Abstract:
ABSTRACT 3-Oxoadipate:succinyl-coenzyme A (CoA) transferase and 3-oxoadipyl-CoA thiolase carry out the ultimate steps in the conversion of benzoate and 3-chlorobenzoate to tricarboxylic acid cycle intermediates in bacteria utilizing the 3-oxoadipate pathway. This report describes the characterization of DNA fragments with the overall length of 5.9 kb from Pseudomonas sp. strain B13 that encode these enzymes. DNA sequence analysis revealed five open reading frames (ORFs) plus an incomplete one. ORF1, of unknown function, has a length of 414 bp. ORF2 (catI) encodes a polypeptide of 282 amino acids and starts at nucleotide 813. ORF3 (catJ) encodes a polypeptide of 260 amino acids and begins at nucleotide 1661. CatI and CatJ are the subunits of the 3-oxoadipate:succinyl-CoA transferase, whose activity was demonstrated when both genes were ligated into expression vector pET11a. ORF4, termed catF, codes for a protein of 401 amino acid residues with a predicted mass of 41,678 Da with 3-oxoadipyl-CoA thiolase activity. The last three ORFs seem to form an operon since they are oriented in the same direction and showed an overlapping of 1 bp between catI and catJ and of 4 bp between catJ and catF. Conserved functional groups important for the catalytic activity of CoA transferases and thiolases were identified in CatI, CatJ, and CatF. ORF5 (catD) encodes the 3-oxoadipate enol-lactone hydrolase. An incomplete ORF6 of 1,183 bp downstream of ORF5 and oriented in the opposite direction was found. The protein sequence deduced from ORF6 showed a putative AMP-binding domain signature.

Abstract:
ABSTRACT In the present study, we sought to elucidate the contribution of the Cryptococcus neoformans catalase gene family to antioxidant defense. We employed bioinformatics techniques to identify four members of the C. neoformans catalase gene family and created mutants lacking single or multiple catalase genes. Based on a phylogenetic analysis, CAT1 and CAT3 encode putative spore-specific catalases, CAT2 encodes a putative peroxisomal catalase, and CAT4 encodes a putative cytosolic catalase. Only Cat1 exhibited detectable biochemical activity in vitro, and Cat1 activity was constitutive in the yeast form of this organism. Although they were predicted to be important in spores, neither CAT1 nor CAT3 was essential for mating or spore viability. Consistent with previous studies of Saccharomyces cerevisiae, the single (cat1, cat2, cat3, and cat4) and quadruple (cat1 cat2 cat3 cat4) catalase mutant strains exhibited no oxidative-stress phenotypes under conditions in which either exogenous or endogenous levels of reactive oxygen species were elevated. In addition, there were no significant differences in the mean times to mortality between groups of mice infected with C. neoformans catalase mutant strains (the cat1 and cat1 cat2 cat3 cat4 mutants) and those infected with wild-type strain H99. We conclude from the results of this study that C. neoformans possesses a robust antioxidant system, composed of functionally overlapping and compensatory components that provide protection against endogenous and exogenous oxidative stresses.

Abstract:
Abstract Recent research has attempted to examine the proportion of interviewer variance that is due to interviewers systematically varying in their success in obtaining cooperation from respondents with varying characteristics (i.e., nonresponse error variance), rather than variance among interviewers in systematic measurement difficulties (i.e., measurement error variance) - that is, whether correlated responses within interviewers arise due to variance among interviewers in the pools of respondents recruited, or variance in interviewer-specific mean response biases. Unfortunately, work to date has only considered data from a CATI survey, and thus suffers from two limitations: Interviewer effects are commonly much smaller in CATI surveys, and, more importantly, sample units are often contacted by several CATI interviewers before a final outcome (response or final refusal) is achieved. The latter introduces difficulties in assigning nonrespondents to interviewers, and thus interviewer variance components are only estimable under strong assumptions. This study aims to replicate this initial work, analyzing data from a national CAPI survey in Germany where CAPI interviewers were responsible for working a fixed subset of cases.

