Memoria para optar el título de Bioquímico
Cupriavidus necator JMP134 (pJP4), antes denominada Ralstonia eutropha JMP134 (pJP4), es una bacteria ambiental capaz de utilizar los compuestos contaminantes 2,4-diclorofenoxiacetato (2,4-D) y 3-clorobenzoato (3-CB) como única fuente de carbono y energía. Las vías metabólicas que permiten dichas capacidades se encuentran codificadas en los genes cromosomales benABC y benD, y en la vía de degradación de clorocatecoles, codificados por los genes tfd del plasmidio pJP4. Se ha descrito que el regulador TfdR/S (familia LysR) activa la expresión de los genes tfd. El regulador de la vía de degradación de catecol, CatR, tiene en común aspectos genéticos, funcionales y evolutivos con el regulador TfdR/S. Estos antecedentes hacen posible proponer que CatR podría estar influyendo en la expresión de los genes tfd. En esta memoria se estudió el papel de CatR en la degradación de 3-CB y 2,4-D. Para esto se identificaron los putativos genes catR en el genoma de C. necator, encontrándose dos, catR1 y catR2, cuyo contexto genómico sugiere que podrían participar diferencialmente en el metabolismo de cloroaromáticos. Estos genes fueron clonados e introducidos en C. necator. Adicionalmente, se obtuvo una cepa de C. necator mutante del gen catR1 mediante una estrategia de doble recombinación homóloga. Esta mutante fue complementada con los genes catR1 o catR2. Estas cepas fueron estudiadas mediante curvas de crecimiento en medios de cultivos con distintas concentraciones de 3-CB, 2,4-D o benzoato (BZ). En cada caso se determinó la densidad óptica en fase estacionaria y la velocidad de crecimiento. Además, se evaluó la degradación de 3-CB y BZ mediante la detección del compuesto por HPLC. Estos estudios permitieron determinar que tanto CatR1 como CatR2 aumentan la degradación de 3-CB, puesto que la sobreexpresión de sus genes aumentó la velocidad de crecimiento (μ) en ±2 veces. Consistente con lo anterior, la ausencia de CatR1 disminuyó la velocidad de crecimiento en ±3 veces en 3-CB y la tasa de degradación de este sustrato disminuyó ±3 veces, lo cual también fue observado durante el crecimiento en BZ. Adicionalmente, los estudios de complementación realizados para corregir la ausencia de CatR1, con los genes catR1 y catR2 por separado, mostraron recuperar el fenotipo silvestre en ambos sustratos. Por otra parte, la degradación de 2,4-D fue mejorada por CatR2, puesto que la sobreexpresión del gen catR2 impidió la disminución en el rendimiento celular que se observa en la cepa silvestre al aumentar la concentración de 2,4-D. En cambio, la velocidad de crecimiento en 2,4-D a altas concentraciones de sustrato se vio desfavorecida por la sobreexpresión del gen catR1. Los resultados de este trabajo indican que los reguladores CatR1 y CatR2 modifican el crecimiento en estos compuestos, probablemente a través de la modulación de la expresión de los genes ben y tfd. Lo anterior se basa en que ambos reguladores poseen cercanía estructural con el regulador TfdR y compartirían la capacidad de interactuar con las respectivas regiones promotoras. En este contexto, se propone un modelo de la regulación ejercida por CatR1 y CatR2 en C. necator. Este modelo sugiere que estos reguladores modulan diferencialmente los genes tfd, de modo que CatR2 activa los módulos génicos tfd I-II y el gen tfdA, mientras que CatR1 activa sólo el módulo tfd I. En base a modelamiento molecular, se predijo que las diferencias observadas entre el regulador CatR1 y CatR2 podrían ser explicadas por la afinidad de unión a DNA y no por la unión del inductor
Cupriavidus necator JMP134 (pJP4) (Ralstonia eutropha) is a soil bacterium that is able to use the pollutants 2,4-dichlorophenoxyacetic acid (2,4-D), and 3-chlorobenzoate (3-CB), as sole carbon and energy source. The metabolic pathways are encoded in the chromosome by the benABC and benD genes and in the pJP4 plasmid, by the chlorocatechol degrading, ortho ring cleavage pathway tfd genes. It has been reported that the TfdR/S regulator (LysR family) activates the expression of the tfd genes. The regulator of the catechol degradative pathway, CatR, shares genetic, functional and evolutionary aspects with the TfdR/S regulator, making possible that CatR may influence the expression of tfd genes. In this work, the role of CatR in 3-CB or 2,4-D degradation was studied. Two putative genes encoding CatR regulators were identified in the genome of C. necator, catR1 and catR2, genetic contexts at which suggest that they may be differentially involved in chloroaromatic metabolism. These genes were cloned and overexpressed in C. necator. In addition, a catR1 gene mutant was obtained by a double recombination strategy. This mutant was complemented with either the catR1 or catR2 genes. These strains were studied by growth curves in cultures grown at different concentrations of 3-CB, 2,4-D or benzoate (BZ). In each case, the optical density at the stationary phase and the growth rate were determined. In addition, the degradation of 3-CB or BZ was monitored by HPLC. These studies showed that both regulators increase degradation of 3-CB, since overexpression of catR1 or catR2 genes provoked a two-fold increase in growth rate. Accordingly, a three-fold decrease in growth rate and 3-CB degradation rate was determined in the absence of catR1. The same was observed during growth in BZ. In addition, complementation with catR1 or catR2 genes recovered wild type growth phenotype with both substrates. On the other hand, the degradation of 2,4-D was improved for CatR2, because the overexpression of catR2 gene avoided the decrease in cell yield at higher concentrations of 2,4-D. In contrast, the degradation of 2,4-D at higher concentrations of this substrate was impaired by overexpression of catR1. The results of this work show that regulators CatR1 and CatR2 modify growth on 2,4-D or 3-CB, probably through modulation of the expression of the ben and tfd genes. The latter assumption is based on that both regulatory proteins are structurally related to TfdR and that both could interact with the respective promoter regions. In this context, a model for the regulation by CatR1 and CatR2 in C. necator is proposed. This model suggests that both regulatory proteins differentially modulates tfd genes, with CatR2 activating both tfd modules and tfdA gene, whereas CatR1 only activates module tfd- I. Based on molecular modelling, it was predicted that differences between CatR1 and CatR2 would be explained by DNA binding affinity and not by inducer binding

