Transportporen erfolgt über verschiedene Templatverfahren, wobei synthetische Template meist sehr aufwendig und teuer synthetisiert werden müssen. Aufgrund des hohen synthetischen, finanziellen und materiellen Aufwandes sind daher nur Synthesen im kleinen Maßstab möglich, welche den breiten Einsatz in verschiedenen Anwendungsfeldern stark limitieren. Dem gegenüber stehen Biotemplate, welche sich durch ihre hohe Verfügbarkeit, ihre Nachhaltigkeit und ihre geringen Kosten, welche bis zu 3000-fach [3-6] geringer als synthetische Template sind, auszeichnen. Zudem besitzt Holz als Bio-Templat eine über Jahrmillionen auf Stofftransport optimierte Struktur, welche Holz zu einem vielversprechenden Templat für die Synthese hierarchischer Kohlenstoffe nicht nur aus ökologischer und ökonomischer, sondern vor allem auch aus wissenschaftlicher Sicht macht. Über die Verknüpfung des CDC-Prozesses mit Holz als Bio-Templat können so hierarchisch strukturierte Kohlenstoffmaterialien mit hohen Oberflächen und einem guten Transportsystem synthetisiert werden, welche die Möglichkeit zum Einsatz in einem breiten Anwendungsbereich bieten. Durch einen einfachen Zwei-Stufen-Prozess bestehend aus Imprägnierung eines flüssigen SiC-Präkursors und anschließender Hochtemperturchlorierung (Abbildung 1) war es möglich, hierarchisch strukturierte Kohlenstoffe unter Erhalt der typischen Holzmikrostruktur mit großen Transportporen und zusätzlichem mikoporösem Charakter, durch den eingebrachten CDC-Kohlenstoff, zu synthetisieren. Die Porengrößen und -verteilung sind dabei stark von der Holzart, der Chlorierungstemperatur und den Parametern des Imprägnierprozesses abhängig. Es konnte ein linearer Zusammenhang zwischen eingebrachter Siliziumcarbidmenge und der spezifischen Oberfläche des resultierenden Kohlenstoffmaterials ermittelt werden, welcher ein gezieltes Design der Holz-CDC-Materialien in Bezug auf Oberfläche und Porenvolumen/-größe ermöglicht. Neben der makroporösen Zellstruktur des Holzes konnte zudem die makroskopische Form während des gesamten Prozesses vollständig erhalten werden, welches die gezielte Synthese von Formkörpern, wie Monolithen, ermöglicht. Die Synthese von Holz-CDC-Materialien bietet daher einen großen ökonomischen Vorteil gegenüber herkömmlichen Kohlenstoffsynthesen, in denen meist pulverförmige Produkte entstehen, welche dann zur Nutzung in verschiedenen Anwendungen durch Presswerkzeuge oder den Zusatz von Bindermaterialien in Formkörper gebracht werden müssen. Zur weiteren Steigerung der Oberfläche und des Porenvolumens wurden Voraktivierungen am Holztemplat durchgeführt. Hierbei wurden sowohl physikalische Aktivierungsmethoden mit Wasserdampf oder Kohlenstoffdioxid, wie auch chemische Aktivierungsmethoden mit Säuren und Basen untersucht. Über den Aktivierungsprozess wurde eine zusätzliche Porosität in die Holzmatrix eingebracht, wodurch nach anschließendem Imprägnierprozess und Hoch-temperaturhalogenierung Holz-CDC-Materialien mit trimodalem Porensystem bestehend aus Mikro-, Meso- und Makroporen mit Oberflächen von bis zu 1800 m^2/g und Porenvolumina bis zu 1,0 cm^3/g erzielt werden konnten. Aufgrund ihrer guten Leitfähigkeit, hohen Oberfläche und porösen Eigenschaften stellen Kohlenstoffe interessante Kathodenmaterialien für die Lithiumschwefelbatterie dar. Trotz intensiver Forschungen in den letzten 10 Jahren konnten die Herausforderungen einer hohen Zyklenstabilität, Ratenstabilität und Zellkapazität, sowie geringer Elektrolytmengen bis heute nicht zufriedenstellend gelöst werden. Hierarchisch strukturierte Kohlenstoffmaterialien, welche „Reaktions- und Transportporen“ besitzen, stellten sich als vorteilhaftes Kathodenmaterial heraus. Die longitudinal ausgerichteten Makroporen (Transportporen) der Holz-CDCs ermöglichen einen schnellen Ionentransport, welcher auch bei hohen Lade- und Entladeraten stabile Kapazitäten ermöglicht. Dem gegenüber setzen die Mikroporen (Reakionsporen) die Löslichkeit der Polysulfide herab, welches eine gute Ratenstabilität über 100 Zyklen zur Folge hat. Es konnten mit den synthetisierten Holz-CDC-Materialien stabile Kapazitäten über 580 mAh/gSchwefel mit hohen Stromdichten von 20 mA/cm^2 (2C) und sehr geringen Elektrolytmengen von nur 6,8 µl/mgSchwefel erzielt werden. Diese Daten zeigen eine deutliche Verbesserung zu den in der Literatur bisher veröffentlichten Werten [3,7]. Neben dem Einbringen einer zusätzlichen Porosität werden durch den chemischen Aktivierungsprozess Oberflächenfunktionalitäten an der Kohle gebildet. Diese Oberflächen-funktionalitäten können vor allem in der Adsorption von polaren Verbindungen essentiell für eine hohe Adsorptionskapazität sein. Quecksilber stellt ein giftiges Element dar, welches anthropogen durch die Kohleindustrie jährlich mit ca. 4000 t freigesetzt wird. Die Entfernung von Quecksilber aus Industrieabgasen erfolgt über dessen Lösung in Wasser und anschließende adsorptive Prozesse, wobei Kohle als Adsorbens Einsatz findet. Untersuchungen der Holz-CDC-Materialien zeigten hohe Quecksilber-Adsorptionskapazitäten von 242 mgHg/gKohle. Gegenüber herkömmlichen kommerziellen Aktivkohlen [8] mit 12 mg/g und neuartigen Aktivkohlen auf Bio-Basis [8] mit 150 mg/g, zeigen die untersuchten Holz-CDC-Materialien 1,5- bis 200-fach höhere Aufnahmekapazitäten. Bei diesen ersten proof-of-principle-Untersuchungen konnte das hohe Potential holzbasierter CDC-Materialien für die Anwendung in Adsorptionsprozessen gezeigt werden, welches eine deutliche Steigerung der Kapazität durch weitere zukünftige Optimierungen des Materials verspricht. Holz kann ebenfalls Anwendung zur Synthese hochporöser Kohlenstoffstäbchen finden. Hierbei wird die Holzstruktur vollständig mit Siliziumcarbid gefüllt. Nach der anschließenden Entfernung des Holztemplates über Calcination bleibt die Negativstruktur des Holzgerüstes als stäbchenförmige Strukturen erhalten. Durch nachfolgende Reinigung und Hochtemperatur-chlorierung können die SiC-Stäbchen in rein mikroporöse CDC-Stäbchen umgewandelt werden, welche sehr hohe spezifische Oberflächen von bis zu 3680 m^2/g und Porenvolumina von bis zu 1,6 cm^3/g besitzen. Aufgrund ihres unpolaren Charakters und der hohen spezifischen Oberfläche sind diese Strukturen besonders für die Adsorption von aromatischen, gering bzw. nicht polaren Verbindungen geeignet. Das adsorptive Verhalten der Stäbchenstrukturen wurde bei der Adsorption von Methylenblau, einer in der Literatur häufig verwendeten Beispielsubstanz für die Adsorption voluminöser aromatischer Verbindungen, und von Diclofenac untersucht. Diclofenac ist ein Schmerzmittel, welches vor allem bei Rheuma eingesetzt wird und mit ca. 63 t/Jahr in Deutschlands Wassersysteme eingetragen wird. Die schlechte Abbaubarkeit und die unzureichende Entfernung von Diclofenac über herkömmliche Abwasseraufbereitungsanlagen haben in den letzten Jahren zu einer deutlichen Anreicherung des Medikamentes in der Umwelt geführt. Die Entfernung von Diclofenac hat, neben der Entfernung anderer aromatischer Medikamente, wie Ibuprofen und Carbamazepin, in den letzten 10 Jahren daher deutlich an Bedeutung gewonnen. CDC-Stäbchen zeigen im Vergleich zu herkömmlichen Kohlenstoffen, wie Printex oder Hydraffin P800, fast doppelt so hohe Aufnahmekapazitäten für Methylenblau unter ähnlich schnell ablaufender Adsorptionkinetik. Auch Diclofenac kann an den CDC-Stäbchen mit 580 mg/g deutlich besser adsorbieren als an Hydraffin P800, welche eine Kapazität von 490 mg/g zeigt. Bedenkt man, dass es sich bei Hydraffin P800 (Firma: Donau Carbon) um eine für die Adsorption von organischen Wasserschadstoffen optimierte Aktivkohle handelt, wird das hohe Potential der unoptimierten CDC-Stäbchen deutlich. Abstract:
L’augmentation de la moyenne de vie a modifié le spectre des maladies cardiaques vers l'insuffisance cardiaque, un désordre progressif incurable, marqué par une perte massive de cardiomyocytes. En raison de la rareté des donneurs, la transplantation cardiaque, devient de plus en plus difficile. En 2003, la découverte des cellules souches cardiaques humaines (hCSC) pouvant supporter la réparation du myocarde après un infarctus a soulevé l’espoir d’approches thérapeutiques plus prometteuses. La transplantation des cellules autologues (issues du patient) a montré une efficacité limitée en raison de la mauvaise qualité des cellules après infarctus. De ce fait, l’utilisation des CPC allogéniques provenant de donneurs sains semble aujourd’hui le choix le plus réaliste et représente un potentiel produit thérapeutique abordable et disponible pour tous. Cependant les cellules allogéniques soulèvent des problèmes immunologiques importants qu’il faut résoudre avant leur utilisation clinique. Concernant la réponse immune innée, nos résultats indiquent que les hCPC allogéniques ont une susceptibilité modeste à la lyse NK et pourraient même être protégées contre cette lyse en contexte inflammatoire. Contrairement à la réponse immunitaire innée favorable contre les hCPC allogéniques, la présence des alloanticorps anti-HLA peut être néfaste. Nos résultats ont montré que les DSA-HLA I spécifiques de l’haplotype des hCPC peuvent induire leur lyse par CDC et ADCC et cette lyse est fortement augmentée par les conditions inflammatoires. L’ensemble de ces résultats indique que les hCPC allogéniques sont des cellules à faible risque immunologique en ce qui concerne la réponse immune innée, par contre une élimination rapide par CDC ou tardive par ADCC des hCPC pourrait se produire chez les patients ayant des DSA-HLA I spécifiques de l’allèle(s) exprimé(s) par les hCPC incitant ainsi le dépistage des DSA-HLA avant l’infusion thérapeutique des hCPC
The increase in average life changed the spectrum of heart disease to heart failure, a progressive incurable disorder, characterized by a massive loss of cardiomyocytes. Due to the scarcity of donors, cardiac transplantation, only effective treatment known to date is becoming increasingly difficult. In 2003, the discovery of human cardiac stem cells (HCS/PC) can support the repair of the myocardium after myocardial raised hopes of promising therapeutic approaches. Transplantation of autologous (from the patient) showed limited efficiency because of the poor quality of the cells after infarction. Therefore, the use of stem / progenitor heart (CPC) from allogeneic healthy donors seems today the most realistic choiceand represents a potential therapeutic product available to all. However allogeneic cells raise significant immunological problems that must be resolved before clinical use. Regarding the innate immune response, our results indicate that allogeneic HCPC have a modest susceptibility to NK lysis and could even be protected against this lysis in the inflammatory context. In addition, allogeneic HCPC seem able to inhibit the activity of NK cells. Unlike the favorable innate immune response against the allogeneic HCPC, the presence of anti-HLA alloantibodies can be harmful. Our results showed that the DSA-specific HLA class I haplotype of the HCPC can induce their lysis by CDC and ADCC and this lysis is greatly increased by inflammatory conditions. However, the DSA-specific HLA II do not induce HCPC lysis. All these results indicate that allogeneic HCPC are cells with low immunological risk regarding the innate immune response. On the contrary, rapid elimination by CDC or ADCC of HCPC may occurs in patients with DSA- HLA class I specific allele (s) expressed by the HCPC thus prompting the screening of HLA-DSA prior to therapeutic infusion of HCPC