Abstract:
Background: Feline demodicosis is considered an uncommon dermatopathy in cats that is mainly caused by the mite Demodex cati, but in few cases D. gatoi may be involved. Although the clinical aspects and pathogeny of feline demodicosis are not completely understood, its clinical expression is believed to be associated with the presence of primary immunosuppressive agents. Thus, the aim of this study is to report the diagnosis and treatment of an infested cat by D. cati, associated with mixed intestinal parasitic infection. Case: An approximately 1-year-old male short hair cat which was grown in a rural area was presented at the Veterinary Hospital of the Federal University of Pampa (Unipampa) in Uruguaiana, Rio Grande do Sul. The cat had alopecia, itching, excoriations on neck and head, and softened feces as clinical signs. Performed skin scrapings revealed eggs, larvae and adult forms of D. cati. In order to fnd possible immunosuppressive agents, exams for detection of feline immunodefciency virus (FIV) and feline leukemia virus (FeLV) were also performed. They revealed, however, negative results for FIV and FeLV infections. Moreover, coproparasitologic analysis revealed the presence of the intestinal parasites Trichuris sp., Ancylostoma sp., and Spirometra mansonoides. Thus, it was decided to proceed the treatment utilizing 0.2 mg/kg of moxidectin subcutaneously every four days and weekly shower with antiseptic shampoo containing 0.15% triclosan. In addition, it was prescribed oral doses of febendazol (50 mg/kg/day) for four days. After 20 days of treatment, the skin lesions had decreased signifcantly and the skin scrapings were negative for D. cati. In this way, the showers with 0.15% triclosan was suspended. Nevertheless, the treatment with moxidectin was maintained for more fours weeks until totalclinical healing was achieved with hair growth and absence of skin lesions. Furthermore, after the treatment with febendazol the animal feces had reached a normal consistency and new coproparasitologic analysis revealed negative results for the presence of intestinal parasites. Discussion: The diagnosis of Demodex cati was based on clinical history, physical examination (alopecia, excoriation in the head and neck) and microscopic visualization of the mite in skin scrapings. The treatment used with moxidectin, acaricidal quite safe and effective, widely used in treatment of canine demodicosis, but no reports about the use in cats. The mite D. cati is considered normal resident of cat’s skin, and the pathogenesis of demodicosis is still unknown. It is known that the playback exacerbated mite can present with skin lesions is commonly associated with immunosuppression states. The presence of three different intestinal parasites (Spirometra mansonoides, Trichuris sp., and Ancylostoma sp.) might have been essential for an immunosuppressive status of the cat and clinical development of demodicosis. Thus, this report supports the hypothesis that a primary immunosuppressive agent is necessary for clinical development of feline demodicosis. Nevertheless, further researches are necessary in order to determine the prevalence of feline demodicosis and correlate the presence of cestodes and development of this disease. In addition, this report highlights the therapeutic success utilizing na association of moxidectin and topic treatment with the treatment of possible immunosuppressive agents. Keywords: dermatopathy, imunossuppression, cat, therapy

Abstract:
Whether computer assisted data collection methods should be used for survey data collection is no longer an issue. Most professional research organizations, commercial, government and academic, are adopting these new methods with enthusiasm. Computer assisted telephone interviewing (CATI) is most prevalent, and computer assisted personal interviewing (CAPI) is rapidly gaining in popularity. Also, new forms of electronic reporting of data using computers, telephones and voice recognition technology are emerging. This paper begins with a taxonomy of current computer assisted data collection methods. It then reviews conceptual and theoretical arguments and empirical evidence on such topics as: (1) respondents and interviewer acceptance of new techniques, (2) effect of computer assisted interviewing on data quality, (3) consequences for survey costs and (4) centralized vs. decentralized deployment of CATI.

