Abstract:
In order to improve the measuring precision of the one-dimension line-matrix CCD, a new sub-pixel edge detection method is proposed. Firstly the date collected by the line-matrix CCD is converted to digital data by virtual oscillograph. Base on threshold comparison, the fitting part of the edge signal is picked up for further processing; secondly, the pixels are expanded following the edge direction of the edge point, and the edge signal is fitted through the two order multinomial; finally, comparing the two order curve with the threshold voltage through the least square fit method, the precise position of the edge point can achieve the sub-pixel edge location precision.

Abstract:
Objective. To compare the detection of microcalcifications on mammograms of an anthropomorphic breast phantom acquired by a direct digital flat-panel detector mammography system (FPM) versus a stereotactic breast biopsy system utilizing CCD (charge-coupled device) technology with either a 1024 or 512 acquisition matrix (1024 CCD and 512 CCD).Materials and Methods. Randomly distributed silica beads (diameter 100–1400 m) and anthropomorphic scatter bodies were applied to 48 transparent films. The test specimens were radiographed on a direct digital FPM and by the indirect 1024 CCD and 512 CCD techniques. Four radiologists rated the monitor-displayed images independently of each other in random order.Results. The rate of correct positive readings for the “number of detectable microcalcifications” for silica beads of 100–199 m in diameter was 54.2%, 50.0% and 45.8% by FPM, 1024 CCD and 512 CCD, respectively. The inter-rater variability was most pronounced for silica beads of 100–199 m in diameter. The greatest agreement with the gold standard was observed for beads 400 m in diameter across all methods.Conclusion. Stereotactic spot images taken by 1024 matrix CCD technique are diagnostically equivalent to direct digital flat-panel mammograms for visualizing simulated microcalcifications 400 m in diameter.

Abstract:
The use of loss function in studying the reduction in determinant of information matrix due to missing observations has effectively produced designs that are robust to missing observations. Modified central composite designs are constructed for non-standard models using principles of the loss function or equivalently first compound of (I ) matrix associated with hat matrix . Although central composite designs (CCDs) are reasonably robust to model mis-specifications, efficient designs with fewer design points are more economical. By classifying the losses due to missing design points in the CCD portions, where there are multiple losses associated with specified CCD portions, the design points having less influence may be deleted from the full CCD. This leads to a possible increase in design efficiency and offers alternative designs, similar in the structure of CCDs, for non-standard models.

AbstractThe observational data of July 11, 1991 eclipse solar corona obtained by both electropolarimeter (EP) and CCD-matrix were processed. Using these data, the solar corona photometry was carried out. The results of EP data are compared with the ones of CCD data. It must be noted here that the CCD data give us only characteristics of the inner corona, while the EP data show the features of both the inner and middle corona up to 4R⊙. Standard flattening indexϵis evaluated from both data. The dependence of the flattening index on the distance from the solar limb is investigated. The isophotes in Na and Ca lines are plotted. Based on these data some ideas and conclusions on the type of the solar corona are presented.

Abstract:
Paracrine and autocrine actions of the insulin-like growth factors (IGFs) are inferred by local expression within the bowel. CCD-18Co cells, IEC-6 cells, and immunoneutralization were used to analyze whether IGFs have direct autocrine or paracrine effects on proliferation of cultured intestinal fibroblasts and epithelial cells. Growth factor expression was analyzed by ribonuclease protection assay and RT-PCR. Extracellular matrix (ECM) was analyzed for effects on cell proliferation. CCD-18Co cells express IGF-II mRNAs and low levels of IGF-I mRNA. Conditioned medium from CCD-18Co cells (CCD-CM) stimulated proliferation of IEC-6 and CCD-18Co cells. Neutralization of IGF immunoreactivity in CCD-CM reduced but did not abolish this effect. RT-PCR and immunoneutralization demonstrated that other growth factors contribute to mitogenic activity of CCD-CM. Preincubation of CCD-CM with ECM prepared from IEC-6 or CCD-18Co cells reduced its mitogenic activity. ECM from CCD-18Co cells enhanced growth factor-dependent proliferation of IEC-6 cells. IEC-6 cell ECM inhibited IGF-I action on CCD-18Co cells. We conclude that IGF-II is a potent autocrine mitogen for intestinal fibroblasts. IGF-II interacts with other fibroblast-derived growth factors and ECM to stimulate proliferation of intestinal epithelial cells in a paracrine manner.