Les transporteurs d'ADP/ATP (Ancp) et de phosphate inorganique (PiC) ainsi que la porine VDAC représentent les principaux maillons d'une machinerie de transport d'ADP, d'ATP et de phosphate inorganique à travers la double membrane mitochondrial. Actuellement, les connaissances relatives aux mécanismes moléculaires mis en jeu dans cette machinerie accusent un retard. L'objectif de ce projet de Thèse a été d'amener de nouveaux éléments sur l'état conformationnel de ces protéines étudiées dans la levure Saccharomyces cerevisiae, modèle biologique où la génétique est facilitée. La majeure partie de ce projet a concerné l'Ancp de levure qui assure l'échange d'ADP cytosolique contre de l'ATP matriciel néosynthétisé suite aux processus de phosphorylation oxydative. Il peut être inhibé spécifiquement par deux poisons naturels, le carboxyatractyloside (CATR) et l'acide bongkrékique (BA) qui stabilisent la protéine dans deux états conformationnels distincts adoptés lors du mécanisme de transport. Afin de mieux comprendre cette dynamique, une étude comparative des deux conformères a été réalisée en combinant l'échange hydrogène/deutérium couplé à la spectrométrie de masse (HDX-MS) aux outils biochimiques directement liés à l'étude des Ancp. Les résultats obtenus corrèlent l'hypothèse d'une structure malléable du transporteur dans laquelle des changements conformationnels importants interviennent. D'autre part, ce projet a permis d'obtenir les premières données d'accessibilité au solvant du PiC. Enfin, l'étude en HDX-MS de VDAC, nous a permis de montrer que son accessibilité au solvant en solution de détergent était en accord avec les données structurales publiées récemment
The ADP/ATP (Ancp) and phosphate inorganic (PiC) carriers as well as the VDAC porin catalyze import and export of ADP, ATP and Pi across the mitochondrial membranes. Characterization of this mechanism at the molecular level is not well understood. The main goal of this work concerns the study of the conformational state of these proteins in detergent solution purified from the yeast Saccharomyces cerevisiae where the genetic is easier. The major part of this project concerned the Ancp located in the inner mitochondrial membrane that catalyzes the transmembrane exchange of ADP and ATP between the cytosolic and the matrix compartments. ADP/ATP transport is achieved by interconversion of Ancp between two conformational states that are fixed by the specific transport inhibitors carboxyatractyloside (CATR) and bongkrekic acid (BA). Conformational dynamics of the Ancp was investigated by hydrogen/deuterium exchange coupled to mass spectrometry (HDX-MS). The data were compared to those obtained using molecular and biochemical approaches. Our results suggest a flexible structure of the carrier and a real synergy between each side of the carrier during the transport mechanism of nucleotides. On the other hand, these work provided the first data about the solvent accessibility of PiC. Finally, the study of VDAC in HDX-MS allowed us to show that its solvent accessibility in detergent solution corroborated the structural data published previously

Abstract:
Feral and fearful cats and kittens in animal shelters are not likely to be adopted as companion animals because they emit fearful or aggressive behaviors in the presence of humans. The purpose of the fearful to friendly (F2F) research was to investigate a shaping procedure to increase friendly behaviors of feral and fearful domestic cats and kittens with the goal of achieving animal shelters’ adoptability criteria. The results showed the F2F procedure was a safe and very effective procedure to quickly tame feral kittens deemed unadoptable. The day after implementing F2F, three out of four kittens approached me and accepted petting and holding without any additional training.

Abstract:
During recent years of schizophrenia research, many etiologies have been emphasized, some of them implicating infectious and autoimmune diseases. Many different infectious agents have been examined, but the root seems to stem from the secondary autoimmune deregulation, which can be caused by different infectious agents. Among the effects that autoimmune deregulation has on the body, one prominent effect is on the brain, resulting in either severe or mild encephalitis. The mild encephalitis that has been implicated as one of the causes of schizophrenia-spectrum disorders has been associated with different pathogens, many of which can be transmitted by the household cat. Thus in the present research we have used the schizotypy personality construct model as an analog for schizophrenia spectrum disorders, and the relationship between current schizotypy and childhood household cat interactions were examined. An online questionnaire was completed by 356 undergraduate students and assessed the current schizotypy using the Schizotypal Personality Questionnaire Brief Revised (SPQ-BR), as well as questions about cat ownership and cat bites (puncturing skin) prior to age 13. While no significant relationship was found between childhood cat ownership and current schizotypy, individuals endorsing a cat bite prior to age 13 (N = 66) reported a significantly higher level of current overall schizotypy, which was largely driven by the Disorganized factor of the SPQ-BR.. This relationship should be explored further by examining the antibodies and sera of individuals with the schizophrenia spectrum disorder.

A segunda parte do relatório é uma monografia sobre «Abordagem cat-friendly» na qual é também apresentado um caso clínico. A abordagem cat-friendly inclui o conhecimento do comportamento do gato, técnicas de contenção, realização de procedimentos e dicas para organização do CAMV (centro de atendimento médico veterinário) de modo a que o paciente felino se sinta o mais confortável e menos stressado possível; Abstract: Feline medicine and surgery The present report was written following the externship of the master’s degree in Veterinary Medicine of Universidade de Évora, performed at Hospital do Gato, between September of 2019 and February of 2020. Abstract:
Purpose: To examine how IR-related press releases affect share price for stock companies, and to explore how said companies practice Investor Relations. Methods: The study was conducted using an event study as well as e-interviews. The event study has a quantitative deductive research approach where the market model is used for calculating the abnormal return based on press releases. The e-interviews have a qualitative research approach and follow a semi structured interview guide. The study includes all listed stock companies within the Swedish construction industry and includes press releases from the last decade. Theory: The study is based on the efficient market hypothesis and its semi strong form, theories within Investor Relations and previous research. Results: The event study shows a significant negative abnormal return during a four day period starting the day after the press release event. The negative abnormal return could derive from investor relations being used by companies as damage control, or that the investors’ expectations are too high as a result of IR. No significant differences in how companies practice IR was found in e-interviews. To the contrary, companies show similar tendencies in IR practice. It can be concluded that stock companies, through investor relations, are able to affect their share price.
Syfte: Att undersöka hur IR-relaterade pressmeddelanden påverkar aktiekursen hos börsnoterade företag samt att undersöka hur företagen arbetar med Investor Relations. Metod: Studien är utförd med hjälp av en eventstudie tillsammans med e-intervjuer. Eventstudien har en kvantitativ deduktiv ansats där marknadsmodellen används för beräkning av avvikande avkastning baserat på pressmeddelanden. E-intervjuerna har en kvalitativ ansats och följer en semistrukturerad intervjuguide. Studien innefattar alla börsnoterade företag i den svenska byggindustrin och pressmeddelanden från det senaste decenniet. Teori: Den effektiva marknadshypotesen och dess halvstarka form, teorier inom investor relations tillsammans med tidigare genomförda forskning har legat till grund för studien. Resultat: Resultatet från eventstudien visar på en signifikant negativ avvikande avkastning med start dagen efter händelsedagen till och med fyra dagar efter händelsedagen. Den negativa avvikande avkastningen kan bero på att arbetet med IR försöker dämpa befintlig negativitet hos investerare, alternativt kan det bero på att investerares förväntningar är för höga på grund av positiv IR. Utmärkande strategier för hur IR-arbetet går till i praktiken bland de observerade företagen har inte funnits i e-intervjuerna, istället liknar företagens IR-arbete varandra i stort. Slutsatsen dras att börsnoterade företag kan påverka sin aktiekurs med hjälp av investor relations.

Abstract:
Every day a large numbers of transactions occur by people with different backgrounds. Insiders’ are a part of them and are considered to have an insight in companies that is not accessible to outsiders. This affects the market conditions for the participants when trading stocks, where individual participants regularly have the possibility to earn abnormal returns at the expense of others. Although Sweden, Germany and the United States continually keep developing the insider trading regulation, research shows that insiders’ still have the ability to earn abnormal returns. The difference in insiders’ ability to earn abnormal returns is believed to be because of that the countries are from different legal families, which in turn influence the market conditions where the insider trading occurs. The purpose is to examine whether legal insider trading generates abnormal return relative to index and analyze whether there are differences between the studied countries and what the differences may be due. Method: The study takes ledge in previous research using a quantitative method where 90 sorted companies in Sweden, Germany and the U.S. are used to gather insider transactions between 2006 and 2012. The data is then analyzed with statistical methods to find answers to the purpose of the study. The results shows that insiders have abnormal return for both the buy and sell transactions, which prove that there is an information gap between insiders and external stakeholders. The legal origin may have an effect on the relationship when it is linked to insiders’ abnormal return through ownership concentration and accounting quality, which in turn affects insiders’ ability to generate abnormal returns. The insiders’ position has also shown to have an impact on the abnormal returns, but is not completely consistent with the theory of information hierarchy.
Det sker varje dag stora mängder av transaktioner av personer med skilda bakgrunder. Insiders är en del av dem och anses ha insyn i företag som inte är tillgänglig för utomstående. Detta påverkar förutsättningarna för marknadens aktörer vid aktiehandel där enskilda aktörer regelbundet kan tjäna på bekostnaden av andra. Sverige, Tyskland och USA utvecklar kontinuerligt regleringen för insiderhandeln men tidigare forskning visar, trots harmonisering i lagstiftningen, på skillnader mellan länderna. Detta antas bero på att länderna härstammar från olika legala familjer, vilket påverkar förutsättningarna på ländernas respektive marknader, och därför påverkar insiders förmåga till att skapa överavkastning.  Syfte är att undersöka om laglig insiderhandel genererar överavkastning jämfört med index och analysera om det finns skillnader mellan de undersökta länderna och vad skillnaderna kan bero på. Studien tar avsats i tidigare forskning och använder en kvantitativ metod där 90 utsorterade bolag i Sverige, Tyskland och USA används för att inhämta insidertransaktioner mellan 2006 och 2012. Materialet analyseras sedan med hjälp av statistiska verktyg för att få svar på arbetes syfte. Resultatet är att insiders har överavkastning för både köp och säljtransaktioner och bevisar att det finns ett informationsgap mellan insiders och utomstående intressenter. Det legala ursprunget kan ha en påverkan på förhållandet då det kopplas till insiders överavkastning genom ägarkoncentrationen och redovisningskvalitén som i sin tur påverkar insiderns förmåga till att skapa överavkastning. Insiderns position visas också ha en påverkan på överavkastningen där en högre position ger högre överavkastning, vilket går i linje med informationshierarkin.