Abstract:
Abstract Prospect theory states that the influential power of avoiding negative outcomes is stronger than that of achieving positive outcomes. In a survey context, this theory has been tested with respect to not only participation in a CATI survey, but also giving consent to data linkage in CATI surveys. No study, however, has tested the theory with respect to participation in a CAPI or web survey. This study does so in a mixed-mode panel context; it also tests the moderating effects of time-in-panel, response history, and mode protocol. Results show that the framing of the survey participation request influences participation propensity in a way consistent with prospect theory, but only for relatively recent panel entrants. The opposite effect is found for long-term panel participants. No difference is found between mode protocols.

Investigations of the relationships between the gut microbiota and gastrointestinal parasitic nematodes are attracting growing interest by the scientific community. These studies have however been carried out mainly in humans and experimental animals, while knowledge of the make-up of the gut commensal microbiota in presence or absence of infection by parasitic nematodes in domestic animals is limited. In this study, we investigate the qualitative and quantitative impact that infections by a widespread parasite of cats (i.e. Toxocara cati) exert on the gut microbiota of feline hosts. The faecal microbiota of cats with patent infection by T. cati (= Tc+), as well as that of negative controls (= Tc-) was examined via high-throughput sequencing of the V3-V4 hypervariable region of the bacterial 16S rRNA gene, followed by bioinformatics and biostatistical analyses of sequence data. A total of 2,325,366 useable high-quality sequences were generated from the faecal samples analysed in this study and subjected to further bioinformatics analyses, which led to the identification of 128 OTUs and nine bacterial phyla, respectively. The phylum Firmicutes was predominant in all samples analysed (mean of 53.0%), followed by the phyla Proteobacteria (13.8%), Actinobacteria (13.7%) and Bacteroidetes (10.1%). Among others, bacteria of the order Lactobacillales, the family Enterococcaceae and genera Enterococcus and Dorea showed a trend towards increased abundance in Tc+ compared with Tc- samples, while no significant differences in OTU richness and diversity were recorded between Tc+ and Tcsamples (P = 0.485 and P = 0.581, respectively). However, Canonical Correlation and Redundancy Analyses were able to separate samples by infection status (P = 0.030 and P = 0.015, respectively), which suggests a correlation between the latter and the composition of the feline faecal microbiota.

This dissertation is the result of an ethnographic journey undertaken in three spiritual centers of Umbanda , located in different districts of Natal/RN. The aim of the study was to understand how the representations of exus (spirit category) are established in the studied religious environment. In order to study this, mythical narratives of songs and of people were analyzed. In addition, the incorporated spirit entities, loaded with codes and values, were possible to be read and understood. The present work was developed according to a theoretical and methodological approach that interprets culture as a constant construction process, with representations and significations. The empirical research focuses on the ethnographic observations and the narratives; providing the experimentation of the models of discursive dialogues and polyphony. It was observed that the studied empirical field is marked by the cult of the Jurema which influences the mythical construction of exus . Therefore, this study enabled a reflection on the meeting of notions of two distinct worlds ( Umbanda and Jurema ). This reflection is crucial to the understanding of the position that exus occupy in the local umbandista system