Abstract:
Imaging model of double CCD camera based on pinhole camera model and distortion model is introduced, and the fundamental matrix is presented. Distortion model is led into the fundamental matrix calibration. Based on the imaging model of Z. Zhangs calibration method, fundamental matrix is carried out. Pixel coordinate for calibration is modified by cameras interior parameters and distortion coefficients. Base on the imaging model of double CCD camera, measurement model for cam spacing is established. Cam spacing is measured by the feature of cam edge. In experiments, accurate pixel coordinates of model plane were used in calibration. The distance between a point and its corresponding epipolar line was estimated as the error. The measurement results show that the accuracy of proposed method satisfied the monitoring requirements of the axial positioning error of the cam piece.

Abstract:
Cell-cell or cell-matrix interactions have crucial roles in the maintenance of the physical cohesion of any tissue. In addition, growing body of evidence indicates that these two adhesion systems do not act independently, but rather are functionally interconnected by a permanent crosstalk. This dialog usually operates via common molecules that trigger convergent signaling as well as by actomyosin network which, by providing physical link, contributes to establishment of intracellular force counterbalancing tension applied by extracellular surrounding. Blood vessels endothelium is a particular tissue in term of mechanical conditions. Apart from intracellular compression, endothelial lining needs to resist hemodynamic forces as well as rigidity of the basal membrane - two mechanical inputs acting from opposite sides of the endothelial layer. Cerebral Cavernous Malformation (CCM) is a sporadically acquired or inherited disease of venous capillaries within neuro-vascular unit characterized by defects in all aspects of local microenvironment. Loss of intra-endothelial junctions and mural cell coverage, aberrant organization of basal lamina as well as stagnant blood flow are features of CCM lesions. Thereby, CCM became for us an interesting model to study mechanotrasduction process and in this context, the cross-talk between integrin and cadherin mediated adhesion structures. Indeed, CCM proteins are well recognized players involved in a control of VE-cadherin mediated intracellular junctions. In addition, CCM1 was found to interact with ICAP-1, a negative regulator of β1 integrin, raising the possibility that this complex most likely acts as molecular node regulating β1 integrin/ VE-cadherin convergent signaling pathways.Studies performed during this thesis have demonstrated that CCM complex coordinates cadherin- and integrin-mediated tensional homeostasis by repressing β1 integrin activation and maintaining a balance of activity between the two isoforms of RhoA-associated kinases ROCK1 and ROCK2. We have found that β1 integrin sustained over-activation upon CCM proteins loss contributes to increased ECs sensitivity to mechanical cues, such as ECM physical reorganization or hemodynamic force that in turn activates ROCK1-dependent contractility. This establishes a positive feedback mechanical loop that breaks tensional homeostasis and switches on the malignant phenotype

Abstract:
Genome-wide association studies (GWAS) have identified up to 30 loci that associate with increased risk of coronary artery disease or myocardial infarction. Here, I tested the function of one locus that changed the amino acid sequence of a mitochondrial matrix protease called paraplegin (SPG7) that performs critical quality assurance functions. Loss-of-function mutations in this protease are associated with hereditary spastic paraplegia. Here, I show that this variant that changes an arginine to a glutamine at position 688 within the protease domain is a gain-of-function. Cells bearing this variant have increased mitochondrial fusion and number, produce higher levels of reactive oxygen species and have increased cellular proliferation. Importantly, when expressed in yeast, the Q688 variant of SPG7 rescues the growth arrest caused by a protease-deficient mutation in AFG3L2. My study identifies a novel functional variant of SPG7 and highlights the need to go beyond the GWAS paradigm.

Pless, Elin. "Investigation of interactions with extracellular matrix proteins mediated by the CCP modules of the metabotropic GABAB receptor." Thesis, University of Edinburgh, 2010. http://hdl.handle.net/1842/5714.