Sensory neocortex contains a significant number of inhibitory neurons that use gamma-aminobutyric acid (GABA) as their neurotransmitter. Functional roles for these neurons have been identified in physiological studies. For example, in primary somatosensory cortex (SI), blockade of GABAa receptors with bicuculline leads to expansion of receptive fields (RFs). The magnitude of RF enlargement varies between SIpopulations of GABAergic neurons were identified by labeling specific calcium binding proteins.

This paper details the making of Stray Cats, a University of New Orleans thesis film. It examines the process of creating the film in three parts. Part one will detail the pre-production, including the conceptualization, writing, and preparation for the project. Part two will cover the production, including the directing, production design, cinematography, and on-set operations. Part three will describe the post-production process, including the editing, color correction, sound design, and musical score. These three components will then be referenced in regards to the theme of the film, and will conclude by analyzing whether or not the final film achieves the initial vision.

Parathyroid hormone (PTH) secretion maintains free-ionised extracellular calcium (Ca2+o) homeostasis under the control of the calcium-sensing receptor (CaR). In humans and dogs, blood acidosis and alkalosis is associated with increased or suppressed PTH secretion respectively. Furthermore, large (1.0 pH unit) changes in extracellular pH (pHo) alter Ca2+o sensitivity of the CaR in CaR-transfected HEK-293 cells (CaR-HEK). Indeed, it has been found in this laboratory that even pathophysiological acidosis (pH 7.2) renders CaR less sensitive to Ca2+o while pathophysiological alkalosis (pH 7.6) increases its Ca2+o sensitivity, both in CaR-HEK and parathyroid cells. If true in vivo, then CaR’s pHo sensitivity might represent a mechanistic link between metabolic acidosis and hyperparathyroidism in ageing and renal disease. However, in acidosis one might speculate that the additional H+ could displace Ca2+ bound to plasma albumin, thus increasing free-Ca2+ concentration and so compensating for the decreased CaR responsiveness. Therefore, I first demonstrated that a physiologically-relevant concentration of albumin (5% w/v) failed to overcome the inhibitory effect of pH 7.2 or stimulatory effect of pH 7.6 on CaR-induced intracellular Ca2+ (Ca2+i) mobilisation. Determining the molecular basis of CaR pHo sensitivity would help explain cationic activation of CaR and permit the generation of experimental CaR models that specifically lack pHo sensitivity. With extracellular histidine and free cysteine residues the most likely candidates for pHo sensing (given their sidechains’ pK values), all 17 such CaR residues were mutated to non-ionisable residues. However, none of the resulting CaR mutants exhibited significantly decreased CaR pHo sensitivity. Even co-mutation of the two residues whose individual mutation appeared to elicit modest reductions (CaRH429V and CaRH495V) failed to exhibit any change in CaR pHo sensitivity. I conclude therefore, that neither extracellular histidine nor free cysteine residues account for CaR pHo sensitivity. Next, it is known that cytosolic cAMP drives PTH secretion in vivo and that cAMP potentiates Ca2+o-induced Ca2+i mobilisation in CaR-HEK cells. Given the physiological importance of tightly controlled PTH secretion and Ca2+o homeostasis, here I investigated the influence of cAMP on CaR signalling in CaR-HEK cells. Agents that increase cytosolic cAMP levels such as forskolin and isoproterenol potentiated Ca2+o-induced Ca2+i mobilisation and lowered the Ca2+o threshold for Ca2+i mobilisation. Indeed, forskolin lowered the EC50 for Ca2+o on CaR (2.3 ± 0.1 vs. 3.0 ± 0.1 mM control, P<0.001). Forskolin also potentiated CaR-induced ERK phosphorylation; however protein kinase A activation appeared uninvolved in any of these effects. Pertussis toxin, used to block CaR-induced suppression of cAMP accumulation, also lowered the Ca2+o threshold for Ca2+i mobilisation though appeared to do so by increasing efficacy (Emax). Furthermore, mutation of the CaR’s two putative PKA consensus sequences (CaRS899 and CaRS900) to a non-phosphorylatable residue (alanine) failed to alter the potency of Ca2+o for CaR or attenuate the forskolin response. In contrast, phosphomimetic mutation of CaRS899 (to aspartate) did increase CaR sensitivity to Ca2+o. Together this suggests that PKA-mediated CaRS899 phosphorylation could potentiate CaR activity but that this does not occur following Ca2+o treatment in CaR-HEK cells. Together, these data show that cAMP regulates the Ca2+o threshold for Ca2+i mobilisation, thus helping to explain differential efficacy between CaR downstream signals. If true in vivo, this could help explain how multiple physiological signal inputs may be integrated in parathyroid cells.

This is a product development project which has developed a prototype in order to facilitate for people using mobility devices when they want to buy a larger quantity of groceries. The prototype is a shopping cart composed with the upper part of a walker.Today there are many different types of mobility devices used by elderly and people with physical impairment. According to Myndigheten för samhällsskydd och beredskap (MSB) are accidental falls the most common cause of accidents in Sweden.This project has been using Computer Aided Design (CAD) as a tool to create a visual image of the prototype. The advantage of this is to modify the functionality and performance of the prototype on the computer before the modification takes place in the real model. Using human manikins, the CAD model was rendered in natural surroundings to get as true a picture as possible. The prototype is adjustable, why various anthropometric measurements were used to fit to the individual, but not less than the 40th percentile for women, corresponding to a height of 161 cm.The project has resulted in a CAD model and a functional prototype. The prototype has been tested by a user group at Alla Hjärtans Hus (AHH) and received positive feedback. The prototype also received constructive criticism from users, so suggestions can be applied. Suggestions for further development of the prototype have been developed. The prototype needs to be modified and components must be added before it can be used in grocery retailers.