O trabalho busca dissertar sobre as técnicas de data mining mais difundidas: regressão logística, árvore de decisão e rede neural, além de avaliar se tais técnicas oferecem ganhos financeiros para instituições privadas que contam com processos ativos de conquista de clientes. Uma empresa do setor financeiro será utilizada como objeto de estudo, especificamente nos seus processos de aquisição de novos clientes para adesão do Crédito Direto ao Consumidor (CDC). Serão mostrados os resultados da aplicação nas três técnicas mencionadas, para que seja possível verificar se o emprego de modelos estatísticos discriminam os clientes potenciais mais propensos dos menos propensos à adesão do CDC e, então, verificar se tal ação impulsiona na obtenção de ganhos financeiros. Esses ganhos poderão vir mediante redução dos custos de marketing abordando-se somente os clientes com maiores probabilidades de responderem positivamente à campanha. O trabalho apresentará o funcionamento de cada técnica teoricamente, e conforme os resultados indicam, data mining é uma grande oportunidade para ganhos financeiros em uma empresa.
The paper intends to discourse about most widespread data mining techniques: logistic regression, decision tree and neural network, and assess whether these techniques provide financial gains for private institutions that have active processes for business development. A company of the financial sector is used as object of study, specifically in the processes of acquiring new customers for adhesion to consumer credit (in Brazil CDC). This research will show the results of the three above mentioned techniques, to check whether the statistical models point out relevant differences between prospects´ intentions to adhere to consumer credit. In the meantime, the techniques are checked whether they leverage financial gain. These gains are expected to came from better focused and directed marketing efforts. The paper presents the operation of each technique theoretically, and as the results indicate, data mining is a great opportunity for a company boost profits.