Full text
Abstract:
GABAB receptors are G-protein coupled receptors for the major inhibitory neurotransmitter in the mammalian central nervous system, γ-aminobutyric acid (GABA). The receptor is linked to a variety of disorders including epilepsy, pain, spasticity, drug addiction and cognitive impairment and is, therefore of major importance for drug discovery. The most abundant receptor isoforms GABABR1a and R1b differ by the presence in R1a of a pair of Nterminal extracellular complement control protein modules (CCP1 and CCP2) which - in other proteins - are generally involved in mediating specific protein-protein recognition. The CCP1 module contains disulphides but is natively disordered. In the current work, the yeast two-hybrid system was used to confirm an interaction of CCP1 of GABABR1a with the extracellular protein fibulin-2. Further work with the yeast twohybrid system extablished the novel interaction of the abundant extracellular matrix protein laminin, with GABABR1a CCP1, via its laminin globular (LG) domains. The laminin interaction was further characterised by surface plasmon resonance, demonstrating that several different domains are involved in the binding to the GABAB receptor CCPs. The primary binding site is located on laminin α5 LG4-5, but the E10 domains of the β1 chain and LG1-3 on α1 may also be involved. The pharmacological properties of the GABABR1a and R1b isoforms were studied by transient expression in Xenopus laevis oocytes. It was demonstrated that the agonist baclofen, as well as the antagonist CGP55845, appear to be more potent at GABABR1b compared to GABABR1a. Intriguingly, when recorded in the precence of laminin, GABABR1b/R2 expressing oocytes exhibited an increased baclofen-evoked response while the response in GABABR1a/R2 was completely abolished. In conclusion, the work demonstrates that laminin is a binding partner for GABABR1a CCPs. Such an interaction between the metabotropic GABA receptor and the extracellular matrix may lie behind the recently reported roles of GABA in neuronal migration and the laying down of neuronal circuitry during the development of parts of the central nervous system.

The image acquisition methods applied to nuclear medicine and radiobiology are a valuable research study for determination of thyroid anatomy to seek disorders associated to follicular cells. The Monte Carlo (MC) simulation has also been used in problems related to radiation detection in order to map medical images since the improvement of data processing compatible with personnel computers (PC). This work presents an innovative study to find out the adequate scintillation inorganic detector array that could be coupled to a specific light photo sensor, a charge coupled device (CCD) through a fiber optic plate in order to map the follicles of thyroid gland. The goal is to choose the type of detector that fits the application suggested here with spatial resolution of 10 m and good detector efficiency. The methodology results are useful to map a follicle image using gamma radiation emission. A source - detector simulation is performed by using a MCNP4B (Monte Carlo for Neutron Photon transport) general code considering different source energies, detector materials and geometries including pixel sizes and reflector types. The results demonstrate that by using MCNP4B code is possible to searching for useful parameters related to the systems used in nuclear medicine, specifically in radiobiology applied to endocrine physiology studies to acquiring thyroid follicles images.

Abstract:

Abstract:
This article considers some classical and more modern results obtained in random matrix theory (RMT) for applications in statistics. In the classic paradigm of parametric statistics, data are generated randomly according to a probability distribution indexed by parameters. From this data, which is by nature random, the properties of the deterministic (and unknown) parameters may be inferred. The ability to infer properties of the unknown Σ (the population covariance matrix) will depend on the quality of the estimator. The article first provides an overview of two spectral statistical techniques, principal components analysis (PCA) and canonical correlation analysis (CCA), before discussing the Wishart distribution and normal theory. It then describes extreme eigenvalues and Tracy–Widom laws, taking into account the results obtained in the asymptotic setting of ‘large p, large n’. It also analyses the results for the limiting spectra of sample covariance matrices..