Abstract:
Toxocara canis and T. cati are parasitic nematodes of dogs and cats, transmitted by the ingestion of embryonated eggs, by the transmamary and transplacentary routes or by predation of paratenic hosts harboring third-stage larvae in their bodies. The transplacentary route occurs only in T. canis infection. Many parasites have mechanisms to change the host\'s behavior in order to ensure their transmission. Several researchers have demonstrated the occurrence of behavioral changes, using mice as a model of paratenic host for T. canis. However, there are no studies on the occurrence of changes in behavior of Rattus norvegicus experimentally infected with T. cati. The aims of this study were to investigate the distribution of T.cati larvae in R. norvegicus and determine the myotropic and neurotropic phases in the infection of this parasite, and to compare behavior of male and female rodents experimentally infected with either T. canis or T. cati, in the acute and chronic periods of infection. The variables measured were: anxiety, motor activity and muscle strength. Initially we used 21 rats aged eight weeks, infected with 300 eggs of T. cati. On days 3, 5, 8, 10, 15, 30 and 60 post-infection three animals were killed to count the larvae in the organs and muscle. In the second experiment 50 females and 50 males of R. norvegicus, with six to eight weeks of age were used. For each sex the animals were divided into three groups: T. canis - 20 rats infected with 300 eggs of T. canis, T.cati -20 rats infected with 300 eggs of T. cati and control - 10 rats infection free. On days 5, 15, 40 and 70 post-infection, the infected animals and control groups were submitted to evaluation of behavioral variables and determination of forepaws\' muscle strength. It was verified that the neurotropic phase of larvae occurred mainly in the 15th dpi and 30 dpi. Myotropic phase occurred during all experimental period, but especially 15th and 60th dpi. In regard to muscle strength in females, a significant difference in the three groups was observed only after 40th post-infection. In males there was significant difference among the three groups throughout the experiment. In the behavioral variables, only the female of the group infected with T. canis showed significant differences after 40th dpi when compared to control group. Males showed no significant differences in the performance evaluation. It can be concluded that T. cati migration behavior was different, R. norvegicus regardless of sex showed greater decrease in muscle strength when infected with T. cati and only infected females by T. canis showed behavioral changes remaining more time under conditions of exposure in the open field.

This study aims to show how craziness is represented in the Brazilian novels Quincas Borba (1891), by the Rio writer Machado de Assis, O louco do Cati (1942), by the writer Dyonélio Machado, from Rio Grande do Sul, and O Grande Mentecapto (1979), by Fernando Sabino. Considering these three narratives, located in a period that extends from the end of the nineteenth century to the end of the twentieth century, the objective is to investigate the strategies and narrative resources used by the narrators/authors to present the protagonists Rubião, Cati, and Geraldo Viramundo as crazy individuals, unsuited to the social reality that they encounter and, consequently, marginalized. Understanding the novel Quincas Borba, by Machado de Assis, as a formal and thematic matrix for the representation of craziness, this research also has the intention of highlighting the dialogue that this novel establishes with its successors O louco do Cati and O Grande Mentecapto, pointing out similarities and differences found among them. As a theoretical support, Michael Foucault’s studies on psychiatry and its relation to power and knowledge, as well as the critical fortune of each author and work discussed, contribute to this thesis. The results show that the three novels use elements typical of the Dionysian literary tradition, such as: the masquerading game of narrators, dialogism, polyphony, humor through the tragicomic narrative form, among other resources, especially in the case of Machado de Assis. Through the creation of a singular narrative form, each of the three works represents craziness so that it is capable of problematizing the lack of logic of discourses, institutions, politics, religiosity, psychiatric presuppositions and current social organization, at different times. In this trajectory of the Brazilian Literature, Machado de Assis was the one who gave the first and very significant steps, insofar as the protagonists’ craziness undresses the Brazilian social craziness and exposes the games of power concealed under human relations and under the sociabilities.

The pegmatite rocks in Rio Grande do Norte are responsible for much of the production of industrial minerals like quartz and feldspar. Quartz and feldspar are minerals from pegmatite which may occur in pockets with metric to centimetric dimensions or as millimetric to sub millimetric intergrowths. The correct physical liberation of the mineral of interest, in case of intergrowths, requires an appropriate particle size, acquired by size reduction operations. The method for treating mineral which has a high efficiency fines particles recovery is flotation. The main purpose of the present study is to evaluate the recovery of quartz and potassium feldspar using cationic diamine and quaternary ammonium salt as collectors by means of dissolved air flotation DAF. The tests were performed based on a central composite design 24, by which the influence of process variables was statistically verified: concentration of the quaternary ammonium salt and diamine collectors, pH and conditioning time. The efficiency of flotation was calculated from the removal of turbidity of the solution. Results of maximum flotation efficiency (60%) were found in the level curves, plotted in conditions of low concentrations of collectors (1,0 x 10-5 mol.L-1). These high flotation efficiencies were obtained when operating at pH 4 to 8 with conditioning time ranging from 3 to 5 minutes. Thus, the results showed that the process variables have played important roles in the dissolved air flotation process concerning the flotability of the minerals.