A presente dissertação pretende descrever e analisar as potencialidades futuras oferecidas por sistemas CATV operando serviços de comunicação iterativos e de faixa larga. Neste contexto, visando determinar as configurações mais adequadas para a parte óptica da rede, são analisadas diversas alternativas de implementação envolvendo os dispositivos constituintes básicos: laser, modulador externo, EDFA, e fibra, sendo neste último caso, tanto a utilização de fibra padrão quanto a de dispersão deslocada. Dentro do espírito de serviços de faixa larga, implementa- se em laboratório um sistema WDM com duas portadoras ópticas. A primeira, em 1310 nm, é modulada em intensidade com 2.48 Gbits/s. a segunda, em 1550nm, é modulada em AM- VSB. As interferências mútuas são então medidas e confrontadas com resultados previstos teoricamente, validando este último modelo. A formulação teórica permite então prever a deterioração mutua em um WDM com duas portadoras digitais em 2.48Gbit/s. Os resultados obtidos sugerem a viabilidade deste último arranjo para portar vídeo digitalizado. Dando seqüência, são apresentadas novas soluções para compressão de vídeo, técnicas de modulação e tecnologias de transporte. Finalmente, os resultados obtidos anteriormente, tanto os práticos quanto os teóricos e os simulados, são usados para propor novas arquiteturas de redes de CATV explicitando as sugestões para o caso de redes HFC (preferencialmente destinado a operadoras de TV a cabo) e também para configurações FTTC, estas últimas, preferencialmente destinadas às operadoras telefônicas.
This work is aiming at discussing the future possibilities the CATV systems are likely to offer, concerning iterative communications and broadband services. In what concerns the system optical section, a search is carried out in order to reach suitable arrangements and configurations concerning the basic used components, namely: laser, external modulator, EDFA and the optical fiber. Both standard and dispersion shifted fiber utilization are considered. Considering broadband utilization, a practical experiment is carried out. A two carrier WDM system is tested in the laboratory. The first carrier - 1310 nm - is digitally modulated at 2.48 Gbit/s, while the second one - 1550 nm - carries an AM-VSB modulation. Crosstalk is measured and the obtained results are compared with those from a proposed theoretical model. A satisfactory agreement validates the model. Next, the same model is then used for predicting crosstalk level in another two carrier WDM, however now, with both carriers being digitally modulated at 2.48 Gbit/s. the obtained results suggest that this last arrangement is worthwhile to be used for efficiently carrying digital video. A discussion on new solutions for video compression, modulation formats and transport techniques is follwed. The work is closed with a number of new CATV network solutions being suggested. Previously obtained results and performed suggestions are used for achieving two basiv lines of applications. The first one is based on HFC arrangements, mainly conceived for cable TV operators. The second on is based on FTTH arrangements, being this last suggestion better suited for traditional telephone companies.

Obesidade na clínica de felinos é um problema frequente e tornou-se motivo de grande preocupação para a medicina veterinária. A obesidade é definida como acúmulo excessivo de tecido corporal adiposo. O objetivo do presente estudo observacional prospectivo foi determinar os principais fatores de riscos relacionados ao excesso de peso em gatos com ênfase na interação tutor-gato. Para tanto, foram selecionados gatos, divididos em dois grupos, a saber: grupo dos gatos obesos (n=18) nesse grupo foram incluídos gatos com escore de condição corporal entre 8 e 9; grupo dos gatos não obesos (n=18) com escore de condição corporal 5. Foram excluídos do estudo os gatos que apresentaram outras comorbidades, além da obesidade. A pesquisa dos prováveis fatores de risco relacionados a obesidade foi realizada mediante a utilização de um questionário respondido por todos os tutores dos 36 gatos. Nenhuma das variáveis qualitativas estudadas nos aspectos demográficos dos gatos obesos e não obesos demonstrou significância, ou seja, não houve associação entre obesidade e o sexo, o status sexual, as raças e o tamanho do pelame. Assim como, nenhuma das variáveis qualitativas analisadas nos aspectos demográficos dos tutores de gatos obesos e não obesos demonstrou significância; Não houve relação entre obesidade e o sexo do tutor, ao estado civil, a profissão (inclusive os clínicos veterinários), a escolaridade e a renda média familiar dos tutores. Com relação aos aspectos antrozoológicos, contatou-se que ambos os grupos têm uma relação estreita de afeto com seus tutores, contudo a porcentagem do grau de afeto tende a ser maior no grupo obeso, apesar de não ter sido evidenciada diferença estatística. Sobre os aspectos ambientais, oito variáveis foram presumivelmente consideradas importantes na manutenção e no controle do peso dos gatos. Constatou-se que felinos do grupo não obeso costumam brincar sozinhos e com outros animais, enquanto que gatos obesos são menos interativos. Ainda, o enriquecimento ambiental, assim como o manejo alimentar com dieta úmida influenciaram na condição física desses gatos. Constatou-se que o número de felinos não obesos que recebe ração úmida é bem maior que o daqueles pacientes com excesso de peso. Finalmente, o escore de condição corporal atribuído pelos tutores de felinos com excesso de peso tende a ser subestimado quando comparado ao do profissional especializado. Os principais riscos ocasionados pelo excesso de peso identificados neste estudo foram o tipo de alimentação oferecido, principalmente dietas secas, e a inatividade física. Apesar de os aspectos antrozoológicos não demonstrarem valor estatístico, o grau de afeto tende a ser levemente superior quando se analisa valores percentuais.
Feline obesity is a frequent problem and has become a major concern in veterinary medicine. Obesity is defined as excessive accumulation of body fat. The purpose of this prospective observational clinical study was to determine the main risk factors related to obesity in cats with emphasis on owner-cat interaction. Thus, the studied cats were divided into two groups: obese cats (n=18) with a body condition score between 8 and 9; and non-obese cats (n=18) with a body condition score of 5. All cats were healthy at the time of the study was conducted and cats with any other disease, besides obesity were excluded. The study of the risk factors related to obesity was performed using a questionnaire answered by all owners of the 36 cats. None of the qualitative variables in demographic aspects of obese and non-obese cats showed significance, and there was no association between obesity and gender, sexual status, race and coat size. Also, none of the qualitative variables in demographic aspects of the owners of obese cats and non-obese cats showed significance. Besides that, there was no relationship between obesity and the gender of the cat owner, marital status, profession (including veterinarians), schooling and the average family income of the owner. Regarding the antrozoological aspects, we concluded that both groups of cats a close affectionate relationship with their owners, but the percentage of the degree of affection tends to be higher in the obese group, despite there being no statistical difference. Considering the environmental aspects, eight variables were presumably important in maintaining and controlling the weight of cats. It was found that non obese cats usually play alone and with other cats, while obese cats were less interactive. Moreover, environmental enrichment as long as the moisture of the diet influenced the physical condition of these cats. Non obese cats usually had more moist food than obese cats. Finally, the body condition score given by the owners of obese cats tends to be underestimated when compared to the experts score. The main risks to the overweight findings in this study were type of food offered, especially dry diets and the physical inactivity. Also, despite the anthrozoological aspects evaluated have not shown any significance as risk factors for obesity in cats, the degree of affection between owner-cat tend to be slightly higher in obese cats group.

This dissertation study describes and presents an alternative method for delivering digital video, the MPEG-4 compression format, through the use of an analog cable network for direct viewing on television sets. The purpose of the study is to propose this technology as an alternative to the present cable network technologies in Uruguay and elsewhere in South America. Delivery standards that might be used over such a CATV network include DVB-C (digital video broadcast for cable), and IP (internet protocol) networks. The study compares the current technologies to determine which would be the best way to provide such delivery over CATV networks. It is the argument of this study that the delivery of MPEG-4 format streaming video is no different than the delivery of any type of digital signal: in order for the signal to be delivered, it must pass through a transmission medium such as fiber-optic cable, twisted-wire pair, coaxial cable, etc. The dissertation makes the case that MPEG-4 is a feasible alternative technology which should be considered for new CATV applications in this region. This study is from a technical point of view, when it comes to implementation there are other elements that should be taken into account.