Abstract:

Abstract:
The primary argument of this chapter is that children’s and youth’s defiance may be misread and misinterpreted unless a greater understanding of the interplay of genders, races, and ethnicities is grasped. It analyzes various types of aggression to illustrate that the norms that determine harms from aggressive behavior need to be articulated and critiqued. The chapter sets out central characteristics of Oppositional Defiant Disorder (ODD) and Conduct Disorder (CD), then analyzes the larger context for understanding defiant behavior. Research on features of aggression in children’s play is included, and this leads to an analysis of how to understand the harms of aggression. The author also examines the matrix of raced, gendered, and classed intersections in the interpretation and reproduction of norms for behavior. This analysis of the construct of aggression makes it more difficult to interpret certain behavior as maladaptive defiant traits. The chapter ends with considerations as to why children (and adults) might have reasons for being defiant.

Abstract:
Heart failure may be a consequence of ischemic or non-ischemic cardiomyopathy. Etiologies for LV systolic dysfunction in ischemic cardiomyopathy include; 1) transmural scar, 2) nontransmural scar, 3) repetitive myocardial stunning, 4) hibernating myocardium, and 5) remodeled myocardium. The LV remodeling process, which is activated by the renin-angiotensin system (RAS), stimulates toxic catecholamine actions and matrix metalloproteinases, resulting in maladaptive cellular and molecular alterations5, with a final pathway to interstitial fibrosis. These responses to LV dysfunction and interstitial fibrosis lead to progressive worsening of LV function. Established treatment options for ischemic cardiomyopathy include medical therapy, revascularization, and cardiac transplantation. While there has been continuous progress in the medical treatment of heart failure with beta-blockers, angiotensin-converting enzyme (ACE) inhibition, angiotensin II type 1 receptor (AT1R) blockers, and aldosterone to beneficially influence morbidity and mortality, the 5-years mortality rate for heart failure patients remains as high as 50%. Revascularization procedures include percutaneous transluminal coronary artery interventions (PCI) including angioplasty and endovascular stent placement and coronary artery bypass grafting (CABG). Whereas patents with heart failure due to non-coronary etiologies may best benefit from medical therapy or heart transplantation, coronary revascularization has the potential to improve ventricular function, symptoms, and long term survival, in patients with heart failure symptoms due to CAD and ischemic cardiomyopathy.

Abstract:
Vascular calcification (VC) is a common feature of patients with advanced CKD and it could be, at least in part, the cause of increased cardiovascular mortality in these patients. From a morphologic point of view, there are at least two types of pathologic calcium phosphate deposition in the arterial wall—namely, intima calcification (mostly associated with atherosclerotic plaques) and media calcification (associated with stiffening of the vasculature, resulting in significantly adverse cardiovascular outcomes). Although VC was viewed initially as a passive phenomenon, it appears to be a cell-mediated, dynamic, and actively regulated process that closely resembles the formation of normal bone tissue, as discovered recently. VC seems to be the result of the dysregulation of the equilibrium between promoters and inhibitors. The determinants are mostly represented by altered calcium and phosphorus metabolism, secondary hyperparathyroidism, vitamin D excess, high fibroblast growth factor 23, and high levels of indoxyl sulphate or leptin; meanwhile, the inhibitors are vitamin K, fetuin A, matrix G1a protein, osteoprotegerin, and pyrophosphate. A number of non-invasive imaging techniques are available to investigate cardiac and vascular calcification: plain X-rays, to identify macroscopic calcifications of the aorta and peripheral arteries; two-dimensional ultrasound for investigating the calcification of carotid arteries, femoral arteries, and aorta; echocardiography, for assessment of valvular calcification; and, of course, computed tomography technologies, which constitute the gold standard for quantification of coronary artery and aorta calcification. All these methods have a series of advantages and limitations. The treatment/ prevention of VC is currently mostly around calcium-mineral bone disease interventions, and unproven. There are interesting hypotheses around vitamin K, Magnesium, sodium thiosulphate and other potential agents.

Abstract:
Full text
Abstract:

Abstract:

Abstract:
Abstract The stability behavior of a virtual thrust chamber demonstrator with low injection pressure loss is studied numerically. The approach relies on an eigenvalue analysis of the Linearized Euler Equations. An updated form of the stability prediction procedure is outlined, addressing mean flow and flame response calculations. The acoustics of the isolated oxidizer dome are discussed as well as the complete system incorporating dome and combustion chamber. The coupling between both components is realized via a scattering matrix representing the injectors. A flame transfer function is applied to determine the damping rates. Thereby it is found that the procedure for the extraction of the flame transfer function from the CFD solution has a significant impact on the stability predictions.