Dans le cadre de la recherche de nouvelles cibles pour le traitement du cancer, les phosphatases Cdc25 sont des candidats intéressants dont l'inhibition devrait permettre de ralentir la croissance tumorale et éventuellement d'améliorer les traitements actuellement en usage. Les objectifs de ce projet de thèse sont de concevoir et synthétiser de nouveaux composés capables d'inhiber les CDC25, et de déterminer l'efficacité des meilleurs composés dans les lignées cellulaires du cancer du sein. L'évaluation du potentiel inhibiteur des composés est réalisée in vitro par une méthode fluorimétrique très sensibilité (substrat 3-OMFP). Les effets des composés sont évalués dans les lignées cellulaires MCF-7 et MDA-MB-231 : La viabilité des cellules est évaluée par la méthode colorimétrique du MTT, la cytotoxicité est évaluée par coloration au bleu trypan et par observation microscopique avec le système de vidéomicroscopie Incucyte. La mort cellulaire est caractérisée par la détection de marqueurs apoptotiques (caspases) et de marqueurs des dommages de l'ADN (H.2AX Histone PARP) par western blot. L'analyses des mécanismes liés à la mort cellulaire sont explorées par cytométrie en flux via la détection d'espèces réactives de l'oxygène (ERO) avec les sondes H2DCFDA et Redox Sensor Red. L'inhibition de CDC25 dans les cellules est évaluée indirectement par détection des formes phosphorylées dse CDK en Western Blot. L'évaluation in vitro de 93 molécules synthétisées nous a permis d'identifier de nouveaux composés actifs. Ils appartiennent à diverses familles chimiques comprenant des stéroïdes, thiophènes, coumarines, imidazoles ainsi que des dérivés de quinone. Les dérivés coumariniques ont montré une intéressante inhibition de CDC25, non décrite jusqu'à présent. Une nouvelle structure coumarine-soufre-quinone, nommée SV37, a été conçue pour optimiser le potentiel d'inhibition. Ce composé présente un fort potentiel inhibiteur des CDC25 in vitro (CI50 < 5uM pour CDC25 A et C). L'effet de SV37 sur la croissance cellulaire a été évalué sur les lignées cellulaires MCF-7, MDA-MB-231, hTERT-HME1 et HepG2 sur lesquelles une inhibition de la croissance cellulaire est observée (CI50 de 9 à 18 µM). L'analyse de la viabilité des cellules traitées à la CI50 indique l'absence de mort cellulaire pour la lignée MCF7, tandis que pour la lignée MDA-MB-231 la diminution de la croissance cellulaire est liée à une augmentation de la mort cellulaire. Afin d'étudier les mécanismes liés à la mort cellulaire, nous nous sommes concentrés sur l'étude du modèle triple négatif MDA-MB- 231. Les modifications morphologiques de ces cellules sont caractérisées par l'apparition d'altérations cellulaires compatibles avec la mort cellulaire. Le clivage des caspase-3 et 7 a été observé dès 16h de traitement, ce qui suggère l'induction d'une mort apoptotique. Par ailleurs, des ERO ont été détectées 15 min après le début du traitement, cette émission d'ERO a pu être totalement bloquée par un prétraitement avec la N-acétylcystéine (NAC). La détection de marqueurs des dommages de l'ADN, entre 16 et 28 heures après début du traitement, corrobore l'activation des caspases et les observations réalisées en vidéo-microscopie. Après traitement à CI50, une accumulation des pCDK dans les cellules a été observée après 4 et 8 heures, suggérant une inhibition de l'activité CDC25. De plus, les cellules prétraitées avec la NAC n'ont montré aucune accumulation de pCDK après traitement par SV37. Ces résultats suggèrent un lien direct entre la production de ROS induit par le composé SV37 et l'inhibition des CDC25. Ce projet a permis de définir les coumarines en tant que nouvelle classe de composés inhibitrice des phosphatases CDC25. Ces travaux ont de plus permis l'identification d'un composé coumarine-soufre-quinone SV37 qui constitue une structure de base pour le développement d'inhibiteurs de plus en plus efficaces,
Within the context of research for new targets for cancer therapy, Cdc25 phosphatases are interesting candidates, the inhibition of which being able to slow down tumor growth and eventually improve the cancer treatments currently in use. The objectives of this PhD project are to design and synthesize new compounds able to inhibit CDC25 and to determine efficiency of identified compounds in breast cancer cell lines. In vitro evaluation of inhibitory potential of compound is realized through a high sensitivity fluorometric method (3-OMFP substrate). Cellular effects were evaluated in MCF-7 and MDA-MB-231 cell lines. Effects on cell viability are assessed through MTT assays, and cytotoxicity is evaluated through trypan blue assays and microscopic observations with Incucyte videomicroscopy system. Cell death was characterized by detection of apoptotic markers (caspases) and DNA damages markers (PARP Histone H.2AX) by Western Blotting. The analyses of mechanisms underlying cell death were explored through cytometric detection of reactive oxygen species (ROS) with H2DCFDA and Redox Sensor Red probes. Inhibition of CDC25 in cells was indirectly evaluated through detection of phosphorylated forms of CDK by Western Blotting. In vitro evaluation of 93 synthesized compounds allowed us to find new active compound in various chemical families including steroid, thiophene, coumarinic, imidazole and quinone derivatives. The coumarinic derivatives showed potent CDC25 inhibition. A new coumarin-sulfurquinone combined structure, named SV37, was designed to optimize efficiency of inhibition. In vitro tests on this compound, showed a strong CDC25 inhibitory potential (IC50 under 5µM for CDC25 A and C). Effect of SV37 on cell growth was evaluated on various cell lines (MCF-7, MDA-MB-231, hTERT-HME1 and HepG2). Results indicate inhibition of cell growth (IC50 values from 9 to 18 µM). Analysis of cell viability indicates no remarkable cell death in MCF7 at IC50 value whereas in MDA-MB-231 the cell growth decrease was characterized by an increase of cell death. For deeper investigations on the cell death and on the underlying mechanisms, we focused the study on the triple negative model MDA-MB-231. The morphological changes of MDA-MB-231 cells during the treatment were characterized by the appearance of cellular alterations compatible with a cellular demise and culminating with a disruption of cells after 20h. Caspase-3 and 7 cleavages were observed 16h after beginning of the treatment, suggesting an apoptotic cell death. A ROS induction was observed 15 min after the beginning of the treatment and was totally prevented by Nacetylcysteine (NAC) pretreatment. DNA damage markers were detected between 16 and 28 hours after beginning of treatment, a timing falling with caspase activation and with the appearance of cell demise observed by video microscopy. Accumulation of pCDK in cells was observed after 4 and 8 hr of treatment by SV37 at IC50 suggesting an inhibition of CDC25 activity, and cells pretreated with NAC showed no accumulation of pCDK after SV37 treatment. This strongly suggests a direct link between ROS generation by the compound SV37 and the accumulation of pCDK. This project increased knowledge on inhibitors of CDC25 phosphatases and allowed the identification of coumarine compound as new CDC25 inhibitors. This work will enable the development of ever more efficient inhibitors, leading to efficient inhibition of CDC25 and inhibition of tumor development

Différents matériaux carbonés nanostructurés ont été synthétisés et mis en oeuvre comme matériaux supercapacitifs à double couche électrochimique (EDLC) ou comme substrats de matériaux pseudocapacitifs avec pour objectif d'augmenter leur densité de puissance. Nous avons ainsi développé une méthode de synthèse simple et originale de carbures de silicium (SiC) qui procède par une réduction topotactique d'un composite silice/carbon par le magnésium. Du fait de la température de synthèse inférieure à 800°C, SiC résultant conserve la morphologie et/ou la structure poreuse du précurseur composite. Par cette approche, nous pouvons moduler la structure poreuse ordonnée de SiC à façon, développer des porosités hiérarchiques méso/macro, préparer des feuillets ou des fibres de SiC. Les différentes formes de SiC ont été converties par chloration en autant de carbones, opération introduisant une microporosité supplémentaire. En électrolyte organique, ces carbones à porosité hierarchique combinent à la fois des capacités importantes issues de la microporosité mais également des performances inégalées en terme de puissance du fait de la méso ou macro-porosité associée. Dans une approche différente, des feuillet de graphène ont été décorés par voie sol-gel non-hydrolytique (micro-onde en milieu alcool benzilique) par des nanoparticules de FeOx. Le composite FeOx/graphene résultant combine simultanément les comportements EDLC et pseudocapacitif du graphène et de FeOx. Du fait de sa structure particulière, le composite FeOx/graphene conserve les performances en puissance du graphène auxquelles s'ajoutent celles d'énergie de FeOx. Nous avons également décoré des nanofibres de carbone avec des carbones mésoporeux. Après dépôt de MnO2 birnessite, les composites gagnent à la fois en capacité et en puissance en particulier avec des carbones présentant des pores supérieurs à 10nm
Various nanostructured carbon materials were synthesized and further served as active materials of electrical double layer capacitor or substrates of pseudocapacitive materials in order to improve power capability of corresponding supercapacitor. On the one hand, a simple synthesis of porous silicon carbides (SiCs) was achieved by performing a topotactic thermal reduction by magnesium (Mg) of a silica/ carbon composite. Thanks to the low synthetic temperature (below 800 ºC), the SiCs well preserved the pristine skeletons of their silica/carbon precursors. Successively, the SiCs with diverse porous structures from their silica/carbon precursor emerged, e.g. ordered tunable mesoporous SiCs, 3D-hierarchical meso and macroporous SiC, SiC nanosheet and SiC nanofiber. Furthermore, the porous SiCs derived from magnesio-thermal reduction were reduced to hierarchical carbons with newborn narrow distributed microporosity by chlorination. In an organic electrolyte, the hierarchical carbon combines the high specific capacitance from narrow distributed microporosity and the outstanding rate capability from ordered-arranged meso or macroporosity that make it promising for high power and energy density capacitor. On the other hand, a “benzyl alcohol route” has been used to decorate RGO nanosheets with FeOx nanoparticles. The resulting FeOx/ RGO composite, due to their hybrid nanostructure, combine both EDLC capacitive and pseudocapacitive bahaviors of RGO and FeOx, respectively. Thanks to the laminated RGO and nano FeOx particles film, the resulting composite gains the same power capability as RGO and a higher energy density than raw FeOx. Furthermore, mesoporous carbon was introduced to adorn the CNF surface through self-assemble of resol, carbon nanofiber(CNF) and Pluronic@127. After further coating with birnessite-MnO2, the composite electrode gains extra capacitance and power improvement in presence of superficially coating mesoporous carbon with pore size larger than 10nm

Abstract:
This master thesis deals with polyhydroxyalkanoates (PHA) and their role in bacterial biofilms. In the theoretical part the polyhydroxyalkanoates, bacterial biofilm and the relationship between them were reviewed. The experimental part focused on differences in PHA production by planktonic and biofilm cells. In order to study selected topic, bacterial strains of Burkholderia cepacia and Burkholderia sacchari were cultivated using a CDC biofilm reactor. The attention was paid to quantity and especially to the form in which PHA occurs in planktonic and biofilm cells. Results of Raman spectroscopy have shown that PHA exists exclusively in native amorphous form in planktonic bacterial cells. On the other hand, in biofilm PHA occurs also in a partially crystalline form. In addition, the resistance of planktonic and biofilm cells against various stress factors and the effect of osmotic stress on PHA production was tested too. According to the results of the experiment, when the bacteria were exposed to different stress factors (high temperature, low temperature, presence of detergent and so forth) biofilm cells showed a higher stress resistance than planktonic cells. Apart from slowing cell growth and reproduction, increased osmotic pressure in the culture medium also caused decrease of PHA production. In addition, planktonic cells responded to external stimuli more sensitively than biofilm ones.