AbstractThis chapter discusses the Stichting Amsterdamse Zwerfkatten (Amsterdam Stray Cat Foundation, afterwards SAZ), who work with and for stray cats. In their practices and views they challenge common assumptions about cat subjectivity and agency, the cats’ right to a habitat and social relations, as well as the idea that there is a strict difference between cats and humans. Their approach offers an alternative way of thinking about cat agency and human agency, networks of cat-human relations, sharing the city with cats, and working towards more freedom for cats and humans. In the chapter I examine these relations from the perspective of agency, care, and politics, and investigate whether or not this can function as a model for building new communities with other animals, which centres their agency.

Abstract This work presents an interface for tolerance analysis in a CAD system. A method for picking up necessary information from a 2D drawing is developed and implemented as an interface in a commercial CAD system. The interface communicates with an external calculation program which determines unknown tolerance limits using the normal distribution model. Results from the calculation program is in the end used by the interface to present measures with tolerances on the drawing. The advantage of using CATI in preliminary design is discussed, and a strategy for treating interrelated tolerance chains is presented.

Abstract:
Feline coronavirus (FCoV) is ubiquitous in the domestic cat (Felis catus) population. The aim of this study was to determine the prevalence and potential predisposing factors of FCoV in cats of an animal shelter in Latvia and to compare the prevalence between cats in the quarantine area and resident cats in the adoption area. Oropharyngeal and faecal swabs and blood samples were collected from 40 domestic shorthair cats from an animal shelter in Jelgava, Latvia. Swabs were analyzed for FCoV RNA by reverse transcriptase-polymerase chain reaction (RT-PCR). Blood serum samples were tested for FCoV specific antibodies by indirect enzyme-linked immunosorbent assay (ELISA). FCoV RT-PCR positivity in oropharyngeal and rectal swabs was 7.5% (3/40) and 72.5% (29/40), respectively. Additionally, FCoV seroprevalence was 67.5% (27/40). The proportion of cats shedding FCoV within the adoption (72.7%) and quarantine (72.2%) areas was similar (p = 0.55). The prevalence of FCoV faecal shedding in young cats was significantly higher (p < 0.05) than in adult cats. Sex had no significant effect on FCoV RT-PCR positivity. Further studies on larger cat population including different population types are needed to determine the overall prevalence and epidemiological patterns of FCoV infection in Latvia.

Abstract:
On the basis of the current state of research, this study identifies and validates futureoriented competencies. Using quantitative social research methods, it then investigates demands in industry. This involved, first, an evaluation of literature and the identification of competencies for research and innovation (R&I) activities. Next, from clusters of the identified competencies I derived 14 different types. On this basis, I generated a competency profile that informs the development of a tool for R&I, the Wheel of Competencies. With this newly developed tool specific competency profiles can be generated and analysed. Second, I operationalized and implemented the competence components in a questionnaire. On this basis, 200 CEOs and heads of research and development (R&D) departments of medium-sized and large enterprises in Germany were surveyed in November and December 2021 using computer-assisted telephone interviews (CATI). All enterprises have at least 50 employees and an in-house R&D department. In addition, enterprises have to belong to one of the following industry sectors: automotive, chemical, electrical or mechanical engineering. The results show that certain competencies are in very high demand across all industries, while others are more specific to an industry sector. Overall, the results indicate that the competencies in demand address the dynamic complexity in collaborative R&I processes. The results presented here make an important evidencebased contribution to curriculum development in engineering education based on future-oriented competencies and illustrates which transfer activities and collaborative formats are increasingly relevant.