Postural control incorporates multiple neural and mechanical systems at various levels of the motor control system, yet the question of how all these systems interact remains unanswered. This dissertation describes development of a biologically based, three-dimensional mathematical model of the forelimb of the domestic cat that integrates skeletal anatomy, muscular architecture, and neural control. Previous work has shown that muscle architecture profoundly affects its function. However, even though the forelimbs of quadrupeds contribute to posture and locomotion differently from hindlimbs, most models of quadruped motion are based upon hindlimb mechanics. The proposed work consists of three main steps: (1) architectural and anatomical characterization, which involves acquisition of muscle attachment data, measurement of whole muscle and muscle fiber properties, and estimation of limb kinematic parameters; (2) model development and implementation, wherein the data will be integrated into a mathematical model using special-purpose software; and (3) model validation, including verification of model estimates against experimentally obtained measurements of muscle moment arms, and prediction of limb kinetics, namely end-point forces arising from perturbations to the limb. It was found that the forelimb does indeed possess structure, particularly at the shoulder and antebrachium, that allows for more diverse movements. The neural wiring in these regions is more complex than in the hindlimb, and there exists substantial muscular structure in place for non-sagittal motion and object suppression and retrieval. Other results showed that the kinematics of the limb alone produce a restorative response to postural disturbance but that the magnitude is reduced, indicating that neural input acts as a modulatory influence on top of the intrinsic mechanism of limb architecture. Furthermore the model demonstrated many of the essential features found in the experiments. This study represents the implementation of the first forelimb model of the cat incorporating mechanical properties and serves as a key component of a full quadruped model to explore posture and locomotion.

Cat ownership in Australia is declining compared to an increasing trend of cat ownership in the United Kingdom, United States and Europe. The decline in Australia may be linked to concerns over perceived impacts of cat predation and an associated dislike of cats. However, while there are numerous studies on feral cats and their impacts on declining native fauna, the impact of pet cats on suburban wildlife or fauna in remnant bushland is relatively unknown although there is a wide perception of risk. The primary aim of this thesis was to apply the precautionary principle to the question of the putative impact of pet cats on the abundance and diversity of small mammals in urban bushland adjacent to low-density suburbia in the City of Armadale, a municipality on the south-east fringe of Perth, Western Australia. At the time of writing, Western Australia is yet to introduce state legislation governing cat control although many local councils within the state have either implemented or are in the process of implementing cat regulations. The precautionary principle was deemed an ideal approach to this question, because it provides a rationale for deciding on possible actions where both the potential risk to environmental values and the uncertainty about possible impacts are high. In such cases the precautionary principle requires two broad lines of action: firstly, detailed consultation with stakeholders to determine their perceptions of risk and the actions they are prepared to take to reduce it and, secondly, research to reduce uncertainty. With regard to stakeholder consultation, local residents were surveyed in regard to their attitudes and current cat husbandry practices. A substantial proportion of respondents within this municipality believed cat regulations were necessary (75% of owners and 95% of non-owners). At least 70% of both owners and non-owners agreed with the propositions that cats not owned by licensed breeders should be desexed, local councils should restrict the maximum number of cats that can be owned on one property and that pet cats entering nature reserves are harmful to wildlife. Most (c.85%) cat owners agreed that they would license their cats if that became compulsory. Although fewer owners (c.60%) were prepared to keep their cats on their property at all times to protect wildlife, over 80% were willing to confine their cats at night if it was required. Owners seemed to be substantially motivated by the value of these measures in reducing injury to cats and facilitating the return of lost animals rather than concern over wildlife protection. Attempts to reduce uncertainty involved (i) assessing roaming patterns of pet cats to determine the sizes of appropriate buffer zones around nature reserves, and (ii) determining species diversity, species richness and abundance of small mammals in remnant bushland adjacent to sub-divisions with varying regulations governing cat husbandry. Radio tracking results to assess cat roaming patterns showed substantial variation in home range size between cats in high density suburbia (ranged between 0.01 ha - 0.64 ha) and those in low density suburbia (ranged from 0.07 ha - 2.86ha). Larger home range sizes of cats in the rural areas (up to 2.9 ha) suggest buffer zones of up to 500 metres around nature reserves are needed to exclude almost all roaming cats. The abundance and species richness of small mammals were investigated in four areas of remnant bushland. Two were adjacent to subdivisions where cat ownership was unrestricted, one next to a subdivision where cat ownership was prohibited and the remaining one next to a subdivision where compulsory night curfew and bells on pet cats were enforced. No definitive evidence of predatory impact by pet cats on the small mammals was found. Mammal species diversity was not significantly different between sites and species richness and absolute abundance were not higher in sites where cats were restricted. Vegetation comparisons showed significant differences in the structure and species composition of the vegetation between most sites and the mammal species richness and abundance appeared linked to ground cover density in the various sites. This factor, not cat restrictions, appeared to be the primary determinant of species richness, species diversity and absolute numbers of small mammals in these sites. This study in the City of Armadale has shown that the implementation of proposed cat legislation must have a 'whole of ecosystem' approach, i.e. protecting identified remnant bushland containing biodiversity from threatening processes such as plant disease and inappropriate fire, especially arson, as well as possible predations from pet cats. Habitat restoration and protection may be more important conservation activities than regulation of cats. Regulation of cats can be done at differing levels of intensity and cost, bearing in mind that this community is receptive to regulation of some aspects of cat ownership. Community education on the values of cat confinement in regards to cat welfare might increase chances of compliance.

However, while there are numerous studies on feral cats and their impacts on declining native fauna, the impact of pet cats on suburban wildlife or fauna in remnant bushland is relatively unknown although there is a wide perception of risk. The primary aim of this thesis was to apply the precautionary principle to the question of the putative impact of pet cats on the abundance and diversity of small mammals in urban bushland adjacent to low-density suburbia in the City of Armadale, a municipality on the south-east fringe of Perth, Western Australia. At the time of writing, Western Australia is yet to introduce state legislation governing cat control although many local councils within the state have either implemented or are in the process of implementing cat regulations. The precautionary principle was deemed an ideal approach to this question, because it provides a rationale for deciding on possible actions where both the potential risk to environmental values and the uncertainty about possible impacts are high. In such cases the precautionary principle requires two broad lines of action: firstly, detailed consultation with stakeholders to determine their perceptions of risk and the actions they are prepared to take to reduce it and, secondly, research to reduce uncertainty. With regard to stakeholder consultation, local residents were surveyed in regard to their attitudes and current cat husbandry practices. A substantial proportion of respondents within this municipality believed cat regulations were necessary (75% of owners and 95% of non-owners). At least 70% of both owners and non-owners agreed with the propositions that cats not owned by licensed breeders should be desexed, local councils should restrict the maximum number of cats that can be owned on one property and that pet cats entering nature reserves are harmful to wildlife. Most (c.85%) cat owners agreed that they would license their cats if that became compulsory. Although fewer owners (c.60%) were prepared to keep their cats on their property at all times to protect wildlife, over 80% were willing to confine their cats at night if it was required. Owners seemed to be substantially motivated by the value of these measures in reducing injury to cats and facilitating the return of lost animals rather than concern over wildlife protection. Attempts to reduce uncertainty involved (i) assessing roaming patterns of pet cats to determine the sizes of appropriate buffer zones around nature reserves, and (ii) determining species diversity, species richness and abundance of small mammals in remnant bushland adjacent to sub-divisions with varying regulations governing cat husbandry. Radio tracking results to assess cat roaming patterns showed substantial variation in home range size between cats in high density suburbia (ranged between 0.01 ha - 0.64 ha) and those in low density suburbia (ranged from 0.07 ha - 2.86ha). Larger home range sizes of cats in the rural areas (up to 2.9 ha) suggest buffer zones of up to 500 metres around nature reserves are needed to exclude almost all roaming cats. The abundance and species richness of small mammals were investigated in four areas of remnant bushland. Two were adjacent to subdivisions where cat ownership was unrestricted, one next to a subdivision where cat ownership was prohibited and the remaining one next to a subdivision where compulsory night curfew and bells on pet cats were enforced. No definitive evidence of predatory impact by pet cats on the small mammals was found. Mammal species diversity was not significantly different between sites and species richness and absolute abundance were not higher in sites where cats were restricted. Vegetation comparisons showed significant differences in the structure and species composition of the vegetation between most sites and the mammal species richness and abundance appeared linked to ground cover density in the various sites. This factor, not cat restrictions, appeared to be the primary determinant of species richness, species diversity and absolute numbers of small mammals in these sites. This study in the City of Armadale has shown that the implementation of proposed cat legislation must have a 'whole of ecosystem' approach, i.e. protecting identified remnant bushland containing biodiversity from threatening processes such as plant disease and inappropriate fire, especially arson, as well as possible predations from pet cats. Habitat restoration and protection may be more important conservation activities than regulation of cats. Regulation of cats can be done at differing levels of intensity and cost, bearing in mind that this community is receptive to regulation of some aspects of cat ownership. Community education on the values of cat confinement in regards to cat welfare might increase chances of compliance.