This study’s aim was to explore face, content, concurrent validity and psychometric properties of the Swedish Child Dissociative Checklist (CDC) as well as and to provide guidelines towards validation. The CDC is used in Sweden when dissociation is highly probable, and validation could motivate wider use. Qualitative data was gathered from psychologists-in-training (N = 4) clinical psychologists (N = 5) and parents (N = 23) through a focus group, interviews and questionnaires, respectively. Quantitative data using the CDC and items from the Child Behaviour Checklist (CBCL) was obtained from parents. Results suggest good face, content and convergent validity. Factor analysis provides suggestions for future investigations into quantitative validation. Guidelines when validating is revising certain items, investigating the possibility of developing two versions of the CDC, utilizing an adequate sample size as well as assessment with a clinical group.
Studiens syfte var att undersöka face, innehålls- och konvergent validitet samt psykometriska egenskaper hos den svenska versionen av Child Dissociative Checklist (CDC), samt utforma riktlinjer för validering. I Sverige har CDC använts vid hög närvaro av dissociativa symptom, och validering kan motivera bredare användning. Kvalitativ data samlades in från psykologstudenter (N = 4), kliniska psykologer (N = 5) och föräldrar (N = 23) genom fokusgrupp, intervjuer respektive frågeformulär. Kvantitativa data erhölls från föräldrar med CDC och items från Child Behaviour Checklist (CBCL). Resultaten tyder på god face, innehålls- och konvergent validitet. Faktoranalys bidrar med förslag på framtida tillvägagångssätt för kvantitativ validering. Riktlinjer vid validering är revidering av vissa items, utforskning av möjligheten att utveckla två versioner av CDC, tillgång till ett stort sample samt jämförelse med en klinisk grupp.

From something seen as clear-cut and uncontroversial this doctoral thesis intends to scrutinize in-depth the viability of the Judge s ex officio, ie, without the need of previous provocation by any of the litigants (ex officio), of the unfair terms in consumer-related contracts, and as corollary of such a possibility, of what was intended by the legislator of Consumer Protection Law when qualifying such a rule as a public one. The controversy was underestimated by this candidate. This underestimation has never been caused by disrespect or contemptuous treatment of the legal controversy, on the contrary. The challenges and difficulties arisen as something thought not possess such profound implications, not only within the material law, but also in procedural law (feared), only made the study and the search for answers more enticing. The Judge s expression in itself (ex officio) about the material law turned itself to certain immanentism of the material law with procedural law. So, as researcher of the Consumer Protection Law, it arises impacts or not on the Judge s expression of an unfair terms or any other contra legem interpretation it is not as simple as it might seem. Modulations, forces and various decisional effects had to be addressed in the search for a better answer. Variation which is amplified both on individual as well as on class actions. In parallel with such a confrontation (discovery), also came the complicating element of demystifying what would be the definition of public order. In Brazilian law, the rule is to perceive public order as something imperative, law binding, unattainable. Those features do not cover encompass all situations of the Brazilian Consumer Law (CDC) and forced the researcher to seek more details in foreign sources. The search for definitions, characteristics and effects in foreign law systems widened up even more what the research sought to analyze. Nevertheless the research s scope, it was realized the confrontation of the features of public order would of use to Brazilian law in itself as the focus on different foreign systems would aggrandize and justify the option for the CDC. Finally, as during this investigation the Superior Court of Justice (STJ) pronouncement number nº. 381, whose content goes against this thesis, came to light, its analysis and confrontation turned out to be inevitable
De algo tido como claro e pacífico a tese de doutorado que se propõe busca estudar a fundo a possibilidade da decretação de ofício, ou seja, sem provocação das partes, das cláusulas abusivas nos contratos relacionais de consumo e, como corolário a tal possibilidade, o que pretendeu o legislador do Código de Proteção e Defesa do Consumidor ao qualificar tal norma como de ordem pública. É confessado por este candidato que o tema, a controvérsia, foi subestimado. Tal subestimação jamais foi causada por um possível desrespeito ou um tratamento desdenhoso da controvérsia estritamente jurídica. Muito pelo contrário. Os desafios e as dificuldades que surgiram com algo que se imaginava não possuir um grau tão profundo de implicações não só dentro do direito material, mas principalmente, e mais desafiador sob o direito instrumental (temido) só tornaram o estudo e a busca por resposta mais apaixonantes. Da decretação em si sob o direito material retornou-se a certo imanentismo do direito material com o direito processual. Logo, como materialista do direito consumerista surgem impactos que a mera decretação de ofício ou não de uma cláusula ou qualquer outro ato jurídico contra legem não é simples como aparenta. Modulações, forças e diversos efeitos sentenciais tiveram que ser tratados na busca de uma melhor resposta. Variante que se amplifica tanto nas ações individuais, quanto nas coletivas. Paralelo a tal enfrentamento e a reboque do mesmo veio o complicador de desmistificar o que define a ordem pública. No direito pátrio as doutrinas só preveem a ordem pública como algo imperativo, norma cogente, indisponível. Característica que não engloba todas as situações do CDC e que obrigou o pesquisador a buscar fontes estrangeiras. As buscas no direito estrangeiro por definições, características e efeitos ampliaram mais ainda o que sempre se buscou delimitar na tese. Não obstante, ao olhar sobre a delimitação da tese, cientificamente, percebeu-se que o enfrentamento das características da ordem pública seria de grande valia ao direito pátrio e que o estudo sobre sistemas estrangeiros diferentes só engradeceria e justificaria ainda mais a opção do CDC. Por fim, como no decorrer do estudo surgiu à edição da Súmula 381 do STJ, cujo teor vai de encontro a presente tese, inevitável tornou-se seu estudo e enfrentamento

Abstract:
In der vorliegenden Arbeit wurden mesoporöse Materialien erfolgreich mit magnetischen bzw. katalytisch aktiven Nanopartikeln funktionalisiert, wobei zwei unterschiedliche Synthesestrategien verfolgt wurden. Zum einen erfolgte eine direkte Integration der katalytisch aktiven CeO2-Nanopartikel in poröse, thermisch stabile SiC-Matrizes mithilfe der inversen Mikroemulsionsmethode. Die Größe dieses wässrigen, cersalzhaltigen Nanoreaktors konnte über den RW-Wert (molares Wasser-Tensid-Verhältnis) im Bereich von 2,0-9,9 nm mit einer engen Größenverteilung variiert werden. Für katalytische Untersuchungen wurden die Cerhydroxidpartikel aus dem Mikroemulsionssystem ausgefällt und bei 100-600 °C calciniert. Dabei konnte eine größenabhängige Aktivität der Nanopartikel in der Rußverbrennung nachgewiesen werden, wobei eine Herabsetzung der Rußverbrennungstemperatur um bis zu 239 K nachgewiesen werden konnte. Weiterhin konnten polymere CeO2-SiC Vorläuferverbindungen durch Zugabe einer flüssigen, präkeramischen Vorläuferverbindung (Polycarbosilan) zum Mikroemulsionssystem hergestellt werden, wobei flüssig prozessierbare, transparente Lösungen resultierten. Anschließend erfolgte nach Entfernung der flüchtigen Bestandteile die Pyrolyse zur Bildung der CeO2-SiC-Keramiken. In Abhängigkeit von den Pyrolysebedingungen konnten kristalline SiC-Strukturen mit spezifischen Oberflächen von bis zu 240 m2/g nachgewiesen werden. In weiteren Untersuchungen konnte die Modularität dieses neuartigen Synthesekonzeptes demonstriert werden, indem Platin als zusätzliche Aktivkomponente in das bestehende Mikroemulsionssystem integriert wurde. Im Gegensatz zu den platinfreien Systemen erfolgte eine Vernetzungsreaktion infolge der Pt-katalysierten Vernetzungsreaktion des allylgruppenhaltigen Polycarbosilans, mit spezifischen Oberflächen von bis zu 896 m2/g. Anhand von TEM-Untersuchungen konnte eine hohe Dispersion der CeO2-Aktivkomponente mit Partikelgrößen von 2-3 nm gezeigt werden. Durch die Zugabe von Divinylbenzol als Kreuzvernetzungskomponente konnte neben einer weiteren Erhöhung der Oberfläche auf 992 m2/g auch die Hydrophobizität des Polymerkomposits signifikant erhöht werden. In einer zweiten Synthesestrategie wurden intermetallische MPt-Systeme (M=Fe, Co, Ni) durch Infiltration geeigneter Vorläuferverbindungen und anschließender Thermolyse in geordneten mesoporösen SiO2- bzw. Kohlenstoffmaterialien synthetisiert. Die Phasenbildung in Abhängigkeit von den Thermolysebedingungen wurde mithilfe der Röntgenpulverdiffraktometrie untersucht. Dabei wurden nach der Reduktion bei 400 °C ungeordnete fcc-MPt-Legierungen mit superparamagnetischen Eigenschaften erhalten. Dagegen resultierte für FePt-Systeme nach der Reduktion bei 750 °C bis 800 °C eine deutliche Zunahme der Raumtemperaturkoerzitivitäten auf bis zu 28,35 kOe (FePt@CMK-3) bzw. 15,60 kOe (FePt@SBA-15) infolge der Bildung der intermetallischen fct-FePt Verbindung. Weiterhin wurden die strukturellen sowie magnetischen Eigenschaften der FePt-Nanopartikel in Abhängigkeit vom Massenanteil sowie der Porengröße bzw. -geometrie der porösen Silicatemplate untersucht. Dabei konnte eine starke Abhängigkeit der Raumtemperaturkoerzitivität von der Porenstruktur sowie den jeweiligen Reduktionsbedingungen nachgewiesen werden. Ein weiterer Aspekt dieser Arbeit war die Synthese hochporöser CDC-Kohlenstoffmaterialien (CDC: carbide derived carbon) durch die Chlorierung nichtoxidischer SiC-Keramiken. Hierbei wurde das Silicium der mesoskopisch geordneten SiC-Strukturen durch Umsetzung mit Chlor bei unterschiedlichen Thermolysebedingungen extrahiert. Die resultierenden CDC-Materialien wiesen neben sehr hohen spezifischen Oberflächen von bis zu 2865 m2/g bzw. Porenvolumina von 2,21 cm3/g auch eine von der SiC-Struktur sowie den Chlorierungsbedingungen abhängige mesoskopische Ordnung auf. Die mesoporösen CDC-Materialien eignen sich als Sorbentien mit hohen Butan- bzw. Wasserstoffspeicherkapazitäten von 0,692 gButan/gCDC (25 °C: 80 Vol% Butan) bzw. 2,58 gew% (77 K: 1 bar). Daneben resultieren überaus hohe gravimetrische Methanspeicherkapazitäten von 0,191 g Methan/gCDC im Hochdruckbereich (25 °C, 100 bar), die deutlich größer sind als die bekannter Metallorganischer Gerüstverbindungen
Ordered mesoporous materials were successfully functionalized with magnetic and catalytic active nanoparticles. Two different synthesis strategies were employed. Cerium oxide nanoparticle containing silicon carbide composites were synthesized by direct incorporation of catalytic active CeO2 nanoparticles in preceramic polycarbosilane using inverse microemulsion technique and subsequent pyrolysis. Resulting ceramic composites offer specific surface up to 240 m2/g and a narrow pore sizes in the range of 4-6 nm. Additionally porous Pt containing CeO2-SiC composites were prepared demonstrating the versitibilty of this new synthesis strategy. Catalytic activity of ceria nanoparticles were shown in soot combustion reaction. In a second approach intermetallic MPt nanoparticles (M= fe, Co, Ni) were synthesized inside the pores of ordered mesoporous silica and carbon materials. Crystalline structure and particles size were controlled by the porous template and the annealing conditions. Very high room temperature coercivities up to 28.4 koe were obtained for intermetallic FePt nanoparticles in mesoporus carbon matrices. Catalytic activity of FePt silica composites were demonstrated in selective acetylene hydration. Furthermore high porous, mesostructured carbon materials (CDC: carbide derived carbon) were prepared by chlorination of ordered mesoporous silica resulting extraordinary high specific surface areas up 2865 m2/g, high pore volunina up to 2.21 cm3/g and mesoscopic ordering. These new carbon structures are appropriate as high performance energy storage materials