Abstract:
The secondary effect discovery of synthetic sorbents opened another research direction for many field of studies. However, the sorption parameters of lignocellulosic sorbents are rarely reported most importantly, kenaf shive. This paper centered at the sorption behavior of optimized kenaf shive sorbents using Response Surface Methodology (RSM) via surface deposit technique. Five-level Central Composite Design (CCD) experimental matrix was used to analyze the effect of particle sizes 125‒1000μm, stirring time (5–30 min) and methyltrimethoxysilane (MTMS) concentration (5–20% v/v) as individual and combined variables process in the developed sorbents. The unmodified shive was compared with the modified, and it reveals a positive shift in the sorption capability. Instrumental analysis such as FTIR (Fourier Transform Infra-Red), XRD (X-ray Diffraction), DT-TGA (Differential Thermal-Thermogravimetric analysis) and BET (Brunaure-Emmett–Teller) were carried out on the optimized sorbent and the results were in conformity with the sorption results. The sorption behavior deployed fits the pseudo-first-order and Langmuir isotherm with regression coefficient R2=0.9496 and R2=0.9400. The sorption property was found to be spontaneous and exothermic, however, the activation energy studies shows physic-sorption phenomenon with 25.3kJmol−1 and R2=0.9360.

Abstract:
This chapter discusses the standardization of instruments and typologies in social work assessment and introduces, from a multidimensional perspective, a new standardized instrument evaluating the level of complexity associated with the social intervention process in a sample of chronic kidney disease (CKD) patients. The authors evaluated the matrix's metric properties by internal consistency and defined a rating index through the best cutoff points, using receiver operator curve and Youden Index. Matrix construction and validation used focus groups of experts in blinded classification of 100 CKD patients and indicator weighting. The matrix shows good internal consistency and reliability (Cronbach's alpha = .742). Cutoff points indicate three levels of complexity classification. The matrix is a good instrument to identify the complexity associated with the social intervention process in the area of Nephrology, and is a relevant contribution to the social information management of social workers, the health teams and the administration of health units.

Abstract:
Chronic kidney disease (CKD) is a worldwide health problem affecting 9.1% of the world’s population. The treatments to prevent the progression of CKD remain limited, however. Resident fibroblasts in the kidneys play crucial roles in the pathological conditions commonly recognized in CKD, such as renal fibrosis, renal anemia, and peritubular capillary loss. Fibroblasts in the kidney provide structural backbone by producing extracellular matrix proteins and produce erythropoietin for normal hematopoiesis under physiological conditions. In the diseased condition, however, fibroblasts differentiate into myofibroblasts that produce excessive extracellular matrix proteins at the cost of the inherent erythropoietin-producing abilities, resulting in renal fibrosis and renal anemia. Pericytes, which are mesenchymal cells that enwrap peritubular capillaries and highly overlap with resident fibroblasts, detach from peritubular capillary walls in response to kidney injury, resulting in peritubular capillary loss and tissue hypoxia. Several reports have demonstrated the beneficial roles of fibroblasts in the regeneration of renal tubules Renal fibroblasts also have the potential to differentiate into a proinflammatory state, producing various cytokines and chemokines and prolonging inflammation by forming tertiary lymphoid tissues, functional lymphoid aggregates, in some pathological conditions. In this article, we describe the heterogenous functions of renal fibroblasts under healthy and diseased conditions.

Abstract:
Compressive covariance sensing (CCS) can recover the second-order statistics of a signal that has undergone compression, and this can be achieved without the requirement of sparsity conditions. Instead, certain structure information in the statistical domain is to be captured during compression. In particular, least squares compressive covariance sensing is considered which requires the estimation of the covariance of the available compressed signal in order to recover the covariance matrix of the original signal. Different covariance estimation methods are applied and the CCS performance compared, in the presence of white Gaussian noise, in terms of the normalized mean square error between the true and recovered covariance.