El uso de este servicio apuntará a mejorar la calidad de vida de la población que podrá tener acceso a éste. Con la implementación de este proyecto se espera dar servicio y a la vez educar a la población sobre la importancia de la salud bucal, conjuntamente, se pretende incentivar el uso de estos servicios móviles dentales por el Estado de Chile, liberando recursos para lograr ampliar la cobertura de atención primaria y de urgencia, y por ende impulsar un desarrollo sustentable como país. El Objetivo de esta evaluación de proyecto nace a raíz de varios motivos: Primero, la necesidad de sumar alternativas no convencionales y de calidad en atención primaria. La seguridad en las personas es un tema común en nuestra sociedad, más aun cuando en los últimos años ha aumentado los robos de vehículos a través del “portonazo” no solo en el barrio alto, sino que en todo el país, afectando a todo tipo de familias. Durante el año 2016 este crimen se ha trasladado a comunas emergentes, dejando inseguros a gran parte de la población que cuenta con uno o más automóviles particulares, un 5,9% de ellos fue víctima de intento de robo o hurto de vehículos durante los últimos doce meses, una cifra no menor considerando el parque automotriz de RM es 1,8M vehículos particulares equivalente a 40% del parque total nacional (fuente INE 2015). El año 2015 las autoridades informaron que entre 4 y 5 autos eran robados cada 1 hora (102 autos por día en Chile) con una tasa de crecimiento de 4,2% respecto del 2014 (fuente Carabineros de Chile – La Tercera). Este alto crecimiento en los robos de vehículos particulares ha generado una industria en este tipo de delincuencia que supera los US $200M anuales, con más de 35.000 vehículos robados anualmente. Sabemos que a nivel mundial, la industria automotriz y la seguridad satelital han diseñados aparatos de rastreo en tiempo real para vehículos de alta gama, para la industria militar y las de uso estándar aplicada fuertemente en flotas. Entendiendo este contexto y dado el crecimiento en robo de vehículos particular, más la alta inseguridad ciudadana instalada, hemos visualizado una oportunidad de negocio para nuestro producto Car Recovery, el que ofrece monitorear y recuperar vehículos robados en tiempo real, a través de seguimiento en línea por un equipo especializado en vigilancia, acompañado de una plataforma robusta de operación 24/7, nuestra ventaja competitiva, es el equipo de rescatistas que acudirán motorizados al recupero del vehículo particular, mientras en paralelo nuestros operadores telefónicos se pondrán en contacto con seguridad ciudadana y Carabineros. Abstract:

The final goal of this document is the construction of multi-band terminal for the CVIS project to allow the communication among cars and between the cars and the roadside infrastructure. For the construction of this multi-band terminal, this document takes as starting point, a new compact disk antenna described in [1]. It consists in a circular patch antenna shorted by a central metallic post. This allows reducing the dimensions of a classical circular patch antenna so it is very useful for our application given that the terminal is going to be place on the top of a car and it should be small enough to be attractive for the final users. Specifically, we are going to analyze the behaviour of this kind of antenna when it radiates in the TM01 mode, because the radiation pattern of this mode is particularly interesting for the applications of the CVIS project. This document is divided into two main parts. In the first one, a study in depth of the behaviour of the antenna proposed in [1] was performed. Firstly, through an analytical model and afterward with the help of two simulation tools (WIPL-D and EMDS), we analyze the influence of the main parameters of the antenna (outer and inner radiuses, height, electrical permittivity and the position of the feed) on its properties (resonant frequency, bandwidth, entrance impedance, the shape of the radiation pattern and so on). A general methodology for the design of this kind of antennas was proposed, and it was put in practice with the design of a prototype for a band around 2 GHz. In addition to the conclusions about the influence of the different parameters of the antenna, another important conclusion was done. It was discovered that the use of the simulation tool WIPL-D Lite was not suitable for the simulation of this kind of antenna. In the second part, this document tackles the construction of a dual-frequency antenna for the bands of 2.4-2.484 GHz and 5.75-5.95 GHz. For this, the present document studies the possibility of stacking two compact circular patch antennas, so we put the one which covers the higher band (the smaller) on the top of the one which covers the lower band (the lager). The two patch antennas have a coaxial feed and the feed of the upper antenna goes inside the central post of the lower antenna to minimize the influence of it on the radiation pattern of the lower antenna. This proposal works out not to be feasible, because we need a lower antenna with a large inner radius to allow the variation of the feed position of the upper antenna in a wide range. This is necessary to get a good matching for the upper antenna. The problem is that the radiation pattern becomes very asymmetric when we increase the inner radius of the patch antenna. To solve that, two alternative were analyzed in this document. The first consists in putting a second feed symmetrically placed with respect to the central post. It leads to a more symmetric radiation pattern so we can choose a larger inner radius. In addition, the introduction of the second feed increases the bandwidth of the antenna. The second alternative is a simplification of the first one. It consists in replacing the upper antenna with a monopole on the top of the lower antenna. It is simpler but it prevents the possibility of stacking other patch antennas to cover more frequency bands in a future. Due to this disadvantage, the first alternative was chosen. Finally, a proposal based on the first alternative, which fulfils quite well all the requirements which were raised in the wording of this master thesis, was presented and studied in depth.