Abstract:
The publication of the Curriculum Development Centre's discussion paper 'Core Curriculum for Australian Schools' in June 1980 stimulated discussion of the concept of core curriculum in Australia. The driving force came from the Foundation Director of the CDC, Dr Malcolm Skilbeck. This study discusses the themes and directions to which Skilbeck was committed through a study of his work prior to his return to Australia in 1975 and his subsequent writings. The study considers Skilbeck's work against general thinking on educational matters in Australia and overseas. The initial discussion centres on Skilbeck's work in the United Kingdom prior to 1975. This concludes that his views were moulded by his own research on the American progressive educator John Dewey and that Dewey's ideals of a democratic society moulded and sustained by a democratic core curriculum have been dominant in all Skilbeck's subsequent thinking. The study reviews the establishment, working and conclusions of the CDC Core Curriculum and Values Education Working Party. In two subsequent chapters, the study looks at Skilbeck's approach to cultural mapping and school-based curriculum development as the two fundamental Planks of his approach to the development and implementation of a core curriculum for Australian schools. The study shows that Skilbeck's concept of cultural mapping is helpful but does not succeed in providing an effective basis for the articulation of national guidelines. In consequence, the CDC did not succeed in providing a framework sufficient to hold together the infinite range of possibilities opened UP by school-based action. The study considers the limited published reactions to the CDC Paper. It notes that the termination of the CDC by the Committee for Review of Commonwealth Functions in early 1931 prevented the fuller dissemination and debate of the topic during 19S1 and subsequently. The study notes that responses were disaapointingly few and in many cases failed to address the central questions raised by the CDC paper, in particular the idea of national curriculum guidelines and their application through school-based curriculum development. The major responses came in the State of Victoria where local circumstances encouraged discussion of the issues raised by the CDC. The study concludes that the CDC discussion paper was a valuable stimulus to discussion of curricular foundations at the time it was released but represented a point of view that was not fully understood or appreciated at the time. It laid the foundation for the renaissance of the general concept as 'democratic curriculum' in 1986 and provides important indications of the potential for the development of the Participation and Equity Program.

A presente dissertação de mestrado teve como objetivo principal verificar a aplicabilidade da metodologia Regressão Logística Geograficamente Ponderada (GWLR) para a construção de modelos de credit scoring. As fórmulas do melhor conjunto de modelos locais estimados via GWLR foram comparadas entre si, em termos de valor dos coeficientes e significância das variáveis, e frente ao modelo global estimado via Regressão Logística. Foram utilizados dados reais referentes às operações de Crédito Direto ao Consumidor (CDC) de uma instituição financeira pública nacional concedidas a clientes domiciliados no Distrito Federal (DF). Os resultados encontrados demonstraram a viabilidade da utilização da técnica GWLR para desenvolver modelos de credit scoring. Os modelos estimados para cada região do DF se mostraram distintos em suas variáveis e coeficientes (parâmetros) e três dos cinco indicadores do modelo via GWLR se mostraram superiores aos do modelo via Regressão Logística. ________________________________________________________________________________________________ ABSTRACT
This master thesis aimed to verify the applicability of the methodology Geographically Weighted Logistic Regression (GWLR) to develop credit scoring models. The formulas of the best set of local models estimated by GWLR were compared in terms of value of the coefficients and significance of the variables, and against the global model estimated by Logistic Regression. It was used a real granting data of Direct Credit Consumer from a national public financial institution to borrowers domiciled in the Federal District (FD) of Brazil. The results demonstrated the feasibility of using the technique GWLR to develop credit scoring models. The estimated models for each region of FD have showed to be different in their variables and coefficients (parameters) and three out of five indicators calculated for the developed model by GWLR were superiors than indicators of the developed model by Logistic Regression.