Abstract:
Since the introduction of the lipid emulsion formulation in 1986, propofol has become established for induction as well as for maintenance of anaesthesia in veterinary practice1 including cats2;3-8. Propofol is rapidly metabolized by hepatic glucuronidation in most species and it has also been shown to undergo extrahepatic metabolism9-13, so that total body clearance may exceed liver blood flow in certain species. Because of their highly carniverous diet, cats are little exposed to antiherbivory compounds so that they have become deficient in UGP-glucuronosyltransferase (UGT)14. Consequently, a number of drugs are eliminated slowly15;16, often giving rise to prolonged half-lives of the parent drugs. Cats are therefore sensitive to the adverse effects of many drugs and toxins that are normally glucuronidated before elimination. It is therefore likely that the disposition of propofol may differ markedly from that of humans and other animal species17. Adam et al18 reported that for the cremophor propofol formulation in cats, volumes of distribution were smaller and elimination halflives were longer than those of pigs, rats and rabbits. In addition, pulmonary uptake has been demonstrated to occur in cats,19 however propofol’s pharmacokinetics have not been studied formally. The purpose of this study was to determine the pharmacokinetic behaviour of propofol after single intravenous injections. In comparison with man, the apparent central volume of distribution in domestic cats is small (0.56L.kg-1 body weight vs. 0.228L.kg-1) for the human pharmacokinetic parameter set of Marsh et al20 and the clearance (0.0086 L.kg-1.min-1 vs. 0.027 L.kg- 1.min-1) is approximately 2½ times slower in cats when compared with humans. Slow clearance should not influence recovery from anaesthesia following standard induction doses, because the early decreases in blood concentrations are predominantly due to redistribution of drug to various tissues (similar to the disposition of thiopentone which exhibits a slow total body clearance21. However it is possible that drug may accumulate within the body after prolonged infusions, resulting in delayed recovery times. This phenomenon is best described by calculating “context-sensitive” decrement-times by computer simulation22-24. Computer software♣ were used to calculate the 20%, 50% and 80% context-sensitive decrement times for the cat pharmacokinetic model. For comparative purposes, similar calculations were performed for an adult human male (weight 70 kg) using the pharmacokinetic parameter-set of Marsh et al20. Assuming that recovery from anaesthesia occurs after a 50% decrease in blood concentrations has taken place, it is apparent from the 50% context-senstive decrement-time graph that for infusions lasting up to 20 minutes (during which concentrations are kept constant), recovery can be expected to be rapid and predictable. However if infusions are administered for longer than 20 minutes, the recovery times of the “average” cat increase rapidly, reaching a plateau of 36 minutes, while recovery times of the human remain short, albeit increasing slowly. Awakening times become dramatically prolonged and unpredictable in both cats and humans if propofol concentrations are required to decrease by 80% for recovery to occur. Under these circumstances the 80% decrement time after a two-hour infusion is approximately two hours in cats and 45 minutes in humans. On the other hand, if dosing is conservative, so that blood concentrations need to decrease by only 20% for awakening to occur, then recovery times are short and predictable, being only a few minutes, regardless of the duration of the preceding infusion. These findings are in accordance with those of Pascoe et al25 who reported that cats took longer to recover after a short (30 min) infusion than after a long (150 min) infusion. In their crossover study, the propofol infusion rates were adjusted so that the cats were maintained at a light level of anaesthesia at which they responded sluggishly to pedal stimulation. It is therefore likely that propofol concentrations were kept steady and were similar during the 30-minute as well as during the 150-minute infusions. Delayed recovery has also been reported when propofol was administered to cats on consecutive days26. Conclusions and clinical relevance: We recommend that propofol infusions be administered to cats only for fairly short procedures and that for prolonged surgery, maintenance of anaesthesia should be accomplished using other drugs. Background: Hyperthyroidism has substantial effects on the circulatory system. The cardiac biomarkers NT-proBNP and troponin I (cTNI) have proven useful in identifying cats with myocardial disease but have not been as extensively investigated in hyperthyroidism.
Hypothesis: Plasma NT-proBNP and cTNI concentrations are higher in cats with primary cardiac disease than in cats with hyperthyroidism and higher in cats with hyperthyroidism than in healthy control cats.

Animals: Twenty-three hyperthyroid cats, 19 cats with HCM without congestive heart failure, and 19 euthyroid, normotensive healthy cats eight years of age or older. Fourteen of the hyperthyroid cats were re-evaluated three months after administration of 131I.

Methods: A complete history, physical examination, complete blood count, serum biochemistries, urinalysis, blood pressure measurement, serum T4 concentration, plasma concentrations of NT-proBNP and cTNI, and echocardiogram was prospectively obtained from each cat.

Results: Hyperthyroid and HCM cats had plasma NT-proBNP and cTNI concentrations that were significantly greater than healthy older cats, but there was no significant difference between hyperthyroid and HCM cats with respect to concentration of either biomarker. In hyperthyroid cats that were re-evaluated three months after 131I treatment, plasma NT-proBNP and cTNI concentrations as well as ventricular wall thickness decreased.

Conclusions and Clinical Relevance: Although there may be a role for NT-proBNP in monitoring the cardiac response to treatment of hyperthyroidism, neither NT-proBNP nor cTNI can be used to distinguish hyperthyroid cats from cats with HCM. Therefore, the thyroid status of older cats should be ascertained prior to interpreting results of cardiac biomarker testing.

The aim of this thesis is to explore how roles, identities and hierarchies of age and gender are reproduced or transformed through power relations, within the space of the family car. This study encompasses a detailed analysis of the mechanisms of power within family car space. The theoretical approach is informed by relational conceptualisations of power, space and age, and by post-structuralist concepts of the performative nature of gender. The central argument is that social categories of gender and age are produced and re-created relationally through the particular discursive practices of power which are reiteratively performed within family car space. In some cases, these practices are enabled through the appropriation of the resources of in-car technology, which allows adults to control the aural and physical spaces of the family car.

Normal levels of CO₂ are responsible for the maintenance of approximately 30% of sympathetic neurogenic vascular tone in intact cats. The central medullary chemoreceptors have been implicated as the major source of this CO₂ dependent neurogenic vascular tone. However, it is possible that spinal cord CO₂ sensitivity could also have mediated a portion of neurogenic vascular tone. Cats with acute and chronic spinal transections can maintain near normal levels of systemic arterial blood pressure, and show cardiovascular and sympathetic reflex changes in response to a variety of stimuli. Thus, it seemed likely that the acute spinal cat could exhibit the spinal component of CO₂ mediated sympathetic neurogenic vascular tone. Therefore the effects of systemic CO₂ increases on the perfusion pressures of vascularly isolated hindlimbs autoperfused at constant flow (as an indication of vascular resistance and sympathetic vascular tone) were studied in the acute cervical spinal cat. The contributions of the lumbar sympathetic system and the adrenal glands to perfusion pressure responses to CO₂ were evaluated. Experiments were carried out in mongrel cats with acute cervical (C2) transections. Each cat had one leg denervated by cutting and stripping the lumbar sympathetic chain from L₁-L₇. In all cats each hindlimb was vascularly isolated and perfused with blood taken from the abdominal aorta. Bilateral adrenalectomy was performed on 8 animals. CO₂ administration for 5 minutes resulted in biphasic increases in the perfusion pressures of both legs which were designated peak1 (P1) and peak2 (P2). Increasing PCO₂ from 16 to 38mm Hg, and from 16 to 62mm Hg resulted in significant P1 and P2 responses of the innervated leg. This also resulted in a significant P2 response, and an observable but insignificant P1 response, of the denervated leg. Adrenalectomy reduced P1 and P2 responses of the innervated leg, and abolished the P1 response and reduced the P2 response of the denervated leg. In non-adrenalectomized cats increasing PCO₂ also resulted in a significant increase in systemic arterial pressure (SAP) with no changes in heart rate (HR). In adrenalectomized cats increasing PCO₂ resulted in an observable but non-significant increase in SAP and a significant decrease in HR. These results suggested that: 1) The P1 response was primarily a sympathetic neurogenic response to increased CO₂. 2) The P2 response was primarily a hormonal response to CO₂ in the denervated leg, and a combination of hormonal and sympathetic neurogenic responses to CO₂ in the innervated leg. 3) The adrenal glands were mostly involved in the P2 response to CO₂, but possibly had a small role in the P1 response. 4) Other non-adrenal vasoconstrictor hormones may have played a role in the P2 response to C0₂. 5) Likely, CO₂ initially activated the sympathetic system to directly increase neurogenic tone, perhaps by stimulating sympathetic afferent or efferent neurons, or hypothetical spinal chemosensitive regions. Progressively the adrenal and possibly other unidentified vasoconstrictor hormone systems became activated, either directly by CO₂ or indirectly by CO₂ mediated sympathetic activation. These hormone systems may have also played a role in CO₂ mediated maintenance of vascular tone.