The main objective of this dissertation is to understand how the main transformations that boosted the great independentist turn of the Catalan nationalism took place in the inauguration speeches of the leaders from the key party CDC, while presidential candidates of the Generalitat, the Catalan government. With the rise of new popular demands for independence as of 2010, and after the (illegal) referendum a few years later, that won with an imperative victory of the "yes" to independence, there was a sharp shift on the emphasis of the party: from moderate and autonomist, CDC began to treat the independence of Catalonia as the main objective of its governments. With the inaugurations speeches from 1980 and 1999 of Jordi Pujol, from 2010 and 2012 of Artur Mas, and the one from 2016 of Carles Puigdemont, the research intends to find the most important points that constituted the character of the transformation, as well as the role of ethnic, civic and economic nationalism in these changes. With the use of documental analysis, the dissertation is going to address as preliminary analisys the historical, social and economic contexto f the speeches, the profiles of the authors, in addition to key concepts and the internal logic of each of the speeches, in order to reach a more concise final analysis of the central problem of the work. It was comprehended that even though it is quite difficult to estabilish a cause-and-effect-relationship, it was noted the significative role of the consequences of the economic crisis of 2008 and the changes on the Statute of Autonomy of 2006 in the essence of the Catalan nationalism, that ended up inferring in a new dynamic bewteen civil society and political elites, generating the transformations inside CDC, alterating their main policies and strengthening their political use of nationalism.
O presente trabalho tem como principal objetivo compreender de que maneira se deram as principais transforma??es que impulsionaram a virada independentista do nacionalismo catal?o nos discursos dos l?deres do partido-chave CDC, enquanto candidatos ? presid?ncia da Generalitat, o governo catal?o. Com o aumento das novas reivindica??es populares por independ?ncia a partir de 2010, e depois da realiza??o do referendo (ilegal) alguns anos mais tarde, o qual obteve uma vit?ria imperativa do ?sim? ? independ?ncia, notou-se uma brusca mudan?a de ?nfase do partido: de moderado e autonomista, o CDC passou a tratar a independ?ncia da Catalunha como principal objetivo de seus governos. La sclérose en plaques (SEP) est une maladie auto-immmune notamment marquée par une défaillance de la mise en place des mécanismes de réparation tissulaire, ne permettant pas de rétablir l'homéostasie. L'utilisation de la résolution de l'inflammation, médiée par les. macrophages phagocytant des cellules apoptotiques. a récemment été évoquée afin de proposer une nouvelle famille de médicaments basée 1 sur cette résolution de l'inflammation. Dans ce cadre, nous proposons de résoudre l'inflammation dans un modèle murin de SEP, à l'aide du surnageant de co-culture de macrophages ayant phagocyté des cellules apoptotiques. induisant la production de facteurs pro-résolutifs (SuperMApo). Nous démontrons ici que l'injection de SuperMApo dans un modéle de SEP permet de contrôler la maladie via une reprogrammation des macrophages et des pDC, mais pas des cDC. dans la rate. Ces cellules démontrent en effet une capacité accrue à générer des T régulateurs (Treg) et moins de Th1, et leur présence est nécessaire pour l'effet du SuperMApo. Cette reprogrammation se fait à deux niveaux moléculaires. Premièrement. un contrôle épigénétique de l'expression des gènes induit notamment une modification de la méthylation globale de I' ADN et des promoteurs de miRNA impliqués dans la régulation des réponses immunes. Deuxièmement, une diminution de l'activation des acteurs de la voie de transcription pro-inflammatoire NFκB (nuclear factor kappa 8) a lieu. résultant d'une baisse de l'activation de ces cellules. Ces travaux démontrent donc que cibler la résolution de l'inflammation est une stratégie intéressante afin de contrôler la SEP
Multiple sclerosis (MS) is an autoimmune disease notably delined bv a default of mechanisms of tissue reparation which are insufficient to establish homeostasis. The active resolution of innammation mediated by macrophage effcrocytosis of apoptotic cells has emerged to propose a new branch of pharmacology named "resolution pharmacology". ln this study. we propose to resolve innammation in a murine model of MS. using the supermatanl issued from the culture between apoptotic cells and macrophages. inducing efferocytosis and the production of pro-resolutive factors (SuperMApo). Here. we demonstrate that injection of SupcrMApo in a model of MS allow the control of the disease correlated with a reduction of the inflammatory inliltrate in the central nervous system. The control of the disease is associated with a reprograrnming of macrophages and pDC, but not cDC, in the spleen, demonstrated by a higher capacity to generate Treg and a lower ability to generate Th 1, and their presence is necessary to the anti-innammatory effect of SupcrMApo. Reprogramming of macrophages and pDC occurs at two main levels. First. an epigenetic control of gene expression induces a modulation of global DNA methylation and a modulation of DNA methylation at the promoter of miRNA implicated in the regulation of immune responses. Secondly, a decreased activation of NFκB activation pathway is observed resulting to a decreased activation of these cells. Thus, this work demonstrates that targeting the resolution of inflammation is an interesting strategy to treat MS

Pendant mon doctorat, nous nous sommes concentrés sur l’amélioration de la vérification des interfaces asynchrones sur les systèmes multi-horloges. Notre missionétait d’optimiser la vérification de la traversée de domaine d’horloge (CDC) sur RTLpour éviter les bugs potentiels qui pourraient affecter l’intégrité du silicium. Nousavons introduit de nouveaux aspects pour élever la qualité de la vérification et avonsatteint avec succès leurs objectifs. Nous avons commencé par améliorer les méthodes de vérification structurelle existantes en introduisant le flot de vérification CDCUPF-Aware. Nous avons également cherché à intégrer la vérification fonctionnelledynamique avec la vérification statique CDC à travers le flot de vérification semiformelle CDC. Cela a conduit à la modélisation du "Qualificateur Universel", uneméthode générique pour vérifier tous les types de synchroniseurs de données CDC,et du "Flot Hybride", une façon d’utiliser des assertions pour accélérer la contraintede l’arbre d’horloge pour la vérification statique CDC. De plus, en collaboration avecAccellera, notre équipe a été pionnière dans la création de la première norme IEEEpour les modèles CDC, qui servent de représentations abstraites concises de blocset de sous-blocs pour la vérification hiérarchique CDC. Dans les travaux futurs, nousvisons à intégrer toutes les solutions proposées dans un flot cohérent et unifié. During my PhD, we focused on improving the verification of asynchronous interfaceson multi-clock systems. Our mission was to optimize Clock Domain Crossing (CDC)verification on RTL to avoid potential bugs that may affect silicon integrity. We introduced novel aspects to elevate the quality of verification and successfully met theirgoals. We started by enhancing existing structural verification methods introducingthe UPF-Aware CDC Verification Flow. We were also interested to integrate thedynamic functional verification with CDC static verification through the CDC SemiFormal Verification Flow. This led to the modeling of the "Universal Qualifier", ageneric method to verify all types of CDC data synchronizers, and the "Hybrid Flow",a way to use assertions to accelerate the clock tree constraining for the CDC staticverification. In addition, in collaboration with Accellera, our team pioneered the firstIEEE standard for CDC models, which serve as concise CDC abstracted representations of blocks and sub-blocks for CDC hierarchical verification. In future work, weaim to integrate all proposed solutions into a coherent and unified flow. We also planto address metastability injection and explore its potential in dynamic simulation andformal verification

Abstract:

The complex work of designing new ASICs today and the increasing costs of time to market (TTM) delays are putting high responsibility on the research and development teams to make fault free designs. The main purpose of implementing a static rule checking tool in the design flow today is to find errors and bugs in the hardware definition language (HDL) code as fast and soon as possible. The sooner you find a bug in the design, the shorter the turnaround time becomes, and thereby both time and money will be saved.

There are a couple of tools in the market that performs static HDL analysis and they vary in both price and functionality. In this project mainly Atrenta Spyglass was evaluated but similar tools were also evaluated for comparison purpose.

The purpose of this master thesis was to evaluate the need of implementing a rule checking tool in the design flow at the Digital ASIC department PDU Base Station development in Kista, who also was the commissioner for this project. Based on the findings in this project it is recommended that a static rule checking tool is introduced in the design flow at the ASIC department. However, in order to determine which of the different tools the following pointers should be regarded:

• If the tool is only going to be used as for lint checks (elementary structure and code checks) on RTL, then the implementation of Mentors Design Checker is advised.

• If the tool is going to be used for more sophisticated structural checks, clock tree/reset tree propagation, code checks, basic constraints checks, basic Clock Domain Crossings (CDC) checks, then Synopsys LEDA is advised.

• If the tool is going to be used as for advanced structural checks, extensive clock tree/reset tree propagation, code checks, constraints checks, functional Design For Test (DFT) checks (as testmode signal propagation) and functional CDC checks on RTL as well as on netlist level, then Atrenta Spyglass is advised.

The areas regarding checks that could be of interest for Ericsson is believed to be regular lint checks for RTL (naming, code and basic structure), clock/reset tree propagation (netlist and RTL), constraints and functional DFT checks (netlist and RTL).

Abstract This study focuses on the process of communication between government agencies and the public during crisis situations, and the development of an effective response strategy when a significant threat to public health and/or safety is believed to exist. My specific research interests are (1) the nature of the decision-making process that influences the communicative choices made during such events, and (2) how decision-makers make sense of an evolving, ambiguous, and unpredictable situation, in order to establish credibility with the public, determine the appropriate response strategy, and gain the public's trust in order to influence its behavior. This is a qualitative research study based on a series of in-depth interviews conducted with key staff members of the Centers for Disease Control and Prevention (CDC) regarding the CDC's organizational response to the 2009-2010 H1N1 influenza pandemic. As global public health threats have the potential to significantly affect critical areas of the U.S. economy, national security policies are evolving to include strategic planning for issues related to global public health threats. However, despite having faced several serious public health threats during the past decade, governments worldwide and the global public health community continue to struggle with developing sufficient contingency plans and effective response strategies to meet the challenges of unexpected, highly unpredictable, and potentially devastating public health crises. My research addresses gaps identified in exploring the experience of crisis response participants in order to understand the process of response development. Additionally, I identify practices, processes, and recommendations that will be useful for future response teams confronted with equally challenging emerging threat and/or crisis scenarios.