Abstract:
More than a tool that provides mobility, the car is also a commodity with symbolic values related to the sense of self-regard. To various degrees, people pride themselves on being car owners or users. Based on a literature review primarily of a plethora of psychological theories, this thesis proposes a framework that defines the concept of car pride, examines the process of its formation, and classifies different types of car pride. Car pride is measured using data collected from a survey in Shanghai. Although household income and location are found to be significant predictors, car pride cannot be effectively explained by individual socio-economic characteristics. Car pride and ownership are found to be interdependent. The 2SLS model finds a significant impact of ownership on pride. Conversely, car pride can influence whether someone plans to buy a car, more so for first-time car buyers. In terms of the relationship between car pride and car use, the influence of car use on pride is weak, but car pride significantly stimulates car use. The relationship between car pride and other attitudinal factors is also investigated. A positive correlation between pride and dependence indicates people with high car pride are usually more dependent on cars. Public acceptance of car ownership restriction policies can also be subject to the influence of car pride. People with high car pride, though having strong desire for cars, tend to support policies that restrict car ownership, possibly because such policies make car ownership more exclusive and special. Car pride in Shanghai is compared with Beijing to reveal regional similarities and differences of car pride. It is found although overall car pride is similar in the two cities, car pride in Beijing has much more variation. Overall, this thesis provides a systematic understanding on its psychological structure, and its role in shaping people’s behavior (in terms of car ownership and use) and attitudes (in terms of car dependence and acceptance of car ownership restriction policies). It is suggested car pride can have important implications in addressing transportation issues such as traffic congestion and rapid motorization.

Imagine to be able to catch a little more sleep on your way to work or school, drive home after a night at the bar or watch a movie on a long road trip. All these things have never been possible before without someone else driving the car, until recent years. Autonomous cars or self-driving cars is being introduced to society more and more and will be the next big step in the progression of personal cars. There are a number of factors that decides how fast this new technology will be adopted. Safety, reliability, ethics and cost to name a few. This project will focus on the cost aspect of self-driving cars by examine if ultrasonic sensors can be used to develop a cheap self-driving car and thereby reach a broad customer base. To determine this a small scale prototype car was built and tested in a highway cruising situation and the results showed that the prototype was able to drive itself.
Tänk dig att kunna sova några extra minuter på väg till jobbet eller skolan, köra hem efter en natt i baren eller titta på film under en lång bilresa. Dessa saker har tidigare bara varit möjligt genom att ha en annan person som kör bilen, tills nu. Självstyrande bilar håller på att introduceras till samhället mer och mer och kommer vara det nästa stora steg som bilindustrin kommer ta. Det finns flera faktorer som bestämmer hur snabbt den här teknologin kommer adopteras. Säkerhet, pålitlighet, etik och kostnad för att nämna några. Det här projektet kommer att fokusera på kostnadsaspekten gällande självstyrande bilar, genom att undersöka om ultraljudssensorer kan användas vid utvecklandet av en självstyrande bil och på så sätt kunna hålla nere kostnaderna och nå en bredare kundbas. För att fastställa detta byggdes en småskalig prototypbil och testades i en simulation av motorvägskörning. Resultatet av testerna visar att bilen kunde köra sig själv med endast information från ultraljudssenorerna.

Abstract:
This report contains improvements of the radio controlled car which is developing an electronic differential and a speed controller. The report contains theory of engine modelling, steering and testing of these theories. Ackerman steering is used to get different steering radius of the inner and outer wheel when cornering. In order to regulate the velocity of the rear wheels they are individually controlled by implementing two separate controllers one for each wheel. To control the system PD-controllers is used for a fast rise time, two types of PD-controllers are tested, feed-back and feed-forward. A demonstrator was constructed to execute the two tests, one of the test was to determine the electronic differentials efficiency by measuring the cornering ability of the car with and without differential at different speeds. The second test was performed to determine which regulator gave the fastest controlling of the speed. This by exposing both controllers with different test scenarios. Based on our tests the conclusion is drawn that the electronic differential had a very positive impact on the turning radius. While testing, the controllers showed only a small difference, where the feed-forward controller was slightly faster in some of the tests.
Rapporten behandlar förbättringar av den radiostyrda bilen med fokus på utveckling av en elektronisk differential samt reglering av hastigheten. I rapporten kommer teori kring motormodellering och styrning, samt tester av dessa teorier. Ackerman-styrning användes för att få olika radie på inner och ytter hjulen vid kurvtagning. För att kunna reglera hasigheten på bakhjulen individuellt implementerades två separata regulatorer. För reglering av hastighet på bakhjulen användes PD-regulatorer för en snabb stigtid, två typer av PD-regulatorer testades en feed-back och en feed-forward. En demonstratör konstruerades för att möjliggöra tester av forskningsfrågorna. Det första testet utfördes för att undersöka den elektroniska differentialaxelns inverkan på bilens förmåga att ta kurvor i olika hastigheter. Det andra testet genomfördes för att undersöka vilken regulator som gav den snabbade regleringen av hastigheten. Detta genom att utsätta regulatorerna för olika test senarior. Utifrån testerna kunde slutsatsen dras att den elektroniska differentialen har en mycket stor positiv inverkan på bilens sväng radie. Medan testerna av regulatorerna endast gav en marginell skillnad men Feed-Forward regulator var något snabbare i en del av testerna.

As new challenges presents themselves continuously in today’s society the need for autonomous driving solutions is ever growing and with it: the need for creative solutions. This thesis investigates the pneumatic solution to making a car jump. The analysis of theoretical and testing results have demonstrated a correlation between the weight of the car and height of the jump. With the selected configuration it has been proven unattainable to make the car jump. Further research should look into the optimization of the pneumatic system, specifically the mass flow rate throughout the whole system.
I dagens samhälle presenteras nya utmaningar dagligen och behovet av autonoma lösningar i fordon ökar ständigt. I takt med det ökar behovet av kreativa lösningar. Detta examensarbete undersöker pneumatiska lösningen bakom att få en bil att hoppa. Analysen av teoritiska och experimentiella resultat visar en korrelation mellan bilens vikt och den möjliga hopphöjden. Med den valda konfigurationen är det inte möjligt att utföra hoppet och åtgärder tas upp i disskusionen. Vidare forskning bör undersöka optimeringen av det pneumatiska systemet, specifikt på hur det maximala massflödet kan uppnås i systemet.

El monto de inversión inicial que representa el proyecto es de S/ 77 096 soles, como inicio de operaciones aportado por los cuatro socios fundadores de APP MOBILE CAR. En la actualidad presentamos un escenario de mercado automotriz optimista con una industria cada vez más competitiva y dinámica donde la nueva tendencia van de la mano con era digital (App) que van revolucionando el mercado automotriz. Un estudio realizado por la empresa Brandwatch indica lo siguiente: Para el año 2020, un 40% de los nuevos consumidores de automóviles serán millennials. De ellos, un 88% navega por internet, complementando que hasta Abril del presente 2018 el mercado peruano ha crecido un 33.4 %. Esto representa estabilidad, solidez y crecimiento para APP MOBILE CAR donde se refleja optimismo de los consumidores generado por la estabilidad que estamos viviendo.
The present project is based on the analysis of the following problem: people who have a car and require maintenance service, but have limited time for what is proposed to provide a quick, efficient and trustworthy alternative solution, transferring the service which offers a fixed-site automotive workshop, to the point of user's location, whether this address or work. The initial investment amount represented by the project is S / 77 096 soles, as start of operations contributed by the four founding partners of APP MOBILE CAR. Currently we present an optimistic automotive market scenario with an increasingly competitive and dynamic industry where the new trend goes hand in hand with the digital era (App) that are revolutionizing the automotive market. A study conducted by the company Brandwatch indicates the following: By the year 2020, 40% of new car consumers will be millennials. Of these, 88% surf the Internet, complementing that until April of the present 2018 the Peruvian market has grown by 33.4%.