Abstract:
Les phosphatases à double spécificité de la famille CDC25 jouent un rôle prépondérant à différents points du cycle cellulaire en activant les complexes CDK/Cyclines. Afin de restreindre l'activation de ces complexes, les CDC25s, au nombre de trois (A, B et C) dans les cellules de mammifères, sont finement régulées au cours du cycle cellulaire tant au niveau de leur activité, de leur localisation que de leur stabilité. La phosphatase CDC25B, en activant initialement le complexe CDK1/Cycline B au niveau des centrosomes, est considérée comme le starter de la mitose. Bien que dégradée par le protéasome, peu d'informations sur la régulation de sa dégradation étaient connues. Le but de ce travail a donc été de caractériser le(s) mécanisme(s) impliqué(s), ainsi que les déterminants moléculaires présents sur CDC25B, régulant sa stabilité au cours du cycle cellulaire. Une étude in cellulo de différents mutants ponctuels de la protéine nous a permis d'identifier le motif DDGFVD comme étant nécessaire à l'interaction de CDC25B avec la protéine F-Box TrCP. La perte de cette interaction entraîne une stabilisation de CDC25B à la transition métaphase-anaphase. Cette stabilisation anormale de CDC25B engendre un retard de sortie de mitose accompagné de défauts cellulaires caractéristiques d'une instabilité génétique accrue et d'une fragmentation du matériel péricentriolaire. Par ailleurs, par vidéomicroscopie nous avons observé que les cellules exprimant le mutant stabilisé de CDC25B présentent des caractéristiques morphologiques différentes des cellules exprimant la protéine sauvage, avec une vitesse de déplacement plus importante. Sachant que la surexpression de CDC25B est détectée dans de nombreux cancers généralement très agressifs, et que, comme nous l'avons montrée, une stabilisation de CDC25B en mitose induit une instabilité génétique, nous pouvons supposer que dans certains cancers cette surexpression pourrait résulter d'un défaut de dégradation de la phosphatase. Aussi, une meilleure compréhension des mécanismes de dégradation de CDC25B permettrait d'envisager le développement d'approches thérapeutiques ayant pour but d'agir sur sa stabilité
CDC25 proteins are highly conserved dual specificity phosphatases that play an essential role by activating the CDK/Cyclin complexes all along the cell cycle. To restrain CDK/Cyclin activities, these phosphatases must be tightly regulated in terms of activity, localization and stability. One of the three mammalian members, CDC25B, is considered as the starter of the mitosis through the activation of CDK1/Cyclin B complexes at the centrosomes, at the G2-M transition. This protein is known to be degraded by the proteasome but the exact mechanisms involved in this process are still unclear. To obtain a deeper insight into the regulation of CDC25B stability, we have investigated the molecular determinants and the exact mechanisms involved in CDC25B degradation in vitro as well as in cellulo. Analysis of various mutants of CDC25B led us to identify the DDGFVD motif as a motif required for the interaction of CDC25B with the F-box protein TrCP. The lack of interaction causes a stabilisation of the phosphatase in metaphase-anaphase transition. This stabilisation entails a delay in mitotic exit and several cellular defects related to genetic instability, and the fragmentation of pericentriolar matrix during mitosis. Videomicroscopy's observations indicate that cells expressing the stabilized mutant of the CDC25B exhibit an increased mobility compared to cells expressing wild type protein. Since CDC25B is frequently overexpressed in many cancers cells and that a stabilisation of the protein entails genetic instability, we propose that in some cancers this overexpression could be a consequence of a lack of CDC25B degradation. A better understanding of mechanisms regulating CDC25B degradation could lead to new therapeutical approaches focused on the control of CDC25B stability

A hansenÃase à uma doenÃa granulomatosa, infecto-contagiosa causada pelo Mycobacterium leprae. Trata-se de uma infecÃÃo crÃnica com amplo espectro de respostas imunes celulares em humanos. Possui alto poder infectante e baixo poder patogÃnico. Este estudo tem como objetivo quantificar e comparar leucÃcitos e subpopulaÃÃes de linfÃcitos T totais (CD3+), T auxiliares (CD3+CD4+), T citotÃxicos (CD3+CD8+), B (CD19+) e NK (CD3-CD16+CD56+) em sangue perifÃrico de indivÃduos com hansenÃase e controles saudÃveis. Os pacientes foram provenientes do Centro de Dermatologia D. LibÃnia, Fortaleza-CE, Brasil. A determinaÃÃo do nÃmero de linfÃcitos em cada subpopulaÃÃo foi realizada por citometria de fluxo. A anÃlise estatÃstica foi realizada pelo programa GraphPad Prism 5.0 para Windows com significÃncia estabelecida para valores de p<0,05. à um estudo do tipo caso controle de carÃter observacional, realizado a partir da anÃlise do sangue perifÃrico de indivÃduos com diagnÃstico de hansenÃase e de indivÃduos saudÃveis. A populaÃÃo de pacientes com hansenÃase, sem tratamento foi composta de 15 pessoas. A populaÃÃo de controles saudÃveis foi composta por 29 pessoas. As mÃdias das contagens de LinfÃcitos NK (CD3-CD16+CD56+) no grupo de pacientes com hansenÃase e nos controles saudÃveis, dadas em cÃlulas/mm3, foram, 147(Â113,4) e 378,1 (Â231,7) respectivamente, p = 0,0008. As mÃdias das contagens de LinfÃcitos B (CD19+) no grupo de pacientes com hansenÃase e nos controles saudÃveis, dadas em cÃlulas/mm3, foram, 233,3 (Â85,89) e 115,3 (Â53,01) , respectivamente, p < 0,0001. NÃo foram encontradas diferenÃas estatÃsticas significantes entre as amostras de leucÃcitos, de linfÃcitos T CD3+, linfÃcitos T CD4+ e linfÃcitos T CD8+. Os dados do presente estudo sinalizam que as cÃlulas NK parecem desempenhar papel de relevÃncia na resposta ao M. leprae. O linfÃcito B jà ocupa papel de destaque na resposta imunolÃgica ao M. leprae, sobretudo nas formas lepromatosas, e este estudo reforÃa a importÃncia destas cÃlulas.
Leprosy is an infectious and granulomatous disease caused by Mycobacterium leprae. The aim of this study was to quantify and compare levels of leucocytes and lymphocyte subpopulations (CD3+, CD3+CD4+, CD3+CD8+, CD19+, CD3-CD16+CD56+) in peripheral blood of patients with leprosy and healthy controls. Patients were followed at Centro de Dermatologia D. LibÃnia, Fortaleza-CE, Brasil. Flow cytometry was used to determine numbers of lymphocytes. Statistical analisys was done with GraphPad Prism 5.0 software for windows. P values under 0.05 were considered siginificant.This was an observational case-control study. Fifteen leprosy patients without treatment were evaluated and 29 healthy individuals were included in control group. NK cells (CD3-CD16+CD56+) mean in leprosy patients was 147(Â113,4) and in controls was 378,1 (Â231,7). Comparisson stablished a p value of 0.0008. B lymphocytes (CD19+) mean in leprosy patients was 233,3 (Â85,89) and in controls was 115,3 (Â53,01), with p < 0,0001 . No differences were observed in CD3+ T lymphocytes, CD4+ T lymphocytes and CD8+ T lymphocytes. This study suggests that NK cells may play a role in innate response to M. leprae.

Leprosy is an infectious and granulomatous disease caused by Mycobacterium leprae. The aim of this study was to quantify and compare levels of leucocytes and lymphocyte subpopulations (CD3+, CD3+CD4+, CD3+CD8+, CD19+, CD3-CD16+CD56+) in peripheral blood of patients with leprosy and healthy controls. Patients were followed at Centro de Dermatologia D. Libânia, Fortaleza-CE, Brasil. Flow cytometry was used to determine numbers of lymphocytes. Statistical analisys was done with GraphPad Prism 5.0 software for windows. P values under 0.05 were considered siginificant.This was an observational case-control study. Fifteen leprosy patients without treatment were evaluated and 29 healthy individuals were included in control group. NK cells (CD3-CD16+CD56+) mean in leprosy patients was 147(±113,4) and in controls was 378,1 (±231,7). Comparisson stablished a p value of 0.0008. B lymphocytes (CD19+) mean in leprosy patients was 233,3 (±85,89) and in controls was 115,3 (±53,01), with p < 0,0001 . No differences were observed in CD3+ T lymphocytes, CD4+ T lymphocytes and CD8+ T lymphocytes. This study suggests that NK cells may play a role in innate response to M. leprae.
A hanseníase é uma doença granulomatosa, infecto-contagiosa causada pelo Mycobacterium leprae. Trata-se de uma infecção crônica com amplo espectro de respostas imunes celulares em humanos. Possui alto poder infectante e baixo poder patogênico. Este estudo tem como objetivo quantificar e comparar leucócitos e subpopulações de linfócitos T totais (CD3+), T auxiliares (CD3+CD4+), T citotóxicos (CD3+CD8+), B (CD19+) e NK (CD3-CD16+CD56+) em sangue periférico de indivíduos com hanseníase e controles saudáveis. Os pacientes foram provenientes do Centro de Dermatologia D. Libânia, Fortaleza-CE, Brasil. A determinação do número de linfócitos em cada subpopulação foi realizada por citometria de fluxo. A análise estatística foi realizada pelo programa GraphPad Prism 5.0 para Windows com significância estabelecida para valores de p<0,05. É um estudo do tipo caso controle de caráter observacional, realizado a partir da análise do sangue periférico de indivíduos com diagnóstico de hanseníase e de indivíduos saudáveis. A população de pacientes com hanseníase, sem tratamento foi composta de 15 pessoas. A população de controles saudáveis foi composta por 29 pessoas. As médias das contagens de Linfócitos NK (CD3-CD16+CD56+) no grupo de pacientes com hanseníase e nos controles saudáveis, dadas em células/mm3, foram, 147(±113,4) e 378,1 (±231,7) respectivamente, p = 0,0008. As médias das contagens de Linfócitos B (CD19+) no grupo de pacientes com hanseníase e nos controles saudáveis, dadas em células/mm3, foram, 233,3 (±85,89) e 115,3 (±53,01) , respectivamente, p < 0,0001. Não foram encontradas diferenças estatísticas significantes entre as amostras de leucócitos, de linfócitos T CD3+, linfócitos T CD4+ e linfócitos T CD8+. Os dados do presente estudo sinalizam que as células NK parecem desempenhar papel de relevância na resposta ao M. leprae. O linfócito B já ocupa papel de destaque na resposta imunológica ao M. leprae, sobretudo nas formas lepromatosas, e este estudo reforça a importância destas células.

Efforts by community-based organizations (CBOs), such as community development corporations (CDCs), have incorporated small business development into their overall neighborhood revitalization efforts with mixed success, especially when working with minority, low- and moderate-income entrepreneurs. These entrepreneurs often lack the technical skills and access to capital that they need to start, expand, or maintain their businesses. They face linguistic and social barriers that isolate them from traditional lenders and small business assistance providers. This thesis seeks to 1) provide a context for community-based organizations' roles in small business development for those unfamiliar with the challenges involved in minority, low- and moderate-income business development and 2) provide CBOs interested in pursuing small business development with some comparative guidelines for their own efforts. The thesis identifies three elements that contribute to the success of small businesses-social, human, and financial capital. Minority, low- and moderate-income entrepreneurs often face deficiencies in one or more of these elements. Community development corporations have attempted to provide technical assistance to businesses in their communities to address these issues, but are often faced with their own constraints of limited staffing and resources. One collaboration of ten Boston area CDCs is attempting to break down these constraints and combine and share the expertise of the member CDCs. This collaboration known as the CDC Community Business Network (CBN) provides one-on-one neighborhood-based assistance yet also provides access to the larger experiences and contacts of the other member CDCs. Even with the shared resources of its member CDCs, CBN still faces its own challenges of limited skills, capacity, and funding; an unclear role among other technical assistance providers; and the need to develop effective collaborations with other stakeholders in small business development. The thesis examines two other small business assistance networks and looks to them for lessons in addressing the deficiencies in the three types of capital and the challenges facing CBN. The two comparable networks offer alternative organizational and operational approaches to small business assistance. This thesis asserts that small business assistance organizations must have an understanding of the deficiencies in social, human, and financial capital in order to effectively assist minority, low- and moderate-income entrepreneurs.

Specific Aims: The purpose of this study was to assess pharmacists’, pharmacy technicians’, and pharmacy interns’ knowledge of current Centers for Disease Control and Prevention (CDC) immunization guidelines for pregnant women. Methods: Questionnaires administered to volunteers during the Arizona Pharmacy Association (AzPA) 2013 Annual Convention and Trade Show collected data showing the volunteers’ level of knowledge about current immunization guidelines; data on professional roles (pharmacist, pharmacy intern, or pharmacy technician), years in practice, current immunization certification status and activity, and practice setting were also collected. Main Results: Questionnaires were completed by 112 volunteers, including 48 pharmacists, 25 pharmacy technicians, and 39 pharmacy interns. The overall percentage of correct answers from all participants was 33%. Pharmacists, pharmacy technicians, and pharmacy interns had correct answer percentages of 41%, 16%, and 34%, respectively. Pharmacy practitioners who were state certified to perform immunizations performed statistically significantly better than the non-certified group (44.2% correct versus 33% correct, P=0.012). Practitioners who work at a practice site that provides immunizations were compared with practitioners who do not, with results trending toward statistical significance, but falling just short (45.7% correct versus 36% correct, P=0.054). Conclusion: The general level of knowledge about CDC immunization guidelines appears to be inadequate among the volunteer group of pharmacy practitioners, possibly leading to missed opportunities for needed immunizations.

Abstract:
Silicon was selectively removed from a silicon carbonitride (SiCN) aerogel by hot chlorine gas treatment, leading to a N-doped carbon aerogel (N-CDC aerogel). The combined effects of pyrolysis and etching temperature were studied with regard to the change in the composition of the material after etching as well as the microstructure of the produced hierarchically porous material. Upon removal of Si from amorphous SiCN, carbon and nitrogen, which are not bonded together in the starting material, react, creating new C–N bonds. The removal of silicon also gives rise to a high amount of micropores and hence a high specific surface area, which can be beneficial for the functionality of the carbonaceous material produced. The mesoporous structure of the aerogel allows us to complete the etching at low temperature, which was found to be a crucial parameter to maintain a high amount of nitrogen in the material. The combination of a high amount of micropores and the mesopore transport system is beneficial for adsorption processes due to the combination of a high amount of adsorption sites and effective transport properties of the material. The N-CDC aerogels were characterized by nitrogen physisorption, X-ray photoelectron spectroscopy (XPS), thermogravimetry (TG/DTA), and infrared spectroscopy (DRIFT) and they were evaluated as CO2 absorbers and as electrodes for electric double-layer capacitors (EDLCs).

Abstract:
This master thesis examines the possibility to improve the efficiency of the newSwedish occupational pension system. Concerns have been raised that the occupational pension system will yield unsatisfactory pensions that will cause problems for future generations. By implementing a solidarity reserve, a buffer, the market risk can possibly be mitigated, hence enabling a higher expected return for the beneficiaries. A model was created that simulates two pension plans. The first, a benchmark plan with features derived from the Swedish occupational pension system, while the second plan holds a collective solidarity reserve and increased investment risk. The model was run a large number of times over different scenarios and relevant data was extracted from each run. The data is then analyzed with respect to the efficacy of the solidarity reserve as a tool for risk mitigation and with respect to key considerations to take into account while implementing a solidarity reserve. The results from the project is that the plan that utilizes a solidarity reserve with a higher risk level yields higher mean payouts for the beneficiaries and that the two plans yield similar outcomes in adverse scenarios. Thus, the conclusion from the project is that the solidarity reserve has the potential of being a useful tool for risk mitigation. This allows a higher risk profile, which enables higher expected return for the beneficiaries. The primary consideration to take into account is the investment strategy used for the buffer and the authors propose that a hedged strategy can be successful in order to lower the correlation with the market in case of an adverse market event. The thesis also proposes a number of areas for further research, among others, a detailed study of how a solidarity reserve would function in an extremely adverse scenario. Optimization of the premium contribution levels for the buffer, the size of the buffer and the disbursement strategy of the buffer are also proposed as useful topics for further research.

IKEA, attraverso un ambizioso piano strutturale, si sta impegnando al massimo per rendersi più comoda e accessibile su tutti i canali, studiando nuovi punti di incontro con la propria clientela. Per far fronte a questi compiti, l’organizzazione fa affidamento su una solida partnership con i propri fornitori, tra cui società di trasporto e aziende specializzate in montaggio e installazioni. I principali servizi offerti sono prelievo, consegna, montaggio e installazione, ma i mercati ne forniscono anche di aggiuntivi customizzati alle esigenze locali. Vi sono inoltre i Customer Support Centers, sempre a disposizione per fornire assistenza. Una supply chain altamente efficace, reattiva e innovativa è un fattore critico di successo per tutti i canali. A tale scopo, all’interno dell’elaborato, viene effettuata un’analisi che mette a confronto l’andamento del parcel, riguardante gli ordini di piccole dimensioni, con quello del truck, in uscita dai negozi. Lo studio prosegue concentrandosi sul lead time offerto al cliente e sulla saturazione caratterizzante entrambi i flussi, e si conclude con un’analisi volta alla quantificazione degli oversales rilevati dagli store nella preparazione degli ordini. In ambo gli scenari sono state rilevate problematiche, che hanno spinto l’introduzione della variazione di peso allo scopo di equilibrare i flussi IKEA. Attraverso l’applicazione di tale manovra è stato possibile massimizzare le performance offerte dall’organizzazione e creare flessibilità nella rete logistica. La supply chain svolge un ruolo cruciale ai fini del successo di IKEA, ed è importante che l’efficienza venga mantenuta lungo tutti i flussi logistici. Quando ciò non avviene è indispensabile intervenire prontamente e compensare i disequilibri.
